Abstract
본 발명은 균주 접종을 통해 유익성분을 함유하는 발효스프레드에 관한 것으로서, 보다 상세하게는 FT4-2 균주를 과일 또는 채소의 원액에 접종하여 배양 후 발효체와 혼합하여 상기 균주의 유익성분을 극대화하여 상기 발효체를 이용한 발효스프레드를 통해 해당 유익성분의 손실없는 섭취가 가능한 균주 접종을 통해 유익성분을 함유하는 발효스프레드에 관한 것이다. 유익성분을 극대화시키고 저당도로 제조된 발효잼 또는 발효스프레드와 이를 제조하는 방법을 통해, β-글루칸(β-glucan) 생성능, 알코올 생성능 및 β-글루코시다제(β-glucosidase) 분비능이 있으며, 우레아제를 생성하지 않는 특징을 가짐으로써, 인간 정상 세포의 면역기능을 활성화시켜 암세포의 증식과 재발을 억제하고 혈당과 혈중 콜레스테롤을 감소시키며 지질대사를 개선하여 체지방 형성과 축적을 억제할 수 있는 효과가 존재한다.
Description
본 발명은 균주 접종을 통해 유익성분을 함유하는 발효스프레드에 관한 것으로서, 보다 상세하게는 FT4-2 균주를 과일 또는 채소의 원액에 접종하여 배양 후 발효체와 혼합하여 상기 균주의 유익성분을 극대화하여 상기 발효체를 이용한 발효스프레드를 통해 해당 유익성분의 손실 없는 섭취가 가능한 균주 접종을 통해 유익성분을 함유하는 발효스프레드에 관한 것이다.
된장은 콩을 주재료로 사용하여 발효 숙성시킨 장류로서 단백질과 지방함량이 높아 찌개류, 반찬류 등을 만들때 사용하는 기호식품으로 애용되고 있다.
종래의 된장과 간장 제조방법은 각 지역마다 차이가 있으나, 통상적으로 메주콩을 깨끗한 물에 거품이 나지 않을 때까지 세척하고 하루 동안 물에 불린 다음, 다시 세척 후 흐르는 물에 깨끗이 세척하여 삶아준다. 삶아진 메주콩을 방아에 찧고 메주를 만든 후, 볏짚을 두툼하게 깔아 메주를 올려주고 다시 볏짚을 깔아 층층이 쌓아준다. 띄워진 메주를 항아리에 넣고 소금물과 고추, 감초, 숯 등을 넣은 후에 약 50일 숙성 후에 액체는 한번 더 끓인 후에 항아리에 넣어 숙성시키고, 된장은 다른 항아리에 넣어 다시 숙성시킨다. 여기서, 액체는 간장이고 덩어리는 된장이 된다.
또한, 상기 메주의 상태에서 빻아 메주가루, 엿기름, 쌀가루, 고춧가루, 소금 등과 섞어 항아리에 넣어 숙성시키는 고추장이 있다.
최근, 상기와 같은 된장, 고추장, 청국장, 간장 등과 같은 우리 전통 장류가 건강에 대한 관심이 높아짐에 따라 건강식품으로 각광받고 있다.
또한, 경제발전으로 인한 생활수준의 향상에 따라 맛뿐만 아니라, 건강과 영양까지 고려한 기능성 장류의 수요가 증가하고 있는 추세이며, K-한류의 열풍에 따라 전세계인들의 식탁에 해당 장류를 응용하여 편하게 취식할 수 있는 제품들이 폭발적으로 늘어나고 있는 추세이다.
이에 전통 장류를 이용하여 서구인들의 입맛에 익숙하되 유익성분을 극대화시키고 저당도로 제조된 발효잼 또는 발효스프레드와 이를 제조하는 방법이 개발되고 있는 실정이다.
본 발명의 목적은 전통 장류를 이용하여 서구인들의 입맛에 익숙하되 유익성분(β-글루칸(β-glucan) 생성능, 알코올 생성능 및 β-글루코시다제(β-glucosidase) 분비능)을 극대화시키되, 우레아제를 생성하지 않는 특징을 가지는 저당도로 제조된 발효잼 또는 발효스프레드와 이의 제조 방법을 제공하는 것이다.
