본 발명은 베타-갈락토시다아제(beta-galactosidase, β-galactosidase)가 발현되도록 형질전환된 재조합 코리네박테리움 글루타미쿰(Corynebacterium glutamicum)을 이용한 푸코실락토오스(fucosyllactose, FL)의 생산방법에 관한 것으로, 더욱 상세하게는 FL 생산을 위해 코리네박테리움 글루타미쿰에 베타-갈락토시다아제가 발현되도록 형질전환시킴으로써 기질로 락토오스(lactose)을 사용하고 남는 잔여 락토오스를 제거하여 고순도로 푸코실락토오스의 생산을 용이하게 하는 방법에 관한 것이다.
사람의 모유에는 약 200여종 이상의 다양한 모유올리고당 (Human milk oligosaccharides, HMOs)이 존재하며, 이는 유당 및 지방 다음으로 모유에서 세 번째로 많은 성분이다. 모유올리고당은 면역 기능을 강화시키거나, 아이의 발달과 행동에 좋은 영향을 주는 등의 다양한 이점을 가지며, 그 중에서도 2'-푸코실락토오스(2'-fucosyllactose, 2'-FL)와 3-푸코실락토오스(3-fucosyllactose, 3-FL)는 다양한 생리활성에 관여하는 주요 HMO인 것으로 보고되어 있다.
기존 연구에서 푸코실락토오스를 생산하는 방법은 직접 모유로부터 추출하는 방법과 화학적 또는 효소적 방법으로 합성하는 방법이 있다. 직접 모유로부터 추출하는 방법은 비윤리적이라는 것과, 모유 수급이 제한적이고 낮은 생산성이라는 문제가 있고, 화학적 합성법은 고가의 기질, 낮은 이성체 선택성(stereo-selectivity)과 생산수율, 그리고 독성 유기용매의 사용 등으로 인한 문제가 있다. 또한, 효소적 합성법은 전구체가 되는 푸코오스의 공여체로 이용되는 GDP-L-푸코오스가 매우 고가라는 점과 푸코오스 전이효소의 정제비용이 많이 든다는 문제점이 있다.
이에 상기와 같은 모유로부터의 직접 추출법, 화학적 또는 효소적 합성방법은 푸코실락토오스를 대량으로 생산하기에는 한계가 있고, 이를 해결하기 위한 방안으로 미생물을 이용하는 방법이 있다. 하지만 이역시 대부분 재조합 대장균을 이용한 생산방법임에 따라 대장균에 대한 소비자들의 인식이 해로운 병원균이라는 부정적인 견해가 많고, 대장균 세포막 성분이 엔도톡신으로 작용할 수 있기 때문에 분리, 정제 비용이 많이 소요된다는 문제가 있으며, 락토오스 제한 배양상태에서는 락토오스 투과효소(lactose permease)의 작용에 의해 락토오스 킬링(lactose killing)이라는 대장균 세포가 사멸되는 현상이 나타나므로 사용하는 것이 다소 제한적이라는 한계가 있다.
이에 기존의 여러 방법들을 보완할 수 있으면서 푸코실락토오스의 생산 및 정제과정에서의 불필요한 요소들을 최소화하고 이로 인한 고순도의 푸코실락토오스의 생산 및 정제를 위한 새로운 기술에 대해 필요성이 높은 실정이다.
본 발명은 숙주세포로서 대장균보다 안전한 코리네박테리움 글루타미쿰 (Corynebacterium glutamicum)을 이용하되, 기질로 사용하고 남는 세포 외부의 잔여 락토오스를 제거하기 위해 베타-갈락토시다아제(beta-galactosidase, β-galactosidase)가 발현되도록 형질전환시킴으로써 고농도, 고수율, 고생산성으로 푸코실락토오스(2fucosyllactose, FL)를 생산하는 방법을 개발하여 제공하고자 한다.

한편, 본 발명의 재조합 코리네박테리움 글루타미쿰 (Corynebacterium glutamicum)에 있어서, 상기 재조합 코리네박테리움 글루타미쿰(Corynebacterium glutamicum)은, 바람직하게 포스포만노뮤타아제 (Phosphomannomutase)가 과발현되도록 형질전환되며, GTP-만노오스-1-포스페이트 구아닐트랜스퍼라아제 (GTP-mannose-1-phosphate guanylyltransferase)가 과발현되도록 형질전환된 것이 좋다.
또한, 본 발명은 락토오스가 첨가된 배지에, 상기 재조합 코리네박테리움 글루타미쿰(Corynebacterium glutamicum)을 배양하는 것을 특징으로 하는 푸코실락토오스(fucosyllactose)의 생산방법을 제공한다.
한편, 본 발명의 푸코실락토오스(fucosyllactose)의 생산방법에 있어서, 상기 배지는, 바람직하게 글루코오스를 더 포함하고 있는 것이 좋다.
본 발명은 베타-갈락토시다아제가 발현되도록 형질전환된 재조합 코리네박테리움 글루타미쿰을 이용하여 기질로 사용되고 남는 세포 외부의 잔여 락토오스를 제거하여 고농도, 고수율, 고생산성으로 푸코실락토오스(fucosyllactose, FL)를 생산할 수 있다.
