Verfahren zur Herstellung eines Benzolsulfonsäureabkömmlings. Das Hauptpatent betrifft ein Verfahren zur Herstellung eines chemotherapeutisch wertvollen Benzolsulfonsäureabkömmlings.
Gegenstand des vorliegenden Patentes ist ein Verfahren zur Herstellung eines Benzol- sulfonsäureabkömmlings, welches dadurch gekennzeichnet ist, dass man ein Benzolsul- fonsäure-3',5'-dijodanilid, welches in 4-Stel- lung einen zur Aminogruppe verseifbaren Rest enthält, verseift.
Geeignete zur Amino- gruppe verseifbare Reste sind zum Beispiel die Acylaminogruppen und die Azomethin- gruppen, die nach an sich bekannten Me thoden der Verseifung unterworfen werden können. Das so erhältliche 4-Aminobenzol- sulfonsäure 3',5'-dijodanilid bildet farblose Kristalle vom Schmelzpunkt 199 . Es soll therapeutische Anwendung finden.
<I>Beispiel 1:</I> 54 g 4 - Acetylamino - benzolsulfonsäure- 3',5'-dijodanilid werden mit 300 cm' 15 % iger Natronlauge 1 Stunde unter Rückflussküh- lung zum Sieden erhitzt. Sodann wird die Umsetzungslösung mit 300 cm' Wasser ver dünnt, mit etwas Tierkohle kurz aufgekocht und filtriert. Durch Ansäuern des Filtrates mit Essigsäure wird das 4-Aminobenzolsul- fonsäure - 3',5'- dijodanilid ausgefällt.
Nach dem Umlösen aus Methanol erhält man die Substanz in farblosen Kristallen vom Schmelzpunkt 199 .
Das Ausgangsmaterial erhält man auf folgende Weise: Zu einer Lösung von 69 g 3,5-Dijodanilin in 200 cm' Pyridin fügt man unter Rühren 50 g 4-Acetylamino-benzol- sulfonsäurechlorid hinzu und erhitzt das Ge misch 30 Minuten im siedenden Wasserbad. Das Umsetzungsgemisch wird anschliessend in Wasser eingerührt, wobei das 4-Acetyl- amino-benzolsulfonsäure-3',5'-dijodanilid als weisser, kristalliner Niederschlag ausfällt. Dieses Rohprodukt kann direkt zur Versei fung verwendet werden.
Durch Umlösen aus Aceton erhält man die Verbindung in farb- losen Kristallen vom Schmelzpunkt 277 .
Beispiel <I>2:</I> 4- (4"-Nitro-benzylidenamino) -benzolsul- fonsäure-3',5'-dijodanilid wird mit verdünnter Salzsäure übergossen und durch. Kochen rasch verseift. Nach dem Abkühlen wird der festgewordene Rückstand abgesaugt. Er wird mit kalter verdünnter Natronlauge ausge zogen. Der zurückbleibende 4-Nitrobenzalde- hyd wird abfiltriert. Das Filtrat wird mit Ammoniumchlorid versetzt und auf diese Weise das 4-Aminobenzolsulfonsäure-3',5'- dijodanilid abgeschieden.
Es schmilzt nach dem Umkristallisieren aus Methanol bei 199 .
Den Ausgangsstoff erhält man durch Um setzung von 4- (4'-Nitro - benzylidenamino)- benzolsulfonsä,urechlorid (dargestellt durch Behandeln von 4 - Nitro - benzylidenanilin mit Chlorsulfonsäure, aus Methylenchlorid schwachgelbe Kristalle vom Schmelzpunkt 111 ) mit 3,5-Dijodanilin in Pyridin. Das 4-(4"-Nitro-benzylidenamino) -benzolsulfon- säure-3',5'-dijodanilid wird als gelbe, allmäh lich erstarrende Masse erhalten,
die ohne wei- tere Reinigung der Verseifung unterworfen werden kann.
