Abstract

"BIBLIOTECAS COMBINATORIAIS DE PROTEìNAS TENDO A ESTRUTURA DE ANDAIME DE DOMìNIOS TIPO LECTINA TIPO C". Uma nova família de bibliotecas de proteína compreendendo CTLDS (<UM>C<MV>-<UM>t<MV>ype <UM>L<MV>ectin-<UM>L<MV>ike <UM>D<MV>omains) nas quais regiões de alça de polipeptídeo internas revestindo os sítios de ligação de ligando nos CTLDs foram substituídas com conjuntos de segmentos de polipeptídeo completamente ou parcialmente aleatorizados. Os CTLDs de tetranectina foram escolhidos como estrutura para a modalidade preferida da invenção; e vetores de fagomídeo versátil úteis na geração e manipulação de bibliotecas de CTLD de tetranectina humana e de murino são descritos como parte desta invenção. Os CTLDs de tetranectina em forma monomérica bem como trimérica são eficientemente exibidos como fusões de gene III em forma totalmente funcional pelo vetor de exibição de fago fd recombinante. Os derivados de CTLD com afinidade para novos ligandos podem facilmente ser isolados de bibliotecas de vetores exibindo CTLDS, nos quais regiões de alça foram aleatorizadas, empregando-se um ou mais ciclos de enriquecimento por avaliação ou seleção seguida por amplificação da subpopulação enriquecida em cada ciclo. A eficiência com a qual produtos de proteína contendo CTLDs com novas propriedades de ligação podem ser produzidos, por exemplo, por expressão bacteriana em redobramento in vitro, em formatos mono-, tri ou multiméricos fornece importantes vantagens em termos de simplicidade, custo e eficiência de geração, produção e aplicações diagnósticas ou terapêuticas em comparação com derivados de anticorpo recombinante.
