Abstract
CCN2の発現を促進させるための組成物が開示される。この組成物は、緑茶エキスを含んでなる。
Description
本特許出願は、先に出願された日本国における特許出願である特願2022-146409号(出願日:2022年9月14日)に基づく優先権の主張を伴うものである。この先の特許出願における全開示内容は、引用することにより本明細書の一部とされる。
本発明は、緑茶エキスを含んでなる、CCN2の発現を促進するための組成物に関する。
CCNファミリーは、基本的な生物学的プロセス(例えば、細胞の増殖、接着、遊走、分化、創傷治癒、血管新生等)に関与するタンパク質である。このCCNファミリーには6種の異なるサブタイプ(CCN1~6)が存在することが知られており、各サブタイプの生理的意義や疾患との関連について様々な研究がなされている。
その中でもCCN2は、結合組織成長因子(connective tissue growth factor:CTGF)として知られる因子であり、例えば、線維芽細胞の増殖促進、細胞外基質産生促進、結合組織増殖促進、軟骨細胞の増殖、細胞分化の促進、創傷治癒、および線維性疾患に関与することが報告されている(特許文献1)。また、この他にも、CCN2は腎疾患や癌に関与することも報告されている(特許文献2)。したがって、CCN2を適切に制御することにより、様々な疾患の治療や身体機能の改善につながることが期待される。
このようなCCN2を制御する例として、例えば、チョウマメの抽出物、およびニオイタコノキの抽出物が、ヒト皮膚線維芽細胞において、CCN1およびCCN2の発現を促進させることが報告されている(特許文献1)。また、CCN3遺伝子発現またはCCN3活性レベルを調節することにより、CCN2の活性を調節する方法も報告されている(特許文献2)。
本発明は、CCN2の発現を促進させるための組成物を提供する。
本発明者らは、緑茶エキスがCCN2の発現を促進させることを見出した。本発明はこれらの知見に基づくものである。

CCN2は、結合組織成長因子(connective tissue growth factor:CTGF)として知られる因子であり、したがって、本発明の、CCN2の発現を促進させるための組成物は、特に限定されるわけではないが、結合組織に直接的または間接的に作用してもよい。したがって、本発明の組成物は、結合組織に作用する薬剤として用いることもできる。このような薬剤は、特に限定されるわけではないが、美肌効果(例えば、ハリがある肌の維持、しわ・たるみの予防・改善、血管安定化等)、アトピー性皮膚炎の改善、創傷治癒促進、発毛促進、軟骨再生、軟骨の減少が原因で発症する疾患(例えば、変形性関節症、関節リウマチ等)の改善等の様々な効果が期待できる。このような薬剤は、医師等により行われる治療目的で用いてもよく、美容目的で用いてもよく、保健機能食品等の食品として用いてもよい。
本発明の別の態様によれば、クエン酸回路の機能を活性化するための組成物が提供される。
一般に、クエン酸回路は、トリカルボン酸回路、TCA回路、クレブス回路等ともよばれ、ほとんどの生物の酸化的代謝に共通する経路である。本発明におけるクエン酸回路の機能の活性化とは、クエン酸回路全体または一部の活性を高める作用を意味し、このようなクエン酸回路の機能の活性化は、特に限定されるわけではないが、例えば、クエン酸回路に直接的または間接的に関与する酵素を活性化させることにより達成されてもよい。本発明におけるクエン酸回路の機能の活性化は、単位時間当たりに生成される生成物の量が増加した場合のすべてを含む。このような生成物としては、特に限定されるわけではないが、好ましくは、クエン酸、イソクエン酸、コハク酸、フマル酸、リンゴ酸、NADH、AMP、ATPから選択される1種以上である。本発明の特に好ましい態様によれば、このような生成物はATPである。
