abgebende Verbindung, wie z. B. das Xylidin selbst oder sein Hydrochlorid, mit einer das Radikal einer Halogenessigsäure abgebenden Verbindung behandelt und das erhaltene Halogenessigsäurexylidid mit Cyelohexyl- amin zum (N-Cyclohexyl-aminoacetyl)-xyli- did-2,6 umsetzt.
Diese neue Verbindung bildet farblose Kri stalle vom Schmelzpunkt 60 bis 620. Das Hy- drochlorid derselben bildet farblose Kristalle vom Schmelzpunkt 232 bis 2340.
Die neue Verbindung besitzt eine rasche und gute Wirkung als lokales Anästhetikum, während ihre Toxizität. verhältnismässig ge ring ist. Gut geeignet zur Anwendung als Lokalanästhetikum sind auch die Salze dieser Substanz mit organischen oder anorganischen Säuren, z. B. Weinsäure, Zitronensäure, Salz säure. Besonders geeignet ist das Hydrochlo- rid. Dieses neue Lokalanästhetikum kann ohne Zusatz von Vasokonstringens angewendet wer den. Die wässerigen Lösungen seiner Salze sind besonders gut. haltbar, auch in Verbin dung mit Adrenalin.
Die erste Stufe des Verfahrens, die -Her stellung des Halogenessigsäure-xylidids, kann zum Beispiel durch Einwirkung von Halogen essigsäurehalogenid, wie Chloressigsäurechlo- rid, auf vic. m-Xylidin in der Kälte oder auch durch Behandlung desselben mit Halogen essigsäure erfolgen, wobei zur Beschleunigung der Reaktion zweckmässig ein Katalysator oder Kondensationsmittel verwendet wird, bei spielsweise Phosphorpentoxyd oder Phosphor- trichlorid.
Beispiel: 1 Mol vicinales m-Xylidin wird in 800 ems Eisessig aufgelöst. Die Mischung wird auf 100 C abgekühlt, worauf auf einmal 1,1 Mol Chloracetylchlorid zugesetzt wird. Während einiger Minuten wird die Mischung kräftig umgerührt und auf einmal 1000 em3 halb gesättigte Natriumacetatlösung zugesetzt oder eine andere puffierende oder alkalisierende Substanz. Das Reaktionsgut wird während einer halben Stunde durchgeschüttelt.
Der sich bildende Niederschlag wird abfiltriert, mit Wasser ausgewaschen und getrocknet. Die Ausbeute an Chloressigsäure-xylidid beträgt <B>70</B> bis<B>80%</B> der theoretischen Menge. Das er haltene Produkt ist genügend rein zur Weiter verarbeitung.
In 1000 cm' trockenem Benzol oder einem andern wasserfreien organischen Lösungsmit tel werden 1 Mol Chloressigsäure-xylidid-2,6 und 21/2 bis 3 Mol Cyclohexylamin gelöst. Die Mischung wird am Rückflusskühler 4 bis 5 Stunden gekocht.
Das abgeschiedene Cyclo- hexylamin-hydrochlorid wird abfiltriert. Die Benzollösung wird zweimal mit 3n-Salzsäure durchgeschüttelt, zuerst mit 800 ems und dann mit 400 cms. Die Säureextrakte werden ver- einigt und mit 30 % iger Natronlauge versetzt, bis die Ausfällung nicht weiter vermehrt wird.
Die ausgefällte Substanz wird in. Äther auf genommen. Die Ätherlösung wird mit Pott asche getrocknet, worauf das Lösungsmittel abgetrieben wird. Das erhaltene (N-Cyclo- hexyl-aminoacetyl)-xylidid-2,6 kann durch Umkristallisierung aus Petroleumäther gerei nigt werden.

In 1000 cm 'of dry benzene or another anhydrous organic solvent, 1 mole of 2,6-chloroacetic acid-xylidide and 2 1/2 to 3 moles of cyclohexylamine are dissolved. The mixture is boiled on the reflux condenser for 4 to 5 hours.
The separated cyclohexylamine hydrochloride is filtered off. The benzene solution is shaken twice with 3N hydrochloric acid, first with 800 ems and then with 400 cms. The acid extracts are combined and 30% sodium hydroxide solution is added until the precipitation does not increase any further.
The precipitated substance is taken up in ether. The ether solution is dried with pot ash, whereupon the solvent is driven off. The (N-cyclohexyl-aminoacetyl) -xylidid-2,6 obtained can be cleaned by recrystallization from petroleum ether.
