Description

   <Desc/Clms Page number 1>

Procedure for the preparation of monocamphorates of the Solanaceae alkaloids.

As is to be expected from a dicarboxylic acid, camphor acid forms two series of salts: neutral and acid salts. Surprisingly, it has now been found that alkaloids, even if they are used in excess, only come together with camphoric acid in a molecular ratio of 1: 1.

It does not matter whether acid and base are brought together in organic solvents such as ether, alcohol, etc. or in aqueous solution in the form of salts.

The possibility of producing uniform, nicely crystallized salts in this way means a significant relief z. B. when working with solutions of unknown alkaloid content. The present process makes use of this phenomenon for the preparation of monocamphorates of the solanaceae alkaloids. The compounds obtainable according to the same, in particular those of hyoscyamine and scopolamine, are distinguished by particularly excellent therapeutic effects on the vagus.

Example 1: 2 g of camphoric acid and 3 g of hyoscyamine are each dissolved in absolute ether and the solutions are combined. The battle-acid salt immediately precipitates in a microcrystalline form. It has a
 EMI1.1
 show around 1450.

Example 3: Sodium battle-acid is added to a neutral atropine chlorohydrate solution, it is evaporated, taken up with alcohol and the atropine monocamphorate is easily obtained.

Example 4: The hyoscyamine amphorate can be obtained from a neutral hyoscyamine sulphate solution by precipitating with camphoric barium, filtering off the barium sulphate and evaporating.

Hyoseyamine and seopolamine monoeamphorate are easily soluble in water and alcohol, both have a bitter taste and slightly acidic reaction to litmus.

Atropine amphorate is very similar to hyoseyamine amphorate, it does not affect the direction of the polarized light.
