Abstract
エルゴチオネイン生産糸状菌を培養することによるエルゴチオネイン生産において、エルゴチオネイン生産性を高めることを目的とする。 エルゴチオネイン生産糸状菌において、エルゴチオネイン生産を亢進する遺伝子変異を特定した。配列番号1の433位~440位の保存領域を含む、配列番号1に対応する配列において、配列番号1のW404~Q515の機能ドメインに対応する機能ドメインに含まれる領域を欠失させた核酸、及び当該核酸を含む糸状菌を提供する。
Description
本開示は、エルゴチオネイン(以下、ERGと表記する場合がある。)生産性を高めるように改変された核酸、並びにERG生産性を高めるように遺伝子改変された微生物に関する。さらに遺伝子改変された微生物を用いたERG生成方法にも関する。
エルゴチオネインは、ライ麦角菌(Claviceps purpurea)から単離された含硫アミノ酸として発見され、植物や動物の生体内にも存在することが見いだされている。しかしながら、植物や動物は、エルゴチオネインを合成することはできず、生体内のエルゴチオネインは、担子菌類などの微生物により合成されたエルゴチオネインに由来すると考えられている。特に担子菌類の一部のキノコ、例えば、ヒラタケ、シイタケ、マイタケ、エリンギなどの食用キノコにも含まれており、特にタモギタケに多く含まれていることが知られている。エルゴチオネインは、ヒスチジンからの生合成経路が知られており、硫黄原子はシステインから供給される。エルゴチオネインは、高い抗酸化性を有しており、また、エラスターゼ阻害作用、チロシナーゼ阻害作用が報告されており、美白やしわ予防といった美容・食品分野で特に注目されている。また、エルゴチオネインが生体酸化防御システムに関与することが判明してきており、医療分野での応用も試みられている。エルゴチオネインは、熱安定性やpH安定性が高く、高温でもその抗酸化作用を維持することができ、生理作用を期待して食品へ配合される他、抗酸化剤としての食品への使用も期待されている。
エルゴチオネインの製造方法としては、タモギタケなどの担子菌からの抽出、化学合成、微生物を用いた発酵が用いられている。タモギタケなどの担子菌からの抽出は、原材料の取得に時間がかかり、大量生産には適していない。大量生産には化学合成が適しているが高価な合成試薬を用いる必要があり、また、精製コストが高くなるという問題がある(特許文献1)。したがって、C1化合物資化性の細菌や酵母を用いた発酵(特許文献2、非特許文献1)、さらにはエルゴチオネイン生合成遺伝子を過剰発現させた微生物を用いた発酵(特許文献3)が製造方法としては有力であり、微生物発酵物をそのまま、または簡易な抽出操作により得られた抽出物を使用できる点で有力である。

微生物培養によるERG生産において、ERG生産性を高めることを目的とする。
本発明者らが、麹菌のERG生産性を高めることを目的として、鋭意研究を行ったところ、機能未知の遺伝子において、特定の遺伝子変異を導入することにより、エルゴチオネイン生産性が亢進したことを見出し本発明に至った。
そこで、本発明は以下に関する:
[1] 配列番号1に含まれる保存領域を含む、配列番号1に対応する配列において、配列番号1のW404~Q515の機能ドメインに対応する機能ドメインに含まれる領域に、当該機能ドメインの機能を変調する変異を含む配列;又は

をコードし、エルゴチオネイン産生微生物に導入された場合にエルゴチオネイン産生を亢進する、核酸。
[2] 前記保存領域が、配列番号1の433位~440位の保存領域である、項目1に記載の核酸。

本発明によれば、AO090005000664に対応する遺伝子において、配列番号1のW404~Q515の機能ドメインに対応する機能ドメインに含まれる領域に、当該機能ドメインの機能を変調する変異を含む微生物は、ERG生産を亢進することができる。
本発明において遺伝子変異を導入する微生物としては、エルゴチオネイン生産性を有する微生物である。そのような微生物としては、糸状菌が好ましく、より好ましくはアスペルギルス(Asperugillus)属、ペニシリウム(Penicillium)属、ラサムソニア属(Rasamsonia)、タラロマイシス(Talaromyces)属に属する任意の菌を使用することができる。アスペルギルス属の糸状菌の一例として、アスペルギルス・オリゼー(Aspergillus oryzae)、アスペルギルス・ソーヤ(Aspergillus sojae)、アスペルギルス・ルチュエンシス(Aspergillus luchuensis)、アスペルギルス・タマリ(Aspergillus tamarii)、アスペルギルス・ニガー(Aspergillus niger)、アスペルギルス・ニジュランス(Aspergillus nidulans)が挙げられる。糸状菌の菌株としては、例えば、アスペルギルス・オリゼーRIB40株、RIB326株など公的に入手可能な菌株、種麹屋などから市販されている種麹に含まれる菌株、又は清酒、醤油醸造蔵等の飲食品製造環境などから分離して得られる菌株を使用することができる。分離して得られる菌株を使用することができる。
本発明において、変異を導入する遺伝子は、アスペルギルス・オリゼーRIB40株におけるAO090005000664と名付けられた遺伝子、及びそのオルソログ遺伝子に関する。この遺伝子は機能未知の翻訳領域を有しており、657残基のアミノ酸配列(配列番号1)からなるタンパク質をコードする塩基配列(配列番号2)からなる。AO090005000664遺伝子の、アスペルギルス・ソーヤのオルソログ遺伝子の塩基配列は、Accession No.BaCA02000013のREGION 19840...21877に対応し(配列番号4)、そのアミノ酸配列は配列番号3で表される。AO090005000664遺伝子の、アスペルギルス・タマリ、アスペルギルス・ルチュエンシス、アスペルギルス・ニガー、アスペルギルス・ニジュランスでのオルソログ遺伝子は、それぞれ、BDV40DRAFT_264902(アミノ酸配列:配列番号5、塩基配列:配列番号6)、AAWM_07753(アミノ酸配列:配列番号7、塩基配列:配列番号8)、An16g07990(アミノ酸配列:配列番号9、塩基配列:配列番号10)、及びANIA_10201(アミノ酸配列:配列番号11、塩基配列:配列番号12)である。アスペルギルス属において高く保存されており、アスペルギルス・ソーヤでは98%、アスペルギルス・タマリでは96%、アスペルギルス・ルチュエンシスでは67%、アスペルギルス・ニガーでは67%、アスペルギルス・ニジュランスでは62%のアミノ酸配列の同一性を有する。これらの遺伝子は、アスペルギルス・ソーヤでは100%、アスペルギルス・タマリでは98%、アスペルギルス・ルチュエンシスでは79%、アスペルギルス・ニガーでは79%、アスペルギルス・ニジュランスでは75%のアミノ酸配列の相同性を有する。
AO090005000664には、他の種と高度に保存される複数の保存領域が存在する。このような保存領域として、特に配列番号1の411~479位に対応する領域(配列番号13)が挙げられる。より保存性の高い領域という観点から、保存領域は、配列番号1の411位~467位に対応する領域(配列番号14)が好ましく、411位~462位(配列番号15)に対応する領域がより好ましく、411位~444位(配列番号16)に対応する領域がさらに好ましく、427位~444位(配列番号17)に対応する領域がさらに好ましく、433位~440位(配列番号18)に対応する領域がさらにより好ましい。433位~440位(配列番号18)に対応する領域は、アスペルギルス属で極めて高く保存されており、より好ましくは完全に一致しうる。

