Abstract
本発明は、ヒト皮膚の老化防止(皮膚のハリや弾力保持)、紫外線または活性酸素種発生による細胞障害から細胞を保護する新規のスペルミジン産生促進剤、スペルミジンの産生を促進する皮膚外用剤、抗紫外線(UV)剤または抗酸化剤に関する。具体的には、マグネシウム化合物を有効成分として用いることで、スペルミジン産生促進剤、スペルミジンの産生を促進する皮膚外用剤、抗紫外線(UV)剤または抗酸化剤を調製することができる。
Description
本発明はスペルミジン産生促進剤およびそれを用いる化粧、美容または非治療的方法に関する。
皮膚は、生体の最外層に存在することから、常に外界からの刺激にさらされている。皮膚老化の原因は、加齢に加えて、外界からの刺激である、乾燥、酸化、太陽光(紫外線)等による影響も皮膚老化に関わる直接的な因子として挙げられる。これらの刺激は、肌荒れを引き起こす原因になる。
ポリアミンは、第一級アミノ基を2つ以上もつ直鎖状の生体内脂肪族炭化水素である。ポリアミンは、動物・植物・微生物まで、あらゆる生体中に含まれ、細胞分裂や蛋白合成などの活動に関与している成長因子である。代表的なポリアミンとして、プトレシン、スペルミジン及びスペルミンを挙げることができる。ポリアミンの生理作用としては、(1)細胞増殖作用、(2)細胞分化促進作用、(3)免疫必須因子、(4)抗アレルギー作用、(5)蛋白質合成促進作用、(6)核酸との相互作用による構造の安定化、(7)酵素活性調節作用等が知られている。加齢によって、体内のポリアミンが減少し、老化に伴うポリアミンの低下がヒトの健康障害と並行して起こることが知られており、ポリアミンの植物およびヒトの健康と疾病における多様な機能に関する報告がされている(非特許文献1)。
納豆や大豆などに含まれるポリアミンがアンチエイジング効果物質として注目され、細胞を活性化できる機能性食品として食品分野への応用、また、ヒトの皮膚組織にもポリアミンが存在することから、化粧品機能性原料としての応用も期待される。
一方、マグネシウム塩とカルシウム塩とからなる皮膚バリア機能回復促進剤が皮膚バリア機能を極めて短時間のうちに回復させることで、皮膚の表皮機能の低下による表皮増殖性異常等を防止することは報告されている(特許文献1)。しかし、マグネシウム化合物が、ヒト細胞においてポリアミンの一種であるスペルミジンを増加させる作用を有することは、これまで知られていなかった。

本発明は上記事情に鑑みてなされたもので、スペルミジン産生能を促進させてスペルミジン量を増加させるスペルミジン産生促進剤を提供することを目的とする。さらに本発明は、スペルミジンの産生を促進することで、外部刺激である紫外線、活性酸素種による細胞障害から細胞を保護する、抗紫外線(UV)剤および抗酸化剤を提供することを目的とする。
本発明者らは、ヒトケラチノサイト由来のHaCaT細胞を用いて、細胞内ポリアミンの合成に関わる因子のマグネシウム化合物の効果を検討したところ、マグネシウム化合物に優れたスペルミジン産生促進作用を有することをはじめて見出した。そして、マグネシウム化合物を有効成分として含有することを特徴とするスペルミジン産生促進剤を発明した。さらにマグネシウム化合物が紫外線および活性酸素種による細胞障害から有効に細胞を保護することを見出し、マグネシウム化合物を含有する抗紫外線(UV)剤および抗酸化剤を発明した。
そこで、本発明は以下の発明に関する:

マグネシウム化合物を用いて細胞内のスペルミジンの産生を促進することができる。さらには、スペルミジンの産生を促進することで、本発明のスペルミジン産生促進剤は、ヒト皮膚の老化、とくに外部刺激による細胞障害から細胞を有効に保護することができる。
本発明の一つの態様において、本発明は、マグネシウム化合物を有効成分として含有することを特徴とする、スペルミジン産生促進剤に関する。