상기 목적을 달성하기 위하여, 본 발명은 FT4-2 균주 접종 및 증균과정을 거쳐 생성되는 효모액, 상기 효모액에 소정 비율로 혼합되는 발효체 및 상기 효모액 및 발효체와 함께 혼합되는 대체당을 포함하는 균주 접종을 통해 유익성분을 함유하는 발효스프레드를 제공하는 것이 가능하다.
상기 효모액은 과일 또는 채소를 착즙한 원액에 효모를 접종하는 것이 바람직하다.
상기 채소는 토마토를 포함할 수 있다.

보다 구체적으로, YPD를 이용하여 FT4-2를 접종 후 섭씨30도에서 24시간 효모균(유익균)을 배양하는 전배양단계, 상기 전배양이 종료된 배양체를 멸균수로 3회 이상 세척하는 세척단계, 상기 세척완료된 배양체를 과일 또는 채소를 착즙한 원액에 접종하여 효모액을 형성하는 접종단계 및 상기 접종단계를 통해 형성된 효모액에 발효체 및 대체당을 소정 비율로 혼합하는 혼합단계를 포함하는 것을 특징으로 하는 균주 접종을 통해 유익성분을 함유하는 발효스프레드 제조방법이 제공될 수 있다.
이 경우, 상기 접종단계는 상기 배양체를 과일 또는 채소를 착즙한 원액에 접종 후 섭씨 30도에서 48시간 효모균을 배양하는 후배양단계를 더 포함할 수 있다.

도 1은 본 발명의 일 실시예를 나타내는 균주 접종을 통해 유익성분을 함유하는 발효스프레드의 구성을 표현하는 개략도이며, 도 2는 본 발명의 일 실시예를 나타내는 균주 접종을 통해 유익성분을 함유하는 발효스프레드의 제조방법을 나타내는 흐름도이다.
도 1 및 도 2를 참고하면, 본 발명의 균주 접종을 통해 유익성분을 함유하는 발효스프레드의 구체적인 제조방법의 플로우 및 제조되는 발효스프레드의 구성을 확인하는 것이 가능하다.
이하에서는 본 발명의 균주 접종을 통해 유익성분을 함유하는 발효스프레드 및 이의 제조방법에 대하여 보다 상세하게 기술하도록 한다.
본 발명은 FT4-2(기탁번호 KCCM12520P, 한국미생물보존센터) 효모 균주(10) 접종 및 증균과정을 거쳐 생성되는 효모액(100), 상기 효모액(100)에 소정 비율로 혼합되는 발효체(200) 및 상기 효모액(100) 및 발효체(200)와 함께 혼합되는 대체당(300)을 포함하는 균주 접종을 통해 유익성분을 함유하는 발효스프레드(400)가 제공될 수 있다.
보다 구체적으로는, 최근 된장, 고추장, 청국장, 간장 등과 같은 우리 전통 장류가 건강에 대한 관심이 높아짐에 따라 건강식품으로 각광받고 있으며, 또한, 경제발전으로 인한 생활수준의 향상에 따라 맛뿐만 아니라, 건강과 영양까지 고려한 기능성 장류의 수요가 증가하고 있는 추세이며, K-한류의 열풍에 따라 전세계인들의 식탁에 해당 장류를 응용하여 편하게 취식할 수 있는 제품들이 폭발적으로 늘어나고 있는 추세이다.
이에 전통 장류를 이용하여 서구인들의 입맛에 익숙하되 유익성분을 극대화시키고 저당도로 제조된 발효잼 또는 발효스프레드와 이를 제조하는 방법이 개발되고 있는 실정이다.
