上述の通り、アシタバから得られる黄汁にはカルコンが含まれているが、カルコンは難水溶性であり、濃度が高くなると、製剤とした際の分散安定性が悪くなる。よって、従来のカルコンを含有する製剤は、カルコンを低濃度で含有するものであった。
しかしながら、上述のようにカルコンは様々な生理活性を有する成分であるため、カルコン濃度が高く、且つ、分散安定性の高い製剤とすることが求められていた。
そこで、本発明は、カルコン濃度が高く、且つ、分散安定性の高い製剤を提供することを目的とする。
本発明者らは、鋭意検討の結果、高濃度のカルコンに対して、特定の可溶化成分を共存させることにより、分散安定性が改善されることを見出し、かかる知見に基づいて、本発明を完成するに至った。
本発明は、以下の態様を含む。

本発明によれば、高濃度のカルコンに対して、特定の可溶化成分を共存させることにより、分散安定性が改善された製剤を提供することができる。
[カルコン含有製剤]
本発明において、製剤は、(A)カルコン、及び、(B)ショ糖脂肪酸エステル、グリセリン脂肪酸エステル、ポリオキシエチレンソルビタン脂肪酸エステル、酵素分解レシチン、及び、サポニンからなる群より選択される少なくとも1種の可溶化成分を含有する。
本発明において、製剤は、(A)カルコン、及び、(B)ショ糖脂肪酸エステル、グリセリン脂肪酸エステル、ポリオキシエチレンソルビタン脂肪酸エステル、酵素分解レシチン、及び、サポニンからなる群より選択される少なくとも1種の可溶化成分を含有する。
(A成分:カルコン)
上述のように、アシタバ(明日葉、学名:Angelica keiskei)の葉、茎、根等の部位には、粘度の高い濃黄色の汁(本明細書において、黄汁ともいう)が含まれている。黄色を呈する主成分としては、カルコンが知られている。

カルコンは、基本骨格として、下記式(1)で表される2’,4-ジヒドロキシカルコン骨格を有することが知られている。
カルコンは、上記式1において、上記基本骨格における3’位にゲラニル基を有するか、及び/又は、4’位に低級アルコキシ基を有することが好ましい。
カルコン類としては、例えば、アシタバ由来のカルコンが好ましく、4-ヒドロキシデリシン、及び、キサントアンゲロールからなる群より選択される少なくとも1種のカルコンがより好ましい。
カルコン類のうち、4-ヒドロキシデリシンは、上記基本骨格における4’位にメトキシ基を有するものであり、下記式(2)で表される。
キサントアンゲロールは、上記基本骨格における3’位にゲラニル基を有するものであり、下記式(3)で表される。
アシタバ由来のカルコンにおいて、4-ヒドロキシデリシンの1質量部に対するキサントアンゲロールの含有量は、本発明の効果を奏する限り、制限はされないが、例えば、0.1~10質量部、0.5~5質量部、1.0~3質量部、1.5~2.5質量部等が挙げられる。
カルコンは、特に製造方法や入手方法は限定されないが、例えば、アシタバ由来であってもよく、公知の方法を用いて合成して用いてもよく、市販品を用いてもよい。
アシタバ由来のカルコンとして、アシタバ由来の黄汁を用いることも可能である。アシタバ由来の黄汁は、アシタバの全草又は一部、例えば、葉部、茎部、根部、種子、及び、花部からなる群より選択される少なくとも1種から得ることができる。アシタバ由来の黄汁固形分としては、例えば、アシタバの全草、根、茎、葉等に対して、粉砕、破砕、摩砕、圧搾等の処理を行った処理物を用いることが挙げられる。
また、アシタバ由来のカルコンとして、上記処理物や黄汁に対して、溶媒で抽出した抽出物を用いることも可能である。アシタバ由来のカルコンとして抽出物を用いる場合は、その抽出方法は特に制限されるものではない。アシタバの抽出物は、アシタバの全草又は一部、例えば、葉部、茎部、根部、種子、及び、花部からなる群より選択される少なくとも1種の新鮮な植物体又は乾燥物から、あるいはそれらを適宜裁断し、破砕又は粉砕した粉末から、溶媒により抽出される。