スペルミジンは、図1に示すようにオルニチンからプトレシンが作られ、そこへアミノプロピル基が付加され、スペルミジン、スペルミンが合成される。その合成には、プトレシンの他、S-アデノシルメチオン(SAM)が必要であり、SAMはメチル基の供与体でもある。SAMは、アデノシルメチオニンデカルボキシラーゼ1(AMD1)の作用を受けて脱炭酸アデノシルメチオニン(Decarboxylated S-adenosylmethionine:dc-SAM)になり、プトレシンからスペルミジンおよびスペルミンが合成される際にはスペルミジンシンターゼ(SRM)の作用を受けて、dc-SAMからアミノプロピル基が提供される。
本発明者らは、ヒトケラチノサイト由来のHaCaT細胞を用いて、細胞内ポリアミンの合成に関わる因子のマグネシウム化合物の効果を検討したところ、アデノシルメチオニンデカルボキシラーゼ1(AMD1)およびスペルミジンシンターゼ(SRM)の発現量が促進されることを見出した。したがって、本発明では、スペルミジンの合成に関与するアデノシルメチオニンデカルボキシラーゼ1(AMD1)およびスペルミジン合成酵素であるスペルミジンシンターゼ(SRM)の発現量を指標としてスペルミジンの産生量を評価した。
さらに本発明者らは、マグネシウム化合物が、外部刺激である紫外線、活性酸素種に対する皮膚障害から皮膚のバリア機能の増進および細胞を保護する効果を有することに着目した。かかる細胞保護の効果にはマグネシウム化合物のスペルミジン産生促進作用が大きく関与していることが示唆された。
その作用機序について、図2に示す。マグネシウム流入チャネルとして機能する、細胞表面のNIPAL2とNIPAL4を介して、マグネシウム化合物が細胞内に取り込まれることにより、細胞内のAMD1およびSRMの発現およびスペルミジンの産生が促進され、さらにスペルミジンの産生が外的刺激による細胞障害からの保護に有効であると考えられる。
本明細書において、スペルミジン産生促進剤とは、細胞のスペルミジンの産生および/またはスペルミジンの産生能を促進する効果を有することを意味する。
スペルミジンの産生および/またはスペルミジンの産生能を促進する効果は、細胞をスペルミジン産生促進剤を含む培養液で培養した後の細胞内のスペルミジンの産生量またはアデノシルメチオニンデカルボキシラーゼ1(AMD1)またはスペルミジンシンターゼ(SRM)の発現量を測定することによって評価することができる。
スペルミジンの産生および/またはスペルミジンの産生能を促進する効果は、細胞をスペルミジン産生促進剤を含む培養液で培養した後の細胞内のスペルミジンの産生量またはアデノシルメチオニンデカルボキシラーゼ1(AMD1)またはスペルミジンシンターゼ(SRM)の発現量を測定することによって評価することができる。
本発明のスペルミジン産生促進剤に用いられるマグネシウム化合物としては、ハロゲン化物及び複ハロゲン化物、酸化物及び酸化物の関連化合物、硫化物、窒化物、炭化物、酸素酸塩、有機マグネシウム化合物等のマグネシウム含有化合物が挙げられる。特に好ましくは、塩化マグネシウム、乳酸マグネシウムまたは硫酸マグネシウムである。
本発明のスペルミジン産生促進剤は、たとえば、表皮細胞、皮膚繊維芽細胞および/またはケラチノサイトにおけるスペルミジンの産生を促進するために用いられるが、これらに限定されない。本発明では、特に、表皮細胞および/またはケラチノサイトにおいて効果を有する。
本発明の一つの態様は、マグネシウム化合物を有効成分として含有することを特徴とする、スペルミジンの産生を促進する皮膚外用剤に関する。
本発明のスペルミジン産生促進剤は、マグネシウム化合物を用いることで、スペルミジンの産生を促進し、ヒト皮膚の老化防止(皮膚のハリや弾力保持)、紫外線などの外部刺激、活性酸素種による細胞障害から細胞を保護するための治療等に有効に適用され得る。よって、スペルミジンの産生を促進する皮膚外用剤(医薬品、医薬部外品、化粧料を含む)および/またはスペルミジンの異常分解またはスペルミジン産生能の低下による疾病を治療するための治療剤が提供される。
