Abstract
本発明は、血清が低減された培地に添加することで、動物細胞の増殖を促進することができる素材を提供することを目的とする。 特定の分子量分布やNSIを有する植物性蛋白質素材が低濃度の添加で動物細胞の増殖を促進することを見出した。
Description
本発明は、動物細胞増殖促進剤に関する。
動物細胞培養において効率的に細胞を増殖させることで、様々な分野での研究および開発の範囲が広がる。例えば、再生医療や細胞を用いた培養肉の分野において効率よく進められることが期待される。また、生命維持のためのホルモン、抗体、酵素など必要な物質の産生性向上も可能である。
従来の技術としては、ウシや馬などの動物由来の血清や動物血清由来の成分を増殖因子として培地に添加することが操作基準のように汎用されている。
しかしながら、近年注目されている再生医療分野及び細胞を用いた培養肉の研究分野においては、組成分が完全に明らかになっていない動物血清中に含まれる不要物質の安全性や動物個体による品質安定性などの問題が懸念されている。
そこで、近年無血清培地及び化学合成した増殖因子の開発が進められ、動物血清の代わりに培養培地への使用・添加に利用されている。
従来の技術としては、ウシや馬などの動物由来の血清や動物血清由来の成分を増殖因子として培地に添加することが操作基準のように汎用されている。
しかしながら、近年注目されている再生医療分野及び細胞を用いた培養肉の研究分野においては、組成分が完全に明らかになっていない動物血清中に含まれる不要物質の安全性や動物個体による品質安定性などの問題が懸念されている。
そこで、近年無血清培地及び化学合成した増殖因子の開発が進められ、動物血清の代わりに培養培地への使用・添加に利用されている。
動物血清の代わりに培養培地へ利用されている増殖因子となる成分は化学合成であり高価である。可食成分ではないこと、および増殖因子が高価なため製造コストが高くなり将来の食卓に新たな選択肢となりうる培養肉の開発への活用が制限されていることが課題である。また、窒素源としてアミノ酸や低分子のペプチドが培地に添加されているが、これらの成分は細胞の栄養素として細胞増殖に寄与し、低分子成分のみでは血清の持つ機能を全て代替することは困難である。
特許文献1では、大豆蛋白質加水分解物及び酵母抽出物を無血清培地に添加して、細胞増殖率を高めることが提案されている。また、特許文献2ではβ-コングリシニン濃縮物の加水分解物を含む動物細胞培養用培地に関する技術が開示されている。

本発明は、血清が低減された培地に添加することで、動物細胞の増殖を促進することができる素材を提供することを目的とする。
本発明者らは、上記の課題の解決に対し鋭意検討を重ねた。様々な蛋白質素材を検討する中で、従来の大豆蛋白質等とは異なる、特定の分子量分布やNSIを有する蛋白質素材が低濃度の添加で動物細胞の増殖を促進することを見出し、本発明を完成するに至った。
すなわち本発明は、

(動物細胞増殖促進剤)
本発明の動物細胞増殖促進剤は、下記a)~c)の全特徴を有する植物性蛋白質素材を含むことを特徴とする。

c)分子量分布の測定結果で、2,000以上20,000未満の面積比率が30%以上、かつ20,000以上の面積比率が70%以下。
本発明の動物細胞増殖促進剤の固形分中、上記植物性蛋白質素材は好ましくは50質量%以上であり、より好ましくは60質量%以上、70質量%以上、80質量%以上、90質量%以上、95質量%以上、100質量%でありうる。
本発明の動物細胞増殖促進剤は、下記a)~c)の全特徴を有する植物性蛋白質素材を含むことを特徴とする。
a)固形分中の蛋白質含量が70質量%以上、
b)NSIが80以上、
c)分子量分布の測定結果で、2,000以上20,000未満の面積比率が30%以上、かつ20,000以上の面積比率が70%以下。
本発明の動物細胞増殖促進剤の固形分中、上記植物性蛋白質素材は好ましくは50質量%以上であり、より好ましくは60質量%以上、70質量%以上、80質量%以上、90質量%以上、95質量%以上、100質量%でありうる。
(蛋白質素材)
本発明の蛋白質素材は、植物性蛋白質素材、動物性蛋白質素材を用いることができ、植物性蛋白質素材を用いることが好ましい。
本発明の蛋白質素材の概念は、植物性あるいは動物性蛋白質を主成分とし、各種加工食品や飲料に原料として使用されている食品素材である。該植物性蛋白質素材の由来の例として、大豆、エンドウ、緑豆、ルピン豆、ヒヨコ豆、インゲン豆、ヒラ豆、ササゲ等の豆類、ゴマ、キャノーラ種子、ココナッツ種子、アーモンド種子等の種子類、とうもろこし、そば、麦、米などの穀物類、野菜類、果物類などが挙げられる。より具体的な実施形態では、該植物性蛋白質素材は、豆類の蛋白質から調製される。さらに具体的な実施形態では、該植物性蛋白質素材は大豆蛋白質、エンドウ豆蛋白質、緑豆蛋白質又は空豆蛋白質から調製される。さらにより具体的な実施形態では、該植物性蛋白質素材は大豆蛋白質又はエンドウ豆蛋白質から調製される。一例として大豆由来の蛋白質素材の場合、脱脂大豆や丸大豆等の大豆原料からさらに蛋白質を濃縮加工して調製されるものであり、一般には分離大豆蛋白質、濃縮大豆蛋白質や粉末豆乳、あるいはそれらを種々加工したものなどが概念的に包含される。
