Abstract
본 발명은 본 발명은 카테킨 하이드레이트를 유효성분으로 포함하는 만성폐쇄성폐질환 예방, 개선 또는 치료용 조성물에 관한 것으로, 본 발명의 카테킨 하이드레이트(catechin hydrate)는 미세먼지로 인한 인간기관지상피세포의 세포사멸을 유의하게 억제하고, 미토콘드리아 기능을 회복시키며, 폐 손상을 억제시키는 효과가 있어 폐기종(emphysema), 만성기관지염(chronic bronchitis), 천식(asthma) 또는 폐렴(Pneumonia)과 같은 만성폐쇄성폐질환의 예방, 개선 또는 치료용 조성물로 유용하게 이용될 수 있다.
Description
본 발명은 카테킨 하이드레이트를 유효성분으로 포함하는 만성폐쇄성폐질환 예방 또는 치료용 약학적 조성물에 관한 것이다.
만성폐쇄성폐질환(Chronic obstructive pulmonary disease, COPD)은 세계보건기구(WHO) 보고에 따르면 전 세계 주요 사망원인 4위의 질환으로 10년 후엔 3위로 상승할 것으로 예상되는 심각한 폐 질환이다. 환경오염 증가와 산업발달에 따른 다양한 위험물질에 대한 노출 증가가 주요 원인이며, 특히 흡연이 주된 원인으로 규정되는 난치성 만성질환이다. 특히 COPD는 65세 이상의 고령인구에서 발병률이 급증하는데 노화도 발병의 주요 위험인자이다. 하지만 진단율 자체가 매우 낮고 뚜렷한 치료제가 없는 실정이어서 다른 심각한 만성질환들의 사망률이 감소하는 것에 반해 COPD의 사망률은 증가하는 추세이다. 따라서 COPD를 치료할 수 있는 치료제 개발에 대한 연구가 전 세계적으로 활발하게 진행되고 있다.
COPD는 흡연, 대기오염, 화학물질, 직업성 인자, 유전적 소인 등 다양한 원인에 의해서 발병하는데, 이중 흡연이 주요한 원인으로 지목되고 있으며, 실제 COPD 환자의 80% 이상이 흡연자로 밝혀진 바 있다(Biol Pharm Bull, 2012, 35:1752-1760). COPD의 발병 기전에는 기관지와 폐 조직 등에서의 만성 염증, 폐에서의 단백분해효소 활성화, 산화적 스트레스(oxidative stress) 등이 관여하며, 염증에 관여하는 세포는 주로 호중구와 대식세포, T림프구 등이다. 이러한 염증세포들은 TNF-α, IFN-γ, IL-1β, IL-6, IL-8, IL-18 등의 다양한 염증성 사이토카인, 조직 손상을 야기하는 여러 단백분해효소들을 생성한다(대한결핵 및 호흡기학회 호흡기학 서울:군자출판사; 2007, p 301-5;Am J Respir Crit Care Med, 1997 155, 1441-1447; Am J Physiol Lung CellMol Physiol, 2010, 298:L262-L269). 담배연기로 유발된 COPD 동물 모델에서는 호중구의 유입, KC(keratinocyte chemoattractant), TNF-α(tumour necrosis factorα), MIP-2(macropage inflammatory protein 2), MIP-1α 및 MCP-1(monocyte chemoattractant protein), MMP12(matrix metalloproteinase 12)와 GM-CSF(granulocyte macrophage colony-stimulating factor)이 증가함이 관찰되었다(Clin. Sci. 2014, 126(3):207).
COPD의 대표적인 임상 증상은 기관지가 좁아지면서 호흡이 힘들어진다. COPD는 매우 천천히 진행되기 때문에 상태가 극도로 악화될 때까지 진단이나 치료를 받지 않은 환자가 많아 치료가 어려운 질환이다. COPD를 구성하는 주요 임상 질환은 폐기종(emphysema)과 만성 기관지염(chronic bronchitis)이 있으며, 장기간 가래 및 기침 증상을 나타내는 만성 기관지염 환자들의 대부분은 COPD로 최종 진단 받을 확률이 높다. 반면 폐기종은 폐의 폐포(alveoli) 구조가 파괴되어 산소와 이산화탄소의 정상적인 교환이 일어나지 못하게 된다.
COPD 치료를 위한 약물 요법으로는 흡입제, 먹는 약, 주사제, 호흡재활 치료, 산소 요법이 있으며, 외과적 치료로서, 중증COPD 환자에게 폐 이식 또는 폐 용적 축소술을 시행하기도 하는데, 최근 식물 등의 생약을 이용한 호흡기 질환 예방, 치료용 성분에 대한 다양한 연구가 진행되고 있는 실정이다.
이에 본 발명자들은 카테킨 하이드레이트(catechin hydrate)가 만성폐쇄성 폐질환의 마커인 세포자멸사 및 미토콘드리아 기능 장애의 대표적인 인자들의 유도를 유의미하게 억제하는 것을 확인하고 본 발명을 완성하였다.
본 발명의 목적은 카테킨 하이드레이트(catechin hydrate) 또는 이의 약학적으로 허용가능한 염을 유효성분으로 포함하는 만성폐쇄성폐질환 예방 또는 치료용 약학적 조성물을 제공하는 데 있다.
또한, 본 발명의 다른 목적은 카테킨 하이드레이트(catechin hydrate) 또는 이의 식품학적으로 허용가능한 염을 유효성분으로 포함하는 만성폐쇄성폐질환 예방 또는 개선용 식품 조성물을 제공하는 데 있다.
