Abstract
인간 HVEM에 높은 친화도로 결합하며, HVEM의 BTLA에 대한 결합을 저해하되 HVEM의 LIGHT에 대한 결합을 촉진함으로써, 단독 또는 다른 치료제와 병용하여 고형암 또는 혈액암을 치료하는데 사용될 수 있는, 항-HVEM 모노클로날 항체, 및 이와 관련된 조성물 및 방법이 제공된다.
Description
본 개시는 항-HVEM 항체에 관한 것으로서, 보다 상세하게는, HVEM의 특정 리간드에 대한 결합을 선택적으로 저해하거나 촉진하는 항-HVEM 항체, 및 이와 관련된 조성물 및 방법에 관한 것이다.
최근 새로운 동시-억제 쌍(co-inhibitory pair)인 HVEM(Herpes Virus Entry Mediator, TNFRSF14)과 BTLA(B and T lymphocyte attenuator)가 항-종양 면역 반응에서 주목을 받고 있다. 이들 두 분자는 T 세포를 포함한 많은 면역 세포에 의해 발현될 수 있으며, BTLA를 통한 신호전달은 이들 면역 세포의 활성화 억제와 연관되어 있다. 또한, HVEM 네트워크는 LIGHT(Tumor necrosis factor superfamily member 14, TNFSF14), 림포톡신 α(Lymphotoxin α, LT α) 또는 CD160과 같은 다른 부가적인 파트너를 포함한다. 국제공개 WO2008/145754호 및 WO2008/146101호는 혈액 악성종양 및 자가면역 질환을 치료하는데 사용할 수 있는 HVEM에 대한 항체로서, HVEM이 BTLA, LIGHT 및 gD에 결합하는 것을 차단하지 않거나, HVEM이 LIGHT에 결합하는 것을 차단하는 모노클로날 항체를 개시하고 있다.
국제공개 WO2014/183885호 및 WO2014/184360호는 BTLA/HVEM 상호작용의 길항제(antagonist)로서, Vγ9Vδ2 T 세포의 증식을 증가시키는 항체 또는 이의 단편을 개시하고 있다. 상기 항체는 HVEM에 대한 모노클로날 항체로 Collection Nationale de Cultures de Microorganismes에 수탁번호 제CNCM 1-4752호로 기탁된 하이브리도마에 의해 생산되는 HVEM 18.10이다. HVEM 18.10은 생쥐 IgG1 항-인간 HVEM mAb(murine 항-human HVEM mAb)로서 복수(ascites)로 생산되며 단백질 A 결합 및 용출에 의해 정제된 것이다. 상기 항체는 HVEM이 BTLA, LIGHT, gD에 결합하는 것을 모두 저해하는 특성을 갖는다. 이 특허는 상기 항체가 혈액 악성종양, 예를 들어 림프종이나 고형암 치료에 사용 가능하다고 개시하고 있다.
국제공개 WO2020/222235호는 HVEM과 BTLA 간의 상호작용을 저해하고 세포 내에서 HVEM을 통한 하류 신호전달(downstream signaling)을 활성화하는 HVEM에 특이적으로 결합하는 항체를 개시하고 있다.
지금까지 HVEM이 BTLA나 gD에 결합하는 것을 저해하되 LIGHT에 결합하는 것을 촉진하는 항-HVEM 항체는 알려져 있지 않았다.
본 개시의 목적은 인간 HVEM에 높은 친화도로 결합하면서, HVEM의 BTLA 및/또는 gD에 대한 결합을 저해하되 LIGHT에 대한 결합을 촉진하는, 항-HVEM 항체를 제공하는 것이다.
본 개시의 다른 목적은 상기 항체를 코딩하는 단리된 핵산을 제공하는 것이다.

정의
용어 '항체'는 전체 항체(whole antibody) 및 임의의 항원 결합 단편 또는 그의 단일쇄를 포함한다. '항체'는 디설파이드(disulfide) 결합에 의해 상호 연결된 적어도 2개의 중쇄(H) 및 2개의 경쇄(L) 또는 이의 항원 결합 부분을 포함하는 당단백질을 포함한다. 각각의 중쇄는 중쇄 가변부위(VH 또는 HV) 및 중쇄 불변부위로 구성된다. 중쇄 불변부위는 4개의 영역, 즉 CH1, 힌지(hinge), CH2 및 CH3으로 이루어진다. 각각의 경쇄는 경쇄 가변부위(VL 또는 LV) 및 경쇄 불변부위로 이루어진다. 경쇄 불변부위는 하나의 영역, 즉 CL로 이루어진다. 용어 '인간 항체'는 인간 또는 인간 세포에 의해 생산되거나 인간 항체 레파토리 또는 다른 인간 항체-코딩 서열을 이용하는 비-인간 공급원으로부터 유래된 항체의 아미노산 서열에 상응하는 아미노산 서열을 보유하는 항체이다.
용어 '인간화 항체'는 분자의 항원 결합부는 실질적으로 비인간 종으로부터의 면역글로불린으로부터 유래한 반면, 분자의 나머지 면역글로불린 구조는 인간 면역글로불린의 구조 및/또는 서열에 기초하는 것인 분자를 가리킨다.
용어 '키메라 항체'는 중쇄 및/또는 경쇄의 일부는 특정한 공급원 또는 종으로부터 유래된 반면, 중쇄 및/또는 경쇄의 나머지는 다른 공급원 또는 종으로부터 유래된 항체를 지칭한다.
