Verfahren zur Darstellung einer Aminobenzolsulfonsäureamidverbindung. Gemäss der im Patent Nr.199315 be schriebenen Erfindung sind gewisse Amino- acyl- und Oxyacyl-aminobenzolsulfonsäure- amide sowie deren Substitutionsproduktedurch ihre Wirkung gegen bakterielle Infektions krankheiten ausgezeichnet.
Gegenstand des vorliegenden Patentes ist ein Verfahren zur Herstellung einer Amino- benzolsulfonsäureamidverbindung, welches da durch gekennzeichnet ist, dass man auf ein 4-(4'-Halogenbenzolsulfonamido)-benzolsulfon- amid Ammoniak bei erhöhter Temperatur im Druckgefäss einwirken lässt. Die Reaktion kann in Gegenwart eines Lösemittels, wie Wasser, Methyl- oder Äthylalkohol erfolgen. Vorzugsweise findet das 4-(4'-Brombenzol- sulfonamido)-benzolsulfonamid bei der Um setzung Verwendung. Aus der Reaktions mischung kann das Reaktionsprodukt in üb licher Weise, z.
B. durch Eindampfen unter vermindertem Druck, Auflösen des Rück standes in verdünnter Lauge und Ausfällen des Reaktionsproduktes mit verdünnter Essig säure abgetrennt werden. Das so erhältliche 4-(4'-Aminobenzolsulfonamido) -benzolsulfon- amid bildet nach dein Umkristallisieren, z. B. aus verdünntem Alkohol, farblose Kristalle vom Schmelzpunkt 1310. Es ist in kaltem Wasser schwer löslich, löslich in verdünnten Alkalien. Es soll therapeutische Anwendung finden.
Beispiel: 20 g 4-(4'-Brombenzolsulfonamido)-benzol- sulfonamid (dargestellt durch Kondensation von 4-Brombenzolsulfochlorid mit 4-Amino- benzolsulfonamid in Pyridin, aus verdünntem Alkohol farblose Kristalle vom Schmelzpunkt 209 ) werden im Rührautoklaven mit 120 cm' konzentriertem wässrigen Ammoniak 10 Stun den auf 2001 bei etwa 60-70 Atmosphären Druck erhitzt. Die Reaktionsmischung wird unter vermindertem Druck auf dem Wasser bade zur Trockne gebracht, mit verdünnter Natronlauge gelöst, filtriert und mit Essig säure angesäuert.
Das ausfallende 4-(4'-Amirio- benzolsulforiamido)-benzolsulfonamid bildet nach dem Umkristallisieren aus verdünntem Alkohol farblose Kristalle vom Schmelzpunkt 1310. An Stelle von wässrigem Ammoniak kann man auch methylalkoholisches oder äthylalkoholisches Ammoniak zur Umsetzung verwenden. An Stelle der Bromverbindung können auch andere Halogenverbindungen, z. B. die Jod- oder die Chlorverbindung, An wendung finden. Bei der Chlorverbindung wird die Reaktionstemperatur zweckmässig etwas erhöht.
