Abstract
医薬製剤等に使用される遮光性のハードカプセルシェル皮膜およびハードカプセルシェル及びその製造方法を提供する。 健康上の問題のあるとされる酸化チタンではなく、従来から医薬品として安全に使用されている酸化マグネシウムを遮光剤として皮膜形成組成物によりハードカプセルシェル皮膜を製造する。遮光性ハードカプセルシェルは、この皮膜によって形成される。このハードカプセルシェルは光および紫外線の透過を低減することができるので、シェル内に充填された任意の有効成分の安定性および品質を長期間維持することができる。
Description
本発明は、医薬製剤等に使用される遮光性ハードカプセルシェル皮膜およびハードカプセルシェルに関する。前記シェルは、遮光性ハードカプセルシェル皮膜から形成される。また、本発明は、遮光剤を含有するハードカプセルシェル皮膜形成用の組成物に関する。また、本発明は、遮光性のハードカプセルシェルの製造方法に関する。
さらに、本発明は、ハードカプセルシェル皮膜が、遮光剤として酸化マグネシウムを含む皮膜形成組成物によって生成される、遮光性ハードカプセルシェル皮膜、およびハードカプセルシェルに関するものであり、前記シェルは、前記遮光性ハードカプセルシェル皮膜から形成される。このハードカプセルシェル皮膜およびハードカプセルシェルは、光および紫外線の透過を低減することができ、それにより、ハードカプセルシェルは、前記シェルに充填された任意の有効成分の安定性および品質を長期間維持することができる。
従来から医薬品や健康食品など、さまざまな製品にさまざまな着色が行われている。着色の目的としては、高い識別性、外観上の美しさの付与、品質維持のための遮光性の付与などがある。特に、光に不安定な成分を含む製剤については、一般的な方法は、ハードカプセルシェルのシェルを形成する皮膜に白色の遮光剤を添加して、白色の遮光性ハードカプセルシェルを製造することである。更に必要な場合には、種々の染料のような着色剤であって、可食性色素(例えば、青色1号、黄色5号、赤色3号等々)あるいは可食性である顔料(例えば、三二酸化鉄あるいはアルミニウムレーキ等)を添加して遮光した皮膜に自在な着色を付与している。
対象となる物質に遮光性を与える遮光剤としては種々のものが用いられているが、もっともよく用いられているのは酸化チタン(TiO2, 二酸化チタンともいう)である。酸化チタンは、優れた白色度や隠蔽力、着色力、極めて高い化学的安定性などの特色をもつ白色顔料であり、医薬品や化粧品分野で古くから用いられてきた。
しかし、これらの遮光された製剤中に配合される成分の中には、酸化チタンにより安定性が損なわれるもの、あるいは紫外線により酸化チタンから発生したラジカルのために分解が促進されるものもあり、製剤の品質の維持に問題があった。また、酸化チタン含有皮膜で被覆された製剤は、光照射により、経時的に皮膜の剥離や白浮きが生じ、コーティング状態を保つことが困難となり、その結果、そのような調剤の外観および取り扱いやすさが損なわれる可能性がある。さらに、光や酸素、水分により製剤中の成分が分解される問題も懸念される。
そこで、不透明または半透明のゼラチンハードカプセルを製造するにあたり、酸化チタンの代わりにより安全性が高く、栄養的にも価値のある不透明化剤として、平均粒径3μm以下の天然カルシウム粉末(炭酸カルシウムが主成分)を使用すること(特許文献2)が研究された。
しかし、内閣府の食品安全委員会によれば、2020年に、フランス食品環境労働衛生安全庁(ANSES)により、食品中のナノ材料、いわゆるナノ粒子の特定及び消費者の健康リスク評価に関する意見書が公表され、ナノ粒子が含まれる食品添加物材料として、炭酸カルシウムはその評価対象とされている。したがって、カプセルに対する炭酸カルシウムの使用については安全性の問題が生じている。(ウェブサイトhttps://www.fsc.go.jp/fsciis/foodSafetyMaterial/show/syu05400370475を参照)

また、酸化チタンの発がん性について、国際がん研究機関(IARC: International Agency for Research on Cancer)は、2006年6月に、顔料グレード及びウルトラファインTiO2に関する吸入及び気管内注入による動物実験の結果は、動物におけるTiO2の発がん性を示す証拠として十分であると結論し、グループ2B(ヒトに対する発がん性が疑われる)に分類した。

この点を考慮し、酸化チタンを使用せず、水溶性カルシウム塩から選択された1種以上と、水溶性セルロース系高分子の皮膜基剤とを含有する遮光性皮膜組成物も検討されている(特許文献2)。しかし、特許文献2に記載の水溶性カルシウム塩は、「食品添加物等の規格基準第2 添加物」の使用基準において、その使用量がカルシウムとして食品の1.0%以下でなければならないと制限されているため、白色着色濃度が制限されてしまうという問題があった。
一方、酸化マグネシウムを遮光性物質として利用する例としては、睡眠・鎮静作用を有する薬効成分としてジフェンヒドラミンまたはその酸付加塩を含有する固形剤を、遮光性物質と水溶性高分子物質を含有する皮膜で被覆してなることを特徴とする被覆固形催眠製剤(特許文献3)がある。特許文献3においては、固形剤としてハードカプセルが挙げられており、遮光性物質の一つとして酸化マグネシウムが挙げられている。しかしこの製剤は、カプセルシェルそのものに遮光性を付与したものではなく、薬剤をカプセルシェルに充填したカプセル剤を遮光性物質と水溶性高分子物質を含有する皮膜でコーティングしたものであり、製造工程が複雑になるという欠点がある。
また、特許文献4には、外層に酸化マグネシウムを含有する層を設けたゼラチンカプセルシェルが記載されている。この層は、ゼラチンカプセルの口腔粘膜への張り付きを防止するために付加されたものであり、カプセルシェルの一番外側に設ける必要がある。

