Verfahren zur Herstellung eines Benzolsalfonsäureabkömmlings. Es wurde gefunden, .dass man zu chemo- therapeutisch wertvollen Verbindungen ge langt, wenn man Benzolsulfonsäureanilide herstellt, die min@dee ns in der 3'- und 5' iStellun;
g des Anilidrestes ,durch Halogen, Alkyl oder Halogenalkyl substituiert sind und ausserdem in 4-.Stellung des Benzolrestes eine stiokstoffhaltige Gruppe, z. B. die Amino-, Acylamino-, Alkylamino-, Nitro-, Azo- oder Azomethin.gruppe tragen.
Als Halogenalkylgruppe hat sich die T'rifluor- methylgruppe als besonders geeignet erwie sen. Das Stickstoffatom in 4-Stellung des Benzolrestes kann auch Glied eines hetero- cyelischen Ringes sein.
Dieser Ring kann als Substituent vorhanden sein, oder er kann zu sammen mit dem Benzolring auch ein kon- deneiertes aromatisch-heterocyolisehes Ring system, z. B. ein Chinolinringsystem, bilden.
Verbindungen der gekennzeichneten Art sind im Malaria-Propylaxe-Versuch und im Ma laria: Heilversuch (nach Roehl) beim Kana- rienvogel wirksam, während ähnlich auf gebaute Verbindungen, denen aber die in bestimmter Weise 3',5'-disubstituierte Ani- lid@gruppe fehlt, wie z.
B. das 4-Amino- benzolsulfonamid, das 4-Aminobenzolsulfon- anilid, 2',4'-Diaminoazobenzol-4-,sulfonamid, 4' - :Sulf onamidophenylazo - 7 -, acetylamino-1- oxynaphthaäin, 3,6-,disulfos@aures Natrium und ähnliche in den .gleichen Versuchsanordnun- gen unwirksam sind.
Als besonders ;günstig hat sich die Substitution durch Amino und Acylamino in 4-Stellung und durch Halogen und Halogenalkyl in 3',5'-,Stellung erwiesen.
,Gegenstand des vorliegenden Patentes ist ein Verfahren zur Herstellung eines Benzol- sulfonsäureabkömmlings, welches dadurch gekennzeichnet ist, Uass man ein Benzol- sulfon6äure-3',5'-,dibromanilid, welches in 4-Stellung einen zur Aminogruppe verseif- baren Rest enthält, verseift.
Geeignete, zur Aminogruppe verseifbare Reste sind z. B. die Acylaminogruppen und die Azomethingrup- pen, die nach an sich bekannten Methoden der Verseifung unterworfen werden können. Das so erhältliche 4-Aminobenzolsulfonsäure- 3',5'-dibromanilid bildet farblose Kristalle vom Schmelzpunkt 152 . Es soll therapeu tische Anwendung finden.
<I>Beispiel 1:</I> 45 g 4-Acetylaminobenzolsulfonsäure-43',5'- dibromanilid werden mit 300 cm' 15 % iger Natronlauge 1 ,Stunde unter Rückflussküh- lung zum Sieden erhitzt. Sodann wird die Umsetzungslösung mit 300 cm' Wasser ver dünnt, mit etwas Tierkohle kurz aufgekocht und filtriert.
Durch Ansäuern des Filtrates mit Essigsäure wird das 4-Aminoberrzolsul- fonsäure-3',5'-dibromanilid ausgefällt. Nach dem Umlösesi aus Methanol erhält man die Substanz in farblosen Kristallen vom Schmelz punkt 152 .
Das als Ausgangsmaterial verwendete 4-Acetylaminobenzolsulfonsäure-3',5'-dibrom- anilid erhält man auf folgende Weise:
Zu einer Lösung von 50 g 3,5-Dibromanilin in 200 cm' Pyridin fügt man unter Rührei) 50 g 4-Acetylaminobenzolsulfonsäurechloid und er hitzt das Gemisch 30 Minuten im siedenden Wasserbad. Das Umsetzungsgemisch wird an schliessend in Wasser eingerührt, wobei das 4- Aoetylaminobenzolsulfon6äure-3',5'-dibrom- anilid als weisser, kristalliner Niederschlag ausfällt. Dieses Rohprodukt kann .direkt zur Verneifung verwendet werden.
Durch Um lösen aus Aceton erhält man die Verbindung in Form farbloser Kristalle vom Schmelz punkt 244 .
<I>Beispiel 2:</I> 4-(4"-Nitrobenzylidenamino)-benzolsulfon- säure-3',5'-dibromanilid wird mit verdünnter Salzsäure übergossen und durch Kochen rasch verseift. Nach dem Abkühlen wird der festgewordene Rückstand abgesaugt. Er wird mit kalter verdünnter Natronlauge aus gezogen.
Der zurückbleibende 4-Nitrobenz- aldehyd wird abfiltriert. Das Filtrat wird mit Ammoniumchlorid versetzt und auf diese Weise das 4-Aminobenzolsulfonsäure-3',5'- dibromani,lid abgeschieden. Es schmilzt nach dem Umkristallisieren aus Methanol bei 152'.
