Abstract
Description
本発明は、CD137に結合する抗体分子に関する。この抗体分子は、癌及び感染症などの疾患及び障害の治療及び診断に利用される。
細胞シグナル伝達は、全ての生物の生命に不可欠な部分であり、通常、可溶性リガンド又は表面で発現されるリガンドと相互作用する細胞表面受容体を必要とする。この相互作用は、受容体、リガンド又はその両方に変化をもたらす。例えば、リガンド結合は、受容体を一緒にクラスター化させて二量体又はオリゴマーにする、受容体の構造変化を誘発し得る。次に、このクラスター化効果が、細胞内シグナル伝達経路の活性化をもたらす。CD137などの、腫瘍壊死因子受容体スーパーファミリー(TNFRSF)のメンバーを含む、このように活性化される多くの受容体がある。
上記の背景技術の項において説明されるように、CD137アゴニスト分子の臨床開発は、治療が用量制限高度肝炎(ウレルマブ)又は低い臨床的有効性(ウトミルマブ)のいずれかを伴うため、遅れている。

本発明は、CD137に結合する抗体分子に関する。CD137は、腫瘍壊死因子受容体スーパーファミリーメンバー9(TNFRSF9)又は4−1BBとしても知られている。抗体分子は、好ましくは、ヒトCD137、より好ましくは、ヒト及びカニクイザルCD137、さらにより好ましくは、二量体ヒト及びカニクイザルCD137に結合する。抗体分子によって結合されるCD137の部分は、好ましくは、CD137細胞外ドメインである。ヒト及びカニクイザルCD137の細胞外ドメインは、配列番号112及び113のそれぞれに記載される配列を含むか又はそれからなり得る。本発明の抗体分子は、好ましくは、細胞の表面において発現されるCD137に結合することが可能である。細胞は、好ましくは、免疫細胞、例えば、CD8+又はCD4+ T細胞又は制御性T(Treg)細胞、好ましくは、CD8+ T細胞、又はB細胞、ナチュラルキラー(NK)細胞、ナチュラルキラーT(NKT)細胞、樹状細胞(DC)、又は腫瘍浸潤リンパ球(TIL)である。

実施例1−ヒト及びカニクイザルCD137抗原の産生、特徴付け及び選択
活性化T細胞は、それらの細胞表面においてCD137受容体を発現する。CD137受容体のクラスター化は、受容体シグナル伝達及びさらなるT細胞活性化を誘発するのに不可欠であることが知られている(Chester et al.,2018)。架橋時に、CD137受容体クラスター化及びその後のT細胞活性化を誘発する一方、架橋の非存在下でT細胞活性化を示さないか又は弱いT細胞活性化を示すモノクローナル抗体を単離することが望ましいであろう。この目的を達成するために、本発明者らは、抗CD137mAbは、細胞表面において発現される際、モノマーCD137に結合するはずであるが、二量体CD137抗原及びCD137を過剰発現する細胞によって模倣される、高レベルのCD137への優先的な結合を必要とする可能性があると仮説を立てた。したがって、組み換えモノマー及び二量体CD137、並びに細胞表面で発現されるCD137を、選択に使用するために産生した。
腫瘍壊死因子受容体スーパーファミリー(TNFRSF)メンバーは、それらの同種リガンドに結合される場合に一緒にクラスター化する多量体を形成する傾向が知られている(Croft,2003)。それらの機能性のために集合するこの傾向により、ファージディスプレイなどのインビトロ選択に使用するため、並びに選択されたタンパク質の特徴付けのために溶液中に集合しない可溶性組み換えタンパク質を産生することが困難になる。
完全長ヒトCD137(配列番号119)又はカニクイザルCD137(配列番号120)を発現するDO11.10細胞(National Jewish Health)(それぞれ、「DO11.10−hCD137」及び「DO11.10−cCD137」と示される)(表2を参照)を、選択された抗CD137 mAbの選択及びさらなる特徴付けのために、その天然形態に最も類似した膜結合構造で抗原を表すために産生した。
2.1 抗CD137クローンの選択、スクリーニング及び発現
CDR1、CDR2及びCDR3におけるランダム化を伴うヒト生殖細胞系列のFabドメインを示す合成ナイーブファージミドライブラリー(MSM Technologies)を、実施例1に記載される組み換えCD137抗原及び細胞表面で発現されるCD137抗原を用いた選択に使用した。
抗CD137 mAbの結合を、ヒト及びカニクイザルCD137−mFc−Aviを用いて、ELISAにおいて分析した。
