Abstract
本発明は、ヒトの脳血流の低下抑制又は改善用組成物及び脳血流の低下抑制又は改善方法を提供することを目的とする。
本発明は、ケルセチン又はその配糖体を有効成分として含有する、ヒトの脳血流の低下抑制又は改善用組成物等に関する。
Description
本発明は、脳血流の低下抑制又は改善用組成物に関する。また、本発明は、脳血流の低下抑制又は改善方法等に関する。
加齢や認知症等の要因によって、脳血流が低下することが知られている。脳血流が低下すると、脳活動に必要な酸素及び栄養が充分に届かなくなり、脳循環障害に伴う症状の発生等につながる。
ケルセチンは、フラボノールの一種であり、そのままで、又は、配糖体として玉ねぎなどの植物に含有されている。特許文献1には、α-グルコシルイソクエルシトリンを有効成分として含有する血管内皮機能向上用組成物が記載されている。特許文献1の実施例には、酵素処理イソクエルシトリンを含む組成物の投与によって、上腕の血管径の増大率(FMD値)が改善されたことが記載されている。
しかしながら特許文献1では、ケルセチン又はその配糖体の脳血流への効果は検討されていない。ケルセチン又はその配糖体のヒトにおける脳血流への効果は、これまで報告されていない。
本発明は、ヒトの脳血流の低下抑制又は改善用組成物を提供することを目的とする。また、本発明は、脳血流の低下抑制又は改善方法を提供することを目的とする。
本発明者らは、上記課題を解決するべく鋭意検討した結果、ケルセチン又はその配糖体がヒトの脳血流の低下抑制又は改善作用を有することを見出した。
すなわち、以下に限定されるものではないが、本発明は、以下のヒトの脳血流の低下抑制又は改善用組成物等に関する。
〔1〕ケルセチン又はその配糖体を有効成分として含有する、ヒトの脳血流の低下抑制又は改善用組成物。
〔2〕1日当たりの摂取量中に、ケルセチン又はその配糖体をケルセチン換算で50~500mg含有する、上記〔1〕に記載の組成物。
〔3〕安静時の脳血流の低下を抑制又は脳血流を改善する、上記〔1〕又は〔2〕に記載の組成物。
〔4〕加齢、ストレス、栄養状態及び季節変動からなる群より選択される少なくとも1つに起因する脳血流の低下を抑制する、上記〔1〕~〔3〕のいずれかに記載の組成物。
〔5〕経口用組成物である上記〔1〕~〔4〕のいずれかに記載の組成物。
〔6〕飲食品である上記〔1〕~〔5〕のいずれかに記載の組成物。
〔7〕「脳の血流」、「脳血流」、「脳の血液循環」、「頭の血流」、「頭の血の流れ」、「頭の血の巡り」、「脳疲労」及び「頭のだるさ」からなる群より選択される少なくとも1の機能の表示を付した、上記〔1〕~〔6〕のいずれかに記載の組成物。

本発明によれば、ヒトの脳血流の低下抑制又は改善用組成物を提供することができる。また、本発明によれば、脳血流の低下抑制又は改善方法を提供することができる。
本発明のヒトの脳血流の低下抑制又は改善用組成物は、ケルセチン又はその配糖体を有効成分として含有する。本発明のヒトの脳血流の低下抑制又は改善用組成物を、以下では単に本発明の組成物ということもある。
本明細書において、「ケルセチン」とは、ビタミンPとも称され、ポリフェノールの一種であるフラボノールに属する化合物であるケルセチンを意味する。ケルセチンは、下記式(1)で示される化合物である。
本発明において、「ケルセチン配糖体」とは、上記ケルセチンの配糖体を意味し、具体的にはケルセチンの3位の水酸基に1以上の糖がグリコシド結合した一連の化合物の総称である。ケルセチン配糖体は、下記一般式(2)で示される化合物である。ケルセチン配糖体は、1種の化合物であってもよく、2種以上の化合物であってよい。
上記一般式中の(X)nは、糖鎖を表す。Xは糖(単糖)を表し、nは1以上の整数である。
ケルセチンにグリコシド結合するXで表される糖鎖を構成する糖は、例えば、グルコース、ラムノース、ガラクトース、グルクロン酸等であり、好ましくはグルコース、ラムノースである。また、nは1以上であれば、特に制限されないが、好ましくは1~16、より好ましくは1~8である。nが2以上であるとき、X部分は1種類の糖からなっていてもよく、複数種の糖からなっていてもよい。換言すると、nが2以上であるとき、(X)nは、1種類の糖からなる糖鎖であってもよく、複数種の糖からなる糖鎖であってもよい。本発明におけるケルセチン配糖体は、既存のケルセチン配糖体を、酵素などで処理して糖転移させたものも含む。本発明でいうケルセチン配糖体は、具体的には、ルチン、酵素処理ルチン(ルチンの酵素処理物)、クエルシトリン、イソクエルシトリンなどを含む。本発明において、ケルセチン配糖体として、酵素処理ルチンを使用することが、特に好ましい。酵素処理ルチンの好ましい例として、ルチンを酵素処理してラムノース糖鎖部分を除去したイソクエルシトリン、イソクエルシトリンを糖転移酵素で処理して得られるイソクエルシトリンにグルコース1~7個からなる糖鎖が結合したもの、及びその混合物を主成分とするものが挙げられる。一態様において、ケルセチン配糖体として、ケルセチンの3位の水酸基に1個以上(好ましくは、1~8個)のグルコースがグリコシド結合した化合物(例えば、イソクエルシトリン、イソクエルシトリンに1~7個のグルコースが結合した配糖体)等が好ましく、ケルセチンの3位の水酸基に1~7個のグルコースがグリコシド結合した化合物がより好ましい。