도 1은 본 발명의 베타-갈락토시다아제가 발현되도록 형질전환된 균주(LacZ(intra), 시그널펩타이드를 이용하여 베타-갈락토시다아제가 발현되도록 형질전환된 균주(LacZ(extra)의 세포 성장 패턴을 비교한 결과 그래프이다.
도 2는 본 발명의 베타-갈락토시다아제가 발현되도록 형질전환된 균주(LacZ(intra), 시그널펩타이드를 이용하여 베타-갈락토시다아제가 발현되도록 형질전환된 균주(LacZ(extra)의 락토오스 농도(세포 내/외를 합친 것)를 비교한 결과 그래프이다.
도 3은 본 발명의 베타-갈락토시다아제가 발현되도록 형질전환된 균주(LacZ(intra), 시그널펩타이드를 이용하여 베타-갈락토시다아제가 발현되도록 형질전환된 균주(LacZ(extra)의 2'-푸코실락토오스 생산성을 비교한 결과 그래프이다.
도 4는 본 발명의 온도 유도로 발현되는 베타-갈락토시다아제가 발현되도록 형질전환된 균주(LacZ(intra), 시그널펩타이드를 이용하여 베타-갈락토시다아제가 발현되도록 형질전환된 균주(LacZ(extra)를 35℃에서 온도에 의해 6시간 발현을 유도하고 25℃에서 96시간까지 배양 후 잔여 락토오스 농도 및 2'-푸코실락토오스생산성을 비교한 결과 그래프이다.
모유올리고당은 면역 기능을 강화시키거나, 아이의 발달과 행동에 좋은 영향을 주는 등의 다양한 이점을 가지며, 그 중에서도 2'-푸코실락토오스(2'-fucosyllactose, 2'-FL)와 3-푸코실락토오스(3-fucosyllactose, 3-FL)는 다양한 생리활성에 관여하는 주요 HMO인 것으로 보고되어 있다. 이를 생산하기 위한 기존의 대표적인 방법들(직접 모유로부터 추출하는 방법, 화학적 또는 효소적 합성방법, 주로 대장균을 이용한 생산방법)에는 한계가 있으므로 본 발명에서는 이를 보완할 수 있으면서 푸코실락토오스의 생산 및 정제과정에서의 불필요한 요소들을 최소화하고 이로 인한 고순도의 푸코실락토오스 생산 및 정제를 위한 새로운 기술을 개발하였다.
기존 연구에 의하면 푸코실락토오스를 생산하기 위해 대장균을 사용하였으나, 대장균의 경우 세포 내 베타-갈락토시다아제(beta-galactosidase, β-galactosidase)를 보유하고 있음에 따라 세포 내로 들어오는 락토오스, 즉 푸코실락토오스의 기질이 되는 락토오스가 분해되므로 푸코실락토오스의 생산이 감소할 수 있다. 이에 생산성을 높이고자 베타-갈락토시다아제의 활성을 억제 또는 저해하는 기술이 연구된 바가 있다.
한편, 본 발명에서는 코리네박테리움 글루타미쿰(Corynebacterium glutamicum)을 이용하여 푸코실락토오스를 생산함에 있어, 생산성을 늘리기 위한 방법의 일환으로 락토오스를 최대한 효과적으로 이용하여 푸코실락토오스의 생산성을 늘릴 수 있는 기술을 찾고자 노력하였다.
락토오스는 푸코실락토오스를 생산함에 있어 기질이 되는 중요한 물질이나, 반응 후 잔존하는 미반응 락토오스는 푸코실락토오스의 정제 과정에서 부담(load)으로 작용하고, 이당체인 푸코실락토오스와 미반응 락토오스를 분리·정제 과정에서 고순도로 분리하기란 여의치 않다. 따라서, 미반응 락토오스를 제거하여 푸코실락토오스의 순도를 높이는 기술의 개발이 분리·정제 과정의 효율성 증가를 위해 요구되는 것이다.
한편, 코리네박테리움 글루타미움은 대장균과는 달리, 세포 내 베타-갈락토시다아제가 없는데, 이와 같은 사실로부터 본 발명에서는 미반응 락토오스를 제거하기 위해 베타-갈락토시다아제를 사용해 보는 것을 착안하였다.
그런데, 베타-갈락타오시다아제를 상시 발현시키면, 푸코실락토오스 생산의 기질이 되는 락토오스가 분해되어 오히려 푸코실락토오스의 생산성이 떨어질 수 있는 문제가 발생한다. 즉, 베타-갈락토시다아제의 활성이 상시적으로 유지된다면 푸코실락토오스의 기질이 되는 락토오스가 분해되어 푸코실락토오스의 생산성이 감소할 수 있는 것이다. 따라서, 본 발명에서는 푸코실락토오스의 생산 중, 즉 배양 중에는 베타-갈락토시다아제가 발현되지 않도록 막아 놨다가 푸코실락토오스의 생산이 어느 정도 완료된 후, 베타-갈락토시다아제의 발현을 유도하여 미반응 잔류 락토오스를 제거하고자 하였다. 이를 위해 본 발명에서는 베타-갈락토시다아제의 발현을 유도성 프로모터를 사용하여 조절하고자 한 것이다.