上記のように酸化チタンの代替技術は研究されているものの、酸化チタンの粒子は非常に細かく、その隠蔽力の高さは他に並ぶものが無く、少量で十分な遮蔽力を発揮するため、食品や医薬品を含む多くの産業分野では酸化チタン粉末が遮光剤として広く使用されているのが現状であった。
このように、健康食品業界では酸化チタンのような人体を害する恐れのある物質のカプセルへの添加は望ましくないものの、遮光剤として適切な酸化チタンの代替はなかなか見つからなかった。したがって、これらに代わる遮光剤の開発がより一層望まれていた。
酸化チタンの代替品が満たす必要のあるさらなる要件は、カプセルに対して許容可能な機械的特性を提供することである。
さまざまな相対湿度条件下での保管に十分な安定性を備えたカプセルが望まれる。機械的特性は、例えば、カプセルの破壊および弾性挙動(例えば、チューブ試験による)、上部強度(例えばカプセル上部圧縮試験による)、およびプレ・ロックフォース強度(例えば、ボディとキャップの仮結合時に要する力であるプレ・ロックフォース測定試験による)である。これらは、許容範囲内にある必要がある重要なパラメータになる可能性がある。あるいは、機械的挙動を評価するために、ゲル化組成物によって形成された皮膜について、伸び率(例:フィルム引張試験による)、破断時の変形(例:フィルム引張試験による)、耐衝撃性(例:破壊試験の衝撃エネルギー)、溶出液による貫通時間(例えば、貫通時間測定試験による)、または耐パンク性(例えば、パンクテストによる)等を測定することができる。カプセルの十分な遮光特性と適切な機械的特性の間でバランスを見つける必要がある。

例えば、カプセルのポリマー水溶液に、酸化チタンあるいはその代替品粒子を添加してカプセルを製造すると、カプセル皮膜の厚みは100μm程度しかないため、粒子の混入により脆くなりカプセルが割れやすくなるリスクが発生する。即ち、その粒子は粒子径が大きくなるにつれて、または粒子の添加量が大きくなるにつれて皮膜の伸びが著しく低下してカプセル皮膜が割れやすくなるという問題が発生する。このような機械的特性の低下リスクを回避するために、添加する遮光剤(粒子)の粒子径と添加量を最適化したハードカプセルシェルを提供することが望ましい。
本発明者らは、特に健康食品業界に待望されていた酸化チタンに代わりうる遮光剤を開発するため、前述の技術的課題に取り組み、皮膜形成性ポリマーに添加する遮光剤として酸化マグネシウムの利用を検討した。その結果、酸化マグネシウムにより、酸化チタンを使用したものに匹敵する見た目の美しさや遮光性能が実現できることを見出した。また、他の色材と組み合わせることにより遮光と同時に自在な着色を行うことも可能であることが分かった。各種食用タール系色素や特に健康食品市場向けには、例えばベニバナ色素、カラメル色素、クチナシ等に代表される天然系色素と組み合わせることによって、安全性の高い美しい着色遮光カプセルを提供できる。
そして、粒子の形態の酸化マグネシウムを遮光剤として用いることにより、皮膜の伸び率を過度に低下させることなく上記の必要な機械的特性を有するハードカプセルシェルを提供することが見出された。好ましくは、粒子径と添加量を最適化した酸化マグネシウムを使用することができる。
本発明は、上記の知見に基づいて完成されたものであり、以下の態様を含む。

本発明のハードカプセルシェルは医薬、食品分野において使用することを意図している。したがって、ハードカプセルシェルの製造に使用される材料は、いずれも食品、医薬の分野で安全に使用可能なものに限られる。
酸化マグネシウムは、無色の結晶性鉱物ペリクレースとして自然に発生する。水酸化マグネシウムまたは炭酸マグネシウムを加熱することにより、かさばる白色粉末(軽い)または比較的密度の高い白色粉末(重い)のいずれかとして生成される。これらのマグネシウム塩を中程度の条件下(400~900℃で数時間)に加熱すると、いわゆる軽質酸化マグネシウムが生成される。より厳しい条件(例えば1200℃で12時間)で加熱すると、いわゆる重質酸化マグネシウムが生成される。軽質酸化マグネシウムは、高温で持続加熱すると重質酸化マグネシウムに変換される。
本発明で使用する酸化マグネシウムは、700℃以上、好ましくは800℃以上、より好ましくは900℃以上、さらに好ましくは少なくとも925℃の焼成温度で製造される。特定の実施形態における焼成温度は、950℃および1300℃である。

本発明で用いる酸化マグネシウムは、医薬の分野で制酸剤、緩下剤として、また、食品分野ではマグネシウム強化、サプリメント、栄養補助食品用などに汎用されているものである。しかし、これまで、カプセルシェルの遮光剤として使用されたことはなかった。つまり、ハードカプセルシェルのシェルを形成するハードカプセルシェル皮膜に対し遮光剤として使用されたことはなかった。
前記皮膜形成性ポリマーおよび前記酸化マグネシウムを含む前記ハードカプセルシェル皮膜中の前記酸化マグネシウムは、遮光剤および/または白色化剤として使用される。
ハードカプセルシェル皮膜の遮光剤として使用される酸化マグネシウムは、好ましくは、50μm以下、45μm以下、40μm以下、35μm以下、30μm以下、25μm以下、20μm以下、または15μm以下の粒子サイズを有する。50μmを超える場合は皮膜強度の低下が大きくなる。
ハードカプセルシェル皮膜の遮光剤として使用される酸化マグネシウムは、好ましくは、50μm以下、45μm以下、40μm以下、35μm以下、30μm以下、25μm以下、20μm以下、または15μm以下の粒子サイズを有する。50μmを超える場合は皮膜強度の低下が大きくなる。
粒子サイズ分布は、D10および/またはD90値によっても特徴付けることができる。D10/D90値は、粒子の10%/90%の粒子サイズがD10/D90値よりも小さい粒子サイズであることを表す。この場合、上記の%は累積体積%を意味する。
D50およびD100の値は、それぞれ次のように定義される。すなわち、D50またはD100の値は、それぞれ粒子の50%または100%がD50またはD100値より小さい粒径を有する粒径であることを意味する。
一実施形態では、酸化マグネシウムは、2.0μm以下、好ましくは1.5μm以下、より好ましくは1.0μm以下のD10値を有する粒子サイズ分布を有する。