抗ヒトCD137 mAb FS30−5、FS30−10、FS30−15及びFS30−16を、フローサイトメトリーを用いて、ヒト又はカニクイザルCD137(DO11.10−hCD137又はDO11.10−cCD137)を発現する細胞への結合について試験した。非特異的結合も、CD137発現を欠くDO11.10細胞及びHEK293細胞への結合を試験することによって評価した。結合親和性を、2つの陽性対照抗CD137 mAb、MOR7480.1(実施例1.2を参照及び20H4.9(米国特許第7288638号)の結合親和性と比較し、その可変ドメインをクローニングし、CH2ドメイン中にLALA突然変異を含むヒトIgG1フォーマット(G1AAフォーマット)において発現させて、抗体G1AA/MOR7480.1及びG1AA/20H4.9を得た。
選択されたmAbの生物物理学的特性の評価は、薬剤開発だけでなく、結合及び機能データの解釈のためにも重要である。具体的には、アゴニストT細胞活性化を分析する場合、集合体の存在が、抗体のクラスター化を模倣し、T細胞活性化を誘発し得る。したがって、FS30 mAbのモノマー割合のパーセンテージを、SECによって決定した。
次に、抗CD137 mAbの機能活性を、初代T細胞活性化アッセイにおいて分析した。インビボで、抗CD137 mAbは、Fcy受容体の動員によってアゴニズムを誘発し、それによって、mAbの架橋及びその後のCD137受容体のクラスター化を引き起こす。表面CD137受容体分子をクラスター化するmAbの最大能力を模倣するために、FS30 mAbを、アッセイの前に、抗ヒトCH2抗体(クローンMK1A6[Jefferis et al.,1985;Jefferis et al.,1992]、社内で産生される)を用いて架橋させた。T細胞活性化を、非架橋のmAbと比較した。FS30−5、FS30−10、FS30−15及びFS30−16を、FS30−6(結合ELISA(実施例2.2)においてカニクイザルCD137に対して交差反応性でないことが分かり、したがって異なるエピトープに結合する可能性が高いヒトCD137結合剤である)、並びに陽性対照抗CD137 mAb G1AA/MOR7480.1及びG1AA/20H4.9とともに試験した。LALA突然変異を有するヒトIgG1骨格中の抗ニワトリ卵白リゾチーム(HEL)抗体D1.3(Braden et al.,1996)(G1AA/HelD1.3と示される)を陰性対照として使用した。
ヒトCD8+ T細胞を単離するために、最初に末梢血単核細胞(PBMC)を、血小板成分献血の副産物である白血球除去錐体細胞から単離した。簡潔に述べると、白血球錐体細胞の内容物をPBSでフラッシュし、Ficoll(Sigma-Aldrich、1440−02)勾配上に重ね合わせた。PBMCを遠心分離によって単離し、Ficoll勾配を通過しなかった細胞を回収した。PBMCをPBSでさらに洗浄し、残っている赤血球を、製造業者の説明書に従って、10mlの1×赤血球溶解緩衝液(eBioscience、00−4300−54)の添加によって溶解させた。CD8+ T細胞を、製造業者の説明書に従って、CD8+ T細胞単離キット(ヒト)(Miltenyi Biotec、130−096−495)を用いて、溶離液中に存在するPBMCから単離した。
FS30抗体を、200nMから開始して2×最終濃度で、T細胞培地中で希釈し、架橋剤(抗ヒトCH2抗体MK1A6)を抗体試料に加えて、1:1のモル比で架橋させてから、1:3の滴定を行った。非架橋の抗体試料を、100nM及び25nMのみで試験した。100μlの希釈された抗体又は抗体/架橋剤混合物を、合計で200μlのアッセイ体積及び1倍濃度の抗体のために、プレートにおいてT細胞に加えた。アッセイを、72時間にわたって、37℃、5%のCO2でインキュベートした。上清を収集し、製造業者の説明書に従って、ヒトIL−2 ELISA Ready-SET-Go!キット(eBioscience、88−7025−88)を用いてアッセイした。プレートを、Gen5 Software、BioTekを用いて、プレートリーダーを用いて450nmで読み取った。630nmの吸光度値を、450nmの吸光度値から減算した(補正)。サイトカイン濃度の計算のための標準曲線は、4パラメータロジスティック曲線フィット(Gen5 Software、BioTek)に基づいていた。ヒトIL−2(hIL−2)の濃度を、抗体のlog濃度に対してプロットし、得られた曲線を、GraphPad Prismにおいてlog(アゴニスト)対応答の式を用いてフィッティングした。アッセイの結果は、表5及び図1に示される。