本発明において、機能ドメインとは、AO090005000664遺伝子又はそのオルソログ遺伝子がコードするタンパク質における、配列番号1のW404~Q515の機能ドメイン、又は当該機能ドメインに対応する機能ドメインを指す。かかる機能ドメインは5つのαヘリックスから構成される超二次構造をとるドメインが存在することが示されている(図8A及びB)。この機能ドメインの具体的な機能は不明である一方、機能ドメインを構成する427位~429位の欠失(3DEL)、427~444位の欠失(18DEL)、及び419位から444位(26DEL)、431位~475位(45DEL)、419位~475位(57DEL)、及び404位~515位(112DEL)が欠失された際に、エルゴチオネイン産生を亢進することから、機能ドメインの機能変調が、エルゴチオネイン産生に寄与しうる。特に、機能ドメインを構成するアミノ酸の欠失によりエルゴチオネイン産生が増大する観点から、エルゴチオネイン産生増大には、機能ドメインにおける機能の妨害が必要とされうる。また、配列番号1の428位及び459位は、5つのαヘリックスの接続領域に位置しており、かかる位置における変異は、超二次構造における構造変化を及ぼす可能性が高いと考えられる。
機能ドメインの機能を変調する変異とは、配列番号1のW404~Q515の機能ドメイン、又は当該機能ドメインに対応する機能ドメインの機能に影響を及ぼす変異であればよく、かかる変異は、当該機能ドメインにおける1又は複数のアミノ酸の欠失、付加又は置換でありうる。対応する機能ドメインとは、AO090005000664と名付けられた遺伝子のオルソログ遺伝子における、機能ドメインをいう。かかる機能ドメインの機能に影響を及ぼすことにより、AO090005000664遺伝子又はそのオルソログ遺伝子を有するエルゴチオネイン産生微生物において、エルゴチオネイン生産性を亢進することができる。エルゴチオネイン産生を亢進する観点から、機能ドメインを変調する変異は、W404~Q515の機能ドメインに対応する機能ドメインに含まれる領域の欠失であるか、又は428位に対応する位置のG及び/又は459位に対応する位置のCにおける変異、特に置換である。
欠失されるアミノ酸の数は、欠失が生じた微生物においてERGの生産が亢進される限り任意の数である。一例として、欠失されるアミノ酸の数は、1~120、1~112、1~60、1~57及び1~47からなる群から選ばれる任意の範囲の数が欠失されうる。欠失されるアミノ酸の数は、好ましくは1~30、さらに好ましくは1~26の任意の数である。一例として1、2、3、4、5、6、7、8、9、10、11、12、13、14、15、16、17、18、19、20、21、22、23、24、25、及び26からなる群から選ばれる少なくとも1の数のアミノ酸が欠失される。より具体的には、428位に対応する位置アミノ酸が欠失されるか、又は428位に対応する位置のアミノ酸を含む複数のアミノ酸が欠失されうる。別の態様では、428位に対応する位置のアミノ酸を含まないアミノ酸が欠失されてもよい。
一例として、411位~462位の保存領域(配列番号15)に含まれ、428位を含む任意の配列が欠失されうる。一例として428位、427位~429位、427位~440位(配列番号20)、427位~444位(配列番号17)、及び419位~444位(配列番号21)からなる群から選ばれる少なくとも1の領域に対応する領域が欠失されうる。さらに、別の例として、431位~475位、419位~475位、及び404位~515位からなる群から選ばれる少なくとも1の領域に対応する領域が欠失されてもよい。
本発明において、アミノ酸位置は、AO090005000664遺伝子がコードする配列番号1のアミノ酸位に基づいて示されるが、対応する遺伝子、すなわちオルソログ遺伝子がコードするタンパク質において対応するアミノ酸位置を表す。対応する位置は、配列番号1のアミノ酸配列の相同配列をアライメントすることにより決定することができる。
W404~Q515の機能ドメインに対応する機能ドメインに含まれる領域が欠失された配列に対する同一性は、少なくとも60%、少なくとも65%、少なくとも70%、少なくとも75%、少なくとも80%、少なくとも85%、少なくとも90%、少なくとも95%、少なくとも97%、少なくとも98%、又は少なくとも99%の同一性を有する。かかる同一性を有し、かつ糸状菌においてW404~Q515の機能ドメインに対応する機能ドメインに含まれる領域が欠失された場合に、エルゴチオネイン生産性を高める配列を選択することができる。
エルゴチオネイン生成に寄与するという機能は、エルゴチオネインの生成過程における、何らかの機能を担うことをいう。このような機能として、エルゴチオネインの合成経路や分解経路に寄与する酵素の転写調節に関わる機能であってもよいし、エルゴチオネインの合成経路に寄与する酵素反応であってもよいし、エルゴチオネインの又はその前駆体の分解経路に寄与する酵素反応であってもよい。配列番号1に対応する配列において、配列番号1のW404~Q515の機能ドメインに対応する機能ドメインに含まれる領域が欠失された配列をコードする本発明の核酸が、エルゴチオネイン産生性の糸状菌に導入されることで、エルゴチオネイン産生を亢進する機能を有する。
本発明は、さらに、エルゴチオネイン産生能を亢進された微生物の製造方法にも関する。より具体的に、かかる製造方法は、配列番号1に対応する配列において、配列番号1のW404~Q515の機能ドメインに対応する機能ドメインに含まれる領域を欠失させることを含む。当該欠失される領域は、通常、W404~Q515の機能ドメインに対応する機能ドメインに含まれる領域であるが、エルゴチオネイン産生能を亢進可能な限り、404~511位の隣接する領域も欠失されてもよい。配列番号1の404位~511位の機能ドメインに対応する機能ドメインに含まれる領域が欠失される場合、当該欠失される領域は、配列番号1の428位に対応する位置を含んでもよいし、含まなくてもよい。欠失される領域には、配列番号1の428位に対応する位置が含まれることが好ましい。欠失される領域は、配列番号1の428位、427位~429位、427位~440位、427位~444位、及び419位~444位からなる群から選ばれる少なくとも1の領域に対応する領域である。欠失される領域は、411位~462位(配列番号15)の保存領域のすべてを欠失していてもよいし、さらには404位~515位の機能ドメインすべてを欠失していてもよい。欠失の範囲は、かかる欠失を有する微生物が、エルゴチオネイン産生を亢進可能な限り任意の欠失であってよい。W404~Q515の機能ドメイン内、特に411位~462位(配列番号15)の保存領域内の連続するアミノ酸配列が欠失されるが、不連続のアミノ酸配列が欠失されていてもよい。前記微生物としては、アスペルギルス属、ペニシリウム属、ラサムソニア属及びタラロマイシス属からなる群から選ばれるエルゴチオネイン生成微生物である。
本発明の別の態様では、配列番号1の対応する配列において、配列番号1の428位に対応する位置のG及び/又は459位に対応する位置のCにおいて変異を有し、エルゴチオネイン産生微生物に導入された場合にエルゴチオネイン産生を亢進する、核酸に関してもよい。かかる位置における変異は、より具体的にはG428S及び/又はC459Fの置換であってもよい。かかる位置に変異を有する核酸を有することにより、エルゴチオネイン産生微生物はエルゴチオネイン産生を亢進する。より具体的には、配列番号1の対応する配列は、本明細書により定義される保存領域を有する。
さらに、本発明は以下の:
1)配列番号1、3、5、7、9、及び11からなる群から選ばれるアミノ酸配列において、428位に対応する位置にSを有するか、及び/又は459位に対応する位置にFを有する、アミノ酸配列、又は
2)配列番号1、3、5、7、9、11からなる群から選ばれるアミノ酸配列に対し、少なくとも60%の同一性を有し、かつ428位に対応する位置にSを有するか、及び/又は459位に対応する位置にFを有する、アミノ酸配列
をコードし、エルゴチオネイン産生糸状菌に導入された場合にエルゴチオネイン産生を亢進する、核酸に関してもよい。G428S、C459F変異を有することにより、エルゴチオネイン産生を亢進することができる。
1)配列番号1、3、5、7、9、及び11からなる群から選ばれるアミノ酸配列において、428位に対応する位置にSを有するか、及び/又は459位に対応する位置にFを有する、アミノ酸配列、又は

