Verfahren zur Herstellung von Derivaten von polyhydrischen Alkoholen
Die vorliegende Erfindung bezieht sich auf ein Verfahren zur Herstellung von neuen polyhydrischen Alkoholen der Formel CH3SO20CH2-(CHOH) n-CH20SO2CHS in welcher n 2, 3 oder 4 ist, das dadurch gekennzeichnet ist, dass entweder ein Alkohol der Formel HOCH2-(CHOH)11-CH2OH mit Methansulfonylchlorid in Gegenwart einer als Säureakzeptor wirkenden organischen tertiären Base oder ein bis-Epoxyd der Formel

worin m 0, 1 oder 2 ist, mit Methansulfonsäure umgesetzt, die Reaktionsmischung mit einem Acetylierungsmittel und einer organischen tertiären Base als Säureakzeptor behandelt und das erhaltene Acetylderivat der Formel CH3SO2OCH2-(CHG-CO-CH3)11-CH2OSO2CH3 hierauf der Hydrolyse unterworfen wird.
Von den Verbindungen, die gemäss dem Verfahren der Erfindung hergestellt werden, sind insbesondere
1 ,6-Dimethansulfonyl-mannit,
1 ,4-Dimethansulfonyl-erythrit und
1 ,5-Dimethansulfonyl-ribit von Interesse.

worin m 0 oder 2 ist mit der Methansulfonsäure, wird zweckmässig in einem inerten Lösungsmittel durchgeführt.
Man kann z. B. einen Alkohol der Formel HOCH2-(CHOH)3-CH2OH mit dem Methansulfonylchlorid in Gegenwart einer organischen tertiären Base umsetzen, die Reaktionsmischung mit Essigsäureanhydrid und einer organischen tertiären Base behandeln und das erhaltene Acetylderivat der Formel CH3SO2OCH2 (CHO-COCH3) 3OCH2SO2CH3 der Säurehydrolyse unterwerfen, worauf die Verbindung der Formel CH3SO2OCH2-(CHOH)8-CH2OSO2CH0 entsteht.
Vorzugsweise ist der Säureakzeptor Pyridin oder ein Pyridinabkömmling.
1,6- Dimethansulfonyl - mannit verhindert das Wachstum des Walker-256-Rattentumors bei einer Dosierung, die offensichtlich keine Anzeichen von Toxizität zeigt. Bei Applikation an Menschen wird die Knochenmarkfunktion herabgesetzt. Die genannte Verbindung ist deshalb von beachtlichem Interesse als Arzneimittel zur Behandlung von Leucaemia. Die andern Verbindungen, die gemäss der Erfindung erhalten werden, zeigen ähnliche Wirkungsweisen.
Beispiele
1. Eine gerührte Lösung von D-Mannit (36 g, 0,20 Mol) in siedendem Pyridin (200 ml) wurde auf 30 abgekühlt und auf dieser Temperatur gehalten, während Methansulfonylchlorid (48 g, 0,42 Mol) tropfenweise während 2 Stunden zugegeben wurde.
Nach weiteren 3 Stunden Rühren bei 5o wurde die klare Lösung mit einer eisgekühlten Mischung von Essigsäureanhydrid (82 g, 0,80 Mol) und Pyridin (150 ml) behandelt und hierauf für weitere 20 Stunden ohne Rühren bei Zimmertemperatur belassen.
Die erhaltene Lösung wurde langsam unter kräftigem Umrühren zu Eiswasser (1,5 1) zugegeben, der so ausgefällte Festkörper abgetrennt, gewaschen, getrocknet und aus einer Mischung von Äthanol und Aceton (500 ml: 50 ml) umkristallisiert, wobei rohes 1,4 - Dimethansulfonyl - 2,3,4,5 - tetracetyl-D- mannit (45 g) vom Smp. 145-147" erhalten wurde.
Dieses Produkt (21 g) wurde während 1 Stunde mit 0,55n Chlorwasserstoff in absolutem Methanol (800 ml) unter Rückfluss zum Sieden erhitzt. Das Lösungsmittel wurde hierauf bei 20 mm (während 2 Stunden) entfernt, wobei ein schwachgelbliches öl erhalten wurde, welches sich beim Trocknen (20 Stunden) in einem Vakuumexsikkator über Schwefelsäure und Natriumhydroxyd verfestigte. Der so erhaltene Feststoff wurde sukzessive mit einer Mischung von Methanol und Äther (20 ml: 60 ml) und Äthylacetat (20 ml) verrieben, um anhaftendes Ö1 zu entfernen, wobei 7,5 g von rohem 1,6-Dimethan- sulfonyl-D-mannit erhalten wurde. Nach Umkristallisation aus Äthylacetat/j2ithanol (50 mol: 80 ml) wurde das reine Produkt vom Smp. 132-133" erhalten.
2. 10 g D-Ribit wurden in 100 ml warmem anhydrischem Pyridin gelöst und 15 ml Methansulfonylchlorid unter Rühren während 41/2 Stunden bei einer Temperatur, die 0" nicht überschritt, zugegeben. Die Mischung wurde während weiterer 11/2 Stunden gerührt und bildete dann eine klare Lösung. 20 ml Essigsäureanhydrid gemischt mit 40 mI Pyridin wurden hierauf unter Rühren zugegeben und die Mischung während 3 Stunden bei 0" belassen, wonach derselben ermöglicht wurde, sich auf Raumtemperatur zu erwärmen und diese während weiterer 15 Stunden stehengelassen wurde. Nach Abkühlen auf 0" wurden 50 ml Wasser von 0" zugegeben und das Ganze gerührt, so dass eine klare Lösung erhalten wurde, welche in 500 ml Wasser, welches mit Eis gekühlt war, gegossen wurde.
Das ausgefällte Öl wurde durch Dekantieren getrennt und unter Eiskühlung mit einer Mischung von Äthyl- acetat und Petroläther (60-800) verrieben. Der so erhaltene kristalline Festkörper wurde von anhaftendem Öl befreit, indem er auf einen porösen Teller aufgepresst wurde, wonach er einem Smp. von 80 bis 850 zeigte. Der- 1 ,5-Dimethansulfonyl-2,3 ,4-tri- acetyl-D-ribit wurde schliesslich in reiner Form (Ausbeute 8,2 g) erhalten, indem er wiederholt aus einer Mischung von Äthylacetat und Petroläther und schliesslich aus Methanol umkristallisiert wurde.