또한, 본 발명의 다른 목적은 카테킨 하이드레이트(catechin hydrate) 또는 이의 식품학적으로 허용가능한 염을 유효성분으로 포함하는 호흡기 건강 개선을 위한 건강기능식품 조성물을 제공하는 데 있다.
또한, 본 발명의 다른 목적은 상기 약학적 조성물을 개체에 투여하는 단계를 포함하는 만성폐쇄성폐질환 예방 또는 치료방법을 제공하는 데 있다.
상기와 같은 목적을 달성하기 위해, 본 발명은 카테킨 하이드레이트(catechin hydrate) 또는 이의 약학적으로 허용가능한 염을 유효성분으로 포함하는 만성폐쇄성폐질환 예방 또는 치료용 약학적 조성물을 제공한다.
이어서, 본 발명은 카테킨 하이드레이트(catechin hydrate) 또는 이의 식품학적으로 허용가능한 염을 유효성분으로 포함하는 만성폐쇄성폐질환 예방 또는 개선용 식품 조성물을 제공한다.
나아가, 본 발명은 카테킨 하이드레이트(catechin hydrate) 또는 이의 식품학적으로 허용가능한 염을 유효성분으로 포함하는 호흡기 건강 개선을 위한 건강기능식품 조성물을 제공한다.

본 발명의 카테킨 하이드레이트(catechin hydrate)는 미세먼지로 인한 인간기관지상피세포의 세포사멸을 유의하게 억제하고, 미토콘드리아 기능을 회복시키며, 폐 손상을 억제시키는 효과가 있어 폐기종(emphysema), 만성기관지염(chronic bronchitis), 천식(asthma) 또는 폐렴(Pneumonia)과 같은 만성폐쇄성폐질환의 예방, 개선 또는 치료용 조성물로 유용하게 이용될 수 있다.

본 발명에서 사용되는 용어 “카테킨 하이드레이트(catechin hydrate)”은 식물의 잎과 껍질에서 발견되는 천연 화합물이다. 상기 카테킨 하이드레이트의 분자식은 C15H14O6·XH2O이고, IUPAC 이름은 (2R,3S)-2-(3,4-dihydroxyphenyl)-3,4-dihydro-2H-chromene-3,5,7-triol;hydrate이며, D-(+)-Catechin, Catechuic Acid, Cyanidol 등의 동의어를 가지고, 290.3 g/mol의 분자량을 가진다.
본 발명의 일 실시예에 있어서, 상기 카테킨 하이드레이트(catechin hydrate)은 하기 화학식 1로 표시되는 것일 수 있으나, 이에 제한되는 것은 아니다.

본 발명의 일 실시예에 있어서, 상기 카테킨 하이드레이트(catechin hydrate)는 25 내지 75μM 농도로 포함되는 것일 수 있으며, 바람직하게는 40 내지 60μM 농도로 포함되는 것일 수 있고, 더욱 바람직하게는 45 내지 55μM 농도로 포함되는 것일 수 있으나, 이에 제한되는 것은 아니다.
본 발명의 일 실시예에 있어서, 상기 카테킨 하이드레이트(catechin hydrate)는 2 내지 15 mg/kg의 농도로 투여되는 것일 수 있으며, 바람직하게는 8 내지 12 mg/kg 농도로 투여되는 것일 수 있으나, 이에 제한되는 것은 아니다.
본 발명의 일 실시예에 있어서, 상기 카테킨 하이드레이트는 기관지상피세포의 세포사멸을 억제하는 것일 수 있으나, 이에 제한되는 것은 아니다.

본 발명에서 사용되는 용어, “미세먼지(Particulate Matter, PM)”는 분진(粉塵))는 아황산가스, 질소 산화물, 중금속, 오존 및 일산화 탄소 등의 유해 물질을 포함하는 대기오염 물질로서 자동차 및 공장 등에서 발생하며, 대기 중 장기간 떠다니는 물질을 의미하며, 입경 10μm 이하의 경우 PM10, 입경 5μm 이하의 경우 PM5, 입경 2.5μm 이하의 경우 PM2.5, 입경 1μm 이하의 경우 PM1로 불리고 있다.
본 발명의 일 실시예에 있어서, 상기 만성폐쇄성폐질환은 미세먼지로 인해 유발된 것일 수 있으나, 이에 제한되는 것은 아니다.
본 발명에서 사용되는 용어, “만성폐쇄성폐질환(chronic obstructive pulmonary disease)”은 담배, 대기오염 또는 독성흡입물질에 의해 기도에 염증이 지속돼 기도가 좁아지면서 서서히 기도폐쇄가 일어나는 질환을 의미한다.
본 발명의 일 실시예에 있어서, 상기 만성폐쇄성폐질환(chronic obstructive pulmonary disease)은 만성 기관지염(chronic bronchitis), 폐기종(Emphysema) 또는 천식(asthma)일 수 있다.

본 발명에 따른 산 부가염은 통상의 방법, 예를 들면, 카테킨 하이드레이트를 유기용매, 예를 들면 메탄올, 에탄올, 아세톤, 메틸렌클로라이드, 아세토니트릴 등에 녹이고 유기산 또는 무기산을 가하여 생성된 침전물을 여과, 건조하여 제조되거나, 용매와 과량의 산을 감압 증류한 후 건조하거나 유기용매 하에서 결정화시켜셔 제조할 수 있다.