をコードし、エルゴチオネイン産生糸状菌に導入された場合にエルゴチオネイン産生を亢進する、核酸に関してもよい。G428S、C459F変異を有することにより、エルゴチオネイン産生を亢進することができる。
配列番号1、3、5、7、9、11に対する同一性は、少なくとも60%、少なくとも65%、少なくとも70%、少なくとも75%、少なくとも80%、少なくとも90%、又は少なくとも95%の同一性を有する。特に、アスペルギルス・ソーヤのオルソログ遺伝子及びアスペルギルス・ニガーのオルソログ遺伝子とは、それぞれ68%及び67%の同一性である一方で、G428Sに対応する変異(アスペルギルス・ニガーではG442S)をアスペルギルス・ソーヤ及びアスペルギルス・ニガーに導入することで、ERG産生の亢進が確認されている(図4及び5)。
さらに、本発明は以下の:

配列番号2、4、6、8、10、及び12に対する同一性は、少なくとも60%、少なくとも65%、少なくとも70%、少なくとも75%、少なくとも80%、少なくとも90%、又は少なくとも95%の同一性を有する。特に、アスペルギルス・ソーヤのオルソログ遺伝子及びアスペルギルス・ニガーのオルソログ遺伝子とは、それぞれ68%及び67%の同一性である一方で、G428Sに対応する変異(アスペルギルス・ニガーではG442S)を導入することで、ERG産生の亢進が確認されている(図4及び5)。
本発明の核酸は、糸状菌、例えばアスペルギルス属、ペニシリウム属、ラサムソニア属又はタラロマイシス属に由来してもよい。本発明の核酸は、これらの糸状菌において保存性が高い。アスペルギルス属に属する糸状菌由来の核酸にコードされるアミノ酸配列は、配列番号1に対し75~100%の相同性を有する。ペニシリウム属に属する糸状菌由来の核酸にコードされるアミノ酸配列は、配列番号1に対し68~75%の相同性を有する。ラサムソニア属に属する糸状菌由来の核酸にコードされるアミノ酸配列は、配列番号1に対し70%の相同性を有する。タラロマイシス属に属する糸状菌由来の核酸にコードされるアミノ酸配列は、配列番号1に対し64~68%の相同性を有する。
エルゴチオネイン生成に寄与するという機能は、エルゴチオネインの生成過程における何らかの酵素反応を担うことをいう。エルゴチオネインの合成経路に寄与する酵素反応であってもよいし、エルゴチオネインの又はその前駆体の分解経路に寄与する酵素反応であってもよい。合成経路に寄与する酵素反応を担う場合、本発明の核酸がコードするタンパク質は、エルゴチオネイン又はその前駆体の合成活性を有しうる。分解経路に寄与する酵素反応である場合、本発明の核酸がコードするタンパク質は、エルゴチオネイン又はその前駆体若しくは代謝物の分解活性を有しうる。配列番号1の428位に対応する位置にSを有するか、及び/又は459位に対応する位置にFを有するアミノ酸配列をコードする本発明の核酸は、エルゴチオネイン産生性の糸状菌に導入されることで、エルゴチオネイン産生を亢進する機能を有する。
配列番号1の428位に対応する位置及び459位に対応する位置は、配列番号1のアミノ酸配列の相同配列をアライメントすることにより決定することができる。1の例では近傍の配列から決定することができ、428位の近傍配列であるLSGLQのGの位置が428位に対応する。428位のGがSに変異することでエルゴチオネイン生産性が亢進する。459位の近傍配列であるAPCEDのCの位置が459位に対応する。459位のCがFに変異することでエルゴチオネイン生産性が亢進する。428位に対応する位置及び459位に対応する位置は、別個に変異が導入されていてもよいし、同時に変異が導入されていてもよい。
本発明は、さらに、エルゴチオネイン産生能を亢進された微生物の製造方法にも関する。より具体的に、かかる製造方法は、本発明に係る保存領域をコードする微生物のゲノムに対し、配列番号1の428位に対応する位置及び/又は459位に対応する位置に変異を導入することを含む。428位に対応する位置の変異は、G428Sであり、459位の変異はC459Fであることが好ましい。前記微生物が、アスペルギルス属、ペニシリウム属、ラサムソニア属及びタラロマイシス属からなる群から選ばれるエルゴチオネイン生成微生物である。一例として、配列番号1の部分配列(427位~444位)に対応する以下の保存領域:

