Verfahren zur Herstellung eines Lokalanästhetikums
Durch die Arbeiten von N. Löfgren (Studies on Local Anaesthetics, Xylocaine, 1948) sind wertvolle Lokalanästhetika bekanntgeworden, die sich von Dialkyl-aminoacetaniliden herleiten, welche ihrerseits mindestens in 2- und stellung je einen Alkylrest von höchstens 4 C-Atomen tragen und die ausserdem noch in einer oder mehreren der verbleibenden Stellungen durch je einen solchen Rest substituiert sein können.
Von den untersuchten Verbindungen dieser Art zeichnen sich vor allem das 2,6-Dimethyl-N-diäthylamino- acetanilid durch raschen Wirkungseintritt aus. Die weitere Bearbeitung des Gegenstandes ergab, dass diese günstige Eigenschaft schon beim Ersatz der beiden Methyl- durch Äthylgruppen weitgehend verlorengeht.
Das gleiche gilt für andere, höher substituierte, in 2,6-Stellung Äthylreste tragende Derivate, wie z. B. für das 2,6-Diäthyl-4-methyl-N-diäthylaminoacet-anllid.
Es wurde nun gefunden, dass 2-Methyl-6-äthyl-Ndiäthylaminoacet-anilid ein wertvolles Lokalanästhetikum ist. Es wurde nämlich die überraschende Beobachtung gemacht, dass das 2-Methyl-6-äthyl-N-diäthylaminoacet-anilid und seine Salze, d. h. diejenige Verbindung, bei der nur eine ringständige Methylgruppe durch einen Äthylrest ersetzt ist und die bisher in der Literatur noch nicht beschrieben wurde, einen gleich raschen Wirkungseintritt zeigt wie die 2,6-Dimethylverbindung. Hinsichtlich Wirkungsdauer und örtlicher Verträglichkeit sind beide Stoffe gleichwertig. In seiner Allgemeinverträglichkeit weist jedoch das 2-Methyl 6-äthyl-N-diäthylaminoacet-anilid gegenüber der 2,6 Dimethylverbindung Vorteile auf.
Gegenstand des vorliegenden Patentes ist ein Ver- fahren zur Herstellung eines Lokalanästhetikums, welches dadurch gekennzeichnet ist, dass man 2-Methyl6-äthyl-N-acetanilid, das in der N-Acetylgruppe einen reaktionsfähigen Substituenten trägt, mit Diäthylamin umsetzt, wobei 2-Methyl-6-äthyl-N-diäthylaminoacetanilid gebildet wird.
Die neue Verbindung hat gegenüber dem 2,6 Dimethyl-N-diäthylaminoacet-anilid ausserdem den Vorteil, dass das entsprechende 2-Methyl-6-äthyl-anilin viel leichter zugänglich ist als das 2,6-Dimethyl-anilin.
Letzteres kann nur mit einer Ausbeute von etwa 13% aus m-Xylol erhalten werden. Im Gegensatz dazu lässt sich das 2-Methyl-6-äthyl-anilin nach einem andern Verfahren mit über 900/0iger Ausbeute durch Einwirkung von Äthylen auf das leichtzugängliche o-Toluidin erhalten. Das 2-Methyl-6-äthyl-N-diäthylaminoacetanilid zeigt weiterhin der 2,6-Dimethylverbindung gegenüber den Vorteil, dass es mit Phosphorsäure ein wohlkristallisiertes, wasserlösliches, beständiges und nichthygroskopisches Phosphat bildet. Dadurch lässt es sich unschwer in grosser Reinheit herstellen. Von besonderem Wert für die pharmazeutische und medizinische Praxis ist die leichte Handhabung dieses Salzes, die die rasche Herstellung von Lösungen sehr erleichtert.
Als reaktionsfähigen Substituenten kann die Acetylgruppe beispielsweise ein Halogenatom oder eine Alkoxy-, Aralkoxy- oder Aryloxygruppe aufweisen.
Die Umsetzung z. B. eines 2-Methyl-6-äthyl-N-halogenacet-anilids mit Diäthylamin erfolgt zweckmässig in einem wasserfreien, organischen Lösungsmittel, wie Benzol, Toluol oder andern.
Das 2-Methyl-6-äthyl-N- diäthylaminoacet- anilid stellt farblose Kristalle von F. 55" dar. Es löst sich in Gegenwart von Mineralsäuren oder organischen Säuren unter Bildung der entsprechenden Salze leicht in Wasser. Besonders charakteristisch ist sein Monophosphat, das durch Umsetzung der Base mit Phosphorsäure in alkoholischer Lösung leicht rein mit einem Schmelzpunkt von 178-180 erhältlich ist.
Beispiel
In eine Mischung aus 230 cm3 gesättigter Natriumacetatlösung, 230 cm3 Eisessig und 68 g 2-Methyl-6 äthyl-anilin wird unter Rühren bei 2-8" 85 g Chloracetylchlorid eingetropft, 2 Stunden nachgerührt, dann abgesaugt, mit Wasser gewaschen. Durch Umkristallisieren aus 50%igem Alkohol erhält man das 2-Methyl6-äthyl-chloracet-anilid in langen Nadeln vom F.