용어 '항-HVEM 항체를 코딩하는 단리된 핵산'은 항체 중쇄 및 경쇄(또는 그의 단편)를 코딩하는 하나 이상의 핵산 분자를 지칭한다.
용어 '숙주 세포'는 외인성 핵산이 도입된 세포를 지칭한다. 숙주 세포는 형질전환체 및 형질전환된 세포를 포함하며, 이는 1차 형질전환된 세포 및 계대배양 횟수와 관계없이 그로부터 유래된 자손을 포함한다.

용어 '투여' 또는 '투여하는 것'은 제약 조성물 또는 유효성분을 대상에게 제공하는 단계를 의미한다. 제약 조성물은 다양한 적절한 경로를 통하여 투여될 수 있다.
용어 '치료 유효량'은 유의성 있는 부정적이거나 불리한 부작용을 야기하지 않으면서 질환, 장애 또는 이상을 치료하는 데에 요구되는 작용제를 포함하는 제약 조성물의 수준, 양 또는 농도를 의미한다.
용어 '치료하다', '치료하는 것' 또는 '치료'는 예를 들어 상처를 입거나 손상된 조직의 치유, 또는 기존의 것이거나 인지된 질환, 장애 또는 이상을 변경, 변화, 강화, 개선, 개량 및/또는 미화하는 것에 의해, 원하는 치료상의 결과를 달성하도록 하는, 질환, 장애 또는 이상의 경감 또는 감소(일부 감소, 상당한 감소, 거의 완전한 감소 및 완전한 감소 포함), 해결 또는 예방(일시적이거나 영구적인 것)을 의미한다. '예방'은 질병, 장애 또는 질환의 발병의 지연을 의미한다. 질병, 장애 또는 질환의 발병이 예정된 기간 동안 지연된 경우 예방은 완전한 것으로 간주될 수 있다.
용어 '병용'은 사전에 투여된 치료제가 체내에서 여전히 유효한 동안 제2 치료제가 투여되도록 하는 2종 이상의 상이한 치료제의 임의의 형태를 가리킨다. 예를 들어, 2종의 치료제는 대상에서 동시에 유효하며, 이는 2종의 치료제의 상승 효과를 포함할 수 있다. 상이한 치료제는 단일의 제제 또는 별개의 제제로 동시에 또는 순차적으로 투여될 수 있다.
항-HVEM 항체, 핵산, 숙주세포, 생산방법

본 개시의 다른 양상에 따르면, 상기 항-HVEM 항체를 코딩하는 단리된 핵산이 제공된다. 상기 핵산은 항체의 VL을 포함하는 아미노산 서열 및/또는 VH를 포함하는 아미노산 서열을 코딩할 수 있다.
본 개시의 또 다른 양상에 따르면, 상기 핵산을 포함하는 숙주 세포가 제공된다. 숙주 세포는 항체의 VL을 포함하는 아미노산 서열 및 항체의 VH를 포함하는 아미노산 서열을 코딩하는 핵산을 포함하는 벡터; 또는, 항체의 VL을 포함하는 아미노산 서열을 코딩하는 핵산을 포함하는 제1 벡터 및 항체의 VH를 포함하는 아미노산 서열을 코딩하는 핵산을 포함하는 제2 벡터를 포함할 수 있다. 예를 들어, 숙주 세포는 진핵 세포, 예를 들어 차이니즈 햄스터 난소(Chinese Hamster Ovary, CHO) 세포일 수 있다.
본 개시의 또 다른 양상에 따르면, 항체의 발현에 적합한 조건 하에 상기 항체를 코딩하는 핵산을 포함하는 숙주 세포를 배양하는 것, 및 선택적으로 숙주 세포 또는 그 배양 배지로부터 항체를 회수하는 것을 포함하는, 항-HVEM 항체를 생산하는 방법이 제공된다.
제약 조성물, 용도 및 방법
본 개시의 또 다른 양상에 따르면, 유효성분으로서 상기 항체 및 제약상 허용되는 담체를 포함하는, 암을 치료하기 위한 제약 조성물이 제공된다.
제약상 허용되는 담체는 조성물을 증진 또는 안정화하거나, 조성물의 제조를 용이하게 한다. 제약상 허용되는 담체는 생리학상 상용성인 용매, 분산 매질, 코팅제, 항박테리아제 및 항진균제, 등장화제 및 흡수 지연제 등을 포함한다.
본 개시의 제약 조성물은 관련 기술분야에 공지된 다양한 방법에 의해 투여될 수 있다. 투여 경로 및/또는 방식은 목적하는 결과에 따라 달라진다. 투여가 정맥내, 근육내, 복강내 또는 피하이거나, 표적 부위에 인접하게 투여되는 것일 수 있다. 제약상 허용되는 담체는 정맥내, 근육내, 피하, 비경구, 척수 또는 표피 투여(예를 들어, 주사 또는 주입에 의함)에 적합해야 한다. 조성물은 멸균이고 유체여야 한다. 조성물은 동결건조된 형태일 수 있다. 등장화제, 예를 들어 당, 폴리알콜, 예컨대 만니톨 또는 소르비톨, 및 염화나트륨을 조성물 내에 포함시킬 수 있다. 본 개시의 제약 조성물은 관련 기술분야에 널리 공지되고 통상적으로 실시되는 방법에 따라 제조될 수 있다.