ゲノムに対する変異の導入は、任意の手法を用いて導入されてもよい。定法の形質転換系を用いて導入されてもよいし、ランダム変異を導入した結果導入されてもよいし、ゲノム編集技術を利用して導入されてもよい。一例として、形質転換系では、遺伝子カセットを用いて導入されてもよいし、部位特異的な遺伝子導入を行ってもよいし、ベクターなどによって導入されてもよい。本発明の別の態様では、本発明の核酸を発現可能に組み込まれたベクターに関してもよい。ランダム変異の変異導入方法としては、例えば、物理的にDNAを損傷し変異を導入する紫外線(UV)やX線照射、化学的にDNAを損傷し変異を導入するN―メチル―N’―ニトロ―N―ニトロソグアニジン(NTG)やエチルメタンスルホン酸(EMS)などのアルキル化試薬による処理などが挙げられる。ゲノム編集技術としては、一例として、CRISPR/Casシステムを利用した部位特異的切断と相同組換えを利用することができる。発明は、本発明に係る核酸を含む微生物にも関する。本発明に係る製造方法の結果取得された微生物である。
本発明に係る微生物を培養することにより、高い生産性でエルゴチオネインを製造することができる。本発明に係る微生物は、固体培養又は液体培養のいずれで培養されてもよい。一例として、本発明に係る微生物を培養することにより、高い生産性でエルゴチオネインを製造することができる。本発明に係る微生物は、固体培養又は液体培養のいずれで培養されてもよい。一例として、蒸し米において本発明に係る微生物を培養することで、エルゴチオネイン含有量が高い米麹を製造することができる。かかる麹を用い、本技術分野の周知の工程を経て、醤油、味噌、塩麹、甘酒などを製造することができ、また麹菌を収集してマイコプロテインを得ることもできる。したがって、本発明の1の態様では、本発明は、高含量のエルゴチオネインを含む米麹の製造方法に関していてもよい。生成されたエルゴチオネインは、培養物中から定法に基づいて精製することができる。一例として、培養物を粉砕し、水を蒸発させて乾燥粉末を得ることができる。乾燥されたエルゴチオネイン含有原料に対し、含水エタノールを添加することにより、エルゴチオネインがエタノール溶液中に抽出される。不溶性物質を除去し、エタノール溶液中のエルゴチオネインを、クロマトグラフィーに供することで精製してもよい。
エルゴチオネインは、抗酸化作用をはじめ、エラスターゼ阻害作用、チロシナーゼ阻害作用、ポリフェノールオキシダーゼ(PPO)阻害作用など種々の作用を有することが知られている。したがって、製造されたエルゴチオネインは、精製されて又はそのまま食品、化粧品、医薬部外品、及び医薬品などの製品に配合されてもよいし、製品に配合される濃縮物として調製されてもよい。食品として配合される場合、機能性表示食品とすることもできる。かかる機能性表示として、エルゴチオネインの、エラスターゼ阻害作用に基づく抗しわ作用、チロシナーゼ阻害活性に基づく美肌又は美白作用、過酸化脂質の生成に基づく生活習慣病予防作用、活性酸素除去に基づく認知症やアルツハイマー病の予防作用がなどの機能を表示することができる。製造されたエルゴチオネインは、特にエルゴチオネインの生理作用に着目した、機能性表示食品、栄養機能食品、特定保健用食品、又は栄養補助食品(サプリメント)であってもよい。
本開示において言及される全ての文献はその全体が引用により本明細書に取り込まれる。相同性は、NCBIのblastpのプログラムを用いて、NCBIに登録されたタンパク質データベースを用いて決定される。本開示において相同性及び同一性の%を除き、特に言及がない限り質量%を意味する。
以下に説明する本発明の実施例は例示のみを目的とし、本発明の技術的範囲を限定するものではない。本発明の技術的範囲は特許請求の範囲の記載によってのみ限定される。本発明の趣旨を逸脱しないことを条件として、本発明の変更、例えば、本発明の構成要件の追加、削除及び置換を行うことができる。
実施例1:ERG高生産株の解析
(1)培地
アスペルギルス属の真核生物は、以下の培地を用いて培養した。液体培養ではCzapek-DOX(Cz-DOX)培地(3.5%Czapek-DOX broth、BD社製、pH6.0)、PD培地(2%デキストリン、1%ポリペプトン、0.5%KH2PO4、0.1%NaNO3、0.05%MgSO4、0.1%カザミノ酸、pH6.0)、パフミン培地(5%パフミンSM、pH無調整)を用いた。プレート培養では主にCz-DOX寒天培地(3.5% zapek-DOX broth、BD社製、0.1%trace element、2%Agar添加)又はマルツ寒天培地(4.5%Malt Agar Nissui,日水製薬社製,0.1%trace element、0.5% yeast extract)を用いた。
(1)培地
アスペルギルス属の真核生物は、以下の培地を用いて培養した。液体培養ではCzapek-DOX(Cz-DOX)培地(3.5%Czapek-DOX broth、BD社製、pH6.0)、PD培地(2%デキストリン、1%ポリペプトン、0.5%KH2PO4、0.1%NaNO3、0.05%MgSO4、0.1%カザミノ酸、pH6.0)、パフミン培地(5%パフミンSM、pH無調整)を用いた。プレート培養では主にCz-DOX寒天培地(3.5% zapek-DOX broth、BD社製、0.1%trace element、2%Agar添加)又はマルツ寒天培地(4.5%Malt Agar Nissui,日水製薬社製,0.1%trace element、0.5% yeast extract)を用いた。
(2)ERG高生産株のゲノム解析
ERG高生産株の全ゲノムを解析し、その中で機能未知のAO090005000664という遺伝子におけるG428S又はC459Fの変異がERG生産亢進に寄与することを見出した。AO090005000664のアミノ酸配列(配列番号1)、及び塩基配列(配列番号2)は、図1のとおりである。
ERG高生産株の全ゲノムを解析し、その中で機能未知のAO090005000664という遺伝子におけるG428S又はC459Fの変異がERG生産亢進に寄与することを見出した。AO090005000664のアミノ酸配列(配列番号1)、及び塩基配列(配列番号2)は、図1のとおりである。
AO090005000664のアミノ酸配列について、NCBI(National Center for Biotechnology Information)でBLAST(Basic Logical Alignment Search Tool)のデフォルト設定で相動性検索を行ったところ、アスペルギルス属で高度に保存されており、配列番号1の411~467位において、高度に保存された保存領域が見いだされた(図2)。AO090005000664と、アスペルギルス・オリゼー、アスペルギルス・ソーヤ、アスペルギルス・タマリ、アスペルギルス・ルチュエンシス、アスペルギルス・ニガー、及びアスペルギルス・ニジュランスのオルソログ遺伝子との相同性は、それぞれ100%、98%、96%、67%、67%、及び62%であった。
実施例2:アスペルギルス・オリゼーRIB40株における変異再現株の作製およびERG生産性の確認
(1) G428Sカセットの作製
アスペルギルス・オリゼーRIB40_dKP株(Δku70、ΔpyrG)を宿主として、AO090005000664遺伝子にG428S変異を導入するための、遺伝子カセットを作製した。プライマーを表1に示す。RIB40株のゲノム配列を鋳型とし、AoG428S_F1とR1、AoG428S_F2とR2、AoG428S_F3とR3、AopyrG FとRのプライマーセットで、PCRにより遺伝子断片を増幅した。PCR条件は添付のプロトコルに従った。精製した4つのPCR増幅断片とLinerized pUC19(Takara)(各0.5μL)をNEBuilder HiFi DNA Assembly Master Mix(NEB)2.5μLと混合し、60℃、1時間反応させた。反応物4μLをECOS Competent E. coli JM109(ニッポンジーン)に導入して形質転換を行った。次に、この形質転換体から抽出したプラスミドを鋳型AoG428S_NFとNRのプライマーセットを用いて、PCRにより増幅された遺伝子断片をG428Sカセットとした。PCR産物50μLはエタノール沈殿処理して滅菌水10μLに溶解させた。
(1) G428Sカセットの作製
アスペルギルス・オリゼーRIB40_dKP株(Δku70、ΔpyrG)を宿主として、AO090005000664遺伝子にG428S変異を導入するための、遺伝子カセットを作製した。プライマーを表1に示す。RIB40株のゲノム配列を鋳型とし、AoG428S_F1とR1、AoG428S_F2とR2、AoG428S_F3とR3、AopyrG FとRのプライマーセットで、PCRにより遺伝子断片を増幅した。PCR条件は添付のプロトコルに従った。精製した4つのPCR増幅断片とLinerized pUC19(Takara)(各0.5μL)をNEBuilder HiFi DNA Assembly Master Mix(NEB)2.5μLと混合し、60℃、1時間反応させた。反応物4μLをECOS Competent E. coli JM109(ニッポンジーン)に導入して形質転換を行った。次に、この形質転換体から抽出したプラスミドを鋳型AoG428S_NFとNRのプライマーセットを用いて、PCRにより増幅された遺伝子断片をG428Sカセットとした。PCR産物50μLはエタノール沈殿処理して滅菌水10μLに溶解させた。
(2)C459Fカセットの作製
上述の(1)と同様にしてC459Fカセットを作製した。プライマーはAoG428S_R2をAoC459F_R2に、AoG428S_F3をAoC459F_F3に変えて実施した。