유도성 프로모터 (inducible promoter)는 화합물에 의해서 발현이 유도되는 것, 온도의 변화에 의해 발현이 유도되는 것 등 다양한 형태가 있는데, 본 발명에서는 온도의 변화에 의해 발현이 유도되는 온도 감음형 유도성 프로모터를 사용하는 것이 좋다. 화합물을 사용하면 잔류 화합물이 배지 중에 잔존하게 되어 이 또한 분리·정제 과정에서 부담으로 작용하기 때문이다.
한편, 본 발명에서는 바람직하게 유도성 프로모터에 의해 발현이 조절되는 베타-갈락토시다아제가 균체 밖으로 분비되는 것이 좋다. 본 발명에서, 배지 중에 존재하는 미반응 락토오스는, 분리·정제 문제 때문에 제거하는 것이 좋으나, 균체 내부에 존재하는 락토오스는, 푸코실락토오스 생산을 위한 기질로 사용되기 때문에, 베타-갈락토시다아제에 의해 분해되는 것이 바람직하지 않다. 따라서, 발현을 시킨 베타-갈락토시다아제는 균체 내부에 머무르게 하지 않고 균체 밖으로 분비하는 것이 좋은 것이다. 베타-갈락토시다아제를 균체 외부로 배출하는 것에는 다양한 방법이 있으나, 바람직하게는 시그널펩타이드를 사용하여 균체 외부로 분비하는 것이 좋다.
한편, 본 발명에서, 상기, 균체 밖으로 배출되는, 유도성 프로모터에 의해 발현이 조절되는 베타-갈락토시다아제는, 균체 내에서 활성이 나타나지 않고, 균체 밖으로 배출된 후 활성이 나타나는 것이 더욱 좋다. 이를 통해 균체 내부에 있는 락토오스의 분해를 최대한 억제하고, 균체 외부 즉 배지 중에 있는 잔여 미반응 락토오스를 효율적으로 분해할 수 있는 것이다.
이를 위해, 본 발명에서는 베타-갈락토시다아제를 발현하도록 코리네박테리움 글루타미쿰을 형질전환시키되, 세포질(균체 안쪽)에서 베타-갈락토시다아제 활성이 나타나지 않고 세포외(균체 바깥쪽)에서 그 활성이 나타날 수 있는 시스템이 구축된 재조합 코리네박테리움 글루타미쿰을 제작하였다. 그 일환으로, '시그널 펩타이드(signal peptide)'를 이용하여 베타-갈락토시다아제가 발현되도록 형질전환된 코리네박테리움 글루타미쿰을 제작하였으며, 시그널 펩타이드가 연결됨에 따라 세포질(안쪽)에서는 베타-갈락토시다아제의 활성이 나타나지 않으나, 세포외(바깥쪽)에서는 폴딩되면서 베타-갈락토시다아제의 활성이 나타나 배지 중 미반응 잔류 락토오스가 분해되도록 하였다.
한편, 본 발명에서는 상기에서 기술한 바와 같이, 미반응 잔류 락토오스의 분해를 위해 베타-갈락토시다아제를 도입하였으나, 베타-갈락토시다아제의 도입에 따라 푸코실락토오스의 생산량이 증대하는 예상치 못한 결과를 확인할 수 있었다. 락토오스가 분해되면 분해 산물로 포도당이 생성되는데, 생성된 포도당은 세포 내로 유입되어 푸코실락토오스 합성 반응의 또 다른 기질인 구아노신이인산-L-푸코오스 (GDP-L-fucose)의 생산에 사용된다. GDP-L-fucose는 락토오스와 같이 푸코실락토오스 생산의 전구체인데, 생성된 포도당은 GDP-L-fucose의 생성에 사용되어, 궁극적으로 푸코실락토오스의 생산량을 증대시킬 수 있게 된 것이다. 즉, 본 발명은 미반응 부산물로서 정제 과정에 부담이 되는 락토오스를 분해시켜 제거하면서도, 이를 푸코실락토오스의 생산에도 활용할 수 있어, 푸코실락토오스의 생산량을 증가시킬수 있게 되는 것이다. 정리하자면, 본 발명은 락토오스가 갖는 분리·정제 문제를 해결하면서도 푸코실락토오스의 생산량도 증대시킨 것이다.
