Description
本発明は、生菌数調整剤に関し、より具体的には、皮膚表面に存在するアクネ菌の生菌数を調整して、皮膚症状を改善する生菌数調整剤に関する。
リタ(Sapindus trifoliatus)の果実からの抽出物を含有し、皮膚状態を正常化または正常性を維持するための皮膚洗浄料が提案されている(特許文献1参照)。
特許文献1に記載の皮膚洗浄料では、アトピー性皮膚炎を改善すべく、黄色ブドウ球菌やアクネ菌の生育を抑制することが検討されている。

本発明では上記事情に鑑み、皮膚表面に存在するアクネ菌の生菌数をより確実に調整して、皮膚症状を改善する生菌数調整剤を提供することとした。

以下、本発明の生菌数調整剤について、好適実施形態に基づいて詳細に説明する。
本発明の生菌数調整剤は、被験者の皮膚表面に存在するアクネ菌の生菌数を調整するのに使用される。この生菌数調整剤は、乳酸桿菌発酵液を含む。本発明者らは、鋭意検討した結果、乳酸桿菌発酵液を含む組成物は、皮膚表面に存在するアクネ菌の生菌数を所定の範囲に調整し得ることを見出し、本発明を完成するに至った。そして、アクネ菌の生菌数を所定の範囲に調整することにより、皮膚症状を改善し得ることも判明した。
本発明の生菌数調整剤は、被験者の皮膚表面に存在するアクネ菌の生菌数を調整するのに使用される。この生菌数調整剤は、乳酸桿菌発酵液を含む。本発明者らは、鋭意検討した結果、乳酸桿菌発酵液を含む組成物は、皮膚表面に存在するアクネ菌の生菌数を所定の範囲に調整し得ることを見出し、本発明を完成するに至った。そして、アクネ菌の生菌数を所定の範囲に調整することにより、皮膚症状を改善し得ることも判明した。
具体的には、アクネ菌の生菌数が相対的に少ない被験者は、皮膚症状として乾燥肌を訴える人が多いが、アクネ菌の生菌数を所定の範囲に調整することにより乾燥肌が改善する。一方、アクネ菌の生菌数が相対的に多い被験者は、皮膚症状としてざ瘡および/または脂性肌を訴える人が多いが、アクネ菌の生菌数を所定の範囲に調整することによりざ瘡および/または脂性肌が改善する。
生菌数調整剤は、皮膚表面に存在する皮膚常在菌に占めるアクネ菌の割合を30%以上50%未満程度に調整することが好ましく、35%以上45%以下に調整することがより好ましい。
言い換えれば、皮膚表面に存在する皮膚常在菌に占めるアクネ菌の割合が30%未満の被験者は、皮膚症状として乾燥肌を訴える傾向が強く、皮膚表面に存在する皮膚常在菌に占めるアクネ菌の割合が70%以上の被験者は、ざ瘡を訴える傾向が強く、60%以上の被験者は、脂性肌を訴える傾向が強い。
言い換えれば、皮膚表面に存在する皮膚常在菌に占めるアクネ菌の割合が30%未満の被験者は、皮膚症状として乾燥肌を訴える傾向が強く、皮膚表面に存在する皮膚常在菌に占めるアクネ菌の割合が70%以上の被験者は、ざ瘡を訴える傾向が強く、60%以上の被験者は、脂性肌を訴える傾向が強い。
この皮膚表面に存在する皮膚常在菌に占めるアクネ菌の割合は、皮膚常在菌を採取するためのシール材を皮膚表面に貼付し、シール材の粘着剤層に転写された皮膚常在菌の種類および数を特定することにより行われる。
なお、皮膚常在菌の種類を特定する方法には、例えば、rDNAの特定領域の塩基配列を次世代シーケンサー(NGS)を用いて解析する方法等を使用することができる。また、皮膚常在菌の数を特定する方法には、例えば、対象となる菌の特定の配列に対するPCRを行い定量する方法(qPCR)等を使用することができる。
なお、皮膚常在菌の種類を特定する方法には、例えば、rDNAの特定領域の塩基配列を次世代シーケンサー(NGS)を用いて解析する方法等を使用することができる。また、皮膚常在菌の数を特定する方法には、例えば、対象となる菌の特定の配列に対するPCRを行い定量する方法(qPCR)等を使用することができる。
ここで、乳酸桿菌とは、真正細菌類でラクトバチルス科に属する細菌である。乳酸桿菌は、その形状が細長く、胞子をつくらず、非運動性のグラム陽性菌である。
かかる乳酸桿菌による発酵液には、アミノ酸をはじめとする多種多様な成分が含まれており、これらの成分が複合的に作用すると考えられる。その結果、アクネ菌が少ない場合は、その増殖を促進し、アクネ菌が多い場合には、その増殖を抑制するように働くようである。