(3) A. oryzae RIB40_dKPの形質転換
RIB40_dKP株を10mMウリジン、10mMウラシルを含んだCz-DOX_UraUri培地に塗布し、5日間、30℃で培養した。プレート上で生育させ、分生子をマイクロループで掻き取り、150mL容三角フラスコに入れた10mMウリジン、10mMウラシルを含んだPD培地40mLに植菌した。30℃、180rpmで旋回培養を20時間行い、菌体を回収した。
RIB40_dKP株を10mMウリジン、10mMウラシルを含んだCz-DOX_UraUri培地に塗布し、5日間、30℃で培養した。プレート上で生育させ、分生子をマイクロループで掻き取り、150mL容三角フラスコに入れた10mMウリジン、10mMウラシルを含んだPD培地40mLに植菌した。30℃、180rpmで旋回培養を20時間行い、菌体を回収した。
培養後の菌体をミラクロースで集菌し、KCl buffer (0.6 M KCl, 0.1M NaH2PO4, pH5.5)で洗浄した。回収した菌体を細胞壁溶解酵素20mL(0.5%Lysing enzyme(Sigma-aldrich)、0.3%Yatalase(Takara)、1.5%Cellurase Onozuka(Yakult) /20mL KCl buffer)に懸濁し、30℃、で3時間振とうした後、70μmのセルストレーナーに通してろ液を回収した。
酵素処理後、遠心処理(4℃、3000rpm、10min)によりプロトプラストを沈殿させ、上清を除去した。次に、SolB buffer(1.2M Sorbitol,0.1M KCl,10mM Tris-HCl;pH7.5)30mLを加えてプロトプラストを再懸濁させてエッペンチューブに移し、遠心処理によりプロトプラストを回収した。さらに、30mLのSolB bufferを加えて同様の洗浄操作を再度繰り返した。回収したプロトプラストを0.4~0.8mLのSol Bに懸濁させた。該プロトプラスト懸濁液100μLにG428Sカセット、または、C459Fカセット5~10μg、12.5μLのSolC(50%PEG3350,50mM CaCl2,10mM Tris-HCl;pH7.5)を加えてゆっくり混合した後、氷上で30minインキュベートした。その後、1mLのSolCを加えて、ピペッティングにより混合させた後、室温で10min静置した。その後、10mLのSolBを加え、遠心処理によりプロトプラストを沈殿させ、上清を除去した。再生用寒天培地(3.5%Czapek-DOX broth、1.2Mソルビトール、0.1%trace element、2%Agar)上に全量をのせ、50℃のTop Agar(3.5%Cz-DOX borth、1.2Mソルビトール、0.1%trace element、0.5%Agar)を重層し、30℃で約1週間培養した。
得られた形質転換体は、Cz-DOX寒天培地(3.5%Czapek-DOX borth、0.1%trace element、1.5%Agar)で3回点植して純化した。
(4) 変異再現株のERG生産量の確認
上記で得られた形質転換体、および、コントロールとしてRIB40野生株のERG生産量を比較した。
(4-1)培養・抽出
50mL容三角フラスコにパフミン培地(5%パフミンSM、pH無調整)10mLを調製し、120℃、30分オートクレーブ滅菌した。そこに分生子懸濁液(1.0×107個/ml)を100μL添加し、30℃、160rpm、4日間振盪培養した。培養液を熱水抽出(90℃、1時間)し、上清0.5mLをナノセップ遠心ろ過デバイス(日本ポール社製 3K)でろ過し、適宜超純水で希釈して分析サンプルとした。
上記で得られた形質転換体、および、コントロールとしてRIB40野生株のERG生産量を比較した。
(4-1)培養・抽出
50mL容三角フラスコにパフミン培地(5%パフミンSM、pH無調整)10mLを調製し、120℃、30分オートクレーブ滅菌した。そこに分生子懸濁液(1.0×107個/ml)を100μL添加し、30℃、160rpm、4日間振盪培養した。培養液を熱水抽出(90℃、1時間)し、上清0.5mLをナノセップ遠心ろ過デバイス(日本ポール社製 3K)でろ過し、適宜超純水で希釈して分析サンプルとした。

(5) 形質転換株の配列確認
Cz-DOX寒天培地で4日培養した菌体の分生子を爪楊枝で2掻き程度分、100μLのTEに懸濁させた分生子懸濁液0.8μLを19.2μLのKOD one(Toyobo社製)PCR反応液に加え、コロニーPCRを行った。PCR産物をQIAquick PCR purification kit(QIAGEN社製)で精製した。カラムからの溶出にはElution Buffer(QIAGEN社製)を用い、溶出量は、PCRの系と同様量の50μLで溶出した。精製したPCR産物をFasmac社に委託して配列を確認した。

実施例3:アスペルギルス・ソーヤにおける変異再現株の作製およびERG生産性の確認
実施例1のアスペルギルス・オリゼーRIB40 株AO090005000664 G428S 変異再現株の作製の項に記載されている方法と同様に、アスペルギルス・ソーヤNBRC4239_dKP株(Δku70、ΔpyrG)を宿主として、変異導入、形質転換、ERG生産量の確認を行った。アスペルギルス・ソーヤにおけるAO090005000664遺伝子のオルソログ遺伝子は、アミノ酸同一性で98%、アミノ酸相同性では、100%である。
G428S変異再現の形質転換体19株のうち6株にG428S変異が入っていた。
各菌株のERG生成量を図4に示す。変異導入株のERG生産性を比較したところ、対照株および野生株と比較してERG生産性が向上した。アスペルギルス・ソーヤでも本遺伝子変異がERG生産性向上に関わることが明らかとなった。

