また、本発明者らは先に、β-グルコシダーゼの存在下でクロストリジウム属微生物を培養して、セルロース系バイオマスをワンステップで糖化するBSES法を提案している(特許文献2参照)。
本発明者らは、上記課題に注目して鋭意研究を行い、麦粕分解に適した微生物の探索を行ってきた結果、パエニバシラス(Paenibacillus)属微生物、又はパエニバシラス(Paenibacillus)属微生物を含んだ微生物叢が麦粕のタンパク質層やセルロース繊維を分解することを見出し、本発明を完成するに至った。
本発明においては、パエニバシラス・マセランス(Paenibacillus macerans)の遺伝子配列を有する遺伝子組換え微生物種を用いてもよい。エニバシラス・マセランス(Paenibacillus macerans)の遺伝子配列を有する遺伝子組換え微生物種として、パエニバシラス・マセランスI-6株(NITE P-03556)又はパエニバシラス・マセランスI-7株(NITE P-03557)の遺伝子配列を有する遺伝子組換え微生物種を用いてもよい。

さらに前記Paenibacillus属微生物、好ましくはI-6株(NITE P-03556)及びI-7株(NITE P-03557)と上記遺伝子組換体微生物種、又は前記DNA配列を用いて形質転換した遺伝子組換体Paenibacillus属微生物と上記遺伝子組換体微生物種を共培養し、微生物叢をタンパク質を含むセルロース系繊維物の分解に用いても構わない。
以下に、本発明を実施例により具体的に説明するが、本発明の方法で用いられる微生物は、上述したマーカー、プロモーター、宿主に限定されないことは明らかである。
(麦粕を分解する微生物株の単離と同定)
発明者らが保存している土壌サンプルを用い、麦粕を分解できる微生物のスクリーニングを以下の方法で行った。
まず、中温帯(37~45℃)と高温度帯(50~60℃)の培養条件で、麦粕と水だけで良好な増殖を示し、培養前の麦粕の乾燥重量と、培養後の麦粕の乾燥重量の差分が大きいものを選抜し、さらに嫌気性条件で集積培養を5回以上繰り返して選抜を行った。
その結果、高温度帯のサンプルからは候補を見つけられなかったが、45℃にて良好な増殖と乾燥重量差の大きい培養物を得た。
微生物単離を行う前に、微生物叢に関して知見を得るために、次世代シーケンスを用いて16SrRNA配列を解析するメタゲノム解析を行ったところ、表1に示すように、培養物中には数種類の菌が存在していることが分かった。
分解の主役となるPaenibacillus maceransを分離するために、嫌気性条件下において麦粕アルカリ処理後の上澄み液を培地として用い、ロールチューブ法によりコロニーの単離を行った。各コロニーを形成した微生物に対して、前述した麦粕と水だけで培養を行ったところ、麦粕を効率的に分解するPaenibacillus macerans I-6株(NITE P-03556)及び Paenibacillus macerans I-7株(NITE P-03557)を単離した。
I-6株(NITE P-03556)及びI-7株(NITE P-03557)の分類学上の性質を明らかにするために、16SrRNA及びrpoB遺伝子の塩基配列解析を行った。rpoB遺伝子(RNAポリメラーゼBサブユニット遺伝子(rpoB))は細菌分類において、16SrRNA遺伝子以外で指標とされる遺伝子配列になるため、より確実に細菌同定が可能である。
まず、前記バイオマス繊維物を炭素源として含む培地を用いて、それぞれI-6株(NITE P-03556)及びI-7株(NITE P-03557)を4日間培養した。得られた培養物を4℃にて10,000回転で5分間、遠心分離して、それぞれのバイオマスで培養した菌体を回収した。次いで、得られた菌体を、最終濃度0.5%となるように10%SDS(ラウリル硫酸ナトリウム)と、最終濃度5μg/mLとになるようにプロテナーゼK(1mg/mL)溶液を加え、37℃で1時間反応させた。次いで、最終濃度1%となるように10%臭化セチルトリメチルアンモニウム-0.7M塩化ナトリウム溶液を加え、65℃で10分間反応させた。
次いで、得られた水層に対し、0.6倍容量のイソプロパノールを加えゲノムDNAを析出させ、遠心分離を行ってゲノムDNAを調製した後、調製したゲノムDNAを70%エタノールで洗浄し、乾燥した。
16SrRNA増幅用PCRプライマーは、27Fオリゴヌクレオチドプライマー(5’-AGAGTTTGATCCTGGCTCAG-3’:配列番号1)、及び1492Rオリゴヌクレオチドプライマー(5’-GGCTACCTTGTTACGACTT-3’:配列番号2)を用いた。
