Abstract
abstract the present invention relates to a method and a kit for monitoring, diagnosis, prognosis of acute kidney injury in early stage and determination of treatment in subjects suffering thereof. the method comprises the steps of a) providing a urine sample; b) enriching the urine sample in exosomes present in the urine sample using at least one step of immunopurification; c) detecting an acute kidney injury (aki) marker in the exosome. the invention also relates to a kit for determining the presence and/or level of a specific kidney injury marker, for simple and early determination of the onset of aki in a subject, the kit comprising means for enriching the urine sample in exosomes using at least one step of immunopurification, and means for detecting a predetermined kidney injury marker of a condition. the method and kit of the invention have the advantage to provide a specific, simple and early detection of kidney injury markers that is no possible with the standard methods applied until now. the method and kit of the invention also provide the advantage of identifying the specific structure that is injured in the kidney, since it uses specific antibodies for specific proteins present in specific structures of the kidney. --------------------------------------------------------------------------tradução do resumo resumo patente de invenção: "método para monitorização, diagnóstico e/ou prognóstico de lesão renal aguda no estágio inicial". a presente invenção se refere a um método e a um kit para mo-nitorização, diagnóstico, prognóstico de lesão renal aguda no estágio inicial e determinação de tratamento em sujeitos que sofram dela. o método com-preende as etapas de a) obtenção de uma amostra de urina; b) enriqueci-mento da amostra de urina em exossomos presentes na amostra de urina usando pelo menos uma etapa de imunopurificação; c) detecção de um mar-cador de lesão renal aguda (aki) no exossomo. a invenção também se refere a um kit para determinação da presença e/ou nível de um marcador de lesão renal específico, para determinação simples e precoce do início de aki em um sujeito, o kit compreendendo meios para enriquecimento da amostra de urina em exossomos usando pelo menos uma etapa de imunopurificação, e meios para detecção de um marcador de lesão renal predeterminado de uma condição. o método e o kit da invenção têm a vantagem de proporcionarem uma detecção específica, simples e precoce de marcadores de lesão renal, o que não era possível com os métodos padronizados aplicados até agora. o método e o kit da invenção também proporcionam a vantagem de identificar a estrutura específica que está lesionada no rim, pois usam anticorpos específicos para proteínas específicas presentes em estruturas específicas do rim.