実施例4:アスペルギルス・ニガー(Aspergillus niger)における変異再現株の作製およびERG生産性の確認
実施例1のアスペルギルス・オリゼーRIB40 株AO090005000664 G428S 変異再現株の作製の項に記載されている方法と同様に、アスペルギルス・ニガーA1179株 (別名MA70.15、Genotyping:kusA::amdS、cspA1(short conidiophoresとなる変異)、pyrG-)を宿主として、変異導入、形質転換し、培養日数を6日に変更してERG生産量の確認を行った。
実施例1のアスペルギルス・オリゼーRIB40 株AO090005000664 G428S 変異再現株の作製の項に記載されている方法と同様に、アスペルギルス・ニガーA1179株 (別名MA70.15、Genotyping:kusA::amdS、cspA1(short conidiophoresとなる変異)、pyrG-)を宿主として、変異導入、形質転換し、培養日数を6日に変更してERG生産量の確認を行った。
アスペルギルス・ニガーにおけるAO090005000664遺伝子のオルソログ遺伝子のアミノ酸同一性は67%、アミノ酸相同性では、79%である。ERG高生産性を示したアスペルギルス・オリゼー株の変異点(G428SおよびC459F)は、アスペルギルス・ニガーでも保存されている(G442およびC473)。そこで、アスペルギルス・オリゼーRIB40株で変異効果が確認されたG428Sに相当する442番目のグリシンをセリンに置換した変異株を作製した。各菌株のERG生成量を図5に示す。変異導入株のERG生産性を比較したところ、対照株および野生株と比較してERG生産性が向上した。アスペルギルス・ニガーでも本遺伝子変異がERG生産性向上に関わることが明らかとなった。
実施例5:ERG高生産株の解析
(1)培地
アスペルギルス属の真核生物は、以下の培地を用いて培養した。液体培養ではCzapek-DOX(Cz-DOX)培地(3.5%Czapek-DOX broth、BD社製、pH6.0)、PD培地(2%デキストリン、1%ポリペプトン、0.5%KH2PO4、0.1%NaNO3、0.05%MgSO4、0.1%カザミノ酸、pH6.0)、パフミン培地(5%パフミンSM、pH無調整)を用いた。プレート培養では主にCz-DOX寒天培地(3.5% zapek-DOX broth、BD社製、0.1%trace element、2%Agar添加)又はマルツ寒天培地(4.5%Malt Agar Nissui,日水製薬社製,0.1%trace element、0.5% yeast extract)を用いた。

(2)ERG高生産株のゲノム解析
ERG高生産株の全ゲノムを解析し、その中で機能未知のAO090005000664という遺伝子におけるアミノ酸欠損がERG生産亢進に寄与することを見出した。AO090005000664のアミノ酸配列(配列番号1)、及び塩基配列(配列番号2)は、図1のとおりである。

AO090005000664のアミノ酸配列について、NCBI(National Center for Biotechnology Information)でBLAST(Basic Logical Alignment Search Tool)のデフォルト設定で相動性検索を行ったところ、アスペルギルス属で高度に保存されており、配列番号1の411~467位において、高度に保存された保存領域が見いだされた(図6)。AO090005000664と、アスペルギルス・オリゼー、アスペルギルス・ソーヤ、アスペルギルス・タマリ、アスペルギルス・ルチュエンシス、アスペルギルス・ニガー、及びアスペルギルス・ニジュランスのオルソログ遺伝子との同一性は、それぞれ100%、98%、96%、67%、67%、及び62%であった。
実施例6:アスペルギルス・オリゼーRIB40株のアミノ酸欠損株の作製
(1) アミノ酸欠損カセットの作製およびアスペルギルス・オリゼーRIB40_dKPの形質転換株の取得
RIB40_dKP株(Δku70、ΔpyrG)を宿主として、アミノ酸を欠損させる遺伝子カセットを作製した。欠損させるアミノ酸はG428を中心とした保存性の高いアミノ酸とした(図6)。具体的に428位、427位~429位、427位~440位、427位~444位、及び419位~444位をそれぞれ欠失させた。用いたプライマーを表1に示す。
RIB40株のゲノム配列を鋳型とし、プライマー1と2、プライマー3と4、プライマー5と6のプライマーセットで、PCRにより遺伝子断片を増幅した。PCR条件は添付のプロトコルに従った。精製した3つのPCR増幅断片とLinerized pUC19(Takara)(各0.5μL)をNEBuilder HiFi DNA Assembly Master Mix(NEB)2.5μLと混合し、60℃、1時間反応させた。反応物4μLをECOS Competent E. coli JM109(ニッポンジーン)に導入して形質転換を行った。次に、この形質転換体から抽出したプラスミドをテンプレートにして、プライマー7と8(Ao3DEL)、プライマー9と8(Ao18DEL)、および、プライマー9と10(Ao26DEL)のセットで、PCRより遺伝子断片を増幅した。PCR条件は添付のプロトコルに従った。各PCR反応液にDpnIを添加して37℃、1時間反応させた。次に、DpnI処理した各PCR産物2μlをLigation high Ver.2(Toyobo)5μl、T4 Polynucleotide Kinase 1μl、滅菌水 7μlと混合し16℃、1時間反応させた。各反応物5μLをECOS Competent E. coli JM109 (ニッポンジーン)に導入して形質転換を行った。次に、この形質転換体から抽出した各プラスミドを鋳型として、プライマー11と12のセットを用いて、PCRにより増幅された遺伝子断片をそれぞれアミノ酸欠損カセットAoXDELカセット(X=3、18、26)とした。PCR産物50μLはエタノール沈殿処理して滅菌水10μLに溶解させた。
AoXDELカセットを導入してRIB40_dKP株を形質転換した。形質転換株
はCz-DOX寒天培地に点植により3回植え継いで純化した。