또한, 염기를 사용하여 약학 또는 식품학적으로 허용가능한 금속염을 만들 수 있다. 알칼리 금속 또는 알칼리 토금속 염은 예를 들면 화합물을 과량의 알칼리 금속 수산화물 또는 알칼리 토금속 수산화물 용액 중에 용해하고, 비용해 화합물 염을 여과하고, 여액을 증발, 건조시켜 얻는다. 이때, 금속염으로는 나트륨, 칼륨 또는 칼슘염을 제조하는 것이 제약상 적합하다. 또한, 이에 대응하는 은 염은 알칼리 금속 또는 알칼리 토금속 염을 적당한 은 염(예, 질산은)과 반응시켜 얻는다.

본 발명의 약학적 조성물은 약학적으로 유효한 양으로 투여한다. 본 발명에서 사용되는 용어 "약학적으로 유효한 양"은 의학적 치료에 적용 가능한 합리적인 수혜/위험 비율로 질환을 치료하기에 충분하며 부작용을 일으키지 않을 정도의 양을 의미하며, 유효용량 수준은 환자의 건강상태, 질환의 종류, 중증도, 약물의 활성, 약물에 대한 민감도, 투여 방법, 투여 시간, 투여 경로 및 배출 비율, 치료기간, 배합 또는 동시 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 결정될 수 있다. 본 발명의 조성물은 개별 치료제로 투여하거나 다른 치료제와 병용하여 투여될 수 있고, 종래의 치료제와 순차적으로 또는 동시에 투여될 수 있으며, 단일 또는 다중 투여될 수 있다. 상기한 요소들을 모두 고려하여, 부작용 없이 최소한의 양으로 최대 효과를 얻을 수 있는 양을 투여하는 것이 중요하며, 이는 당업자에 의해 용이하게 결정될 수 있다.

본 발명에 따른 조성물은 약학적으로 유효한 양의 카테킨 하이드레이트 또는 이의 식품학적으로 허용가능한 염을 단독으로 포함하거나 하나 이상의 약학적으로 허용되는 담체, 부형제 또는 희석제를 포함할 수 있다. 상기에서 약학적으로 유효한 양이란 만성폐쇄성폐질환의 증상을 예방, 개선 및 치료하기에 충분한 양을 말한다. 상기에서 "약학적으로 허용되는"이란 생리학적으로 허용되고 인간에게 투여될 때, 통상적으로 위장 장애, 현기증과 같은 알레르기 반응 또는 이와 유사한 반응을 일으키지 않는 조성물을 말한다.

본 발명에서 사용되는 용어 "투여"는 임의의 적절한 방법으로 개체에 소정의 본 발명의 조성물을 제공하는 것을 의미한다.
또한, 본 발명의 약학적 조성물은 정제, 환제, 산제, 과립제, 캡슐제, 현탁제, 내용액제, 유제, 시럽제, 멸균된 수용액, 비수성용제, 현탁제, 유제, 동결건조제제 및 좌제로 이루어진 군으로부터 선택되는 어느 하나의 제형을 가질 수 있다. 좌제의 기제로는 위텝솔(witepsol), 마크로골, 트윈(tween) 61, 카카오지, 라우린지, 글리세롤, 젤라틴 등이 사용될 수 있다.
본 발명의 카테킨 하이드레이트 또는 이의 식품학적으로 허용가능한 염은 임상 투여시에 경구 및 비경구의 여러 가지 제형으로 투여될 수 있으며, 제제화할 경우에는 보통 사용하는 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 제조된다.
경구투여를 위한 고형 제제에는 정제, 환자, 산제, 과립제, 캡슐제, 트로키제 등이 포함되며, 이러한 고형 제제는 하나 이상의 본 발명의 카테킨 하이드레이트 또는 이의 식품학적으로 허용가능한 염에 적어도 하나 이상의 부형제 예를 들면, 전분, 탄산칼슘, 수크로스(sucrose), 락토오스(lactose) 또는 젤라틴 등을 섞어 조제된다. 또한, 단순한 부형제 외에 마그네슘 스티레이트 탈크 같은 윤활제들도 사용된다. 경구 투여를 위한 액상 제제로는 현탁제, 내용액제, 유제 또는 시럽제 등이 해당되는데, 흔히 사용되는 단순 희석제인 물, 리퀴드 파라핀 이외에 여러 가지 부형제, 예를 들면 습윤제, 감미제, 방향제, 보존제 등이 포함될 수 있다.

본 발명의 약학적 조성물은 단독으로, 또는 수술, 방사선 치료, 호르몬 치료, 화학 치료 또는 생물학적 반응조절제를 사용하는 방법들과 병용하여 사용할 수 있다.
본 발명의 약학적 조성물은 카테킨 하이드레이트 또는 이의 식품학적으로 허용가능한 염을 유효성분으로 포함하는 외용제의 제형으로 제공할 수 있다. 본 발명의 약학적 조성물을 피부외용제로 사용하는 경우, 추가로 지방 물질, 유기 용매, 용해제, 농축제 및 겔화제, 연화제, 항산화제, 현탁화제, 안정화제, 발포제(foaming agent), 방향제, 계면활성제, 물, 이온형 유화제, 비이온형 유화제, 충전제, 금속이온봉쇄제, 킬레이트화제, 보존제, 비타민, 차단제, 습윤화제, 필수 오일, 염료, 안료, 친수성 활성제, 친유성 활성제 또는 지질 소낭 등 피부 외용제에 통상적으로 사용되는 임의의 다른 성분과 같은 피부 과학 분야에서 통상적으로 사용되는 보조제를 함유할 수 있다. 또한, 상기 성분들은 피부과학 분야에서 일반적으로 사용되는 양으로 도입될 수 있다. 본 발명의 약학적 조성물이 피부 외용제로 제공될 경우, 이에 제한되는 것은 아니나, 연고, 패취, 겔, 크림 또는 분무제 등의 제형일 수 있다.