ナイーブファージライブラリーの最初のスクリーンによって同定された36のmAbから、5つの抗ヒトCD137 mAbクローン(FS30−5、FS30−10、FS30−15、FS30−16及びFS30−35)が、組み換えヒト及びカニクイザルCD137の両方に結合することが分かった。FS30−5、FS30−10、FS30−15及びFS30−16 mAbクローンは、細胞表面CD137受容体に結合して、架橋時にT細胞活性化を誘発することが示された。これらのクローンは、FS30−35 mAbクローンと一緒に、実施例3に記載されるように、mAb2フォーマットにおける発現及び配列最適化のために選択された。
mAbのCDRベースの抗原結合部位は、Fc抗原結合又は「Fcab」ドメインとして知られている、定常ドメイン中で生成されるさらなる結合部位と組み合わされて、mAb2と呼ばれる二重特異的抗体が得られる。mAb2フォーマットにおける抗CD137結合部分の特徴付けを可能にするために、FS30−5、FS30−10、FS30−15、FS30−16又はFS30−35クローンのいずれかのCDRを含み、且つCH2ドメイン中にLALA突然変異、及びCH3ドメイン中にヒトOX40受容体結合部位を含むIgG1分子からなるmAb2分子を調製した。FS20−22−49AA/HelD1.3と示される抗ヒトOX40/抗HEL mAb2のVHドメインを、FS30クローンの対応するVHドメインで置換し、生成されたVHを、FS30 mAbの対応する軽鎖と共トランスフェクトすることによって、これらのmAb2分子を生成した。IgG1分子のCH2ドメイン中のLALA突然変異は、得られたmAb2分子中に保持されていた。得られたmAb2分子の重鎖及び軽鎖配列は、配列番号83及び13、103及び46、89及び88、92及び91、93及び68に示される。これらのmAb2分子は、FS20−22−49AA/FS30−5、FS20−22−49AA/FS30−10、FS20−22−49AA/FS30−15、FS20−22−49AA/FS30−16及びFS20−22−49AA/FS30−35と示された。mAb2を、HEK293−6E細胞中で一過性発現によって産生し、MabSelect SuReプロテインAカラム(GE Healthcare)を用いて精製した。
CD137は、サイトカイン受容体の腫瘍壊死因子受容体スーパーファミリー(TNFRSF)に属する(Moran et al.,2013)。5つのmAb2分子の抗CD137 Fab結合部位の特異性を分析するために、ヒトCD137及び5つの密接に関連するヒトTNFRSFメンバー(TNFRSF1A、TNFRSF1B、GITR、NGFR及びCD40)へのmAb2の結合を、SPRを用いて試験した。目的は、1μMの濃度で密接に関連する抗原へのmAb2の結合を示さないが、1nMの濃度でCD137受容体への結合を示すことによって、1000倍の特異性を実証することであった。
FS30−5、FS30−10、FS30−16及びFS30−35抗CD137 mAbが、CD137に対する高い親和性及び特異性、並びにT細胞活性化アッセイにおける活性を示した一方、それらは、CDRループ内に1つ以上の潜在的な翻訳後修飾(PTM)部位を含有していた。結合及び活性を保持又は改善しながらこれらの部位において置換され得るアミノ酸残基を同定するために、これらのクローンをさらに操作することが決定された。同定された潜在的なPTM部位は、VH CDR3中のメチオニン残基(FS30−5中のKabat位置M100D及びM100H、FS30−10中のM97、FS30−16中のM100A、及びFS30−35中のM100F)、VH CDR2中の潜在的なアスパラギン酸塩異性化モチーフ(FS30−16中のKabat位置D54G55)及びVL CDR3中の潜在的な脱アミド化部位(FS30−16中のKabat位置Q90G91)を含んでいた。
5.1 組み換えCD137への選択されたmAb2クローンの結合
組み換えヒト、カニクイザル及びマウスCD137−mFC−Avi抗原(表1を参照)及びラットCD137−mFc抗原(R&D Systems、7968−4B−050)への、FS20−22−49AA/FS30−5−37、FS20−22−49AA/FS30−10−3、FS20−22−49AA/FS30−10−12、FS20−22−49AA/FS30−10−16及びFS20−22−49AA/FS30−35−14 mAb2クローンの結合を、Biacore 3000機器(GE Healthcare)を用いて、SPRによって測定した。IgG1フォーマット(G1/MOR7480.1;配列番号99及び101)における抗CD137 MOR7480.1 mAb及びG1AA/20H4.9を、陽性対照として使用した。