Zu einer Dispersion von 48 g 2-Methyl-6-äthylchloracet-anilid in 200 cm3 Alkohol werden 37 g wasserfreies Diäthylamin zugegeben und 4 Stunden am Rückfluss gekocht. Darauf wird der Alkohol und überschüssiges Diäthylamin im Vakuum abdestilliert und der Rückstand in 250 cm3 100/0iger Salzsäure gelöst, mit A-Kohle abgesaugt und das Filtrat mit einer konzentrierten wässrigen Lösung von 85 g Pottasche versetzt. Dann wird mit Äther ausgeschüttelt, die Ätherlösung mit Pottasche getrocknet und das Lösungsmittel abdestilliert. Durch Vakuumdestillation erhält man das 2-Methyl-6-äthyl-N-diäthylaminoacet-anilid in Form weisser Nadeln vom F. 55" und Kp. l 157-160 .
Aus der Lösung von 10 g 2-Methyl-6-äthyl-Ndiäthylaminoacet-anilid in 30 cm3 Alkohol kristallisiert nach Zugabe der berechneten Menge alkoholischer, etwa 2m-Phosphorsäure das 2-Methyl-6-äthyl-N-di äthylaminoacet-anilid-monophosphat aus. Das Salz wird abgesaugt, mit Alkohol gewaschen und getrocknet. Die Ausbeute beträgt 13 g. Smp. 178-180 .

Method for producing a local anesthetic
Through the work of N. Löfgren (Studies on Local Anesthetics, Xylocaine, 1948), valuable local anesthetics have become known which are derived from dialkylaminoacetanilides, which in turn have an alkyl radical of at least 4 carbon atoms in at least 2 and positions and which can also be substituted in one or more of the remaining positions by such a radical.
Of the compounds of this type investigated, 2,6-dimethyl-N-diethylaminoacetanilide in particular is distinguished by its rapid onset of action. Further processing of the object showed that this favorable property is largely lost when the two methyl groups are replaced by ethyl groups.
The same applies to other, more highly substituted derivatives bearing ethyl radicals in the 2,6-position, such as. B. for the 2,6-diethyl-4-methyl-N-diethylaminoacet-anllid.
It has now been found that 2-methyl-6-ethyl-N-diethylaminoacet-anilide is a valuable local anesthetic. The surprising observation was made that 2-methyl-6-ethyl-N-diethylaminoacet-anilide and its salts, i.e. H. that compound in which only one methyl group in the ring has been replaced by an ethyl radical and which has not yet been described in the literature shows the same rapid onset of action as the 2,6-dimethyl compound. With regard to the duration of action and local compatibility, both substances are equivalent. In terms of its general tolerance, however, 2-methyl 6-ethyl-N-diethylaminoacetanilide has advantages over the 2,6-dimethyl compound.
The subject of the present patent is a process for the production of a local anesthetic, which is characterized in that 2-methyl6-ethyl-N-acetanilide, which has a reactive substituent in the N-acetyl group, is reacted with diethylamine, with 2-methyl -6-ethyl-N-diethylaminoacetanilide is formed.
The new compound also has the advantage over 2,6-dimethyl-N-diethylaminoacet-anilide that the corresponding 2-methyl-6-ethyl-aniline is much more easily accessible than 2,6-dimethyl-aniline.
The latter can only be obtained from m-xylene with a yield of about 13%. In contrast, 2-methyl-6-ethyl-aniline can be obtained in another process with a yield of over 900/0 by the action of ethylene on the easily accessible o-toluidine. The 2-methyl-6-ethyl-N-diethylaminoacetanilide also has the advantage over the 2,6-dimethyl compound that it forms a well-crystallized, water-soluble, stable and non-hygroscopic phosphate with phosphoric acid. This makes it easy to produce with great purity. The easy handling of this salt, which makes the rapid preparation of solutions very easy, is of particular value for pharmaceutical and medical practice.
The acetyl group can have, for example, a halogen atom or an alkoxy, aralkoxy or aryloxy group as reactive substituents.
The implementation z. B. a 2-methyl-6-ethyl-N-haloacet-anilide with diethylamine is conveniently carried out in an anhydrous, organic solvent such as benzene, toluene or others.
The 2-methyl-6-ethyl-N-diethylaminoacetanilide is colorless crystals of F. 55 ". In the presence of mineral acids or organic acids it dissolves easily in water with the formation of the corresponding salts. Its monophosphate, the can easily be obtained pure with a melting point of 178-180 by reacting the base with phosphoric acid in alcoholic solution.

37 g of anhydrous diethylamine are added to a dispersion of 48 g of 2-methyl-6-ethylchloroacetanilide in 200 cm3 of alcohol and the mixture is refluxed for 4 hours. The alcohol and excess diethylamine are then distilled off in vacuo and the residue is dissolved in 250 cm3 of 100% hydrochloric acid, suctioned off with activated charcoal and the filtrate is treated with a concentrated aqueous solution of 85 g of potash. Then it is extracted with ether, the ether solution is dried with potash and the solvent is distilled off. 2-Methyl-6-ethyl-N-diethylaminoacet-anilide is obtained by vacuum distillation in the form of white needles with a diameter of 55 "and a boiling point of 157-160.
From the solution of 10 g of 2-methyl-6-ethyl-N-diethylaminoacet-anilide in 30 cm3 of alcohol, 2-methyl-6-ethyl-N-diethylaminoacet-anilide-monophosphate crystallizes after adding the calculated amount of alcoholic, about 2m-phosphoric acid out. The salt is filtered off with suction, washed with alcohol and dried. The yield is 13 g. M.p. 178-180.