이러한 실정을 반영하여 기술하는 본 발명의 목적은, 전통 장류를 이용하여 서구인들의 입맛에 익숙하되 유익성분(β글루칸(β생성능, 알코올 생성능 및 β글루코시다제(β분비능)을 극대화시켜 인간 정상 세포의 면역기능을 활성화시켜 암세포의 증식과 재발을 억제하고 혈당과 혈중 콜레스테롤을 감소시키며 지질대사를 개선하여 체지방 형성과 축적을 억제할 수 있는 효과를 가지되, 우레아제를 생성하지 않아 암모니아 부산물의 생성을 방지할 수 있는 특징을 가지는 저당도로 제조된 발효잼 또는 발효스프레드와 이의 제조 방법을 제공하는 것이다.
이와 같은 효과를 발현하면서도 관능성이 향상된 발효스프레드의 구현을 위해, FT4-2(기탁번호 KCCM12520P, 한국미생물보존센터) 효모 균주(10)를 사용하며, 해당 FT4-2 효모 균주(10)의 수 극대화 및 배양체(20)와 효모액(100) 및 발효체(200)와의 혼합상태에서도 생명력과 효과를 극대화하기 위한 제조방법을 통한 발효스프레드가 제공될 수 있다.
FT4-2 효모 균주(10)는, 블루베리 등으로부터 분리되어 얻을 수 있는 효모이며, 특유의 효과로서, 유익성분(β-글루칸(β-glucan) 생성능, 알코올 생성능 및 β-글루코시다제(β-glucosidase) 분비능)을 극대화시켜 인간 정상 세포의 면역기능을 활성화시켜 암세포의 증식과 재발을 억제하고 혈당과 혈중 콜레스테롤을 감소시키며 지질대사를 개선하여 체지방 형성과 축적을 억제할 수 있는 효과를 가지되, 우레아제를 생성하지 않아 암모니아 부산물의 생성을 방지할 수 있는 효과를 가지므로 발효체를 이용한 스프레드 제조에 탁월한 효과를 발휘하는 효모 균주라 할 것이다. 또한, 와인 제조를 위한 스타터 균주 등 발효식품 관련 산업에 매우 유용하게 이용할 수 있으며, 발효식품 제조용 스타터(starter)란, 발효식품 제조를 위해 발효에 관여하는 미생물을 포함하는 제제 또는 조성물을 의미한다. 이 경우 발효식품 제조 시에 첨가함으로써 발효된 식품에서 생장할 수 있는 미생물 또는 우점종으로 생장할 수 있는 미생물을 제공하기 위하여 사용된다. 상기 식품 발효용 스타터를 사용하여 식품을 제조하는 경우, 상기 식품 발효용 스타터에 포함된 미생물에 의하여, 식품의 품질을 일정하게 조절하거나, 특정한 목적, 일 예로 식품에서 이취를 발생시키지 않거나, 감소시키는 목적을 달성할 수 있다.
도 2를 참고하면, 위의 FT4-2 효모 균주(10)를 활용한 구체적인 본 발명의 발효스프레드의 제조방법의 흐름을 파악하는 것이 가능하며, 아래에서는 이에 대해 보다 상세하게 기술한다.
우선, 미생물의 증식, 또는 배양을 목적으로 미생물이 일반적으로 생존하고 번식하는 자연 환경과 유사한 영양 상태를 유지하도록 고안된 물질로서 자라는데 필요한 모든 영양소를 첨가하여 만든 액체 및 고체의 물질인 YPD(yeast peptone dextrose)배지를 사용할 수 있으며, 배양 배지에서의 미생물 배양은 필수 영양소 선정, 산소 또는 배양에 필요한 기체, 수분, pH 및 온도를 비롯한 여러 가지 중요한 요인에 따라 달라지며, 중요 영양원소로 탄소, 질소, 무기 인산염 및 황, 미량 금속, 물 및 비타민 등이 포함된다. 이러한 YPD(yeast peptone dextrose)배지를 이용하여 FT4-2 효모 균주(10)를 배지에 접종 후 섭씨30도에서 24시간 효모균(유익균)을 배양하여 FT4-2 효모 균주(10)의 수 및 활성화를 극대화시키는 전배양단계(S100), 상기 전배양단계(S100)가 종료된 배양체(20)를 멸균수로 3회 이상 세척하여 잡균 및 이물질을 최대한 제거하고 세척하는 세척단계(S200), 상기 세척완료된 배양체(20)를 과일 또는 채소를 착즙한 원액에 접종하여 효모액(100)을 형성하는 접종단계(S300) 및 상기 접종단계(S300)를 통해 형성된 효모액(100)에 발효체(200) 및 대체당(300)을 소정 비율로 혼합하는 혼합단계(S400)를 포함하는 것을 특징으로 하는 균주 접종을 통해 유익성분을 함유하는 발효스프레드 제조방법이 제공될 수 있다.