摂取されたケルセチン配糖体は、腸管内で糖が切断されてアグリコン(ケルセチン)になってから体内へ吸収される。
摂取されたケルセチン配糖体は、腸管内で糖が切断されてアグリコン(ケルセチン)になってから体内へ吸収される。
本発明において、ケルセチン又はその配糖体を得るための由来、製法については特に制限はない。例えば、ケルセチン又はその配糖体を多く含む植物として、ソバ、エンジュ、ケッパー、リンゴ、茶、タマネギ、ブドウ、ブロッコリー、モロヘイヤ、ラズベリー、コケモモ、クランベリー、オプンティア、葉菜類、柑橘類などが知られており、これらの植物からケルセチン又はその配糖体を得ることができる。
本発明においては、本発明の効果を奏することになる限り、上記の植物等の天然物由来の抽出物を、濃縮、精製等の操作によってケルセチン又はその配糖体の含量を高めたもの、例えば、ケルセチン又はその配糖体を含有する抽出物の、濃縮物又は精製物を用いることができる。濃縮方法又は精製方法は、既知の方法を採用することができる。本発明の一態様においては、精製又は単離されたケルセチン又はその配糖体を使用してもよい。ケルセチン又はその配糖体は、化学合成品を使用することもできる。
本発明においては、本発明の効果を奏することになる限り、上記の植物等の天然物由来の抽出物を、濃縮、精製等の操作によってケルセチン又はその配糖体の含量を高めたもの、例えば、ケルセチン又はその配糖体を含有する抽出物の、濃縮物又は精製物を用いることができる。濃縮方法又は精製方法は、既知の方法を採用することができる。本発明の一態様においては、精製又は単離されたケルセチン又はその配糖体を使用してもよい。ケルセチン又はその配糖体は、化学合成品を使用することもできる。
ケルセチン又はその配糖体は、天然物や飲食品に含まれ、食経験がある化合物である。このため安全性の観点から、ケルセチン又はその配糖体は、例えば毎日摂取することにも問題が少ないと考えられる。本発明によれば、安全性が高い物質を有効成分として含むヒトの脳血流の低下抑制又は改善用組成物を提供することができる。
本発明の組成物は、ヒトの脳血流の低下を抑制又は脳血流を改善するためのものである。
本発明において、脳血流の低下とは、脳全体又は脳の一部組織への血液量及び/又は血流量が低下(減少)することをいう。脳血流の低下抑制は、脳内の血液量及び/又は血流量の低下を抑制することを意味し、脳内の血液量及び/又は血流量を維持することを含む。脳血流の改善は、脳内の血液量及び/又は血流量を改善することを意味し、脳内の血液量及び/又は血流量を増加させること、低下した脳血液量及び/又は脳血流量を回復させること等を含む。回復は、一部を回復させることを含む。本発明の組成物は、ヒトの脳血液量及び/又は脳血流量の低下抑制又は改善のために使用することができる。
本発明において、脳血流の低下とは、脳全体又は脳の一部組織への血液量及び/又は血流量が低下(減少)することをいう。脳血流の低下抑制は、脳内の血液量及び/又は血流量の低下を抑制することを意味し、脳内の血液量及び/又は血流量を維持することを含む。脳血流の改善は、脳内の血液量及び/又は血流量を改善することを意味し、脳内の血液量及び/又は血流量を増加させること、低下した脳血液量及び/又は脳血流量を回復させること等を含む。回復は、一部を回復させることを含む。本発明の組成物は、ヒトの脳血液量及び/又は脳血流量の低下抑制又は改善のために使用することができる。
脳血流は、公知の方法で測定することができる。脳血流は、例えば、近赤外線分光法(NIRS)により測定することができる。NIRSは、非侵襲である、妥当性が高い、簡便である等の利点がある。NIRSの中でも、Time-resolved spectroscopy(TRS法)のNIRSは、定量性と再現性に優れている。TRS法のNIRSでは、照射光に短パルス光を用いることによって、透過光の時間応答波形を測定する。NIRSにより対象の脳血流を測定すると、総ヘモグロビン濃度を指標にして脳血液量を、酸素化ヘモグロビン濃度を指標にして単位時間当たりの脳血流量の変化を測定することができる。また、TRS法を用いることで安静時の脳血液量及び脳血流量を測定することができる。
後記の実施例に示されるように、ケルセチン又はその配糖体を摂取することによって、摂取しない場合と比較して、脳血流の低下を抑制又は脳血流を改善することができる。ケルセチン又はその配糖体は、脳血流の低下抑制又は改善のための有効成分として使用することができる。
