- the resolution of MIP imaging reaches 300nm level, and a beam of short-wavelength visible light is used as the detection light to detect the photothermal effect generated by molecular vibration absorption, thus breaking through the diffraction limit of conventional infrared imaging by an order of magnitude.

- the probe light with a wavelength of 405nm emitted by the detection light source serves as a balance between diffraction-limited resolution and photodamage of high photon energy, and due to its shorter wavelength, it increases scattering, increases the number of photons in the back-detection mode, and further improves the signal-to-noise ratio.
- the focus of the pump beam and the probe beam are optimized by using a Cassegrain objective and a high numerical aperture objective, respectively, for counter-propagation.
- the photothermally modulated detection beam can be collected by the forward or back-detection mode of a high-bandwidth photodiode.
- a pinhole is placed confocally on the back-detection path.
- the photothermal relaxation of the probe beam at the center and edge of a single 2 ⁇ m PMMA microsphere was also measured in the embodiment, as shown in Figure 6 (b)-(d).
- the relaxation at both positions consists of a heating and a dissipation process: one is a fast dissipation on a time scale of tens of nanoseconds, and the other is a long dissipation on a microsecond scale.
- the fast dissipation part is mainly due to photoacoustic transmission, with a 16% drop in intensity at the center of the microsphere and a 68% drop at the edge, which accounts for the difference in photothermal conversion efficiency.
- the other part is a slower exponential decay relaxation, with relaxation times of 3.8 ⁇ s and 6.7 ⁇ s at the center and edge, respectively.
- Photothermal relaxation can be described as the Fourier series of various harmonic components shown in the FFT waveform, as shown in Figure 6 (c), and the intensity ratio between the center and the edge increases at high-order harmonics, as shown in Figure 6 (d). This is consistent with the simulation results.

- the examples use super-resolution infrared imaging of mammalian cells to demonstrate label-free organelle tracing.
- the examples found that a large amount of LD accumulated in the cell body, as shown in Figure 7 (a) and (c). Most of the protein signals were distributed in the cytoplasm and nucleus, showing that the chondrocytes were spindle-shaped. Some protein droplets were also observed, as shown in Figure 7(b).
- This example also provides V-PEARL super-resolution imaging of yeast cells.
- Yeast is a powerful model organism for studying cell biology, especially lipid metabolism. However, with individual yeast cells being approximately 2-4 ⁇ m in size, a major limitation is the lack of tools to visualize tiny LDs (typically 0.05-0.5 ⁇ m).
- the example uses a V-PEARL microscope for the first time to perform subcellular far-field infrared imaging of yeast cells. Yeast cells contain sparse LDs that are not obvious in the reflectance image, as shown in FIG8(a).
- a single LD is visible at 1750 cm -1 , while a protein signal is found in the amide I band at 1650 cm -1 , as shown in FIG8(b) and (c).
- the example also measures the size distribution of intracellular LDs, as shown in FIG8(d), showing a clear difference in the histogram of LD size between the 1st and 4th harmonic demodulations.
- the average size of these LDs is 197 ⁇ 19 nm at 1st order demodulation (conventional MIP) and 185 ⁇ 16 nm at 4th order demodulation.
- V-PEARL showed richer spatial frequency components, especially at higher spatial frequencies, which represents higher spatial resolution, as shown in Figure 8 (e).
- V-PEARL imaging found subtle spatial separation structures between small LDs and a pair of protein droplets (each about 170nm), as shown in Figure 8 (i).
- the resolution of this method is improved by 43.5 times.
- the line profile and Gaussian fitting show a trend of improved resolution at the 2nd and 3rd harmonics.
- only a single peak with a slight shoulder was found in the conventional MIP, as shown in Figure 8 (j).
- the smallest feature observed in the V-PEARL imaging of yeast is ⁇ 86nm, which is the first time that features below 100nm have been resolved using far-field infrared imaging, as shown in Figure 8 (k) and (m).