実施例4に記載される突然変異誘発作業において導入されたアミノ酸突然変異が結合特異性に影響を与えたかどうかを試験するために、選択されたmAb2クローンを、CD137受容体に密接に関連する他のTNFRSFメンバーへの結合について試験した。
CD137−CD137L相互作用は、CD137受容体の活性化のために必要とされる。アゴニスト抗CD137抗体は、リガンド相互作用を模倣することによってCD137の活性化を引き起こし得、それによって潜在的にリガンド結合をブロックし、又はリガンド結合を妨げずに受容体のクラスター化及び活性化を引き起こす。抗体がCD137Lを潜在的に模倣する場合、それは、受容体及びリガンドの相互作用をブロックし得る。MOR7480.1が、リガンド/受容体相互作用をブロックすることは、当該技術分野において公知であるが(米国特許出願公開第2012/0237498号)、20H4.9抗体は、CD137とそのリガンドとの相互作用をブロックしないことが以前に報告されている(米国特許第7288638号)。
mAb2フォーマットにおける抗ヒトCD137 mAbクローンFS30−5−37、FS30−10−3、FS30−10−12、FS30−10−16及びFS30−35−14を(抗OX40 FcabクローンFS20−22−49AAとともに)、ELISAベースの方法を用いて、CD137−CD137L相互作用をブロックするそれらの能力について試験した。IgG1フォーマット(G1/11D4;配列番号110及び111)における抗OX40 mAb 11D4(欧州特許第2242771号)を、アイソタイプ/陰性対照として使用し;抗OX40 FcabクローンFS20−22−49AA及び抗FITC抗体4420のFab領域(Bedzyk et al.,1989;Bedzyk et al.,1990)を含むmAb2 FS20−22−49AA/4420(配列番号98及び97)を、OX40結合のための陰性対照mAb2として使用し;抗CD137 mAb G1/MOR7480.1(配列番号99及び101)並びにG1/20H4.9(配列番号104及び106)を、CD137結合及びリガンドブロッキング活性のための陽性対照として使用した。
mAb2フォーマットにおける抗ヒトCD137 mAbクローンFS30−5−37、FS30−10−3、FS30−10−12及びFS30−10−16を(抗OX40 FcabクローンFS20−22−49AAとともに)、細胞ベースの方法を用いて、CD137−CD137L相互作用をブロックするそれらの能力について試験した。IgG1フォーマット(G1/11D4;配列番号110及び111)における抗OX40 mAb 11D4(欧州特許第2242771号)を、アイソタイプ/陰性対照として使用し;抗OX40 FcabクローンFS20−22−49AA及び抗FITC抗体4420のFab領域(Bedzyk et al.,1989;Bedzyk et al.,1990)を含むmAb2 FS20−22−49AA/4420(配列番号98及び97)を、OX40結合のための陰性対照mAb2として使用し;抗CD137 mAb G1/MOR7480.1(配列番号99及び101)を、CD137結合及びリガンドブロッキング活性のための陽性対照として使用した。
選択されたFS20−22−49AA/FS30−5−37、FS20−22−49AA/FS30−10−3、FS20−22−49AA/FS30−10−12、FS20−22−49AA/FS30−10−16及びFS20−22−49AA/FS30−35−14 mAb2クローンの機能活性を、実施例4に記載されるように、DO11.10−hCD137細胞を用いて、T細胞活性化アッセイにおいて試験した。IgG1フォーマット(G1/4420;配列番号96及び97)における抗FITC抗体4420を、アイソタイプ陰性対照として使用し;抗OX40 mAb G1/11D4(配列番号110及び111)及びmAb2クローンFS20−22−49AA/4420(配列番号98及び97)を、陰性対照として使用し;IgG1(G1/MOR7480.1;配列番号99及び101)及びIgG2(G2/MOR7480.1;配列番号102及び101)フォーマットの両方における抗CD137抗体MOR7480.1(IgG2フォーマットは、抗体が臨床試験において試験されているフォーマットである(Gopal et al.,2017;Tolcher et al.,2016))を、陽性対照として使用した。アッセイの前に、mAb及びmAb2分子を、抗ヒトCH2抗体、MK1A6(実施例2.4を参照)と架橋させ、1つの実験において、非架橋のmAb及びmAb2分子の活性を調べた。マウスIL−2産生を、T細胞活性化の尺度として使用した。

