  Verfahren zur Darstellung des     5,7-Dimethyltocolphosphorsäureesters.       Die     Tocopherole    sind in Wasser unlösliche  Öle. Es wurde nun gefunden, dass man diese  Verbindungen in wasserlösliche Form über  führen kann, wenn man die     phenolische          Hydroxylgruppe    durch Phosphorsäure     ver-          estert    und die Ester in ihre     Alkalisalze    über  führt. Die Einführung des Phosphorsäure  restes geschieht zweckmässig durch Einwir  kung von     Phosphoroxychlorid    und     nach-          heriger    Umsetzung der Reaktionsprodukte  mit Wasser.

   Die entstehenden     Tocopherol-          phosphorsäureester    sind unlöslich in Wasser.  Sie lösen sich aber in äquivalenten Mengen  Alkali. Die erhaltenen Lösungen sind an der  Luft stabil.  



  Gegenstand des vorliegenden     Patentes     ist ein Verfahren zur Darstellung des     5,7-          Dimethyltocolphosphorsäureesters,    welches  dadurch gekennzeichnet ist, dass man auf       5,7-Dimethyltocol    ein     Phosphorylierungs-          mittel    einwirken lässt.  



  Der     5,7-Dimethyltocolphosphorsäureester     bildet eine wachsartige Masse, die in Wasser    unlöslich ist. Er löst sich leicht in 2 Äqui  valenten Natronlauge. Die Lösung schäumt  stark und scheidet mit konzentrierter Lauge  das Salz in Flocken ab. In kristallisierter  Form erhält man das Natrium salz,     wenn    man  die Lösung des     5,7-Dimethyltocolphosphor-          säureesters    in Methylalkohol mit Natron  lauge versetzt. Der neue     5,7-Dimethyltocol-          phosphorsäureester    bildet das Ausgangspro  dukt für die     Gewinnung    des     Natriumsalzes,     welches als Arzneimittel verwendet werden  soll.  



  <I>Beispiel:</I>  2,65 Teile     5,7-Dimethyltocol    werden in  20 Teilen trockenem     Pyridin    gelöst, 1,9 Teile       Phosphoroxychlorid    zugesetzt     und    das . Ge  misch 4 Tage in einem geschlossenen Kolben  stehen gelassen. Darauf     wird    mit Wasser  verdünnt, mit Äther     extrahiert    und die       Ätherlösung        viermal    mit     normaler    Salzsäure  extrahiert. Nach dem Waschen     mit    Wasser  wird die     mit    Natriumsulfat getrocknete  Ätherlösung im Vakuum eingedampft.

   Das      zurückbleibende Harz wird in 50 Teile 0,2  normaler Natronlauge aufgenommen und  etwa vorhandenes     Tocol    mit Äther extrahiert.  Die alkalische Schicht wird angesäuert, mit  Äther extrahiert und die Ätherschicht noch  mals mit     n-Salzsäure    extrahiert. Beim Ab  dampfen des Äthers bleibt der     5,7-Dimethyl-          tocolphosphorsäureester    zurück.

   The tocopherol phosphoric acid esters formed are insoluble in water. However, they dissolve in equivalent amounts of alkali. The solutions obtained are stable in air.



  The subject of the present patent is a process for the preparation of the 5,7-dimethyltocolphosphoric acid ester, which is characterized in that a phosphorylating agent is allowed to act on 5,7-dimethyltocol.



  The 5,7-dimethyltocolphosphoric acid ester forms a waxy mass that is insoluble in water. It dissolves easily in 2 equivalents of sodium hydroxide solution. The solution foams strongly and separates the salt in flakes with concentrated lye. The sodium salt is obtained in crystallized form when sodium hydroxide solution is added to the solution of the 5,7-dimethyltocolphosphoric acid ester in methyl alcohol. The new 5,7-dimethyltocol-phosphoric acid ester is the starting product for obtaining the sodium salt, which is to be used as a drug.



  <I> Example: </I> 2.65 parts of 5,7-dimethyltocol are dissolved in 20 parts of dry pyridine, 1.9 parts of phosphorus oxychloride are added and that. The mixture was left to stand in a closed flask for 4 days. It is then diluted with water, extracted with ether and the ether solution extracted four times with normal hydrochloric acid. After washing with water, the ether solution, dried with sodium sulfate, is evaporated in vacuo.

The alkaline layer is acidified, extracted with ether and the ether layer extracted again with n-hydrochloric acid. When the ether evaporates, the 5,7-dimethyl tocolphosphoric acid ester remains.

PATENTANSPRUCH: Verfahren zur Darstellung des 5,7-Di- methyltocolphosphorsäureesters, dadurch ge kennzeichnet, dass man auf 5,7-Dimethyl- tocol ein Phosphorylierungsmittel einwirken lässt. Der 5,7-Dimethyltocolphosphorsäureester bildet eine wachsartige Masse, die in Wasser unlöslich ist. Er löst sich leicht in 2 Äqui valenten Natronlauge. Die Lösung schäumt stark und scheidet mit konzentrierter Lauge das Salz in Flocken ab.