ケルセチン又はその配糖体は、安静時の脳血流の低下を抑制又は脳血流を改善することができる。一態様において、本発明の組成物は、安静時の脳血流の低下を抑制又は脳血流を改善するために好ましく使用することができる。このため本発明の組成物は、例えば、高齢者、認知機能が低下した人、脳の疲れを感じる人、ストレスを感じる人等の脳血流の低下抑制又は改善のために用いることができる。安静時の脳血流の低下抑制又は改善によって、例えば、定常状態での脳の疲れを感じにくくなる。
ヒトの脳血液量及び脳血流量は、例えば、加齢、ストレス、栄養状態、季節変動等の要因によって低下する。一態様において、本発明の組成物は、加齢、ストレス、栄養状態及び季節変動からなる群より選択される少なくとも1つに起因する脳血流の低下を抑制するために好ましく使用することができる。一態様において、本発明の組成物は、加齢、ストレス及び季節変動からなる群より選択される少なくとも1つに起因する脳血流の低下を抑制するために使用することができる。一態様において、本発明の組成物は、安静時の脳血流の低下を抑制若しくは脳血流を改善するため、又は、加齢、ストレス、栄養状態及び季節変動からなる群より選択される少なくとも1つに起因する脳血流の低下を抑制するために好ましく使用することができる。
本発明の組成物は、例えば、脳血流の低下抑制又は改善が、その予防又は改善に有効である状態又は疾患の予防又は改善のために使用することができる。このような状態又は疾患として、脳血流の低下に起因する又は脳血流の低下を伴う状態又は疾患が挙げられる。上記状態又は疾患として、具体的には、例えば、脳梗塞、めまい、立ちくらみ、脳疲労等が挙げられる。一態様において、本発明の組成物は、例えば、上記の状態又は疾患を予防又は改善するために使用することができる。また、脳血流の低下抑制又は改善によって、例えば、認知機能の低下抑制又は改善、脳疲労の予防又は改善、脳活動の低下抑制、低下した脳活動の回復等の効果が得られることが期待できる。
状態又は疾患の予防は、状態又は疾患の発症を防止すること、発症を遅延させること、発症率を低下させること、発症のリスクを軽減すること等を包含する。状態又は疾患の改善は、対象を状態又は疾患から回復させること、状態又は疾患の症状を軽減すること、状態又は疾患の症状を好転させること、状態又は疾患の進行を遅延させること、防止すること等を包含する。
状態又は疾患の予防は、状態又は疾患の発症を防止すること、発症を遅延させること、発症率を低下させること、発症のリスクを軽減すること等を包含する。状態又は疾患の改善は、対象を状態又は疾患から回復させること、状態又は疾患の症状を軽減すること、状態又は疾患の症状を好転させること、状態又は疾患の進行を遅延させること、防止すること等を包含する。
本発明の組成物は、治療的用途又は非治療的用途のいずれにも適用することができる。非治療的とは、医療行為、すなわち人間の手術、治療又は診断を含まない概念である。
本発明の組成物は、飲食品、医薬品、医薬部外品、化粧料等の形態とすることができる。本発明の組成物は、それ自体が脳血流の低下抑制又は改善のために用いられる飲食品、医薬品、医薬部外品、化粧料等であってもよく、これらに配合して使用される素材又は製剤等であってもよい。
本発明の脳血流の低下抑制又は改善用組成物は、一例として、剤の形態で提供することができるが、本形態に限定されるものではない。当該剤をそのまま組成物として、又は、当該剤を含む組成物として提供することもできる。一態様において、脳血流の低下抑制又は改善用組成物は、脳血流の低下抑制又は改善剤ということもできる。
本発明の組成物は、経口用組成物、非経口用組成物のいずれであってもよいが、好ましくは、経口用組成物である。経口用組成物としては、飲食品、経口用医薬品、経口用医薬部外品が挙げられ、好ましくは飲食品又は経口用医薬部外品であり、より好ましくは飲食品である。非経口用組成物として、非経口用医薬品、非経口用医薬部外品、化粧料が挙げられる。好ましくは化粧料又は非経口用医薬部外品である。
本発明の組成物は、飲食品、医薬品、医薬部外品、化粧料等の形態とすることができる。本発明の組成物は、それ自体が脳血流の低下抑制又は改善のために用いられる飲食品、医薬品、医薬部外品、化粧料等であってもよく、これらに配合して使用される素材又は製剤等であってもよい。
本発明の脳血流の低下抑制又は改善用組成物は、一例として、剤の形態で提供することができるが、本形態に限定されるものではない。当該剤をそのまま組成物として、又は、当該剤を含む組成物として提供することもできる。一態様において、脳血流の低下抑制又は改善用組成物は、脳血流の低下抑制又は改善剤ということもできる。
本発明の組成物は、経口用組成物、非経口用組成物のいずれであってもよいが、好ましくは、経口用組成物である。経口用組成物としては、飲食品、経口用医薬品、経口用医薬部外品が挙げられ、好ましくは飲食品又は経口用医薬部外品であり、より好ましくは飲食品である。非経口用組成物として、非経口用医薬品、非経口用医薬部外品、化粧料が挙げられる。好ましくは化粧料又は非経口用医薬部外品である。