菌種のさらなる確認のため、16SrRNA以外にもrpoB遺伝子のシークエンスを利用した比較を行った。
rpoB遺伝子増幅用PCRプライマーは、rpoB-1698Fオリゴヌクレオチドプライマー(5-AACATCGGTTTGATCAAC-3:配列番号3)、及びrpoB-2041Rオリゴヌクレオチドプライマー(5-CGTTGCATGTTGGTACCCAT-3:配列番号4)を用いた。
PCRは、ExTaqDNAポリメラーゼ(宝酒造社製)により、16SrRNA遺伝子の増幅を行った。PCRの条件は98℃で1分間、55℃で1分間、72℃で2分間を30サイクルの条件において増幅を行った。得られたPCR産物は、0.8%アガロースゲル電気泳動で増幅されたバンドを確認後、QIAGEN PCR精製キット(QIAGEN社製)を用いて、増幅されたPCR産物を精製した。
塩基配列解析及び相同性検索については、非特許文献1及び2に記載された手法に基づき、GenBank/EMBL/DDBJのデータベースを用いて、BLAST(http://www.ncbi.nlm.nih.gov/BLAST/Blast.cgi)により実施した。
得られたP. macerans I-6株(NITE P-03556)及び P. macerans I-7株(NITE P-03557)の16SrRNAシーケンスをそれぞれ配列番号5、配列番号6に示す。またP. macerans I-6株(NITE P-03556)及び P. macerans I-7株(NITE P-03557)のrpoB遺伝子シーケンスをそれぞれ配列番号7と配列番号8に示す。
(Paenibacillus属微生物による麦粕分解能)
実施例1で使用した麦粕の成分を分析したところ、粗タンパク質23.4%、粗脂肪13.3%、ヘミセルロース33.6%、セルロース20%、リグニン5.2%、灰分4.4%と、一般的なセルロースバイオマスとは全く異なる成分、成分比とは異なることが判明した。このことから、Paenibacillus属微生物がタンパク質を含むセルロース系繊維物を分解可能かについて検討を加えた。
NaOHによるアルカリ処理した4%(w/v)麦粕と滅菌蒸留水を用い、P. macerans株(DSM24)及び、Clostridium thermocellum(DSM1313), P. curdlanolyticus, P. cisolokensis(DSM 101873)とともに麦粕分解能を比較した。嫌気培養にするために上層を窒素ガスで置換し、ゴム栓とアルミキャップにて栓をした。
麦粕の分解測定は、前記微生物を1%(v/v)シリンジにて接種し、P. macerans株(DSM 24)及びP. curdlanolyticusは37℃で培養し、P. macerans I-6株及びI-7株は45℃で培養、またP. cisolokensis (DSM101873)は50℃にて培養、Clostridium thermocellumは60℃にて培養を行った。5日間後、残った麦粕を乾燥させ、初発の麦粕の重量の差し引きにより割合を求めた。

結果を図1に示す。なお、この試験は5回の反復試験から分解率の分布を示したものである。図1中のRI-7は、後述する実施例4及び5の遺伝子組換えI-7が示す麦粕分解能である(実施例5を参照)。
(Paenibacillus maceransのプロテアーゼ活性とキシラナーゼ活性)
実施例2の結果より、P. maceransI-6とP.maceransI-7は、セルロースやヘミセルロース分解活性のみならず、恐らく麦粕表面をコーティングしているタンパク質を効果的に分解し繊維質を露出させることで分解を促進している可能性が示唆されている。
そこで、プロテアーゼ活性があるかどうかを確認した。
実施例2と同様に、アルカリ処理した麦粕を用い、プロテアーゼ活性と、セルラーゼ活性、キシラナーゼ活性とについて、P. macerans株タイプストレイン(DSM24)とI-6株、I-7株の菌体外酵素活性を比較した。
微生物の培養後の溶液に対し、1%カゼイン基質溶液を加え、恒温水槽(ヤマト科学社製)にて37℃又は45℃、5分間をプレインキュベーションした。その後、培養液を加え、37℃又は45℃、30分間インキュベーションした。30分後、トリクロロ酢酸(TCA)試薬を加え酵素反応を停止させた。さらに37℃で30分間インキュベーションした後、遠心分離して上清を得た。
この上清に炭酸ナトリウム試液を加えて攪拌し、さらにフォーリン試液を加えて攪拌し、恒温水槽にて37℃、30分間インキュベートした。発色した溶液は分光光度計(島津製作所社製)にて吸光度660nmで測定した。酵素活性1Uは、1分間に1μmolのアミノ酸(チロシン)を遊離する酵素量と定義し、酵素1mgあたりの酵素活性(U/mg)を算出した(表2)。