また、動物性蛋白質素材としては、例えば、カゼイン、カゼインナトリウム、ホエー蛋白等の乳蛋白、卵白等が挙げられる。
植物性蛋白質素材と動物性蛋白質素材は併用して使用することができる。併用する場合は、配合比率は特に限定されないが、植物性蛋白質素材:動物性蛋白質素材=99:1~1:99である。
本発明の蛋白質素材は、植物性蛋白質素材、動物性蛋白質素材を用いることができ、植物性蛋白質素材を用いることが好ましい。

また、動物性蛋白質素材としては、例えば、カゼイン、カゼインナトリウム、ホエー蛋白等の乳蛋白、卵白等が挙げられる。
植物性蛋白質素材と動物性蛋白質素材は併用して使用することができる。併用する場合は、配合比率は特に限定されないが、植物性蛋白質素材:動物性蛋白質素材=99:1~1:99である。
本発明の蛋白質素材について、植物性蛋白質素材を例として、さらに説明する。
a)蛋白質純度
本態様の動物細胞培養用培地に用いられる植物性蛋白質素材は固形分中の蛋白質含量が70質量%以上、例えば、80質量%以上、85質量%以上、又は90質量%以上である。上記範囲に含まれる植物性蛋白質素材の原料としては、分離蛋白質が好ましく、例えば大豆由来の蛋白質素材から調製する場合、分離大豆蛋白質などが挙げられる。
本態様の動物細胞培養用培地に用いられる植物性蛋白質素材は固形分中の蛋白質含量が70質量%以上、例えば、80質量%以上、85質量%以上、又は90質量%以上である。上記範囲に含まれる植物性蛋白質素材の原料としては、分離蛋白質が好ましく、例えば大豆由来の蛋白質素材から調製する場合、分離大豆蛋白質などが挙げられる。
<蛋白質純度の測定>
蛋白質純度はケルダール法により測定する。具体的には、105℃で12時間乾燥した蛋白質素材質量に対して、ケルダール法により測定した窒素の質量を、乾燥物中の蛋白質含量として「質量%」で表す。なお、窒素換算係数は6.25とする。基本的に、小数点以下第2桁の数値を四捨五入して求められる。
蛋白質純度はケルダール法により測定する。具体的には、105℃で12時間乾燥した蛋白質素材質量に対して、ケルダール法により測定した窒素の質量を、乾燥物中の蛋白質含量として「質量%」で表す。なお、窒素換算係数は6.25とする。基本的に、小数点以下第2桁の数値を四捨五入して求められる。
b)蛋白質のNSI
本態様の動物細胞培養用培地に用いられる植物性蛋白質素材は、蛋白質の溶解性の指標として用いられているNSI(Nitrogen Solubility Index:窒素溶解指数)が80以上のものである。より好ましくはNSIが85以上、90以上、95以上、又は97以上のものを用いることができる。例えば、NSIが高い植物性蛋白質素材として、蛋白質が不溶化される処理、例えば酵素分解処理やミネラルの添加処理等、がされていないもの、あるいはされていたとしてもわずかであるもの、を用いることが好ましい。
植物性蛋白質素材のNSIが高いことは、水への分散性が高いことを示し、本態様の動物細胞培養用培地の分散安定性に寄与し得る。NSIが低すぎると沈殿が生じやすくなり、好ましくない。
なお、NSIは後述する方法に基づき、全窒素量に占める水溶性窒素(粗蛋白)の比率(質量%)で表すものとし、本発明においては後述の方法に準じて測定された値とする。
本態様の動物細胞培養用培地に用いられる植物性蛋白質素材は、蛋白質の溶解性の指標として用いられているNSI(Nitrogen Solubility Index:窒素溶解指数)が80以上のものである。より好ましくはNSIが85以上、90以上、95以上、又は97以上のものを用いることができる。例えば、NSIが高い植物性蛋白質素材として、蛋白質が不溶化される処理、例えば酵素分解処理やミネラルの添加処理等、がされていないもの、あるいはされていたとしてもわずかであるもの、を用いることが好ましい。
植物性蛋白質素材のNSIが高いことは、水への分散性が高いことを示し、本態様の動物細胞培養用培地の分散安定性に寄与し得る。NSIが低すぎると沈殿が生じやすくなり、好ましくない。
なお、NSIは後述する方法に基づき、全窒素量に占める水溶性窒素(粗蛋白)の比率(質量%)で表すものとし、本発明においては後述の方法に準じて測定された値とする。
<NSIの測定法>
試料3gに60mlの水を加え、37℃で1時間プロペラ攪拌した後、1,400×gにて10分間遠心分離し、上澄み液(I)を採取する。次に、残った沈殿に再度水100mlを加え、再度37℃で1時間プロペラ撹拌した後、遠心分離し、上澄み液(II)を採取する。(I)液及び(II)液を合わせ、その混合液に水を加えて250mlとする。これをろ紙(NO.5)にてろ過した後、ろ液中の窒素含量(水溶性窒素)をケルダール法にて測定する。同時に試料中の全窒素量をケルダール法にて測定し、全窒素量に対する水溶性窒素の割合を質量%として表したものをNSIとする。基本的に、小数点以下第2桁の数値を四捨五入して求められる。