本発明の組成物は、本発明の効果を損なわない限り、ケルセチン又はその配糖体に加えて、任意の添加剤、任意の成分を含有することができる。これらの添加剤及び成分は、組成物の形態等に応じて選択することができ、一般的に飲食品、医薬品、医薬部外品、化粧料等に使用可能なものが使用できる。本発明の組成物を、飲食品、医薬品、医薬部外品、化粧料等とする場合、その製造方法は特に限定されず、一般的な方法により製造することができる。
例えば本発明の組成物を飲食品とする場合、ケルセチン又はその配糖体に、飲食品に使用可能な成分(例えば、食品素材、必要に応じて使用される食品添加物等)を配合して、種々の飲食品とすることができる。飲食品は特に限定されず、例えば、一般的な飲食品、健康食品、健康飲料、機能性表示食品、特定保健用食品、健康補助食品、病者用飲食品等が挙げられる。上記健康食品、機能性表示食品、特定保健用食品、健康補助食品、病者用飲食品等は、例えば、細粒剤、錠剤、顆粒剤、散剤、カプセル剤、チュアブル剤、ドライシロップ剤、シロップ剤、液剤、飲料、流動食等の各種製剤形態として使用することができる。
一態様において、本発明の組成物は、飲料の形態で提供されて良い。飲料は特に限定されないが、例えば、麦茶飲料、緑茶飲料、ブレンド茶、紅茶飲料、ウーロン茶飲料等の茶系飲料が好ましい。

本発明の組成物を医薬品又は医薬部外品とする場合、例えば、ケルセチン又はその配糖体に、薬理学的に許容される担体、必要に応じて添加される添加剤等を配合して、各種剤形の医薬品又は医薬部外品とすることができる。そのような担体、添加剤等は、医薬品又は医薬部外品に使用可能な、薬理学的に許容されるものであればよく、例えば、賦形剤、結合剤、崩壊剤、滑沢剤、抗酸化剤、着色剤等の1又は2以上が挙げられる。医薬品又は医薬部外品の投与(摂取)形態としては、経口又は非経口(経皮、経粘膜、経腸、注射等)投与の形態が挙げられる。本発明の組成物を医薬品又は医薬部外品とする場合、経口用(経口投与用)医薬品又は医薬部外品とすることが好ましい。経口投与のための剤形としては、液剤、錠剤、散剤、細粒剤、顆粒剤、糖衣錠、カプセル剤、懸濁液、乳剤、チュアブル剤等が挙げられる。非経口投与のための剤形として、注射剤、点滴剤、皮膚外用剤(貼付剤、クリーム剤、軟膏等)等が挙げられる。
本発明の組成物を化粧料とする場合、ケルセチン又はその配糖体に、化粧料に許容される担体、添加剤等を配合することができる。化粧料の製品形態は特に限定されない。
本発明の組成物に含まれるケルセチン又はその配糖体の含有量は特に限定されず、その形態等に応じて設定することができる。本発明の組成物中のケルセチン又はその配糖体の含有量は、例えば、ケルセチン換算で、0.0001重量%以上が好ましく、0.001重量%以上がより好ましく、0.01重量%以上がさらに好ましく、0.1重量%以上が特に好ましく、また、99.9重量%以下が好ましく、95重量%以下がより好ましく、80重量%以下がさらに好ましく、45重量%以下が特に好ましい。一態様において、ケルセチン又はその配糖体の含有量は、ケルセチン換算で組成物中に0.0001~99.9重量%が好ましく、0.001~95重量%がより好ましく、0.01~80重量%がさらに好ましく、0.01~45重量%が特に好ましく、0.1~45重量%が最も好ましい。また、一態様において、本発明の組成物を飲食品とする場合、ケルセチン又はその配糖体の含有量は、ケルセチン換算で、飲食品中に0.0001~99.9重量%が好ましく、0.001~95重量%がより好ましく、0.001~80重量%がさらに好ましく、0.001~45重量%がさらにより好ましく、0.01~45重量%が特に好ましい。ケルセチン又はその配糖体の含有量は、公知の方法に従って測定することができ、例えば、高速液体クロマトグラフィー(HPLC)法などを用いることができる。
本明細書中、ケルセチン換算の量又はこれに類する表現は、ケルセチンの場合はその量を意味し、ケルセチン配糖体の場合は、当該配糖体のモル数に、ケルセチンの分子量を乗じて得られる値を意味する。

本発明の組成物は、通常、対象に摂取又は投与されるものである。本発明の組成物の投与経路は特に限定されず、その形態に応じた適当な方法で摂取又は投与することができる。一態様において、本発明の組成物は、経口で摂取(経口投与)されることが好ましい。本発明の組成物の投与量(摂取量ということもできる)は特に限定されず、脳血流の低下抑制又は改善効果が得られるような量であればよく、投与形態、投与方法、対象の体重などに応じて適宜設定すればよい。