カセット導入位置について、プライマー1と4を用いて、バンドの大きさにより確認した。また、このPCR産物を精製してシーケンス解析を行い、アミノ酸欠損を確認した。

実施例7:アスペルギルス・ソーヤNBRC4239株のアミノ酸欠損株の作製
(1) アミノ酸欠損カセットの作製およびアスペルギルス・ソーヤNBRC4239_dKPの形質転換株の取得
NBRC4239_dKP株(Δku70、ΔpyrG)を宿主として、上述したアスペルギルス・オリゼーRIB40株と同様にアミノ酸欠損させる遺伝子カセットを作製した。NBRC4239株のゲノム配列を鋳型とし、プライマー13と14、プライマー3と15、プライマー5と6のプライマーセットで、PCRにより遺伝子断片を増幅した。PCR条件は添付のプロトコルに従った。精製した3つのPCR増幅断片とLinerized pUC19(Takara)(各0.5μL)をNEBuilder HiFi DNA Assembly Master Mix(NEB)2.5μLと混合し、60℃、1時間反応させた。反応物4μLをECOS Competent E. coli JM109(ニッポンジーン)に導入して形質転換を行った。次に、この形質転換体から抽出したプラスミドをテンプレートにして、プライマー16と17(As1DEL)、プライマー7と8(As3DEL)、プライマー18と8(As14DEL)、プライマー9と8(As18DEL)、および、プライマー19と10(As26DEL)のセットで、PCRより遺伝子断片を増幅した。上述したアスペルギルス・オリゼーRIB40株と同様に取得した各プラスミドを鋳型として、プライマー20と21のセットを用いて、PCRにより増幅された遺伝子断片をそれぞれアミノ酸欠損カセットAsXDELカセット(X=1、3、14、18、26)とした。PCR産物50μLはエタノール沈殿処理して滅菌水10μLに溶解させた。
AsXDELカセットを導入してNBRC4239_dKP株を形質転換した。形質転換株はCz-DOX寒天培地に点植により3回植え継いで純化した。

NBRC4239_dKP株(Δku70、ΔpyrG)を宿主として、上述したアスペルギルス・オリゼーRIB40株と同様にアミノ酸欠損させる遺伝子カセットを作製した。NBRC4239株のゲノム配列を鋳型とし、プライマー13と14、プライマー3と15、プライマー5と6のプライマーセットで、PCRにより遺伝子断片を増幅した。PCR条件は添付のプロトコルに従った。精製した3つのPCR増幅断片とLinerized pUC19(Takara)(各0.5μL)をNEBuilder HiFi DNA Assembly Master Mix(NEB)2.5μLと混合し、60℃、1時間反応させた。反応物4μLをECOS Competent E. coli JM109(ニッポンジーン)に導入して形質転換を行った。次に、この形質転換体から抽出したプラスミドをテンプレートにして、プライマー16と17(As1DEL)、プライマー7と8(As3DEL)、プライマー18と8(As14DEL)、プライマー9と8(As18DEL)、および、プライマー19と10(As26DEL)のセットで、PCRより遺伝子断片を増幅した。上述したアスペルギルス・オリゼーRIB40株と同様に取得した各プラスミドを鋳型として、プライマー20と21のセットを用いて、PCRにより増幅された遺伝子断片をそれぞれアミノ酸欠損カセットAsXDELカセット(X=1、3、14、18、26)とした。PCR産物50μLはエタノール沈殿処理して滅菌水10μLに溶解させた。
AsXDELカセットを導入してNBRC4239_dKP株を形質転換した。形質転換株はCz-DOX寒天培地に点植により3回植え継いで純化した。
(2) 分生子PCR
カセット導入位置について、プライマー13と15を用いて、バンドの大きさにより確認した。また、このPCR産物を精製してシーケンス解析を行い、アミノ酸欠損を確認した。

実施例8:アスペルギルス・オリゼーRIB40株のアミノ酸欠損株の作製
(1) アミノ酸欠損カセットの作製およびアスペルギルス・オリゼーRIB40_dKPの形質転換株の取得
RIB40_dKP株(Δku70、ΔpyrG)を宿主として、アミノ酸を欠損させる遺伝子カセットを作製した。実施例10の予測構造をもとに、ドメインの一部または全体を欠失させるアミノ酸とした。具体的に431位~475位、419位~475位、及び404位~515位をそれぞれ欠失させた。用いたプライマーを表1に示す。
実施例2で作製したプラスミドをテンプレートにして、プライマー22と23(Ao45DEL)、プライマー22と10(Ao57DEL)、および、プライマー24と25(Ao112DEL)のセットで、PCRより遺伝子断片を増幅した。PCR条件は添付のプロトコルに従った。各PCR反応液にDpnIを添加して37℃、1時間反応させた。次に、DpnI処理した各PCR産物2μlをLigation high Ver.2(Toyobo)5μl、T4 Polynucleotide Kinase 1μl、滅菌水 7μlと混合し16℃、1時間反応させた。各反応物5μLをECOS Competent E. coli JM109 (ニッポンジーン)に導入して形質転換を行った。次に、この形質転換体から抽出した各プラスミドを鋳型として、プライマー11と12のセットを用いて、PCRにより増幅された遺伝子断片をそれぞれアミノ酸欠損カセットAoXDELカセット(X=45、57、112)とした。PCR産物50μLはエタノール沈殿処理して滅菌水10μLに溶解させた。

実施例9:アスペルギルス・ニガーFGSC A1179株のアミノ酸欠損株の作製
(1) アミノ酸欠損カセットの作製およびアスペルギルス・ニガーFGSC A1179の形質転換株の取得
FGSC A1179株(ΔkusA、pyrG-)を宿主として、上述したアスペルギルス・オリゼーRIB40株と同様にアミノ酸欠損させる遺伝子カセットを作製した。アスペルギルス・ニガーIAM2533株のゲノム配列を鋳型とし、プライマー26と27、プライマー28と29のプライマーセットで、PCRにより遺伝子断片を増幅した。PCR条件は添付のプロトコルに従った。精製した2つのPCR増幅断片および実施例2で精製したプライマー5と6の遺伝子断片をLinerized pUC19(Takara)(各0.5μL)をNEBuilder HiFi DNA Assembly Master Mix(NEB)2.5μLと混合し、60℃、1時間反応させた。反応物4μLをECOS Competent E. coli JM109(ニッポンジーン)に導入して形質転換を行った。次に、この形質転換体から抽出したプラスミドをテンプレートにして、プライマー30と31(Ang3DEL)、プライマー32と31(Ang18DEL)、プライマー32と33(Ang26DEL)、プライマー34と33(Ang57DEL)、および、プライマー35と36(Ang112DEL)のセットで、PCRより遺伝子断片を増幅した。上述したアスペルギルス・オリゼーRIB40株と同様に取得した各プラスミドを鋳型として、プライマー37と38のセットを用いて、PCRにより増幅された遺伝子断片をそれぞれアミノ酸欠損カセットAsXDELカセット(X=3、18、26、57、112)とした。PCR産物50μLはエタノール沈殿処理して滅菌水10μLに溶解させた。
AngXDELカセットを導入してFGSC A1179株を形質転換した。形質転換株はCz-DOX寒天培地に点植により3回植え継いで純化した。

