Verfahren zur Darstellung basischer Azoverbindungen der Chinolinreihe
Es wurde gefunden, daß basische Azoverbindungen der Chinolinreihe, die wenigstens
in der z- oder 4-Stellung des Chinolinringes eine primäre oder sekundäre' Aminogruppe
enthalten, sich durch hervorragende Wirkung bei der Gewebsdesinfektion sowie gegen
Blutparasiten auszeichnen. Dabei hat sich gezeigt, daß namentlich die letztere Wirkung
gelegentlich noch dadurch verstärkt wird, daß man die tertiären Stickstoffatome
quaternär macht. Zur Darstellung der neuen Verbindungen kann man entweder in beliebigen
Chinolinverbindungen, welche mehrere Aminogruppen, davon mindestens eine in 2- oder
4-Stellung des Pyridinringes, enthalten und die eine primäre Aminogruppe im Benzolkern
aufweisen, diese letztere Aminogruppe diazotieren und mit beliebigen Komponenten
kuppeln. Man kann auch beliebige Chinolinverbindungen, welche mehrere Aminogruppen,
davon mindestens eine in 2- oder 4-Stellung des Pyridinringes, enthalten, als Kupplungskomponenten
mit beliebigen Diazoverbindungen umsetzen. Man kann auch nach den üblichen Methoden.nachträglich
Azoverbindungen der Chinolinreihe primäre oder sekundäre Aminogruppen, davon mindestens
eine in 2- oder 4-Stellung des Pyridinringes, einführen.
14 g p-Nitranilin werden in Gegenwart von z2 ccm konzentrierter Salzsäure in der
üblichen Weise diazotiert. Zu der klaren Lösung läßt man unter Rühren und Kühlen
mit Eis eine Lösung von 26 g 4, 6-Diaminoehinaldindihydrochlorid (Beispiel
i des Patents 591 480) 'in ioo ccm Wasser im Verlauf 1/2 Stunde zufließen.
Die Farbstoffbildung tritt sofort ein und ist nach Zugabe von 309 kristallisiertem
Natriumacetat in i bis 2 Stunden beendet. Danach wird mit viel verdünnter Salzsäure
versetzt, einige Zeit zum Sieden erhitzt, nach dem Erkalten das rotbraune Hydrochlorid
der oben förmulierten Base abgesaugt und dieses mit verdünnter Salzsäure gewaschen.
Durch Verrühren mit heißer,- verdünnter Natronlauge und Auswaschen mit heißem Wasser
erhält man die schwarzviolette Farbstoffbase, die nach dem Umkristallisieren aus
Benzol und Äthanol bei 26o° unter Aufschäumen schmilzt. Die Salze
der
Base sind in Wasser leicht löslich -mit roter Farbe, die Lösungen Werden durch Salzsäure

Wird die heiße methanolische Lösung der Nitrofarbbase allmählich mit
einer wässe= rigen Lösung der berechneten Menge Natriumsulfid versetzt und danach
noch l/2 Stunde im Sieden gehalten, so erhält man die Triaminoazoverbindung, die
nach Zugabe von viel Wasser abgesaugt und mit Wasser gewaschen wird. Die Base ist
hellrot gefärbt und .schmilzt nach dem Umkristallisieren aus Chlorbenzol bei 25r°.
Das salzsaure Salz ist in Wasser leicht löslich mit roter Farbe, die einen Stich
ins Gelbe zeigt. Durch verdünnte Salzsäure wird die Lösung nicht ausgesalzen.
26 ,g q:, 6-Diamiaiochinaldindihydrochlorid werden in ioo ccm Wasser gelöst. Die
infolge hydrolytischer Spaltung schwach sauer reagierende Lösung wird dann unter
Kühlung mit 25 ccm z n-Nitritlösung diazotiert und darauf so lange im Wasserbad
erwärmt, bis die Farbstoftbildung beendet ist. Danach wird in der Wärme mit Salzsäure
kongosauer gemacht, gesättigte Nitriumchloridlösung zugesetzt, nach dem Erkalten
das ausgeschiedene Hydrochlorid obiger Farbbase abgesaugt und mit verdünnter Salzsäure
gewaschen. Man erhält es in einer Menge von 2o g. Die Farb-. stoffbildung verläuft
auch recht glatt, wenn man die Diazotierung und Selbstkuppelung in einer Lösung
der Diaminochinaldinbase in etwa 5o°/oiger Essigsäure unter den angegebenen Bedingungen
vornimmt. Das Salz bildet ein rotes Pulver, das sich in Wasser mit gelbroter Farbe
leicht löst. Ammoniak fällt aus der Lösung die rote Farbbase, die in Aceton und
in Alkohol leicht löslich ist. Wird die Farbbase mit 2 Möl jodmethyl in tlthanol i Stunde auf etwa
i io ° erwärmt, so erhält man däs Dijodmethylat in der berechneten Menge: Es ist
in Wasser ziemlich schwer löslich. Durch Behandeln mit Chlorsilber kann es in das
Chlormethylat verwandelt werden, das sich in Wasser mit roter Farbe leicht löst.
