Abstract
본 발명은 TCTP 에피토프 및 이에 특이적으로 결합하는, 항체 또는 항원 결합 단편에 관한 것이다. 또한, 본 발명은 상기 항체 또는 항원 결합 단편을 포함하는 암 예방 또는 치료용 약학 조성물, 상기 항체 또는 항원 결합 단편을 이용한 암 진단을 위한 정보의 제공 방법, 및 상기 TCTP 에피토프에 특이적으로 결합하는 물질의 스크리닝 방법에 관한 것이다. 본 발명에 따른 TCTP 에피토프는 TCTP에 존재하는 다른 영역 또는 에피토프에 비하여 항체 또는 항원 결합 단편과의 결합능 및 친화도가 매우 우수하다. 또한, 상기 항체 또는 항원 결합 단편은 암세포를 사멸시키는 효과 또는 종양 성장 및 증식 억제 효과가 매우 우수하며, 특히 기존의 항암제에 대하여 내성 또는 불응성을 나타내는 암에 대한 항암 효과가 현저하게 우수할 뿐만 아니라 기존 면역항암제로 사용되고 있던 약물과도 우수한 시너지 효과를 나타내므로, 암의 예방, 치료 또는 진단 등에 유용하게 활용될 수 있다.
Description
본 발명은 TCTP 에피토프 및 이에 특이적으로 결합하는, 항체 또는 항원 결합 단편에 관한 것이다. 또한, 본 발명은 상기 항체 또는 항원 결합 단편을 포함하는 암 예방 또는 치료용 약학 조성물, 상기 항체 또는 항원 결합 단편을 이용한 암 진단을 위한 정보의 제공 방법, 및 상기 TCTP 에피토프에 특이적으로 결합하는 물질의 스크리닝 방법에 관한 것이다.
지난 수년간 암에 대한 집중적인 연구가 이루어졌음에도 불구하고 여전히 암은 전세계적으로 주요한 사망 원인이다. 다수의 암 치료법이 개발되었으나 모든 암종에 대해 그리고 모든 환자에 대해 효과적이지 않다. 암을 치료하기 위해 현재 사용되고 있는 방법은 상대적으로 비선택적이다. 수술에 의해 질병을 갖는 조직을 제거하거나, 방사선 치료에 의해 고형 종양의 크기를 감소시키거나, 화학치료에 의해 암세포를 빠르게 사멸시킨다. 특히, 화학치료는 약물에 대한 내성을 발달시킬 수 있으며, 일부 경우에는 투여가능한 용량을 제한하여 결국 잠재적으로 유효한 약물의 사용을 배제시킬 정도로 심각한 부작용을 야기시킨다. 따라서, 타겟 특이적이고 보다 효과적인 암 치료 방법의 개발이 시급하다.
이와 관련하여, 암세포만을 특이적으로 타겟하는 표적항암제, 또는 환자의 면역체계를 활용하는 면역항암제 등이 개발되고 있다. 예를 들어, 국내에서 허가받은 표적항암제의 종류는 약 30여 종에 달하며, 또한 국내에서 허가받은 면역항암제는 옵디보(성분명: 니볼루맙), 키트루다(성분명: 펨브롤리주맙), 티쎈트릭(성분명: 아테졸리주맙), 임핀지(성분명: 더발루맙), 여보이(성분명: 이필리무맙) 등으로서, 이들은 비소세포폐암, 흑색종, 신세포암, 호지킨 림프종, 두경부암, 방광암 등에 치료 허가를 받는 등 다양한 암종에 대하여 사용되고 있다.
그러나, 이러한 표적항암제와 면역항암제의 사용 비율이 증가함에 따라 이에 대하여 이미 내성 또는 불응성을 나타내는 환자의 비율 또한 증가하고 있다. 따라서, 새로운 타겟의 개발을 발굴하고, 이를 활용하여 기존 항암제에 내성 또는 불응성을 나타내는 암 치료제의 개발이 당업계에 요구되고 있다.
본 발명자들은 항암제의 새로운 타겟을 규명하고, 이를 활용함으로써 기존의 항암제에 내성 또는 불응성을 나타내는 암에 대한 치료제를 개발하는 연구를 진행하였다. 그 결과, 본 발명에 따른 서열번호 1, 서열번호 2, 서열번호 3, 서열번호 4 및/또는 서열번호 5의 아미노산 서열로 표시되는 TCTP 에피토프를 규명하였고, 상기 에피토프를 타겟하는 항체 또는 항원 결합 단편은 기존 항암제에 내성 또는 불응성을 나타내던 암종에 대해서도 우수한 항암 효과를 나타낼뿐만 아니라 기존 면역항암제로 사용되고 있던 약물과도 우수한 시너지 효과를 나타낼 수 있음을 확인하였다.
본 발명의 하나의 양태는 서열번호 1 내지 5 중에서 선택되는 아미노산 서열로 표시되는 TCTP의 에피토프에 특이적으로 결합하는, 항체 또는 항원 결합 단편을 제공한다.