なお、酵素のタンパク質濃度の測定は、Pierce BCA Protein Assay Kit (Thermo Fisher製)を用いて行った。タンパク質濃度定量用の標準物質としては、濃度既知のウシ血清アルブミン(Thermo Fisher製)を使用した。
セルロースやキシラン基質に対するDSM 24とI-6株とI-7株の酵素活性を調べた。50mMの酢酸ナトリウム緩衝液(pH7.0)に上記で得た培養液及び1%カルボキシメチルセルロース及びキシラン基質を加え、45℃で30分間保温した後、遊離した還元糖濃度をソモギネルソン法によって定量した。酵素活性1Uは、1分間に1μmolの還元糖を遊離する酵素量と定義し、酵素1mgあたりの酵素活性(U/mg)を算出した(表2)。
なお、酵素のタンパク質濃度の測定は、Pierce BCA Protein Assay Kit (Thermo Fisher製)を用いて行った。タンパク質濃度定量用の標準物質としては、濃度既知のウシ血清アルブミン(Thermo Fisher製)を使用した。
比較結果を表2に示す。
(クロラムフェニコール抗生物質耐性P. macerans I-6株とI-7株の作出)
P. macerans I-6株とP. macerans I-7株の遺伝子組換体作出の可能性を確認するために、市販のプラスミドベクターを用いて、P. macerans I-6株とI-7株のクロラムフェニコール抗生物質耐性株を作出した。
エレクトロポレーション後、P. maceransI-6株とI-7株はDSM 220液体培地中にて3時間ほど45℃にて後培養を行い、クロラムフェニコール40μg/mL濃度を含むDSM 220プレート培地に塗抹して形質転換体の出現を観察した。
結果を図2に示す。図2(B)に示すpNW33Nが組み込まれたP. macerans I-7株のプレートでは、コロニーが現れているが、図2(A)に示すpNW33Nを含まないI-7株のプレートにはコロニーが形成されていないことが分かる。このことから、P. macerans I-6株とI-7株に関し、プラスミドを用いた遺伝子組換体を容易に取得できることが理解できる。
クロラムフェニコール耐性となった遺伝子組換体P. macerans(RI-7株)に対して麦粕糖化実験を行った。実施例2と同様に、前処理をした麦粕と、クロラムフェニコール40μg/mL濃度とを含む培地にRI-7株を同様に接種した。
クロラムフェニコールが含まれる麦粕培養液でも、遺伝子組換体でないI-7株と同等の糖化率を示した(図1を参照)。この結果から遺伝子組換体においても糖化効率を維持し麦粕を糖化出来ることが明らかとなった。
なお、前記した各実施例においては、pNW33Nプラスミドベクターを用いたが、pI6ORI(NITE-P03555)として寄託されている配列を用いて形質転換することもできる。

- パエニバシラス(Paenibacillus)属微生物を用いて、タンパク質を含むセルロース系繊維物を分解する方法。
- 前記タンパク質を含むセルロース系繊維物が麦粕である、請求項1記載のタンパク質を含むセルロース系繊維物を分解する方法。
- 前記パエニバシラス(Paenibacillus)属微生物が、パエニバシラス・マセランス(Paenibacillus macerans)である、請求項1記載のタンパク質を含むセルロース系繊維物を分解する方法。
- 前記パエニバシラス(Paenibacillus)属微生物が、パエニバシラス・マセランス(Paenibacillus macerans)I-6株(NITE P-03556)又はパエニバシラス・マセランス(Paenibacillus macerans)I-7株(NITE P-03557)である、請求項1記載のタンパク質を含むセルロース系繊維物を分解する方法。
- パエニバシラス・マセランス(Paenibacillus macerans)の遺伝子配列を有する遺伝子組換え微生物種を用いて、タンパク質を含むセルロース系繊維物を分解する方法。
- 前記エニバシラス・マセランス(Paenibacillus macerans)の遺伝子配列を有する遺伝子組換え微生物種として、パエニバシラス・マセランスI-6株(NITE P-03556)又はパエニバシラス・マセランスI-7株(NITE P-03557)の遺伝子配列を有する遺伝子組換え微生物種を用いる、請求項5記載のタンパク質を含むセルロース系繊維物を分解する方法。