In der Lösung entsteht auf Zusatz von wenig Natronlauge keine Fällung. Es schmilzt
bei 3o2° unter Aufschäumen.
14 g 4, 6-Diaminochinaldindihydrochlorid werden in einer Mischung von
50 ccm Wasser und 12 ccm konzentrierter Salzsäure mit einer Lösung von 3,5
g Natriumnitrat in io ccm Wasser diazotiert, wobei Lösung eintritt. Diese Lösung
wird allmählich unter Rühren zu einer methanolischen Lösung von 8,8 g 4, 8-Diaminochinaldin
(Beispiel 5 des Patents 59148o), die 35 g krist. Natriumacetat enthält, hinzugegeben.
Der sich bildende hellrote Farbstoff obiger Konstitution wird in heißer, sehr verdünnter
Salzsäure gelöst und aus dieser Lösung nach dem Filtrieren durch Zusatz von konzentrierter
Salzsäure das salzsaure Salz ausgesalzen, das abgesaugt und mit verdünnter Salzsäure
gewaschen wird. Die Ausbeute an diesem dunkelbraunroten Salz beträgt ig g. Es ist
in Wasser leicht löslich mit roter Farbe. Die Base schmilzt bei 26q.° unter Zersetzung.
Arbeitet man nach den Angaben des Beispiels 3 und ersetzt das 4, 8-Diaminochinaldin
durch g g 6-Methoxy-8-aminochinolin, so er- log hält man den Farbstoff der oben
angegebenen Zusammensetzung als ein dunkelrotes Pulver, das, aus wässerigem Alkohol
umkristallisiert, bei 226 bis 227° schmilzt. Das salzsaure Salz ist in Wasser leicht
löslich mit roter lio Farbe.
Durch Kupplung der nach Beispiel 3 erhaltenen Diazolösung-mit io g Phenol in Gegenwart
von Natronlauge erhält man die rote Farbstoffbase nebenstehender Konstitution in
theoretischer
Ausbeute. Sie ist in heißer Natriumcarbonatlösung et-,vas, in verdünnter Natronlauge
leicht löslich mit gelbroter Farbe. Aus viel Alkohol umkristallisiert, bildet sie
ein hellrotes Pulver, das oberhalb 300'
schmilzt. Das- gelb gefärbte Hydrochlorid
ist in Wasser leicht löslich mit rotgelber Farbe. Durch Behandeln der alkalischen
Lösung der Base mit Dimethylsulfat erhält man den Methyläther, der, aus wässerigem
Alkohol umkristallisiert, ein rotes Pulver bildet, das bei i29° unter Aufschäumen
schmilzt. Läßt man auf das Natriumsalz der Phenolbase Diäthylaminoäthylchlorid einwirken,
so erhält man den basischen Äther, dessen Hydrochlorid sich sehr leicht in Wasser
mit rotgelber Farbe löst und bei 14o bis I41 ° schmilzt.
Eine nach Beispiel 3.dargestellte Diazolösung aus 6,9 g 4, 6-Diaminochinaldinbase
wird mit einer salzsauren Lösung von 4,85 g Dimethylanilin vermischt. Auf- Zugabe
einer konzentrierten wässerigere. Lösung von Natriumacetat tritt sofort Kupplung
ein unter Bildung des obigen Farbstoffs. Nach kurzer Zeit wird mit Natronlauge versetzt,
abgesaugt und mit heißem Wasser gewaschen. Die Base bildet ein rotes Pulver, das,
aus Alkohol umkristallisiert, bei 270 bis 27z ° unter Aufschäumen schmilzt. Das
salzsaure Salz bildet ein blauviolettes Pulver, das in Wasser leicht löslich ist
mit roter Farbe. Durch Einwirkung von 2 Mol Dimethylsulfat auf die methanolische
Lösung der Base in derWärme erhält man das diquaternäre Salz, das auf Zusatz von
alkoholischer Salzsäure beim Äufkochen in Lösung geht. Diese Lösung scheidet auf
Zusatz von Äther das Dichlormethylat der Base aus, das sich mit gelbroter Farbe
leicht löst. Die mit Natronlauge gefällte Base schmilzt, aus Alkohol umkristallisiert,
bei :263' unter Dunkelfärbung.