제약 조성물 중 유효성분의 투여량 수준은, 환자에게 독성이 아니면서 특정한 환자, 조성물 및 투여 방식에 대해 목적하는 치료 반응을 달성하기에 효과적인 유효성분의 양을 수득하도록 달라질 수 있다. 선택된 투여량 수준은 다양한 약동학적 인자, 예컨대 사용된 본 개시의 특정한 조성물, 또는 투여 경로, 투여 시간, 배출 속도, 치료 지속기간, 조합되어 사용된 다른 약물, 화합물 및/또는 물질, 치료할 환자의 연령, 성별, 체중, 상태, 전반적 건강 및 이전 병력 및 기타 인자에 따라 달라진다. 비제한적인 예로서, 투여량은 약 0.0001 내지 100 ㎎/㎏ 체중, 보다 통상적으로는 0.1 내지 20 ㎎/㎏ 체중의 범위이다. 예시적인 치료 요법은 1주마다 1회, 또는 2주마다 1회, 또는 매월 1회, 또는 3 내지 6개월마다 1회 투여를 수반한다.

일 구현예에서, 상기 조성물은 하나 이상의 항암제를 추가로 포함하는 것일 수 있다. 예를 들어, 항암제는 화학요법제 또는 면역요법제일 수 있다. 화학요법제의 비제한적인 예는 알킬화제, 나이트로소우레아제, 항대사물질, 항암 항생제, 식물-기원 알칼로이드, 토포아이소머라제 저해제, 호르몬 약물, 호르몬 길항제, 백혈구감소증(호중구감소증) 치료 약물, 혈소판감소증 치료 약물, 항구토제, 아로마타제 저해제, P-당단백질 저해제, 백금 착물 유도체, 다른 면역치료 약물 및 다른 항암 약물을 포함한다. 병용 투여될 수 있는 예시적인 세포독성제는 항미세소관 작용제, 토포아이소머라제 저해제, 항대사물질, 유사분열 저해제, 알킬화제, 안트라사이클린, 빈카 알카로이드, 인터칼레이팅제, 신호 전달 경로를 방해할 수 있는 작용제, 아포토시스를 촉진하는 작용제, 프로테오좀 저해제 및 방사선(국소 또는 전신 조사)을 포함한다. 추가적인 치료제의 비제한적인 예는 펩타이드, 폴리펩타이드, 단백질, 융합 단백질, 핵산 분자, 소분자, 모방 작용제, 합성 약물, 무기 분자 및 유기 분자를 포함하지만 이들로 제한되지 않는다.
면역요법제는 암세포에 대한 신체의 면역 반응을 간접적으로 또는 직접적으로 증강, 자극 또는 증가시키고/거나 다른 항암 요법의 부작용을 감소시키는 화합물, 조성물 또는 치료를 의미한다. 면역요법제의 비제한적인 예는 사이토카인, 암 백신, 단일클론 항체, 비사이토카인 보조제, 면역 세포(T 세포, NK 세포, 수지상 세포, B 세포 등), 면역관문 억제제, 미생물 등을 포함한다. 구체예에서, 면역요법제는 면역관문 억제제이다. 면역관문 억제제는 펩티드, 항체, 핵산 분자 및 소형 분자를 포함한다. 예를 들어, 면역관문 억제제는 대상에서 CD8+ T 세포의 증식성, 이동성, 지속성 및/또는 세포독성 활성, 및 특히 대상의 CD8+ T 세포의 종양 침윤성을 증강시키기 위해 투여될 수 있다. 전형적으로, 면역관문 억제제는 활성화된 T 림프구, 예를 들어 세포독성 T 림프구-연관 단백질 4(CTLA4) 및 프로그램된 세포사멸 1(PD-1), 또는 살해 세포 면역글로불린-유사 수용체(KIR) 패밀리의 다양한 구성원과 같은, NK 세포에 의해 발현되는 면역억제성 수용체를 차단하는 길항제이거나, 이들 수용체의 주요 리간드, 예를 들어 PD-1 리간드 CD274(PD-L1 또는 B7-H1로 가장 잘 알려짐)를 차단하는 길항제이다. 예를 들어, 면역관문 억제제는 항체 또는 이의 항원-결합 단편이다. 구체적으로, 면역관문 억제제는 항-PD-1 항체, 항-PD-L1 항체, 항-PD-L2 항체, 항-CTLA-4 항체, 항-TIM-3 항체, 항-LAG3 항체, 항-IDO1 항체, 항-TIGIT 항체, 항-VISTA 항체, 항-B7H3 항체, 항-B7H4 항체, 항-BTLA 항체, 및 항-B7H6 항체, 및 이의 항원-결합 단편으로 이루어진 군으로부터 선택되는 하나 이상일 수 있다. 보다 구체적으로, 면역관문 억제제는 이필리무맙(Ipilimumab)(여보이®, BMS/오노), 트레멜리무맙(Tremelimumab)(아스트라제네카), 아테졸리주맙(atezolizumab)(티쎈트릭®, 로슈), 니볼루맙(nivolumab)(옵디보®, BMS/오노), 펨브롤리주맙(Pembrolizumab)(키트루다®, MSD), 아벨루맙(Avelumab)(바벤시오®, 화이자/독일머크), 더발루맙(Durvalumab)(임핀지®, 아스트라제네카/메드이뮨), 및 이의 항원-결합 단편 중에서 선택되는 하나 이상일 수 있으나, 이들로 제한되는 것은 아니다.