本発明のスペルミジン産生促進剤は、マグネシウム化合物を用いることで、スペルミジンの産生を促進し、ヒト皮膚の老化防止(皮膚のハリや弾力保持)、紫外線などの外部刺激、活性酸素種による細胞障害から細胞を保護するための治療等に有効に適用され得る。よって、スペルミジンの産生を促進する皮膚外用剤(医薬品、医薬部外品、化粧料を含む)および/またはスペルミジンの異常分解またはスペルミジン産生能の低下による疾病を治療するための治療剤が提供される。
また、本発明の一つの態様は、マグネシウム化合物を有効成分として含有することを特徴とする、スペルミジンの異常分解またはスペルミジン産生能の低下による疾病を治療するための治療剤に関する。疾病には、例えば、肝炎、歯肉炎、リウマチ、変形関節炎、悪性腫瘍、肝硬変、移植拒否、強皮症、肝硬変、動脈硬化、線維症、乾皮症、乾燥肌、荒れ肌、腎炎、ケロイド、過修復、敗血症等が含まれるが、これらに限定はされない。さらに、本発明の一つの態様は、対象におけるスペルミジンの異常分解またはスペルミジン産生能の低下による疾病を治療するための方法であって、マグネシウム化合物を有効成分として含有する組成物を対象に投与することを含む方法に関する。対象はヒトを含む哺乳動物、または非ヒト哺乳動物でありうるが、好ましくはヒトである。
本発明の一つの態様は、マグネシウム化合物を有効成分として含有することを特徴とする、抗紫外線(UV)剤に関する。
マグネシウム化合物は、外部刺激、とくに紫外線に対する皮膚のバリア機能の増進および細胞の保護作用効果を有するため、抗紫外線(UV)剤としても有効に用いることができる。とくにマグネシウム化合物がスペルミジン産生促進に大きく関与していることが考えられ、本願発明の抗紫外線(UV)剤は、スペルミジン産生促進効果を有する抗紫外線(UV)剤として好適に用いることができる。
マグネシウム化合物は、外部刺激、とくに紫外線に対する皮膚のバリア機能の増進および細胞の保護作用効果を有するため、抗紫外線(UV)剤としても有効に用いることができる。とくにマグネシウム化合物がスペルミジン産生促進に大きく関与していることが考えられ、本願発明の抗紫外線(UV)剤は、スペルミジン産生促進効果を有する抗紫外線(UV)剤として好適に用いることができる。
本発明の一つの態様は、マグネシウム化合物を有効成分として含有し、スペルミジンの産生を促進することにより、活性酸素種発生による細胞障害から細胞を保護する抗酸化剤に関する。
マグネシウム化合物は、スペルミジン産生促進作用を有することから、本願発明の抗酸化剤は、スペルミジン産生促進効果を有する抗酸化剤として好適に用いることができる。

本発明のマグネシウム化合物を皮膚外用剤、抗紫外線(UV)剤または抗酸化剤を組成物に配合する場合、マグネシウム化合物は、外用剤(組成物)全量中、たとえば、マグネシウムイオン相当量で、1μM~50mMが好ましく、1mM~20mMがより好ましい。マグネシウム化合物の重量%では、たとえば、塩化マグネシウム六水和物の場合、0.00002~1重量%が好ましく、0.02~0.4重量%がより好ましい。
本発明のスペルミジン産生促進剤、抗紫外線(UV)剤または抗酸化剤(組成物)は、皮膚外用剤、として適用する場合、上記成分に加えて、さらに必要により、本発明の効果を損なわない範囲内で、通常化粧品や医薬品等の皮膚外用剤に用いられる成分、例えば酸化防止剤、油分、紫外線防御剤、界面活性剤、増粘剤、アルコール類、粉末成分、色材、水性成分、水、各種皮膚栄養剤等を必要に応じて適宜配合することができる。
さらに、エデト酸二ナトリウム、エデト酸三ナトリウム、クエン酸ナトリウム、ポリリン酸ナトリウム、メタリン酸ナトリウム、グルコン酸等の金属イオン封鎖剤、メチルパラベン、エチルパラベン、ブチルパラベン等の防腐剤、カフェイン、タンニン、ベラパミル、トラネキサム酸およびその誘導体、甘草抽出物、グラブリジン、カリンの果実の熱水抽出物、各種生薬、酢酸トコフェロール、グリチルリチン酸およびその誘導体またはその塩等の薬剤、ビタミンC、アスコルビン酸リン酸マグネシウム、アスコルビン酸グルコシド、アルブチン、コウジ酸等の美白剤、グルコース、フルクトース、マンノース、ショ糖、トレハロース等の糖類なども適宜配合することができる。