또한, 본 발명의 카테킨 하이드레이트 또는 이의 식품학적으로 허용가능한 염의 인체에 대한 효과적인 투여량은 환자의 나이, 몸무게, 성별, 투여형태, 건강상태 및 질환 정도에 따라 달라질 수 있으며, 일반적으로 약 0.001-100 mg/kg/일이며, 바람직하게는 0.01-35 mg/kg/일이다. 몸무게가 70㎏인 성인 환자를 기준으로 할 때, 일반적으로 0.07-7000 mg/일이며, 바람직하게는 0.7-2500 ㎎/일이며, 의사 또는 약사의 판단에 따라 일정시간 간격으로 1일 1회 내지 수회로 분할 투여할 수도 있다.

본 발명에 따른 카테킨 하이드레이트 또는 이의 식품학적으로 허용가능한 염을 함유하는 식품 조성물은 식품에 그대로 첨가하거나 다른 식품 또는 식품 성분과 함께 사용될 수 있고, 통상적인 방법에 따라 적절하게 사용될 수 있다. 카테킨 하이드레이트 또는 이의 식품학적으로 허용가능한 염의 혼합량은 그의 사용 목적(예방 또는 개선용)에 따라 적합하게 결정될 수 있다. 일반적으로, 건강식품 및 건강기능식품 중의 상기 조성물의 양은 전체 식품 중량의 0.1 내지 90 중량부로 가할 수 있다. 그러나 건강 유지를 목적으로 하거나 또는 건강 조절을 목적으로 하는 장기간의 섭취의 경우에는 상기 양은 상기 범위 이하일 수 있으며, 안전성 면에서 아무런 문제가 없기 때문에 유효성분은 상기 범위 이상의 양으로도 사용될 수 있다.
본 발명의 식품 조성물은 지시된 비율로 필수 성분으로서 본 발명 카테킨 하이드레이트 또는 이의 식품학적으로 허용가능한 염을 함유하는 외에는 다른 성분에는 특별한 제한이 없으며 통상의 음료와 같이 여러 가지 향미제 또는 천연 탄수화물 등을 추가 성분으로서 함유할 수 있다. 상술한 천연 탄수화물의 예는 모노사카라이드, 예를 들어, 포도당, 과당 등; 디사카라이드, 예를 들어 말토스, 슈크로스 등; 및 폴리사카라이드, 예를 들어 덱스트린, 시클로덱스트린 등과 같은 통상적인 당, 및 자일리톨, 소르비톨, 에리트라이톨 등의 당알코올이다. 상술한 것 이외의 향미제로서 천연 향미제(타우마틴, 스테비아 추출물(예를 들어 레바우디오시드 A, 글리시르히진등) 및 합성 향미제(사카린, 아스파르탐 등)를 유리하게 사용할 수 있다. 상기 천연 탄수화물의 비율은 본 발명의 건강기능성 식품 조성물 100 당 일반적으로 약 1 내지 20 g, 바람직하게는 약 5 내지 12 g이다.
상기 외에 본 발명의 카테킨 하이드레이트 또는 이의 식품학적으로 허용가능한 염을 함유하는 식품 조성물은 여러 가지 영양제, 비타민, 광물(전해질), 합성 풍미제 및 천연 풍미제 등의 풍미제, 착색제 및 중진제(치즈, 초콜릿 등), 펙트산 및 그의 염, 알긴산 및 그의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알코올, 탄산음료에 사용되는 탄산화제 등을 함유할 수 있다. 그 밖에 본 발명의 식품 조성물은 천연 과일쥬스 및 과일쥬스 음료 및 야채 음료의 제조를 위한 과육을 함유할 수 있다.
이러한 성분은 독립적으로 또는 조합하여 사용할 수 있다. 이러한 첨가제의 비율은 그렇게 중요하진 않지만 본 발명의 카테킨 하이드레이트 또는 이의 식품학적으로 허용가능한 염을 함유하는 식품 조성물 100 중량부 당 0.1 내지 약 20 중량부의 범위에서 선택되는 것이 일반적이다.

본 발명의 건강기능식품의 일 예로 건강음료 조성물로 사용될 수 있으며, 이 경우 통상의 음료와 같이 여러 가지 향미제 또는 천연 탄수화물 등을 추가 성분으로 함유할 수 있다. 상술한 천연 탄수화물은 포도당, 과당과 같은 모노사카라이드; 말토스, 수크로스와 같은 디사카라이드; 덱스트린, 사이클로덱스트린과 같은 폴리사카라이드; 자일리톨, 소르비톨, 에리트리톨 등의 당 알콜일 수 있다. 감미제는 타우마틴, 스테비아 추출물과 같은 천연 감미제; 사카린, 아스파르탐과 같은 합성 감미제 등을 사용할 수 있다. 상기 천연 탄수화물의 비율은 본 발 명의 건강음료 조성물 100 mL 당 일반적으로 약 0.01 내지 0.04 g, 구체적으로 약 0.02 내지 0.03 g이 될 수 있다.
