Description
本発明は、口腔内の歯面及び粘膜へのフッ素イオン滞留性に優れ、長期保存後も優れたフッ素イオン滞留性を与える口腔用組成物に関する。
フッ化ナトリウム等のフッ素含有化合物は、再石灰化促進や脱灰抑制等の機能を有する薬用成分として、う蝕予防等の目的で歯磨剤組成物等の口腔用組成物に広く用いられている。フッ素含有化合物を効果的に作用させるには、フッ素イオンを長時間に亘って口腔内の粘膜や歯面に滞留させることが有効であり、使用後の口腔内をうがいして水で漱ぐなどした後でも、口腔内にフッ素イオンを多く残して留めておくことが望まれる。
フッ素イオンの滞留性を向上させる方法として、カルシウムイオンとフッ素イオンを共存させる方法が採用されているが、カルシウムイオンとフッ素イオンは反応性が高く、これらを共存させると、歯牙に対して作用する以前に不溶性物質であるフッ化カルシウムとなり、十分な効果が発揮されず、保存後の効果も十分ではなかった。
特許文献1(特表平10-511956号公報)では、カルシウムイオンとフッ素イオンを口腔への適用直前に当該2つの組成物を混合する方法が提案されているが、この方法では、フッ素イオンとして口腔内に入るため、すぐに唾液で流され十分な滞留効果が発揮されず、また、2成分混合後短時間でフッ化カルシウムが析出するため、フッ素イオンが十分に放出されず効果が発揮されなかった。
特許文献2(特開2009-137863号公報)には、ポリリン酸塩、カルシウム塩、フッ化物塩の複合体を予め調製した組成物が提案されているが、この方法では、特に歯磨製剤において、長期保存後に口腔内のフッ素イオン滞留性が低下する課題があった。
また、特許文献3(国際公開第2013/47826号)は、ピロリドンカルボン酸又はその塩等のラクタム骨格を有し、かつ酸性基を有するラクタム化合物と、フッ素化合物とを含有する口腔用組成物が、象牙質う蝕抑制効果に優れることを提案している。特許文献4(特開2017-48162号公報)では、ラクタム化合物と所定の平均粒子径を有する水不溶性カルシウム塩とを含有する、象牙質う蝕抑制効果が向上した口腔用組成物が提案されている。しかし、これらは、いずれも象牙質に対する効果に留まっており、口腔内の粘膜や歯面への滞留性に関しては未だ改善の余地があり、また、長期保存後に機能低下する傾向があった。

また、特許文献3(国際公開第2013/47826号)は、ピロリドンカルボン酸又はその塩等のラクタム骨格を有し、かつ酸性基を有するラクタム化合物と、フッ素化合物とを含有する口腔用組成物が、象牙質う蝕抑制効果に優れることを提案している。特許文献4(特開2017-48162号公報)では、ラクタム化合物と所定の平均粒子径を有する水不溶性カルシウム塩とを含有する、象牙質う蝕抑制効果が向上した口腔用組成物が提案されている。しかし、これらは、いずれも象牙質に対する効果に留まっており、口腔内の粘膜や歯面への滞留性に関しては未だ改善の余地があり、また、長期保存後に機能低下する傾向があった。
本発明は、上記事情に鑑みなされたもので、口腔内の歯面及び粘膜へのフッ素イオン滞留性に優れ、長期保存後も優れたフッ素イオン滞留性を与える口腔用組成物を提供することを目的とする。
本発明者らは、上記目的を達成するため鋭意検討を行った結果、(A)縮合リン酸又はその塩、(B)カルシウム塩、(C)フッ素含有化合物及び(D)ピロリドンカルボン酸又はその塩を組み合わせて口腔用組成物に配合すると、口腔内の歯面及び粘膜へのフッ素イオン滞留性が優れ、長期保存後においても優れたフッ素イオン滞留性を与えることができることを知見し、本発明をなすに至った。