일 구현예에서, 면역요법제는 각각의 사용 목적에 맞게 통상의 방법에 따라 액제, 현탁제, 산제, 과립제, 정제, 캡슐제, 환제, 엑스제, 에멀젼, 시럽제, 에어로졸 등의 경구 제형, 멸균 주사용액의 주사제 등의 비경구 제형과 같이 다양한 형태로 제형화하여 사용할 수 있다. 면역요법제는 경구 투여되거나, 정맥 내, 복강 내, 피하, 피내, 근육 내, 척추, 척추강 또는 직장 내 국소 투여 또는 주입 등을 포함한 다양한 경로를 통해 비경구 투여될 수 있다. 투여량은 환자의 체중, 연령, 성별, 건강상태, 식이, 투여 시간, 투여 방법, 투여 기간 또는 간격, 배설율, 체질 특이성, 제제의 성질, 질환의 중증도 등에 따라 그 범위가 다양할 수 있으며, 통상의 기술자에 의해 적절하게 선택될 수 있다. 예를 들어, 약 0.1 내지 10,000 ㎎/㎏의 범위일 수 있으나 이에 제한되지 않으며, 하루 일회 내지 수회에 나누어 투여될 수 있다.
본 개시의 제3 측면은 치료 유효량의 상기 항체를 암 치료를 필요로 하는 대상에게 투여하는 단계를 포함하는, 대상에서 암을 치료하는 방법이 제공된다.
투여되는 대상은 인간이나 동물, 예를 들어 인간, 돼지, 개, 고양이, 소, 말, 쥐 등을 제한 없이 포함할 수 있다.
이하, 본 개시를 실시예를 들어 구체적으로 설명하나, 이는 본 개시의 이해를 돕기 위한 것일 뿐 본 개시의 범위를 어떤 식으로든지 제한하는 것은 아니다.
실시예 1: 항체 스크리닝
HVEM(Herpes virus entry mediator; TNFRSF14)에 결합하는 항체를 제작하기 위하여, 파지 디스플레이 라이브러리를 사용하였다. 파지 디스플레이드-scFv 라이브러리는 필라멘트형 박테리오파지의 pIII 말단에 합성된 인간 항체 서열로 구성된 단일쇄 가변 단편(scFv)이 융합된 형태의 라이브러리로, 인간 항체의 중쇄 가변영역(VH) 및 경쇄 가변영역(VL)으로 구성되어 있으며, VL은 카파 경쇄이다.
HVEM에 특이적 친화력을 가지는 인간 항체를 선별하기 위하여, 두 가지 전략으로 패닝을 진행하였다. 첫 번째 전략은 인간 HVEM으로만 3 라운드의 패닝을 진행하는 것이고, 두 번째 전략은 인간 HVEM으로 1차와 2차 라운드의 패닝을 진행하고 생쥐 HVEM으로 3차 라운드의 패닝을 진행하는 것이었다.
각 전략에서 획득한 파지 풀을 E. coli에 감염시켜 단일 콜로니 형태로 만들어 모노클로날 ELISA(enzyme-linked immunosorbent assay)를 수행하였다. 단일 콜로니를 카르베니실린이 포함된 SB 배지(Super Broth Medium)에서 37 ℃에서 O.D 0.5가 될 때까지 배양한 후, 1 mM IPTG(Isopropyl β-D-1-thiogalactopyranoside)를 넣고 30 ℃ 밤새 배양하였다. 원심분리를 통해 상등액을 제거하고 펠렛을 TES 버퍼로 재현탁하여 세포간극 추출물(periplasmic extract)을 획득하였다. 획득한 세포간극 추출물을 블로킹 버퍼와 1:1 혼합하여 ELISA에 사용하였으며 HVEM에 결합한 항체는 HA(hemaglutinin) 태그를 통해 검출하였다. 양성 클론은 O.D가 0.2 이상이며, 백그라운드 대비 O.D가 5배 이상인 클론으로 설정하였다. 전략 1을 통해 인간 HVEM에 결합하는 73개를 선별하였으며 그 중 34개를 무작위로 선정하여 생쥐 HVEM과의 교차 반응을 ELISA를 통해 확인하였다. 34개 중 3개를 제외한 31개가 교차 반응을 보였다. 그리고 전략 2를 통해 인간 HVEM에 특이적인 2개 클론과 인간과 생쥐 HVEM에 교차 반응을 보이는 57개의 클론을 선별하였다. 이렇게 선별된 클론들의 DNA 염기서열 분석을 통해 인간과 생쥐 HVEM에 교차반응을 보이며 다른 염기서열을 갖는 50개의 개별 클론을 확보하였다.