抽出に用いる溶媒としては、水、有機溶媒又は含水有機溶媒が挙げられる。適宜の抽出溶媒を用いることにより、アシタバの水抽出物、有機溶媒抽出物又は含水有機溶媒抽出物を得ることができる。これらの抽出溶媒は、限定はされないが、水、メタノール、エタノール、エチレングリコール、1,3-ブチレングリコール、イソプロピレングリコール、プロピレングリコール、グリセリン、酢酸エチル、イソプロピルアルコール、テトラヒドロフラン、n-プロパノール、メチルエチルケトン、ジオキサン、アセトン、アセトニトリル、酢酸、ジメチルホルムアミド、n-ヘキサン又はこれらの混合物が好ましい。より高い本発明の効果を得ることの観点から、これらの抽出溶媒のうち、水、エタノール、又は、含水エタノールが特に好ましい。溶媒の濃度は、限定はされないが、より高い抽出効率を得ることの観点から、例えば、100%~40%、好ましくは100%~60%、より好ましくは100%~80%の濃度で用いることができる。抽出温度は、限定はされないが、より高い抽出効率を得ることの観点から、例えば、20~100℃とすることができ、22~80℃が好ましく、25~60℃がより好ましい。抽出時間は、限定はされないが、より高い抽出効率を得ることの観点から、例えば、5分以上14日以内とすることができ、10分以上7日以内が好ましく、15分以上5日以内がより好ましい。
抽出溶媒を加える前に、アシタバに、酸、アルカリ、または酵素を加えることで、抽出効率を高めることも可能である。
抽出溶媒を加えて撹拌する操作は、公知の方法を用いることができるが、例えば、アシタバの裁断物又は粉砕物と抽出溶媒とを含む抽出用容器を回転させる方法、前記抽出用容器を磁気式又は機械式の撹拌装置に設置して混合する方法、前記抽出用容器を振とうさせる方法等が挙げられる。
抽出効率を高めるためには、同種又は複数種の抽出溶媒を用いた多段階抽出を行うことも可能である。多段階抽出を行う場合は、第1段階の抽出において得られた残渣に、さらに同種又は複数種の抽出溶媒を加え、室温ないし加熱を行った後、抽出上清からアシタバのエキスを分離抽出することができる。
抽出上清をそのままアシタバ抽出物として用いることができる。さらには、抽出上清からアシタバ由来黄汁固形分を分離抽出することもできる。この分離抽出段階においては、公知の方法を採用することができ、例えば、ろ過、遠心分離、吸引、圧搾等を行うことが可能である。
希釈や濃縮の前後等に、さらに精製処理に付することにより、精製物としてアシタバ由来黄汁固形分を調製することができる。精製処理には、上記の溶媒による抽出以外に、当業者に公知な方法であるクロマトグラフ法、イオン交換クロマトグラフ法等を単独で、または組み合わせて用いることができる。
クロマトグラフ法を用いる場合であっては、例えば、順相若しくは逆相の担体又はイオン交換樹脂を用いるカラムクロマトグラフィー、高速液体クロマトグラフィー、薄層クロマトグラフィー又は遠心液体クロマトグラフィー等のいずれか、又はそれらを組み合わせて用いる方法が挙げられる。クロマトグラフ法を用いる場合の担体、溶出溶媒等の精製条件は、各種のクロマトグラフ法に対応して適宜選択することができる。
(A)成分の含有量は、より高い本発明の効果を得る観点から、製剤全量を基準として、0.5質量%以上であり、0.6質量%以上、0.7質量%以上、0.8質量%以上、0.9質量%以上、1.0質量%以上、1.1質量%以上、1.2質量%以上、1.3質量%以上、1.4質量%以上、1.5質量%以上、1.6質量%以上、1.7質量%以上、1.8質量%以上、1.9質量%以上、1.0質量%以上、2.0質量%以上、2.2質量%以上、2.4質量%以上、2.6質量%以上、2.8質量%以上、3.0質量%以上等とすることも可能である。
また、(A)成分の含有量は、より高い本発明の効果を得る観点から、製剤全量を基準として、40質量%以下とすることができ、35質量%以下、32質量%以下、30質量%以下、28質量%以下、25質量%以下、22質量%以下、20質量%以下、15質量%以下、12質量%以下、10質量%以下、5質量%以下等とすることも可能である。
