Verfahren zur Herstellung von 4-Phenyl-pentamethylenogyd-4-carbonsäurenitril. Im Schweizer Patent Nr. 206719 ist ein Verfahren zur Darstellung von Piperidinderi- vaten angegeben, nach welchem Arylaceto- nitrile mit Di--(ss-halogenalkyl)-aminen unter Benutzung alkalischer Kondensationsmittel in 4-Aryl-piperidin-4-carbonsäurenitrileüber- geführt werden.
Es wurde nun gefunden, dass man aus Arylacetonitrilen mit .ss,ss'-Dihalogendialkyl- ä.thern unter Benutzung alkalischer Konden sationsmittel entsprechend die bisher noch unbekannten 4-Aryl-pentamethylenoxyd - 4 - carbonsäurenitrile erhält.
Gegenstand des vorliegenden Patentes ist ein Verfahren zur Herstellung von 4 - Phenyl - pentamethylen- oxyd-4-carbonsäurenitril, das dadurch ge kennzeichnet ist, dass, man Benzylcyanid mit l.ss'-Dihalogenäthyläthern unter Benutzung eitles alkalischen Kondensationsmittels um setzt. Als alkalische Kondensationsmittel kommen besonders Natriumamid und Na triumphenyl in Betracht.
Das 4-Phenylpenta- m.ethylenoxyd - 4 - carbonsäurenitril bildet tafelförmige Kristalle, siedet bei 147 bis 148 C / 5 mm und ist ein. wertvolles Zwi schenprodukt für die Herstellung von Heil mitteln.
<I>Beispiel:</I> Zu einem Gemisch von<B>117</B> Gewichts teilen Benzylcyanid, 143 Gewichtsteilen ss,ss'-Dichlordiäthyläther und 500 Gewichts teilen Toluol werden unter Rühren und pas sender Kühlung insgesamt 84 Gewichtsteile pulverisiertes Natriumamid in Portionen von etwas '/io der Gesamtmenge so, eingetragen, dass sich die Temperatur zwischen 40-50' hält.
Darauf wird das Gemisch langsam bis zum Sieden erhitzt und so lange unter Rück fluss gekocht, bis kein Ammoniak mehr ent weicht -(1 bis 2 Stunden). Nach dem Er kalten wird die Toluollzösung mit Wasser ausgeschüttelt, zur Lösung des ausgeschiede nen Natriumchlorids abgetrennt und unter vermindertem Druckdestilliert. Das 4-Phenyl- pentamethylenoxy d-4-.carbonsäurenitril gellt bei 2 mm von 125-140' C über.
Nochmals destilliert siedet es bei 3 mm um<B>133</B> C, bei 5 mm um 147-148' C als farblose Flüs sigkeit über, die beim Stehen bei Zimmer temperatur bald vollkommen in tafelförmi gen Kristallen erstarrt. Ausbeute etwa 50'0 der Theorie. Es lässt sich aus Methylalko hol oder Petroläther umkristallisieren und schmilzt bei 49-50' C.
Das so erliältliehe Nitril kann wie folgt weiter umgesetzt werden Durch zweistündiges Erhitzen des Nitrils mit 66%iger Schwefelsäure auf dem Was serbad und Ausfällen mit Wasser wird das 4-Phenylpenta.methylenoxyd - 4 - carbonsäure- amid erhalten. Farblose Kristalle aus 3le- thylalkohol. F = 216-218 C.
Durch sechsstündiäes Erhitzen mit metlivl . #t auf 190-20(l " l-isst alkoholischer Kalilau-- sch die V erseifun- zur 4-Plienyl-pentametliy- lenoxyd-4-carbonsäure durchführen, die aus stark wasserhaltigem Methylalkohol in farb. losen Blättchen kristallisiert und bei 128 bis <B>130'</B> C schmilzt. Das Chlorid bildet sich aus der Säure mit Tbionylchlorid, es siedet bei 3 mm um 140 C und zeigt einen F. von 53-54 C.
Der ss-Diäthylaminoäthylester wird in Form des Hydrochlorids aus Alko hol kristallisiert, als farbloses Kristallpulver von F.<B>181</B> " gewonnen: die freie Base ist 1111b.
4-PIienj#l-pentriinctliylenoxyd - 4 - carbon- säure wird durch konzentrierte Halobenwas- serstoffsäure bei erhöhter Temperatur unter Aufspaltung des Oxydringes in halogenierts Säuren, die nur als ()1e erhalten wurden, übergeführt. Diese lassen sich unter geeig neten Bedingungen mit Ammoniak oder pri mären Aminen zu 4-Phenyl-piperidin-4-c.ar- bonsäure umsetzen.