乳酸桿菌発酵液の原液、すなわち乳酸桿菌による発酵に供される原液としては、特に限定されないが、例えば、豆乳、米、大豆、乳(牛乳)、ブドウ果汁、セイヨウナシ果汁、加水分解コラーゲン、センチフォリアバラ花エキス、ハイビスカス花エキス等が挙げられる。なお、乳酸桿菌発酵液は、1種を単独で使用してもよく、2種以上を併用してもよい。
これらの中でも、乳酸桿菌発酵液としては、乳酸桿菌/豆乳発酵液が好ましい。
なお、乳酸桿菌発酵液は、例えば、乳酸桿菌に属する複数種類(15~25種類程度)の菌を使用して、原液を発酵させることにより得られる。

乳酸桿菌発酵液の含有量は、0.1質量%以上20質量%以下であることが好ましく、0.5質量%以上15質量%以下であることがより好ましく、1質量%以上10質量%以下であることがさらに好ましい。かかる含有量で乳酸桿菌発酵液を含む生菌数調整剤を使用すれば、皮膚表面に存在する皮膚常在菌に占めるアクネ菌の割合をより好ましい範囲に調整し易くなる。なお、乳酸桿菌発酵液の含有量を必要以上に多くしても、それに見合う効果の上昇が認められない。また、本発明者らの検討によれば、かかる含有量で乳酸桿菌発酵液を含む生菌数調整剤を使用すれば、アクネ菌のみならず、表皮ブドウ球菌に対しても強い増殖抑制を示すことが判明している。
本発明の生菌数調整剤は、さらに、酵母菌由来成分を含むことが好ましい。この酵母菌由来成分には、多種のアミノ酸およびビタミンが豊富に含まれており、皮膚状態を良好に保持することにより、乳酸桿菌発酵液に含まれる成分が皮膚表面に作用し易くなると考えられる。また、酵母菌由来成分を併用することにより、皮膚の乾燥状態を改善することもできる。
酵母由来成分は、例えば、エキス剤、粉末剤、凍結乾燥剤等として使用することができる。エキス剤は、例えば、酵素分解法、酸分解法等により酵母を処理することにより得られる。
酵母としては、特に限定されないが、例えば、サッカロミセス属、シゾサッカロミセス属、クルイヴェロミセス属、シゾサッカロミセス属、ラカンセア属、ピキア属等に属する酵母を使用することができる。
これらの中でも、サッカロミセス属に属する酵母、特に、サッカロミセス・セレビシアエ、サッカロミセス・ブラウディーを使用することが好ましい。
酵母としては、特に限定されないが、例えば、サッカロミセス属、シゾサッカロミセス属、クルイヴェロミセス属、シゾサッカロミセス属、ラカンセア属、ピキア属等に属する酵母を使用することができる。
これらの中でも、サッカロミセス属に属する酵母、特に、サッカロミセス・セレビシアエ、サッカロミセス・ブラウディーを使用することが好ましい。
酵母菌由来成分の含有量は、0.001質量%以上0.5質量%以下であることが好ましく、0.005質量%以上0.1質量%以下であることがより好ましく、0.01質量%以上0.05質量%以下であることがさらに好ましい。かかる含有量で酵母菌由来成分を含む生菌数調整剤を使用すれば、酵母菌由来成分による作用・効果が十分に発揮される。
本発明の生菌数調整剤は、さらに、穀物発酵液を含むことが好ましい。この穀物発酵液は、保湿作用を有するため、皮膚表面の湿潤状態を高め、乳酸桿菌発酵液に含まれる成分を皮膚表面に保持し易くなると考えられる。よって、乳酸桿菌発酵液による作用・効果をより好適に発揮させることができる。
穀物としては、特に限定されないが、例えば、米(イネ)、コムギ、オオムギ、トウモロコシ、アワ、ヒエ、キビのような禾穀(かこく)類、ダイズ、リョクトウ、インゲンマメ、ラッカセイ、エンドウのような菽穀(しゅくこく)類等が挙げられる。これらの穀物は、1種を単独で使用しても、2種以上を併用してもよい。
これらの中でも、穀物としては、米であることが好ましい。すなわち、穀物発酵液としては、米発酵液が好適である。

