Abstract

Se proporcionan vectores retrovirales para administrar ARN de interferencia en células así como células infectadas por dichos vectores, las cuales tienen un oncogen como gen objetivo en su genoma. Reivindicación 1: Un vector retroviral para llevar un insecto especifico del gen objetivo en una célula a fin de modificar la expresión de un gen objetivo, que comprende: (a) un promotor; (b) una región poliligante; (c) un insecto especifico del gen objetivo que comprende ARN de doble hebra, en donde dicho ARN de doble hebra comprende una porción sentido que es complementaria a una porción de la hebra antisentido del gen objetivo, y una porción antisentido que es complementaria a la porción sentido, de modo que la porción sentido y la porción antisentido se hibridan, y el ARN de doble hebra se pliega para atrás sobre si mismo. Reivindicación 8: El vector retroviral de acuerdo con la Reivindicación 1, en el cual el vector retroviral es un virus de célula pluripotente murino (MSCV, abreviatura del ingles "Murine Stem Cell Virus"). Reivindicación 9: El vector retroviral de acuerdo con la Reivindicación 1, en el cual el vector retroviral es un Lentivirus modificado en el cual: (a) el promotor de CMV endógeno del Lentivirus ha sido eliminado; y (b) un elemento REV que se une a un elemento de respuesta REV (RRE, abreviatura del ingles "REV response element") es insertado.