試料3gに60mlの水を加え、37℃で1時間プロペラ攪拌した後、1,400×gにて10分間遠心分離し、上澄み液(I)を採取する。次に、残った沈殿に再度水100mlを加え、再度37℃で1時間プロペラ撹拌した後、遠心分離し、上澄み液(II)を採取する。(I)液及び(II)液を合わせ、その混合液に水を加えて250mlとする。これをろ紙(NO.5)にてろ過した後、ろ液中の窒素含量(水溶性窒素)をケルダール法にて測定する。同時に試料中の全窒素量をケルダール法にて測定し、全窒素量に対する水溶性窒素の割合を質量%として表したものをNSIとする。基本的に、小数点以下第2桁の数値を四捨五入して求められる。
c)分子量分布
本態様の動物細胞培養用培地に用いられる植物性蛋白質素材は、ゲルろ過によって分子量を測定した場合に、その分子量分布の面積比率は、2,000Da以上20,000Da未満が30%以上、20,000Da以上が70%以下である。具体的な実施形態では、2,000Da以上20,000Da未満が35%以上、20,000Da以上が65%以下である。
ある特定の実施形態では、植物性蛋白質素材は、ゲルろ過によって分子量を測定した場合に、その分子量分布の面積比率は、2,000Da以上10,000Da未満が10~40%、例えば10~35%、15~35%、10~30%、20~30%であり、10,000Da以上が50~80%、例えば55~75%、60~75%、60~70%、65~75%である。さらに具体的な実施形態では、2,000Da未満の面積比率は15%以下、例えば5%以下、13%以下、9%以下、8%以下、7%以下であり、下限は例えば0%以上、1%以上、1.5%以上、2%以上、3%以上である。
他の特定の実施形態では植物性蛋白質素材は、ゲルろ過によって分子量を測定した場合に、その分子量分布の面積比率は、2,000Da以上20,000Da未満が45~90%、例えば、50~85%、55~80%、55~75%、60~70%である。
さらに具体的な実施形態では、2,000Da未満の面積比率は45%以下、例えば、40%以下、35%以下、33%以下であり、下限は例えば0%以上、1%以上、2%以上、5%以上、10%以上、15%以上である。また、さらにより具体的な実施形態では、10,000Da以上の面積比率は50%未満、例えば、5~45%、10~40%、12~35%である。さらにより具体的な実施形態において、20,000Da以上の面積比率は55%未満、例えば50%以下、40%以下、30%以下、25%以下、20%以下、15%以下である。
本態様の動物細胞培養用培地に用いられる植物性蛋白質素材は、ゲルろ過によって分子量を測定した場合に、その分子量分布の面積比率は、2,000Da以上20,000Da未満が30%以上、20,000Da以上が70%以下である。具体的な実施形態では、2,000Da以上20,000Da未満が35%以上、20,000Da以上が65%以下である。
ある特定の実施形態では、植物性蛋白質素材は、ゲルろ過によって分子量を測定した場合に、その分子量分布の面積比率は、2,000Da以上10,000Da未満が10~40%、例えば10~35%、15~35%、10~30%、20~30%であり、10,000Da以上が50~80%、例えば55~75%、60~75%、60~70%、65~75%である。さらに具体的な実施形態では、2,000Da未満の面積比率は15%以下、例えば5%以下、13%以下、9%以下、8%以下、7%以下であり、下限は例えば0%以上、1%以上、1.5%以上、2%以上、3%以上である。
他の特定の実施形態では植物性蛋白質素材は、ゲルろ過によって分子量を測定した場合に、その分子量分布の面積比率は、2,000Da以上20,000Da未満が45~90%、例えば、50~85%、55~80%、55~75%、60~70%である。
さらに具体的な実施形態では、2,000Da未満の面積比率は45%以下、例えば、40%以下、35%以下、33%以下であり、下限は例えば0%以上、1%以上、2%以上、5%以上、10%以上、15%以上である。また、さらにより具体的な実施形態では、10,000Da以上の面積比率は50%未満、例えば、5~45%、10~40%、12~35%である。さらにより具体的な実施形態において、20,000Da以上の面積比率は55%未満、例えば50%以下、40%以下、30%以下、25%以下、20%以下、15%以下である。
植物性蛋白質素材の分子量分布がこのような範囲にあることは、中程度に低分子化されたものが主成分であることを示す一方、高度に分解された低分子のペプチドは少ないことを示している。なお、分子量分布の測定は、後述する方法に基づくものとする。
<分子量分布>