Den Ausgangsstoff erhält man wie folgt: 32,4 .g 4-(4'-Nitrobenzylidenamino)-ben- zoleulfochlorid (dargestellt durch Behand lung von 4-Nitrobenzylidenaailin mit Chlor- sulfonsäure, aus Methylenchlorid schwach gelbe Kristalle vom .Schmelzpunkt 111 ") werden zu einer Lösung von 25,1 g 3,
5-Di- bromanilin in 100 cm' Pyridin eingetragen und im Wasserbad 30 Minuten erwärmt. Beim Abkühlen kristallisiert das 4-(4"-Nitro- benzylidenamino)-benzolsulfonsäure- 3',5'-di - bromanili@d in gelben Blättchen aus.
Es wird abgesaugt, mit Pyridin, später mit Alkohol und schliesslich mit Aceton ausgewaschen und zeigt dann einen Schmelzpunkt von 239 . Es ist in verdünnter kalter Natronlauge mit gel ber Farbe löslich.
Process for the preparation of a benzene sulfonic acid derivative. It has been found that chemotherapeutically valuable compounds are obtained if benzenesulfonic acid anilides are produced which have the min @ dee ns in the 3 'and 5' positions;
g of the anilide radical, are substituted by halogen, alkyl or haloalkyl and, in addition, in the 4-position of the benzene radical, a nitrogen-containing group, e.g. B. the amino, acylamino, alkylamino, nitro, azo or Azomethin.gruppe carry.
The trifluoromethyl group has proven to be particularly suitable as a haloalkyl group. The nitrogen atom in the 4-position of the benzene radical can also be a member of a heterocyclic ring.
This ring can be present as a substituent, or it can, together with the benzene ring, also form a condensed aromatic-heterocyclic ring system, e.g. B. form a quinoline ring system.
Compounds of the type indicated are effective in the malaria propylaxis attempt and in the malaria: curative attempt (according to Roehl) in the canary bird, while similarly built compounds, but which are in a certain way 3 ', 5'-disubstituted anilid @ group is missing, such as
B. 4-Amino-benzenesulfonamide, 4-aminobenzenesulfonanilide, 2 ', 4'-diaminoazobenzene-4-, sulfonamide, 4' -: sulfonamidophenylazo - 7 -, acetylamino-1-oxynaphthaäin, 3,6-, disulfos @ aures sodium and the like are ineffective in the same experimental set-ups.
Substitution by amino and acylamino in the 4-position and by halogen and haloalkyl in the 3 ', 5' - position has proven particularly favorable.
The subject of the present patent is a process for the production of a benzene sulfonic acid derivative, which is characterized in that a benzene sulfonic acid 3 ', 5' -, dibromanilide which contains a residue which can be saponified to the amino group in the 4-position, saponified.
Suitable radicals which can be hydrolyzed to the amino group are, for. B. the acylamino groups and the azomethine groups, which can be subjected to saponification by methods known per se. The 4-aminobenzenesulfonic acid 3 ', 5'-dibromanilide thus obtainable forms colorless crystals with a melting point of 152. It should find therapeutic application.
<I> Example 1: </I> 45 g of 4-acetylaminobenzenesulfonic acid-43 ', 5'-dibromanilide are heated to the boil with 300 cm' of 15% sodium hydroxide solution for 1 hour under reflux cooling. The reaction solution is then diluted with 300 cm 'of water, briefly boiled with a little animal charcoal and filtered.
The 4-aminoberzolsulfonsäure-3 ', 5'-dibromanilide is precipitated by acidifying the filtrate with acetic acid. After dissolving from methanol, the substance is obtained in colorless crystals with a melting point of 152.
The 4-acetylaminobenzenesulfonic acid 3 ', 5'-dibromo anilide used as starting material is obtained in the following way:
50 g of 4-acetylaminobenzenesulfonic acid chloride are added to a solution of 50 g of 3,5-dibromaniline in 200 cm of pyridine with scrambled eggs, and the mixture is heated in a boiling water bath for 30 minutes. The reaction mixture is then stirred into water, the 4-aoetylaminobenzenesulfonic acid-3 ', 5'-dibromo anilide precipitating out as a white, crystalline precipitate. This crude product can be used directly for rejuvenation.
By dissolving from acetone, the compound is obtained in the form of colorless crystals with a melting point of 244.
<I> Example 2: </I> 4- (4 "-Nitrobenzylidenamino) -benzenesulfonic acid 3 ', 5'-dibromanilide is poured over with dilute hydrochloric acid and quickly saponified by boiling. After cooling, the solidified residue is suctioned off. It is drawn out with cold, dilute caustic soda.
The remaining 4-nitrobenzaldehyde is filtered off. The filtrate is mixed with ammonium chloride and in this way the 4-aminobenzenesulfonic acid 3 ', 5'-dibromani, lid is deposited. It melts at 152 'after recrystallization from methanol.
The starting material is obtained as follows: 32.4 g of 4- (4'-nitrobenzylideneamino) -benzenesulfochloride (prepared by treating 4-nitrobenzylideneaailine with chlorosulfonic acid, from methylene chloride pale yellow crystals with a melting point of 111 ") to a solution of 25.1 g of 3,
5-dibromaniline is introduced into 100 cm 'pyridine and heated in a water bath for 30 minutes. On cooling, the 4- (4 "-nitrobenzylideneamino) -benzenesulfonic acid 3 ', 5'-di - bromanili @ d crystallizes out in yellow leaves.
It is filtered off with suction, washed out with pyridine, later with alcohol and finally with acetone and then shows a melting point of 239. It is soluble in dilute, cold caustic soda with a yellow color.
