in welcher einer der Reste Ri, R2 und Rs eine Carboxylgruppe, die übrigen Alkylreste oder Wasserstoff bedeuten, in umsetzungs fähige Säureabkömmlinge, z. B.
Säurechloride, und Umsetzung dieser Verbindungen mit sekundären Aminen erhält man dialkyl- oder arylalkylsubatitierte Amide von Isoxazolcar- bonsäuren, welche wertvolle therapeutische Eigenschaften aufweisen.
Es wurde nun gefunden, dass auch Di- alkyl- und Arylalkylamide von Säuren, deren Carboxylgruppe durch eine oder mehrere Methylengruppen vom Isoxazolkern getrennt ist, eine hervorragende analeptisohe Wirkung besitzen. Diese ist gegenüber den nach dem Verfahren des Patentes 18580 und seiner Zusätze gewonnenen Verbindungen wesent lich verstärkt.
Zur Darstellung der neuen Verbindungen lässt man in der Carboxylgruppe umsetzungs fähige Abkömmlinge der Homologen der Isoxazolcarbonsäuren, wie Säurechloride, -bro- mide, -anhydride oder -ester, auf die ent sprechenden sekundären Amine einwirken.
Gegenstand des vorliegenden Patentes ist ein Verfahren zur Darstellung von 3,5-Di- methylisoxazol-4-essigsäurediäthylamid, wel ches dadurch gekennzeichnet ist, dass ein in der Carboxylgruppe umsetzungsfähiger Ab kömmling der 3,5-Dimethylisoxazol-4-essig- säure auf Diäthylamin einwirken gelassen wird.
Das bisher unbekannte 3,5-Dimethyliso- xazol-4-essigsäurediäthylamid schmilzt bei 59 . Es ist in Wasser nur wenig, leicht in Benzol und Äther löslich. Es übt schon in Dosen von 0,5 mg pro kg eine Wirkung auf die Atmung des morphinisierten Kaninchens aus, während die letale Dosis an der Maus nur 50 mg pro kg beträgt.
Die neue Verbindung soll als Arznei mittel verwendet werden.
Beispiel: Man lässt im Verlauf einiger Stunden 17,3 Teile 3,5-Ditnethylisoxazol-4-essigsäure- chlorid unter Rühren zu 40 Teilen Benzol, 7,8 Teilen Diäthylamin und 20 Teilen 20 0io- iger Natronlauge zulaufen und hält durch Kühlung mit Kältemischung die Temperatur unter + 10 . Wenn alles umgesetzt ist, fügt man weitere 20 Teile konzentrierte Natron lauge zu, trennt die Benzollösung ab und trocknet sie mit 1 Teil Kaleiumcarbonat, filtriert und dampft ein. Der Rückstand wird bei vermindertem Druck destilliert.
Unter 12 mm Druck geht bei 180 0 ein farbloses <B>01</B> über, das nach einiger Zeit erstarrt.

Acid chlorides, and reaction of these compounds with secondary amines, gives dialkyl- or arylalkyl-substituted amides of isoxazolecarboxylic acids, which have valuable therapeutic properties.
It has now been found that dialkyl and arylalkylamides of acids whose carboxyl group is separated from the isoxazole nucleus by one or more methylene groups also have an excellent analeptic effect. This is significantly increased compared to the compounds obtained by the process of patent 18580 and its additives.
To prepare the new compounds, derivatives of the homologues of the isoxazole carboxylic acids, such as acid chlorides, bromides, anhydrides or esters, capable of converting in the carboxyl group are allowed to act on the corresponding secondary amines.
The subject of the present patent is a process for the preparation of 3,5-dimethylisoxazole-4-acetic acid diethylamide, which is characterized in that a carboxyl group convertible from derivative of 3,5-dimethylisoxazole-4-acetic acid to diethylamine is allowed to act.
The previously unknown 3,5-dimethylisoxazole-4-acetic acid diethylamide melts at 59. It is only slightly soluble in water and easily soluble in benzene and ether. Even at doses of 0.5 mg per kg, it has an effect on the respiration of the morphinized rabbit, while the lethal dose in the mouse is only 50 mg per kg.
The new compound is intended to be used as a medicine.
Example: 17.3 parts of 3,5-diethylisoxazole-4-acetic acid chloride are allowed to run in over a few hours with stirring to 40 parts of benzene, 7.8 parts of diethylamine and 20 parts of 20% sodium hydroxide solution and hold by cooling with a cold mixture the temperature below + 10. When everything has been implemented, a further 20 parts of concentrated sodium hydroxide solution are added, the benzene solution is separated off and dried with 1 part of potassium carbonate, filtered and evaporated. The residue is distilled under reduced pressure.
Under 12 mm pressure, a colorless <B> 01 </B> passes over at 180 0, which solidifies after a while.