一態様において、ケルセチン又はその配糖体の投与量は、ケルセチン換算で、1日当たり、好ましくは0.1mg以上、より好ましくは0.3mg以上、さらに好ましくは1.0mg以上、さらにより好ましくは10mg以上、さらにより好ましくは50mg以上、特に好ましくは55mg以上、最も好ましくは60mg以上であり、また、好ましくは8000mg以下、より好ましくは4000mg以下、さらに好ましくは1000mg以下、さらにより好ましくは500mg以下、特に好ましくは200mg以下、最も好ましくは78mg以下である。一態様において、ケルセチン又はその配糖体の投与量は、ケルセチン換算で、1日当たり、0.1~8000mgが好ましく、0.3~4000mgがより好ましく、1.0~1000mgがさらに好ましく、10~500mgがさらにより好ましく、10~200mgが特に好ましく、10~78mgが最も好ましい。また、一態様において、ケルセチン又はその配糖体の投与量は、ケルセチン換算で、1日当たり、50~500mgが好ましく、55~500mgがより好ましく、55~200mgがさらに好ましく、55~78mgがさらにより好ましく、60~78mgが特に好ましい。
一態様においては、上記量を、例えば、1日1回で、又は、数回(例えば2~3回)に分けて、摂取させる又は投与することが好ましい。一態様においては、上記量のケルセチン又はその配糖体を、ヒトに経口で摂取させる又は投与することが好ましい。上記のケルセチン又はその配糖体の投与量は、体重60kgあたりの投与量であってよい。本発明の一態様において、本発明の組成物は、ヒトに、1日あたり上記量のケルセチン又はその配糖体を摂取させる又は投与するために使用することができる。本発明の一態様において、本発明の組成物は、ヒトに、体重60kgあたり、1日あたり上記量のケルセチン又はその配糖体を摂取させる又は投与するための経口用組成物であってよい。上記投与量は、好ましくは成人1日当たりの投与量である。
一態様においては、上記量を、例えば、1日1回で、又は、数回(例えば2~3回)に分けて、摂取させる又は投与することが好ましい。一態様においては、上記量のケルセチン又はその配糖体を、ヒトに経口で摂取させる又は投与することが好ましい。上記のケルセチン又はその配糖体の投与量は、体重60kgあたりの投与量であってよい。本発明の一態様において、本発明の組成物は、ヒトに、1日あたり上記量のケルセチン又はその配糖体を摂取させる又は投与するために使用することができる。本発明の一態様において、本発明の組成物は、ヒトに、体重60kgあたり、1日あたり上記量のケルセチン又はその配糖体を摂取させる又は投与するための経口用組成物であってよい。上記投与量は、好ましくは成人1日当たりの投与量である。
一態様において、本発明の組成物は、ヒトの1日当たりの摂取量中に、ケルセチン又はその配糖体をケルセチン換算で0.1~8000mg含有することが好ましい。組成物の1日当たりの摂取量中のケルセチン又はその配糖体の含有量は、ケルセチン換算で0.3~4000mgがより好ましく、1.0~1000mgがさらに好ましく、10~500mgがさらにより好ましく、10~200mgが特に好ましく、10~78mgが最も好ましい。また、一態様において、本発明の組成物は、1日当たりの摂取量中に、ケルセチン又はその配糖体をケルセチン換算で、好ましくは50~500mg、より好ましくは55~500mg、さらに好ましくは55~200mg、さらにより好ましくは55~78mg、特に好ましくは60~78mg含有することが好ましい。上記のケルセチン又はその配糖体の含有量は、成人の1日摂取量当たりの含有量であることが好ましい。
本発明の組成物は、継続して摂取又は投与されるものであることが好ましい。ケルセチン又はその配糖体を継続して投与又は摂取することによって、より優れた脳血流の低下抑制又は改善効果が発揮されることが期待される。一態様において、本発明の組成物を、好ましくは1週間以上、より好ましくは4週間以上、さらに好ましくは8週間以上継続して摂取又は投与することが好ましい。
本発明の組成物を摂取させる又は投与する対象(投与対象ということもできる)は、ヒトである。本発明の組成物を摂取させる又は投与する対象として、脳血流の低下抑制又は改善を必要とする又は希望する対象が挙げられる。また、投与対象として、例えば、脳血流の低下に起因する又は脳血流の低下を伴う状態又は疾患(例えば、脳梗塞、めまい、立ちくらみ、脳疲労等)の予防又は改善を必要とする又は希望するヒト等が挙げられる。一態様において、投与対象として、脳血流の低下に起因する又は脳血流の低下を伴う状態又は疾患を有する対象が挙げられる。
本発明の組成物は、幅広い年齢の対象に使用することができる。好ましくは、本発明の組成物は、成人(例えば18歳以上)に投与される。一態様において、本発明における投与対象として、中高年者が好ましい。中高年者は、高齢者を含む。中高年者の中でも、対象として高齢者が好ましい。本発明において、中高年者は、例えば、40歳以上のヒトであってよい。高齢者は、例えば、60歳以上のヒトであってよい。一態様において、本発明の組成物は、中高年者用の脳血流の低下抑制又は改善用組成物として好適に使用される。一態様において、本発明の組成物の投与対象として、例えば、40~80歳のヒトが好ましい。本発明の組成物の投与対象として、例えば、60歳以上のヒトがより好ましく、60歳以上80歳未満のヒトがさらに好ましく、60歳以上75歳未満のヒトが特に好ましい。