シラタマノキ属に属する植物としては、特に限定されないが、例えば、ヒメコウジ、シラタマノキ、アカモノ、ハリガネカズラ等が挙げられる。これらの植物は、1種を単独で使用しても、2種以上を併用してもよい。
植物の使用可能な部位としては、例えば、葉、花、種子、果実、果皮、茎、樹皮、植物全草等が挙げられる。これらの部位は、1種を単独で使用しても、2種以上を併用してもよい。
これらの中でも、植物由来成分としては、ヒメコウジ葉に由来する成分であることが好ましい。
植物の使用可能な部位としては、例えば、葉、花、種子、果実、果皮、茎、樹皮、植物全草等が挙げられる。これらの部位は、1種を単独で使用しても、2種以上を併用してもよい。
これらの中でも、植物由来成分としては、ヒメコウジ葉に由来する成分であることが好ましい。
植物由来成分の含有量は、0.001質量%以上0.5質量%以下であることが好ましく、0.005質量%以上0.1質量%以下であることがより好ましく、0.01質量%以上0.05質量%以下であることがさらに好ましい。かかる含有量で植物由来成分を含む生菌数調整剤を使用すれば、整肌作用を十分に高めることができる。
本発明の生菌数調整剤は、必要に応じて、その他の保湿成分、pH調整剤、増粘剤等を含んでいてもよい。
その他の保湿成分としては、特に限定されないが、例えば、グリセリン、トリメチロールプロパン、ジプロピレングリコール(DPG)、エチレングリコール、プロピレングリコール、トリメチレングリコール、1.3-ブチレングリコール(BG)、ソルビトール、マンニトール、ポリエチレングリコール(PEG)のような多価アルコール、グレープフルーツ種子エキス、コメヌカエキスのような植物由来エキス等が挙げられる。
pH調整剤としては、特に限定されないが、例えば、クエン酸、クエン酸ナトリウム、安息香酸ナトリウム、コハク酸一ナトリウム、水酸化カリウム等が挙げられる。
その他の保湿成分としては、特に限定されないが、例えば、グリセリン、トリメチロールプロパン、ジプロピレングリコール(DPG)、エチレングリコール、プロピレングリコール、トリメチレングリコール、1.3-ブチレングリコール(BG)、ソルビトール、マンニトール、ポリエチレングリコール(PEG)のような多価アルコール、グレープフルーツ種子エキス、コメヌカエキスのような植物由来エキス等が挙げられる。
pH調整剤としては、特に限定されないが、例えば、クエン酸、クエン酸ナトリウム、安息香酸ナトリウム、コハク酸一ナトリウム、水酸化カリウム等が挙げられる。
増粘剤としては、特に限定されないが、例えば、アラビアガム、トラガントガムのような植物系高分子、デキストラン、プルランのような微生物系高分子、コラーゲン、カゼイン、アルブミン、ゼラチンのような動物系高分子、デキストリン、アルギン酸ナトリウム、カルボキシメチルデンプンのようなデンプン系高分子、メチルセルロース、エチルセルロース、カルボキシメチルセルロース、ヒドロキシプロピルメチルセルロースのようなセルロース系高分子、ポリビニルアルコール、ポリビニルアセテートのようなビニル系高分子、ポリアクリル酸ナトリウム、ポリエチルアクリレート、ポリアクリル酸アルカノールアミンのようなアクリル系高子等が挙げられる。
植物由来エキス(特に、コメヌカエキス)を使用する場合、その含有量は、0.001質量%以上0.5質量%以下であることが好ましく、0.005質量%以上0.1質量%以下であることがより好ましく、0.01質量%以上0.05質量%以下であることがさらに好ましい。
本発明の生菌数調整剤のpHは、6以下であることが好ましく、5.5以下であることがより好ましく、4.5~5.5であることがさらに好ましい。生菌数調整剤のpHを上記範囲に調整することにより、アクネ菌が皮膚に悪影響を及ぼすことを防止するとともに、皮膚表面に存在する皮膚常在菌に占めるアクネ菌の割合を好適な範囲に調整し易くなる。
このようにして、本発明の生菌数調整剤によれば、皮膚症状を改善することができる。この皮膚症状としては、好ましくはざ瘡、脂性肌および乾燥肌の少なくとも1つである。