이에 본 발명은 푸코오스 전이효소 (fucosyltransferase)가 발현되도록 형질전환되고, GDP-D-만노오스-4,6-데하이드라타아제 (GDP-D-mannose-4,6-dehydratase, 'Gmd'로도 불리며 이하 동일)가 발현되도록 형질전환되며, GDP-L-푸코오스 신타아제 (GDP-L-fucose synthase, 'WcaG'로도 불리며 이하 동일)가 발현되도록 형질전환되고, 락토오즈 퍼미아제 (lactose permease, 'LacY'로도 불리며 이하 동일)가 발현되도록 형질전환되며, 베타-갈락토시다아제(beta-galactosidase)가 발현되도록 형질전환되고, 포스포만노뮤타아제 (Phosphomannomutase) 및 GTP-만노오스-1-포스페이트 구아닐트랜스퍼라아제 (GTP-mannose-1-phosphate guanylyltransferase)를 보유하고 있되, 상기 베타-갈락토시다아제(beta-galactosidase)는 유도성 프로모터에 발현이 조절되는 것을 특징으로 하는 푸코실락토오스(fucosyllactose) 생산용 재조합 코리네박테리움 글루타미쿰 (Corynebacterium glutamicum)을 제공한다. 이때, 상기 푸코오스 전이효소로는, α-1,2-푸코오스 전이효소 (α-1,2-fucosyltransferase)를 사용하면 2'-푸코실락토오스를 생산할 수 있고 α-1,3-푸코오스 전이효소 (α-1,3-fucosyltransferase)를 사용하면 3-푸코실락토오스를 생산할 수 있다.
한편, 상기 베타-갈락토시다아제(beta-galactosidase)는, 어느 미생물로부터 유래된 것이든 제한되지는 않으나 일 예로 대장균(E.coli), 락토바실러스 속(Lactobacillus sp.), 비피도박테리아 속(Bifidobacteria sp.)인 것이 좋으며, 바람직하게는 비피도박테리움 롱검(Bifidobacterium longum)인 것이 좋으며, 더욱 바람직하게는 비피도박테리움 롱검(Bifidobacterium longum) ATCC 15707인 것이 좋다.
한편, 본 발명의 재조합 코리네박테리움 글루타미쿰 (Corynebacterium glutamicum) 내 베타-갈락토시다아제의 발현 시스템을 구축하기 위한 요소 중 하나로 유전자의 전사를 조절하는 부위인 '프로모터(promoter)'를 사용하게 된다. 통상적으로 프로모터에는 상시 발현 프로모터(constitutive promoter)와 유도성 프로모터(inducible promoter, 다른 말로 '조건부 발현 프로모터'라고도 함)가 있다. 본 발명에서는 유도성 프로모터를 사용하는 것이 좋은데, 유도성 프로모터라면 당 업계에 공지된 것이라면 어느 것이든 사용할 수 있다.
본 발명에서 유도성 프로모터로는 바람직하게 열 유도 프로모터(heat inducible promoter, thermal inducible promoter)를 사용하는 것이 좋다. 더욱 바람직하게는 dnaK의 프로모터(PdnaK)를 사용하는 것이 좋다. 이를 통해 코리네박테리움 글루타미쿰의 일반적인 배양온도 (30℃ 내외)에서는 발현이 되지 않고, 온도를 35℃로 높일 경우 발현될 수 있게 한다. 즉, 코리네박테리움 글루타미쿰의 성장 최적온도는 25~37℃이고, 40℃ 이상의 온도에서는 성장이 둔화되는데, 코리네박테리움 글루타미쿰의 생육과정에서 증식이 끝나면 사멸기에 들어가기 때문에 적절한 시기의 발현은 성장에 큰 영향을 미치지 않을 수 있다.
본 발명에서는 열 유도 프로모터인 PdnaK를 물질대사가 활발한 성장시기인 대수기에 발현을 시켜 온도 변형(temp. shif)을 통해 원하는 시기에 베타-갈락토시다아제의 발현 활성을 유도한 후 최적 온도로 돌아가 배양을 진행하였다. 이를 통해 락토오스가 제거되는 정도를 온도 변화를 통해 확인하였다.
한편, 본 발명의 재조합 코리네박테리움 글루타미쿰 (Corynebacterium glutamicum) 내 베타-갈락토시다아제 발현 시스템을 구축할 시, 베타-갈락토시다아제의 발현은 원래의(native) 베타-갈락토시다아제 발현 카세트를 도입하거나 시그널 펩타이드(signal peptide)가 연결된 베타-갈락토시다아제 발현 카세트를 도입할 수 있다.
바람직하게는 시그널 펩타이드가 연결된 베타-갈락토시다아제 발현 카세트를 도입하는 것이 좋다(유전자의 N-말단에 시그널 펩타이드를 추가하여 발현시키는 것). 시그널 펩타이드를 도입함으로써 베타-갈락토시다아제를 균체 외부로 배출시킬 수 있게 된다. 또한, 균체 내에서 베타-갈락토시다아제의 활성이 나타나지 않고 균체 밖으로 배출된 후 베타-갈락토시다아제의 활성이 나타나게 된다.
본 발명에서는 세포 외부의 잔여 락토오스(lactose)를 제거하기 위해 베타-갈락토시다아제를 세포 밖으로 분비할 수 있게 함으로써 푸코실락토오스의 생산성을 높이면서 베타-갈락토시다아제 활성으로 인해 부산물 (By-product)이 되는 락토오스 농도가 낮아지게 되는 최적 생산을 가능하게 할 수 있게 되었다. 더욱 바람직하게는 서열번호 17의 핵산서열을 갖는 시그널 펩타이드를 사용한 것일 수 있다.