(A)成分の含有量は、より高い本発明の効果を得る観点から、製剤全量を基準として、例えば、0.5~40質量%とすることができ、0.6~40質量%、0.7~40質量%、0.8~40質量%、0.9~40質量%、1.0~40質量%、1.1~40質量%、1.2~40質量%、1.3~40質量%、1.4~40質量%、1.5~40質量%、0.5~35質量%、0.6~35質量%、0.7~35質量%、0.8~35質量%、0.9~35質量%、1.0~35質量%、1.1~35質量%、1.2~35質量%、1.3~35質量%、1.4~35質量%、1.5~35質量%、0.5~32質量%、0.6~32質量%、0.7~32質量%、0.8~32質量%、0.9~32質量%、1.0~32質量%、1.1~32質量%、1.2~32質量%、1.3~32質量%、1.4~32質量%、1.5~32質量%、0.5~30質量%、0.6~30質量%、0.7~30質量%、0.8~30質量%、0.9~30質量%、1.0~30質量%、1.1~30質量%、1.2~30質量%、1.3~30質量%、1.4~30質量%、1.5~30質量%等とすることも可能である。また、別の実施態様において、(A)成分の含有量は、製剤全量を基準として、例えば、0.5~30質量%とすることができ、0.6~25質量%、0.7~20質量%、0.8~15質量%、0.9~10質量%、1.0~5質量%等とすることも可能である。
(B成分:可溶化成分)
後述の実施例に示されるように、高濃度のカルコンに対して、B成分として、ショ糖脂肪酸エステル、グリセリン脂肪酸エステル、ポリオキシエチレンソルビタン脂肪酸エステル、酵素分解レシチン、及び、サポニンからなる群より選択される少なくとも1種の可溶化成分を共存させることにより、分散安定性が改善されることが見出されている。(B)成分は、限定されないが、ショ糖脂肪酸エステル、グリセリン脂肪酸エステル、酵素分解レシチン、及び、サポニンからなる群より選択される少なくとも1種が好ましく、ショ糖脂肪酸エステル、グリセリン脂肪酸エステル、及び、サポニンからなる群より選択される少なくとも1種がより好ましい。これらのB成分は、合成して用いてもよく、市販品を用いてもよい。
後述の実施例に示されるように、高濃度のカルコンに対して、B成分として、ショ糖脂肪酸エステル、グリセリン脂肪酸エステル、ポリオキシエチレンソルビタン脂肪酸エステル、酵素分解レシチン、及び、サポニンからなる群より選択される少なくとも1種の可溶化成分を共存させることにより、分散安定性が改善されることが見出されている。(B)成分は、限定されないが、ショ糖脂肪酸エステル、グリセリン脂肪酸エステル、酵素分解レシチン、及び、サポニンからなる群より選択される少なくとも1種が好ましく、ショ糖脂肪酸エステル、グリセリン脂肪酸エステル、及び、サポニンからなる群より選択される少なくとも1種がより好ましい。これらのB成分は、合成して用いてもよく、市販品を用いてもよい。
ショ糖脂肪酸エステルとしては、本発明の効果を奏する限り、制限はされないが、例えば、ショ糖ラウリン酸エステル、ショ糖ミリスチン酸エステル、ショ糖パルミチン酸エステル、ショ糖ステアリン酸エステル、ショ糖オレイン酸エステル、ショ糖ベヘニン酸エステル、ショ糖エルカ酸エステル等が挙げられ、1種単独でもよく、2種以上を組み合わせて用いても良い。ショ糖脂肪酸エステルの市販品としては、例えば、リョートーシュガーエステル(三菱ケミカル社製)等が挙げられる。
グリセリン脂肪酸エステルとしては、本発明の効果を奏する限り、制限はされないが、例えば、モノグリセリン脂肪酸エステル(モノステアリン酸グリセリル等)、ポリグリセリン脂肪酸エステル(デカグリセリンステアリン酸エステル、ペンタグリセリンステアリン酸エステル、ジグリセリンパルミチン酸エステル等)、ポリグリセリン縮合リシノール酸エステル、有機酸モノグリセリド等が挙げられ、1種単独でもよく、2種以上を組み合わせて用いても良い。グリセリン脂肪酸エステルの市販品としては、例えば、、ポエムDO-100V(理研ビタミン社製)、リョートーポリグリエステル(三菱ケミカル社製)、サンソフトQ(太陽化学社製)等が挙げられる。