また、本発明の別の態様によれば、ATPの産生を促進するための組成物が提供される。本発明におけるATPの産生の促進は、特に限定されるわけではないが、例えば、直接的または間接的な、解糖系の機能の活性化、クエン酸回路の機能の活性化、ミトコンドリア内の電子伝達系に共役して起こる、酸化的リン酸化反応の活性化等により達成され得るがこれに限定されない。本発明におけるATPの産生の促進は、単位時間当たりにこれらが生成される量が増加した場合のすべてを含む。
また、本発明の別の態様によれば、ミオイノシトール1-リン酸および/またはミオイノシトール3-リン酸の産生を促進するための組成物が提供される。
一般に、リン脂質(グリセロリン脂質ともいわれる)は、自然界に遍在し、細胞膜の脂質二重層の重要な成分であり、代謝および細胞シグナル伝達にも関与している。リン脂質としては、具体的には、例えば、ホスファチジルコリン(PC)、ホスファチジルエタノールアミン(PE)、ホスホイノシチド(例えば、イノシトール1-リン酸(PI)、ホスファチジルイノシトール2-リン酸(PIP2)、イノシトール3-リン酸(IP3))およびスフィンゴミエリン(SM)等がよく知られている。
一般に、イノシトールは、ミオイノシトール、D-キロイノシトール、L-キロイノシトール、ムコイノシトール、シロイノシトール、アロイノシトール、エピイノシトール、シスイノシトールおよびネオイノシトールが存在することが知られており、特に、ミオイノシトールは生物活性を有することから、生体において重要な役割を担っている。
本発明におけるミオイノシトール1-リン酸およびミオイノシトール3-リン酸は細胞膜において、その産生が促進されていてもよいがこれに限定されない。本発明におけるミオイノシトール1-リン酸および/またはミオイノシトール3-リン酸の産生の促進は、単位時間当たりにこれらが生成される量が増加した場合のすべてを含む。
また、本発明の別の態様によれば、ヒドロキシプロリンおよび/またはNアセチルグルコサミンの産生を促進するための組成物が提供される。
一般に、細胞外マトリックスとは細胞の外に存在する超分子構造体であり、この細胞外マトリックスは、例えば、コラーゲン、プロテオグリカン、フィブロネクチン、ヒアルロン酸、テネイシン、エンタクチン、エラスチン、フィブリリン、ラミニン等の成分により構成されている。本発明におけるヒドロキシプロリンおよびNアセチルグルコサミンは細胞外マトリックスにおいて、その産生が促進されていてもよいが、これに限定されない。本発明におけるヒドロキシプロリンおよび/またはNアセチルグルコサミンの産生の促進は、単位時間当たりにこれらが生成される量が増加した場合のすべてを含む。
また、本発明の別の態様によれば、ヒドロキシプロリンの産生を促進するための、N-アセチルグルコサミン6-リン酸の産生を促進するための、ミオイノシトール1-リン酸の産生を促進するための、またはミオイノシトール3-リン酸の産生を促進するための組成物が提供され、該組成物は皮膚のハリを改善するために用いてもよい。本発明のさらに別の態様によれば、皮膚のハリを改善するための組成物が提供される。
本発明におけるハリとは、皮膚、特に限定されるわけではないが、好ましくは、顔の皮膚における柔らかさや弾性を意味する。皮膚における弾力は、例えば、キュートメーター、ダーマトルクメーター等により計測することができ、この計測結果に基づいて皮膚のハリを評価してもよい。皮膚のハリは、真皮層におけるコラーゲンやエラスチン等の弾性線維量に影響され得るが、これに限定されない。
一般に、CCN2は、結合組織成長因子(connective tissue growth factor:CTGF)としても知られているタンパク質で、CCNファミリーに属する分泌性の増殖促進因子である。CCNファミリーの生物学的機能としては、細胞の増殖・分化・遊走・接着および細胞外マトリックス形成の刺激や、血管形成への関与等が挙げられる。CCN2は細胞表面のアグリカン、フィブロネクチン、インテグリンと結合可能であり、マトリックスメタロプロテアーゼと協働して、細胞外マトリックスタンパク質の発現を促進し、細胞接着活性を調節するほか、血管新生因子としても働いていることが知られている。なお、本発明におけるCCN2には、その機能を損なわない範囲において、アミノ酸配列の一部に欠失、置換、挿入等の変異を有するものも含まれる。