한편, 본 발명의 재조합 코리네박테리움 글루타미쿰 (Corynebacterium glutamicum)에 있어서, 상기 재조합 코리네박테리움 글루타미쿰(Corynebacterium glutamicum)은, 바람직하게 포스포만노뮤타아제 (Phosphomannomutase)가 과발현되도록 형질전환되며, GTP-만노오스-1-포스페이트 구아닐트랜스퍼라아제 (GTP-mannose-1-phosphate guanylyltransferase)가 과발현되도록 형질전환된 것이 좋다. 코리네박테리움 글루타미쿰은 포스포만노뮤타아제 (Phosphomannomutase, ManB), GTP-만노오스-1-포스페이트 구아닐트랜스퍼라아제 (GTP-mannose-1-phosphate guanylyltransferase, ManC)를 암호화하는 유전자를 자체적으로 보유하여 발현시킬 수 있기 때문에, 굳이 이 효소를 암호화하는 유전자를 도입시켜줄 필요는 없으나, 대량 생산을 위해서는 이 효소를 과발현시켜줄 필요가 있다. 따라서, 본 발명에서는 바람직하게 이들 두 효소를 과발현할수 있도록 코리네박테리움 글루타미쿰을 형질전환하는 것이 필요한 것이다.
한편, 본 발명에서 사용하는 '발현'이라는 용어는, 본 발명의 코리네박테리움 글루타미쿰 균주가 자체적으로 발현시킬 수 없는 효소를, 인위적으로 발현시키기 위해 외부 유래의 유전자를 균주 내로 도입하여 발현시키는 것을 의미하고, '과발현'이라는 용어는 본 발명의 코리네박테리움 글루타미쿰 균주가 자체적으로 해당 효소를 암호화하는 유전자를 가지고 있어, 스스로 발현시킬 수 있으나, 대량생산을 위한 목적으로 이의 발현량을 증대시키기 위해 인위적으로 해당 효소의 발현량을 증대시켜 과발현한 것을 의미한다.
또한, 본 발명은 락토오스가 첨가된 배지에, 상기 재조합 코리네박테리움 글루타미쿰(Corynebacterium glutamicum)을 배양하는 것을 특징으로 하는 푸코실락토오스(fucosyllactose)의 생산방법을 제공한다.
한편, 본 발명의 푸코실락토오스(2'-fucosyllactose)의 생산방법에 있어서, 상기 배지는, 바람직하게 글루코오스를 더 포함하고 있는 것이 좋다. 이와 같이 추가 배지 성분을 더함으로써 균주의 생육이 활발해져 더욱 높은 생산성으로 푸코실락토오스를 생산할 수 있다. 또한, 이를 위해서는 유가식 배양을 통해 글루토오스 또는 락토오스를 지속적으로 공급하면, 세포의 성장을 더욱 증대시키고, 고순도, 고수율, 고생산성으로 푸코실락토오스를 생산할 수 있다. 유가식 배양에 관한 세부 지엽적 기술들은 당업계의 공지 기술을 사용할 수 있으므로, 이에 대해서는 그 기재를 생략하기로 한다.
한편, 하기 실험에 의하면, 본 발명의 베타-갈락토시다아제가 발현되도록 형질전환된 재조합 코리네박테리움 글루타미쿰은 락토오스 분해 활성, 즉 베타-갈락토시다아제 활성이 있음에 따라 형질전환되지 않은 코리네박테리움 글루타미쿰에 비교하여 락토오스가 분해되며, 잔여 락토오스 농도가 낮은 것을 확인할 수 있었다. 특히, 시그널 펩타이드를 이용하여 베타-갈락토시다아제가 발현되도록 형질전환된 재조합 코리네박테리움 글루타미쿰에서 세포 밖으로 베타-갈락토시다아제를 분비하도록 함에 따라, 기질로 사용할 락토오스의 농도도 충분하여 2'-푸코실락토오스의 생산량을 늘리면서도 대조군보다 잔여 락토오스가 낮음에 따라 2'-푸코실락토오스의 원활한 분리 정제가 가능하게 된 것이다. 한편, 2'-푸코실락토오스의 생산에 관해 본 발명의 생산 전략이 효과적으로 검증되어 그 유효성이 확인된바, 3-푸코실락토오스로 적용시에도 일응 동일한 효과가 나올 것으로 판단된다.
이하, 본 발명의 내용을 하기 실시예를 통해 더욱 상세히 설명하고자 한다. 다만, 본 발명의 권리범위가 하기 실시예에만 한정되는 것은 아니고, 그와 등가의 기술적 사상의 변형까지를 포함한다.

[실시예 1: 베타-갈락토시다아제가 발현되도록 형질전환된 재조합 코리네박테리움 글루타미쿰 제작]
본 실시예에서는 코리네박테리움 글루타미쿰(Corynebacterium glutamicum)에서 열 유도 프로모터(heat inducible promoter, thermal inducible promoter)를 이용하고, 베타-갈락토시다아제가 발현되도록 형질전환된 재조합 코리네박테리움을 제작하였다.