口腔用組成物に配合されたフッ素含有化合物のフッ素が、歯面だけでなく口腔粘膜にも十分に滞留すれば口腔内滞留性がより高まると期待されたが、口腔粘膜はムチン等の唾液タンパクで覆われているためにフッ素が吸着し難い。これに対して、本発明では、歯磨剤等の口腔用組成物において、(C)成分に(A)及び(B)成分を併用して組み合わせることで、歯面(エナメル質)へのフッ素イオンの吸着に加えて、口腔粘膜を覆っているムチンへのフッ素イオンの吸着性が高まり、フッ素イオン滞留性を向上して顕著に優れた滞留性を与える作用効果が発揮され、これにより、適用後の口腔内にフッ素イオンを比較的多く残して留めておくことができた。更に、(D)成分を添加することで、長期保存後においても機能低下を防止してフッ素イオンの滞留性向上効果を保持し、特に口腔粘膜へのフッ素イオン滞留性を優れたものとすることができた。
本発明の作用効果は、(C)成分に(A)、(B)及び(D)成分を組み合わせることで得られる。後述の比較例に示すように、(A)又は(B)成分が配合されていないと、歯面及び口腔粘膜へのフッ素イオン滞留性が悪く(比較例1、2)、(A)成分が配合されていない比較例1は、長期保存後のフッ素イオン滞留性も悪く、また、(D)成分が配合されていないと、長期保存後のフッ素イオン滞留性が悪かった(比較例3)。これに対し、実施例に示すように、本発明の(A)、(B)、(C)及び(D)成分が配合された口腔用組成物は、歯面及び口腔粘膜へのフッ素イオン滞留性に優れ、かつ長期保存後のフッ素イオン滞留性に優れていた。
なお、特許文献5(特開2021-95380号公報)は、トリポリリン酸塩又はピロリン酸塩、水溶性カルシウム塩、フッ化ナトリウム、カチオン化セルロースの組み合わせによる、口腔粘膜へのフッ素イオン滞留性及び放出性の改善である。これに対して、本発明は、(A)、(B)、(C)及び(D)成分の組み合わせで、特に(D)成分が特異的に予想外の作用を奏することによって、長期保存後においても歯面及び口腔粘膜へのフッ素イオン滞留性を向上させ、優れたフッ素イオン滞留効果を与えるものである。
本発明の作用効果は、(C)成分に(A)、(B)及び(D)成分を組み合わせることで得られる。後述の比較例に示すように、(A)又は(B)成分が配合されていないと、歯面及び口腔粘膜へのフッ素イオン滞留性が悪く(比較例1、2)、(A)成分が配合されていない比較例1は、長期保存後のフッ素イオン滞留性も悪く、また、(D)成分が配合されていないと、長期保存後のフッ素イオン滞留性が悪かった(比較例3)。これに対し、実施例に示すように、本発明の(A)、(B)、(C)及び(D)成分が配合された口腔用組成物は、歯面及び口腔粘膜へのフッ素イオン滞留性に優れ、かつ長期保存後のフッ素イオン滞留性に優れていた。
なお、特許文献5(特開2021-95380号公報)は、トリポリリン酸塩又はピロリン酸塩、水溶性カルシウム塩、フッ化ナトリウム、カチオン化セルロースの組み合わせによる、口腔粘膜へのフッ素イオン滞留性及び放出性の改善である。これに対して、本発明は、(A)、(B)、(C)及び(D)成分の組み合わせで、特に(D)成分が特異的に予想外の作用を奏することによって、長期保存後においても歯面及び口腔粘膜へのフッ素イオン滞留性を向上させ、優れたフッ素イオン滞留効果を与えるものである。
従って、本発明は、下記の口腔用組成物を提供する。

本発明によれば、口腔内の歯面及び粘膜へのフッ素イオン滞留性に優れ、長期保存後も優れたフッ素イオン滞留性を与えるフッ素含有化合物含有の口腔用組成物を提供できる。本発明の口腔用組成物は、歯面へのフッ素イオン滞留性に加えて、口腔粘膜へのフッ素イオン滞留性にも優れることから、口腔内にフッ素イオンを比較的多く残して留めおくことで、(C)成分の再石灰化促進、脱灰抑制等の効果を十分に発揮させることができ、う蝕予防用として有効である。