上記粒度分布は、40重量%濃度の酸化マグネシウムをUltraTurraxの高速撹拌機で6,600rpmで15分間水中に分散した分散液を作り、水を加えて1×10-4g MgO/L程度まで希釈した後、島津粒度分布アナライザーSALD-2000Jにより680nmのレーザーを使用して測定した。
酸化マグネシウムは、軽質酸化マグネシウム、重質酸化マグネシウム共に遮光剤として使用することができるが、重質酸化マグネシウムがより好ましい。
酸化マグネシウムのグレードの例を表1に示す。

酸化マグネシウムを含有する皮膜形成性ポリマー溶液を調製する場合、最大限の柔軟性を得るためには、酸化マグネシウムをできるだけ濃縮した状態で水に分散することが望ましい。そこで上記のMS-0とMS-2の20%分散液、40%分散液を調製し、8日間にわたりそれぞれの粘度の変化を測定した。分散液は、酸化マグネシウム製品と水の混合物を、ホモジナイザーを用い10000rpmで10分間分散させ、全体をスパチュラでかき混ぜることによって調製した。この分散液に対し、調製時、1日後、8日後の粘度をブルックフィールドデジタル粘度計(BROOKFIELD DV2T)で測定した。結果を表2に示す。
本発明において使用する皮膜形成性ポリマーとしては、医薬品や食品に使用可能なものが挙げられる。例えば、カプセル皮膜の通常の原料であるゼラチン、ポリグルタミン酸等のタンパク質、セルロース誘導体、プルラン、スターチ、デキストリン、寒天、キトサン、酸処理破砕酵母等などの多糖類、ポリビニルアルコール(PVA)、PVA-メチルメタクリル酸共重合体、ポリビニルピロリドン(PVP)等が使用できる。しかし、皮膜形成性ポリマーであればこれに限らない。好ましい皮膜形成性ポリマーは、ゼラチン、セルロース誘導体又はプルランである。
セルロース誘導体の例には、非腸溶性セルロース誘導体および腸溶性セルロース誘導体が含まれる。非腸溶性セルロース誘導体には水溶性セルロースエーテルが含まれ、水溶性セルロースエーテルは、セルロースのヒドロキシル基の水素原子の一部が、アルキル基(例えばC1-4アルキル基)、ヒドロキシアルキル基(例えばC1-4ヒドロキシアルキル基)、又はそれらの組み合わせで置き換えられている水溶性セルロースエーテルであることが好ましい。例としては、メチルセルロース、ヒドロキシプロピルセルロース(HPC)などのヒドロキシ低級アルキルセルロース、およびヒドロキシエチルメチルセルロース、ヒドロキシエチルエチルセルロース、およびヒドロキシプロピルメチルセルロース(HPMC)などのヒドロキシル低級アルキルアルキルセルロースが含まれる。それらの中で、ヒドロキシプロピルメチルセルロース(ヒプロメロース、HPMC)が好ましい。たとえば、適切に使用できるHPMCの例としては、メトキシ基28.0~30.0%にヒドロキシプロポキシ基7.0~12.0%を含有するHPMC2910、メトキシ基27.0~30.0%にヒドロキシプロポキシ基4.0~7.5%を含有するHPMC2906、メトキシ基19.0~24.0%にヒドロキシプロポキシ基4.0~12.0%を含有するHPMC2208、約23.0~32.0%のヒドロキシプロポキシ基および約16.5~20.0%のメトキシ基を含むHPMC1828、およびそれらの混合物が挙げられる。
本発明において、HPMCメトキシおよびヒドロキシプロポキシ含有量は、米国薬局方に従って表される。例えば、HPMC2910、2906および2208はUSP43-NF38に収載されている。さまざまなHPMCグレードを単独または組み合わせて使用できる。好ましいHPMCは、HPMC2906、HPMC2910およびそれらの混合物である。
腸溶性セルロース誘導体には、ヒドロキシプロピルメチルセルロースアセテートサクシネート(HPMCAS)、ポリアクリル酸共重合体、カルボキシメチルエチルセルロース、ヒプロメロースフタル酸エステル(HPMCP)および酢酸フタル酸セルロース(CAP)などの腸溶性ポリマーが含まれる。
腸溶性セルロース誘導体には、ヒドロキシプロピルメチルセルロースアセテートサクシネート(HPMCAS)、ポリアクリル酸共重合体、カルボキシメチルエチルセルロース、ヒプロメロースフタル酸エステル(HPMCP)および酢酸フタル酸セルロース(CAP)などの腸溶性ポリマーが含まれる。
したがって、好ましい実施形態として、セルロース誘導体は、ヒプロメロース、ヒドロキシプロピルメチルセルロースアセテートサクシネート(HPMCAS)、カルボキシメチルエチルセルロース、ヒプロメロースフタル酸エステル(HPMCP)および酢酸フタル酸セルロース(CAP)からなる群から選択され、ヒプロメロースがより好ましい。その実施形態は本明細書に記載されている。
ゼラチンとしては、ウシまたはブタの骨から抽出されたアルカリ処理ゼラチン、ウシの皮、ブタの皮、または魚の鱗から抽出された酸処理ゼラチン、または、上記のアルカリ処理ゼラチンと酸処理ゼラチンを溶解し混合して作った混合ゼラチンが使用できる。
前記皮膜中の皮膜形成性ポリマーの量は、好ましくは、少なくとも75重量%、少なくとも77.5重量%、少なくとも80重量%、少なくとも82.5重量%、少なくとも85重量%、少なくとも87.5重量%、少なくとも90重量%、少なくとも92.5重量%、少なくとも95重量%、または少なくとも97.5重量%である。前記重量%は皮膜の乾燥重量に対する割合である。
皮膜形成性ポリマーの量の範囲は、皮膜の乾燥重量に対して、好ましくは83.3~99.75重量%、より好ましくは90.9~98.0重量%、さらにより好ましくは92.6~95.2重量%である。

