Verfahren zur Herstellung eines Lokalanästhetikums
Durch die Arbeiten von N. Löfgren (Studies on Local Anaesthetics, Xylocaine, 1948) sind wertvolle Lokalanästhetika bekanntgeworden, die sich von Dialkylaminoacetaniliden herleiten, welche ihrerseits mindestens in 2- und 6-Stellung je einen Alkylrest von höchstens 4 C-Atomen tragen und die ausserdem noch in einer oder mehreren der verbleibenden Stellungen durch je einen solchen Rest substituiert sein können.
Von den untersuchten Verbindungen dieser Art zeichnen sich vor allem das 2,6-Dimethyl-N-diäthylamino- acetanilid durch raschen Wirkungseinritt aus. Die weitere Bearbeitung des Gegenstandes ergab, dass diese günstige Eigenschaft schon beim Ersatz der beiden Methyl- durch Athylgruppen weitgehend verlorengeht. Das gleiche gilt für andere, höher substi- tuierte, in 2,6-Stellung Ethylreste tragende Derivate, wie z. B. für das 2,6-Diäthyl-4-methyl-N-diäthyl- aminoacet-anilid.
Es wurde nun gefunden, dass man ein wertvolles Lokalanästhetikum erhält, wenn man das 2-Methyl6-äthyl-anilin in das 2-Methyl-6-äthyl-N-diäthylaminoacet-anilid bzw. seine Salze überführt. Es wurde nämlich die überraschende Beobachtung gemacht, dass das 2-Methyl-6-äthyl-N-diäthylaminoacet-anilid und seine Salze, d. h. diejenige Verbindung, bei der nur eine ringständige Methylgruppe durch einen Athylrest ersetzt ist und die bisher in der Literatur noch nicht beschrieben wurde, einen gleich raschen Wirkungs- eintritt zeigt wie die 2,6-Dimethylverbindung. Hinsichtlich Wirkungsdauer und örtlicher Verträglichkeit sind beide Stoffe gleichwertig. In seiner Allgemeinverträglichkeit weist jedoch das 2-Methyl-6-äthyl-Ndiäthylaminoacet-anilid gegenüber der 2,6-Dimethylverbindung Vorteile auf.
Die neue Verbindung hat gegenüber dem 2,6 Dimethyl-N- diäthylaminoacet-anilid ausserdem den Vorteil, dass das entsprechende 2-Methyl-6-äthyl-anilin viel leichter zugänglich ist als das 2,6-Dimethyl- anilin. Letzteres kann nur mit einer Ausbeute von etwa 130/0 aus m-Xylol erhalten werden. Im Gegensatz dazu lässt sich das 2-Methyl-6-äthyl-anilin nach dem Verfahren des deutschen Patentes Nr. 951501 mit über 90 0/o iger Ausbeute durch Einwirkung von Äthylen auf das leicht zugängliche o-Toluidin erhalten. Das 2-Methyl-6-äthyl-N-diäthyl-aminoacet-anilid zeigt weiterhin der 2,6-Dimethyl-verbindung gegen über den Vorteil, dass es mit Phosphorsäure ein wohlkristallisiertes, wasserlösliches, beständiges und nichthygroskopisches Phosphat bildet. Dadurch lässt es sich unschwer in grosser Reinheit herstellen.
Von be sonderem Wert für die pharmazeutische und medizinische Praxis ist die leichte Handhabung dieses Salzes, die die rasche Herstellung von Lösungen sehr erleichtert.
Man erhält das Produkt, wenn man 2-Methyl-6 äthyl-anilin oder eines seiner Salze, wie z. B. ein Hydrohalogenid, mit Diäthylaminoessigsäure oder mit einem reaktionsfähigen Derivat mit funktionell abgewandelter Carboxylgruppe, wie z. B. ihrem Anhydrid, einem ihrer Ester oder ihrer Halogenide oder ihrem Amid, umsetzt. Im letztgenannten Falle verwendet man zweckmässig das Hydrochlorid des 2-Methyl-6 äthyl-anilins.
Das 2-Methyl-6-äthyl-N-diäthylamino-acet-anilid stellt farblose Kristalle vom F. 55o dar. Es löst sich in Gegenwart von Mineralsäuren oder organischen Säuren unter Bildung der entsprechenden Salze leicht in Wasser. Besonders charakteristisch ist sein Monophosphat, das durch Umsetzung der Base mit Phosphorsäure in alkoholischer Lösung leicht rein mit einem Schmelzpunkt von 178-180" erhältlich ist.