본 명세서에서 사용되는 용어, "항체"는 면역학적으로 특정 항원과 반응성을 갖는 면역글로불린 분자 또는 항원을 특이적으로 인식하는 수용체 역할을 하는 단백질 분자를 의미한다. 따라서, 본 발명에서 "항체"는 모노클로날 항체, 폴리클로날 항체, 전장(whole) 항체(디설파이드 결합에 의해 상호 연결된 적어도 2개의 중쇄 및 2개의 경쇄로 이루어진 항체), 항체 단편, Fab, Fab', F(ab')2, Fv, 단쇄 Fv(scFv), 디아바디, 선형 항체, 이중특이성(bispecific) 항체, 다중특이성 항체, 키메라 항체, 인간화 항체, 인간 항체 또는 항체의 항원-결합 부분을 모두 포함하는 것으로 해석된다. 상기 전장 항체는 IgA, IgD, IgE, IgM 및 IgG를 포함하며, 상기 IgG는 아형(subtype)으로서 IgG1, IgG2, IgG3 및 IgG4를 포함할 수 있다.
본 명세서에서 사용되는 용어, "항원 결합 단편"은 모 항체의 결합 특이성을 적어도 일부 보유하고, 모 항체의 항원 결합 영역의 일부(예를 들면, 하나 이상의 CDR) 또는 가변영역을 포함하는 항체 단편 또는 항체의 유사체를 의미한다. 상기 항원 결합 단편은 예를 들면 Fab, Fab', F(ab')2, Fv 단편, sc-Fv, 유니바디 (unibody), 디아바디, 선형 항체, 나노바디, 도메인 항체 또는 항체 단편으로부터 형성된 다특이적 항체일 수 있다.

본 명세서에서 사용되는 용어, "중쇄"는 중쇄 가변영역(Variable Region)과 중쇄 불변영역(Constant Region)을 포함하는 전장 중쇄 및 이의 단편을 의미한다. 중쇄에는 감마(γ), 뮤(μ), 알파(α), 델타(δ) 및 엡실론(ε) 형이 있다.
본 명세서에서 사용되는 용어, "경쇄"는 경쇄 가변영역과 경쇄 불변영역을 포함하는 전장 경쇄 및 이의 단편을 의미한다. 경쇄에는 카파(κ) 및 람다(λ) 형이 있다.
본 명세서에서 사용되는 용어, "CDR(Complementarity determining region)"은 B 세포 및 T 세포에 의해 생성된 항체의 가변영역의 일부로서, 항원결합부위를 형성하고 있는 초가변영역(Hypervariable region)의 아미노산 서열을 의미한다. 중쇄 및 경쇄의 아미노산 서열에는 각각 비연속적으로 배열된 3개의 CDR, 즉 중쇄 CDRH1, CDRH2, CDRH3 및 경쇄 CDRL1, CDRL2, CDRL3이 포함되어 있다. CDR은 항원의 인식에 관여하는 부위로 항체가 항원 또는 에피토프와 결합함에 있어 주요한 접촉 잔기를 제공하여 항원 특이성의 다양성에 결정적인 역할을 한다.
본 발명에서 항체는 전장 항체이거나 또는 항원 결합능을 갖는 항체 단편이며, 중쇄는 감마(γ), 뮤(μ), 알파(α), 델타(δ) 또는 엡실론(ε)형 중 어느 하나일 수 있고, 경쇄는 카파(κ) 또는 람다(λ) 형일 수 있다.
본 명세서에서 사용되는 용어, "TCTP(Translationally-controlled tumor protein)"는 모든 진핵 생물에서 널리 발현되는 고도로 보존된 단백질로서, 이를 구성하는 아미노산 서열 및 이를 인코딩하는 염기 서열은 기존 데이터베이스에 공지되어 있다(NCBI Reference Sequence: NP_003286.1, NM_003295.4 등). TCTP의 발현은 전사 및 번역 수준, 또는 광범위한 세포외 신호에 의해 고도로 조절되며, 세포 성장, 세포 주기 진행, 다양한 스트레스 조건이나 세포자멸사에 대한 세포 보호 등에 관여한다.