以下、本発明につき更に詳述する。
本発明の口腔用組成物は、(A)縮合リン酸又はその塩、(B)カルシウム塩、
(C)フッ素含有化合物、及び(D)ピロリドンカルボン酸又はその塩を含有する。
本発明の口腔用組成物は、(A)縮合リン酸又はその塩、(B)カルシウム塩、
(C)フッ素含有化合物、及び(D)ピロリドンカルボン酸又はその塩を含有する。
(A)縮合リン酸又はその塩は、フッ素イオンの滞留性向上効果を奏し、また、長期保存後においてもフッ素イオンの滞留性向上効果を保持する作用を奏する。
縮合リン酸又はその塩は、水溶性縮合リン酸又はその塩(20℃の水に対する溶解性;1g/100g以上)を好ましく用いることができる。例えばピロリン酸、トリポリリン酸、テトラポリリン酸等の直鎖状のポリリン酸又はその塩、トリメタリン酸、テトラメタリン酸、ヘキサメタリン酸等の環状のポリリン酸又はその塩が挙げられる。塩としては、ナトリウム塩、カリウム塩等のアルカリ金属塩が挙げられる。これらの中でも、特に長期保存後のフッ素イオン滞留性向上の点から、ピロリン酸塩、トリポリリン酸塩、メタリン酸塩が好ましく、ピロリン酸塩、トリポリリン酸塩がより好ましい。
縮合リン酸又はその塩としては、具体的にピロリン酸カリウム、ピロリン酸ナトリウム、トリポリリン酸ナトリウム、ヘキサメタリン酸ナトリウム等が挙げられ、ピロリン酸カリウムが特に好ましい。
これらは、1種単独でも2種以上を組み合わせてもよく、また、市販品を使用できる。
縮合リン酸又はその塩は、水溶性縮合リン酸又はその塩(20℃の水に対する溶解性;1g/100g以上)を好ましく用いることができる。例えばピロリン酸、トリポリリン酸、テトラポリリン酸等の直鎖状のポリリン酸又はその塩、トリメタリン酸、テトラメタリン酸、ヘキサメタリン酸等の環状のポリリン酸又はその塩が挙げられる。塩としては、ナトリウム塩、カリウム塩等のアルカリ金属塩が挙げられる。これらの中でも、特に長期保存後のフッ素イオン滞留性向上の点から、ピロリン酸塩、トリポリリン酸塩、メタリン酸塩が好ましく、ピロリン酸塩、トリポリリン酸塩がより好ましい。
縮合リン酸又はその塩としては、具体的にピロリン酸カリウム、ピロリン酸ナトリウム、トリポリリン酸ナトリウム、ヘキサメタリン酸ナトリウム等が挙げられ、ピロリン酸カリウムが特に好ましい。
これらは、1種単独でも2種以上を組み合わせてもよく、また、市販品を使用できる。
(A)縮合リン酸又はその塩の配合量は、組成物全体の0.1~1.5%(質量%、以下同様)が好ましく、より好ましくは0.15~1.4%、更に好ましくは0.2~1.0%である。配合量が0.1%以上であると、十分にフッ素イオンの滞留性が向上し、歯面及び口腔粘膜へのフッ素イオン滞留性に優れ、かつ長期保存後も優れたフッ素イオン滞留性を付与できる。1.5%以下であると、フッ素イオン滞留性が十分に維持される。
(B)カルシウム塩は、フッ素イオンの滞留性向上効果を奏する。
カルシウム塩は、20℃の水に対する溶解性が0.5g/100g以上のものを好ましく用いることができる。例えば塩化カルシウム、グリセロリン酸カルシウム、硝酸カルシウム、酢酸カルシウム、クエン酸カルシウム、グルコン酸カルシウム、安息香酸カルシウム、ギ酸カルシウム、フマル酸カルシウム、乳酸カルシウム、酪酸カルシウム、イソ酪酸カルシウム、リンゴ酸カルシウム、マレイン酸カルシウム、プロピオン酸カルシウム、吉草酸カルシウム等が挙げられる。これらの中でも、特に長期保存後のフッ素イオン滞留性向上の点から、塩化カルシウム、グリセロリン酸カルシウムが好ましく、塩化カルシウムがより好ましい。