本発明によれば、CCN2の発現を促進させるための組成物が提供される。CCN2の発現の促進は、CCN2遺伝子の発現の促進、またはCCN2タンパク質の発現の促進のいずれか、またはこの両方であってもよい。
本発明の一つの実施態様によれば、本発明の組成物は、CCN2遺伝子の発現を促進する能力を有していてもよい。このような能力としては、特に限定されるわけではないが、例えば、CCN2遺伝子のmRNAへの転写開始を促進する能力、CCN2遺伝子のmRNAの発現を促進する能力、CCN2遺伝子のmRNAの分解を阻害する能力等が挙げられる。
本発明の一つの実施態様によれば、本発明の組成物は、CCN2タンパク質の発現を促進する能力を有していてもよい。このような能力としては、特に限定されるわけではないが、例えば、CCN2遺伝子のmRNAからタンパク質への翻訳を促進する能力、CCN2タンパク質の細胞外分泌を促進する能力、シグナル伝達機能を促進する能力等が挙げられる。
本発明における緑茶エキスは、茶科チァグァに属する常緑樹である茶木(カメリア・シネンシス;Camellia sinensis)から抽出したもの、またはバチルス・サブチリス(Bacillus subtilis spp.)を接種し醗酵させた茶葉等から抽出したものを含んでなり、前記抽出したものを特定の溶媒で分画して得た分画物を含んでいてもよい。本発明における緑茶エキスは、本発明の目的効果を有するエキスが得られるのであれば、抽出方法、抽出溶媒、抽出された成分または抽出物の形態を問わない。
本発明における緑茶エキスの抽出原料としては、特に限定されるわけではないが、茶木の葉、花、幹、実、根、幹、根の芯材等が挙げられ、好ましくは、葉である。
本発明における緑茶エキスの形態は、特に限定されるわけではないが、好ましくは、粉末状である。
本発明における緑茶エキスの抽出または分画には、水、有機溶媒、またはこれらの混合溶媒を用いてよい。このような有機溶媒としては、特に限定されるわけではないが、例えば、アルコール、イソプロパノール、アセトン、ヘキサン、エチルアセテート、二酸化炭素、またはこれらの二種以上の混合溶媒等が挙げられるが、緑茶の有効成分を本発明の目的効果を有する程度に残存させることが可能な条件下、室温でまたは加温して抽出または分画を行ってもよい。前記アルコールは、特に限定されるわけではないが、C1~C5の低級アルコールであってよい。抽出または分画の回数や方法は、本発明の目的効果を有するエキスが得られるのであれば、特に限定されるわけではないが、例えば、冷浸抽出、超音波抽出、還流冷却抽出、熱水抽出等の方法を用いてもよい。
本発明における緑茶エキスが商業的に販売されている場合、本発明の目的効果を有するエキスが得られるのであれば、それを使用してもよい。
本発明の組成物に含まれる緑茶エキスの配合量は、用途に応じて適宜決定できる。
本発明の別の態様によれば、CCN2発現促進剤を含んでなる組成物が提供され、該組成物は、ヒドロキシプロリンおよび/またはNアセチルグルコサミンの産生を促進するための、クエン酸回路の機能を活性化するための、ATPの産生を促進するための、または皮膚のハリを改善するための組成物である。また、本発明によれば、治療上有効な量のCCN2発現促進剤を含んでなる組成物を、それを必要とする被験体に投与することを含んでなる、ヒドロキシプロリンおよび/またはNアセチルグルコサミンの産生を促進するための、クエン酸回路の機能を活性化するための、ATPの産生を促進するための、または皮膚のハリを改善するための方法が提供される。さらに、本発明によれば、ヒドロキシプロリンおよび/またはNアセチルグルコサミンの産生の促進、クエン酸回路の機能の活性化、ATPの産生の促進、または皮膚のハリの改善を目的とする薬剤の製造における、CCN2発現促進剤の使用が提供される。さらに、本発明によれば、CCN2発現促進剤を含んでなる組成物の、ヒドロキシプロリンおよび/またはNアセチルグルコサミンの産生を促進するための、クエン酸回路の機能を活性化するための、ATPの産生を促進するための、または皮膚のハリを改善するための使用が提供される。さらに、本発明によれば、ヒドロキシプロリンおよび/またはNアセチルグルコサミンの産生を促進するための、クエン酸回路の機能を活性化するための、ATPの産生を促進するための、または皮膚のハリを改善するためのCCN2発現促進剤が提供される。