상기 외에 건강음료 조성물은 여러 가지 영양제, 비타민, 전해질, 풍미제, 착색제, 펙트산, 펙트산의 염, 알긴산, 알긴산의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알코올 또는 탄산화제 등을 함유할 수 있다. 그 밖에 천연 과일주스, 과일주스 음료, 또는 야채 음료의 제조를 위한 과육을 함유할 수 있다. 이러한 성분은 독립적으로 또는 혼합하여 사용할 수 있다. 이러한 첨가제의 비율은 크게 중요하진 않지만 본 발명의 건강음료 조성물 100 중량부당 0.01 내지 0.1 중량부의 범위에서 선택되는 것이 일반적이다.
일 측면에서, 본 발명은 상기 약학적 조성물을 개체에 투여하는 단계를 포함하는 만성폐쇄성폐질환 예방 또는 치료방법에 관한 것이다.
본 발명의 약학적 조성물은 치료학적으로 유효한 양 또는 약학적으로 유효한 양으로 투여될 수 있다.
본 발명에서, 용어 "치료학적으로 유효한 양"은 대상 질환을 예방 또는 치료하는데 유효한 조성물의 약학적으로 허용가능한 염의 양을 의미하며, 본 발명의 조성물의 치료적으로 유효한 양은 여러 요소, 예를 들면 투여방법, 목적부위, 환자의 상태 등에 따라 달라질 수 있다. 따라서, 인체에 사용 시 투여량은 안전성 및 효율성을 함께 고려하여 적정량으로 결정되어야 한다. 동물실험을 통해 결정한 유효량으로부터 인간에 사용되는 양을 추정하는 것도 가능하다. 유효한 양의 결정시 고려할 이러한 사항은, 예를 들면 Hardman and Limbird, eds., Goodman and Gilman's The Pharmacological Basis of Therapeutics, 10th ed. (2001), Pergamon Press; 및 E.W. Martin ed., Remington's Pharmaceutical Sciences, 18th ed. (1990), Mack Publishing Co.에 기술되어있다.
본 발명에서, 용어 “약학적으로 유효한 양”은 의학적 치료에 적용 가능한 합리적인 수혜/위험 비율로 질환을 치료하기에 충분하며 부작용을 일으키지 않을 정도의 양을 의미하며, 유효용량 수준은 환자의 건강상태, 질환의 종류, 중증도, 약물의 활성, 약물에 대한 민감도, 투여 방법, 투여 시간, 투여 경로 및 배출 비율, 치료기간, 배합 또는 동시 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 결정될 수 있다. 본 발명의 조성물은 개별 치료제로 투여하거나 다른 치료제와 병용하여 투여될 수 있고, 종래의 치료제와 순차적으로 또는 동시에 투여될 수 있으며, 단일 또는 다중 투여될 수 있다. 상기한 요소들을 모두 고려하여, 부작용 없이 최소한의 양으로 최대 효과를 얻을 수 있는 양을 투여하는 것이 중요하며, 이는 당업자에 의해 용이하게 결정될 수 있다.
이하, 본 발명을 실시예를 통하여 더욱 상세히 설명하기로 한다. 이들 실시예는 본 발명을 보다 구체적으로 설명하기 위한 것으로서, 본 발명의 범위가 이들 실시예에 한정되는 것은 아니다.
<준비예 1> 재료 및 시약 준비
Catechin hydrate는 Sigma-Aldrich(St. Louis, MO, USA)에서 구매하였고, 세포실험에 사용하기 위해 용매인 Dimethyl sulfoxide(DMSO)에 용해시킨 후 실험을 진행하였다. Thiazolyl Blue Tetrazolium Bromide (MTT), Ovalbumin (OVA)는 Sigma-Aldrich(St. Louis, MO, USA) 사에서 구매하여 사용하였으며, JC-1, MitoSOX은 Thermo Fisher Scientific(Waltham, USA), FITC Annexin V apoptosis detection kit는 BD Biosciences(Franklin Lakes, NJ, USA), 미토콘드리아 내 산소호흡률 측정을 위한 Seahorse XF Cell Mito Stress Test Kit 및 그 외 시약은 Agilent(California, USA), Diesel Particulate Matter (SRM 2975)은 NIST(Maryland, USA)사 에서 구매하여 사용하였다.
<준비예 2> 실험 동물의 준비
본 발명의 동물실험은 경북대학교 동물실험 윤리위원회 심의 동의 하에 실시하였다. 6주령의 암컷 BALB/C mice(샘타코바이오코리아, 경기도 오산)은 케이지 당 5마리씩 무작위로 배치한 후 20 내지 25 ℃, 습도 45 내지 50%에서 주야 주기가 12시간으로 조절된 동물실험실에서 2주간 순화 기간을 거쳤다. 사료는 정상식이로 공급하였고, 섭취와 음수는 자유식이로 공급하였다. 실험군은 비슷한 체중을 가진 그룹으로, 군당 10마리씩 5개 군으로 나누었다.
<실시예 1> Catechin hydrate의 1-Nitropyrene에 의해 유도된 세포사멸에 대한 회복능 평가
Catechin hydrate가 미세먼지 대표 물질인 1-Nitropyrene에 의한 세포사멸에 미치는 영향을 확인하기 위해, 다양한 농도의 catechin hydrate가 전처리된 인간기관지상피세포에 1-Nitropyrene를 처리한 후 세포생존률을 측정하였다.