ポリオキシエチレンソルビタン脂肪酸エステルとしては、本発明の効果を奏する限り、制限はされないが、例えば、例えば、モノラウリン酸ポリオキシエチレンソルビタン、モノミリスチン酸ポリオキシエチレンソルビタン、モノパルミチン酸ポリオキシエチレンソルビタン、モノステアリン酸ポリオキシエチレンソルビタン、及びモノオレイン酸ポリオキシエチレンソルビタン等が挙げられ、1種単独でもよく、2種以上を組み合わせて用いても良い。ポリオキシエチレンソルビタン脂肪酸エステルの市販品としては、例えば、エマゾール(花王社製)等が挙げられる。
酵素分解レシチンとしては、本発明の効果を奏する限り、制限はされないが、レシチンに対して、ホスホリパーゼA2を作用させ、2-位における脂肪酸のエステル結合を加水分解したものであればよい。原料となるレシチンは制限されないが、例えば、大豆レシチン、卵黄レシチン等が挙げられ、1種単独でもよく、2種以上を組み合わせて用いても良い。酵素分解レシチンの市販品としては、例えば、サンレシチン(太陽化学社製)等が挙げられる。
サポニンは、サポゲニンをアグリコンとする配糖体の総称であり、本発明の効果を奏する限り、制限はされないが、例えば、バラ科キラヤの樹皮から抽出されたキラヤサポニン等が挙げられ、1種単独でもよく、2種以上を組み合わせて用いても良い。サポニンの市販品としては、例えば、キラヤニン(丸善製薬社製)等が挙げられる。
(B)成分の総含有量は、より高い本発明の効果を得る観点から、製剤全量を基準として、例えば、0.01~95質量%とすることができ、0.05~94質量%が好ましく、0.1~93質量%がより好ましく、0.5~92質量%が更に好ましい。また、別の実施態様において、(B)成分の総含有量は、製剤全量を基準として、例えば、0.01~40質量%、0.05~40質量%、0.1~40質量%、0.5~40質量%、0.01~30質量%、0.05~30質量%、0.1~30質量%、0.5~30質量%、0.01~25質量%、0.05~25質量%、0.1~25質量%、0.5~25質量%、0.01~20質量%、0.05~20質量%、0.1~20質量%、0.5~20質量%、0.01~15質量%、0.05~15質量%、0.1~15質量%、0.5~15質量%等とすることも可能である。
(B)成分として、ショ糖脂肪酸エステルを含有する場合、ショ糖脂肪酸エステル単独の含有量は、製剤全量を基準として、例えば、0.01~95質量%とすることができ、0.05~94質量%が好ましく、0.1~93質量%がより好ましく、0.5~92質量%が更に好ましく、0.5~50質量%が更により好ましく、0.5~45質量%が更により好ましく、0.5~40質量%が更により好ましく、0.5~35質量%が更により好ましい。また、別の実施態様において、ショ糖脂肪酸エステル単独の含有量は、製剤全量を基準として、例えば、0.01~30質量%、0.05~30質量%、0.1~30質量%、0.5~30質量%、1.0~30質量%、0.01~28質量%、0.05~28質量%、0.1~28質量%、0.5~28質量%、1.0~28質量%、0.01~26質量%、0.05~26質量%、0.1~26質量%、0.5~26質量%、1.0~26質量%等とすることも可能である。
(B)成分として、グリセリン脂肪酸エステルを含有する場合、グリセリン脂肪酸エステル単独の含有量は、製剤全量を基準として、例えば、0.01~95質量%とすることができ、0.05~94質量%が好ましく、0.1~93質量%がより好ましく、0.5~92質量%が更に好ましく、0.5~50質量%が更により好ましく、0.5~45質量%が更により好ましく、0.5~40質量%が更により好ましく、0.5~35質量%が更により好ましい。また、別の実施態様において、グリセリン脂肪酸エステル単独の含有量は、製剤全量を基準として、例えば、0.01~30質量%、0.05~30質量%、0.1~30質量%、0.5~30質量%、1.0~30質量%、0.01~28質量%、0.05~28質量%、0.1~28質量%、0.5~28質量%、1.0~28質量%、0.01~26質量%、0.05~26質量%、0.1~26質量%、0.5~26質量%、1.0~26質量%等とすることも可能である。