これらは、1種単独でも2種以上を組み合わせてもよく、また、市販品を使用できる。
カルシウム塩は、20℃の水に対する溶解性が0.5g/100g以上のものを好ましく用いることができる。例えば塩化カルシウム、グリセロリン酸カルシウム、硝酸カルシウム、酢酸カルシウム、クエン酸カルシウム、グルコン酸カルシウム、安息香酸カルシウム、ギ酸カルシウム、フマル酸カルシウム、乳酸カルシウム、酪酸カルシウム、イソ酪酸カルシウム、リンゴ酸カルシウム、マレイン酸カルシウム、プロピオン酸カルシウム、吉草酸カルシウム等が挙げられる。これらの中でも、特に長期保存後のフッ素イオン滞留性向上の点から、塩化カルシウム、グリセロリン酸カルシウムが好ましく、塩化カルシウムがより好ましい。
これらは、1種単独でも2種以上を組み合わせてもよく、また、市販品を使用できる。
(B)カルシウム塩の配合量は、組成物全体の0.1~2%が好ましく、より好ましくは0.1~1.5%、更に好ましくは0.2~1%である。配合量が0.1%以上であると、十分にフッ素イオンの滞留性が向上し、歯面及び口腔粘膜へのフッ素イオン滞留性に優れる。2%以下であると、フッ素イオン滞留性が十分に維持される。
(C)フッ素含有化合物としては、フッ化ナトリウム、モノフルオロリン酸ナトリウム、フッ化スズ等のフッ素含有化合物を使用できる。これらの中でも、フッ化ナトリウムが、特に長期保存後のフッ素イオン滞留性の点から好ましい。
これらは、1種単独でも2種以上を組み合わせてもよく、また、市販品を使用できる。

(C)フッ素含有化合物の配合量は、フッ素イオンとして、組成物全体の500~5,000ppmが好ましく、より好ましくは1,100~3,000ppmである。配合量がフッ素イオンとして500ppm以上であると、フッ素イオンの滞留性を十分に向上させることができ、5,000ppm以下であると、過剰摂取による斑状歯等の為害作用の発生を防止できる。
フッ素含有化合物の配合量は、供給するフッ素イオン量により異なり、例えばフッ化ナトリウムの場合、供給するフッ素イオンを500ppmにする場合、その配合量は組成物全体の0.11%となる。具体的にフッ化ナトリウムの配合量は、組成物全体の0.11~1.1%が好ましく、より好ましくは0.25~0.7%である。
フッ素含有化合物の配合量は、供給するフッ素イオン量により異なり、例えばフッ化ナトリウムの場合、供給するフッ素イオンを500ppmにする場合、その配合量は組成物全体の0.11%となる。具体的にフッ化ナトリウムの配合量は、組成物全体の0.11~1.1%が好ましく、より好ましくは0.25~0.7%である。
本発明では、(A)成分と(C)成分との量比を示す(A)/(C)が、モル比として好ましくは0.05~1、より好ましくは0.1~0.5である。
また、(B)成分と(C)成分との量比を示す(B)/(C)が、モル比として好ましくは0.1~2、より好ましくは0.2~1である。
(A)/(C)のモル比、(B)/(C)のモル比がそれぞれ上記範囲内であると、更に好ましい。これらモル比の範囲内で(C)成分を配合すると、歯面及び口腔粘膜へのフッ素イオン滞留性がより優れ、かつ長期保存後もフッ素イオンの滞留性向上効果がより優れる。
また、(B)成分と(C)成分との量比を示す(B)/(C)が、モル比として好ましくは0.1~2、より好ましくは0.2~1である。