本発明の一実施形態において、前記ハードカプセルシェル皮膜、およびそれにより形成される前記ハードカプセルシェルが残留水分の形態の水を除いて皮膜形成性ポリマーおよび酸化マグネシウムのみからなる。
前記のハードカプセルシェル皮膜およびそれにより形成される前記ハードカプセルシェルが残留水分の形態の水を除いて皮膜形成性ポリマーおよび酸化マグネシウムのみからなる場合は、乾燥皮膜または乾燥カプセルシェルにおける皮膜形成ポリマーおよび酸化マグネシウムの重量比の範囲は、次のとおりである。
皮膜形成性ポリマーが83.33~99.75重量%および酸化マグネシウムが0.25~16.67重量%、好ましくは、皮膜形成性ポリマーが90.91~98.04重量%および酸化マグネシウムが1.96~9.09重量%、より好ましくは、皮膜形成性ポリマーが92.59~95.24重量%および酸化マグネシウムが4.76~7.41重量%である。酸化マグネシウムの添加により、皮膜の伸長率が低下する傾向があり、衝撃に対し割れやすくなるが、可塑剤を添加することにより伸長率を改善することができる。
本発明の一実施形態において、前記ハードカプセルシェル皮膜、およびそれにより形成される前記ハードカプセルシェルが残留水分の形態の水を除いて皮膜形成性ポリマーおよび酸化マグネシウムのみからなる。
前記のハードカプセルシェル皮膜およびそれにより形成される前記ハードカプセルシェルが残留水分の形態の水を除いて皮膜形成性ポリマーおよび酸化マグネシウムのみからなる場合は、乾燥皮膜または乾燥カプセルシェルにおける皮膜形成ポリマーおよび酸化マグネシウムの重量比の範囲は、次のとおりである。
皮膜形成性ポリマーが83.33~99.75重量%および酸化マグネシウムが0.25~16.67重量%、好ましくは、皮膜形成性ポリマーが90.91~98.04重量%および酸化マグネシウムが1.96~9.09重量%、より好ましくは、皮膜形成性ポリマーが92.59~95.24重量%および酸化マグネシウムが4.76~7.41重量%である。酸化マグネシウムの添加により、皮膜の伸長率が低下する傾向があり、衝撃に対し割れやすくなるが、可塑剤を添加することにより伸長率を改善することができる。
上記の可塑剤としてはキシリトール、フルクトース、ソルビタン脂肪酸エステル、クエン酸トリエチル、グリセリン、ソルビトール(ソルビトール・ソルビタン液)、マンニトール、トレハロース、植物油(ゴマ油、ヒマシ油)、中鎖脂肪酸トリグリセライド、トリアセチン、フタル酸エステル類(フタル酸ジオクチル)、フィトステロール、プロピレングリコール、ポリソルベート、ポリエチレングリコール(マクロゴール)などが挙げられる。好ましい可塑剤は、ソルビタン脂肪酸エステルであり、さらに好ましくはソルビタンモノラウリン酸エステル(CAS1338-39-2、ソルビタンモノラウレート)である。
可塑剤の含有量は、使用する皮膜形成性ポリマーの重量(乾燥重量)に対して好ましくは0.01~10重量%、より好ましくは0.05~2.5重量%、さらにより好ましくは0.3~1.5重量%である。0.01重量%より少ない場合、あるいは10重量%より多い場合は可塑作用が損なわれることがある。
本発明の一実施形態では、前記ハードカプセルシェル皮膜、該皮膜で形成される前記ハードカプセルシェルは、残留水分の形態の水を除いて、皮膜形成性ポリマー、酸化マグネシウム、および可塑剤のみからなる。前記ハードカプセルシェル皮膜、前記該皮膜で形成されるハードカプセルシェルが残留水分の形態の水を除いて皮膜形成性ポリマー、酸化マグネシウムおよび可塑剤のみからなる場合、乾燥皮膜または乾燥カプセルシェルの重量に基づく皮膜形成性ポリマー、酸化マグネシウムおよび可塑剤の可能な含有量の範囲は次のとおりである。
皮膜形成性ポリマー76.92~99.74 重量%、酸化マグネシウム0.25~15.38重量%および可塑剤0.01~7.69重量%;

