Verfahren zur Herstellung eines N-Sulfonylharnstoffes. Im Hauptpatent ist ein Verfahren zur Herstellung von N - Benzolsulfonylharnstoff beschrieben, gemäss welchem Isoharnstoff- methyläther-methylsulfat mit Benzolsulfo- chlorid umgesetzt wird.
Gegenstand des vorliegenden Patentes ist ein Verfahren zur Herstellung des noch nicht bekannten N-(p-Aminobenzolsulfonyl)-N'N'- Dimethylharnstoff, welches dadurch gekenn zeichnet ist, dass man N'N'-Dimethyl-isoharn- stoff-O-methyläther-methylsufat, erhältlich aus unsymmetrischem Dimethylharnstoff und Dimethylsulfat,
mit Acetylsulfanilsäure- chlorid kondensiert und aus dem erhaltenen Acetylsulfanilyldimethyl - isoharnstoff - me- thyläther die Äthermethylgruppe durch Be handeln mit Halogenwasserstoff und die Acetylgruppe durch Behandeln mit Alkali lauge abspaltet.
<I>Beispiel:</I> 300 g Kaliumcarbonat, 300 cm3 gesättigte Kaliumcarbonatlösung, 150 g Acetylsulfanil- säurechlorid und 150g N'N'-Dimethyl-iso- ha.rnstoff-0-methyläther-methylsulfat (aus unsymmetrischem Dimethylharnstoff und Di- methylsulfat) werden in einer geschlossenen Mischvorrichtung 12 Stunden lang verrührt, dann mit Wasser versetzt, abgesaugt und mit Wasser gewaschen.
Man erhält 90 g rohen Acetylsulfanilyl-dimethyl - isoharnstoff- me- thyläther als weisses Pulver vom F.<B>136</B> bis 144 .
Abspaltung der Äthermethylgruppe: 90 g des obigen Produktes werden mit 90 cm3 konzentrierter Salzsäure übergossen. Die Chlormethylentwicklung setzt sogleich leb haft ein und wird bei 45 in etwa 20 Minuten zu Ende geführt. Dabei wird die Masse, die sich anfangs gelöst hatte, wieder fest. Man verdünnt mit Wasser, kühlt ab und wäscht das Produkt mit Wasser.
Man erhält etwa 70 bis 75 g rohen Acetylsulfanilyldimethyl- harnstoff, dessen Kaliumsalz feine Nädelchen bildet. Abspaltung der Acetylgruppe: Durch zweistündiges Erhitzen mit 2 1M1 Natron lauge in wässriger Lösung auf 80 bis 90 wird die Acetylgruppe abgespalten, und man erhält N-(p-Aminobenzolsulfonyl)-N'N'-di- methylharnstoff, der zur Verwendung als Heilmittel bestimmt ist. Er schmilzt bei 158 bis 161 .
Hierbei tritt Zersetzung ein, die Verbindung verfestigt sich bei 171 bis 173 und schmilzt beim weiteren Erhitzen bei 258 bis 255 . Das Natriumsalz bildet feine Na deln und löst sich sehr leicht in Wasser mit neutraler Reaktion (pH = 7). An Stelle von konzentrierter Salzsäure kann man zum Ab spalten der Äthermethylgruppe auch ver dünnte Salzsäure oder gasförmigen Chlor wasserstoff anwenden, ebenso auch Brom wasserstoff- oder Jodwasserstoffsäure.