本発明におけるCCN2発現促進剤は、有効成分としてCCN2の発現を促進するものであれば特に限定されず、CCN2の発現の促進は、CCN2遺伝子の発現の促進、またはCCN2タンパク質の発現の促進のいずれか、またはこの両方であってもよい。

本発明における表皮角化細胞としては、特に限定されるわけではないが、例えば、生体から取得された細胞、その継代された細胞、または株化された細胞を使用してもよく、必要に応じてこれらの細胞を分化させて使用してもよい。本発明における表皮角化細胞としては、特に限定されるわけではないが、具体的には、正常ヒト新生児表皮角化細胞(NHEK)等が挙げられる。
本発明における表皮角化細胞は、いかなる動植物に由来する細胞であってもよく、限定されるわけではないが、好ましくは動物細胞であり、好ましくはヒト細胞である。
本発明の組成物は、経口用組成物、局所用組成物のいずれであってもよいが、好ましくは、局所用組成物である。
本発明の組成物を経口用組成物とする場合、経口用組成物に配合される公知の成分(添加剤)をさらに配合することができる。このような成分としては、特に限定されるわけではないが、例えば、乳化剤、水和剤、溶媒、エモリエント、安定剤、増粘剤、保存剤、滑沢剤、キレート剤、充填剤、賦形剤、粉末、芳香剤、香料、吸収剤、染料、乳白剤、抗酸化剤、防腐剤、ビタミン、アミノ酸、栄養剤、鉱物(電解質)、合成風味剤および天然風味剤等の風味剤、ペクチン酸及びその塩、アルギン酸及びその塩、有機酸、pH調節剤、グリセリン、アルコール、炭酸飲料に用いられる炭酸化剤等が挙げられる。
本発明における経口用組成物は、液体、半固体、固体のいずれであってもよく、特に限定されるわけではないが、好ましくは、錠剤、顆粒剤、丸剤、粉末剤、ドリンク剤といった液剤、キャラメル、またはゲルの形態である。
本発明の組成物を局所用組成物とする場合、局所用組成物に配合される公知の成分(添加剤)をさらに配合することができる。このような成分としては、限定されるわけではないが、例えば、乳化剤、水和剤、溶媒、エモリエント、安定剤、増粘剤、保存剤、滑沢剤、キレート剤、充填剤、賦形剤、粉末、芳香剤、香料、吸収剤、染料、乳白剤、抗酸化剤、ビタミン、アミノ酸等が挙げられる。
本発明における局所用組成物は、液体、半固体、固体のいずれであってもよく、限定されるわけではないが、好ましくは、溶液、懸濁液、分散液、エマルジョン、クリーム、軟膏、ゲル、ローション、フォーム、ペースト、またはスプレーの形態である。
本発明の組成物は、特に限定されるわけではないが、医薬組成物、化粧用組成物、食品組成物のいずれかとしてもよく、好ましくは化粧用組成物である。本発明における化粧用組成物とは、化粧をすることを目的に使用される組成物のことを意味し、経口用組成物、局所用組成物のいずれであってもよい。
本発明における食品組成物は、栄養機能食品、特定保健用食品、機能性表示食品等の保健機能食品であってもよく、また治療食(すなわち、治療の目的を果たすもの、または、医師が食事箋を出し、それに従い栄養士等が作成した献立に基づいて調理されたもの)、食事療法食、介護食等として、製造されてもよい。
以下の例に基づいて本発明を具体的に説明するが、本発明はこれらの例に限定されるものではない。含有量は特記しない限り、質量%で示す。
CCN2が、血管周皮細胞におけるクエン酸回路および細胞外マトリックス等の代謝物に与える影響の評価
(試料の調製)
市販されているリコンビナントCCN2タンパク質(CTGF human、Sigma-Aldrich社製)を滅菌水にて、500μg/mlになるよう希釈し、試料を調製した。

(血管周皮細胞の培養)
