2002年4月19日出願の、R. Anthony Williamson、Dennis R. Burton および Gianluca Moronciniの、米国仮出願第60/371,610号「細胞のプリオンポリペプチドの複製インターフェイスを含有し、その他のタンパク質凝集疾患からのモチーフ・グラフトされたハイブリッドポリペプチおよびその使用(MOTIF-GRANTED HYBRID POLYPEPTIDES CONTAINING THE REPLICATIVE INTERFACE OF CELLULAR PRION POLYPEPTIDE AND FROM OTHER DISEASES OF PROTEIN AGGREGATION AND USES THEREOF」からの優先権を主張する。
本出願において、標的ポリペプチドに特異的に反応する試薬を提供し、かかる標的ポリペプチドは、タンパク質凝集の疾患(コンフォメーションが変化したタンパク質が関与する疾患)、例えば、アミロイド疾患に関連する感染性形態のポリペプチドである。そのような特異性を有するハイブリッド分子、例えばハイブリッドポリペプチドを提供する。該ハイブリッドポリペプチドは、結果として得られるハイブリッド分子があるコンフォメーションのタンパク質に特異的に結合し、その他のコンフォメーションのタンパク質には結合しないように、標的ポリペプチドと特異的に相互作用し、骨格(例えば、抗体または酵素またはその他のレシピエントの一部など)に挿入されたポリペプチドモチーフを含む。典型的には、標的とされるコンフォメーションは、疾患に関与するコンフォメーションである。ポリペプチドモチーフは、骨格のあらゆる所望の機能が保持され、挿入されるモチーフがその標的に対して特異的に結合する能力を保持するように、骨格に挿入される。選択される骨格はモチーフと標的ポリペプチドの複合体の検出を助け、あるいは可能にするようにその活性または結合部位を利用されうる。コンフォメーション特異性を有するポリペプチドの調製方法も提供する。

タンパク質コンフォメーションの疾患については、プリオンタンパク質またはアミロイドタンパク質などの、同一のタンパク質 (またはその部分)が2以上のアイソフォーム(配座異性体)を示し、少なくとも1つの形態が疾患の原因であり、あるいは疾患に関与している。診断、予後、治療および/または薬剤スクリーニングの目的で、疾患関連配座異性体に対して良性の(非-疾患関連)配座異性体と比較してより特異的に相互作用する分子 (即ち典型的には少なくとも、2-、5- 10-倍、一般的に少なくとも 約 100-倍の、より大きいアフィニティーにて反応する分子)を得るのが有用である(あるいは逆もまた同様)。したがって、本明細書において、複数の配座異性体を有するタンパク質の1つの配座異性体に特異的に反応する分子を提供する。典型的にはかかる分子は疾患関連配座異性体と相互作用する。
上述のように、本発明における方法およびハイブリッド分子は、タンパク質凝集またはコンフォメーションの疾患に関与または関連するタンパク質を用いるものである。かかる疾患においては、少なくとも1つの形態のタンパク質は良性であり、別の形態が例えば疾患感染性試薬として、および/または凝集反応において、疾患に関与する。かかる疾患および2以上の異なるコンフォメーションにてアセンブリーする関連タンパク質(ここで少なくとも1つのコンフォメーションがコンフォメーションが変化したタンパク質である)は上記の表1に示すものを含む。
遍在性の細胞のプリオンタンパク質(PrPc)の異常な配座異性体であるPrPScは、感染性プリオン粒子の唯一の同定されている成分である。プリオン増殖の際に、新生のプリオン感染性の形成がPrPc のコンフォメーション再編成を引き起こすことによってPrPScが自己複製するテンプレート依存的プロセスを介して進行すると考えられている。プリオン複製複合体において正確にどのようにして異なるPrPcおよびPrPSc 配座異性体が互いに相互作用し、またおそらくその他の補助分子と相互作用するのか(Kaneko et al. (1998) Proc. Natl. Acad. Sci. U.S.A. 94:10069-10074; Zulianello et al. (2000) J. Virol. 74:4351-4360)は知られていない。異なるプリオン株がその特徴的な性質を複数の継代にわたって保持しているという観察は、プリオン増殖が高度に忠実なプロセスであることを示し、また、PrPcと PrPScとの分子相互作用が非常に特異的であることを示唆する (Prusiner et al. (1998) Proc. Natl. Acad. Sci. U.S.A. 95:13363-13383; Caughey (2001) T.I.B.S. 26:235-242)。
