Diese Erfindung bezieht sich auf ein neues Verfahren zur Herstellung von 4-(6-Methoxy-2-naphthyl)-butan-2-on.
Die US-PS Nr. 4 061 779 beschreibt 4-(6-Methoxy-2-naphthyl)-butan-2-on (British Approved Name Nabumetone) und seine Verwendung bei der Behandlung von rheumatischen und arthritischen Erkrankungen. Eine Anzahl von Verfahren zur Herstellung der Verbindung wird ebenfalls beschrieben.
Es wurde nun ein weiteres Verfahren zur Herstellung von Nabumetone gefunden, das durch eine durch Palladium katalysierte Addition von 3-Buten-2-ol an ein 6-Halogen-2-methoxynaphthalin oder 6-Methoxy-2-naphthoylchlorid gekennzeichnet ist.
Demgemäss bezieht sich die vorliegende Erfindung auf ein Verfahren zur Herstellung von 4-(6-Methoxy-2-naphthyl)-butan-2-on, das dadurch gekennzeichnet ist, dass man eine Verbindung der Formel (I):

Der Ausdruck "Halogen" umfasst Iod, Brom oder Chlor, vorzugsweise Brom oder Iod.
Der Palladiumkatalysator kann Palladium selbst oder eine Palladium(II)-Verbindung aufweisen. Geeignete Palladium(II)-Verbindungen umfassen Palladium(II)-acetat und Palladium(II)-halogenide, wie Palladium(II)-chlorid, -bromid oder -iodid. In einer bevorzugten Ausführungsform der Erfindung enthält die Palladium(II)-Verbindung auch einen geeigneten komplexbildenden Liganden, wie Triphenylphosphin oder Triethylamin, am bevorzugtesten Triphenylphosphin.
Wenn Palladium als Katalysator verwendet wird, besteht eine bevorzugte physikalische Form aus Palladium auf Kohle, wie 5%iges Palladium auf Kohle.
Das Verfahren wird zweckmässig in Gegenwart eines Aktivators für den Katalysator ausgeführt, und Natriumiodid oder Iod ist besonders brauchbar.
Wenn X in Formel (I) für Halogen steht, wird die Reaktion zweckmässig bei einer erhöhten Temperatur von 50 bis 200 DEG C, vorteilhaft um 120 DEG C, in einem inerten Lösungsmittel, wie Dimethylformamid, ausgeführt. Wenn Triethylamin verwendet wird, kann überschüssiges Triethylamin als Lösungsmittel verwendet werden, oder alternativ kann das Lösungsmittel Acetonitril sein.
Wenn X für Halogen steht, ist es auch vorteilhaft, ein alkalisches Material, vorzugsweise Natriumcarbonat, zu verwenden, um zur Neutralisation der während der Reaktion gebildeten Halogenwasserstoffsäure beizutragen. Vorzugsweise wird ein molarer Überschuss von mindestens 20% über die stöchiometrische Menge Natriumcarbonat verwendet, was dazu führt, dass die Ausbeuten an dem gewünschten Produkt verbessert werden.
Wenn X in Formel (I) für die Gruppe der Formel:

steht, wird die Reaktion vorzugsweise bei einer Temperatur von ca. 100 bis 150 DEG C während ca. 0,5 bis 5 Stunden unter Stickstoff und in Gegenwart einer Reaktionsbase, wie eines tertiären Amins, und in einem inerten Lösungsmittel ausgeführt. Geeignete inerte Lösungsmittel sind Xylol oder Toluol, und ein geeignetes Amin ist N-Benzyldimethylamin. Ein Vorteil dieser Reaktion besteht darin, dass Xylol und Toluol billige, leicht zurückgewinnbare Lösungsmittel sind und dass das tertiäre Amin ebenfalls leicht zurückgewinnbar ist.
Verbindungen der Formel (I), in denen X für Halogen steht, sind bekannte Verbindungen oder werden nach Methoden hergestellt, die analog zu denjenigen sind, die für die Herstellung von strukturell ähnlichen bekannten Verbindungen angewandt werden. Die Verbindung der Formel (I), worin X für -COCl steht, ist neu und kann aus 2-Acetyl-6-methoxynaphthalin (das selbst eine bekannte Verbindung ist) gemäss Standardmethoden (siehe G.W. Gray und B. Jones, J. Chem. Soc., 678, 1954 und R. Gay und A. Horeau, Bull. Soc. Chim. France, 622, 1953) hergestellt werden.
Die folgenden Beispiele erläutern die Erfindung.
Beispiel 1

