Abstract

<B>ENSAIO PARA CONFORMAçãO RELACIONADA à DOENçA DE UMA PROTEìNA<D> é revelado um método de ensaio que isola e detecta a presença de uma conformação relacionada 'doença de uma proteína (por exemplo, PrP^ Sc^) presente em uma amostra também contando a conformação da proteína não relacionada à doença (por exemplo, PrP^ c^). A amostra é tratada (por exemplo, colocada em contato com protease) de uma maneira que hidrolisa a conformação relacionada à doença e não hidrolisa a conformação não relacionada a doença. A amostra tratada é colocada em contato com um parceiro de ligação (por exemplo, um anticorpo marcado que liga-se à PrP^ Sc^) e a ocorrência de ligação fornece uma indicação de que PrP^ Sc^ está presente. De forma alternativa a PrP^ Sc^ da amostra tratada é desnaturada ( por exemplo, por contato com guanidina) ou desdobrada. A PrP^ Sc^ desdobrada é colocada em contato com um parceiro de ligação e a ocorrência de ligação indica a presença de PrP^ Sc^ na amostra. Em uma outra modalidade, PrP^ Sc^ e PrP^ c^ sofrem reação com um anticorpo marcado que liga-se a ambas as conformações e uma conformação que liga-se apenas à conformação relacionada à doença, e a presença da conformação relacionada à doença é determinada por comparação das duas.
