Abstract

I ett aspekt angår foreliggende oppfinnelse en mammaisk celle-basert høykapasitets test for profilering og screening av human epitelial natriumkanal (ENaC) klonet fra et humant nyre cDNA-bibliotek og uttrykkes også i andre vev inkludert humant smaksvev. Foreliggende oppfinnelse angår videre amfibie-oocyttbaserte medium-throughput elektrofysiologiske tester for å identifisere human ENaC-modulatorer, foretrukket ENaC-forsterkere. Forbindelser som modulerer ENaC-funksjon i en cellebasert ENaC-test forventes å påvirke saltsmak hos mennesker. Testene beskrevet heri har fordeler foran eksisterende cellulære ekspresjonssystemer. l tilfelle av mammaiske celler kan slike tester gjennomføres i standard 96- eller 384-brønners dyrkingsplater i høykapasitets metode hvor forsterkede testresultater oppnås ved anvendelse av en forbindelse som hemmer ENaC-funksjon, foretrukket et amilorid-derivat slik som fenamil. l tilfelle av de oocytt elektrofysiologiske testene ifølge foreliggende oppfinnelse (toelektroders spennings clamp-teknikk), letter disse testene identifikasjon av forbindelser som spesifikt modulerer human ENaC.
