Procédé de fabrication du 1-(p-chlorophényl)-1-(2-pyridyl)-3-diméthylaminopropane. Le présent brevet a pour objet un procédé de préparation du 1-(p-chlorophényl)-1-(2- pyridyl)-3-diméthyl-aminopropane. Ce procédé est caractérisé en ce qu'on fait réagir une 2-métal-pyridine avec la 4-chloro-p-diméthyl- amino-propiophénone pour obtenir le 4-chloro- phényl- (2-pyridyl) -p-diméthylaminoéthyl- car- binol,
on fait réagir ce carbinol avec un agent halogénant de manière à obtenir l'halogénure correspondant, après quoi on traite cet halo- génure avec un agent. déshydrohalogénant, pour obtenir le 1-(p-chlorophényl)-1-(2- pyridyl)-3-diméthyl-aminopropène-1, qui est transformé en 1- (p - chlorophényl) -1- (2 pyridyl)-3-diméthylaminopropane par hydro génation.
Le produit ainsi obtenu bout à 153 à 158 C sous une pression de 2-3 mm; il possède une activité antihistaminique et anti- anaphylactique très élevée. On peut utiliser le composé obtenu con formément à l'invention sous forme de base libre ou sous forme de ses sels avec des acides minéraux tels que les acides chlorhydrique, bromhydrique, sulfurique et phosphorique ou avec des acides organiques tels que les acides salicylique, tartrique, maléique, succinique, ci trique et lactique.
On peut utiliser ce composé sous diverses formes pharmaceutiques telles que comprimés pour l'administration orale, crèmes pour applications topiques et solutions injectables. Les sels sont de préférence utilisés sous forme de crèmes; les solutions injectables seront for mées de sels non toxiques. <I>Exemple:</I> Le p-chlorophényl-(2-pyridyl)-p-dim6th@rl- aminoéthyl-carbinol est préparé de la façon suivante: Du chlorhydrate de 4-chloro-p-diméthyl- aminopropiophénone (0,1 mole) est dissous dans 50 ml d'eau et refroidi dans -un bain de glace.
La base libre est libérée dans la glace avec une solution à 10 % de carbonate de soude et d'huile est extraite à l'éther. La solu tion éthérée est lavée avec de l'eau et séchée sur du. carbonate dé potasse, puis l'éther est éliminé, en donnant ; la base libre.
On prépare une solution de 0,2 mole de 2-pyridyllithium dans 250 ml d'éther et, après l'avoir refroidie jusqu'à -40 C; l'on ajoute une solution de 18 g de 4-chloro-p- diméthyl- aminopropiophénone dans 50 ml d'éther, goutte à goutte, en agitant, en l'espace d'une demi-heure. Aussitôt la réaction achevée, on laisse la température monter jusqu'à -15 C, le mélange de réaction étant agité à cette-tem- pérature pendant une heure.
Le contenu du flacon est décomposé avec de la glace et de l'acide. chlorhydrique, puis rendu basique avec de l'ammoniaque gazeux. L'huile obtenue est extraite à l'éther, l'éther est ensuite éva poré et le résidu est distillé. Le carbinol est un sirop visqueux jaune, bouillant à une tem pérature de 196-200 C/2 mm.
On prépare comme suit le propène substitué, à savoir le 1-(p-chlorophényl)-1-(2- pyridyl)-3-diméthylaminopropène. Dans un ballon de 500 cms muni d'un agitateur, d'un condenseur à reflux et d'un entonnoir, on place 0,4 mole de tribromure de phosphore et 100 6m3 de benzène sec. On ajoute 15 g de pyridine sèche en l'espace d'une demi-heure et refroidit le ballon dans un bain de sel de glace.
On ajoute goutte à goutte en l'espace de 4 heures, en agitant, une solution de 1 mole du carbinol, 10 em3 de pyridine et 100 em3 de benzène, en maintenant la tempé rature entre 0 et 5 C. On laisse le produit de la réaction reposer pendant 16 heures à la température ambiante, puis on le verse sur de la glace et le rend basique à l'aide d'ammo niaque. On enlève la couche organique et épuise 1a couche aqueuse à l'aide de plusieurs portions de benzène.
On sèche les couches organiques_combinées et chasse le solvant par distillation sous vide, ce qui laisse une huile brin foncé. On dissout le bromure brut dans la quinoléine ou la diméthylaniline et chauffe à 150-160 C pendant plusieurs heures. On verse le mélange brun ainsi obtenu sur de 1 a glace, le reprend à l'éther, le sèche et distille. L'amine non saturée est une huile jaune bouillant à<B>150-1550 Cil</B> mm.
On peut également transformer le carbinol en bromure correspondant à l'aide d'acide bromhydrique anhydre au sein d'un solvant organique.
On réduit l'amine non saturée catalytique- ment, à l'aide d'hydrogène et de nickel de Raney, à l'état de 1-(p-chlorophényl)-1-(2- pyridyl)-3-diméthylaminopropane bouillant à 153-158 C/2-3 mm.