열 유도 프로모터로 알려진 dnak의 프로모터(Pdnak), 코리네박테리움 글루타미쿰(Corynebacterium glutamicum 유래)와 비피도박테리움 롱검(Bifidobacterium longum) ATCC 15707 유래 베타-갈락토시다아제를 표 2개의 프라이머 2개 세트(F-linker-pdnaK-g, R-pdnaK-g; F-lacZ(15707)-g, R-linker-ter-g)로 각각 유전자를 증폭한 후, 증폭된 프로모터(Pdnak), 베타-갈락토시다아제 주형을 이용하여 프라이머 'F-linker-pdnaK-g'와 'R-linker-ter-g'로 1차 overlap PCR을 수행하였다.
증폭한 PdnaK-beta-galactosidase DNA 주형은 코리네박테리움 글루타미쿰 ATCC 13032 유래의 NCgl2392를 Knock-out한 위치에 도입하기 위해 NCgl2392 N-말단(N-terminal)과 C-말단(C-terminal) 부분으로부터 500bps의 유전자를 프라이머 'F-pK19-NCgl2392-1'와 'R-pK19-NCgl2392-2'; 'F-pK19-NCgl2392-3'와 'R-pK19-NCgl2392-4'로 증폭한 후, 증폭된 2개의 주형을 이용하여 프라이머 'F-pK19-NCgl2392-1'와 'R-pK19-NCgl2392-4'로 overlap PCR을 수행하여 'pK19mobsacB-NCgl2392' 벡터를 제작하였다.
증폭한 PdnaK-beta-galactosidase DNA 주형은 제한효소 NotI에 의해 절단된 pK19mobsacB-NCgl2392 벡터에 삽입하여 pK19mobsacB-PdnaK-beta-galactosidase 플라스미드를 구축하였고 플라스미드를 코리네박테리움 글루타미쿰 ATCC 13032에 형질전환하고, 2차 교차 과정을 거쳐 염색체 상에서 1 카피의 PdnaK-beta-galactosidase 유전자를 삽입하였다.
2'-푸코실락토오스의 생산을 확인하기 위해 PdnaK-beta-galactosidase가 도입된 코리네박테리움 글루타미쿰 ATCC 13032에 pFGW(Ps) 플라스미드와 pXIL 플라스미드를 형질전환하였다.
또한, 3-푸코실락토오스의 생산을 확인하기 위해 PdnaK-beta-galactosidase가 도입된 코리네박테리움 글루타미쿰 ATCC 13032에 pFGW(Ab) 플라스미드와 pXIL 플라스미드를 형질전환하였다.
pFGW(Ab) 플라스미드와 pFGW(Ps) 벡터는 특허 제10-2014925호에서 사용하였으며, pFGW(Ab)는 pFGW(Ps) 벡터를 bglII와 XbaI 제한효소 처리하여 Pseudopedobacter saltans (Ps) 유래의 α-1,2-푸코오스 전이효소를 제거하고 pFGW(Ps)를 주형으로 F_tuf_bglII 와 R_tuf_azoT 프라이머를 이용하여 PCR 하고 F-tuf-azoT와 R-azoT를 이용하여 합성된 Azospirillum brasilense 유래의 azoT를 PCR한 후 F-tuf-azoT와 R-azoT로 overlap PCR 하여 얻은 증폭물을 제한효소로 전달된 pFGW(Ps)에 삽입하여 pFGW(Ab)를 제작하였다.
[실시예 2: 시그널 펩타이드(signal peptide)를 이용하여 베타-갈락토시다아제가 발현되도록 형질전환된 재조합 코리네박테리움 글루타미쿰 제작]
본 실시예에서는 코리네박테리움 글루타미쿰(Corynebacterium glutamicum)에서 열 유도 프로모터(heat inducible promoter, thermal inducible promoter)를 이용하고, 베타-갈락토시다아제 발현 벡터에 시그널 펩타이드를 추가하여 베타-갈락토시다아제가 발현되도록 형질전환된 재조합 코리네박테리움을 제작하였다.
열 유도 프로모터로 알려진 dnak의 프로모터(Pdnak), 시그널 펩타이드, 베타-갈락토시다아제를 프라이머 3개 세트(F-linker-pdnaK-g, R-pdnaK-SP-g; F-pdnaK-SP-g, R-SP-lacZ(15707)-g; F-SP-lacZ(15707)-g, R-linker-ter-g)로 각각 유전자를 증폭한 후, 증폭된 프로모터(Pdnak), 시그널 펩타이드 주형을 이용하여 프라이머 'F-linker-pdnaK-g'와 'R-SP-lacZ(15707)-g'로 1차 overlap PCR을 수행한 1차 증폭물과 베타-갈락토시다아제와 'F-linker-pdnaK-g'와 'R-linker-ter-g'로 2차 overlap PCR을 수행하였다.
증폭한 'PdnaK-시그널펩타이드(signal peptide)-베타-갈락토시다아제(beta-galactosidase)' DNA 주형은 코리네박테리움 글루타미쿰 ATCC 13032 유래의 NCgl2392를 Knock-out한 위치에 도입하기 위해 NCgl2392 N-말단(N-terminal)과 C-말단(C-terminal) 부분으로부터 500 bps의 유전자를 프라이머 'F-pK19-NCgl2392-1'와 'R-pK19-NCgl2392-2'; 'F-pK19-NCgl2392-3'와 'R-pK19-NCgl2392-4'로 증폭한 후, 증폭된 2개의 주형을 이용하여 프라이머 'F-pK19-NCgl2392-1'와 'R-pK19-NCgl2392-4'로 overlap PCR을 수행하여 'pK19mobsacB-NCgl2392' 벡터를 제작하였다.
