Abstract

"MéTODO E SISTEMA PARA PREPARAçãO DE AMOSTRAS DE TECIDO PARA EXAME HISTOLóGICO E PATOLóGICO". Material biológico viável é criogenicamente preservado (criopreservação) por imersão do material em um tanque de fluido de resfriamento, e circulação do fluido de resfriamento acima do material em uma velocidade e temperatura predeterminadas substancialmente constantes para congelar o material. O material pode ser mergulhado diretamente no fluido de resfriamento sem preparação, ou preparado quimicamente antes do congelamento. Um método de acordo com a presente invenção congela o material biológico de forma rápida o suficiente para evitar a formação de cristais de gelo nas estruturas celulares (vitrificação) e permite que as amostras mantenham a estrutura anatómica e permaneçam bioquimicamente ativas após o descongelamento. A temperatura do fluido de resfriamento é preferivelmente entre -200<198>C e -30O<198>C, que é quente o suficiente para minimizar a formação de fraturas de stress e outros artefatos em membranas celulares devido a mudanças térmicas. Células congeladas que usam um método de acordo com a presente invenção mostrou ter um dano celular e intercelular significativamente menor que as células congeladas por outros métodos de criopreservação usados para técnicas patológicas e histológicas. Devido ao fato da presente invenção poder congelar material biológico de forma que o material seja vitrificado, a atividade bioquímica dentro da célula não é perdida após o congelamento e assim várias modalidades do presente método podem ser empregadas em um sistema para preparar material biológico para as novas técnicas de criopatologia e imunohistoquímica nas áreas de pesquisa e cuidados com pacientes.