(B)成分として、ポリオキシエチレンソルビタン脂肪酸エステルを含有する場合、ポリオキシエチレンソルビタン脂肪酸エステル単独の含有量は、製剤全量を基準として、例えば、0.01~99質量%とすることができ、0.05~94質量%が好ましく、0.1~93質量%がより好ましく、1.0~92質量%が更に好ましく、3~91質量%が特に好ましい。また、別の実施態様において、ポリオキシエチレンソルビタン脂肪酸エステル単独の含有量は、製剤全量を基準として、例えば、50~99質量%、60~99質量%、70~99質量%、80~99質量%、50~95質量%、60~95質量%、70~95質量%、80~95質量%、50~92質量%、60~92質量%、70~92質量%、80~92質量%等とすることも可能である。
(B)成分として、酵素分解レシチンを含有する場合、酵素分解レシチン単独の含有量は、製剤全量を基準として、例えば、0.01~16質量%とすることができ、0.05~14質量%が好ましく、0.1~12質量%がより好ましく、0.5~10質量%が更に好ましい。
(B)成分として、サポニンを含有する場合、サポニン単独の含有量は、製剤全量を基準として、例えば、0.01~16質量%とすることができ、0.05~14質量%が好ましく、0.1~12質量%がより好ましく、0.5~10質量%が更に好ましい。また、別の実施態様において、サポニン単独の含有量は、製剤全量を基準として、例えば、0.01~25質量%、0.05~25質量%、0.1~25質量%、0.5~25質量%、1.0~25質量%、0.01~20質量%、0.05~20質量%、0.1~20質量%、0.5~20質量%、1.0~20質量%、0.01~15質量%、0.05~15質量%、0.1~15質量%、0.5~15質量%、1.0~15質量%等とすることも可能である。
(A)成分と(B)成分との含有比率は、特に限定されないが、本発明の効果を顕著に奏する観点から、(A)成分の1質量部に対して、(B)成分の総含有量は、例えば、0.1~100質量部とすることができ、0.5~80質量部が好ましく、0.8~60質量部がより好ましく、1.0~50質量部が更に好ましく、1~45質量部が特に好ましい。また、別の実施態様において、(A)成分の1質量部に対して、(B)成分の総含有量は、例えば、0.1~40質量部、0.5~40質量部、1.0~40質量部、1.5~40質量部、0.1~30質量部、0.5~30質量部、1.0~30質量部、1.5~30質量部、0.1~20質量部、0.5~20質量部、1.0~20質量部、1.5~20質量部、0.1~15質量部、0.5~15質量部、1.0~15質量部、1.5~15質量部等とすることも可能である。
(B)成分として、ショ糖脂肪酸エステルを含有する場合、(A)成分の1質量部に対して、ショ糖脂肪酸エステルの総含有量は、例えば、0.1~100質量部とすることができ、0.5~90質量部が好ましく、0.8~80質量部がより好ましく、1.0~70質量部が更に好ましく、1.0~60質量部が特に好ましい。
(B)成分として、グリセリン脂肪酸エステルを含有する場合、(A)成分の1質量部に対して、グリセリン脂肪酸エステルの総含有量は、例えば、0.1~100質量部とすることができ、0.5~90質量部が好ましく、0.8~80質量部がより好ましく、0.8~70質量部が更に好ましく、0.8~60質量部が特に好ましい。
(B)成分として、ポリオキシエチレンソルビタン脂肪酸エステルを含有する場合、(A)成分の1質量部に対して、ポリオキシエチレンソルビタン脂肪酸エステルの総含有量は、例えば、0.1~50質量部とすることができ、0.5~48質量部が好ましく、0.8~46質量部がより好ましく、1.0~44質量部が更に好ましい。