本発明のハードカプセルシェル皮膜は、使用する皮膜形成性ポリマーの種類、使用目的に応じて、ゲル化剤、ゲル化補助剤、pH調整剤、甘味料、酸味料、保存剤、香料、結合剤、増粘剤、酸化マグネシウム以外の着色剤等を適宜含んでいてもよい。
着色剤は通常、染料または顔料のいずれかに区別され、染料は、適用媒体に可溶な着色剤として定義される。すなわち、染料は、ハードカプセルシェル皮膜の皮膜形成性ポリマー溶液に可溶である。一方、顔料は、適用媒体、すなわちハードカプセルシェル皮膜の皮膜形成性ポリマー溶液に不溶性である着色剤として定義される。
ハードカプセルシェル皮膜に含まれる着色剤は、好ましくは白色以外の色を有する。

好ましい実施形態では、酸化マグネシウムは、白色を提供するハードカプセルシェル皮膜中の唯一の成分である。好ましい実施形態では、ハードカプセルシェル皮膜は、白色を提供する他の成分を含まない。

前記ハードカプセルシェル皮膜およびそれによるハードカプセルシェルが、残留水分の形態の水を除き、皮膜形成性ポリマーおよび酸化マグネシウム、ならびに任意選択で可塑剤および任意選択で白色着色剤以外の着色剤のみからなる場合、前記ハードカプセルシェル皮膜、およびそれによるハードカプセルシェルは、好ましくは、それぞれ、乾燥皮膜または乾燥カプセルシェルの重量に基づいて、少なくとも以下の組成を有する。
少なくとも75重量%の皮膜形成性ポリマー、

本発明の皮膜形成性組成物に使用するゲル化補助剤としては、使用するゲル化剤に適したナトリウム塩、カリウム塩、カルシウム塩等がある。これらは適量を単独、あるいは組み合わせて使用しても良い。ゲル化補助剤の量は、最大10重量%、好ましくは最大5重量%であり、重量%は、使用する皮膜形成性ポリマーの重量(乾燥重量)に基づく。
増粘剤としては、アルギン酸、ファーセレラン、タラガム、カラヤガム、スクシノグリカン、キサンタンガム、アラビアガム、カラヤガム、グアーガム、タマリンドシードガム、タラガム、トラガントガム、CMC-Ca、CMC-Na、グルコサミン、プルラン、ポリアクリル酸ナトリウム、カードラン、及び加工デンプンなどがある。増粘剤や結合剤の使用により、ゲル強度が上がり皮膜の強度を補うことが可能である。
本発明のさらなる主題は、シェルが、本明細書で定義されるようなハードカプセルシェル皮膜によって形成され、そのすべての実施形態も含むハードカプセルシェルである。
本発明のさらなる主題は、ハードカプセルシェルを調製するための方法であり、ハードカプセルシェルは、ディップ成形によって調製され、ハードカプセルシェルのシェルは、ハードカプセルシェル皮膜によって形成される。ハードカプセルシェル皮膜には、ここに定義されるカプセルシェル皮膜、およびそのすべての実施形態で示されるハードカプセルシェル皮膜が含まれる。
本発明のさらなる主題は、ハードカプセルシェルを調製するための方法であり、ハードカプセルシェルは、ディップ成形によって調製され、ハードカプセルシェルのシェルは、ハードカプセルシェル皮膜によって形成される。ハードカプセルシェル皮膜には、ここに定義されるカプセルシェル皮膜、およびそのすべての実施形態で示されるハードカプセルシェル皮膜が含まれる。
本発明に係るハードカプセルシェルは、ハードカプセルシェルの調製で知られている任意のディップ成形法によって製造することができる。一例としてゼラチンカプセルの場合を説明する。まず、高温水に20から40重量部のゼラチンを均一に溶解し、ゼラチン色由来の淡黄で透明な浸漬液を調製する。酸化マグネシウム及び場合により可塑剤は、分散液と水溶液の状態で各々を浸漬液に加える。これを粘度調整したのちに、ステンレス製の成型ピンを浸漬液に浸し、続いて引き上げる。引き上げたピン上に付着する浸漬液を直ちに冷却・ゲル化させてカプセルに適した皮膜厚みを確保する。これを乾燥し、不要な部分をカットしたボディーとキャップ一対を嵌合して製造する。
乾燥条件としては、60℃オーブンで15分の高温乾燥、60℃オーブンで30分の高温乾燥、あるいは室温での自然乾燥などが挙げられる。
ハードカプセルシェルは、一以上のポリマー皮膜の層、好ましくは一以上の皮膜形成性ポリマーの皮膜の層を含むことができる。このようなポリマー皮膜の層を、本明細書では単に「層」と呼ぶ。 本発明の意味における一層、および複数の層の定義に関して、各々の層は、別の層を作製するプロセスステップとは別の独自のプロセスステップで作製される。
一つの層を調製するための典型的なプロセスステップは、ディップ成形、バンディング、またはスプレーコーティングなどのコーティングであり得る。
一つの層のみを有するハードカプセルシェルの場合、この一つの層は、本明細書に記載されたハードカプセルシェル皮膜である。これは、ハードカプセルシェルにはそれ以上の層がないことを意味する。 これは、たとえば、バンドやコーティングが施されておらず、その一つの層のみであることを意味する。
好ましくは、一層のみのハードカプセルシェルの場合、この一層が前記ハードカプセルシェル皮膜であり、この一層がディップ成形により形成される。

二層以上のハードカプセルシェルの場合、好ましくは、ハードカプセルシェルは二、三、四または五層を有する。 二層以上の場合、層の組成は同一であっても異なっていてもよいが、好ましくは、互いに接触する層の組成は異なる。 特定の実施形態においては、すべての層が互いに異なる組成を有する。
二層以上の場合、少なくとも最内層はディップ成形により形成することが好ましい。