(A)/(C)のモル比、(B)/(C)のモル比がそれぞれ上記範囲内であると、更に好ましい。これらモル比の範囲内で(C)成分を配合すると、歯面及び口腔粘膜へのフッ素イオン滞留性がより優れ、かつ長期保存後もフッ素イオンの滞留性向上効果がより優れる。
(D)ピロリドンカルボン酸又はその塩は、(A)及び(B)成分によるフッ素イオンの滞留性向上効果を経時においても保持し、長期保存後もフッ素イオンの滞留性向上効果を発揮させる作用を奏する。
ピロリドンカルボン酸又はその塩としては、ピロリドンカルボン酸や、そのナトリウム塩、カリウム塩等のアルカリ金属塩が挙げられる。具体的には、ピロリドンカルボン酸ナトリウム等のピロリドンカルボン酸のアルカリ金属塩を使用し得る。
ピロリドンカルボン酸又はその塩は、市販品を使用できる。例えば、ピロリドンカルボン酸ナトリウムは、味の素ヘルシーサプライ(株)製のAJIDEW(登録商標)N-50等を用いることができる。
ピロリドンカルボン酸又はその塩としては、ピロリドンカルボン酸や、そのナトリウム塩、カリウム塩等のアルカリ金属塩が挙げられる。具体的には、ピロリドンカルボン酸ナトリウム等のピロリドンカルボン酸のアルカリ金属塩を使用し得る。
ピロリドンカルボン酸又はその塩は、市販品を使用できる。例えば、ピロリドンカルボン酸ナトリウムは、味の素ヘルシーサプライ(株)製のAJIDEW(登録商標)N-50等を用いることができる。
(D)ピロリドンカルボン酸又はその塩の配合量は、組成物全体の0.1~10%が好ましく、より好ましくは0.5~5%である。配合量が0.1%以上であると、長期保存後もフッ素イオンの滞留性が十分に向上する。10%以下であると、適度な製剤粘度が十分に確保される。10%を超えると、製剤の粘度上昇によって押出し性が悪化する場合がある。
本発明の口腔用組成物は、ペースト状、ジェル状、液体状等の形態、特にペースト状、液体状の形態で、練歯磨、ジェル状歯磨、液体歯磨、潤製歯磨等の歯磨剤、洗口剤、マウスウォッシュ、マウススプレー剤等に調製して使用できる。特に歯磨剤組成物であることが好ましく、練歯磨等の歯磨剤として好適である。
調製方法は、形態や剤型に応じた常法を採用できる。歯磨剤組成物は、公知の方法で調製でき、例えば、(A)~(D)成分、水、及び任意成分を通常の方法で配合し、得ることができる。
この場合、口腔用組成物の目的、剤型等に応じて、上述した成分以外にも適宜なその他の任意成分を必要に応じて配合できる。任意成分は、例えば、界面活性剤、研磨剤、増粘剤、湿潤剤、甘味剤、香料、防腐剤、着色剤、有効成分、溶剤等が挙げられる。
以下に任意成分の具体例を示す。これら任意成分は、1種単独で又は2種以上を組み合わせて配合でき、また、各任意成分の配合量は、本発明の効果を妨げない範囲で通常量とし得る。
調製方法は、形態や剤型に応じた常法を採用できる。歯磨剤組成物は、公知の方法で調製でき、例えば、(A)~(D)成分、水、及び任意成分を通常の方法で配合し、得ることができる。
この場合、口腔用組成物の目的、剤型等に応じて、上述した成分以外にも適宜なその他の任意成分を必要に応じて配合できる。任意成分は、例えば、界面活性剤、研磨剤、増粘剤、湿潤剤、甘味剤、香料、防腐剤、着色剤、有効成分、溶剤等が挙げられる。
以下に任意成分の具体例を示す。これら任意成分は、1種単独で又は2種以上を組み合わせて配合でき、また、各任意成分の配合量は、本発明の効果を妨げない範囲で通常量とし得る。
界面活性剤:
界面活性剤は、アニオン性界面活性剤、ノニオン性界面活性剤、両性界面活性剤が挙げられ、具体的には下記(i)~(iii)に示す。
(i)アニオン性界面活性剤