8,2 g 1 ,5-Dimethansulfonyl-2,3,4-triacetyl-D-ribit wurden mit 250 ml n2 anhydrischer Lösung von Salzsäure in Methanol während 1 Stunde erhitzt und das Lösungsmittel und die Salzsäure unter vermindertem Druck verflüchtigt, wonach roher 1 ,5-Di- methansulfonyl-D-ribit erhalten wurde.
3. 36 g Erythrit wurden in 230 ml warmem Pyridin gelöst, auf 0" abgekühlt und die Mischung mit 50 ml Methylsulfonylchlorid tropfenweise unter Rühren während einer Zeit von 4 Stunden behandelt. Die Mischung wurde auf ungefähr 0" gehalten und das Rühren während ll,/2 Stunden nach der Zugabe des Methansulfonylchlorids fortgesetzt. 112 ml Essigsäureanhydrid, gemischt mit 150 ml Pyridin, wurden hierauf unter Rühren bei von zugegeben, die Mischung auf Raumtemperatur kommen lassen und hierauf dieselbe während 18 Stunden stehengelassen.
Die Mischung wurde hierauf mit 2,5 1 Wasser, welches Eis enthielt, verdünnt und den erhaltenen Niederschlag abfiltriert. Dieser wurde mit 1,5 1 siedendem Dioxan extrahiert und der unlösliche Rückstand verworfen. Der Extrakt enthielt 39 g rohes 1 ,4-Di- methansulfonyl-2,3-diacetyl-erythrit. 39 g dieses Produktes wurden zu 1,5 1 0,54n methanolischer Salzsäure zugegeben und die Mischung unter Rühren während 51/2 Stunden zum Sieden unter Rückfluss erhitzt. Nach Abfiltrieren des unlöslichen Rückstandes wurden das Methanol und die Salzsäure unter reduziertem Druck entfernt und das ölige Produkt schliesslich im Vakuum über Schwefelsäure und Natriumhydroxyd getrocknet.
Das kristalline Produkt wurde zweimal aus Äthylacetat umkristallisiert, wobei 10,5 g von reinem 1,4-Dimethansulfonyl-erythrit vom Smp. 122-124" erhalten wurden.
Es können als Ausgangsstoffe natürlich auch die entsprechenden optisch aktiven Verbindungen, das heisst die Stereoisomeren, sowie die razemischen Mischungen verwendet werden.

wherein m is 0, 1 or 2, reacted with methanesulfonic acid, the reaction mixture is treated with an acetylating agent and an organic tertiary base as an acid acceptor and the resulting acetyl derivative of the formula CH3SO2OCH2- (CHG-CO-CH3) 11-CH2OSO2CH3 is then subjected to hydrolysis.
Of the compounds made according to the process of the invention, in particular are
1, 6-dimethanesulfonyl mannitol,
1, 4-dimethanesulfonyl-erythritol and
1, 5-dimethanesulfonyl-ribitol of interest.