(B)成分として、酵素分解レシチンを含有する場合、(A)成分の1質量部に対して、酵素分解レシチンの総含有量は、例えば、0.1~16質量部とすることができ、0.5~14質量部が好ましく、0.8~12質量部がより好ましく、1.0~10質量部が更に好ましい。
(B)成分として、サポニンを含有する場合、(A)成分の1質量部に対して、サポニンの総含有量は、例えば、0.1~100質量部とすることができ、0.5~90質量部が好ましく、0.8~80質量部がより好ましく、0.8~70質量部が更に好ましく、0.8~60質量部が特に好ましい。
(C成分)
上記成分の他、本発明の効果を顕著に奏する観点から、更に、(C)成分として、プロピレングリコール、水飴、還元水飴、グリセリン、糖アルコール、単糖類、デキストリン、及び、マルトデキストリンからなる群より選択される少なくとも1種を含有することが好ましく、プロピレングリコール、水飴、還元水飴、グリセリン、糖アルコール、及び、単糖類からなる群より選択される少なくとも1種を含有することがより好ましく、プロピレングリコール、水飴、グリセリン、マルチトール、エリスリトール、ソルビトール、キシリトール、グルコース、フルクトース、ガラクトース、及び、マンノースからなる群より選択される少なくとも1種を含有することが更に好ましい。これらのB成分は、合成して用いてもよく、市販品を用いてもよい。

水飴の市販品としては、例えば、テトラスイート(日本コーンスターチ社製)等が挙げられる。還元水飴の市販品としては、例えば、エスイー600(物産フード社製)等が挙げられる。デキストリンの市販品としては、例えば、ファイバーソル2(難消化性デキストリン)、マックス1000(いずれも松谷化学社製)、CAVAMAX(シクロデキストリン、シクロケム社製)等が挙げられる。マルトデキストリンの市販品としては、例えば、マックス2000N、TK-16(松谷化学社製)等が挙げられる。
(C)成分の総含有量は、より高い本発明の効果を得る観点から、製剤全量を基準として、例えば、0.01~95質量%とすることができ、0.05~94質量%が好ましく、0.1~93質量%がより好ましく、0.5~92質量%が更に好ましい。また、別の実施態様において、(C)成分の総含有量は、製剤全量を基準として、例えば、1.0~95質量%、5~95質量%、10~95質量%、15~95質量%、20~95質量%、25~95質量%、30~95質量%、35~95質量%、40~95質量%、45~95質量%、50~95質量%等とすることも可能である。

本発明において、製剤は、水性製剤(基剤又は担体として水性ないしは親水性のものを主に含む)であってもよく、油性製剤(基剤又は担体として油性ないしは疎水性のものを主に含む)であっても良い。
水性製剤の場合の水の含有量は、製剤全量を基準として、例えば、50質量%以上が好ましく、75質量%以上がより好ましく、90質量%以上がさらにより好ましい。また、95質量%以上、又は98質量%以上であってもよい。また、基剤又は担体が水のみからなっていてもよい。
油性製剤の場合の水の含有量は、製剤全量を基準として、例えば、50質量%未満が好ましく、30質量%以下がより好ましく、20質量%以下が更に好ましく、10質量%以下が特に好ましい。

飲食品組成物に含まれる製剤の含量は、0.01~100質量%であることが好ましく、0.1~50質量%であることがさらに好ましく、0.2~25質量%であることがさらにより好ましい。
<医薬組成物>

また、このような種々の剤型の医薬製剤を調製するには、他の薬学的に許容される賦形剤、結合剤、増量剤、崩壊剤、界面活性剤、滑沢剤、分散剤、緩衝剤、保存剤、嬌味剤、香料、被膜剤、担体、希釈剤、他の薬効成分等を適宜組み合わせることができる。また、これらの投与形態のうち、好ましい形態は経口投与であり、経口用液体製剤は、嬌味剤、緩衝剤、安定化剤等を加えて常法により調製することができる。