2-Brom-6-methoxy-naphthalin (0,82 g), Palladiumacetat (0,009 g), Triphenylphosphin (0,03 g), Natriumcarbonat (0,1 g) und 3-Buten-2-ol (0,43 g) wurden in DMF (3 ml) gelöst und ca. 17 Stunden lang auf 120 DEG C erhitzt. Das Gemisch wurde in verdünnte HCl gegossen und mit Ethylacetat extrahiert, worauf der Extrakt gewaschen, getrocknet und eingedampft wurde und ein \l ergab. Dieses wurde auf zwei präparative Dünnschicht-Siliciumdioxidplatten aufgebracht und im Verlauf von 4 Stunden mit einem Gemisch aus 20% Ethylacetat und 80% Benzin vom Siedebereich 80 bis 100 DEG C eluiert. Die richtige Zone, die mit einer authentischen Probe identifiziert wurde, wurde von dem Träger abgekratzt und mit Methanol extrahiert. Das Lösungsmittel wurde verdampft, wobei das rohe Produkt (0,05 g von einer Platte), Schmelzpunkt 77 bis 79 DEG C, erhalten wurde.
Beispiel 2
Ein Gemisch aus 2-Brom-6-methoxy-naphthalin (4,1 g, 17,3 mMol), Palladiumacetat (0,045 g, 0,2 mMol), Triphenylphosphin (0,15 g, 0,55 mMol), Natriumcarbonat (1,1 g, 10,38 mMol), 3-Buten-2-ol (2,15 g, 30 mMol) und Dimethylformamid (15 ml) wurde unter einer Stickstoffatmosphäre 17 Stunden lang auf 120 DEG C erhitzt. Das abgekühlte Reaktionsgemisch wurde unter Rühren in Wasser (250 ml) gegossen, und der resultierende Niederschlag wurde abfiltriert und mit Wasser (30 ml) gewaschen. Das rohe Produkt wurde erneut in Methylendichlorid (50 ml) suspendiert, ge trocknet (MgSO4) und durch Glasfaserpapier filtriert, und das Filtrat wurde zu einer hellbraunen gummiartigen Festsubstanz eingedampft (3,42 g, 87% Gewichtsausbeute).
Die Analyse dieser Festsubstanz zeigte das Vorhandensein von Nabumetone in 35,4%iger Reinheit.
Beispiel 3
6-Methoxy-2-naphthoylchlorid (2,2 g, 10 mMol), Palladiumacetat (0,0225 mg, 0,1 mMol), 3-Buten-2-ol (0,88 g, 12,25 mMol) und N-Benzyldimethylamin (1,69 g, 12,25 mMol) wurden unter einer Stickstoffatmosphäre in p-Xylol (20 ml) 2,5 Stunden lang auf 130 DEG C erhitzt. Das Reaktionsgemisch wurde auf Raumtemperatur abgekühlt, und das ausgefällte N-Benzyldimethylamin-hydrochloridsalz wurde abfiltriert und mit p-Xylol (5 ml) gewaschen. Das mit den Waschflüssigkeiten vereinigte Filtrat wurde nacheinander mit 2-normaler Salzsäure (2 x 15 ml), gesättigter Natriumhydrogencarbonatlösung (20 ml) und Wasser (20 ml) gewaschen. Die organische Phase wurde getrocknet (MgSO4), filtriert und zu einer beigefarbigen halbfesten Substanz eingedampft (1,7 g, 74,7% Gewichtsausbeute).
Die Analyse durch HPLC zeigte, dass sie 31,5% Nabumetone enthielt, was eine Aktivitätsausbeute von 23,5%, bezogen auf das Naphthoylchlorid, ergab.
Beispiel 4
Ein Gemisch aus 2-Brom-6-methoxy-naphthalin (4,74 g, 20 mMol), But-3-en-2-ol (2,16 g, 30 mMol), Palladiumacetat (0,081g, 0,36 mMol), Natriumiodid (0,5 g, 3,3 mMol), Natriumcarbonat (1,27 g, 12 mMol) und Dimethylformamid (20 ml) wurde unter einer Stickstoffatmosphäre 5 Stunden lang auf 140 DEG C erhitzt. Das Reaktionsgemisch wurde auf Raumtemperatur abgekühlt und unter Rühren in Wasser (250 ml) gegossen.
Nach 15 Minuten wurde der hellgraue Niederschlag abfiltriert und mit Wasser (2 x 40 ml) gewaschen und unter Vakuum bei 45 DEG C getrocknet. Das Produkt wog 4,2 g und enthielt 68,5% Nabumetone, was eine Aktivitätsausbeute von 63,1% ergab.