Eine dianotierte Lösung von 14 g 4, 6-Diaminochinaldindihydrochlorid wird zu einer
Lösung von 5,5 g:2, 6-Diaminopyridin in verdünnter Salzsäure bei Gegenwart von Natriumacetat
hinzugegeben. Nach Beendigung der Kupplung wird unter Erwärmen im Wasserbad mit
Ammoniak übersättigt, der Niederschlag abgesaugt, in heißer, verdünnter Essigsäure
aufgenommen, die Lösung filtriert- und das Filtrat mit Kochsalzlösung versetzt.
- Dabei scheidet sich das Hydrochlorid der neuen Farbstoffbase in einer Menge von
z g, z g als dunkelbraunes Pulver aus, das in Wasser mit roter Farbe löslich ist.
Die Base stellt ein rotes Pulver dar, das, aus Chlorbenzol umkristallisiert, bei
28o bis 28z unter Aufschäumen schmilzt.
Zu einer-Lösung von q.,35 g r-Phenp1L3-methyl-5-pyrazolon in 62,5 ccm 2 n-Natronlauge
wird eine dianotierte Lösung von 7 g 4, 6-Diaminochinaldindihydrochlorid in 37,5
ccm a n-Salzsäure hinzugefügt. Nach beendeter Kupplung wird mit viel Wasser verdünnt,
essigsauer gemacht, aufgekocht, .filtriert, das Filtrat mit Salzsäure versetzt und
nach dem Erkalten das salzsaure Salz des gebildeten Farbstoffs abgesaugt. Es ist
ein rotgelbes Pulver, das beim Aufkochen mit Wasser leicht in Lösung geht. Die aus
der heißen Lösung mit Ammoniak gefällte rote Base wird nach dem Trocknen in Nitrobenzol
mit 2 Mol Dimethylsulfat 2o Minuten lang im Ölbad unter Rühren auf r50° erhitzt.
Nach dem Erkalten wird mit Äther verdünnt, der Niederschlag abgesaugt, in heißem
Wasser gelöst und aus der Lösung mit Natronlauge die nunmehr darin unlösliche orange
gefärbte Base gefällt. Nach dem Umkristallisieren aus Alkohol und Wasser schmilzt
sie bei 285'
unter Zersetzung. Mit Salzsäure bildet sie ein hellbraunes Salz,
dem obige Konstitution zuzuerteilen ist und das in Wasser leicht löslich ist.
18,6 g Anilin, gelöst in 4oo ccm 2 n-Salzsäure, werden mit 149 Natriumnitrit dianotiert.
Die
Diazolösung wird darauf mit 32,9 2, 4-Dioxychinolin in 400 ccm 2 n-Natronlauge unter
Rühren zusammengebracht. Nach beendeter Kupplung wird angesäuert, der gebildete
Farbstoff abgesaugt und in heißer, verdünnter Natronlauge gelöst. Die filtrierte
Lösung scheidet beim Ansäuern mit Essigsäure die gelbe Farbbase aus, die abgesaugt,
mit Wasser gewaschen und gut getrocknet wird. 25 g dieses Dioxyazofarbstoffs werden
alsdann mit roo ccm Phosphoroxychlorid 2 Stunden zum schwachen Sieden erhitzt. Nach
dem Erkalten wird das Reaktionsprodukt unter Rühren in Zoo ccm mit Eis versetztes
konzentriertes wässeriges Ammoniak eingegossen, das ausgeschiedene rohe Chlorierungsprodukt
in Äther aufgenommen und der Äther nach dem Trocknen mit Chlorcalcium abdestilliert.
Es hinterbleibt das reine Benzolazodichlorchinölin als eine feste Masse, die in
den gewöhnlichen organischen Lösungsmitteln leicht löslich ist. 12 g davon werden
mit 6o ccm alkoholischem Ammoniak 6 Stunden auf 2oo° erhitzt. Der Bombeninhalt wird
zur Trockne verdampft, der Trockenrückstand mit verdünnter gssigsäure ausgekocht,
der Auszug heiß filtrierte ünd das Filtrat ammoniakalisch gemacht. - Dabei fällt
die gelbe Base obiger Konstitution in. einer Menge von 7,5 g aus. Die Base schmilzt
nach denn Umkristallisieren aus- wässerigem Methanol bei 245°. Wird sie in Methanol
unter Rühren und Erwärmen mit alkoholischer Salzsäure angesäuert, so bildet sich
nach vorübergehender Lösung ein gelber Brei des salzsauren Salzes, das nach dem
Verdünnen mit Aceton abgesaugt und mit Aceton gewaschen wird. Es löst sich beim
Erwärmen mit Wasser mit gelber Farbe. Durch wenig Salzsäure oder Natriumdhlorid