市販されているヒト胎盤由来血管周皮細胞(hPC-PL、PromoCell社製)を3.3×104cells/シャーレとなるように100mmシャーレに接種し、血管周皮細胞用培地(Pericyte Growth Medium 2、PromoCell社製)中で37℃、5%のCO2雰囲気下でセミコンフルエントに達するまで培養した。
市販されているヒト胎盤由来血管周皮細胞(hPC-PL、PromoCell社製)を3.3×104cells/シャーレとなるように100mmシャーレに接種し、血管周皮細胞用培地(Pericyte Growth Medium 2、PromoCell社製)中で37℃、5%のCO2雰囲気下でセミコンフルエントに達するまで培養した。
(試料の添加)
試料を添加する前日に培地を血管周皮細胞用培地9mLに置換し、37℃、5%のCO2雰囲気下で培養した。その翌日、最終濃度が150ng/mLになるよう血管周皮細胞用培地で希釈した1500ng/mLのリコンビナントCCN2タンパク質を1mL、培地に添加した。なお、コントロールは、リコンビナントCCN2タンパク質を含まない血管周皮細胞用培地1mLとした。
試料を添加する前日に培地を血管周皮細胞用培地9mLに置換し、37℃、5%のCO2雰囲気下で培養した。その翌日、最終濃度が150ng/mLになるよう血管周皮細胞用培地で希釈した1500ng/mLのリコンビナントCCN2タンパク質を1mL、培地に添加した。なお、コントロールは、リコンビナントCCN2タンパク質を含まない血管周皮細胞用培地1mLとした。
(メタボロームサンプルの回収と定量分析)
リコンビナントCCN2タンパク質を添加して6時間後、シャーレから培地を除去し、ヒューマン・メタボローム・テクノロジーズ社のプロトコルに沿ってサンプルを調整した。その後、回収したサンプルをヒューマン・メタボローム・テクノロジーズ社に送付し、メタボローム解析を実施した。得られた結果に対して、ウェルチのt検定による検定(P<0.001の場合、***と記載、P<0.01の場合、**と記載)を行った。
リコンビナントCCN2タンパク質を添加して6時間後、シャーレから培地を除去し、ヒューマン・メタボローム・テクノロジーズ社のプロトコルに沿ってサンプルを調整した。その後、回収したサンプルをヒューマン・メタボローム・テクノロジーズ社に送付し、メタボローム解析を実施した。得られた結果に対して、ウェルチのt検定による検定(P<0.001の場合、***と記載、P<0.01の場合、**と記載)を行った。

結果を図に示す。リコンビナントCCN2タンパク質処理することにより、クエン酸回路およびエネルギーに関連する10種(クエン酸、イソクエン酸、コハク酸、フマル酸、リンゴ酸、NADH、AMP、ATP)、リン脂質(細胞膜)の合成・再合成に関連する2種(ミオイノシトール-1-リン酸、ミオイノシトール-3-リン酸)および細胞外マトリクスに関連する2種(ヒドロキシプロリン、N-アセチルグルコサミン6-リン酸)を含む257種の代謝物が、コントロールと比較して、有意に上昇することが示された。

緑茶エキスが、CCN2遺伝子の発現に与える影響の評価
(表皮角化細胞の培養)
市販されている正常ヒト新生児表皮角化細胞(NHEK(NB)、クラボウ社製)を5×105cells/フラスコとなるようにT75フラスコに接種し、角化細胞用培地(Humedia-KG2、クラボウ社製)中で37℃、5%のCO2雰囲気下でセミコンフルエントに達するまで培養し、種細胞とした。この角化細胞をトリプシン処理により回収し、24ウェルプレートに10×104cells/ウェルもしくは、12×104cells/ウェルとなるように接種し、角化細胞用培地(Humedia-KG2、クラボウ社製)中で37℃、5%のCO2雰囲気下で培養した。翌日、10×104cells/ウェルに接種した細胞は、添加剤フリー角化細胞用培地(Humedia-KB2、クラボウ社製)に培地を置換し、翌日まで培養した。一方、12×104cells/ウェルに接種した細胞は、1.8mMのCaCl2を含む角化細胞用培地に交換し、37℃、5%のCO2雰囲気下で2日間培養し、細胞分化を誘導した。