本発明によれば、従来から医薬分野や食品分野で使用されている酸化マグネシウムという、健康上害のない成分を用い、発がん性が懸念される酸化チタンを使用することなく、ハードカプセルの内容物の遮光を行うことができる。酸化マグネシウムは酸化チタンと遜色のない遮光性を有するため、通常の白色化にも、光に不安定な医薬や食品の製剤化にも好適に使用することができる。
また、本発明の遮光性ハードカプセルシェルは酸化マグネシウムを含有していても溶解性が低下することなく、かつ長期間にわたって安定な遮光性を維持することができ、さらに染料または顔料のような有彩色の着色剤と共に使用すると不透明な有彩色に着色された皮膜を与えることができる。
したがって、特に健康食品市場向けには、例えばクチナシ等に代表される天然系色素と組み合わせることによって、安心安全な美しい着色遮光カプセルシェルを提供できる。

(H) 酸化マグネシウムが重質酸化マグネシウムである(A)~(G)のいずれか1項に記載のハードカプセルシェル皮膜。
(I) 酸化マグネシウムの含有量が皮膜成形性ポリマーの乾燥重量の0.25~20重量%である(A)~(H)のいずれか1項に記載のハードカプセルシェル皮膜。
(J) 酸化マグネシウムの含有量が皮膜成形性ポリマーの乾燥重量の2~10重量%である、(A)~(I)のいずれか1項に記載のハードカプセルシェル皮膜。
(K) 酸化マグネシウムの含有量が皮膜成形性ポリマーの乾燥重量の5~8重量%である、(A)~(J)のいずれか1項に記載のハードカプセルシェル皮膜。
(L)皮膜形成性ポリマーは、ゼラチン、ポリグルタミン酸、セルロース誘導体、プルラン、スターチ、デキストリン、寒天、キトサン、酸処理破砕酵母、ポリビニルアルコール(PVA)、PVA-メチルメタクリル酸共重合体、ポリビニルピロリドン(PVP)からなる群から選択される(A)~(K)のいずれか1項に記載のハードカプセルシェル皮膜。
(M)さらに可塑剤を含む(A)~(L)のいずれか1項に記載のハードカプセルシェル皮膜。
(N) 可塑剤は、ソルビタン脂肪酸エステル、クエン酸トリエチル、グリセリン、ソルビトール、マンニトール、トレハロース、植物油、中鎖脂肪酸トリグリセライド、トリアセチン、フタル酸エステル類、フィトステロール、プロピレングリコール、ポリソルベート、ポリエチレングリコールからなる群から選択される(M)に記載のハードカプセルシェル皮膜。
(O) 可塑剤の含有量は皮膜成形性ポリマーの乾燥重量の0.01~10重量%である(M)または(N)のハードカプセルシェル皮膜。
(P) 可塑剤がソルビタン脂肪酸エステルである(N)のハードカプセルシェル皮膜。
(Q) 可塑剤がソルビタンモノラウリン酸エステルである、(P)に記載のハードカプセルシェル皮膜。
(R) 白色以外の色の着色剤を加えて着色した(A)~(Q)のいずれか1項に記載のハードカプセルシェル皮膜。
(S)ハードカプセルシェルであってシェルが(A~(R)のいずれか一項に記載のハードカプセルシャル皮膜により形成されるハードカプセルシェル。
(T)(S)に記載されたハードカプセルシェルの製造方法であって、前記ハードカプセルシェルはディップ成形により製造される、前記製造方法。
(A)酸化マグネシウムの粒度の分析
IKA社製T50 ULTRA TURRAXを用いて、酸化マグネシウムをソルビタンモノラウリン酸エステル(SML)とともにイオン交換水中で最高速度6600rpmで15分間分散させ、酸化マグネシウム 40重量%、SML 2.7重量%、イオン交換水 57.3重量%を含む分散液を得た。酸化マグネシウムとして、富田製薬株式会社の「酸化マグネシウム食品グレード」(MS-0)を使用した。この製品の焼成温度は600℃である。
上記で作成した分散液を必要量イオン交換水に滴下して、島津製SALD-2000Jレーザー反射式粒子径分布測定器により波長680nmで分散液の粒度分布を測定した。その結果D10、D50、D90の値はそれぞれ0.801、2.577、5.739 であった。図1から、粒子サイズは50μm未満、45μm未満、40μm未満、35μm未満、30μm未満であることがわかる。

