in welcher einer der Reste R, R2 und R, eine Carboxylgruppe, die übrigen Alkylreste oder Wasserstoff bedeuten, in umsetzungs fähige Säureabkömmlinge, z. B.
Säurechlo ride, und Umsetzung dieser Verbindungen mit sekundären Aminen erhält man dialkyl- i oder arylalkyl@substituierte Amide von Isox- azolcarbonsäuren, welche wertvolle thera- peutische Eigenschaften aufweisen.
Es wurde nun gefunden, dass auch Di- alkyl- und Arylalkylamide von Säuren, deren Carbogylgruppe durch eine oder mehrere Methylengruppen vom Isoxazolkern getrennt ist, eine hervorragende analeptische Wir- kung besitzen. Diese ist gegenüber den nach dem Verfahren,des Patentes Nr. 185280 und seiner Zusätze gewonnenen Verbindungen wesentlich verstärkt.
Zur Darstellung,der neuen Verbindungen ,läBt man in der Carboxylgruppe umsetzungs- fähige Abkömmlinge der Homologen der Isoxazolcarbonsäuren, wie Säurechloride, -bromide, -anhydride oder -ester, auf die entsprechenden: sekundären Amine einwirken.
Gegenstand .des vorliegenden Patentes. ist ein Verfahren zur Darstellung von 3.,5-Di- methyln-soxazol-4-essigsäuremethylanilid, wel ches dadurch gekennzeichnet ist, dass ein in der Carboxylgruppe umsetzungsfähiger Ab kömmling der 3,5-Dimethylisoxazol-4-essig- säure auf Methylanilin einwirken ,gelassen wird.
Das bisher unbekannte 3,5-Dimethyl- isoxazol-4-essigsä.uremethylanilid bildet Pris men vom Schmelzpunkt<B>63-65'.</B> Sie sind in Wasser nur wenig, leicht in Alkohol, Benzol und Äther löslich.
Die neue Verbindung soll als Arzneimit tel verwendet werden.
<I>Beispiel</I> .T Man erhitzt 18,3 Teile 3,5-Dimethyl- isoxazol-4-essigsäureäthylester mit 10,7 Tei- l-en Methylanilin während 8 Stunden in einer Stickstoffatmosphäre auf<B>160'.</B> Die Reaktionsmasse wird mit Säure und Äther gesclhüttelt, die ätherische Lösung mit wenig Wasser gewaschen, mit Chlorcaleium ge trocknet und eingedampft. Der Rückstand wird unter vermindertem Druck destilliert.
Unter 13 mm Druck geht bei 190-200' ein helles viskoses 01 über. Es wird in wenig absolutem Äther gelöst und vorsichtig mit Petroläther versetzt, worauf derbe Prismen auskristallisieren.
<I>Beispiel 2:</I> 11,4 Teile 3,5-Dimethylisoxazol-4-essig- säure werden mit 33 Teilen Thionylchlorid versetzt und 4 Stunden bei 40' gehalten.
Man destilliert das überschüssige Thionyl- chl.orid im Vakuum ab und vertreibt die letzten Reste desselben durch Zufügen und Abdestillieren von Toluod. Das zurückblei bende Säurechlorid nimmt man in 50 Tei- len Benzol auf und lässt es zu einer Lösung von 14,5 Teilen Methylanilin in 50 Teilen Benzol laufen. Man hält noch 1-2 Stunde bei 60 . Dabei fällt ein dicker Niederschlag von Methy lanilin-hydrochlorid aus.
Es wird ab- genutscht. Das Filtrat wäscht man zweimal mit gesättigter Kochsalzlösung aus und dampft das Lösungsmittel ab. Man nimmt den Rückstand in wenig absolutem Äther auf, fügt Petroläther hinzu, solange noch keime bleibende Trübung entsteht. Auf )ja- impfen fallen gut ausgebildete Kristalle aus. Sie werden nochmals aus wenig absolutem Äther umkristallisiert.

Acid chlorides, and reaction of these compounds with secondary amines, gives dialkyl- i or arylalkyl @ substituted amides of isoxazole carboxylic acids, which have valuable therapeutic properties.
It has now been found that dialkyl and arylalkylamides of acids whose carbogyl group is separated from the isoxazole nucleus by one or more methylene groups also have an excellent analeptic effect. Example 2: 11.4 parts of 3,5-dimethylisoxazole-4-acetic acid are mixed with 33 parts of thionyl chloride and kept at 40 ° for 4 hours.
The excess thionyl chloride is distilled off in vacuo and the last remains of it are driven off by adding and distilling off toluod. The remaining acid chloride is taken up in 50 parts of benzene and allowed to run into a solution of 14.5 parts of methyl aniline in 50 parts of benzene. Hold at 60 for another 1-2 hours. A thick precipitate of Methy laniline hydrochloride falls out.
It is sucked off. The filtrate is washed twice with saturated sodium chloride solution and the solvent is evaporated off. The residue is taken up in a little absolute ether, and petroleum ether is added as long as the cloudiness remains germic. Well-formed crystals precipitate out. They are recrystallized again from a little absolute ether.