プリオン疾患、例えば、スクレイピーおよびウシ海綿状脳症は、細胞のプリオンタンパク質(PrPc)の異常な配座異性体であるPrPScと密接に関係している。PrPcの第1のαヘリックスと結合するモノクローナル抗体、例えば、モノクローナル抗体D13またはD18は、PrPcとPrPScとのヘテロダイマー結合を妨げることによってプリオン増殖を阻害する(Williamson et al. (1998) J. Virol. 72:9413-9418を参照されたい;同時係属米国特許出願第09/627,218号も参照されたい;これらFabのそれぞれの重鎖と軽鎖の核酸とコードされるタンパク質配列を示す配列番号29-36を参照されたい)。PrPcとPrPScとを識別する抗体またはその他の特異的結合分子はこの問題を解決するために有用である。正常またはPrP-ヌル動物の広範なPrP抗原(感染性プリオン、PrPc、およびαヘリックスまたはβ−シートに富むコンフォメーションに再フォールディングされた組換えおよび合成PrP分子を含む)での免疫化はしかし、くりかえし失敗しており、疾患関連形態のPrPをもっぱら認識する高-アフィニティー抗体を得ることはできていない(Williamson et al. Proc. Natl. Acad. Sci. U.S.A. 93:7279; Peretz et al. (1997) J. Mol. Biol. 273:614; Williamson et al. (1998) J. Virol. 72:9413)。かかる抗体についてのより最近の報告(Korth et al. (1997) Nature 390:74)も中途段階である(Fischer (2000) Nature 408:479)。プリオン増殖はテンプレート依存的プロセスであり、ここでPrPScがPrPcのコンフォメーション再編成を引き起こす(Prusiner et al. (1998) Proc. Natl. Acad. Sci. 本発明において、PrPScに特異的に結合するポリペプチドおよびかかるポリペプチドならにびその他のコンフォメーションが変化したタンパク質疾患(疾患はタンパク質凝集をともなう)の感染性または疾患誘発性配座異性体に結合するハイブリッドポリペプチドの調製方法を提供する。したがって、優先的に(特異的に)1つの配座異性体(一般的に疾患関連配座異性体)に別の配座異性体と比べてより強いアフィニティー、典型的には少なくとも 0.5、1、2、3、5、10-倍強いアフィニティーで結合するポリペプチドを提供する。1以上のアミノ酸欠失を含み、その結果得られる分子の構造の変化が生じるが1つの配座異性体、例えばPrPSc、に対する結合能力は有意に変化しておらず、プリオンタンパク質複合体を形成するか、1つの配座異性体におけるコンフォメーション変化を誘発する、例えば、PrPScにおけるコンフォメーション変化を誘発するペプチドも考えられる。
多くの動物からのプリオンが同定され配列決定されている;例示的なプリオンを配列番号5-13に示す。あらゆる公知のプリオンタンパク質が本発明において考慮される;かかるプリオンの配列は公共のデータベースおよび刊行物から入手できる。例えば、ニワトリ、ウシ、ヒツジ、ラットおよびマウス PrP 遺伝子が Gabriel et al. (1992) Proc. Natl. Acad. Sci. U.S.A. 89:9097-9101に開示され公表されている; シリアンハムスターについての配列はBasler et al. (1986) Cell 46:41-428lに公表されている; ヒツジPrP 遺伝子はGoldmann et al. (1990) Proc. Natl. Acad. Sci. 動物プリオンに加えて、その突然変異形態もポリペプチドモチーフのソースとして考えられる。多数の突然変異体形態が公知であり、ヒトにおいて特徴付けされている。それらには、コドン 102におけるプロリン (P)からロイシン (L)への突然変異(遺伝子的にLODスコアが3を超えるGSSの進展と関係していることが示された)が含まれる。