Abstract
MÉTODOS PARA PRODUZIR METIONINA, PARA APERFEIÇOAR O USO DE DMDS PARA PRODUÇÃO DE METIONINA, E, PARA IDENTIFICAR UM ALELO MUTANTE, COMPOSIÇÃO, PRODUTO CONTENDO METIONINA, MICROORGANISMO, GENE ISOLADO, OACETIL-HOMOSSERINA SULFIDRILASE MUTANTE OU ENZIMAS DE O-SUCCINIL-HOMOSSERINA SULFIDRILASE, E ORGANISMO. A presente invenção proporcionar organismos e processos aperfeiçoados para a produção de metionina. A invenção demonstra que um organismo AmetE AmetH, por exemplo, mutantes de C. glutamicum ou E. coli, pode usar uma fonte de sulfeto terminada com metila, p. ex., dissulfeto de dimetila (DMDS), como uma fonte tanto de um grupo metila como de enxofre, contornando a necessidade de atividade MetH/MetE/ e MetF e a necessidade de reduzir sulfato, para a necessidade de reduzir sulfato, para a síntese de metionina. Também se descreve nesta patente dados que implicam MetY (também denominado MetZ) como um enzima que incorpora uma fonte de sulfeto terminada com metila, p. ex., DMDS, em metionina. Uma cepa AmetF AmetB de C. glutamicum pode usar uma fonte de sulfeto terminada com metila, p. ex., DMDS, como uma fonte, tanto de um grupo metila como de sulfeto. Adicionalmente, a produção de metionina por meio de organismos prototróficos manipulados que superproduzem O-acetil-homosserina foi aperfeiçoada com a adição de uma fonte de sulfeto terminada com metila, p. ex., DMDS.
