"ENSAIO DE ALTA EFICáCIA à BASE DE CéLULAS MAMìFERAS PARA A DETERMINAçãO DO PERFIL E A SELEçãO DE MODULADORES PUTATIVOS DE UM CANAL DE SóDIO EPITELIAL, MéTODOS DE MONITORAR A ATIVIDADE DE UM CANAL DE SóDIO EPITELIAL, E DE IDENTIFICAR UM COMPOSTO MODULADOR DE SABOR SALGADO, UM COMPOSTO QUE MODULA HENAC, E UM MODULADOR DE ENAC HUMANO, E, OóCITO". Em um aspecto, a presente invenção refere-se a um ensaio de alta eficácia baseado em células mamíferas para a formação de perfil e seleção de canal de sódio epitelial humano (hENaC, human epithelial sodium channel clonado de uma biblioteca de c-DNA de rim humano e também é expresso em outros tecidos, incluindo tecido do paladar humano. A presente invenção refere-se adicionalmente a ensaios eletrofisiológicos de média eficácia baseados em oócitos anfíbios para identificar moduladores de ENAC humano, de preferência, acentuadores de ENaC. Espera-se que compostos que modulam função de ENaC em um ensaio de ENaC à base de células venham a afetar o sabor salgado em humanos. Os ensaios aqui descritos apresentam vantagens importantes sobre sistemas de expressão celular existentes. No caso de células mamíferas, referidos ensaios podem ser realizados em placas de cultura convencionais de 96 ou 384 poços em modo de alta eficácia, sendo que se obtém resultados de ensaio acentuados com o uso de um composto que inibe função de ENaC, de preferência, um derivado de amilorida, como fenamila. No caso dos ensaios eletrofisiológicos de oócitos inventivos (técnica de pinça de voltagem com dois eletrodos), estes ensaios facilitam a identificação de compostos que modulam especificamente ENaC humano. Os ensaios da invenção proporcionam uma seleção robusta útil para detectar compostos que facilitam (acentuam) ou inibem função de hENaC. Compostos que acentuam ou bloqueiam atividade de canal ENaC humano deveriam, portanto, modular o sabor salgado em humanos.