この突然変異はPrPをコードする生殖系列DNAにおけるホスホジエステル結合を介してデオキシグアノシン (G)に結合したメチル化デオキシシトシン (C) (CpG)の脱アミノにより、その結果、デオキシシトシンとデオキシチミジン (T)の置換がおこったためであると考えられる。コドン 178におけるアスパラギン酸 (D)からアスパラギン (N)の置換を伴う突然変異がCJDの多くの家系で同定されている。D178N 突然変異はその浸透率が不完全あるが、不眠症の多くのイタリアの家系と関連している。同じ突然変異が、表現型的にはFFIとは異なり、CJDに類似しているが、ただし多くの場合、持続時間が長く鋭波脳波活動が欠如している疾患を患ういくつかの家系で報告された。同じ突然変異が2種類の表現型を与えるというこの知見は、この表現型的不均質の分子基礎を見いだす一連の研究を促した。15名のFFIおよび15名のCJD患者におけるPRNP 遺伝子型の詳細な分析により、D178N 突然変異に加えて、すべてのFFI患者が突然変異体アレルの位置129にメチオニンを有し、すべてのCJD患者がこの位置においてバリンを有することが示された。これらの結果はすべてのFFIおよび CJD症例について確認された。それゆえこれによって2種類のハプロタイプ、FFI における129M、D178N ハプロタイプ、そしてCJD における129V、D178N ハプロタイプが見いだされた。1つのFFI 家系が突然変異体アレルにおいてオクタペプチドリピート欠失を有していることから、すべての公知のFFI 家系が共通の起源を有するようではない。この知見は、表典型の差異はPRNP コドン 129以外の遺伝子的影響によって引き起こされるという可能性に強く問題を投げかけた。突然変異体アレルにおけるコドン 129 のメチオニンまたはバリンはそれぞれFFIおよびCJD178患者に必須であるが、正常アレルにおけるコドン129 がメチオニンやバリンであり得る。それゆえ、FFIおよびCJD 表現型はD178N 突然変異をともなう突然変異体アレルのコドン129により決定され、その結果、2種類のタイプのPrPが発現する。また、FFIは通常表現型においてCJDより早く表れるため、コドン 129は表現型の持続も調節している。
タンパク質凝集の疾患、特にアミロイド疾患からの標的ポリペプチドの疾患関連アイソフォームに特異的に相互作用するハイブリッドポリペプチドの産生方法を本発明において提供する。標的ポリペプチドは疾患関連または疾患誘発性アイソフォームのポリペプチドであって、良性形態から、悪性の疾患誘発性または凝集性アイソフォームに変換したものである。
疾患に特異的なハイブリッド分子を調製するために、疾患関連コンフォメーションと相互作用するポリペプチドのコンフォメーションの部分を同定する。例えばこれは系統的に、ポリペプチドのフラグメントをコンフォメーション変化に関与する能力について試験することによって行う。例えば、異常(即ち疾患誘発性)配座異性体と相互作用するフラグメントの能力を試験する。所望の能力を有するポリペプチドのフラグメントは特異的試薬として用いることが出来、疾患配座異性体に特異的に相互作用する能力を保持するように骨格に挿入することが出来る。骨格は例えば、Fabまたは酵素またはその他の分子である。
選択されたポリペプチドモチーフが挿入され(または結合され)、その結果得られるハイブリッドポリペプチドが所望の結合特異性を有するようなあらゆる分子、例えば、ポリペプチドは、本発明におけるハイブリッド分子の一部として使用するために考慮されうる。ポリペプチドは、標的ポリペプチドの結合のためのモチーフが正確に存在するのに少なくとも十分な数(10、20、30、50、100またはそれ以上のアミノ酸)を含むあらゆるアミノ酸配列に挿入されうる。骨格の目的は、それに結合する形態において標的ポリペプチドに対してモチーフを提示することである。骨格はさらなる特性(例えば検出可能なマーカーまたは標識として役立つ能力など)を有するように、またはさらなる結合特異性を備えさせることによりそのアッセイ(標的タンパク質の特定のアイソフォームを検出するアッセイまたは治療薬スクリーニングアッセイまたはその他のアッセイおよび方法)における使用を可能または容易にするように、設計または選択してもよい。