(表皮角化細胞の培養)
市販されている正常ヒト新生児表皮角化細胞(NHEK(NB)、クラボウ社製)を5×105cells/フラスコとなるようにT75フラスコに接種し、角化細胞用培地(Humedia-KG2、クラボウ社製)中で37℃、5%のCO2雰囲気下でセミコンフルエントに達するまで培養し、種細胞とした。この角化細胞をトリプシン処理により回収し、24ウェルプレートに10×104cells/ウェルもしくは、12×104cells/ウェルとなるように接種し、角化細胞用培地(Humedia-KG2、クラボウ社製)中で37℃、5%のCO2雰囲気下で培養した。翌日、10×104cells/ウェルに接種した細胞は、添加剤フリー角化細胞用培地(Humedia-KB2、クラボウ社製)に培地を置換し、翌日まで培養した。一方、12×104cells/ウェルに接種した細胞は、1.8mMのCaCl2を含む角化細胞用培地に交換し、37℃、5%のCO2雰囲気下で2日間培養し、細胞分化を誘導した。
(試料の添加)
添加剤フリー角化細胞用培地に培地を置換した表皮角化細胞(未分化細胞)、および分化誘導を実施した表皮角化細胞(分化細胞)に、緑茶エキスを1/1000量(v/v)、それぞれ添加剤フリー角化細胞用培地および1.8mMのCaCl2を含む角化細胞用培地培地に加え、培地中濃度が0.1%となるように添加して、37℃、5%のCO2雰囲気下で培養を続けた。未分化細胞および分化細胞のそれぞれについて、緑茶エキス未処理のコントロール群を用意した。
添加剤フリー角化細胞用培地に培地を置換した表皮角化細胞(未分化細胞)、および分化誘導を実施した表皮角化細胞(分化細胞)に、緑茶エキスを1/1000量(v/v)、それぞれ添加剤フリー角化細胞用培地および1.8mMのCaCl2を含む角化細胞用培地培地に加え、培地中濃度が0.1%となるように添加して、37℃、5%のCO2雰囲気下で培養を続けた。未分化細胞および分化細胞のそれぞれについて、緑茶エキス未処理のコントロール群を用意した。
(細胞からのRNAの抽出)
緑茶エキス処理24時間後に培地を除去し、市販のRNA抽出試薬(RNeasy Mini Kit、Qiagen社製)を用いて、細胞の溶解とRNAの抽出を行った。

結果を図13に示す。図13の結果から、緑茶エキスが、未分化および分化表皮角化細胞のいずれにおいてもCCN2遺伝子の発現を有意に上昇させることが示された。

- 緑茶エキスを含んでなる、CCN2の発現を促進するための組成物。
- ヒドロキシプロリンおよび/またはNアセチルグルコサミンの産生を促進するための、請求項1に記載の組成物。
- クエン酸回路の機能を活性化するための、請求項1に記載の組成物。
- ATPの産生を促進するための、請求項1に記載の組成物。
- 皮膚のハリを改善するための、請求項1に記載の組成物。
- 局所用組成物である、請求項1~5のいずれか一項に記載の組成物。
- 溶液、懸濁液、分散液、エマルジョン、クリーム、軟膏、ゲル、ローション、フォーム、ペースト、またはスプレーの形態である、請求項6に記載の組成物。
- 化粧用組成物である、請求項1~5のいずれか一項に記載の組成物。
- ヒドロキシプロリンおよび/またはNアセチルグルコサミンの産生を促進するための、CCN2発現促進剤を含んでなる、組成物。
- クエン酸回路の機能を活性化するための、CCN2発現促進剤を含んでなる、組成物。
- ATPの産生を促進するための、CCN2発現促進剤を含んでなる、組成物。
- 皮膚のハリを改善するための、CCN2発現促進剤を含んでなる、組成物。
- CCN2発現促進剤が緑茶エキスである、請求項9~12のいずれか一項に記載の組成物。
- 局所用組成物である、請求項9~12のいずれか一項に記載の組成物。
- 溶液、懸濁液、分散液、エマルジョン、クリーム、軟膏、ゲル、ローション、フォーム、ペースト、またはスプレーの形態である、請求項14に記載の組成物。
- 化粧用組成物である、請求項9~12のいずれか一項に記載の組成物。