이 경우, 상기 접종단계(S300)는 상기 배양체(20)를 과일 또는 채소를 착즙한 원액에 접종 후 섭씨 30도에서 48시간동안 FT4-2 효모 균주(10)를 배양하여 효모액(100)을 생성하는 후배양단계(S310)를 포함할 수 있으며, 이 과정을 통해 호모액 내 생균 수를 접종 0시간에 2.0 x 10n(n=6)마리에서, 접종 48시간 후 1.5 x 10n(n=8)마리로 약 100배 가량 생균 수를 대폭 증식시키는 것이 가능하다.
이러한 상기 후배양단계(S310)를 포함하는 접종단계(S300)가 종료 후, 섭씨 영하80도 이하로 상기 효모액(100)을 동결시키는 동결단계(S500)를 통해 상기 효모액의 추가 발효를 방지하고, 변질을 예방할 수 있다.
이 경우, 상기 동결단계(S500)는 동결 전 섭씨 120도로 15분 이상 상기 효모액(100)을 가열하여 멸균하는 멸균단계(S550)를 더 포함하는 것을 특징으로 하는 균주 접종을 통해 유익성분을 함유하는 발효스프레드 제조방법이 제공될 수 있다.
균주 접종을 통해 유익성분을 함유하는 발효스프레드의 성능 및 관능을 극대화하기 위하여 상기 효모액(100)은 과일 또는 채소를 착즙한 원액에 전배양된 FT4-2 효모 균주(10)를 접종하는 것을 특징으로 하는 균주 접종을 통해 유익성분을 함유하는 발효스프레드 및 제조방법이 제공될 수 있으며, 이 경우 상기 채소는 토마토를 포함하는 것이 가능하고, 상기 발효체(200)는 된장 또는 고추장을 포함하는 것을 특징으로 하는 균주 접종을 통해 유익성분을 함유하는 발효스프레드 및 제조방법이 제공될 수 있다.
구체적인 균주 접종을 통해 유익성분을 함유하는 발효스프레드의 성능 및 관능의 향상에 대하여는 아래의 표의 내용을 참고할 수 있다.
[실시예 1]우레아제(Urease) 생성여부확인
세포손상을 주는 기전으로 헬리코박터 파일로리(Helicobacter pylori) 감염 시 우레아제에 의해 생성되는 암모니아(ammonia)에 의해 세포 손상이 생기고, 헬리코박터 파일로리의 분비 독소인 Vac A(vacuolating cytotoxin gene A)에 의해 상피세포의 공포화가 발생하는데 호중구에서 분비된 염증매개물질과 활성 산소화물에 의해서도 세포가 손상된다. 상피세포의 손상은 세포의 위축성 변화를 일으키고 세포손상에 대한 보상기전으로 세포증식이 증가하며 활발하게 증식하는 세포는 발암물에 의하여 유전자가 손상을 받을 기회가 많아진다. 따라서 우레아제 생성 여부를 확인하기 위한 실험을 진행하였고, 측정 결과 FT4-2, FT4-7 및 FT4-140 균주에서 우레아제가 생성되지 않음을 확인하였다(표 1)

[실시예 2] β-글루코시다제 효소활성
β-글루코시다제(β-glucosidase)는 비배당체의 형성과 이소플라본배당체의 가수분해에 필요한 촉매작용을 한다고 알려져 있다. β-글루코시다제를 생산하는 미생물로는 아스퍼질러스 나이거(Aspergillus niger), 트라이코더마 레제이(Trichoderma reesei), 푸사리움 옥시스포룸(Fusarium oxysporum) 등의 곰팡이와 사카로마이세스 세레비지애(Saccharomyces cerevisiae), 스키조사카로마이세스 폼베(Schizosaccharoyces pombe) 등의 효모 그리고 토양세균으로 스트렙토마이세스 