さらに、次に記載の各態様で提供されてもよい。
(1)被験者の皮膚表面に存在するアクネ菌の生菌数を調整するのに使用される生菌数調整剤であって、乳酸桿菌発酵液を含む、生菌数調整剤。
(2)上記(1)に記載の生菌数調整剤において、前記乳酸桿菌発酵液の含有量は、0.1質量%以上20質量%以下である、生菌数調整剤。
(3)上記(1)または(2)に記載の生菌数調整剤において、さらに、酵母菌由来成分を含む、生菌数調整剤。
(4)上記(3)に記載の生菌数調整剤において、前記酵母菌由来成分の含有量は、0.001質量%以上0.5質量%以下である、生菌数調整剤。
(5)上記(1)~(4)のいずれか1項に記載の生菌数調整剤において、さらに、穀物発酵液を含む、生菌数調整剤。
(6)上記(5)に記載の生菌数調整剤において、前記穀物発酵液の含有量は、0.001質量%以上0.5質量%以下である、生菌数調整剤。
(7)上記(1)~(6)のいずれか1項に記載の生菌数調整剤において、さらに、シラタマノキ属に属する植物に由来する成分を含む、生菌数調整剤。
(8)上記(7)に記載の生菌数調整剤において、前記シラタマノキ属に属する植物に由来する成分の含有量は、0.001質量%以上0.5質量%以下である、生菌数調整剤。
(9)上記(1)~(8)のいずれか1項に記載の生菌数調整剤において、前記生菌数調整剤のpHは、6以下である、生菌数調整剤。
(10)上記(1)~(9)のいずれか1項に記載の生菌数調整剤において、前記皮膚表面に存在する皮膚常在菌に占める前記アクネ菌の割合を30%以上50%未満に調整可能である、生菌数調整剤。
(11)上記(1)~(10)のいずれか1項に記載の生菌数調整剤において、ざ瘡、脂性肌および乾燥肌の少なくとも1つの症状を改善する、生菌数調整剤。

以下に、実施例を挙げて、本発明をさらに具体的に説明するが、本発明は、これらの実施例に限定されるものではない。
1.生菌数調整剤の準備
[組成]
・乳酸桿菌/豆乳発酵液 :0.1質量%以上20質量%以下
・サッカロミセス・セレビシアエエキス :0.001質量%以上0.5質量%以下
・コメ発酵液 :0.001質量%以上0.5質量%以下
・ヒメコウジ葉エキス :0.001質量%以上0.5質量%以下
・コメヌカエキス :0.001質量%以上0.5質量%以下
・水 :50質量%以上95質量%以下
・その他の成分 :残部
その他の成分は、保湿成分、pH調整剤、増粘剤等である。
なお、生菌数調整剤のpHは、約5であった。
[組成]
・乳酸桿菌/豆乳発酵液 :0.1質量%以上20質量%以下