ラウリル硫酸塩等のアルキル硫酸塩、アシルサルコシン塩、アシルメチルタウリン塩、アシルグルタミン酸塩、α-オレフィンスルホン酸塩(テトラデセンスルホン酸塩)が挙げられる。上記塩としては、ナトリウム塩、カリウム塩等のアルカリ金属塩が好ましい。
(ii)ノニオン性界面活性剤
ポリオキシエチレン硬化ヒマシ油、ポリオキシエチレンアルキルエーテル、ポリグリセリン脂肪酸エステル、ソルビタン脂肪酸エステル、脂肪酸モノグリセライドが挙げられる。
(iii)両性界面活性剤
ヤシ油脂肪酸アミドプロピルベタイン等の脂肪酸アミドプロピルベタイン、N-脂肪酸アシル-N-カルボキシメチル-N-ヒドロキシエチルエチレンジアミン塩が挙げられる。
界面活性剤の配合量は、組成物全体の0.001~10%が好ましく、特に0.1~5%が好ましい。
界面活性剤は、アニオン性界面活性剤、ノニオン性界面活性剤、両性界面活性剤が挙げられ、具体的には下記(i)~(iii)に示す。
(i)アニオン性界面活性剤
ラウリル硫酸塩等のアルキル硫酸塩、アシルサルコシン塩、アシルメチルタウリン塩、アシルグルタミン酸塩、α-オレフィンスルホン酸塩(テトラデセンスルホン酸塩)が挙げられる。上記塩としては、ナトリウム塩、カリウム塩等のアルカリ金属塩が好ましい。
(ii)ノニオン性界面活性剤
ポリオキシエチレン硬化ヒマシ油、ポリオキシエチレンアルキルエーテル、ポリグリセリン脂肪酸エステル、ソルビタン脂肪酸エステル、脂肪酸モノグリセライドが挙げられる。
(iii)両性界面活性剤
ヤシ油脂肪酸アミドプロピルベタイン等の脂肪酸アミドプロピルベタイン、N-脂肪酸アシル-N-カルボキシメチル-N-ヒドロキシエチルエチレンジアミン塩が挙げられる。
界面活性剤の配合量は、組成物全体の0.001~10%が好ましく、特に0.1~5%が好ましい。
研磨剤:
無水ケイ酸等のシリカ系研磨剤、ゼオライト、炭酸カルシウム、水酸化アルミニウム等が挙げられる。研磨剤の配合量は、通常、組成物全体の2~50%、特に10~40%である。

増粘剤:
キサンタンガム、アルギン酸ナトリウム、ヒドロキシエチルセルロース、カラギーナン、カルボキシメチルセルロースナトリウム、ポリビニルピロリドン、ポリアクリル酸ナトリウム、カチオン化セルロース、カルボキシビニルポリマー等の有機増粘剤、増粘性シリカ等の無機増粘剤が挙げられる。増粘剤の配合量は、通常、組成物全体の0.1~10%、特に0.1~8%である。
キサンタンガム、アルギン酸ナトリウム、ヒドロキシエチルセルロース、カラギーナン、カルボキシメチルセルロースナトリウム、ポリビニルピロリドン、ポリアクリル酸ナトリウム、カチオン化セルロース、カルボキシビニルポリマー等の有機増粘剤、増粘性シリカ等の無機増粘剤が挙げられる。増粘剤の配合量は、通常、組成物全体の0.1~10%、特に0.1~8%である。
湿潤剤:
ソルビトール、キシリトール、エリスリトール等の糖アルコール、グリセリン、プロピレングリコール、ポリエチレングリコール(PEG)等の多価アルコールが挙げられる。湿潤剤の配合量は、通常、組成物全体の2~50%である。

メントール、アネトール、カルボン、オイゲノール、リモネン、n-デシルアルコール、シトロネロール、α-テルピネオール、シトロネリルアセテート、シネオール、リナロール、エチルリナロール、ワニリン、チモール、スペアミント油、ペパーミント油、レモン油、オレンジ油、セージ油、ローズマリー油、桂皮油、ピメント油、桂葉油、シソ油、冬緑油、丁子油、ユーカリ油が挙げられ、これらは1種単独で又は2種以上を組み合わせて使用することができる。香料の配合量は、通常、組成物全体の0.01~2%である。
防腐剤:
パラオキシ安息香酸エステル、安息香酸ナトリウムが挙げられる。
着色剤:
青色1号、青色2号、緑色3号、黄色4号、黄色5号、赤色106号、赤色227号、カラメル、酸化チタン、雲母チタンが挙げられる。
パラオキシ安息香酸エステル、安息香酸ナトリウムが挙げられる。
着色剤:

デキストラナーゼ等の酵素、アラントイン、グリチルリチン酸塩、グリチルレチン酸、塩化ナトリウム、イソプロピルメチルフェノール、グルコン酸銅、塩化セチルピリジニウム、塩化ベンザルコニウム、塩化ベンゼトニウム、ヒノキチオール、ゼオライト、乳酸アルミニウム、硝酸カリウム、オウバクエキスが挙げられる。これら有効成分の配合量は、本発明の効果を妨げない範囲で有効量とし得る。
デキストラナーゼ等の酵素、アラントイン、グリチルリチン酸塩、グリチルレチン酸、塩化ナトリウム、イソプロピルメチルフェノール、グルコン酸銅、塩化セチルピリジニウム、塩化ベンザルコニウム、塩化ベンゼトニウム、ヒノキチオール、ゼオライト、乳酸アルミニウム、硝酸カリウム、オウバクエキスが挙げられる。これら有効成分の配合量は、本発明の効果を妨げない範囲で有効量とし得る。
溶剤:
精製水、エタノール等の炭素数2又は3の低級1価アルコールが挙げられる。なお、エタノールは、配合する場合は組成物全体の25%以下でもよく、また、配合しなくてもよい。

表1~4に示す組成の歯磨剤組成物(練歯磨)を通常の方法で調製し、下記方法で評価した。結果を表に併記した。

(1)歯面へのフッ素イオン滞留性の評価方法
歯磨剤組成物を精製水で4倍希釈した液をヒドロキシアパタイト(HA)ディスク(直径7mm、厚さ3.5mm、PENTAX社製)に3分間作用させ、直ちに精製水で洗浄した。HAディスクを乾燥させた後、HAディスク上面を0.1NのHCl 120μLで2分間脱灰させ、フッ素イオンを抽出し、溶液中に含まれるフッ素イオンをイオンメーター(Orion 1115000 4-Star、サーモフィッシャーサイエンティフィック(株)製)で測定し、フッ素イオン濃度(ppm)を求め、下記基準に基づき評価した。
歯面へのフッ素イオン滞留性の評価基準
◎:フッ素イオン濃度が2.0ppm以上
〇:フッ素イオン濃度が1.5ppm以上2.0ppm未満
△:フッ素イオン濃度が1.0ppm以上1.5ppm未満
×:フッ素イオン濃度が1.0ppm未満

◎:フッ素イオン濃度が2.0ppm以上
〇:フッ素イオン濃度が1.5ppm以上2.0ppm未満
△:フッ素イオン濃度が1.0ppm以上1.5ppm未満
×:フッ素イオン濃度が1.0ppm未満
(2)口腔粘膜へのフッ素イオン滞留性の評価方法
10mL遠心チューブにムチン(Sigma-Aldrich製)を0.2g添加し、更に、歯磨剤組成物(精製水による4倍希釈液)を10mL加え、3分間作用させた。その後、10,000rpmで10分間遠心分離し、上清を除去した。ムチンにクエン酸カリウム緩衝液を10mL加えて1分間撹拌し、ムチンに吸着・滞留したフッ素イオンを溶出させた。10,000rpmで10分間遠心分離し、上清を回収し、溶液に含まれるフッ素イオン濃度(ppm)をフッ素イオンメーター(Orion 1115000 4-Star、サーモフィッシャーサイエンティフィック(株)製)で測定し、口腔粘膜へのフッ素イオン滞留性を下記基準に基づき評価した。
口腔粘膜へのフッ素イオン滞留性の評価基準
◎:フッ素イオン濃度が100ppm以上
〇:フッ素イオン濃度が60ppm以上100ppm未満
△:フッ素イオン濃度が30ppm以上60ppm未満
×:フッ素イオン濃度が30ppm未満