分子量分布の測定はHPLC(もしくは高速液体クロマトグラフィー)により行った、すなわち、溶離液で蛋白質素材を0.1質量%濃度に調整し、0.2μmフィルターでろ過したものを試料液とした。2種のカラム直列接続によってゲルろ過システムを組み、はじめに表1に記載している分子量マーカーとなる既知の蛋白質等をチャージし、分子量と保持時間の関係において検量線を作成した。次に試料液をチャージし、各分子量画分の含有量比率%をクロマト全体の吸光度のチャート面積に対する、特定の分子量範囲(時間範囲)の面積の割合によって求める(1stカラム:「TSK gel G3000SWXL」(SIGMA-ALDRICH社)、2ndカラム:「TSK gel G2000SW XL」(SIGMA-ALDRICH社)、溶離液:1%SDS+1.17%NaCl+50mMリン酸バッファー(pH7.0)、23℃、流速:0.4ml/分、検出:UV220nm)。

(植物性蛋白質素材の「分解・分子量分布調整処理」)
本態様の動物細胞培養用培地に用いられる植物性蛋白質素材は、蛋白質を分解させる「分解処理」と、蛋白質の分子量分布の調整する「分子量分布調整処理」を組み合わせて適用することにより得られ得る。上記「分解処理」の例として、酵素処理、pH調整処理(例えば、酸処理、アルカリ処理)、加熱処理、冷却処理、高圧処理、有機溶媒処理、ミネラル添加処理、超臨界処理、超音波処理、電気分解処理及びこれらの組み合わせ等が挙げられる。上記「分子量分布調整処理」の例として、ろ過、ゲルろ過、クロマトグラフィー、遠心分離、電気泳動、透析及びこれらの組み合わせ等が挙げられる。「分解処理」と「分子量分布調整処理」の順序及び回数は特に限定されず、「分解処理」を行ってから「分子量分布調整処理」を行ってもよいし、「分子量分布調整処理」を行ってから「分解処理」を行ってもよいし、両処理を同時に行ってもよい。また、例えば2回以上の「分子量分布調整処理」の間に「分解処理」を行う、2回以上の「分解処理」の間に「分子量分布調整処理」を行う、各々複数回の処理を任意の順に行う、等も可能である。なお、「分解処理」によって所望の分子量分布が得られる場合は、「分子量分布調整処理」を行わなくてもよい。これらの処理を組み合わせて、複数回行う際、原料から全ての処理を連続で行ってもよいし、時間をおいてから行ってもよい。例えば、ある処理を経た市販品を原料として他の処理を行ってもよい。なお、上記特性を満たす限り、分子量分布調整処理を経た植物性蛋白質素材と、分子量分布調整処理を経ていない蛋白質を混合して、特定の植物性蛋白質素材としてもよい。この場合、両者の比率(処理を経た蛋白質素材:処理を経ていない蛋白質)は上記特性を満たす範囲で適宜調整可能であるが、質量比で例えば1:99~99:1、例えば50:50~95:5、75:25~90:10等が挙げられる。ある実施形態では、本態様の動物細胞培養用培地に用いられる植物性蛋白質素材は、「分解・分子量分布調整処理」を経た植物性蛋白質素材からなる。
本態様の動物細胞培養用培地に用いられる植物性蛋白質素材は、蛋白質を分解させる「分解処理」と、蛋白質の分子量分布の調整する「分子量分布調整処理」を組み合わせて適用することにより得られ得る。上記「分解処理」の例として、酵素処理、pH調整処理(例えば、酸処理、アルカリ処理)、加熱処理、冷却処理、高圧処理、有機溶媒処理、ミネラル添加処理、超臨界処理、超音波処理、電気分解処理及びこれらの組み合わせ等が挙げられる。上記「分子量分布調整処理」の例として、ろ過、ゲルろ過、クロマトグラフィー、遠心分離、電気泳動、透析及びこれらの組み合わせ等が挙げられる。「分解処理」と「分子量分布調整処理」の順序及び回数は特に限定されず、「分解処理」を行ってから「分子量分布調整処理」を行ってもよいし、「分子量分布調整処理」を行ってから「分解処理」を行ってもよいし、両処理を同時に行ってもよい。また、例えば2回以上の「分子量分布調整処理」の間に「分解処理」を行う、2回以上の「分解処理」の間に「分子量分布調整処理」を行う、各々複数回の処理を任意の順に行う、等も可能である。なお、「分解処理」によって所望の分子量分布が得られる場合は、「分子量分布調整処理」を行わなくてもよい。これらの処理を組み合わせて、複数回行う際、原料から全ての処理を連続で行ってもよいし、時間をおいてから行ってもよい。例えば、ある処理を経た市販品を原料として他の処理を行ってもよい。なお、上記特性を満たす限り、分子量分布調整処理を経た植物性蛋白質素材と、分子量分布調整処理を経ていない蛋白質を混合して、特定の植物性蛋白質素材としてもよい。この場合、両者の比率(処理を経た蛋白質素材:処理を経ていない蛋白質)は上記特性を満たす範囲で適宜調整可能であるが、質量比で例えば1:99~99:1、例えば50:50~95:5、75:25~90:10等が挙げられる。ある実施形態では、本態様の動物細胞培養用培地に用いられる植物性蛋白質素材は、「分解・分子量分布調整処理」を経た植物性蛋白質素材からなる。
