Abstract
MéTODO PARA AMPLIFICAR SEQuêNCIAS DE áCIDO NUCLEICO ESPECìFICAS. A presente invenção proporciona um método de detectar um polimorfismo em um indivíduo. Em uma forma o método compreende as seguintes etapas: (1) obter uma amostra contendo ácido nucleico do indivíduo; (2) amplificar a amostra de ácido nucleico da etapa (1) por um processo envolvendo termociclagem e precursores, a amplificação ocorrendo na presença de uma endonuclease de restrição termoestável que retém atividade durante termociclage, os precursores sendo selecionados de forma que eles introduzam ou no ácido nucleico amplificado do ácido nucleico não incluindo o polimorfismo ou de ácido nucleico incluindo o polimorfismo uma seq³ência reconhecida pela endonuclease de restrição termoestável; e (3) analisar o produto da etapa (2) para determinar a presença ou ausência do polimorfismo.