A further process for the preparation of nabumetones has now been found, which is characterized by a palladium-catalyzed addition of 3-buten-2-ol to a 6-halogen-2-methoxynaphthalene or 6-methoxy-2-naphthoyl chloride.

The term "halogen" includes iodine, bromine or chlorine, preferably bromine or iodine.
The palladium catalyst can have palladium itself or a palladium (II) compound. Suitable palladium (II) compounds include palladium (II) acetate and palladium (II) halides, such as palladium (II) chloride, bromide or iodide. example 1

The cooled reaction mixture was poured into water (250 ml) with stirring and the resulting precipitate was filtered off and washed with water (30 ml). The crude product was resuspended in methylene dichloride (50 ml), dried (MgSO4) and filtered through glass fiber paper and the filtrate was evaporated to a light brown gummy solid (3.42 g, 87% weight yield).
Analysis of this solid showed the presence of nabumetone in 35.4% purity.
Example 3
6-methoxy-2-naphthoyl chloride (2.2 g, 10 mmol), palladium acetate (0.0225 mg, 0.1 mmol), 3-buten-2-ol (0.88 g, 12.25 mmol) and N -Benzyldimethylamine (1.69 g, 12.25 mmol) was heated to 130 ° C in p-xylene (20 ml) under a nitrogen atmosphere for 2.5 hours. The reaction mixture was cooled to room temperature and the precipitated N-benzyldimethylamine hydrochloride salt was filtered off and washed with p-xylene (5 ml). The filtrate combined with the washing liquids was washed successively with 2N hydrochloric acid (2 × 15 ml), saturated sodium hydrogen carbonate solution (20 ml) and water (20 ml). The organic phase was dried (MgSO4), filtered and evaporated to a beige semi-solid substance (1.7 g, 74.7% weight yield).
Analysis by HPLC showed that it contained 31.5% nabumetone, giving an activity yield of 23.5% based on the naphthoyl chloride.
Example 4
A mixture of 2-bromo-6-methoxy-naphthalene (4.74 g, 20 mmol), but-3-en-2-ol (2.16 g, 30 mmol), palladium acetate (0.081 g, 0.36 mmol) ), Sodium iodide (0.5 g, 3.3 mmol), sodium carbonate (1.27 g, 12 mmol) and dimethylformamide (20 ml) were heated at 140 ° C. for 5 hours under a nitrogen atmosphere. The reaction mixture was cooled to room temperature and poured into water (250 ml) with stirring.
After 15 minutes, the light gray precipitate was filtered off and washed with water (2 × 40 ml) and dried under vacuum at 45 ° C. The product weighed 4.2 g and contained 68.5% nabumetone, giving an activity yield of 63.1%.