증폭한 'PdnaK-signal peptide-beta-galactosidase' DNA 주형은 제한효소 NotI에 의해 절단된 'pK19mobsacB-NCgl2392' 벡터에 삽입하여 'pK19mobsacB-PdnaK-signal peptide-beta-galactosidase' 플라스미드를 구축하였고 플라스미드를 코리네박테리움 글루타미쿰 ATCC 13032에 형질전환하고, 2차 교차 과정을 거쳐 염색체 상에서 1 카피의 'PdnaK-signal peptide-beta-galactosidase' 유전자를 삽입하였다.
2'-푸코실락토오스의 생산을 확인하기 위해 'PdnaK-signal peptide-beta-galactosidase'가 도입된 코리네박테리움 글루타미쿰 ATCC 13032에 pFGW(Ps) 플라스미드와 pXIL 플라스미드를 형질전환하였다.
또한, 3-푸코실락토오스의 생산을 확인하기 위해 'PdnaK-signal peptide-beta-galactosidase'가 도입된 코리네박테리움 글루타미쿰 ATCC 13032에 pFGW(Ab) 플라스미드와 pXIL 플라스미드를 형질전환하였다.
[실험예 1: 본 발명 실시예 2의 시그널 펩타이드(signal peptide)를 이용하여 베타-갈락토시다아제가 발현되도록 형질전환된 재조합 코리네박테리움 글루타미쿰의 2'-푸코실락토오스 생산 확인]
본 실험예에서는 상기 실시예 1과 2에서 구축한 균주 중 2'-푸코실락토오스 생산을 위한 균주를 사용하여 2'-푸코실락토오스 생산을 확인하였다.
회분식 배양은 최소배지 ((NH4)2SO4 20 g/L, Urea 5 g/L, KH2PO4 1 g/L, K2HPO4 1 g/L, MgSO4·7H2O 0.25 g/L, MOPS 42 g/L, CaCl2 10 mg/L, Biotin 0.2 mg/L, Protocatechuic acid 30 mg/L, FeSO4·7H2O 10 mg/L, MnSO4·H2O 10 mg/L, ZnSO4·7H2O 1 mg/L, CuSO4 0.2 mg/L, NiCl2·6H2O 0.02 mg/L, Glucose 40 g/L, Lactose 10 g/L, pH7.0) 50 mL가 담긴 250 mL 플라스크를 이용하였다.
초기 24시간 25℃에서 배양한 후 온도를 35℃로 변경하여 6시간 동안 베타-갈락토시다아제 발현을 유도하면서 배양을 진행하였다. 온도 변경 후 배양 온도인 25℃로 낮추어 배양을 진행해 최종 배양은 96시간으로 완료하였다. 배양을 하며 베타-갈락토시다아제의 발현이 세포 성장에 미치는 영향을 조사하였으며(도 1), 온도 변경을 통한 베타-갈락토시다아제 발현 유도 후 락토오스(세포 내에서의 락토오스의 양+세포 외에서의 락토오스의 양)의 감소 패턴과 2'-푸코실락토오스의 생산을 비교하였다(도 2, 3).
그 결과, 베타-갈락토시다아제가 형질전환되지 않은 균주(control)와 비교하여 형질전환된 두 균주는 도 1과 같이 세포 성장이 잘 이루어지면서도, 도 2 및 도 3과 같이 35℃로의 온도 변경으로 베타-갈락토시다아제가 발현됨에 따라 잔여 락토오스가 감소되는 것으로 나타났고, 2'-푸코실락토오스의 생산량도 증가한 것으로 나타났다.
구체적으로 결과를 살펴보면, 도 2와 같이, 시그널 펩타이드를 이용하여 형질전환시킨 균주(LacZ(extra)) 및 시그널 펩타이드를 이용하지 않고 형질전환시킨 균주(LacZ(intra)) 모두 대조군보다 락토오스의 농도가 낮게 나타났다. 이는, 베타갈락토시다아제의 발현에 의해 락토오스가 분해되었음을 확인시켜 주는 결과이다. 또한, 도 3에서 보면, 대조군 대비 시그널 펩타이드를 이용하여 형질전환시킨 균주(LacZ(extra)) 및 시그널 펩타이드를 이용하지 않고 형질전환시킨 균주(LacZ(intra)) 모두 2'-푸코실락토오스의 생산량이 증가한 것을 확인할 수 있었다. 이는, 잔여 락토오스를 효과적으로 분해 또는 활용하여, 2'-푸코실락토오스의 생산량을 증가시킨 것으로 해석할 수 있다.