본 명세서에서 사용되는 용어, "에피토프(epitope)"는 항원결정기(antigenic determinant)로서, 항체 또는 B 세포, T 세포 등의 면역 세포가 항원을 식별하게 하는 항원의 특정한 부분을 의미한다. 본 발명에 따른 TCTP의 에피토프는 서열번호 1, 서열번호 2, 서열번호 3, 서열번호 4 및 서열번호 5로 이루어진 그룹에서 선택되는 하나 이상의 아미노산 서열로 표시되는 것일 수 있다. 또한, 상기 TCTP 에피토프는 상기 서열번호 1, 서열번호 2, 서열번호 3, 서열번호 4 및/또는 서열번호 5의 아미노산 서열을 코딩하는 염기 서열로 표시되는 것일 수 있다
구체적으로, 상기 서열번호 1 내지 5의 아미노산 서열은 TCTP 단백질(NCBI Reference Sequence: NP_003286.1)을 구성하는 172개 아미노산 서열의 일부일 수 있다. 더욱 구체적으로, 상기 서열번호 1의 아미노산 서열로 표시되는 TCTP 에피토프는 TCTP 단백질의 71 내지 80번째 아미노산일 수 있고, 상기 서열번호 2의 아미노산 서열로 표시되는 TCTP 에피토프는 TCTP 단백질의 61 내지 80번째 아미노산일 수 있고, 상기 서열번호 3의 아미노산 서열로 표시되는 TCTP 에피토프는 TCTP 단백질의 56 내지 80번째 아미노산일 수 있고, 상기 서열번호 4의 아미노산 서열로 표시되는 TCTP 에피토프는 TCTP 단백질의 51 내지 80번째 아미노산일 수 있고, 상기 서열번호 5의 아미노산 서열로 표시되는 TCTP 에피토프는 TCTP 단백질의 51 내지 100번째 아미노산일 수 있다.
본 발명에서 개시되는 서열은 서열목록에 기재된 서열과 실질적인 동일성을 나타내는 서열을 포함한다. 상기 실질적인 동일성이란 두 개의 서열을 최대한 대응되도록 정렬한 후 당업계에서 통상적으로 이용되는 알고리즘을 사용하여 분석한 결과, 80%, 90% 또는 95% 이상의 서열간 상동성을 나타내는 것을 의미한다.
또한, 본 발명의 항체 또는 항원 결합 단편은 TCTP 에피토프를 특이적으로 인식하고 결합할 수 있는 범위 내에서 본 명세서에 기재된 항체 또는 항원 결합 단편의 서열, 상기 서열과 실질적인 동일성을 나타내는 서열 뿐만 아니라, 이의 생물학적 균등물을 포함할 수 있다. 예를 들어, 항체 결합 친화도 및/또는 생물학적 특성 등을 개선시키기 위한 서열의 추가적인 변이를 포함할 수 있으며, 분자의 전체적 활성을 변경시키지 않는 범위 내에서 추가적인 변이를 포함할 수 있다.
본 발명의 다른 하나의 양태는 상기 항체 또는 항원 결합 단편을 코딩하는 핵산을 제공한다.

본 발명의 항체 또는 항원 결합 단편을 코딩하는 핵산 분자는 상기한 뉴클레오타이드 서열에 대하여 실질적인 동일성을 나타내는 뉴클레오타이드 서열을 포함한다. 상기 실질적인 동일성이란 두 개의 서열을 최대한 대응되도록 정렬한 후 당업계에서 통상적으로 이용되는 알고리즘을 사용하여 분석한 결과, 80%, 90% 또는 95% 이상의 서열간 상동성을 나타내는 것을 의미한다.

본 발명에서 사용되는 용어, "항암제 내성"은 항암제를 이용하여 암 환자를 치료할 때 치료 초기부터 효과가 없거나, 또는 초기에는 치료 효과가 있으나 계속적인 치료 과정에서 암 치료 효과가 감소 또는 상실되는 것을 의미한다.
본 발명에서 사용되는 용어, "항암제 불응성"은 항암제를 이용하여 암 환자를 치료할 때 치료 초기부터 효과가 없거나, 또는 치료에 대한 반응이 장기간 동안 지속되지 않는 것을 의미한다.

본 발명의 약학 조성물은 상기 항체 또는 항원 결합 단편을 유효량으로 포함하여, 암 예방 또는 치료를 필요로 하는 대상체에게 투여될 수 있다.

이하 본 발명을 실시예에 의해 보다 상세하게 설명한다. 그러나 이들 실시예는 본 발명을 예시적으로 설명하기 위한 것으로, 본 발명의 범위가 이들 실시예에 의해 제한되는 것은 아니다.
실시예 1. 항암 활성을 나타내는 TCTP region 규명
실시예 1-1. TCTP region의 암세포 생존능 확인
우수한 항암 활성을 나타내는 TCTP 에피토프를 규명하기 위하여, 먼저 상기 에피토프를 포함하는 region을 규명하였다.
구체적으로, 172개의 아미노산으로 구성된 TCTP 단백질(full length)에 대하여 약 50개의 아미노산으로 구성되도록 region을 나누었고, 해당 region을 포함하는 펩타이드 단편을 제조하였다. 즉, TCTP 단백질의 1 내지 49번째 아미노산으로 구성된 region 1 (TCTP 1-49), 50 내지 100번째 아미노산으로 구성된 region 2 (TCTP 50-100), 101 내지 150번째 아미노산으로 구성된 region 3 (TCTP 101-150), 및 151 내지 172번째 아미노산으로 구성된 region 4 (TCTP 151-172)를 포함하는 펩타이드를 각각 제조하였다.