実施例10:AO090005000664遺伝子から発現されるタンパク質の構造解析
SWISS-MODEL(https://swissmodel.expasy.org/)のホモロジーモデリング機能を使用し、AO090005000664遺伝子について、PAAG_04735の予測立体構造をテンプレートに用いて立体構造予測を行った(図8)。テンプレートの立体構造としては、AO090005000664と相同性が高い遺伝子のうち、立体構造が明らかとなっているもの、もしくは、予測立体構造が公開されているものを検索し、同一性43%、Paracoccidioides lutzii由来のPAAG_04735の予測立体構造を選択した。なお、PAAG_04735の予測立体構造はAlphaFold2で予測されたものである。現在、AO090005000664遺伝子と100%同一性を有するOAory_01008280の予測構造がAlphaFoldProteinStructureDatabase(https://alphafold.com/)で公開されているが、ホモロジーモデリングした予測構造は、これと類似していた。なお、ホモロジーモデリング当時、OAory_01008280の予測構造は公開されていなかった。
SWISS-MODEL(https://swissmodel.expasy.org/)のホモロジーモデリング機能を使用し、AO090005000664遺伝子について、PAAG_04735の予測立体構造をテンプレートに用いて立体構造予測を行った(図8)。テンプレートの立体構造としては、AO090005000664と相同性が高い遺伝子のうち、立体構造が明らかとなっているもの、もしくは、予測立体構造が公開されているものを検索し、同一性43%、Paracoccidioides lutzii由来のPAAG_04735の予測立体構造を選択した。なお、PAAG_04735の予測立体構造はAlphaFold2で予測されたものである。現在、AO090005000664遺伝子と100%同一性を有するOAory_01008280の予測構造がAlphaFoldProteinStructureDatabase(https://alphafold.com/)で公開されているが、ホモロジーモデリングした予測構造は、これと類似していた。なお、ホモロジーモデリング当時、OAory_01008280の予測構造は公開されていなかった。
本発明者らの研究により、AO090005000664遺伝子がコードするアミノ酸配列において427位~429位の欠失(3DEL)、427~444位の欠失(18DEL)、及び419位から444位(26DEL)、431位~475位(45DEL)、419位~475位(57DEL)、及び404位~515位(112DEL)をそれぞれ欠失させた(図7A及びC)。なお431位~475位は、5つのαヘリックスから構成される超二次構造ドメインのうち、2本目のαヘリックス及び3本目のヘリックスに相当した。404位~515位は、当該超二次構造ドメインのすべてに相当した。
また、AO090005000664遺伝子と98%同一性を有するオルソログ遺伝子がコードするアミノ酸配列から428位の欠失(1DEL)、427位~429位の欠失(3DEL)、427~440位の欠失(14DEL)、427~444位の欠失(18DEL)、及び419位から444位の欠失(26DEL)を有するように改変された遺伝子を有するA.sojaeにおいて、エルゴチオネイン産生が亢進することが示された(図7B)。
さらに、AO090005000664遺伝子に対し、67%同一性しか有さないAn16g07990遺伝子から、441位~443位の欠失(3DEL)、441位~458位の欠失(18DEL)、433位~458位の欠失(26DEL)、433位~489位の欠失(57DEL)、および418位~529位の欠失(112DEL)を有するように改変された遺伝子を有するA.nigerにおいて、エルゴチオネイン産生が亢進することが示された(図7D)。
また、AO090005000664遺伝子と98%同一性を有するオルソログ遺伝子がコードするアミノ酸配列から428位の欠失(1DEL)、427位~429位の欠失(3DEL)、427~440位の欠失(14DEL)、427~444位の欠失(18DEL)、及び419位から444位の欠失(26DEL)を有するように改変された遺伝子を有するA.sojaeにおいて、エルゴチオネイン産生が亢進することが示された(図7B)。
さらに、AO090005000664遺伝子に対し、67%同一性しか有さないAn16g07990遺伝子から、441位~443位の欠失(3DEL)、441位~458位の欠失(18DEL)、433位~458位の欠失(26DEL)、433位~489位の欠失(57DEL)、および418位~529位の欠失(112DEL)を有するように改変された遺伝子を有するA.nigerにおいて、エルゴチオネイン産生が亢進することが示された(図7D)。
AO090005000664遺伝子及びその対応する遺伝子について、G428位を含むW404~Q515に含まれる超二次構造ドメインの機能は依然として不明であるが、当該超二次構造ドメインの機能を少なくとも部分的に阻害することにより、エルゴチオネイン生産性を高めることができることが示された。なお、AO090005000664遺伝子における位置が言及されており、遺伝子が異なればその対応する位置が異なることは当業者であれば十分に理解することができる。
Claims (20)
- 配列番号1に含まれる保存領域を含む、配列番号1に対応する配列において、配列番号1のW404~Q515の機能ドメインに対応する機能ドメインに含まれる領域に、当該機能ドメインの機能を変調する変異を含む配列;又は
当該配列に少なくとも90%の同一性を有する配列
をコードし、エルゴチオネイン産生微生物に導入された場合にエルゴチオネイン産生を亢進する、核酸。 - 前記保存領域が、配列番号1の433位~440位の保存領域である、請求項1に記載の核酸。
- 前記当該機能ドメインの機能を変調する変異が、W404~Q515の機能ドメインに対応する機能ドメインに含まれる領域の欠失である、請求項1に記載の核酸。
- 前記欠失された領域が、配列番号1の428位に対応する位置を含む、請求項1に記載の核酸。
- 前記欠失された領域が、1~120個のアミノ酸の欠失からなる、請求項1に記載の核酸。
- 前記欠失された領域が、配列番号1の427位~429位、427位~440位、427位~444位、及び419位~444位からなる群から選ばれる少なくとも1の領域に対応する領域である、請求項4に記載の核酸。
- 前記欠失された領域が、配列番号1の431位~475位、419位~475位、及び404位~515位からなる群から選ばれる少なくとも1の領域に対応する領域である、請求項3に記載の核酸。
- 前記当該機能ドメインの機能を変調する変異が、配列番号1の428位に対応する位置のG及び/又は459位に対応する位置のCにおいて変異を有する、請求項1に記載の核酸。
- 前記核酸が、アスペルギルス属、ペニシリウム属、ラサムソニア属、タラロマイシス属からなる群から選ばれる微生物由来の核酸である、請求項1に記載の核酸。
- 請求項1~9のいずれか一項に記載の核酸を含むベクター。

- 請求項11に記載の微生物を培養することを含む、エルゴチオネインの製造方法。
- エルゴチオネイン産生能を亢進された微生物の製造方法であって、配列番号1の433位~440位の保存領域を含む、配列番号1に対応する配列において、配列番号1のW404~Q515の機能ドメインに対応する機能ドメインに含まれる領域に、当該機能ドメインの機能を変調する変異を導入することを含み、前記微生物が、アスペルギルス属、ペニシリウム属、ラサムソニア属及びタラロマイシス属からなる群から選ばれるエルゴチオネイン生成微生物である、前記製造方法。
- 前記当該機能ドメインの機能を変調する変異が、W404~Q515の機能ドメインに対応する機能ドメインに含まれる領域の欠失である、請求項13に記載の製造方法。
- 前記保存領域が、配列番号1の433位~440位の保存領域である、請求項13に記載の製造方法。
- 前記欠失された領域が、配列番号1の428位に対応する位置を含む、請求項14に記載の製造方法。