한편, 도 2를 보면, 시그널 펩타이드를 이용하여 형질전환시킨 균주(LacZ(extra))의 락토오스 농도 (세포 내 + 세포 외)가, 시그널 펩타이드를 이용하지 않고 형질전환시킨 균주(LacZ(intra))에 비해 높게 나타났는데, 이는 시그널 펩타이드의 사용으로 말미암아 효소의 접힘이 세포 내에서 일어나지 않고(unfolding), 세포막을 통해 밖으로 분비되며 시그널 펩타이드와 분리가 되고 접힘(folding)이 되어 활성형이 되므로 세포 내 존재하는 락토오스 분해가 일어났을 가능성이 낮은 것으로 해석할 수 있고, 이는 본 발명에서 예상하는 바와 일치한 결과이었다. 즉, 시그널 펩타이드를 붙여서 발현시켜 세포 밖으로 베타-갈락토시다아제를 분비하도록 하여 세포 내에서 분해되는 락토오스를 최소화함으로써 세포 내 존재하는 LacZ(extra)의 락토오스 농도가 LacZ(intra)가 높아 발생한 결과인 것이다.
정리하자면, 2'-푸코실락토오스의 원활한 분리 정제를 위해서는 잔여 락토오스의 농도가 적은게 좋은데, 베타-갈락토시다아제가 형질전환되지 않은 균주(control)과 비교할시 형질전환된 두 균주 모두 잔여 락토오스의 농도가 감소되는 것을 확인하였다. 다만, 세포 내에 존재하는 락토오스가 너무 많이 분해되면, 2'-푸코실락토오스의 생산에 사용되는 기질이 줄어들기 때문에 2'-FL의 전체적인 생산성 측면에서는 바람직하지 못하다. 그런데 본 발명의 실험에 의할 경우, LacZ(extra) 실험군은 잔류 락토오스의 농도가 control보다는 낮으면서도, LacZ(intra)보다 높게 나타나고, 2'-FL의 생산량 또한 LacZ(intra)보다 높게 나타나, 2'-FL의 생산에 있어, LacZ(extra) 모델이 보다 효과적인 모델로 판단할 수 있었다(도 2, 3, 4).
한편, 상기와 같은 결과는 2'-푸코실락토오스의 생산에 관한 것이지만, 본 발명의 생산 전략이 효과적으로 검증되어 그 유효성이 확인된바, 3-푸코실락토오스로 적용시에도 일응 동일한 효과가 나올 것으로 판단된다.
Claims (6)
- 푸코오스 전이효소 (fucosyltransferase)가 발현되도록 형질전환되고, GDP-D-만노오스-4,6-데하이드라타아제 (GDP-D-mannose-4,6-dehydratase)가 발현되도록 형질전환되며, GDP-L-푸코오스 신타아제 (GDP-L-fucose synthase)가 발현되도록 형질전환되고, 락토오즈 퍼미아제 (lactose permease)가 발현되도록 형질전환되며, 베타-갈락토시다아제(beta-galactosidase)가 발현되도록 형질전환되고, 포스포만노뮤타아제 (Phosphomannomutase) 및 GTP-만노오스-1-포스페이트 구아닐트랜스퍼라아제 (GTP-mannose-1-phosphate guanylyltransferase)를 보유하고 있되,상기 베타-갈락토시다아제(beta-galactosidase)는 유도성 프로모터에 발현이 조절되는 것을 특징으로 하는 푸코실락토오스(fucosyllactose) 생산용 재조합 코리네박테리움 글루타미쿰 (Corynebacterium glutamicum).
- 제1항에 있어서,상기 유도성 프로모터에 의해 발현이 조절되는 베타-갈락토시다아제는,균체 밖으로 배출되는 것을 특징으로 하는 푸코실락토오스(fucosyllactose) 생산용 재조합 코리네박테리움 글루타미쿰 (Corynebacterium glutamicum).
- 제2항에 있어서,상기, 균체 밖으로 배출되는, 유도성 프로모터에 의해 발현이 조절되는 베타-갈락토시다아제는,균체 내에서 활성이 나타나지 않고, 균체 밖으로 배출된 후 활성이 나타나는 것을 특징으로 하는 푸코실락토오스(fucosyllactose) 생산용 재조합 코리네박테리움 글루타미쿰 (Corynebacterium glutamicum).
- 제1항에 있어서,상기 재조합 코리네박테리움 글루타미쿰(Corynebacterium glutamicum)은,포스포만노뮤타아제 (Phosphomannomutase)가 과발현되도록 형질전환되며, GTP-만노오스-1-포스페이트 구아닐트랜스퍼라아제 (GTP-mannose-1-phosphate guanylyltransferase)가 과발현되도록 형질전환된 것을 특징으로 하는 푸코실락토오스(fucosyllactose) 생산용 재조합 코리네박테리움 글루타미쿰 (Corynebacterium glutamicum).
- 락토오스가 첨가된 배지에, 제1항 또는 제2항의 재조합 코리네박테리움 글루타미쿰(Corynebacterium glutamicum)을 배양하는 것을 특징으로 하는 푸코실락토오스(fucosyllactose)의 생산방법.
- 제5항에 있어서,상기 배지는,글루코오스를 더 포함하고 있는 것을 특징으로 하는 푸코실락토오스(fucosyllactose)의 생산방법.