一態様において、本発明の組成物の投与対象は、健常者であってよく、健常な中高年者が好ましい。本発明の組成物は、健常者の脳血流の低下を抑制又は脳血流を改善するため、脳血流低下に起因する又は脳血流の低下を伴う状態又は疾患を予防するために使用することができる。本発明において、健常者とは、認知症、アルツハイマー病、精神疾患、脳血管疾患に罹患しておらず、かつ、脳神経系疾患、精神疾患の既往がない者をいう。本発明においては、認知症、アルツハイマー病、精神疾患、脳血管疾患により投薬を受けている者、及び、脳神経系疾患、精神疾患の既往がある者、あるいはそれらの疾患の疑いがある者は健常者に含まれない。健常者は、日常生活行動に支障のないヒトであることが好ましい。

本発明の組成物には、脳血流の低下抑制又は改善により発揮される機能の表示が付されていてもよい。本発明の組成物には、例えば、「脳の血流」、「脳血流」、「脳の血液循環」、「頭の血流」、「頭の血の流れ」、「頭の血の巡り」、「脳疲労」、「頭のだるさ」等の1又は2以上の機能の表示が付されていてもよい。
上記機能の表示には、本発明の組成物によって上記の機能が改善されることが記載されていてもよい。
一態様において、本発明の組成物は、上記の表示が付された飲食品であることが好ましい。また上記の表示は、上記の機能を得るために用いる旨の表示であってもよい。当該表示は、組成物自体に付されてもよいし、組成物の容器又は包装に付されていてもよい。
上記機能の表示には、本発明の組成物によって上記の機能が改善されることが記載されていてもよい。
一態様において、本発明の組成物は、上記の表示が付された飲食品であることが好ましい。また上記の表示は、上記の機能を得るために用いる旨の表示であってもよい。当該表示は、組成物自体に付されてもよいし、組成物の容器又は包装に付されていてもよい。
本発明は、以下の方法及び使用も包含する。
ケルセチン又はその配糖体をヒトに摂取させる又は投与する、脳血流の低下抑制又は改善方法。

上記の方法及び使用において、ケルセチン又はその配糖体、その好ましい態様等は、上述した本発明の脳血流の低下抑制又は改善用組成物と同じである。上記方法及び使用においては、1日に1回以上、例えば、1日1回~数回(例えば2~3回)、ケルセチン又はその配糖体を対象に摂取させる又は投与することが好ましい。一態様において、ケルセチン又はその配糖体は、脳血流の低下抑制又は改善によって、例えば、脳梗塞、めまい、立ちくらみ、脳疲労等の状態又は疾患を予防又は改善するために使用することができる。
上記方法及び使用においては、脳血流の低下抑制又は改善効果が得られる量(有効量ということもできる)のケルセチン又はその配糖体を使用すればよい。ケルセチン又はその配糖体の好ましい投与量や投与対象等は上述した本発明の脳血流の低下抑制又は改善用組成物と同じである。ケルセチン又はその配糖体は、そのまま摂取又は投与してもよく、これを含む組成物として摂取又は投与してもよい。例えば、本発明の組成物を摂取又は投与してもよい。
ケルセチン又はその配糖体は、ヒトの脳血流の低下抑制又は改善のために使用される飲食品、医薬品、医薬部外品等の製造のために使用することができる。一態様において、本発明は、ヒトの脳血流の低下抑制又は改善用組成物を製造するための、ケルセチン又はその配糖体の使用も包含する。

以下、本発明を実施例によりさらに詳しく説明するが、これにより本発明の範囲を限定するものではない。
<実施例1>
ケルセチン配糖体の脳血流への効果を評価した。脳血流の測定は、短パルス光を用いることによって、透過光の時間応答波形を測定するTime-resolved spectroscopy(TRS法)の近赤外線分光法(NIRS)により行った。

ケルセチン配糖体には、サンエミック(商品名、三栄源エフ・エフ・アイ株式会社)を使用した。サンエミックは、ケルセチンに1~7個のグルコースが付加した2種以上のケルセチン配糖体の混合物である。被験食品には、ケルセチン配糖体をイソクエルシトリン(ケルセチンに1個のグルコースが付加したケルセチン配糖体)として110mg(ケルセチン換算で71.6mg)含有する500mL麦茶飲料を用いた。対照食品は、ケルセチン配糖体を含有しない500mL麦茶飲料とした。

被験者を被験食品群又は対照食品群の2群に割り付けた。被験食品群(40名)と対照食品群(40名)からなるプラセボ対照ランダム化二重盲検並行群間比較試験を実施した。被験食品群の被験者は、被験食品を1日500mLずつ、40週間継続摂取した。対照食品群の被験者は、対照食品を1日500mLずつ、40週間継続摂取した。