이후, 상기 펩타이드 처리에 따른 암세포의 생존능 및 사멸 정도를 확인하였다. 암세포는 기존 항암제 중에서 면역항암제에 불응성을 나타내는 흑색종 B16 P3 세포주를 사용하였다. 암세포 생존능은 ERK 인산화를 통해 확인하였는데, ERK는 암세포 생존과 관련한 신호전달 단백질로서, ERK가 인산화되어 활성화되면 암세포의 생존능이 증가한다. 또한, T 면역세포에 의해 매개되는 암세포의 사멸을 확인하였는데, 이는 T 세포를 첨가하여 추가 배양을 한 뒤 active-caspase-3를 측정함으로써 분석하였다.
그 결과, 도 1에서 볼 수 있듯이, 각 region을 포함하는 TCTP 펩타이드 중에서, 50 내지 100번째 아미노산으로 구성된 TCTP (50-100)를 포함하는 펩타이드를 처리하는 경우, 인산화된 ERK의 양이 현저히 증가하였다. 즉, TCTP (50-100)는 암세포의 생존능을 현저히 증가시킬 수 있음을 확인하였다.
또한, 도 2에서 볼 수 있듯이, 각 region을 포함하는 TCTP 펩타이드 중에서, 50 내지 100번째 아미노산으로 구성된 TCTP (50-100)를 포함하는 펩타이드를 처리하는 경우, T 세포에 의해 사멸된 암세포의 수가 현저히 감소하였다.
상기 결과를 통해, TCTP 단백질의 50 내지 100번째 아미노산 서열이 암과 관련된 질환과의 연관성이 높고, 항암 효능을 위한 필수 region임을 확인하였다.
실시예 1-2. TCTP region과 항체의 결합능 확인
TCTP 단백질의 각 region과 TCTP를 타겟하는 항체의 결합능을 확인하였다.
TCTP를 타겟하는 항체로는 상업적으로 이용가능한 항체(abcam, cat #: ab133568)와 TCTP (50-100) 단백질을 마우스에 투여하여 면역반응을 통해 형성된 항-TCTP 항체 2종(#1 및 #2)을 사용하였다. 구체적으로, TCTP 단백질의 50-100 서열에 대한 펩타이드를 마우스에 투여하여 항-TCTP 항체를 분비하는 하이브리도마 클론 2종을 제조하였고, 각각의 클론으로부터 항-TCTP 항체 #1 및 항-TCTP 항체 #2를 제조하여 사용하였다.
이후, 각 region과 항체의 결합능은 흡광도를 측정함으로써 확인하였다.
그 결과, 도 3에서 볼 수 있듯이, 각 region을 포함하는 TCTP 펩타이드 중에서, 50 내지 100번째 아미노산으로 구성된 TCTP (50-100)를 포함하는 펩타이드가 항-TCTP 항체와의 결합능이 가장 우수하였다.
상기 결과를 통해, TCTP 단백질의 50 내지 100번째 아미노산 서열이 항-TCTP 항체와의 결합능이 가장 우수하므로, TCTP 에피토프를 포함하고 있다는 점이 확인되었다. 나아가, 암과 관련된 질환의 예방, 치료 또는 진단에 가장 유용하게 활용될 수 있다는 결과를 얻었다.
실시예 1-3. 항-TCTP 항체의 암세포 사멸능 확인
상기 실시예 1-2를 통해, 항-TCTP 항체는 TCTP region 중에서도 50 내지 100번째 아미노산 서열로 구성된 TCTP (50-100)에 결합능이 가장 우수함을 확인하였다. 본 실시예에서는, TCTP (50-100)에 결합하는 항체가 항암 활성을 나타낼 수 있는지를 확인하고자 하였다.
구체적으로, TCTP를 타겟하는 항체로는, 상업적으로 이용가능한 항체(abcam, cat #: ab133568)와 TCTP (50-100) 단백질을 마우스에 투여하여 면역반응을 통해 형성된 항-TCTP 항체 2종(#1 및 #2)을 사용하였다.
암세포는 흑색종 B16 P3 세포주를 사용하였으며, 이 세포주는 본 출원인에 의해 면역항암제에 불응성을 나타내도록 준비된 세포주이다. 상기 세포주는 모세포인 B16 암세포를 쥐에 이식하고 PD-1 항체 치료를 한 후에 살아 남는 세포를 다시 배양하는 방법으로 제조되는데, 여기서 명명되는 P3는 상기의 치료와 세포 배양을 3번 반복하여 얻었다는 것을 의미한다. 암세포 생존능은 ERK 인산화를 통해 확인하였는데, ERK는 암세포 생존과 관련되는 신호전달 단백질로서, ERK가 인산화되어 활성화되면 암세포의 생존능이 증가한다. 또한, T 면역세포에 의해 매개되는 암세포의 사멸을 확인하였는데, 이는 T 세포를 첨가하여 추가 배양을 한 뒤 active-caspase-3를 측정함으로써 분석하였다.