실시예 2: scFv 항체 정제 및 HVEM 항원 결합능 검증
확보된 HVEM 항체의 항원 결합능은 ELISA를 통해 확인하였다. 선별된 클론들을 HB2151(non-suppressor E. coli)에 형질전환시켜 scFv 형태의 항체를 정제하여 진행하였다. 형질전환시킨 HB2151을 SB 배지(+카르베니실린)에서 O.D 0.5가 될 때까지 37 ℃에서 배양하고 최종농도가 1 mM이 되도록 IPTG를 첨가한 후 30 ℃에서 밤새 배양하였다. 배양액을 원심분리를 통해 상등액을 제거하고 펠렛을 획득하였다. 획득한 펠렛을 TES 버퍼로 재현탁하고 원심분리를 통해 상등액을 확보하였다.
scFv가 포함된 세포간극 추출물로부터의 scFv의 정제를 위해 말단에 포함된 6×his 태그를 이용하여 Ni-NTA 정제를 하였다. 정제를 통해 50종 중 44종의 scFv를 확보하였다.
정제된 항체를 사용하여, 인간 HVEM 및 생쥐 HVEM 항원 각각에 대한 결합능(affinity)을 측정하였다. 측정법으로는 ELISA를 사용하였다. 인간 HVEM 항원에 대한 결합능이 10 nM 이하, 생쥐 HVEM 항원에 대한 결합능이 50 nM 이하인 항체를 33종 선별하였다. 선별된 클론과 그 결합능을 아래 표 2에 나타내었다.

실시예 3: 전체 크기 인간 항체 제작
선별된 scFv 항체의 VH, VL과 인간 항체의 중쇄 불변영역(CH) 및 경쇄 불변영역(CL)을 사용하여, 전체 크기 인간 항체를 제작하였다.
전체 크기 인간 항체의 중쇄(HC)를 제작하기 위하여, 시그널 서열, 선별된 scFv 항체의 VH 부위, 인간 항체 IgG4의 CH를 융합하였다. 시그널 펩타이드는 항체의 고발현 및 배지로의 배출을 위하여, 인간 IL-2의 시그널 펩타이드를 사용하였다. 인간 항체 IgG4의 CH는 자연상의 중쇄 CH를 기본으로 사용하였으며, 힌지 부위의, EU numbering 시스템 기준, 228번 세린은 프롤린으로 대체하였다.
전체 크기 인간 항체의 경쇄(LC)을 제작하기 위하여, 시그널 서열, 선별된 scFv 항체의 VL 부위, 인간 항체 CL을 융합하였다. 시그널 펩타이드는 항체의 고발현 및 배지로의 배출을 위하여, 인간 IL-2의 시그널 펩타이드를 사용하였다. 인간 항체 경쇄의 CL은 자연상의 카파 경쇄의 CL을 사용하였다. 해당 항체를 제작하기 위하여 융합된 CH, CL의 아미노산 서열은 각각 서열번호 89 및 서열번호 90과 같다.
항체를 발현하는 벡터를 제작하기 위하여, 인간 IL-2 시그널 펩타이드 및 CH, CL을 클로닝하여, 카세트 벡터를 제작하였다. 항체의 VH, VL은 scFv 항체 서열로부터 PCR법을 사용하여 증폭하여 클로닝에 사용하였다. 항체의 중쇄를 발현하는 벡터를 제작하기 위하여, 중쇄 카세트 벡터를 AfeI 제한 효소로 절단하고, 이에 PCR법을 사용하여 증폭된 VH를 클로닝하였다. 항체의 경쇄를 발현하는 벡터를 제작하기 위하여, 카파 경쇄 카세트 벡터를 AfeI, BsiWI 제한 효소로 절단하고, 이에 PCR법을 사용하여 증폭된 VL을 클로닝하였다. HVEM011, HVEM021 항체에 대해서는 CHO 세포 코돈 최적화가 진행된 DNA 서열을 합성한 후에 이를 PCR 법으로 증폭하여 클로닝에 사용하였다.
항체 단백질의 제조를 위하여, 각 항체의 중쇄(HC)를 발현하는 벡터와 경쇄(LC)를 발현하는 벡터를 사용하였다. ExpiCHO-S 세포주에 각 항체를 발현하는 벡터 2종(HC, LC)을 동시형질전환(co-transfection)을 실시하여 항체의 발현을 유도하였다. 세포를 포함하는 부유물을 제거한 배양액은, MabselectSure 및 Hitrap SP FF 정제 컬럼을 사용하여 정제를 수행하였다. 정제된 단백질은 SEC-HPLC를 통한 순도 분석, 엔도톡신 분석을 수행한 이후에 인 비트로 시험, 세포 기반 시험, 동물 시험에 사용되었다.
실시예 4: 항체/항원 결합능 분석
전체 크기 인간 항체의 HVEM에 대한 결합능 분석은 ELISA 시험법을 사용하여 수행하였다. 인간 HVEM-hFc, 게잡이 원숭이 HVEM-hFc, 생쥐 HVEM-mFc 단백질을 각각 0.2 ㎍/㎖로 제작한 이후, 96-하프 웰 면역흡착검사용 플레이트에 코팅하였다. 비특이적 결합을 억제하기 위하여, 3% BSA(bovine serum albumin)가 포함된 인산염 완충 염수(PBS)에 1시간 반응시킨 후에, 순차 희석된 항체를 1시간동안 처리하였다. 결합된 항체의 양을 측정하기 위하여 HRP(horse radish peroxidase)가 결합된 항-인간 IgG4 항체를 사용하였으며, HRP의 활성은 TMB(3,3',5,5'-Tetramethylbenzidine) 기질 용액을 사용하여 발색반응을 유도하고, 1N 황산으로 반응을 종료시켰다. 발색반응의 정도는 SpectraMax i3(Molecular Devices, 10192-220)를 사용하여 확인하였다. 각 단계의 중간에는 0.1% 트리톤 X-100이 포함된 PBS(PBST)를 사용하여 3회 세척을 수행하였다. 항체의 결합능은 SigmaPlot을 사용하여 도출하였다. 항-HVEM 항체의 인간 HVEM에 대한 결합 친화도는 도 1a, 항-HVEM 항체의 마우스 HVEM에 대한 결합 친화도는 도 1b와 같다. 항-HVEM 항체의 인간, 게잡이 원숭이 및 마우스 HVEM에 대한 교차 반응성에 대한 결과는 각각 도 2a, 도 2b, 도 2c 및 도 2d에 나타내었다.