抗体は本発明において考えられる骨格またはレシピエントポリペプチドの一例である。抗体およびそのフラグメントは、目的のポリペプチドモチーフを含むハイブリッドポリペプチドを作るための骨格として機能しうる。ポリペプチドモチーフは、ポリペプチドモチーフのコンフォメーションの保持およびその結果得られるハイブリッドポリペプチドの表面上への提示を可能とするいずれの好適な領域、例えばCDR3ループ(例えば、米国特許第5583202号および、米国特許第5568762号を参照されたい)に挿入してもよい。ポリペプチドモチーフを免疫グロブリン分子の重鎖または軽鎖可変ドメインに挿入すると、標的ポリペプチドに対して特異性を有するハイブリッド免疫グロブリンが生じる。
上述のように、その他の分子、例えば、酵素および発光分子を骨格として用いることが出来る。これらには、触媒および/または結合活性に十分な酵素の全部または一部あるいは発光を提供するのに十分な発光分子の全部または一部が含まれる。骨格として使用される分子にはこれらに限定されないが以下が含まれる:ルシフェラーゼ(発光タンパク質を含む)、アルカリホスファターゼ、β-ガラクトシダーゼおよびその他のシグナル生成酵素、化学発光生成因子、例えば、セイヨウワサビペルオキシダーゼ; 蛍光タンパク質、例えば、周知の赤色、緑色および青色蛍光タンパク質;および色素生産性分子、例えば、色素生産性タンパク質。
上述のように、プリオンタンパク質およびそれ由来のモチーフを含むハイブリッド分子は、本発明において提供するハイブリッド分子の一例である。ハイブリッド分子上に特異的結合を付与するのに十分な少なくとも1つのアミノ酸配列を含むプリオンタンパク質からのあらゆるモチーフが考慮されうる。モチーフには少なくとも5アミノ酸から全分子が含まれ、結合特性を保持するその変異体も含まれる。
アミノ酸89-112および136-158に対応するマウス PrP 配列を、オーバーラップポリメラーゼ連鎖反応(PCR)を用いてHIV-1 gp120に特異的なヒト組換え抗体であるIgG1 b12のHCDR3にグラフトした(Burton et al. (1994) Science 266:1024-1207;配列番号1-4を参照されたい)。抗体 b12を移植されるPrP 配列に対するレシピエント分子として選択した。というのは、この親抗体は抗原結合部位の表面から垂直に延びた比較的長いHCDR3 (18 アミノ酸)を有するからである(Ollmann et al. (2001) Science 293:1155-1159)。抗体表面からPrP 配列をできるだけ遠ざけるために、各グラフトを親 HCDR3の最初のN-末端残基と4つのC-末端残基の間に配置した(図1参照)。さらに2つのグリシン残基をPrP 配列の各末端に組み込んだ。その結果得られたPrP-IgG(89-112 および 136-158) をCHO細胞で発現させ、均質となるまで精製した(Maruyama et al. (1999) J. Virol. 73:6024-6030)。
本発明において提供するあらゆるハイブリッドポリペプチドをコードする核酸分子を提供する。かかる分子は好適な宿主細胞における発現のためにプラスミドおよびベクターに導入するとよい。
核酸分子を含むプラスミドおよびベクターも提供する。コードされるタンパク質を発現する細胞を含む、ベクターを含む細胞を提供する。細胞は、細菌細胞、酵母細胞、真菌細胞、植物細胞、昆虫細胞または動物細胞のいずれでもよい。ハイブリッドポリペプチドの産生方法、例えば、細胞がコードされるポリペプチドを発現する条件下で細胞を培養し、発現したタンパク質を回収する方法を本発明において提供する。細胞は、タンパク質の発現に用いられ、タンパク質は分泌されるか細胞質内に発現される。ハイブリッドポリペプチドは、タンパク質合成の標準方法を用いて化学的に合成することも出来る。
ペプチドアナログは、テンプレートペプチドの性質と類似の性質を有する非ペプチド薬物として医薬業界で一般に使用されている。かかるタイプの非ペプチド化合物は「ペプチド疑似体」(Peptide mimeticsまたはPeptidominetics)と呼ばれている(Luthman et al.、A Textbook of Drug Design and Development、14:386-406、2nd Ed.、Harwood Academic Publishers (1996); Joachim Grante (1994) Angew. Chem. Int. Ed. Engl.、33:1699-1720; Fauchere (1986) J. Adv. Drug Res.、15:29; Veber and Freidinger (1985) TINS、p. 392; およびEvans et al. (1987) J. Med. Chem. 30:1229)。治療上有用なペプチドと構造的に類似したペプチド疑似体は、同等またはより高い治療または予防効果を得るために用いられる。ペプチド疑似体の調製およびその構造は当業者に公知である。ハイブリッドポリペプチドのペプチド疑似体を本発明において提供する。