被験食品又は対照食品を摂取開始前に、被験者の脳血流測定を実施した(摂取前検査)。摂取前検査時の測定値を、Pre値とした。被験食品又は対照食品の40週間の摂取が終了した時点で、再度被験者の脳血流測定を実施した(40週目検査)。40週目検査時の測定値を、40W値とした。
被験者を被験食品群又は対照食品群の2群に割り付けた。被験食品群(40名)と対照食品群(40名)からなるプラセボ対照ランダム化二重盲検並行群間比較試験を実施した。被験食品群の被験者は、被験食品を1日500mLずつ、40週間継続摂取した。対照食品群の被験者は、対照食品を1日500mLずつ、40週間継続摂取した。
被験食品又は対照食品を摂取開始前に、被験者の脳血流測定を実施した(摂取前検査)。摂取前検査時の測定値を、Pre値とした。被験食品又は対照食品の40週間の摂取が終了した時点で、再度被験者の脳血流測定を実施した(40週目検査)。40週目検査時の測定値を、40W値とした。
(脳血流測定)
安静時の前頭葉の脳血流変化を、NIRSにより測定した。脳血流の測定には、非侵襲性脳酸素モニターtNIRS-1(浜松ホトニクス株式会社)を用いた。この装置は、照射光に短パルス光を用いることによって、透過光の時間応答波形を測定するため、定量性と再現性に優れている。tNIRS-1のディスポーザブルパット(パットと記載することもある)を被験者の額に貼り付け、脳血液量及び脳血流の測定を行った。
図3Aは、脳血流測定のためにパットを張り付けた部位を示す模式図である。図3Bは、パットとプローブの接続位置を示す模式図である。図3Aに示すように、パット1、パット2を額の上矢状静脈洞3、髪4、眉毛5、血腫のある部分を避け、左右それぞれに貼り付けた。図3Bに示すように、パット1に、照射プローブ6a(外側)と検出プローブ7a(内側)を繋いだ。パット2に、照射プローブ6b(外側)と検出プローブ7b(内側)を繋いだ。額の左側に貼り付けたパット(図3Aのパット1)をチャンネル1(CH1)、額の右側に貼り付けたパット(図3Aのパット2)をチャンネル2(CH2)とした。

額面積とパッドの大きさから、貼り付け位置の選択余地は無く、毎回ほぼ同じ場所に貼り付けて測定を行った。頭蓋から3cmの深さ、脳実質表面から1cm程度の深さを中心とした強湾バナナ状の部位が測定される。また、本製品は、貼り付け位置や測定回ごとのばらつきが1-2%程度で従来のModified Beer-Lambert(MBL)法のNIRSと比べて非常に小さく、再現性に優れていることが確認されている。
半座位で5分間安静後、安静状態で1分間脳血流を測定した。サンプルタイムは5秒間とし、5秒間の移動平均値として記録される12個の測定値のうち、後半の10個の測定値について平均値を算出し、評価に用いた。評価した指標は、総ヘモグロビン(tHb)濃度、酸素化ヘモグロビン(O2Hb)濃度である。tHb濃度は脳血液量(CBV)、O2Hb濃度は脳血流量(CBF)及び脳活動を反映する指標である。
半座位で5分間安静後、安静状態で1分間脳血流を測定した。サンプルタイムは5秒間とし、5秒間の移動平均値として記録される12個の測定値のうち、後半の10個の測定値について平均値を算出し、評価に用いた。評価した指標は、総ヘモグロビン(tHb)濃度、酸素化ヘモグロビン(O2Hb)濃度である。tHb濃度は脳血液量(CBV)、O2Hb濃度は脳血流量(CBF)及び脳活動を反映する指標である。
(評価と解析)
NIRSによる脳血流測定については、左右差が認められた。右前頭葉の血流はストレス状態で増加することが知られており、ストレスの影響を反映したと考えられたため、測定部位が額の左側(左前頭葉)の測定結果を用いて評価を行った。
被験食品又は対照食品を40週間摂取後のtHb濃度及びO2Hb濃度の変化量(Pre値からの変化量)を求めた。具体的には、被験者ごとに、40週目検査のtHb濃度の測定値(40W値)から、摂取前検査のtHb濃度の測定値(Pre値)を引いて(40W値-Pre値)、tHb濃度の変化量(Δ値)(μmol/L)を算出した。これを、摂取40週のtHb濃度変化量(ΔtHb)とした。被験食品群の被験者の上記Δ値の平均値を、被験食品群の摂取40週のtHb濃度変化量(ΔtHb)とした。対照食品群の被験者の上記Δ値の平均値を、対照食品群の摂取40週のtHb濃度変化量(ΔtHb)とした。O2Hb濃度についても、tHb濃度と同じ方法で、被験食品群及び対照食品群について、摂取40週のO2Hb濃度変化量(ΔO2Hb)を求めた。
摂取前検査時の変化量(Δ値)は0とした。
脱落者や解析除外対象者を除いて、解析は60歳以上75歳未満の68名のデータで行った。内訳は、被験食品群が33名、対照食品群が35名であった。対応のあるt検定により、群内比較を行った(*:p<0.05、**:p<0.01、Pre値に対して)。有意差水準は、5%とした(*:p<0.05)。データ集計及び統計解析には、Microsoft Excel 2013、JMP ver.14.0.0(SAS Institute Inc., Cary, NC, USA)を用いた。