医薬組成物に含まれる製剤の含量は、0.01~100質量%であることが好ましく、0.1~50質量%であることがさらに好ましく、0.2~25質量%であることがさらにより好ましい。

本発明の製剤は、飼料であり得る。飼料としては、ウサギ、ラット、マウス等に用いる小動物用飼料、犬、猫等に用いるペットフード等の飼料等が挙げられ、ペットが摂食できる限り、上記飲食品組成物と同様の形態に調製できる。

表1及び図2に示す通り、実施例の被験製剤では、カルコンが、製剤全量を基準として、0.5質量%以上である場合であっても、カルコンの分散性が改善された。限定はされないが、成分Bとしてポリソルベートを用いた場合は、カルコンの分散性改善効果だけでなく、透明性の改善効果も確認された。
次に、限定はされないが、成分Bをショ糖脂肪酸エステルとし、更に各種の(C)成分を含有させて、同様に溶解性評価を行った。各成分の種類、及び、含有量等は下記表の通りである。各実施例の被験製剤の溶解状態を、表2及び図3に示す。糖アルコールとしては、エリスリトールを用いた(以下の実施例でも同様)。
表2及び図3に示す通り、実施例の被験製剤では、カルコンが、製剤全量を基準として、0.5質量%以上である場合であっても、(B)可溶化成分を含有させることで、カルコンの分散性が改善された。限定はされないが、更に多価アルコールを含有させることにより、カルコンの分散性改善効果だけでなく、透明性の改善効果も確認された。
次に、限定はされないが、成分Cをプロピレングリコール(PG)とし、更に(B)可溶化成分を含有させて、同様に溶解性評価を行った。各成分の種類、及び、含有量等は下記表の通りである。各実施例の被験製剤の溶解状態を、表3及び図4~5に示す。
表3及び図4~5に示す通り、実施例の被験製剤では、カルコンが、製剤全量を基準として、0.5質量%以上である場合であっても、(B)可溶化成分を含有させることで、カルコンの分散性が改善された。限定はされないが、更に多価アルコールを含有させることにより、カルコンの分散性改善効果だけでなく、透明性の改善効果も確認された。
次に、限定はされないが、成分Bをグリセリン脂肪酸エステルとし、更に各種の(C)成分を含有させて、同様に溶解性評価を行った。各成分の種類、及び、含有量等は下記表の通りである。各実施例の被験製剤の溶解状態を、表4及び図6に示す。

次に、限定はされないが、成分Bをサポニン(キラヤ由来)とし、更に各種の(C)成分を含有させて、同様に溶解性評価を行った。各成分の種類、及び、含有量等は下記表の通りである。各実施例の被験製剤の溶解状態を、表5及び図7に示す。
表5及び図7に示す通り、実施例の被験製剤では、カルコンが、製剤全量を基準として、0.5質量%以上である場合であっても、(B)可溶化成分を含有させることで、カルコンの分散性が改善された。
次に、限定はされないが、成分Cをプロピレングリコール(PG)とし、更に(B)可溶化成分を含有させて、同様に溶解性評価を行った。各成分の種類、及び、含有量等は下記表の通りである。各実施例の被験製剤の溶解状態を、表6及び図8に示す。

次に、限定はされないが、成分Bをショ糖脂肪酸エステルとし、更に各種の(C)可溶化成分を含有させて、同様に溶解性評価を行った。各成分の種類、及び、含有量等は下記表の通りである。各実施例の被験製剤の溶解状態を、表7及び図9に示す。

[試験例2.甘味評価試験]
別の実施態様において、アシタバエキスを配合することによる、甘味の増強効果を評価した。

(官能評価方法)
0.4%ショ糖水溶液を調整し、アシタバエキス(実施例47記載の被験製剤)を、それぞれ0.005%、0.02%(質量%)となるように溶解した飲料について、訓練されたパネリスト7名によって官能評価を行った。
0.4%ショ糖水溶液に対して、(i)甘味の強さ(食した直後)、および、(ii)甘味の継続に関して相対評価を行った。
なお、甘味の継続としては食後約1~2秒後に残っている甘味の強さについてコントロールと比較して評価した。評価基準は以下に示した。