그 결과, 도 4에서 볼 수 있듯이, 각 항-TCTP 항체를 처리하는 경우, 인산화된 ERK의 양이 현저히 감소하였다. 즉, TCTP (50-100)에 우수한 정도로 결합하는 항-TCTP 항체는 암세포의 생존능을 현저하게 감소시킬 수 있음을 확인하였다.
또한, 도 5에서 볼 수 있듯이, 각 항-TCTP 항체를 처리하는 경우, 사멸된 암세포의 수가 현저히 증가하였다.
상기 결과를 통해, TCTP 단백질의 50 내지 100번째 아미노산 서열이 암과 관련된 질환과의 연관성이 높고, 항암 효능을 위한 필수 region임을 확인하였다.
실시예 1-4. 항-TCTP 항체의 암세포 성장 억제 효과
상기 실시예 1-2를 통해, 항-TCTP 항체는 TCTP region 중에서도 50 내지 100번째 아미노산 서열로 구성된 TCTP (50-100)에 결합능이 가장 우수함을 확인하였다. 본 실시예에서는, TCTP (50-100)에 결합하는 항체가 in vivo에서 항암 활성을 나타낼 수 있는지를 확인하고자 하였다.
구체적으로, TCTP를 타겟하는 항체로는, TCTP (50-100) 단백질을 마우스에 투여하여 면역반응을 통해 형성된 항-TCTP 항체 2종(#1 및 #2)을 사용하였다. 대조항체로는 IgG 항체 또는 항-PD-1 항체(Bio X Cell사에서 구입한 항-마우스 PD-1 항체)를 사용하였다.
이후, 각 항체 처리에 따른 암세포의 성장 억제 정도를 확인하였다. 암세포는 기존 항암제 중에서 면역항암제에 불응성을 나타내는 흑색종 B16 P3 세포주를 사용하였다. 상기 세포주를 마우스에 이식하고, 상기 항체를 투여한 후, 성장한 암세포의 크기를 확인하였다.
그 결과, 도 6에서 볼 수 있듯이, 항-TCTP 항체를 투여하는 경우, 대조항체인 IgG 항체 또는 항-PD-1 항체를 투여하는 경우에 비하여, 암세포의 성장이 현저히 감소하였다.
상기 결과를 통해, TCTP 단백질의 50 내지 100번째 아미노산 서열이 암과 관련된 질환과의 연관성이 높고, 항암 효능을 위한 필수 region임을 확인하였다.
실시예 2. 항암 활성을 나타내는 TCTP 에피토프 규명
실시예 2-1. TCTP 에피토프와 항체의 결합 확인
상기 실시예 1의 내용을 종합한 결과, TCTP region 중에서도 항체와의 결합능이 우수하고, 암의 생존 및 사멸에 관련이 높은 region은 TCTP 단백질의 50 내지 100번째 아미노산 서열로 구성된 TCTP 50-100임을 확인한바, 상기 region에 포함된 에피토프 서열 중에서 가장 우수한 항암 활성을 나타낼 수 있는 서열을 더욱 구체적으로 규명하고자 하였다.
TCTP 50-100 region에 대하여 랜덤하게 region을 나누었고, 해당 region을 포함하는 펩타이드 단편을 제조하였다. 즉, TCTP 단백질의 51 내지 100번째 아미노산으로 구성된 TCTP 51-100, TCTP 단백질의 71 내지 100번째 아미노산으로 구성된 TCTP 71-100, TCTP 단백질의 51 내지 60번째 아미노산으로 구성된 TCTP 51-60, TCTP 단백질의 51 내지 70번째 아미노산으로 구성된 TCTP 51-70, TCTP 단백질의 51 내지 80번째 아미노산으로 구성된 TCTP 51-80, TCTP 단백질의 56 내지 80번째 아미노산으로 구성된 TCTP 56-80, 및 TCTP 단백질의 61 내지 80번째 아미노산으로 구성된 TCTP 61-80을 포함하는 펩타이드를 각각 제조하였다. 이들의 구성은 도 7에 나타내었다.
TCTP를 타겟하는 항체로는, TCTP (50-100) 단백질을 마우스에 투여하여 면역반응을 통해 형성된 항-TCTP 항체 2종(#1 및 #2)을 사용하였다.
이후, 상기 펩타이드와 항-TCTP 항체간의 결합을 검출하기 위해 ELISA를 수행하였다. 96-웰 ELISA microtiter 플레이트를 PBS에 용해된 5㎍/ml 농도의 스트렙타비딘으로 4℃에서 밤새 코팅하였다. 플레이트를 2회 세척하고 3% MPBS로 37℃에서 1시간 동안 블로킹하였다. 이후 PBS에 용해된 10 μg/ml 농도의 각 펩타이드 100 ㎕를 플레이트에 넣고 37℃에서 1시간 동안 인큐베이션하였다. 플레이트를 3회 워싱한 후, 항-TCTP 항체 100 ㎕를 넣고 실온에서 2시간 동안 인큐베이션하였다. 워싱 후, HRP-conjugated anti-mouse IgG(H&L)를 플레이트에 넣고 37℃에서 30분 동안 인큐베이션하였다. 플레이트를 추가로 6회 워싱하고 기질 용액(substrate solution)을 웰에 첨가하여 1분 동안 반응을 진행하였다. 흡광도는 분광계를 사용하여 450nm에서 측정하였다.