(B)皮膜の伸長率の分析
ヒプロメロース(HPMC2906)の室温水溶液に、室温の40重量%酸化マグネシウム(MS-0)の水分散液を加えてヒプロメロースの(固形分)濃度が20重量%、ヒプロメロース重量に対する酸化マグネシウムの割合が0.5、7.5、15重量%である溶液3種類を作成した。ヒプロメロースは、信越化学工業製のHPMC2906を使用した。上記の液を良く攪拌して脱泡したのち、CAMAG皮膜アプリケーターを使用し、以下の方法で皮膜を作成した。20×20×0.3cmのガラス板を70℃に予熱し、皮膜アプリケーターを使用して、皮膜を乾燥時に100umの厚さに引っ張り、すぐに60℃のオーブンに入れて15分間乾燥させた。それを縦1cm×横10cmの短冊に切り抜き、相対湿度2.5%RHの湿度制御ボックス内に7日保管して水分調整を行った。その後INSTRON社製万能試験機モデル68TM-5により、5mm/minの速度で引っ張り、皮膜が切断されるまでの皮膜の伸長率%を測定した(表3)。酸化マグネシウム含量が増加すると皮膜の伸張率は減少していく。
ヒプロメロース(HPMC2906)の室温水溶液に、室温の40重量%酸化マグネシウム(MS-0)の水分散液を加えてヒプロメロースの(固形分)濃度が20重量%、ヒプロメロース重量に対する酸化マグネシウムの割合が0.5、7.5、15重量%である溶液3種類を作成した。ヒプロメロースは、信越化学工業製のHPMC2906を使用した。上記の液を良く攪拌して脱泡したのち、CAMAG皮膜アプリケーターを使用し、以下の方法で皮膜を作成した。20×20×0.3cmのガラス板を70℃に予熱し、皮膜アプリケーターを使用して、皮膜を乾燥時に100umの厚さに引っ張り、すぐに60℃のオーブンに入れて15分間乾燥させた。それを縦1cm×横10cmの短冊に切り抜き、相対湿度2.5%RHの湿度制御ボックス内に7日保管して水分調整を行った。その後INSTRON社製万能試験機モデル68TM-5により、5mm/minの速度で引っ張り、皮膜が切断されるまでの皮膜の伸長率%を測定した(表3)。酸化マグネシウム含量が増加すると皮膜の伸張率は減少していく。
酸化マグネシウムとしてMS-2を使用し、上記と同様に皮膜の伸長率を測定した。
以下の(C)~(D)および(F)の実施例では、いずれも(B)で使用したヒプロメロースを使用した。
以下の(C)~(E)の実施例では酸化マグネシウムとしてMS-0 およびMS-2を使用し、(F)ではMS-0を使用した。
以下の(C)~(E)の実施例では酸化マグネシウムとしてMS-0 およびMS-2を使用し、(F)ではMS-0を使用した。
(C)可塑剤の作用
40重量%の酸化マグネシウム(MS-0)分散液を用い、ヒプロメロースに対して酸化マグネシウムが0.5、7.5、15重量%の分散液を調製し、ヒプロメロース重量に対してソルビタンモノラウリン酸エステル(SML;CAS1338-39-2)を表2の処方で含む5種類の皮膜を作成し、(B)と同様の方法で乾燥、水分調整した後に万能試験機にて皮膜伸長率を測定した。(表4)
40重量%の酸化マグネシウム(MS-0)分散液を用い、ヒプロメロースに対して酸化マグネシウムが0.5、7.5、15重量%の分散液を調製し、ヒプロメロース重量に対してソルビタンモノラウリン酸エステル(SML;CAS1338-39-2)を表2の処方で含む5種類の皮膜を作成し、(B)と同様の方法で乾燥、水分調整した後に万能試験機にて皮膜伸長率を測定した。(表4)
酸化マグネシウムとしてMS-2を使用し、上記と同様に皮膜の伸長率を測定した。
(D)酸化マグネシウムの遮光効果
ヒプロメロース溶液に、ヒプロメロースの重量に対して酸化マグネシウム(MS-0)を5%、10%、15%、20%を添加し(B)と同様に製造した皮膜と、酸化マグネシウムを含まない皮膜について島津製作所製UV-1900iにて波長190nmから1100nmの光透過率を測定した(図2A、2Bおよび表5)。
同様に、ヒプロメロース溶液に酸化マグネシウムを0.25,0.5、1、2、7.5%および15%分散させ、(B)と同様にして製造した皮膜についても光透過率を測定した(図3、表5)。
ヒプロメロース溶液に、ヒプロメロースの重量に対して酸化マグネシウム(MS-0)を5%、10%、15%、20%を添加し(B)と同様に製造した皮膜と、酸化マグネシウムを含まない皮膜について島津製作所製UV-1900iにて波長190nmから1100nmの光透過率を測定した(図2A、2Bおよび表5)。
同様に、ヒプロメロース溶液に酸化マグネシウムを0.25,0.5、1、2、7.5%および15%分散させ、(B)と同様にして製造した皮膜についても光透過率を測定した(図3、表5)。
酸化マグネシウムとしてMS-2を使用し、上記と同様に光透過率を測定した。
(E)酸化マグネシウムを含むゼラチン皮膜の透過率
ルセロ社製豚皮ゼラチン(酸処理ゼラチン)、及び新田ゼラチン製牛骨ゼラチン(アルカリ処理ゼラチン)を100/0、80/20、60/40、50/50、40/60、0/100%の重量比で30重量%になるように溶かした6種類の50℃ゼラチン水溶液に、(A)で作成した酸化マグネシウム(MS-0)分散液をゼラチン重量に対して10重量%を添加し、良く攪拌して脱泡したのち、皮膜作成機を使って乾燥後の厚みが100μmとなる皮膜を作成した(図4B)。(D)と同じ方法で皮膜の光透過率を測定した(図4A、表6)。
ルセロ社製豚皮ゼラチン(酸処理ゼラチン)、及び新田ゼラチン製牛骨ゼラチン(アルカリ処理ゼラチン)を100/0、80/20、60/40、50/50、40/60、0/100%の重量比で30重量%になるように溶かした6種類の50℃ゼラチン水溶液に、(A)で作成した酸化マグネシウム(MS-0)分散液をゼラチン重量に対して10重量%を添加し、良く攪拌して脱泡したのち、皮膜作成機を使って乾燥後の厚みが100μmとなる皮膜を作成した(図4B)。(D)と同じ方法で皮膜の光透過率を測定した(図4A、表6)。