(評価基準)
1 弱い
2 弱く感じる
3 コントロール(0.4%ショ糖水溶液)と同程度
4 強く感じる
5 かなり強い
0.4%ショ糖水溶液を調整し、アシタバエキス(実施例47記載の被験製剤)を、それぞれ0.005%、0.02%(質量%)となるように溶解した飲料について、訓練されたパネリスト7名によって官能評価を行った。
0.4%ショ糖水溶液に対して、(i)甘味の強さ(食した直後)、および、(ii)甘味の継続に関して相対評価を行った。
なお、甘味の継続としては食後約1~2秒後に残っている甘味の強さについてコントロールと比較して評価した。評価基準は以下に示した。
(評価基準)
1 弱い
2 弱く感じる
3 コントロール(0.4%ショ糖水溶液)と同程度
4 強く感じる
5 かなり強い
表11に示す通り、カルコン含有製剤において、飲食品の甘味を増強する効果が確認された。試験例1に記載のように、当該製剤中のカルコンは、4-ヒドロキシデリシン、及び、キサントアンゲロールを含有しており、4-ヒドロキシデリシンの1質量部に対するキサントアンゲロールの含有量は、約1.7~1.9質量部である。
[製造例]
以下、各種の実施態様を挙げて本発明を説明するが、本発明の範囲はこれらにより限定されるものではない。なお、特に断りのない限り、「%」、「部数」等の数値は、質量基準の数値を表す。配合原料におけるアシタバエキスには、上記実施例におけるカルコンが含まれている。

上記処方において、1~3までを50℃で撹拌溶解する(A相)。4~6までを室温で撹拌溶解する(B相)。B相をパドルでかき混ぜながらA相を加え混合して、水系化粧水を調製する。
上記処方において、A、Bをそれぞれビーカーに秤取し、混合する。Aを撹拌しながらBを徐々に添加し、混合して、マウスウォッシュを調製する。
上記処方において、1~4を70℃まで加温し溶解する(A相)。5~7を70℃まで加温し溶解する(B相)。A相をホモミキサーでかき混ぜながらB相を加え乳化する(2500rpm、3分間)。その後パドルでかき混ぜながら30℃まで冷却して、バスミルクを調製する。
上記処方において、1~6までを室温で撹拌溶解する(A相)。7と8を室温で撹拌溶解する(B相)。B相をパドルでかき混ぜながらA相を加え溶解して、ヘアトニックを調製する。
上記処方において、各配合原料を混合し、殺菌してボトルに充填して、漬物用調味液を調製する。
上記処方において、各配合原料を混合し、殺菌してボトルに充填して、麺つゆを調製する。
上記処方において、各配合原料を混合し、140℃、45秒で殺菌してペットボトルに充填する。

上記処方において、烏龍茶を90℃の水で10分間抽出し、その他の原料を添加して、137℃、40秒殺菌してペットボトルに充填する。
上記処方において、ルイボスを90℃の水で10分間抽出し、その他の原料を添加して、140℃、50秒殺菌してペットボトルに充填する。
Claims (6)
- (A)カルコン、及び、
(B)ショ糖脂肪酸エステル、グリセリン脂肪酸エステル、ポリオキシエチレンソルビタン脂肪酸エステル、酵素分解レシチン、及び、サポニンからなる群より選択される少なくとも1種の可溶化成分
を含有し、
前記カルコンが、製剤全量を基準として、0.5質量%以上である、製剤。 - 前記カルコンが、アシタバ由来である、請求項1に記載の製剤。
- さらに、(C)プロピレングリコール、水飴、還元水飴、グリセリン、糖アルコール、単糖類、デキストリン、及び、マルトデキストリンからなる群より選択される少なくとも1種を含有する、請求項1又は2に記載の製剤。
- 前記(A)成分1質量部に対して、前記(B)成分の含有量が、0.1~100質量部である、請求項1~3のいずれか1項に記載の製剤。
- 前記(A)成分1質量部に対して、前記(C)成分の含有量が、0.01~100質量部である、請求項3又は4に記載の製剤。
- 請求項1~5のいずれか1項に記載の製剤を含有する、飲食品、化粧品、日用品、又は、医薬品。