구체적으로 인간기관지상피세포인 BEAS-2B에 작약추출물 및 Catechin hydrate를 12.5, 25 또는 50μM 농도로 1시간 동안 전처리한 후, 50μM의 1-Nitropyrene을 24시간 동안 처리하여 세포사멸을 유도하였다. 이후 MTT((3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) 시약을 2시간 처리하여 595nm의 흡광도에서 세포 생존률을 확인하였다.
그 결과, 도 1에 나타낸 바와 같이 1-Nitropyrene는 BEAS-2B의 세포사멸을 유발하여 세포생존률이 감소하는 것으로 나타났다. 또한, 12.5μM 또는 25μM 농도의 Catechin hydrate가 처리된 BEAS-2B 세포의 경우 1-Nitropyrene에 의한 세포생존률의 감소를 유의하게 증가시키지 못한 반면, 50μM 농도의 Catechin hydrate는 1-Nitropyrene에 의해 감소된 세포생존률을 유의하게 회복시키는 것으로 나타났다.
<실시예 2> Catechin hydrate의 1-Nitropyrene에 의해 유도된 세포자멸사 억제능 평가
Catechin hydrate가 1-Nitropyrene에 의한 세포사멸에 미치는 영향을 확인하기 위해, 다양한 농도의 Catechin hydrate가 전처리된 인간기관지상피세포에 1-Nitropyrene를 처리한 후 유세포 분석법을 이용하여 Apoptosis를 측정하였다.
구체적으로, 인간기관지상피세포인 BEAS-2B에 Catechin hydrate를 25 또는 50μM 농도로 1시간 동안 전처리한 후, 50μM의 1-Nitropyrene을 24시간 동안 처리하여 세포자멸사를 유도하였다. 이후 세포를 얻어 DNA손상과 Apoptosis가 유발된 세포에 결합하는 Propidium Iodide(PI)과 FITC Annexin V 염색시약을 세포와 함께 15분간 반응시킨 후 유세포 분석기를 통해 세포자멸사가 유발된 세포를 검출하였다.
그 결과, 도 2에 나타낸 바와 같이 Catechin hydrate는 25 또는 50μM 농도에서 1-Nitropyrene에 의해 증가된 세포자멸사를 감소시켰으며, 특히 50μM 농도의 Catechin hydrate의 경우 정상대조군(1-Nitropyrene가 처리되지 않은 세포)과 유사한 수준으로 세포자멸사를 감소시키는 것을 확인하였다.
<실시예 3> Catechin hydrate의 1-Nitropyrene 유도 cleaved-PARP, Cytochrome C 생성 억제 활성 평가
Catechin hydrate가 1-Nitropyrene에 의한 세포사멸에 미치는 영향을 확인하기 위해, 세포사멸인자인 cleaved-PARP와 Cytochrome C의 발현량을 측정하였다.
구체적으로, 인간기관지상피세포인 BEAS-2B에 12.5, 25 또는 50μM 농도로 Catechin hydrate를 1시간 동안 전처리한 후, 50μM의 1-Nitropyrene을 24시간 동안 처리하여 세포를 용해시켜 수확하였다. 이후 단백질 정량을 통하여 일정량을 단백질을 포함하도록 하였고, 단백질 혼합물 중 특정 단백질 분자를 검출하는 데 사용되는 방법인 웨스턴 블롯(western blot)으로 cleaved-PARP 및 Cytochrome C의 발현량을 평가하였다.
발현량 평가를 위한 웨스턴 블롯은 겔 전기영동을 사용하여 단백질 분자를 크기에 따라 분리 한 후 단백질을 겔에서 멤브레인으로 옮기며, 멤브레인을 cleaved-PARP에 특이적으로 결합하는 1차 항체와 배양하여 반응시킨 이후 1차 항체를 특이적으로 인식하고 결합하는 2차 항체와 함께 배양하였다. 2차 항체는 빛을 생성하여 쉽게 감지할 수 있으며, 이러한 단계를 통해 단백질 혼합물 중에서 cleaved-PARP 및 Cytochrome C의 발현량을 검출하였고, image J 소프트웨어 프로그램을 사용하여 발현량을 정량하였다.
그 결과, 도 3에 나타낸 바와 같이 1-Nitropyrene은 BEAS-2B의 세포사멸을 유발하여 cleaved-PARP과 Cytochrome C의 발현을 증가시켰으나, 12.5, 25 및 50μM 농도의 Catechin hydrate를 전처리하는 경우 cleaved-PARP 및 Cytochrome C의 발현을 현저하게 억제하는 것을 확인하였다.
<실시예 4> Catechin hydrate의 1-Nitropyrene에 의해 유도된 미토콘드리아 호흡률 감소에 대한 회복능 평가
상기 실시예 1 내지 3을 통해 Catechin hydrate가 인간기관지상피세포 세포사멸을 억제하는 것을 확인하였으므로, 세포자멸사의 상위 메커니즘인 미토콘드리아 기능 장애를 억제하는지 확인하기 위해 다양한 농도의 Catechin hydrate가 전처리된 세포의 미토콘드리아 호흡률 변화를 측정하였다.