蛋白質を分解又は変性させる処理の条件、例えば酵素、pH、有機溶媒、ミネラル等の種類や濃度、温度、圧力、出力強度、電流、時間等は、当業者が適宜設定できる。酵素の場合、使用される酵素の例として、「金属プロテアーゼ」、「酸性プロテアーゼ」、「チオールプロテアーゼ」、「セリンプロテアーゼ」に分類されるプロテアーゼが挙げられる。反応温度は20~80℃、好ましくは40~60℃で反応を行うことができる。pH調整処理の場合、例えばpH2、2.5、3、3.5、4、4.5、5、5.5、6、6.5、7、7.5、8、8.5、9、9.5、10、10.5、11、11.5、12の任意の値を上限、下限とするpH範囲、例えばpH2~12の範囲で処理し得る。酸処理の場合、酸を添加する方法であっても、また、乳酸発酵などの発酵処理を行う方法であってもよい。添加する酸の例として、塩酸、リン酸等の無機酸、酢酸、乳酸、クエン酸、グルコン酸、フィチン酸、ソルビン酸、アジピン酸、コハク酸、酒石酸、フマル酸、リンゴ酸、アスコルビン酸等の有機酸が挙げられる。また、レモンなどの果汁、濃縮果汁、発酵乳、ヨーグルト、醸造酢などの酸を含有する飲食品を用いて酸を添加してもよい。アルカリ処理の場合、水酸化ナトリウム、水酸化カリウム等のアルカリを添加し得る。変性剤処理の場合、塩酸グアニジン、尿素、アルギニン、PEG等の変性剤を添加し得る。加熱又は冷却処理の場合、加熱温度の例として、60℃、70℃、80℃、90℃、100℃、110℃、120℃、125℃、130℃、135℃、140℃、145℃、150℃の任意の温度を上限、下限とする範囲、例えば60℃~150℃が挙げられる。冷却温度の例として、-10℃、-15℃、-20℃、-25℃、-30℃、-35℃、-40℃、-45℃、-50℃、-55℃、-60℃、-65℃、-70℃、-75℃の任意の温度を上限、下限とする範囲、例えば-10℃~-75℃が挙げられる。加熱又は冷却時間の例として、5秒、10秒、30秒、1分、5分、10分、20分、30分、40分、50分、60分、70分、80分、90分、100分、120分、150分、180分、200分の任意の時間を上限、下限とする範囲、例えば5秒間~200分間が挙げられる。高圧処理の場合、圧力の条件の例として、100MPa、200MPa、300MPa、400MPa、500MPa、600MPa、700MPa、800MPa、900MPa、1,000MPaの任意の圧力を上限、下限とする範囲、例えば100MPa~1,000MPaが挙げられる。有機溶媒処理の場合、用いられる溶媒の例として、アルコールやケトン、例えばエタノールやアセトンが挙げられる。ミネラル添加処理の場合、用いられるミネラルの例として、カルシウム、マグネシウムなどの2価金属イオンが挙げられる。超臨界処理の場合、例えば、温度約30℃以上で約7MPa以上の超臨界状態の二酸化炭素を使用して処理できる。超音波処理の場合、例えば100KHz~2MHzの周波数で100~1,000Wの出力で照射して処理し得る。電気分解処理の場合、例えば蛋白質水溶液を100mV~1,000mVの電圧を印加することにより処理し得る。具体的な実施形態において、蛋白質を分解させる処理は、加熱処理、及びそれらの組み合わせから選択される。
蛋白質の分子量分布を調整する処理の条件、例えばろ材の種類、ゲルろ過の担体、遠心分離回転数、電流、時間等は、当業者が適宜設定できる。ろ材の例として、ろ紙、ろ布、ケイ藻土、セラミック、ガラス、メンブラン等が挙げられる。ゲルろ過の担体の例として、デキストラン、アガロース等が挙げられる。遠心分離の条件の例として、1,000~3,000×g、5~20分間等が挙げられる。
なお、従来の市販の大豆蛋白質素材である「フジプロE」、「フジプロCL」、「フジプロAL」、「ニューフジプロ4500」、「プロリーナRD-1」、「プロリーナ900」、「プロリーナHD101R」などは、いずれも上記a)~c)の全特性を満たす植物性蛋白質素材に該当しない。
(動物細胞培養用培地)
本発明の動物細胞培養用培地は、上記動物細胞増殖促進剤を含む。
本発明の動物細胞培養用培地は、血清が低減された培地であることが好ましい。
血清が低減された培地の1つの実施形態としては血清低減培地でありうる。本発明では血清低減培地とは、通常の血清培地よりも血清が低減されたものであり、好ましくは、培地中の血清量が10%未満であり、より好ましくは、9.5%、9%、8%、7%、6%、5%、4%、3%、2%、1%、0.5%、0.2%、0.1%等のものが挙げられる。
また、血清が低減された培地の別の実施形態としては無血清培地でありうる。
本発明の動物細胞培養用培地は、上記動物細胞増殖促進剤を含む。
本発明の動物細胞培養用培地は、血清が低減された培地であることが好ましい。
血清が低減された培地の1つの実施形態としては血清低減培地でありうる。本発明では血清低減培地とは、通常の血清培地よりも血清が低減されたものであり、好ましくは、培地中の血清量が10%未満であり、より好ましくは、9.5%、9%、8%、7%、6%、5%、4%、3%、2%、1%、0.5%、0.2%、0.1%等のものが挙げられる。
また、血清が低減された培地の別の実施形態としては無血清培地でありうる。
培地の種類は特に限定されず市販されている培地を使用することができる。好ましい培地は培養する動物細胞の種類によって異なり、当業者が適宜選択することができる。