where m is 0 or 2 with the methanesulfonic acid, is conveniently carried out in an inert solvent.
You can z. B. react an alcohol of the formula HOCH2- (CHOH) 3-CH2OH with the methanesulfonyl chloride in the presence of an organic tertiary base, treat the reaction mixture with acetic anhydride and an organic tertiary base and the resulting acetyl derivative of the formula CH3SO2OCH2 (CHO-COCH3) 3OCH2SO2CH3 of the acid hydrolysis subject, whereupon the compound of the formula CH3SO2OCH2- (CHOH) 8-CH2OSO2CH0 is formed.
Preferably the acid acceptor is pyridine or a pyridine derivative.
1,6-dimethanesulfonyl-mannitol prevents the growth of Walker-256 rat tumor at a dosage that apparently shows no signs of toxicity. When administered to humans, bone marrow function is reduced. The aforesaid compound is therefore of considerable interest as a medicament for the treatment of leucemia. The other compounds obtained according to the invention show similar modes of action.
Examples
1. A stirred solution of D-mannitol (36 g, 0.20 mol) in boiling pyridine (200 ml) was cooled to 30 and maintained at that temperature while methanesulfonyl chloride (48 g, 0.42 mol) was added dropwise over 2 hours was admitted.
After stirring for a further 3 hours at 50, the clear solution was treated with an ice-cold mixture of acetic anhydride (82 g, 0.80 mol) and pyridine (150 ml) and then left for a further 20 hours at room temperature without stirring.
The resulting solution was slowly added to ice water (1.5 l) with vigorous stirring, the solid thus precipitated was separated off, washed, dried and recrystallized from a mixture of ethanol and acetone (500 ml: 50 ml), with crude 1.4 - Dimethanesulfonyl-2,3,4,5-tetracetyl-D-mannitol (45 g) of m.p. 145-147 ".
This product (21 g) was refluxed under reflux for 1 hour with 0.55N hydrogen chloride in absolute methanol (800 ml). The solvent was then removed at 20 mm (for 2 hours), a pale yellowish oil being obtained which solidified on drying (20 hours) in a vacuum desiccator over sulfuric acid and sodium hydroxide. The solid thus obtained was successively triturated with a mixture of methanol and ether (20 ml: 60 ml) and ethyl acetate (20 ml) in order to remove adhering oil, 7.5 g of crude 1,6-dimethanesulfonyl-D -mannite was obtained. After recrystallization from ethyl acetate / ethanol (50 mol: 80 ml) the pure product with a melting point of 132-133 "was obtained.
2. 10 g of D-ribitol were dissolved in 100 ml of warm anhydrous pyridine and 15 ml of methanesulfonyl chloride were added with stirring over 41/2 hours at a temperature not exceeding 0 ". The mixture was stirred and formed for a further 11/2 hours then a clear solution. 20 ml of acetic anhydride mixed with 40 ml of pyridine were then added with stirring and the mixture left at 0 "for 3 hours, after which it was allowed to warm to room temperature and allowed to stand for a further 15 hours. After cooling to 0 ", 50 ml of 0" water were added and the whole was stirred, so that a clear solution was obtained, which was poured into 500 ml of water which had been cooled with ice.
The precipitated oil was separated by decanting and triturated with a mixture of ethyl acetate and petroleum ether (60-800) while cooling with ice. The crystalline solid obtained in this way was freed from adhering oil by being pressed onto a porous plate, after which it had a melting point of 80 to 850. The 1,5-dimethanesulfonyl-2,3,4-triacetyl-D-ribitol was finally obtained in pure form (yield 8.2 g) by repeatedly recrystallizing from a mixture of ethyl acetate and petroleum ether and finally from methanol has been.

8.2 g of 1,5-dimethanesulfonyl-2,3,4-triacetyl-D-ribitol were heated with 250 ml of n2 anhydrous solution of hydrochloric acid in methanol for 1 hour and the solvent and the hydrochloric acid were evaporated under reduced pressure, after which crude 1 , 5-dimethanesulfonyl-D-ribitol was obtained.
3. 36 g of erythritol was dissolved in 230 ml of warm pyridine, cooled to 0 "and the mixture treated with 50 ml of methylsulfonyl chloride dropwise with stirring over a period of 4 hours. The mixture was kept at approximately 0" and the stirring was maintained for 11/16 Continued for 2 hours after the addition of the methanesulfonyl chloride. 112 ml of acetic anhydride mixed with 150 ml of pyridine were then added with stirring at from, the mixture was allowed to come to room temperature and then left to stand for 18 hours.
The mixture was then diluted with 2.5 l of water which contained ice and the resulting precipitate was filtered off. This was extracted with 1.5 l of boiling dioxane and the insoluble residue discarded. The extract contained 39 g of crude 1,4-dimethanesulfonyl-2,3-diacetyl-erythritol. 39 g of this product were added to 1.5 liters of 0.54N methanolic hydrochloric acid and the mixture was heated to boiling under reflux for 51/2 hours while stirring. After the insoluble residue had been filtered off, the methanol and the hydrochloric acid were removed under reduced pressure and the oily product was finally dried in vacuo over sulfuric acid and sodium hydroxide.
The crystalline product was recrystallized twice from ethyl acetate, 10.5 g of pure 1,4-dimethanesulfonyl-erythritol with a melting point of 122-124 "being obtained.
The corresponding optically active compounds, that is to say the stereoisomers, and the racemic mixtures can of course also be used as starting materials.