NIRSによる脳血流測定については、左右差が認められた。右前頭葉の血流はストレス状態で増加することが知られており、ストレスの影響を反映したと考えられたため、測定部位が額の左側(左前頭葉)の測定結果を用いて評価を行った。
被験食品又は対照食品を40週間摂取後のtHb濃度及びO2Hb濃度の変化量(Pre値からの変化量)を求めた。具体的には、被験者ごとに、40週目検査のtHb濃度の測定値(40W値)から、摂取前検査のtHb濃度の測定値(Pre値)を引いて(40W値-Pre値)、tHb濃度の変化量(Δ値)(μmol/L)を算出した。これを、摂取40週のtHb濃度変化量(ΔtHb)とした。被験食品群の被験者の上記Δ値の平均値を、被験食品群の摂取40週のtHb濃度変化量(ΔtHb)とした。対照食品群の被験者の上記Δ値の平均値を、対照食品群の摂取40週のtHb濃度変化量(ΔtHb)とした。O2Hb濃度についても、tHb濃度と同じ方法で、被験食品群及び対照食品群について、摂取40週のO2Hb濃度変化量(ΔO2Hb)を求めた。
摂取前検査時の変化量(Δ値)は0とした。

図1Aは、被験食品群及び対照食品群の総ヘモグロビン(tHb)濃度の実測値(μmol/L)を示すグラフであり、図1Bは、総ヘモグロビン濃度の変化量(ΔtHb)を示すグラフである。図2Aは、被験食品群及び対照食品群の酸素化ヘモグロビン濃度(O2Hb)の実測値(μmol/L)を示すグラフであり、図2Bは、酸素化ヘモグロビン濃度の変化量(ΔO2Hb)を示すグラフである。図1B及び図2Bに示す変化量は、いずれもPre値からの変化量である。図1A、図1B、図2A及び図2B中、黒四角は被験食品群、白四角は対照食品群である。横軸の「Pre」は摂取前検査を、「40W」は、40週目検査を示す。図1A、図1B、図2A及び図2Bにおいて、結果は、平均値±標準偏差で示した(**:p<0.01、Pre値に対して)。
結果を、図1A、図1B、図2A及び図2Bに示す。図1A、図1B、図2A及び図2Bは、いずれも額の左側の測定結果である。
図1Aは、被験食品群及び対照食品群の総ヘモグロビン(tHb)濃度の実測値(μmol/L)を示すグラフであり、図1Bは、総ヘモグロビン濃度の変化量(ΔtHb)を示すグラフである。図2Aは、被験食品群及び対照食品群の酸素化ヘモグロビン濃度(O2Hb)の実測値(μmol/L)を示すグラフであり、図2Bは、酸素化ヘモグロビン濃度の変化量(ΔO2Hb)を示すグラフである。図1B及び図2Bに示す変化量は、いずれもPre値からの変化量である。図1A、図1B、図2A及び図2B中、黒四角は被験食品群、白四角は対照食品群である。横軸の「Pre」は摂取前検査を、「40W」は、40週目検査を示す。図1A、図1B、図2A及び図2Bにおいて、結果は、平均値±標準偏差で示した(**:p<0.01、Pre値に対して)。
左前頭葉のtHb濃度とO2Hb濃度は、摂取前検査から40週目検査にかけて対照食品群で有意に低下した。一方、被験食品群では、tHb濃度及びO2Hb濃度の低下は抑制された(図1A、図1B、図2A及び図2B)。対照食品群でtHb濃度とO2Hb濃度が低下した理由として、ストレス状態、加齢の影響、栄養状態、季節変動等が考えられた。ストレスの原因として、コロナ禍によるストレスが考えられた。一方で、被験食品群では、脳血液量及び脳血流量が増加し、その結果として、tHb濃度(図1A及び図1B)、及び、O2Hb濃度(図2A及び図2B)の低下が抑制されたと考えられる。以上から、加齢やストレス、栄養状態、季節変動等の要因によって脳血液量及び脳血流量は低下するが、ケルセチン配糖体摂取によって当該低下が抑制されたと考えられた。上記の結果から、ケルセチン又はその配糖体は、脳血流の低下抑制又は改善作用を有することが分かった。

Claims (9)
- ケルセチン又はその配糖体を有効成分として含有する、ヒトの脳血流の低下抑制又は改善用組成物。
- 1日当たりの摂取量中に、ケルセチン又はその配糖体をケルセチン換算で50~500mg含有する、請求項1に記載の組成物。
- 安静時の脳血流の低下を抑制又は脳血流を改善する、請求項1又は2に記載の組成物。
- 加齢、ストレス、栄養状態及び季節変動からなる群より選択される少なくとも1つに起因する脳血流の低下を抑制する、請求項1又は2に記載の組成物。
- 経口用組成物である請求項1又は2に記載の組成物。
- 飲食品である請求項1又は2に記載の組成物。
- 「脳の血流」、「脳血流」、「脳の血液循環」、「頭の血流」、「頭の血の流れ」、「頭の血の巡り」、「脳疲労」及び「頭のだるさ」からなる群より選択される少なくとも1の機能の表示を付した、請求項1又は2に記載の組成物。
- ケルセチン又はその配糖体をヒトに摂取させる又は投与する、脳血流の低下抑制又は改善方法。
- ヒトの脳血流の低下を抑制又はヒトの脳血流を改善するための、ケルセチン又はその配糖体の使用。
