Abstract

La présente invention concerne une composition antipyrétique à usage vétérinaire apte au traitement d'animaux d'élevage intensif destinés à la consommation hu-maine, en particulier de porcs, de volailles ou de veaux.
Pour des raisons de sécurité publique faciles à
comprendre, les principes actifs vétérinaires 'pouvant étre administrés aux animaux destinés à la consommation humaine, doivent satisfaire à des normes très strictes, et étre ins-crits sur une liste positive susceptible d'ëtre modifiée.
Une seule molécule antipyrétique est actuellement proposée en médecine vétërinaire, à savoir l'aspirine. Cette molécule présente, cependant, un certain nombre d'inconvénients bien connus des spécialistes parmi lesquels on peut noter son caractère ulcérogène qui exclut tout trai-l5 terrent de longue durée du bétail.
De plus, la molécule d'aspirine se distingue par une faible stabilité et une tendance marquée à l'hydrolyse, ce aussi bien en solution aqueuse que lorsqu' elle est incor-porée sous forme pulvérulente à des aliments granulés : elle ne peut donc pas être stockée sous une forme galénique prête à l'emploi et doit étre absorbée très rapidement après sa préparation.
On a, par ailleurs, récemment découvert que l'aspirine peut entra3ner chez l'enfant une dégénérescence hépatique très grave dite <c syndrome de Reye » qui tend à
freiner son utilisation en médecine pédiatrique.
Ces différents inconvénients font qu'il est sou-haitable que les vétérinaires puissent disposer d'un principe actif antipyrétique pouvant ëtre substitué à l'aspirine.
Conformément à l'invention, vn a eu l'idée de proposer, à cet effet, la molécule de paracétamol vu 4-hydroxy-acétanilide de formule .

En effet, dans ce domaine particulier, on ne peut essentiellement envisager que deux formes galéniques.

I1 est en outre connu que 1e paracétamol s'hydrolyse et s'oxyde en milieu aqueux. Par suite, il est exclu de conserver des solutions aqueuses de paracétamol.

La présente invention a, par suite, pour objet de proposer une composition antipyrétique à usage vétérinaire apte au traitement d'animaux d'élevage intensif destinés à la consommation humaine, en particulier de porcs, de volaille ou t0 de veaux, se présentant sous la forme galénique d'une solu-tion concentrée stable et soluble dans l'eau froide en toutes proportions requises pour ces traitements, susceptible d'étre introduite dans les circuits de circulation de l'eau de bois-son des animaux dont disposent de nombreux élevages ou d'étre pulvérisée sur des aliments granulés.
On a de manière surprenante pu satisfaire à tous ces impératifs en dissolvant le paracétamol dans un solvant figurant dans la liste positive énumérant les substances dont l'utilisation est autorisée en médecine vétérinaire à savoir un polyéthylène glycol à bas poids moléculaire et/ou du poly-propylène glycol.
L'invention concerne, par suite, une composition antipyrêtique caractérisée en ce qu'elle est constituée par une solution buvable renfermant de 15 à 25 % en poids, de préférence environ 20 % en poids de paracétamol dissous dans un solvant constitué par un polyéthylène glycol à bas poids moléculaire et/ou du polypropylène glycol.
La solution conforme à l'invention renferme, de préférence en proportions pondérales, ~0 à 85 % de polyéthy lène glycol et 0 à 45 % de polypropylène glycol.
Selon l'invention, cette composition peut, le cas échéant, renfermer jusqu'à 5 % d'excipients, par exemple des agents d'appétence ou encore des antioxydants vu des conser-vateurs.
Le polyéthylène glycol à bas poids moléculaire pouvant être mis en oeuvre conformément à l'invention doit étre suffisamment peu visqueux et va en règle générale du S
PEG 150 au PEG 300 ;' on a pu obtenir des résultats particu-liér.ement satisfaisants en mettant en ouvre du PEG 200.
La composition antipyrétique conforme à
l'Z.nvention se distingue par sa stabilité en solution aqueuse à toute concentration mais également par sa stabilité dans le temps et par l'absence de tout phénomène de dégradation ou de précipitation méme après un stockage prolongé.
Pour vérifier cette dernière propriété, on a étu dié les variations de la concentration en paracétamol pendant un stockage de 42 mois â température ambiante, d'une part, et à une température de 37 °C, d'autre part, de compositions conformes à l'invention renfermant à l'origine 20 % en poids de paracétamol dissous dans du polyëthylène glycol 200.
Ce test a été effectué pour trois lots de compo-citions A, B et C.
Les résultats obtenus sont rassemblés dans les tableaux ci-dessous.

La présente invention concerne également un pro-cédé d'incorporation da la composition antipyrétique susmen-tionnée à l'eau de boisson d'animaux dans un élevage équipé
d'un circu~.t de circulation d'eau pouvant ètre activé automa-tiquement gràce à des organes de commande associés en parti-cu ber à des sondes de niveau ou à des détecteurs de présence d'un animal.
Selon l'invention, la première étape d'un tel procédé consiste à dëterminer une concentration de consigne de l'eau dc boisson en paracétamol en tenant compte de la po-sologie prescrite, des tables de croissance ainsi que de la consommation d'eau moyenne des animaux à traiter.
La concentration de consigne ayant ainsi été dé-terminée, il convient ensuite d'ajouter dans le circuit de circulation la quantité de solution concentrée de paracétamol 2o dissous dans un polyol du type susmentionné permettant d'obtenir cette concentration au niveau des auges ou des pi-pettes où viennent se désaltérer les animaux à traiter.
Selon une première variante de l'invention, le circuit de circulation d'eau équipant l'élevage est relié à
un réservoir de grandes dimensions contenant la quantité
d' eau nécessaire à un groupe d' animaux pendant une durée dé-terminée, en particulier pendant une journée.
Conformément à cette première variante de l'invention, après avoir déterminé la concentration de cvnsi gne de l'eau de boisson en paracétamol .

Dans le cadre de celle-ci, il est bien entendu impératif que la formulation à base de paracétamol soit par-faitement solubilisée, ce à tous les stades de dilution, pour exclure tout risque de colmatage du circuit de circulation d'eau. I1 est à noter qu'un tel colmatage pourrait en parti-culier se produire au niveau du tube d'alimentation de la pompe doseuse qui plonge au fond du bac contenant la solution mère, au niveau de la pompe doseuse et également au niveau des organes de commande permettant l'activation automatique du circuit de circulation d'eau et des pipettes lorsque cette solution est destinée à des volailles.
