abgebende Verbindung, wie z. B. das Xylidin selbst oder sein Hydrochlorid, mit einer das Radikal einer a-Halogen-propionsäure ab gebenden Verbindung behandelt. und das er haltene a-Halogen-propionsäure-xylidid mit Diäthylamin zum a-Diäthylamino-propionyl- xylidid-2,6@ umsetzt.
Diese neue Verbindung bildet farblose Kristalle vom Schmelzpunkt 54 bis 550; Siede punkt 1730 bei 3 mm. Das Hydroehlorid der selben bildet farblose Kristalle vom Schmelz punkt 227 bis 2280.
Die neue Verbindung besitzt eine rasche und gute Wirkung als lokales Anästhetikum, während ihre Toxizität verhältnismässig gering ist. Gut geeignet zur Anwendung als Lokal anästhetikum sind auch die Salze dieser Sub stanz mit organischen oder anorganischen Säuren, z. B. Weinsäure, Zitronensäure, Salz säure. Besonders geeignet ist das Hydrochlo- rid. Dieses neue Lokalanästhetikum kann ohne Zusatz von Vasokonstringens angewendet wer den. Die wässerigen Lösungen seiner Salze sind besonders gut haltbar, auch in Verbin dung mit Adrenalin.
Die erste Stufe des Verfahrens, die Her stellung des Halogenpropionsäure-xylidids, kann zum Beispiel durch Einwirkung eines a - Halogen - propionsäurehalogenids, wie von a-Brom-propionsäurebromid, auf vic. m-Xyli- din in der Kälte oder aber durch Behandlung desselben mit einer ,a-Halogen-propionsäure erfolgen, wobei zur Beschleunigung der Reak tion mveckinässig ein Katalysator oder Kon densationsmittel verwendet wird, beispiels weise Phosphorpentoxyd oder Phosphortri chlorid.
<I>Beispiel:</I> 1 Mol vicinales m-Xylidin wird im fünf fachen Volumen eines wasserlöslichen organi schen Lösungsmittels, z. B. Aceton, Dioxan usw., gelöst.. Die Lösung wird auf 100 C ab gekühlt, worauf 1,2 Mol a-Brom-propionylbro- mid zugesetzt werden.
Während einiger Mi nuten wird die Mischung kräftig umgerührt und auf einmal 1000 cm@ halbgesättigte Na triumacetatlösung zugesetzt öder eine andere pufferende oder alkalisierende Substanz. Das Reaktionsgut wird während einer halben Stunde durchgeschüttelt. Der sich bildende Niederschlag wird abfiltriert, mit Wasser aus gewaschen und getrocknet. Das erhaltene Pro dukt ist genügend rein zur Weiterverarbei tung.
1 Mol des erhaltenen a-Broin-propionsäure- xylidids und 2i#, bis 3 Mol Diäthylamin wer- den in 1000 cml trockenem Benzol aufgelöst und am Rückflusskühler während 4 bis 5 Stunden erhitzt.
Das abgeschiedene Diäthyl- aminhydrobroinid wird abfiltriert. Die Ben- zollösung wird zweimal mit 3n-Salzsäure durchgeschüttelt, zuerst mit 800 cm' und dann mit 400 cin3. Die Säureextrakte werden vereinigt und mit 30%iger Natronlauge ver setzt, bis die Ausfällung nicht weiter ver mehrt wird. Die ausgefällte Substanz wird in Äther aufgenommen. Die Ätherlösung wird mit Pottasche getrocknet, worauf das Lö sungsmittel abgetrieben wird.
Das erhaltene a-Diäthylamino-propionyl-xylidid-2,6 wird durch Destillation im Vakuum gereinigt.

The separated diethylamine hydrobroinide is filtered off. The benzene solution is shaken twice with 3N hydrochloric acid, first with 800 cm 'and then with 400 cm3. The acid extracts are combined and mixed with 30% sodium hydroxide solution until the precipitation is no longer increased. The precipitated substance is taken up in ether. The ether solution is dried with potash, whereupon the solvent is driven off.
The a-diethylamino-propionyl-xylidide-2,6 obtained is purified by distillation in vacuo.