その他の成分は、保湿成分、pH調整剤、増粘剤等である。
なお、生菌数調整剤のpHは、約5であった。
2.生菌数調整剤の使用
被験者(女性111名)に、生菌数調整剤を顔に塗ることを、約4週間継続して行ってもらった。

3.皮膚常在菌の測定
上記被験者の顔の皮膚に皮膚常在細菌採取用のシール材を貼付してもらった。所定時間経過後、シール材を剥がしてもらい、シール材の粘着剤層に転写された皮膚常在菌の種類および数を特定した。
皮膚常在菌の種類を特定する方法には、rDNAの特定領域の塩基配列を次世代シーケンサー(NGS)を用いて解析した。また、皮膚常在菌の数を特定する方法には、対象となる菌(アクネ菌、表皮ブドウ球菌およびコリネバクテリウム菌)の特定の配列に対するPCRを行い定量する方法(qPCR)を使用した。
なお、この測定は、生菌数調整剤を使用する前後で行ってもらった。
上記被験者の顔の皮膚に皮膚常在細菌採取用のシール材を貼付してもらった。所定時間経過後、シール材を剥がしてもらい、シール材の粘着剤層に転写された皮膚常在菌の種類および数を特定した。
皮膚常在菌の種類を特定する方法には、rDNAの特定領域の塩基配列を次世代シーケンサー(NGS)を用いて解析した。また、皮膚常在菌の数を特定する方法には、対象となる菌(アクネ菌、表皮ブドウ球菌およびコリネバクテリウム菌)の特定の配列に対するPCRを行い定量する方法(qPCR)を使用した。
なお、この測定は、生菌数調整剤を使用する前後で行ってもらった。
4.アンケート調査
上記被験者に対して、生菌数調整剤を使用した後、ざ瘡、脂性肌および乾燥肌の変化について、以下の基準に從って回答してもらった。
A:改善
B:変化なし
C:悪化

5.結果
5-1.ざ瘡
生菌数調整剤を使用する前において、皮膚表面に存在する皮膚常在菌に占めるアクネ菌の割合が70%以上である被験者は、ざ瘡に悩んでいた。
生菌数調整剤を使用した後、57%の被験者が改善したと回答した。

5-3.乾燥肌
生菌数調整剤を使用する前において、皮膚表面に存在する皮膚常在菌に占めるアクネ菌の割合が30%未満である被験者は、乾燥肌に悩んでいた。
生菌数調整剤を使用した後、50%の被験者が改善したと回答した。

生菌数調整剤を使用した後、50%の被験者が改善したと回答した。
また、ざ瘡や脂性肌を訴える傾向が高い被験者(すなわち、アクネ菌の割合が多い被験者)では、アクネ菌の割合(平均値)は、生菌数調整剤の使用前69%から使用後59%にまで減少した。そして、この差は統計的に有意であった。
一方、乾燥肌を訴える傾向が高い被験者(すなわち、アクネ菌の割合が少ない被験者)では、アクネ菌の割合(平均値)は、生菌数調整剤の使用前15%から使用後22%にまで増加した。そして、この差は統計的に有意であった。
なお、乳酸桿菌発酵液、酵母菌由来成分、穀物発酵液、シラタマノキ属に属する植物に由来する成分およびその他の成分の種類を変更、および/またはその含有量を変更して、生菌数調整剤を調製して、同様に評価すると同様の効果が得られる。

- 被験者の皮膚表面に存在するアクネ菌の生菌数を調整するのに使用される生菌数調整剤であって、