酸化マグネシウムとしてMS-2を使用し、上記と同様に光透過率を測定した。
(F)カプセルの溶出試験
40重量%酸化マグネシウム水分散液を用い、20重量%のヒプロメロースおよびヒプロメロース重量に対して酸化マグネシウム7.5重量%、SML0.5重量%を含む分散液を調製した。カプスゲル社製ハードカプセル製造機により、80℃に熱したカプセルの金属金型を浸漬してヒプロメロースを熱ゲル化させ、サイズ1号のヒプロメロースカプセルを製造した。このカプセルにアセトアミノフェン粉末280mgを充填し、富山産業製モデルNTR-6300A溶出試験機を用い、37℃のUSP pH1.2溶液、シンカーあり、パドル回転数50rpmの条件で溶出試験を実施した。参照品として、同じ製造条件で酸化マグネシウムを添加しないサイズ1号の20ヒプロメロースカプセルを製造した。カプセル6個の溶出率の平均値は次のとおりであった(表7)。
40重量%酸化マグネシウム水分散液を用い、20重量%のヒプロメロースおよびヒプロメロース重量に対して酸化マグネシウム7.5重量%、SML0.5重量%を含む分散液を調製した。カプスゲル社製ハードカプセル製造機により、80℃に熱したカプセルの金属金型を浸漬してヒプロメロースを熱ゲル化させ、サイズ1号のヒプロメロースカプセルを製造した。このカプセルにアセトアミノフェン粉末280mgを充填し、富山産業製モデルNTR-6300A溶出試験機を用い、37℃のUSP pH1.2溶液、シンカーあり、パドル回転数50rpmの条件で溶出試験を実施した。参照品として、同じ製造条件で酸化マグネシウムを添加しないサイズ1号の20ヒプロメロースカプセルを製造した。カプセル6個の溶出率の平均値は次のとおりであった(表7)。
本発明によれば、健康上の問題のあるとされる酸化チタンではなく、従来から医薬品として安全に使用されている酸化マグネシウムを遮光剤として使用することによりカプセルシェルに遮光性を付与することができる。本発明により得られた遮光性カプセルは溶解性が保持され、長期間にわたって安定な遮光性を維持することができ、色素と共に使用すると不透明な着色皮膜を与えることができる。したがって、特に医薬、動物薬、農薬、化粧品、健康食品などの内容物を充填するカプセルに対し安全に使用でき、非常に有用である。
Claims (19)
- ハードカプセルシェル皮膜からなるハードカプセルシェルであって、
ハードカプセルシェル皮膜は皮膜形成性ポリマーおよび酸化マグネシウムを含み、白色着色剤の酸化チタンを含まないハードカプセルシェル皮膜であって、
ハードカプセルシェルは一層のみを有し、この一層が前記ハードカプセルシェル皮膜である、または
ハードカプセルシェルは一層より多い層を有し、最外層とは異なる少なくとも一層は前記ハードカプセルシェル皮膜である
前記ハードカプセルシェル。 - 前記ハードカプセルシェル皮膜は、白色着色剤の炭酸カルシウムを含まない、請求項1に記載のハードカプセルシェル。
- 前記ハードカプセルシェル皮膜は、酸化マグネシウム以外の白色着色剤を含まない、請求項1または2に記載のハードカプセルシェル。
- 酸化マグネシウムの粒子径が50μm以下である請求項1~3のいずれか1項に記載のハードカプセルシェル。
- 酸化マグネシウムが、0.1~2.0μmのD10値を有する粒子サイズ分布を有する、請求項1~4のいずれか1項に記載のハードカプセルシェル。
- 酸化マグネシウムが、4~45μmのD90値を有する粒子サイズ分布を有する、請求項1~5のいずれか1項に記載のハードカプセルシェル。
- 皮膜形成性ポリマーの含有量が、カプセルシェル皮膜の乾燥重量に基づいて少なくとも75重量%である、請求項1~6のいずれか1項に記載のハードカプセルシェル。
- 酸化マグネシウムが重質酸化マグネシウムである請求項1~7のいずれか1項に記載のハードカプセルシェル。
- 酸化マグネシウムの含有量が皮膜成形性ポリマー重量の0.25~20重量%である請求項1~8のいずれか1項に記載のハードカプセルシェル。
- 酸化マグネシウムの含有量が皮膜成形性ポリマーの乾燥重量の2~10重量%である、請求項1~9のいずれか1項に記載のハードカプセルシェル。
- 酸化マグネシウムの含有量が皮膜成形性ポリマーの乾燥重量の5~8重量%である、請求項1~10のいずれか1項に記載のハードカプセルシェル。
- 皮膜形成性ポリマーは、ゼラチン、ポリグルタミン酸、セルロース誘導体、プルラン、スターチ、デキストリン、寒天、キトサン、酸処理破砕酵母、ポリビニルアルコール(PVA)、PVA-メチルメタクリル酸共重合体、ポリビニルピロリドン(PVP)からなる群から選択される請求項1~11のいずれか1項に記載のハードカプセルシェル。
- さらに可塑剤を含む請求項1~12のいずれか1項に記載のハードカプセルシェル。
- 可塑剤は、キシリトール、フルクトース、ソルビタン脂肪酸エステル、クエン酸トリエチル、グリセリン、ソルビトール、マンニトール、トレハロース、植物油、中鎖脂肪酸トリグリセライド、トリアセチン、フタル酸エステル類、フィトステロール、プロピレングリコール、ポリソルベート、ポリエチレングリコールからなる群から選択される請求項13に記載のハードカプセルシェル。
- 可塑剤の含有量は皮膜成形性ポリマーの乾燥重量の0.01~10重量%である請求項13または14のハードカプセルシェル。
- 可塑剤がソルビタン脂肪酸エステルである請求項14のハードカプセルシェル。
- 可塑剤がソルビタンモノラウリン酸エステルである、請求項16に記載のハードカプセルシェル。
- 白色以外の色の着色剤を加えて着色した請求項1~17のいずれか1項に記載のハードカプセルシェル。
- 請求項1に記載のハードカプセルシェルの製造方法であって、皮膜形成性ポリマーおよび酸化マグネシウムを含むハードカプセルシェル皮膜がディップ成形により製造される、前記ハードカプセルシェルの製造方法。