Beispiel
49 g Natrium werden in 500 cm3 Ligroin (Kp760 120130 ) unter Zusatz von 0,1 g Ölsäure bei 100-110" so dispergiert, dass die Natrium-Teil chengrösse 15 m, u nicht überschreitet. Dazu tropft man im Verlauf von 15 Minuten 112,5 g Chlorbenzol, kühlt auf 200 ab, fügt 135 g 2-Methyl-6-äthylanilin hinzu, rührt 10 Minuten nach und tropft unter Kühlung bei 20-40" 72,5 g N-Diäthylaminoessigsäuremethylester ein. Nachdem man 30 Minuten nachgerührt hat, zersetzt man mit Wasser, fügt verdünnte Salzsäure hinzu, trennt die Kohlenwasserstoffschicht ab, setzt aus der salzsauren Lösung die Base durch Zufügen von Kaliumcarbonatlösung in Freiheit und äthert aus.
Bei der Destillation im Vakuum erhält man 2 Fraktionen: Kp7 90-95", 2-Methyl-6 äthyl-anilin, 70 g Kp5 165-170", N-Diäthylamino essigsäure-(2-methyl 6-äthyl-anilild), 106 g = 93,5 O/o d. Th. Phosphat F. 178-180".
Method for producing a local anesthetic
Through the work of N. Löfgren (Studies on Local Anesthetics, Xylocaine, 1948), valuable local anesthetics have become known which are derived from dialkylaminoacetanilides, which in turn have an alkyl radical of at least 4 carbon atoms in at least 2 and 6 positions and which can also be substituted in one or more of the remaining positions by such a radical.
Of the compounds of this type investigated, 2,6-dimethyl-N-diethylaminoacetanilide in particular is distinguished by its rapid onset of action. Further processing of the subject showed that this favorable property is largely lost when the two methyl groups are replaced by ethyl groups. The same applies to other, more highly substituted derivatives bearing ethyl radicals in the 2,6-position, such as. B. for the 2,6-diethyl-4-methyl-N-diethyl aminoacet anilide.
It has now been found that a valuable local anesthetic is obtained if 2-methyl-6-ethyl-aniline is converted into 2-methyl-6-ethyl-N-diethylaminoacet-anilide or its salts. The surprising observation was made that 2-methyl-6-ethyl-N-diethylaminoacet-anilide and its salts, i.e. H. the compound in which only one methyl group in the ring has been replaced by an ethyl radical and which has not yet been described in the literature shows the same rapid onset of action as the 2,6-dimethyl compound. With regard to the duration of action and local compatibility, both substances are equivalent. In terms of its general tolerance, however, 2-methyl-6-ethyl-N-diethylaminoacetanilide has advantages over the 2,6-dimethyl compound.
The new compound also has the advantage over 2,6-dimethyl-N-diethylaminoacet-anilide that the corresponding 2-methyl-6-ethyl-aniline is much more easily accessible than 2,6-dimethyl-aniline. The latter can only be obtained with a yield of about 130/0 from m-xylene. In contrast to this, 2-methyl-6-ethyl-aniline can be obtained by the process of German Patent No. 951501 with a yield of over 90% by the action of ethylene on the easily accessible o-toluidine. The 2-methyl-6-ethyl-N-diethyl-aminoacet-anilide also has the advantage over the 2,6-dimethyl compound that it forms a well-crystallized, water-soluble, stable and non-hygroscopic phosphate with phosphoric acid. This makes it easy to produce with great purity.
The easy handling of this salt, which makes the rapid preparation of solutions very easy, is of particular value for pharmaceutical and medical practice.
The product is obtained if 2-methyl-6-ethyl-aniline or one of its salts, such as. B. a hydrohalide, with diethylaminoacetic acid or with a reactive derivative with a functionally modified carboxyl group, such as. B. their anhydride, one of their esters or their halides or their amide. In the latter case, the hydrochloride of 2-methyl-6-ethyl-aniline is expediently used.
The 2-methyl-6-ethyl-N-diethylamino-acet-anilide is colorless crystals of F. 55o. It dissolves easily in water in the presence of mineral acids or organic acids with the formation of the corresponding salts. Its monophosphate is particularly characteristic, which can easily be obtained in pure form with a melting point of 178-180 "by reacting the base with phosphoric acid in an alcoholic solution.

49 g of sodium are dispersed in 500 cm3 of ligroin (Kp760 120130) with the addition of 0.1 g of oleic acid at 100-110 "in such a way that the sodium particle size does not exceed 15 μm. 112 is added dropwise over the course of 15 minutes , 5 g of chlorobenzene, cools to 200, adds 135 g of 2-methyl-6-ethylaniline, stirs for 10 minutes and 72.5 g of methyl N-diethylaminoacetate are added dropwise with cooling at 20-40 ". After stirring for 30 minutes, the mixture is decomposed with water, dilute hydrochloric acid is added, the hydrocarbon layer is separated off, the base is released from the hydrochloric acid solution by adding potassium carbonate solution and ether is removed.
Distillation in vacuo gives 2 fractions: bp7 90-95 ", 2-methyl-6-ethyl-aniline, 70 g bp5 165-170", N-diethylamino acetic acid (2-methyl 6-ethyl-aniline), 106 g = 93.5 o / o d. Th. Phosphate F. 178-180 ".