動物細胞としては、例えば、線維芽細胞、表皮細胞、乳腺細胞、脂肪細胞、筋芽細胞、平滑筋細胞、内皮細胞、骨芽細胞、肝細胞、血管内皮細胞、または、それらの前駆細胞が挙げられる。また、その他として造血幹細胞、間葉系幹細胞、神経幹細胞などの体性幹細胞、多能性幹細胞、人工多能性幹細胞が挙げられる。
また、抗体、酵素(ウロキナーゼ等)、ホルモン(インシュリン等)、サイトカイン(インターフェロン、インターロイキン、腫瘍壊死因子、コロニー刺激因子、成長因子等)、その他の生理活性蛋白質・ペプチドなどの有用物質を産生する動物細胞が挙げられ、かかる動物細胞としては、皮膚細胞、軟骨細胞、肝細胞、膵臓細胞、腎細胞等の非形質転換細胞や、コードする遺伝子や有用物質の生合成に関与する遺伝子を導入した形質転換細胞が例示される。形質転換細胞の例としては、マウスミエローマ細胞、チャイニーズハムスター卵巣細胞などの抗体産生細胞などが挙げられる。これらの動物細胞を培養して有用物質を効率的に製造することができる。また、皮膚細胞、軟骨細胞、肝細胞、膵臓細胞、ES細胞、iPS細胞などを培養し、効率的に増殖させることにより、これらを再生医療の分野にも利用することができる。
動物細胞の由来としては、好ましくは哺乳動物由来の細胞であり、例えば、マウス、ラット、ウサギ、ヒト、ウマ、ウシ、サル、ブタ、ニワトリ、カモ、イヌ、ヒツジ、ネコ、ヤギなどが挙げられる。
動物細胞としては、例えば、線維芽細胞、表皮細胞、乳腺細胞、脂肪細胞、筋芽細胞、平滑筋細胞、内皮細胞、骨芽細胞、肝細胞、血管内皮細胞、または、それらの前駆細胞が挙げられる。また、その他として造血幹細胞、間葉系幹細胞、神経幹細胞などの体性幹細胞、多能性幹細胞、人工多能性幹細胞が挙げられる。

動物細胞の由来としては、好ましくは哺乳動物由来の細胞であり、例えば、マウス、ラット、ウサギ、ヒト、ウマ、ウシ、サル、ブタ、ニワトリ、カモ、イヌ、ヒツジ、ネコ、ヤギなどが挙げられる。
本発明の動物細胞増殖促進剤の培地中の量は、培養する動物細胞の種類によって異なるが、望ましくは動物細胞培養用培地中、蛋白質素材として0.015質量%以上である。好ましくは0.02質量%以上、0.04質量%以上である。上限は、好ましくは0.5質量%以下であり、より好ましくは0.4質量%以下であり、さらに好ましくは0.3質量%以下、0.2質量%以下、0.15質量%以下、0.1質量%以下とすることもできる。具体例として、0.015~0.5質量%、0.015~0.4質量%、0.015~0.3質量%、0.015~0.2質量%、0.015~0.15質量%、0.015~0.1質量%、0.02~0.5質量%、0.02~0.4質量%、0.02~0.3質量%、0.02~0.2質量%、0.02~0.15質量%、0.02~0.1質量%、0.04~0.5質量%、0.04~0.4質量%、0.04~0.3質量%、0.04~0.2質量%、0.04~0.15質量%、0.04~0.1質量%が挙げられる。本発明の蛋白質素材が上記範囲の量で含有することにより、動物細胞培養の増殖を促進することができる。
特に、本発明の蛋白質素材は上記のように、0.1質量%以下の低い濃度においても、動物細胞の増殖を促進することができる点で優れている。
本発明において、動物細胞の増殖を促進するとは、本発明の蛋白質素材を添加した培地で培養した細胞の増殖が、無添加の培地で培養した細胞の増殖よりも高いことをいう。
具体的には、無添加の培地で培養した細胞増殖能を1としたときに、植物性蛋白質素材を添加した培地で培養した細胞増殖能が、1.07以上であることをいう。好ましくは、1.08以上であり、より好ましくは1.10以上であり、さらに好ましくは1.13以上であり、さらに好ましくは1.15以上である。
なお、細胞増殖の測定方法は後述する。
特に、本発明の蛋白質素材は上記のように、0.1質量%以下の低い濃度においても、動物細胞の増殖を促進することができる点で優れている。
本発明において、動物細胞の増殖を促進するとは、本発明の蛋白質素材を添加した培地で培養した細胞の増殖が、無添加の培地で培養した細胞の増殖よりも高いことをいう。
具体的には、無添加の培地で培養した細胞増殖能を1としたときに、植物性蛋白質素材を添加した培地で培養した細胞増殖能が、1.07以上であることをいう。好ましくは、1.08以上であり、より好ましくは1.10以上であり、さらに好ましくは1.13以上であり、さらに好ましくは1.15以上である。
なお、細胞増殖の測定方法は後述する。
(動物細胞の増殖促進効果)
本発明の動物細胞培養用培地中で動物細胞を培養すると、本発明の蛋白質素材により動物細胞の細胞増殖が促進される。これにより、増殖が促進される動物細胞が培養肉の製造に有用な物質を産生できるものであれば、動物細胞による培養肉の産生も促進され得る。
また、増殖が促進される動物細胞がヒトの生体に有用な物質を産生する種類のものである場合は、動物細胞による該有用物質の産生も促進されうる。動物細胞から産生される各種有用物質としては抗体の他、酵素やホルモン等が挙げられる。