인간 HVEM 항원에 대한 결합능은 Octet BLI 측정법을 이용하여 추가로 측정하였다. Octet BLI를 사용하여 측정한 인간 HVEM 항원에 대한 HVEM 항체의 결합력은 도 3a, 도 3b (HVEM011), 및 도 3c (HVEM021)에 나타내었다.
실시예 5: 항원/항체 경쟁 결합에 의한 에피토프 규명
HVEM 항체의 경쟁 결합을 측정하기 위하여, 세포 기반 어세이를 수행하였다. HVEM 항체와 BTLA 단백질의 HVEM 경쟁 결합을 측정하기 위하여, Jurkat-Lucia™NFAT(InvivoGen) 세포주에 BTLA 단백질을 안정 발현시키고 이를 사용하였다. 각각 5×105개의 세포에 1 ㎍/㎖의 인간 HVEM-Fc 단백질과 HVEM 항체 10nM 혹은 100nM을 처리하고 2시간동안 얼음에서 반응시켰다. 세포주에 결합된 인간 HVEM-Fc를 측정하기 위하여, Alexa488이 결합된 항-인간항체 IgG1(Invitrogen A10631)을 사용하였다. 유세포분석을 위하여, 염색이 완료된 세포주는 2% 포름알데하이드가 포함된 PBS를 사용하여 20분간 얼음에서 고정시킨 후, 유세포분석 시까지 냉장 보관하였다. 유세포분석은 FACSVerse™ 유동 세포계수기(BD Biosciences)를 사용하여 측정하였으며, 결합된 인간 HVEM-Fc의 양은 FlowJo_v10 프로그램을 사용하여 분석하였다. 해당 결과는 도 4와 같으며, 사용된 HVEM 항체는 모두 세포 표면의 HVEM에 대해서 BTLA와 경쟁적 결합이 관찰되었다.
HVEM 항체와 LIGHT 단백질의 HVEM 경쟁 결합을 측정하기 위하여, Jurkat-Lucia™NFAT(InvivoGen) 세포주에 HVEM 단백질을 안정 발현시키고 이를 사용하였다. 각각 5×105개의 세포에 0.25 ㎍/㎖의 인간 LIGHT 단백질과 HVEM 항체 10nM 혹은 100nM을 동시에 처리하고 2시간동안 얼음에서 반응시켰다. 세포주에 결합된 인간 LIGHT 단백질을 측정하기 위하여, 알로피코시아닌(Allophycocyanin, APC)이 결합된 항-His 항체(Abcam ab72579)를 사용하였다. 유세포분석을 위하여, 염색이 완료된 세포주는 2% 포름알데하이드가 포함된 PBS를 사용하여 고정시키고, 유세포분석 시까지 냉장 보관하였다. 유세포분석은 FACSVerse™ 유동 세포계수기(BD Biosciences)를 사용하여 측정하였으며, 결합된 인간 LIGHT 단백질의 양은 FlowJo_v10 프로그램을 사용하여 분석하였다. 그 결과를 도 5에 나타내었다. HVEM002 항체 등 일부의 항체만 LIGHT 단백질과 경쟁적 결합이 관찰되며, HVEM011, HVEM021을 포함하는 항체들은 LIGHT 결합을 증가시킴이 관찰되었다.
HVEM 항체와 허피스 바이러스 당단백질 D(gD)의 HVEM 경쟁 결합을 측정하기 위하여, Jurkat-Lucia™NFAT(InvivoGen) 세포주에 HVEM 단백질을 안정 발현시키고 이를 사용하였다. 각각 5×105개의 세포에 10 μM의 gD 단백질을 2시간동안 반응시키고, 이에 HVEM 항체 1nM을 추가하고 1시간 반응을 진행하였다. 세포주에 결합된 HVEM 항체를 측정하기 위하여, 플루오레세인 이소티오시아네이트(FITC)가 결합된 항-인간항체 IgG4(Abcam ab99821)를 사용하였다. 유세포분석을 위하여, 염색이 완료된 세포주는 2% 포름알데하이드가 포함된 PBS를 사용하여 고정시키고, 유세포분석 시까지 냉장 보관하였다. 유세포분석은 FACSVerse™ 유동 세포계수기(BD Biosciences)를 사용하여 측정하였으며, 결합된 HVEM 항체의 양은 FlowJo_v10 프로그램을 사용하여 분석하였다. 그 결과를 도 6에 나타내었다. 시험에서 사용된 대부분의 HVEM항체들은 허피스 바이러스 당단백질 D와 세포 표면의 HVEM에 대해서 경쟁적 결합이 관찰되었다.