그 결과, 도 8의 A에서 볼 수 있듯이, 항-TCTP 항체 #1은 각 region을 포함하는 TCTP 펩타이드 중에서, TCTP 51-100, TCTP 51-80, TCTP 56-80 및 TCTP 61-80를 포함하는 펩타이드와의 결합 정도가 우수하였다. 또한, 도 8의 B에서 볼 수 있듯이, 항-TCTP 항체 #2도 각 region을 포함하는 TCTP 펩타이드 중에서, TCTP 51-80, TCTP 56-80 및 TCTP 61-80를 포함하는 펩타이드와의 결합 정도가 우수하였다. 다만, TCTP 51-70을 포함하는 펩타이드는 항-TCTP 항체 #1 또는 #2와 결합하지 않았다.
상기 결과를 종합하면, TCTP 51-100, TCTP 51-80, TCTP 56-80, TCTP 61-80 및 TCTP 71-80이 항-TCTP 항체가 TCTP 단백질의 결합에 필요로 하는 에피토프임을 알 수 있었다. 이러한 에피토프의 아미노산 서열은 하기에 기재하였다.

실시예 2-2. 항-TCTP 항체의 암세포 생존 억제 효과 확인
서열번호 1, 2, 3, 4 또는 5를 에피토프로 갖는 항-TCTP 항체의 항암 효과를 확인하기 위하여, 상기 실시예 1-1에 따른 방법으로 암세포의 ERK 인산화 정도를 분석하였다. ERK는 암세포 생존과 관련한 신호전달 단백질로서, ERK가 인산화되어 활성화되면 암세포의 생존능이 증가하고, ERK가 인산화되지 못하여 불활성화 상태를 나타내면 암세포의 생존능이 감소한다.
구체적으로, 암세포는 기존 항암제 중에서 면역항암제에 불응성을 나타내는 대장암 CT26 P3 세포주, 및 면역항암제에 불응성을 나타내는 비소세포폐암 또는 자궁암 TC-1 P3 세포주를 사용하였다.
TCTP를 타겟하는 항체로는, TCTP (50-100) 단백질을 마우스에 투여하여 면역반응을 통해 형성된 항-TCTP 항체 2종(#1 및 #2)을 사용하였다. 대조항체로는 TCTP 1-50 region을 타겟으로 하는 시노바이올로지컬(Sinobiological) 사의 항-TCTP 항체(대조항체 1; Cat No. 14662-MM08) 또는 TCTP 101-150 region을 타겟으로 하는 산타크루즈(Santacruz) 사의 항-TCTP 항체(대조항체 2; Cat No. sc-133131)를 사용하였다. 한편, 상기 대조항체가 타겟하는 TCTP의 region은 상기 실시예 2-1에 따른 방법으로 확인하였고, 이의 결과는 도 9의 A 및 B에 나타내었다.
그 결과, 도 10의 A 및 B에서 볼 수 있듯이, 서열번호 1, 2, 3, 4 또는 5에 결합하는 항-TCTP 항체를 처리하는 경우, 인산화된 ERK의 양이 현저히 감소하였다.
반면, 대조항체를 처리하는 경우, 항체 미처리군과 비교 시 인산화된 ERK의 양이 유사하거나 오히려 증가하였다.
상기 결과를 통해, 서열번호 1, 2, 3, 4 또는 5는 TCTP 단백질에 이의 타겟 항체가 결합하는데 필수적인 에피토프 서열이며, 상기 에피토프에 결합하는 항체는 면역항암제 불응성 암에 대해서도 우수한 항암 효과를 나타냄을 확인하였다.
실시예 2-3. 항-TCTP 항체의 화학항암제 민감성 증가 효과 확인
서열번호 1, 2, 3, 4 또는 5를 에피토프로 갖는 항-TCTP 항체의 항암 효과를 확인하기 위하여, 화학항암제에 의한 세포사멸 민감성 증가 효과를 확인하였다.
구체적으로, 암세포는 기존 항암제 중에서 면역항암제에 불응성을 나타내는 대장암 CT26 P3 세포주, 자궁경부암 세포 HeLa, 및 면역항암제에 불응성을 나타내는 비소세포폐암 또는 자궁암 TC-1 P3 세포주를 사용하였다. 상기 세포를 현탁하여 12-웰 세포배양 접시에 1-웰 당 30000개의 세포를 0.1% FBS가 포함된 배지에 6시간 배양하였고, 이후 50 ng/ml 농도로 각 항체를 처리하여 24시간 배양하였다. 화학항암제인 옥살리플라틴(oxaliplatin) 또는 시스플라틴(cisplatin)을 50 μM 농도로 처리하여 24시간 추가 배양한 뒤, 세포사멸 정도를 FACS 분석으로 측정하였다.