本発明の動物細胞培養用培地中で動物細胞を培養すると、本発明の蛋白質素材により動物細胞の細胞増殖が促進される。これにより、増殖が促進される動物細胞が培養肉の製造に有用な物質を産生できるものであれば、動物細胞による培養肉の産生も促進され得る。
また、増殖が促進される動物細胞がヒトの生体に有用な物質を産生する種類のものである場合は、動物細胞による該有用物質の産生も促進されうる。動物細胞から産生される各種有用物質としては抗体の他、酵素やホルモン等が挙げられる。
以下に実施例を記載することで本発明を説明する。尚、例中の部及び%は特に断らない限り質量基準を意味するものとする。
(植物性蛋白質素材の調製)
以下の植物性蛋白素材を入手、調製した。これら植物性蛋白素材の蛋白質含量は、どれも80質量%以上であった。詳細な分析値を表2に示した。
植物性蛋白質素材A:分離大豆蛋白質の分解・分子量分布調整処理品(不二製油株式会社テスト製造品、原料分離大豆蛋白質:フジプロF(不二製油株式会社市販品))
植物性蛋白質素材B:分離大豆蛋白質の分解・分子量分布調整処理品(不二製油株式会社テスト製造品、原料分離大豆蛋白質:フジプロF(不二製油株式会社市販品))
分離大豆蛋白質加水分解物:不二製油株式会社市販品
以下の植物性蛋白素材を入手、調製した。これら植物性蛋白素材の蛋白質含量は、どれも80質量%以上であった。詳細な分析値を表2に示した。

(実施例1)マウス横紋筋由来筋芽細胞への増殖促進効果
植物性蛋白質素材Aのマウス横紋筋由来筋芽細胞への増殖促進効果を、以下の方法に従い確認した。
植物性蛋白質素材Aのマウス横紋筋由来筋芽細胞への増殖促進効果を、以下の方法に従い確認した。
(細胞の培養方法)
Penicillin-Streptomycin-Amphotericin B Suspensionを100倍希釈となるように添加した5%FBS(ウシ胎児血清)を含む、ダルベッコ改変イーグル培地(以下、DMEMと称する。)を調製した。これを用いてマウス横紋筋由来筋芽細胞株C2C12(以下、C2C12と称する。)を5×104cell/mLに調製した細胞浮遊液を取得した。得られた細胞浮遊液を96well-plateに0.1mL/well、各8wellに添加した。37℃、24時間(CO2濃度=5%)平衡化後、各濃度の植物性蛋白質素材Aを添加した5%FBSを含むDMEMに置換した。これを37℃、48~72時間培養(CO2濃度=5%)した後に、細胞増殖測定を行った。
なお、植物性蛋白質素材を添加していない5%FBSを含むDMEMまたは10%FBSを含むDMEMでの培養も行った。
なお、植物性蛋白質素材Aは5%FBSを含むDMEMに対して0.2g/L(0.02%相当)~1.5g/L(0.15%相当)添加した。

なお、植物性蛋白質素材を添加していない5%FBSを含むDMEMまたは10%FBSを含むDMEMでの培養も行った。

(細胞増殖能の評価方法)
(株)同仁化学研究所製のCell Counting Kit-8を用いて、細胞増殖測定を行った。
すなわち、培養後の各培地に、水溶性テトラゾリウム塩(WST-8)を添加し、37℃、60分間インキュベーション後、吸光度450nmで測定した。
測定原理は、細胞中の脱水酵素により産生されるNADHは電子伝達体を介して水溶性テトラゾリウム塩(WST-8)をオレンジ色の水溶性ホルマザンに還元する。このホルマザンの450nmの吸光度は生細胞数に比例し、生細胞数を計測することができる。
(株)同仁化学研究所製のCell Counting Kit-8を用いて、細胞増殖測定を行った。
すなわち、培養後の各培地に、水溶性テトラゾリウム塩(WST-8)を添加し、37℃、60分間インキュベーション後、吸光度450nmで測定した。
測定原理は、細胞中の脱水酵素により産生されるNADHは電子伝達体を介して水溶性テトラゾリウム塩(WST-8)をオレンジ色の水溶性ホルマザンに還元する。このホルマザンの450nmの吸光度は生細胞数に比例し、生細胞数を計測することができる。
結果を図1に示した。
通常の10%FBSを含むDMEMでの細胞増殖能と比べ、FBSの濃度を半減した5%FBSを含むDMEMでの増殖能が明らかに低下した。一方、5%FBSを含むDMEMに対して0.2g/L(0.02%相当)~1.5g/L(0.15%相当)の植物性蛋白質素材AをDMEMに添加することで、低下した増殖能が有意に回復した。
従って、植物性蛋白質素材Aは血清低減培地において、C2C12の増殖効果があることが確認された。
通常の10%FBSを含むDMEMでの細胞増殖能と比べ、FBSの濃度を半減した5%FBSを含むDMEMでの増殖能が明らかに低下した。一方、5%FBSを含むDMEMに対して0.2g/L(0.02%相当)~1.5g/L(0.15%相当)の植物性蛋白質素材AをDMEMに添加することで、低下した増殖能が有意に回復した。
従って、植物性蛋白質素材Aは血清低減培地において、C2C12の増殖効果があることが確認された。
(実施例2)
植物性蛋白質素材として、植物性蛋白質素材Bを使用し、5%FBSを含むDMEMに対して、1.0g/L添加した以外は実施例1と同様にして細胞を培養し、増殖能を評価した。結果を図2に示した。