실시예 6: HVEM 면역관문 억제능
HVEM 항체에 의한 HVEM-BTLA 면역관문 억제능을 측정하고자, HVEM-BTLA 면역관문이 작동하는 세포 기반 어세이 시스템을 구축하였다. 우선 BTLA를 발현하는 T 세포를 제작하기 위하여, Jurkat-Lucia™NFAT(InvivoGen) 세포주에 BTLA를 안정 발현시켰다. 이를 위하여, 인간 BTLA를 발현하는 렌티바이러스를 감염시키고, BTLA의 발현이 >90% 이상의 세포에서 안정적으로 유지되는 조건으로 감염된 Jurkat-Lucia™NFAT 세포주를 사용하였다. 이 세포주를 Jurkat-luciaNFAT-hBTLA라 명명하였다.
Jurkat-luciaNFAT-hBTLA 세포주를 활성화시키기 위하여, 5 ㎍/㎖로 희석된 항-CD3 작용제 항체(OKT3)를 50 ㎕를 사용하여 밤샘 코팅된 96웰 세포배양 플레이트를 사용하였다. HVEM-BTLA 면역관문에 의한 면역 억제 반응을 유도하기 위하여, 인간 HVEM-Fc 단백질을 세포 배양액에 처리하였다. 인간 HVEM-Fc 40nM과 5×105 세포를 섞은 후, OKT3 항체 코팅된 플레이트에서 24시간동안 100 ㎕의 배양액에서 배양을 진행하였다. 항체의 HVEM-BTLA 면역관문 억제능을 측정할 때에는, HVEM 항체 및 아이소타입 컨트롤 항체들을 사용하였으며, 인간 HVEM-Fc 및 Jurkat-luciaNFAT-hBTLA 세포주와 함께 항체를 첨가한 후에 배양을 진행하였다. 24시간 반응 후에 배양액을 수거하였으며, 배양액에 분비된 루시아 루시퍼레이즈의 활성은 QUANTI-Luc™(InvivoGen) 시약을 사용하여 측정하였다. 배양액 10 ㎕와 QUANTI-Luc™ 시약 50 ㎕를 혼합한 후에, 루미노미터를 사용하여, 발광을 측정하였다.
그 결과를 도 7에 나타내었다. 본 실험에서 Jurkat-luciaNFAT-hBTLA 세포주는 인간 HVEM-Fc 단백질 처리시 면역관문이 활성화되어서 Jurkat-luciaNFAT-hBTLA의 루시퍼레이즈 활성이 낮게 관찰되었으나, HVEM 항체가 처리될 경우, 면역관문이 비활성화됨에 따른 Jurkat-luciaNFAT-hBTLA의 루시퍼레이즈 활성이 회복되는 것이 관찰되었다.
실시예 7: 동물 모델에서의 항암 효능 검증
해당 HVEM 항체의 항종양 동물 효능을 검증하기 위하여, 아래의 모델을 사용하였다.
생쥐 호모그래프트 모델인 Pdx1-Cre; Kras(G12D/+); Trp53-/-(KPC) mPA6115 모델은 췌장암 모델에서의 효능 검증을 위하여, 비히클(PBS) 대조군 대비 HVEM 항체 투여군에서의 암조직의 부피를 측정하여 비교하였다. 2~3 ㎜의 지름을 가지는 암조직을 피하에 이식하여 모델을 구축하였다. 암조직의 부피가 약 100 ㎣가 될 때에 투여를 개시하였다. 투여는 주 2회, 총 4회 투여가 되었으며, 투여된 항체량은 생쥐의 몸무게를 기준으로 하여 10 ㎎/㎏의 용량을 사용하였으며, 투여 경로는 복강투여이다. 암조직 부피의 측정은 칼리퍼(Caliper)를 사용하여, 암조직 최대 길이(L), 암조직 최대길이에 대한 폭(W)를 측정하고, (V=L×W×W/2)의 식을 사용하여 계산하였다. 그 결과를 도 8에 나타내었다. 군분리 및 최초 투여 14일 후 측정시, 음성대조군 대비 암성장 억제율은 41.69%를 나타내었다.
항-CTLA-4 및 항-PD-1 항체와의 병용 효능을 알아보기 위하여, CT-26 생쥐 모델을 사용하였다. 5×105개의 CT-26 세포를 피하에 이식하고 암조직의 부피가 50 ㎣ 도달시 군분리 후에 투여를 개시하였다. 투여는 HVEM 항체 10 ㎎/㎏, RMP1-14 항-PD-1 항체(BioXCell) 10 ㎎/㎏, 9H10 항-CTLA-4 항체(BioXCell) 5 ㎎/㎏의 용량으로 단독 혹은 병용으로 투여하였으며, 아이소타입 대조군 항체를 HVEM 항체의 음성 대조군으로 사용하였다. 항체 투여는 주 2회, 총 4회 진행하였다. 그 결과를 도 9에 나타내었다. 9H10 항-CTLA-4 항체 단독 및 병용 투여군은 높은 암성장 억제율을 관찰하였으며, 항-PD-1 및 HVEM 항체 병용시 9H10 항-CTLA-4 항체 단독 투여 대비 암성장 억제율을 증가시켰다. HVEM 항체의 병용투여는 항-PD-1 병용투여 대비하여 유의미한 암성장 억제율 증가가 관찰되었다(암세포 이식후 29일, 최초투여 후 23일 기준).