구체적으로, 인간기관지상피세포인 BEAS-2B에 Catechin hydrate를 12.5, 25 또는 50μM 농도로 1시간 동안 전처리한 후, 50μM의 1-Nitropyrene을 6시간 동안 처리하여 미토콘드리아 기능 장애를 유도하였다. 이후 Seahorse XF 장비를 활용하여 mito stress assay를 진행하였고, 분석 동안 미토콘드리아의 전자수송사슬(Electron transport chain) 내 복합체를 억제하거나 활성화시키는 Oligomycin, Carbonyl cyanide-4(trifluoromethoxy) phenylhydrazone(FCCP), Rotenone/antimycin A가 세포에 차례대로 첨가되어 미토콘드리아 산소 소비율(oxygen consumption rate(OCR)) 및 기능 지표들을 측정하였다.
그 결과, 도 4에 나타낸 바와 같이 Catechin hydrate는 12.5, 25 및 50μM 농도에서 1-Nitropyrene에 의해 감소된 미토콘드리아 최대 호흡량(maximal respiration) 및 예비 용량(spare capacity)을 유의하게 회복시키는 것을 확인하였다.
<실시예 5> Catechin hydrate의 1-Nitropyrene에 의해 유도된 미토콘드리아 막전위 손상에 대한 회복능 평가
상기 실시예 1 내지 3을 통해 Catechin hydrate가 인간기관지상피세포 세포사멸을 억제하는 것을 확인하였으므로, 세포자멸사의 상위 메커니즘인 미토콘드리아 기능 장애를 억제하는지 확인하기 위해 다양한 농도의 Catechin hydrate가 전처리된 세포의 미토콘드리아 막전위 손상 수준을 측정하였다.
구체적으로, 인간기관지상피세포인 BEAS-2B에 Catechin hydrate를 25 또는 50μM 농도로 1시간 동안 전처리한 후, 50μM의 1-Nitropyrene을 6시간 동안 처리하여 미토콘드리아 기능 장애를 유도하였다. 이후 세포는 4% 파라포름알데히드로 20분간 고정된 후 미토콘드리아 막 전위 지표로 활용되는 JC-1 염색시약이 2μM의 농도로 20분간 처리되었다. 이후 washing 과정을 거쳐 세포 내 핵을 염색시키는 4,6-diamidino-2-phenylindole(DAPI) 시약으로 염색되어 형광현미경으로 관찰되었다. 관찰된 이미지의 Red/Green intensity는 image J 소프트웨어 프로그램을 사용하여 측정하였다.
그 결과, 도 5에 나타낸 바와 같이 Catechin hydrate는 25 및 50μM 농도에서 Red/Green intensity 비율을 회복시킴으로써 1-Nitropyrene에 의해 유도된 미토콘드리아 막전위 손상을 유의하게 회복시키는 것을 확인하였다.
<실시예 6> Catechin hydrate의 1-Nitropyrene에 의해 유도된 미토콘드리아 내 ROS 생성량 증가에 대한 회복능 평가
상기 실시예 1 내지 3을 통해 Catechin hydrate가 인간기관지상피세포 세포사멸을 억제하는 것을 확인하였으므로, 세포자멸사의 상위 메커니즘인 미토콘드리아 기능 장애를 억제하는지 확인하기 위해 다양한 농도의 Catechin hydrate가 전처리된 세포의 미토콘드리아 내 활성 산소(reactive oxygen species, ROS) 생성량을 측정하였다.
구체적으로, 인간기관지상피세포인 BEAS-2B에 Catechin hydrate를 25 또는 50μM 농도로 1시간 동안 전처리한 후, 50μM의 1-Nitropyrene을 6시간 동안 처리하여 미토콘드리아 기능 장애를 유도하였다. 이후 세포는 4% 파라포름알데히드로 20분간 고정된후 미토콘드리아 ROS 생성 지표로 활용되는 MitoSOX 염색시약이 5μM의 농도로 10분간 처리되었다. 이후 washing 과정을 거쳐 세포 내 핵을 염색시키는 4,6-diamidino-2-phenylindole(DAPI) 시약으로 염색되어 형광현미경으로 관찰되었다. 관찰된 이미지의 Red/DAPI intensity는 image J 소프트웨어 프로그램을 사용하여 측정하였다.
그 결과, 도 6에 나타낸 바와 같이 Catechin hydrate는 25 및 50μM 농도에서 1-Nitropyrene에 의해 유도된 미토콘드리아 ROS 과생성을 유의하게 억제시키는 것을 확인하였다.
<실시예 7> Catechin hydrate 경구투여 및 DEP 비강투여를 통한 만성폐쇄성폐질환 유도
Catechin hydrate의 생체 내 미세먼지 유발 폐 손상에 대한 영향 평가를 위해 도 7을 참조하여 만성폐쇄성폐질환 동물모델을 제작하였다.
구체적으로, 2, 10mg/kg의 농도로 멸균증류수에 용해시킨 Catechin hydrate를 실험용 마우스에 3일 동안 경구투여 하였다. 이후 전신면역반응을 유도하기 위해 Al(OH)3과 PBS에 희석시킨 오브알부민(ovalbumin, OVA)을 마우스 당 20μg씩 주 1회, 총 3주간(실험시작 4,11,18일째) 복강투여 하였다. 실험 4주째에는 폐에 직접적인 염증반응유도를 위해 PBS에 희석시킨 OVA 용액(OVA+PBS+Al(OH)3)을 마우스 당 20μg씩 연속 2일(실험시작 25,26일) 비강투여 하였고, 희생 하루 전 폐에 직접적인 미세먼지 노출을 위해 PM2.5에 해당하는 디젤배기입자(Diesel exhaust particle, DEP) 100μg을 OVA가 포함된 PBS에 용해시킨 DEP 용액(DEP+OVA+PBS)을 마우스에 비강투여하였다. 전체 실험 기간 동안 Catehcin hydrate는 주 3회씩 경구투여 되었으며, 경구투여 1시간 후 OVA 복강투여 및 비강투여를 통해 면역반응이 유도되었다. 또한 모든 비강투여는 4% isoflurane에 약 2분간 노출되어 호흡 마취된 후 진행되었다. DEP를 투여하고 24시간이 경과된 후 모든 마우스를 희생시켰으며, 희생된 마우스로부터 폐 조직을 수득하여 실험에 사용하였다.