레티쿨리(Streptomyces reticuli), 바실러스 서틸리스(Bacillus subtilis), 바실러스 폴리믹사(Bacillus polymyxa), 바실러스 서큘란스(Bacillu circulans), 아그로박테리움 튜머파시엔스(Agrobacterium tumefaciens), 셀룰로모나스 종(Cellulomonas sp.) CS1-1 및 초고온성 고세균인 피로코커스 푸리오수스(Pyrococcus furiosus) 등 다양한 계통군에 속하는 균주들이 알려져 있고, 이들이 생산하는 β-글루코시다제에 대한 효소학적 특성과 유전자도 다수 밝혀졌다. 전통발효식품에 주로 존재하는 대두 이소플로본 배당체인 제니스틴과 다이드진의 체내흡수를 위해 비배당체 형태로 전환하는 생물전환 공정에 활용가능성을 갖는 β-글루코시다제 활성을 확인한 결과, FT4-2 균주가 β-글루코시다제의 활성이 가장 우수한 것으로 확인되었다(표 2)

[실시예 3]β-글루칸 함량 분석
피부 생리활성 물질 중 하나인 β-글루칸(β-glucan)은 글루코오스가 β-(1-3)-D-글리코사이드 결합으로 연결되어 있는 균일 다당류이며, 효모 세포벽의 가장 많은 양의 구성성분으로 효모 세포벽에서 추출된다. β-글루칸은 항암, 항콜레스테롤, 면역증강 및 피부 재생 등과 같은 여러 가지 생리활성 촉진효과가 있다고 보고되어 건강식품 첨가물로서 이용이 증가되고 있다. 특히, 효모로부터 얻어지는 β-글루칸은 피부를 보호하고 손상 시 피부를 재생시키는 역할을 하며, 인체 내에서 면역시스템을 강화시키는데 중요한 역할을 하는 것으로 알려져 있다. 또한, β-(1,3)- 또는 β-(1,6)- 결합의 분자구조 보다 β-(1,3)-/β(1-6)-결합의 분자구조를 갖는 β-글루칸이 생리활성 촉진이 우수하다고 알려져 있다. 사카로마이세스 세레비지애 FT4-2 균주에서 생산된 효모 균체의 β-글루칸 함량을 측정한 결과, 세포건조무게(g) 당 32.56 mg으로 확인되었다(표 3)

[실시예 4]관능평가
전술한 우수한 인체 유익성능을 가지는 FT4-2 효모 균주를 접종하여 효과적인 균배양을 달성하며, 추후 발효체(200)와 혼합되어 우수한 관능평가를 가지는 채소 또는 과일 착즙의 다양한 실시예를 통해 100인 이상의 관능평가를 실시하였다. 딸기, 블루베리, 토마토, 사과 및 무의 착즙을 배양체로 활용한 결과, 추후 된장 또는 고추장 등의 발효체와 혼합 후 발색 및 관능평가 시 잘 어울리며 발효스프레드의 맛을 향상시키는 착즙 원액으로 토마토 착즙이 최적의 결과를 나타내었다.

위의 표의 내용과 같이 토마토의 경우, 관능평가에서 월등히 우수한 효과를 발휘함을 볼 수 있다.
또한, 본 발명에서의 대체당으로는 알룰로오스, 에리스리톨, 자일리톨, 말티톨 등이 사용되는 것이 가능하다.
구체적인 배합비로서는, 아래와 같은 배합실시예 표를 통해 최적의 관능평가를 가지는 배합비를 가지는 본 발명의 실시예가 적합하다 할 것이다. 따라서, 효모액:발효체: 대체당의 비율이 6~7 : 1 : 2~3을 이루는 것이 바람직하다 할 것이다. 즉, 60~70%의 중량비를 효모액이 차지하며, 대체당이 20~30%의 중량비, 발효체가 10%의 중량비를 가짐이 발효스프레드의 당산비 및 관능 밸런스가 가장우수한 결과를 가져온다고 할 것이다.