本発明において提供するハイブリッド分子は様々な用途を有する。それらはサンプル中の1つの配座異性体の存在を検出するためのアッセイに用いることが出来る。サンプルとしては例えば、体液または組織サンプルあるいは食品サンプルまたは土壌サンプルその他のサンプルが挙げられる。これらは疾患の治療薬としても利用できる;それらは候補薬物スクリーニングおよび/または薬物および治療薬または診断薬の設計にも利用できる。
本発明において提供するハイブリッドポリペプチドとタンパク質凝集の疾患(またはコンフォメーションの疾患)に関与する疾患誘発性(または疾患関連)または感染性形態のポリペプチドとの特異的相互作用に基づいて、サンプル、例えば食品または体液または組織サンプル中の疾患誘発性または感染性形態の標的ポリペプチドの存在を検出するためにかかるポリペプチドを利用することが出来る。例えば、PrPScに特異的に相互作用するハイブリッドポリペプチドを用いて血液およびその他の組織をスクリーニングすることが出来る。
ハイブリッドポリペプチドへのPrPScの結合を阻害するかその量を減少させることができる被験化合物は、PrPSc媒介疾患のインビボでの予防または治療のための候補となる。PrPSc媒介疾患には、変異体、孤発性および/または医原性を含むクロイツフェルトヤコブ病 (CJD)、ゲルストマン-ストラウスラー-シェインカー疾患 (GSS)、致死性(fatal)家族性不眠症 (FFI)、クル、スクレイピー、ウシ海綿状脳症(BSE)、およびその他のPrPScの形成を伴う疾患が含まれる。かかる方法によりハイブリッドポリペプチドとPrPScとのインビトロ相互作用を阻害または減少させることが出来るとして同定された被験化合物は、PrPSc 疾患の進行防止または治療の能力についてPrPSc疾患のインビボモデルにおいて試験するとよい。
アッセイが固体支持体、例えば常磁性ビーズで行われる特定の態様において、サンプルからのポリペプチド、あるいは、一般的には、ハイブリッドポリペプチドは、結合、例えばイオン結合または共有、非共有結合またはその他の化学的相互作用によって、支持体またはマトリックス材料の表面に結合される。固定化は直接行ってもリンカーを介してもよい。固定化はあらゆる好適な支持体上に行うことが出来、これらに限定されないが、シリコンチップ、および本明細書に記載するその他の支持体および当業者に周知の支持体が含まれる。複数のポリペプチドを支持体に結合させてもよく、例えばシリコンチップまたはその他のチップの表面のアレイ (即ち2以上のパターン)としてハイスループット・プロトコールおよび形式を含むアッセイに使用すればよい。
アッセイを試験するために標準的プリオン調製物を作り、アッセイの信頼性を高めることができる。標準的プリオン調製物の製造の詳細は公知である(例えば、米国特許第5639581号、米国特許第5908969号および米国特許第5792901号を参照)。調製物は例えば、被験動物のプリオンを含む脳材料を有する宿主動物など、あらゆる動物から得られる。例えば、ヒトプリオンタンパク質遺伝子を含むトランスジェニックマウスによりヒトプリオンを作ることが出来、かかるマウスの脳を用いて標準的ヒトプリオン調製物を作ることが出来る。さらに調製物は「標準化」されたものであり、一般的に実質的に同一の動物から得られる(例えば、100; 1,000、またはそれ以上の動物)。例えば、非常に高コピー数のヒトPrP遺伝子(すべての多型および突然変異)を含む100匹のマウスは自然に疾患を発症し、それぞれからの脳組織を組み合わせて標準的プリオン調製物が作られる。
ハイブリッド分子、例えば、ハイブリッドポリペプチド、およびアッセイを行うためのその他の試薬および材料は組み合わせとして提供でき、所望により標識、アッセイを行うための説明書を含むキットとして提供してもよい。例えばハイブリッドポリペプチドは溶液中に液体分散物として提供してもよいし、実質的に乾燥粉末、例えば凍結乾燥形態で提供してもよい。例えば上述のような支持体プレートなどの固体支持体および1以上のバッファーも別々の要素としてキットに含めてもよい。
G.実施例
免疫沈降