<실시예 8> Catechin hydrate의 DEP에 의해 유도된 cleaved-caspase3, Cytochrome C, IL-1β 생성 억제 활성 평가
DEP 비강투여로 유도된 만성폐쇄성폐질환 마우스 모델 내 Catechin hydrate의 세포사멸 마커 발현 수준 억제 활성을 평가하기 위해 세포사멸인자인 cleaved-caspase3, Cytochrome C 및 염증성 사이토카인 IL-1β 발현량을 웨스턴 블롯(western blot)으로 평가하였다. 각각의 동물모델에서 수득된 폐 조직은 조직 파쇄기를 이용하여 파쇄시킨 후 lysis된 상층액만을 분리하여 단백질 정량을 통해 일정량의 단백질을 포함하도록 하였다.
세포사멸 마커 발현 수준 측정을 위한 웨스턴 블롯은 겔 전기영동을 사용하여 단백질 분자를 크기에 따라 분리 한 후 단백질을 겔에서 멤브레인으로 옮기며, 멤브레인을 cleaved-PARP에 특이적으로 결합하는 1차 항체와 배양하여 반응시킨 이후 1차 항체를 특이적으로 인식하고 결합하는 2차 항체와 함께 배양하였다. 2차 항체는 빛을 생성하여 쉽게 감지할 수 있으며, 이러한 단계를 통해 단백질 혼합물 중에서 leaved-caspase3, Cytochrome C 및 염증성 사이토카인 IL-1β을 검출하였다.
그 결과, 도 8에 나타낸 바와 같이 DEP 투여군은 OVA 투여군 대비 cleaved-caspase3, Cytochrome C 및 IL-1β의 발현을 증가시켰으나, 이에 Catechin hydrate 저농도 투여군 및 고농도 투여군은 DEP에 의해 증가된 세포사멸 마커의 발현을 현저하게 억제하는 것을 확인하였다.
<실시예 9> Catechin hydrate의 DEP에 의해 유도된 Mucin 5AC 발현 억제 활성 평가
만성폐쇄성폐질환 마우스 모델에서는 기도 내부에 정상적으로 유지되고 있던 점액이 비정상적으로 과생성되어 기관지 염증을 나타낸다. 따라서 고정된 폐 조직 내에서 Catechin hydrate 투여에 따른 점액생성에 관련된 단백질인 Mucin 5AC의 발현 수준을 측정하였다.
희생을 위해 개복된 마우스의 폐 조직 내부를 10% formalin을 250mmH2O의 압력으로 5분간 고정시킨 후 폐 조직을 회수하였다. 얻어진 폐 조직은 24시간 동안 10% formalin에 보관된 후 OCT solution에 고정하기 위해 30% sucrose에서 24시간 동안 보관되었다. 이후 조직 고정에 사용되는 OCT solution을 사용하여 폐 조직을 동결 및 고정시킨 후 -20℃에서 4μm 두께로 section하여 면역형광염색에 사용하였다. 면역형광염색을 위해 얻어진 폐 조직 절편을 4% formaldehyde로 고정한 후 하루 동안 Mucin 5AC에 특이적으로 결합하는 1차 항체와 배양하여 반응시켰다. 형광이 붙은 2차 항체를 1차 항체와 반응하도록 함께 배양하여 형광현미경으로 단백질 발현을 관찰하였다.
그 결과, 도 9에 나타낸 바와 같이 DEP 투여군은 OVA 투여군 대비 Mucin 5AC의 발현을 증가시켰으나, Catechin hydrate 저농도 투여군 및 고농도 투여군은 이러한 점액생성 관련 단백질인 Mucin 5AC의 발현 수준을 현저하게 감소시키는 것으로 나타났으며, 이러한 결과는 Catechin hydrate는 투여량에 관계 없이 DEP에 의해 증가된 Mucin 5AC의 생성을 유의하게 억제한다는 것을 의미한다.
<실시예 10> Catechin hydrate의 DEP에 의해 유도된 cleaved-caspase 3 발현 억제 활성 평가
DEP 비강투여로 유도된 만성폐쇄성폐질환 마우스 모델 내 Catechin hydrate의 세포사멸 마커 발현 억제 활성 평가를 위해 고정된 폐 조직 내에서 Catechin hydrate 투여에 따른 세포사멸인자인 cleaved-caspase3의 발현 수준을 측정하였다.

그 결과, 도 10에 나타낸 바와 같이 DEP 투여군은 OVA 투여군 대비 Cleaved-caspase 3의 발현을 증가시켰으나, Catechin hydrate 저농도 투여군 및 고농도 투여군은 이러한 세포사멸인자 Cleaved-caspase 3의 증가된 발현을 현저하게 감소시키는 것을 확인하였다.