TCTP를 타겟하는 항체로는, TCTP (50-100) 단백질을 마우스에 투여하여 면역반응을 통해 형성된 항-TCTP 항체 2종(#1 및 #2)을 사용하였다. 대조항체로는 TCTP 1-50 region을 타겟으로 하는 시노바이올로지컬(Sinobiological) 사의 항-TCTP 항체(대조항체 1; Cat No. 14662-MM08) 또는 TCTP 101-150 region을 타겟으로 하는 산타크루즈(Santacruz) 사의 항-TCTP 항체(대조항체 2; Cat No. sc-133131)를 사용하였다.
그 결과, 도 11의 A 및 B에서 볼 수 있듯이, 서열번호 1, 2, 3, 4 또는 5에 결합하는 항-TCTP 항체를 처리하는 경우, 암세포에서 화학항암제에 대한 민감성이 증가함에 따라 세포 사멸이 증가하였다. 이러한 효과는 대장암, 자궁경부암, 비소세포폐암 또는 자궁암 등 다양한 암종에 대하여 공통적으로 나타나는 효과임을 확인하였다.
반면, 대조항체를 처리하는 경우, 항체 미처리군에 비하여 사멸된 암세포의 비율이 유사하거나 오히려 감소하였다.
상기 결과를 통해, 서열번호 1, 2, 3, 4 또는 5는 TCTP 단백질에 이의 타겟 항체가 결합하는데 필수적인 에피토프 서열이며, 상기 에피토프에 결합하는 항체는 면역항암제 불응성 암에 대해서도 우수한 항암 효과를 나타냄을 확인하였다.
실시예 2-4. 항-TCTP 항체의 in vivo 항암 효과 확인
서열번호 1, 2, 3, 4 또는 5를 에피토프로 갖는 항-TCTP 항체의 in vivo 항암 효과를 확인하였다.
구체적으로, 암세포는 기존 항암제 중에서 면역항암제에 불응성을 나타내는 흑색종 B16 P3 세포주, 및 대장암 MC38 세포주를 사용하였다.
면역항암제에 불응성을 나타내는 흑색종 모델마우스는 일반 생쥐 모델마우스의 피하조직에 B16 P3 흑색종 세포 500000개를 복부 피하이식하여 종양을 형성하여 제조하였다. 종양의 크기가 약 40~50 mm3에 도달하였을 때 각 군(실험군 당, 6마리) 종양의 크기가 최대한 균일하게 분포하도록 분배하고, 항-TCTP 항체를 200 μg을 이틀 간격으로 2주간 총 5회 복강 주사하였다.
대장암 모델마우스는 일반 생쥐 모델마우스의 피하조직에 MC38 대장암 세포 500000개를 복부 피하이식하여 종양을 형성하여 제조하였다. 종양의 크기가 약 25~30 mm3에 도달하였을 때 각 군(실험군 당, 6마리) 종양의 크기가 최대한 균일하게 분포하도록 분배하고, 항-TCTP 항체를 5mg/kg의 농도로 이틀 간격으로 2주간 총 5회 복강 주사하였다.
이후, 종양 이식일(0일)을 기준으로 표시된 날짜에 각각의 종양의 크기를 측정하여 그래프로 나타내었으며, 종양의 크기는 캘리퍼스(calipers)를 이용하여 종양의 장축 및 단축을 측정하고 다음의 계산식을 이용하여 종양 크기를 산출하였다:
종양 크기 = {(장축의 길이)X(단축의 길이)의 제곱}/2
본 실험의 결과에 대해서는 자료의 정규성을 가정하고 모수적인 다중비교 (parametric multiple comparison precedures)를 이용하여 분석하였다. 모수적 이원분산분석(Two-way ANOVA) 결과가 유의하였을 경우, 시닥 다중 비교 테스트(Sidak's multiple comparison test)를 이용하여 사후검정을 실시하였다. 통계학적 분석은 Prism 8.0.1(GraphPad Software Inc., San Diego, CA, USA)를 이용하여 실시하였고, P 값이 0.05 미만일 경우, 통계학적으로 유의한 것으로 판정하였다.
TCTP를 타겟하는 항체로는, TCTP (50-100) 단백질을 마우스에 투여하여 면역반응을 통해 형성된 항-TCTP 항체 2종(#1 및 #2)을 사용하였다. 대조항체로는 IgG 항체 또는 항-PD-1 항체(Bio X Cell사에서 구입한 항-마우스 PD-1 항체)를 사용하였다.