植物性蛋白質素材Bにおいても、血清低減培地において、C2C12の増殖効果があることが確認された。
植物性蛋白質素材として、植物性蛋白質素材Bを使用し、5%FBSを含むDMEMに対して、1.0g/L添加した以外は実施例1と同様にして細胞を培養し、増殖能を評価した。結果を図2に示した。
植物性蛋白質素材Bにおいても、血清低減培地において、C2C12の増殖効果があることが確認された。
(実施例3、比較例1)
植物性蛋白質素材として、植物性蛋白質素材Aあるいは分離大豆蛋白質加水分解物を使用し、5%FBSを含むDMEMに対して、0.5g/L添加した以外は実施例1と同様にして細胞を培養し、増殖能を評価した。結果を図3に示した。
植物性蛋白質素材Aにおいて、血清低減培地において、C2C12の増殖効果が高いことが確認されたのに対し(実施例3、図中、蛋白質素材Aと表記。)、分子量分布が本発明で規定する数値を満たさない分離大豆蛋白質加水分解物はC2C12の増殖効果が低く(比較例1、図中、分離大豆蛋白質加水分解物と表記。)、本発明の合格基準を満たさない結果となった。
植物性蛋白質素材として、植物性蛋白質素材Aあるいは分離大豆蛋白質加水分解物を使用し、5%FBSを含むDMEMに対して、0.5g/L添加した以外は実施例1と同様にして細胞を培養し、増殖能を評価した。結果を図3に示した。
植物性蛋白質素材Aにおいて、血清低減培地において、C2C12の増殖効果が高いことが確認されたのに対し(実施例3、図中、蛋白質素材Aと表記。)、分子量分布が本発明で規定する数値を満たさない分離大豆蛋白質加水分解物はC2C12の増殖効果が低く(比較例1、図中、分離大豆蛋白質加水分解物と表記。)、本発明の合格基準を満たさない結果となった。
(実施例4、比較例2)
本発明の植物性蛋白素材の無血清培地における効果を以下の手順で確認した。
本発明の植物性蛋白素材の無血清培地における効果を以下の手順で確認した。
グルタミンを2mMとなるように添加した5 μM/mL Fibronectin(接着因子)を含む無血清培地(BIO-MPM-1、Biological Industries Ltd.社製)を調製した。これを用いてヒト近位尿細管細胞株HK2を1×104cells/mLに調製した細胞浮遊液を取得した。得られた細胞浮遊液を96well-plateに0.1mL/well、各8wellに添加した。37℃、24時間(CO2濃度=5%)平衡化後、各濃度の植物性蛋白質素材Aを添加した無血清培地(BIO-MPM-1)に置換した。これを37℃、48~72時間培養(CO2濃度=5%)した後に、細胞増殖測定を行った。
なお、植物性蛋白質素材AはBIO-MPM-1無血清培地に対して0.125g/L(0.0125%相当)~5g/L(0.5%相当)添加した。また、細胞増殖能の評価を実施例1と同様に行った。評価結果を図4に示した。
植物性蛋白質素材Aの添加量が0.5g/L、1.25g/L、2.5g/L及び5g/L(実施例4)において、無血清培地において、ヒト近位尿細管細胞株HK2の増殖効果が高いことが確認された。
なお、植物性蛋白質素材AはBIO-MPM-1無血清培地に対して0.125g/L(0.0125%相当)~5g/L(0.5%相当)添加した。また、細胞増殖能の評価を実施例1と同様に行った。評価結果を図4に示した。
植物性蛋白質素材Aの添加量が0.5g/L、1.25g/L、2.5g/L及び5g/L(実施例4)において、無血清培地において、ヒト近位尿細管細胞株HK2の増殖効果が高いことが確認された。
Claims (20)
- 下記a)~c)の全特徴を有する蛋白質素材を含む動物細胞増殖促進剤。
a)固形分中の蛋白質含量が70質量%以上、
b)NSIが80以上、
c)分子量分布の測定結果で、2,000Da以上20,000Da未満の面積比率が30%以上、かつ20,000Da以上の面積比率が70%以下。 - 蛋白質素材が、分子量分布の測定結果で、2,000Da以上20,000Da未満の面積比率が45~90%である、請求項1記載の動物細胞増殖促進剤。
- 分子量分布の測定結果で、2,000Da未満の面積比率が45%以下である、請求項2記載の動物細胞増殖促進剤。
- 請求項1記載の動物細胞増殖促進剤を蛋白質素材として、培地中0.015重量%以上含む動物細胞培養用培地。
- 請求項2記載の動物細胞増殖促進剤を蛋白質素材として、培地中0.015重量%以上含む動物細胞培養用培地。
- 請求項3記載の動物細胞増殖促進剤を蛋白質素材として、培地中0.015重量%以上含む動物細胞培養用培地。
- 動物細胞培養用培地が、培地中の血清量が10%未満の血清が低減された培地である、請求項4記載の動物細胞培養用培地。
- 動物細胞培養用培地が、培地中の血清量が10%未満の血清が低減された培地である、請求項5記載の動物細胞培養用培地。

- 血清が低減された動物細胞培養用培地が、無血清の動物細胞培養用培地である、請求項19記載の動物細胞の増殖を促進する方法。