이상으로 본 발명의 특정한 부분을 상세히 기술하였는 바, 당업계의 통상의 지식을 가진 자에게 있어서 이러한 구체적인 기술은 단지 바람직한 구현예일 뿐이며, 이에 본 발명의 범위가 제한되는 것이 아닌 점은 명백하다. 따라서, 본 발명의 실질적인 범위는 첨부된 청구항과 그의 등가물에 의하여 정의된다고 할 것이다.
본 발명은 균주 접종을 통해 유익성분을 함유하는 발효스프레드에 관한 것으로서, 보다 상세하게는 FT4-2 균주를 과일 또는 채소의 원액에 접종하여 배양 후 발효체와 혼합하여 상기 균주의 유익성분을 극대화하여 상기 발효체를 이용한 발효스프레드를 통해 해당 유익성분의 손실 없는 섭취가 가능한 균주 접종을 통해 유익성분을 함유하는 발효스프레드에 관한 것이므로, 산업상 이용가능성이 있다고 할 것이다.

Claims (10)
- FT4-2 효모 균주(10) 접종 및 증균과정을 거쳐 생성되는 효모액(100);상기 효모액(100)에 소정 비율로 혼합되는 발효체(200); 및상기 효모액(100) 및 발효체(200)와 함께 혼합되는 대체당(300);을 포함하는 균주 접종을 통해 유익성분을 함유하는 발효스프레드(400).
- 제1항에 있어서,상기 효모액(100)은,과일 또는 채소를 착즙한 원액에 전배양된 FT4-2 효모 균주(10)를 접종하는 것을 특징으로 하는 균주 접종을 통해 유익성분을 함유하는 발효스프레드.
- 제2항에 있어서,상기 채소는 토마토를 포함하는 것을 특징으로 하는 균주 접종을 통해 유익성분을 함유하는 발효스프레드.
- 제1항에 있어서,상기 발효체(200)는 된장 또는 고추장을 포함하는 것을 특징으로 하는 균주 접종을 통해 유익성분을 함유하는 발효스프레드.
- YPD를 이용하여 FT4-2 효모 균주(10)를 접종 후 섭씨30도에서 24시간 효모균(유익균)을 배양하는 전배양단계(S100);상기 전배양단계(S100)가 종료된 배양체(20)를 멸균수로 3회 이상 세척하는 세척단계(S200);상기 세척완료된 배양체(20)를 과일 또는 채소를 착즙한 원액에 접종하여 효모액(100)을 형성하는 접종단계(S300); 및상기 접종단계(S300)를 통해 형성된 효모액(100)에 발효체(200) 및 대체당(300)을 소정 비율로 혼합하는 혼합단계(S400);를 포함하는 것을 특징으로 하는 균주 접종을 통해 유익성분을 함유하는 발효스프레드 제조방법.
- 제5항에 있어서,상기 접종단계(S300)는,상기 배양체(20)를 과일 또는 채소를 착즙한 원액에 접종 후 섭씨 30도에서 48시간 FT4-2 효모 균주(10)를 배양하여 효모액(100)을 생성하는 후배양단계(S310);를 더 포함하는 것을 특징으로 하는 균주 접종을 통해 유익성분을 함유하는 발효스프레드 제조방법.
- 제6항에 있어서,상기 후배양단계(S310)를 포함하는 접종단계(S300)가 종료 후, 섭씨 영하80도 이하로 상기 효모액(100)을 동결시키는 동결단계(S500);를 더 포함하는 것을 특징으로 하는 균주 접종을 통해 유익성분을 함유하는 발효스프레드 제조방법.
- 제7항에 있어서,상기 동결단계(S500)는,동결 전 섭씨 120도로 15분 이상 상기 효모액(100)을 가열하여 멸균하는 멸균단계(S510);를 더 포함하는 것을 특징으로 하는 균주 접종을 통해 유익성분을 함유하는 발효스프레드 제조방법.
