Process for the production of a new polyethylene glycol derivative. The subject of the present patent is a process for the production of a polyethylene glycol derivative, which is characterized in that a compound of the formula YS-CI-I2-R, in which Y is a radical which is split off during the reaction and R is an esterified carboxyl group,
Reacts with a reactive ester of such a mixture of polyethylene glycol monomethyl ethers that in the subsequent saponification of the esterified carboxyl group, a mixture of ethers of ß-oxy-ethyl-mer captoacetic acid with polyethylene glycol monomethyl ethers is obtained, which is a light yellow oil monomethyl ether is soluble in cold and boiling water as well as in alcohol, acetone and benzene.
The end product is to be used as an intermediate product in the manufacture of medicinal products.
The cleavable residue Y is. preferably a metal such as an alkali metal atom, an esterified carboxyl group, especially a carbalkylene group. Suitable reactive esters are primarily those of hydrogen halide acids or organic sulfonic acids, such as 1-vlethanesulfonic acid or p-toluenesulfonic acid.
Example: 2.3 parts by weight of sodium are dissolved in 400 parts by volume of alcohol. While passing in a weak stream of nitrogen or hydrogen, 12 parts by weight of ethyl thioglycolate are added and, after a short time, 69 parts by weight of the benzenesulfonic acid ester of a technical polyethylene glycol monomethyl ether with an average molecular weight of 690.
After refluxing for nine hours, most of the alcohol is distilled off, the residue is taken up in water and extracted with chloroform. Evaporation of the chloroform solution gives a yellow oil which is dissolved in 200 parts by volume of alcohol and refluxed with 200 parts by volume of 2N sodium hydroxide solution for three hours.
After the main amount of alcohol has been distilled off, acidification with hydrochloric acid and extraction with chloroform, 60 parts by weight of the mixture of ethers of β-oxy-ethyl-mercapto-acetic acid with polyethylene glycol monomethyl ethers of the formula CH3 (0) are obtained 'CH2 - CH2)
n0 'CH2 - CH2' S 'CH' # COOH where n is an average value of about 10 to 11, as a yellow oil. The crude product can easily be obtained in pure form by adsorption from aqueous solution on a basic exchange resin and by eluting with dilute sodium hydroxide solution. The product purified in this way is a light yellow oil and is soluble in cold and boiling water, in alcohol, acetone and benzene.