- CD137に結合する抗体分子であって、前記抗体分子の抗原結合部位が、
(i)配列番号30、32、38、17、19及び22のそれぞれに記載される抗体FS30−10−16;
(ii)配列番号30、32、34、17、19及び22のそれぞれに記載される抗体FS30−10−3;
(iii)配列番号30、32、36、17、19及び22のそれぞれに記載される抗体FS30−10−12;
(iv)配列番号62、64、66、17、19及び23のそれぞれに記載される抗体FS30−35−14;又は
(v)配列番号7、9、11、17、19及び21のそれぞれに記載される抗体FS30−5−37
の相補性決定領域(CDR)1から6を含み;
前記CDR配列が、ImMunoGeneTics(IMGT)番号付けスキームに従って定義される、抗体分子。 - 前記抗体分子が、
(i)配列番号54及び48のそれぞれに記載される抗体FS30−10−16;
(ii)配列番号28及び48のそれぞれに記載される抗体FS30−10−3;
(iii)配列番号44及び48のそれぞれに記載される抗体FS30−10−12;
(iv)配列番号60及び70のそれぞれに記載される抗体FS30−35−14;又は
(v)配列番号5及び15のそれぞれに記載される抗体FS30−5−37
の重鎖可変(VH)ドメイン及び軽鎖可変(VL)ドメインを含む、請求項1に記載の抗体分子。 - 前記抗体分子の前記抗原結合部位が、配列番号30、32、38、17、19及び22のそれぞれに記載される抗体FS30−10−16のCDR1〜6を含む、請求項1又は2に記載の抗体分子。
- 前記抗体分子が、配列番号54及び48のそれぞれに記載される抗体FS30−10−16のVH及びVLドメインを含む、請求項1から3のいずれか1項に記載の抗体分子。
- 前記抗体分子が、多重特異的抗体分子である、請求項1から4のいずれか1項に記載の抗体分子。
- 前記抗体分子が、前記抗体分子の定常ドメイン中に位置する第2の抗原に結合する第2の抗原結合部位を含む、請求項5に記載の抗体分子。
- 前記定常ドメインが、CH3ドメインである、請求項6に記載の抗体分子。
- 前記第2の抗原が、免疫細胞抗原、腫瘍抗原、又は病原性抗原である、請求項6又は7に記載の抗体分子。
- 前記第2の抗原結合部位が、第1の配列、第2の配列、及び/又は第3の配列を含み、前記第1の配列、第2の配列及び第3の配列がそれぞれ、前記定常ドメインのAB、CD、及びEF構造ループ中に位置する、請求項6から8のいずれか1項に記載の抗体分子。
- 前記抗体分子が、前記第2の抗原の存在下で、免疫細胞上のCD137を活性化することが可能である、請求項6から9のいずれか1項に記載の抗体分子。
- 前記免疫細胞が、T細胞である、請求項10に記載の抗体分子。
- 前記抗体分子が、1つ以上のFcγ受容体への前記抗体分子のCH2ドメインの結合を低減又は抑制するように修飾されている、請求項1から11のいずれか1項に記載の抗体分子。
- 前記抗体分子が、1つ以上のFcγ受容体に結合しない、請求項1から12のいずれか1項に記載の抗体分子。
- 請求項1から13のいずれか1項に記載の抗体分子及び生物活性分子を含むコンジュゲート。
- 請求項1から13のいずれか1項に記載の抗体分子をコードする1つ又は複数の核酸分子。
- 請求項15に記載の核酸分子を含むベクター又は一連のベクター。
- 請求項15に記載の核酸分子、又は請求項16に記載のベクターを含む組み換え宿主細胞。
- 請求項1から13のいずれか1項に記載の抗体分子を産生する方法であって、前記抗体分子の産生のための条件下で、請求項17に記載の組み換え宿主細胞を培養することを含む、方法。
- 請求項1から14のいずれか1項に記載の抗体分子又はコンジュゲートと、薬学的に許容される賦形剤とを含む医薬組成物。
- 治療法によるヒト身体の治療のための方法に使用するための、請求項1から14のいずれか1項に記載の抗体分子又はコンジュゲート。
- 個体における癌又は感染症を治療する方法に使用するための、請求項1から14のいずれか1項に記載の抗体分子又はコンジュゲート。