그 결과, 도 12의 A 및 B에서 볼 수 있듯이, 서열번호 1, 2, 3, 4 또는 5에 결합하는 항-TCTP 항체를 처리하는 경우, 대조항체로서 IgG 또는 항-PD-1 항체를 투여한 것에 비하여 암의 성장이 약 2배 이상 현저히 감소하였다. 또한, 상기 항-TCTP 항체를 항-PD-1 항체와 병용하여 처리하는 경우, 항-TCTP 항체를 단독으로 처리하는 것에 비하여 암의 성장이 더 감소하였다. 이러한 효과는 흑색종 또는 대장암 등 다양한 암종에 대하여 공통적으로 나타나는 효과임을 확인하였다.
상기 결과를 통해, 서열번호 1, 2, 3, 4 또는 5는 TCTP 단백질에 이의 타겟 항체가 결합하는데 필수적인 에피토프 서열이며, 상기 에피토프에 결합하는 항체는 면역항암제 불응성 암에 대해서도 우수한 항암 효과를 나타낼뿐만 아니라 기존 면역항암제로 사용되고 있던 약물과도 우수한 시너지 효과를 나타낼 수 있음을 확인하였다.
Claims (17)
- 서열번호 1 내지 5 중에서 선택되는 아미노산 서열로 표시되는 TCTP의 에피토프에 특이적으로 결합하는, 항체 또는 항원 결합 단편.
- 제1항의 항체 또는 항원 결합 단편을 코딩하는 핵산.
- 제2항의 핵산을 포함하는 벡터.
- 제3항의 벡터로 형질전환된 세포.
- 제1항의 항체 또는 항원 결합 단편을 포함하는, 암 예방 또는 치료용 약학 조성물.
- 제5항에 있어서,상기 암은 항암제 내성 또는 불응성인 것인, 약학 조성물.
- 제5항에 있어서,상기 암은 골암, 폐암, 두부암, 경부암, 갑상선암, 부갑상선암, 비소세포성폐암, 위암, 간암, 췌장암, 피부암, 피부 또는 안구내 흑색종, 직장암, 항문부근암, 결장암, 자궁암, 유방암, 난소암, 나팔관암종, 자궁내막암종, 자궁경부암종, 질암종, 음문암종, 호지킨병, 식도암, 소장암, 내분비선암, 부신암, 연조직 육종, 요도암, 음경암, 전립선암, 방광암, 신장암, 수뇨관암, 신장세포 암종, 신장골반 암종, 중추신경계 종양, 중추신경계 림프종, 척수 종양, 교모세포종, 뇌간 신경교종 및 뇌하수체 선종으로 이루어진 군으로부터 선택된 것인, 약학 조성물.
- 제1항의 항체 또는 항원 결합 단편을 포함하는, 암 진단용 조성물.
- 제8항의 조성물을 포함하는, 암 진단용 키트.
- 제1항의 항체 또는 항원 결합 단편을 이용하여, 암이 의심되는 대상체로부터 분리된 생물학적 시료에서 TCTP 단백질을 검출하거나 정량하는 단계를 포함하는, 대상체의 암 진단을 위한 정보의 제공 방법.
- 제1항의 항체 또는 항원 결합 단편을 이용하여, 암을 치료하려는 대상체로부터 분리된 생물학적 시료에서 TCTP 단백질을 검출하거나 정량하는 단계를 포함하는, 암 치료 대상체 선별을 위한 정보의 제공 방법.
- 제1항의 항체 또는 항원 결합 단편을 이용하여, 항암제에 대하여 내성 또는 불응성을 나타낼 것으로 의심되는 대상체로부터 분리된 생물학적 시료에서 TCTP 단백질을 검출하거나 정량하는 단계를 포함하는, 대상체의 항암제 내성 또는 불응성 예측을 위한 정보의 제공 방법.
- 서열번호 1 내지 5 중에서 선택되는 아미노산 서열로 표시되는 TCTP의 에피토프; 및 상기 TCTP의 에피토프와 결합 가능한 후보물질을 혼합하는 단계를 포함하는, 상기 TCTP의 에피토프에 특이적으로 결합하는 물질의 스크리닝 방법.
- 제13항에 있어서,상기 TCTP의 에피토프에 특이적으로 결합하는 물질은 항체 또는 항원 결합 단편인, 방법.
- 서열번호 1 내지 5 중에서 선택되는 아미노산 서열로 표시되는 TCTP의 에피토프를 이용하여, 상기 TCTP의 에피토프에 특이적으로 결합하는 물질을 스크리닝하는 단계; 및 상기 스크리닝된 물질을 항암제로 판단하는 단계;를 포함하는, 항암제의 스크리닝 방법.
- 제15항에 있어서,상기 TCTP의 에피토프에 특이적으로 결합하는 물질은 항체 또는 항원 결합 단편인, 항암제의 스크리닝 방법.
- 제15항에 있어서,상기 스크리닝 단계는 상기 TCTP의 에피토프와 이에 결합 가능한 후보물질을 혼합하는 단계를 포함하는 것인, 항암제의 스크리닝 방법.