CT-26 암세포주에 생쥐 HVEM의 발현을 안정적으로 증가시킨 세포주를 제작하고 이를 이용하여 생쥐 모델을 제작하였다. CT-26 세포주에 생쥐 HVEM을 발현하는 벡터를 사용하여 형질전환시킨 후에, 생쥐 HVEM이 높게 안정발현이 이루어지는 세포를 분리하여 세포주를 제작하고, 이를 CT-26-mHVEM#2-2이라 명명하였다. 5×105개의 CT-26-mHVEM#2-2 세포를 피하에 이식하고, 3일 후 항체 투여를 개시하였으며, 각각의 항체는 10 ㎎/㎏ 기준으로 주 2회, 총 5회 복강 투여를 진행하였다. 아이소타입 대조군은 음성 대조군으로 사용하였으며, 병용투여 물질로 RMP1-14/hIgG4를 사용하였다. RMP1-14/hIgG4는 항-PD-1 RMP1-14 항체의 가변영역과 인간 항체의 불변 영역을 융합한 키메라 항체이다. 그 결과를 도 10에 나타내었다. HVEM 항체는 항-PD-1 항체와 병용투여 시에, 항-PD-1 항체 단독처리군 대비하여 유의미한 암성장 억제율 증가가 관찰되었다.
실시예 8: 인간화 동물 모델에서의 항암 효능 검증
인간 면역세포 및 암세포에 대한 항암 효능을 알아보기 위하여, 인간화 생쥐 모델을 사용하였다. 인간화 암모델 생쥐를 제작하기 위하여, NSGA 면역부전 생쥐, SNU407 혹은 PC3 인간 암세포주 및 인간 혈액에서 분리한 PBMC(peripheral blood mononuclear cell)를 사용하였다. SNU407 인간 암세포주는 HVEM을 높게 발현하며, PD-L1 발현이 매우 낮게 관찰됨을 확인하였다. PC3 인간 암세포주는 HVEM 및 PD-L1의 발현이 모두 관찰됨을 확인하였다.
NSGA 면역부전 생쥐에 인간 암세포주 SNU407을 이식하여 인간 암조직을 만들고, 3일 후에 인간 혈액에서 분리된 PBMC를 이식하여, 인간 면역세포가 생쥐에 정착하도록 유도하였다. PBMC 이식 1주일 후에 항체 투여를 개시하였으며, 각각의 항체는 10 ㎎/㎏을 기준으로 주 2회, 총 5회 복강투여를 실시하였다. 아이소타입 대조군은 음성 대조군으로 사용하였으며, 이식된 인간 암세포의 PD-L1 결합하는 항체의 대조군으로 아테졸리주맙(Atezolizumab)을 사용하였다. HVEM 항체 2종의 항체를 비교 검증하였다.
그 결과를 도 11에 나타내었다. HVEM의 발현이 높은 SNU407 모델에서 아테졸리주맙에 비하여 HVEM 항체의 암성장 억제 효과가 높음을 확인하였다.
NSGA 면역부전 생쥐에 인간 암세포주 PC3를 이식하여 인간 암조직을 만들고, 3일 후에 인간 PBMC를 이식하여, 인간 면역세포가 생쥐에 정착하도록 유도하였다. PBMC 이식 후 4일 뒤에 항체 투여를 개시하였으며, 각각의 항체는 10 ㎎/㎏으로 투여되었으며, 병용투여를 기준으로 하여, 총 20 ㎎/㎏의 항체 혼합물이 투여되었다. 항체는 주 2회, 총 11회 복강투여를 실시하였다.
그 결과를 도 12에 나타내었다. HVEM 및 PD-L1의 발현이 모두 관찰되는 PC3 모델에서 HVEM 항체 및 PD-L1 항체의 병용투여는 단독투여에 비하여 암성장 억제 효과가 증가됨을 확인하였다.
Claims (20)
- 500 pM 이하의 해리상수(Kd)로 인간 HVEM((Herpes Virus Entry Mediator)에 결합하고, HVEM의 BTLA(B and T lymphocyte attenuator)에 대한 결합을 저해하되 HVEM의 LIGHT에 대한 결합을 촉진하는, 항-HVEM 모노클로날 항체.
- 제1항에 있어서, 50 pM 이하의 해리상수(Kd)로 인간 HVEM에 결합하는 항체.
- 제1항 또는 제2항에 있어서, HVEM의 gD에 대한 결합을 저해하는 항체.

- 제10항에 있어서, 숙주 세포 배양물로부터 항-HVEM 항체를 회수하는 것을 추가로 포함하는 방법.
- 유효성분으로서 제1항 내지 제7항 중 어느 한 항의 항체 및 제약상 허용되는 담체를 포함하는, 암을 치료하기 위한 제약 조성물.

- 제14항 내지 제18항 중 어느 한 항에 있어서, 다른 치료제와 동시 또는 순차적으로 단일 또는 분리된 제제로 투여하기 위한 조성물.
- 치료 유효량의 제1항 내지 제7항 중 어느 한 항에 따른 항체를 암 치료를 필요로 하는 대상에게 투여하는 단계를 포함하는, 대상에서 암을 치료하는 방법.