10mL遠心チューブにムチン(Sigma-Aldrich製)を0.2g添加し、更に、歯磨剤組成物(精製水による4倍希釈液)を10mL加え、3分間作用させた。その後、10,000rpmで10分間遠心分離し、上清を除去した。ムチンにクエン酸カリウム緩衝液を10mL加えて1分間撹拌し、ムチンに吸着・滞留したフッ素イオンを溶出させた。10,000rpmで10分間遠心分離し、上清を回収し、溶液に含まれるフッ素イオン濃度(ppm)をフッ素イオンメーター(Orion 1115000 4-Star、サーモフィッシャーサイエンティフィック(株)製)で測定し、口腔粘膜へのフッ素イオン滞留性を下記基準に基づき評価した。

◎:フッ素イオン濃度が100ppm以上
〇:フッ素イオン濃度が60ppm以上100ppm未満
△:フッ素イオン濃度が30ppm以上60ppm未満
×:フッ素イオン濃度が30ppm未満
(3)長期保存後のフッ素イオン滞留性の評価方法
歯磨剤組成物を50gずつチューブ容器(素材:最内層が直鎖状低密度ポリエチレンからなる直径26mmのラミネートチューブ、大日本印刷(株)製)3本に詰め、40℃で1ヶ月間保存した。保存後に常温に戻した歯磨剤組成物について、上記(2)と同じ方法で口腔粘膜へのフッ素イオン滞留性を評価し、長期保存品のフッ素イオン濃度(滞留フッ素イオン濃度(ppm))を測定して求めた。
上記(2)の方法で求めた未保存の歯磨剤組成物のフッ素イオン濃度を、初期品の滞留フッ素イオン濃度(ppm)とし、フッ素イオン滞留率(%)を下記式により算出し、長期保存後のフッ素イオン滞留性を下記基準に基づき評価した。

長期保存後のフッ素イオン滞留性の評価基準
◎:フッ素イオン滞留率が80%以上
〇:フッ素イオン滞留率が50%以上80%未満
△:フッ素イオン滞留率が20%以上50%未満
×:フッ素イオン滞留率が20%未満
歯磨剤組成物を50gずつチューブ容器(素材:最内層が直鎖状低密度ポリエチレンからなる直径26mmのラミネートチューブ、大日本印刷(株)製)3本に詰め、40℃で1ヶ月間保存した。保存後に常温に戻した歯磨剤組成物について、上記(2)と同じ方法で口腔粘膜へのフッ素イオン滞留性を評価し、長期保存品のフッ素イオン濃度(滞留フッ素イオン濃度(ppm))を測定して求めた。
上記(2)の方法で求めた未保存の歯磨剤組成物のフッ素イオン濃度を、初期品の滞留フッ素イオン濃度(ppm)とし、フッ素イオン滞留率(%)を下記式により算出し、長期保存後のフッ素イオン滞留性を下記基準に基づき評価した。

Claims (9)
- (A)縮合リン酸又はその塩、
(B)カルシウム塩、
(C)フッ素含有化合物
及び
(D)ピロリドンカルボン酸又はその塩
を含有する口腔用組成物。 - (A)縮合リン酸又はその塩が、ピロリン酸塩、トリポリリン酸塩及びメタリン酸塩から選ばれる1種以上である請求項1記載の口腔用組成物。
- (A)縮合リン酸又はその塩が、ピロリン酸カリウムである請求項2記載の口腔用組成物。
- (B)カルシウム塩が、塩化カルシウム及びグリセロリン酸カルシウムから選ばれる1種以上である請求項1記載の口腔用組成物。
- (C)フッ素含有化合物が、フッ化ナトリウム、モノフルオロリン酸ナトリウム及びフッ化スズから選ばれる1種以上である請求項1記載の口腔用組成物。
- (A)/(C)がモル比として0.05~1、(B)/(C)がモル比として0.1~2である請求項1記載の口腔用組成物。
- (C)成分の含有量が、フッ素イオンとして500~5,000ppmである請求項1記載の口腔用組成物。
- (A)成分の含有量が0.1~1.5質量%、(B)成分の含有量が0.1~2質量%、(D)成分の含有量が0.1~10質量%である請求項1記載の口腔用組成物。
- 歯磨剤組成物である請求項1~8のいずれか1項記載の口腔用組成物。