正常または79A スクレイピープリオン感染マウス (脳内接種の130-150日後に屠殺)からの全脳をリン酸緩衝食塩水 (PBS)中で10% (w/v)にてホモジナイズし、等量の200 mM NaCl、50 mM Tris-HCl (pH 8.0)、1% NP40 (またはTriton X-100)および1% デオキシコール酸で希釈し、再びホモジナイズし、超音波処理にかけた。正常またはプリオン感染脳のホモジネートを500 g で15分間遠心し、上清をアリコットとし-20℃で保存した。
SMB 細胞を162 cm2 組織培養フラスコにて集密するまで培養し、PBSで2回洗浄し、フラスコあたり1 mlの細胞溶解バッファー (10 mM Tris-HCl、pH 8.0、100 mM NaCl、10 mM EDTA、0.5% w/v Nonidet P40、0.5% w/v デオキシコール酸ナトリウム)を用いて溶解した。細胞溶解液から1000 g、5分、4℃での遠心によって破片を除いた。免疫沈降実験を、3 mgの総溶解液タンパク質および10μg 抗体を終容量 1 mlにて用いて上述のようにして行った。
2段階 オーバーラップ伸長 PCR (McLane et al. (1995) Proc. Natl. Acad. Sci. U.S.A. 92:5214-5218; 図1参照)を用いてアミノ酸残基 119-136、121-144および121-158 (または136-158および 89-112、実施例4参照)に対応するマウス PrP 配列を、独立にFab b12またはIgG b12 (実施例4参照)のHCDR3 ドメインを置換するようにグラフトした (Burton et al. (1994) Science 266:1024)。

モチーフ-グラフトされた抗体の調製
2段階 オーバーラップ伸長 PCR 19を用いて、アミノ酸残基 89-112、136-158および141-158に対応するマウス PrP 配列を別々に抗体b12にHCDR3ドメインを置換するようにグラフトした。オリゴヌクレオチドプライマーを2倍のポリアクリルアミドゲル電気泳動精製(Operon Technologies)にかけ、それらは以下の配列を含むものであった:

正常または RMLまたは79A スクレイピープリオン感染マウス (脳内接種130-150日後に屠殺)からの全脳をTris緩衝食塩水(TBS; 0.05M Tris、0.2M NaCl、pH 7.4、1% NP-40および1% DOC含有)中で10% (w/v)にてホモジナイズし、等容量のTBSに希釈し、再びホモジナイズし、超音波処理にかけた。正常またはプリオン感染脳ホモジネートを500 g 、15分、4℃で遠心した。遠心したプリオン感染ホモジネートの一部をプロテイナーゼ K (50μg/ml) で1時間 37℃で消化した。PMSF をすべてのサンプルに終濃度2 mMとなるよう添加した。それぞれの免疫沈降について、終濃度 0.3μg/ml〜10μg/mlの抗体をアッセイバッファー (TBS、 3% NP-40 および 3% Tween 20含有)で終容量500μl に調整した反応混合物中のおよそ1 mgの総タンパク質を含む脳ホモジネートのアリコットと共に2時間室温でインキュベートした。ポリクローナルヤギ抗ヒト IgG F(ab')2 (ヒト PrP-グラフトされたハイブリッドポリペプチド検出用)またはポリクローナルヤギ抗マウス IgG F(ab')2 (Fab D13 およびIgG 6H4 検出用)のいずれかに結合したトシル活性化常磁性ビーズ (Dynal) をハイブリッドポリペプチドとホモジネートの混合物に添加し、一晩4℃でインキュベートした。ビーズを洗浄バッファー (TBS 、2% NP-40および2% Tween 20含有)で4回、TBSで1回洗浄し、磁石で分離した。ペレット化したビーズを20μlのローディングバッファー (150 mM Tris-HCl、pH 6.8、6% ドデシル硫酸ナトリウム (SDS)、0.3% ブロモフェノールブルー、30% グリセロール)に再懸濁し、100℃で5分加熱した。サンプルを12% SDS-PAGEゲルで泳動し、ニトロセルロースメンブレンにトランスファーした。メンブレンを、0.1 % Tween 20 を含有するTBS (TBST)中の5% (w/v) 脱脂粉乳で1時間室温でブロッキングし、ブロットされたPrPをFab D13 またはIgG 6H4 抗体1μg/mlにて検出した。TBST 中で5回洗浄後、ブロットされたPrP タンパク質を、ブロッキングバッファーに1:10,000に希釈されたセイヨウワサビペルオキシダーゼ結合ヤギ抗マウスIgG (Pierce)との30分室温でのインキュベーションにより検出した。メンブレンをTBSTで5回洗浄し、増強化学発光試薬 (Amersham)を用いてフィルムに現像した。
Claims (46)
- プリオンポリペプチド由来のポリペプチドモチーフであって、ハイブリッド免疫グロブリンまたはそのFabフラグメント中にあって、プリオンタンパク質のPrPSc形態に結合するのに十分な数のプリオンポリペプチド由来の連続するアミノ酸残基を含むポリペプチドモチーフ;および、
該モチーフが由来するポリペプチド以外のポリペプチド由来のさらなるアミノ酸を含む骨格を含むハイブリッドポリペプチドであって、
該骨格が免疫グロブリンまたはそのフラグメントであって;
該ポリペプチドモチーフがシリアンハムスタープリオンポリペプチドの残基87−158に対応するプリオンタンパク質の一部分に由来する少なくとも18,20,25,30,35,40,45,50,55,60,65,70,75,80残基を含むものであり;
該モチーフがネイティブな非感染性コンフォメーションとして存在するものであり;
該シリアンハムスタープリオンポリペプチドの配列が配列番号:5に示されたものであり;
該ポリペプチドモチーフが免疫グロブリンの重鎖CDR3領域に挿入されており;
結果として得られるハイブリッドポリペプチドが、プリオンポリペプチドのPrPc形態と比較してプリオンポリペプチドのPrPSc形態に対して高い親和性でもって結合するものである、
ハイブリッドポリペプチド。 - 多量体である請求項1のハイブリッドポリペプチド。
- さらなるアミノ酸が、モチーフ部分のN−末端に少なくとも約5アミノ酸およびC−末端に少なくとも約5アミノ酸を含む、請求項1のポリペプチド。
- さらなるアミノ酸が、モチーフ部分のN−末端に少なくとも約15アミノ酸およびC−末端に少なくとも約15アミノ酸を含む、請求項1のポリペプチド。
- 二量体である、請求項1のハイブリッドポリペプチド。
- 三量体である、請求項1のハイブリッドポリペプチド。
- ポリペプチドモチーフが骨格に挿入されている請求項1のハイブリッドポリペプチド。
- 骨格がIgG、IgM、IgA、IgDまたはIgE免疫グロブリンからの定常領域を含む請求項1のハイブリッドポリペプチド。
- 骨格が、Fab、およびF(ab)2または一本鎖Fvである請求項1のハイブリッドポリペプチド。
- プリオンポリペプチドが、ヒト、ハムスター、マウス、ラット、シカ、ヒツジ、ヤギ、オオジカ、クドゥ、ウマ、イヌ、ネコ、ラクダおよびブタの中から選択される動物に由来する、請求項1ないし9のうちいずれかのハイブリッドポリペプチド。
- プリオンポリペプチドがプリオンをコードするアレルの突然変異体形態によってコードされる、請求項10のハイブリッドポリペプチド。
- シリアンハムスタープリオンポリペプチドまたは他の種由来のプリオンポリペプチドに対応する残基の少なくとも残基121−131、121−141、121−136、121−144、121−158、87−112、87−118、87−130、87−169、126−158、131−158、136−158または141−158を含む請求項1ないし9のいずれかのハイブリッドポリペプチド。
- ポリペプチドモチーフが、シリアンハムスタープリオンポリペプチドまたは他の種由来のプリオンポリペプチドに対応する残基の唯一の残基121−131、121−141、121−136、121−144、121−158、87−112、87−118、87−130、87−169、126−158、131−158、136−158、または残基141−158を含む、請求項1ないし12のいずれかのハイブリッドポリペプチド。
- シリアンハムスタープリオンポリペプチドまたは他の種由来のプリオンポリペプチドに対応する残基の少なくとも残基136−158、89−105、89−112または95−112を含む、請求項1ないし12のいずれかのハイブリッドポリペプチド。
- ポリペプチドモチーフがシリアンハムスタープリオンポリペプチドまたは他の種由来のプリオンポリペプチドに対応する残基の唯一の残基136−158、89−105、89−112または95−112を含む、請求項1ないし12のいずれかのハイブリッドポリペプチド。
- PrPc形態のプリオン由来の少なくとも1つのアルファ−ヘリックスを含む残基を含む請求項1ないし15のいずれかのハイブリッドポリペプチド。
- 骨格が抗体b12またはそのフラグメントを含み、プリオンの残基121−158またはその結合部分が配列番号4に示される抗体b12の重鎖の残基119−131の代わりに挿入されている請求項1ないし16のいずれかのハイブリッドポリペプチド。
- 骨格が抗体b12またはそのフラグメントを含み、プリオンの残基87−112またはその結合部分が配列番号4に示される抗体b12の重鎖の残基119−131の代わりに挿入されている請求項1ないし17のいずれかのハイブリッドポリペプチド。
- 骨格が抗体b12の重鎖および軽鎖を含み、重鎖が配列番号4のアミノ酸配列を含み、軽鎖が配列番号2のアミノ酸配列を含む請求項1ないし18のいずれかのハイブリッドポリペプチド。
- シリアンハムスタープリオンポリペプチドまたは他の種由来のプリオンポリペプチドに対応する残基の残基119−158の領域からの少なくとも18、20、25、30、35の連続する残基を含む請求項1ないし19のいずれかのハイブリッドポリペプチドであって:
該領域からの残基がポリペプチドにおけるプリオン由来の残基のみであり、
残基がプリオン配列とシリアンハムスタープリオン配列とのアラインメントにより、配列番号5に示すシリアンハムスタープリオンの残基119−158に対応するポリペプチド。 - プリオンがヒト、ハムスター、マウス、ラット、シカ、ヒツジ、ヤギ、オオジカ、クドゥ、ウマ、イヌ、ネコ、ラクダおよびブタからなる群から選択される動物プリオンである請求項1または請求項20のハイブリッドポリペプチド。
- Fabフラグメントである請求項1のハイブリッドポリペプチド。
- 免疫グロブリンである請求項1のハイブリッドポリペプチド。
- 請求項1ないし23のいずれかのハイブリッドポリペプチドをコードする核酸分子。
- 請求項24の核酸分子を含むベクター。
- 発現ベクターである請求項25ベクター。
- 真核ベクターである請求項25のベクター。
- 作動可能にリンクしたヌクレオチド配列にコードされるポリペプチドの分泌を促すヌクレオチド配列を含む請求項26または27のベクター。
- 哺乳類ベクター、酵母ベクターまたは細菌ベクターである請求項25ないし28のいずれかのベクター。
- ウイルスベクター、ピチアベクターまたは大腸菌ベクターである請求項25のベクター。
- 請求項25ないし30のいずれかのベクターを含む細胞。
- 原核細胞である請求項31の細胞。
- 真核細胞である請求項31の細胞。
- 細菌細胞、酵母細胞、植物細胞、昆虫細胞および動物細胞から選択される請求項31の細胞。
- 哺乳類細胞である請求項33の細胞。
- 以下の工程を含むPrPSc形態のプリオンポリペプチドの検出方法:
感染性アイソフォームのプリオンポリペプチドを含むと考えられるサンプルと、請求項1ないし23のいずれかのハイブリッドポリペプチドとを接触させる工程;および、
サンプル中のPrPScに対する結合を検出する工程。 - サンプルが体液、組織または器官である請求項36の方法。
- サンプルがPrPc形態のプリオンポリペプチドの全てまたは少なくとも約20、25、30、35、45、50、55、60、65、70、75、80、85、90、95、100またはそれ以上の連続するアミノ酸残基を含む請求項36または37の方法。
- サンプル中のプリオンがヒト、ハムスター、マウス、ラット、シカ、ヒツジ、ヤギ、オオジカ、クドゥ、ウマ、イヌ、ネコ、ラクダおよびブタの中から選択される動物プリオンである請求項36ないし38のいずれかの方法。
- サンプルが、血液、尿、汗、唾液、脳脊髄液、精子サンプル、血清、血漿および滑液の中から選択される体液である請求項36ないし39のいずれかの方法。
- 請求項1ないし23のいずれかのハイブリッドポリペプチドの複数を含む固体支持体。
- 以下の工程を含むPrP Sc 形態のプリオンポリペプチドを含む細胞の検出方法:
動物または組織由来の細胞を含むサンプルと請求項1ないし23のいずれかのハイブリッドポリペプチドと接触させる工程、ここでハイブリッドポリペプチドは検出可能に標識されているか、検出可能な骨格を含む;および、
標識された細胞を検出する工程。 - 標識が蛍光標識である請求項42の方法。
- 検出をフローサイトメトリーまたはスキャニングサイトメトリーによって行う請求項43の方法。
- 細胞を複数の異なるハイブリッドポリペプチドと接触させる請求項42の方法。
- ハイブリッドポリペプチドが標的ポリペプチド上の異なるエピトープに結合する請求項45の方法。