皮膚外用剤は、外皮に適用される化粧料、医薬部外品等、特に好適には化粧料に広く適用することが可能であり、その剤型も、皮膚に適用できるものであればいずれでもよく、溶液系、可溶化系、乳化系、粉末分散系、水-油二層系、水-油-粉末三層系、軟膏、化粧水、ゲル、エアゾール等、任意の剤型が適用される。
使用形態も任意であり、例えば化粧水、乳液、クリーム、パック等のフェーシャル化粧料やファンデーション、口紅、アイシャドウ等のメーキャップ化粧料、芳香化粧料、浴用剤等に用いることができる。
また、メーキャップ化粧品であれば、ファンデーション等、トイレタリー製品としてはボディーソープ、石けん等の形態に広く適用可能である。さらに、医薬部外品であれば、各種の軟膏剤等の形態に広く適用が可能である。そして、これらの剤型および形態に、本発明のスペルミジン産生促進剤の採り得る形態が限定されるものではない。
本発明のスペルミジン産生促進剤を飲食品や飼料等に配合する場合、マグネシウム化合物の配合量は、それらの種類、目的、形態、利用方法などに応じて、適宜決めることができ、例えば、飲食品全量中に錠剤やカプセル剤等の場合は1~90質量%、その他の飲食品では0.001~50質量%の量で含むことができる。成人一日当たりマグネシウム化合物の摂取量が約1~370mg程度になるように調製することが好ましい。特に、保健用飲食品等として利用する場合には、本発明の有効成分を所定の効果が十分発揮されるような量で含有させることが好ましい。
飲食品や飼料の形態としては、例えば、顆粒状、粒状、ペースト状、ゲル状、固形状、または、液体状に任意に成形することができる。これらには、飲食品等に含有することが認められている公知の各種物質、例えば、結合剤、崩壊剤、増粘剤、分散剤、再吸収促進剤、矯味剤、緩衝剤、界面活性剤、溶解補助剤、保存剤、乳化剤、等張化剤、安定化剤やpH調製剤などの賦形剤を適宜含有させることができる。
本発明のスペルミジン産生促進剤、抗紫外線(UV)剤または抗酸化剤を医薬製剤として用いる場合、該製剤は経口的にあるいは非経口的(静脈投与、腹腔内投与、等)に適宜に使用される。剤型も任意で、例えば錠剤、顆粒剤、散剤、カプセル剤等の経口用固形製剤や、内服液剤、シロップ剤等の経口用液体製剤、または、注射剤などの非経口用液体製剤など、いずれの形態にも公知の方法により適宜調製することができる。これらの医薬製剤には、通常用いられる結合剤、崩壊剤、増粘剤、分散剤、再吸収促進剤、矯味剤、緩衝剤、界面活性剤、溶解補助剤、保存剤、乳化剤、等張化剤、安定化剤やpH調製剤などの賦形剤を適宜使用してもよい。
本発明のスペルミジン産生促進剤、抗紫外線(UV)剤または抗酸化剤を、皮膚外用剤、飲食品、飼料、医薬製剤等として用いる場合、タンパク質非酵素的糖化作用が関与する種々の症状や疾病、病態等の治療、予防、改善等に役立つ。具体的適用例としては、例えば、ヒト皮膚の老化防止(皮膚のハリや弾力保持)、紫外線および活性酸素種による細胞障害から細胞を保護する治療等に好適に用いられる。また上記症状や疾病、病態等の治療、予防、改善等の生理機能をコンセプトとして、その旨を表示した皮膚外用剤、機能性飲食品、疾病者用食品、特定保健用食品等に応用することができる。
本発明の一つの態様は、マグネシウム化合物を有効成分として含有するスペルミジン産生促進剤を適用することを含む、美容方法または非治療的方法に関する。
本明細書に開示のスペルミジン産生促進剤、皮膚外用剤、抗紫外線(UV)剤または抗酸化剤は、ヒト皮膚の老化防止(皮膚のハリや弾力保持)、紫外線および活性酸素種による細胞障害から細胞を保護する治療等のための美容又は非治療的方法において用いることができる。本発明にかかる美容又は非治療的方法においては、上記皮膚外用剤組成物を所望の効果を得たい部位、例えば顔、首、手、腕、脚等に適用する。適用は、手や塗布具等によってする塗布など、任意の様式で行うことができる。適用の回数、頻度、量等は、所望の効果等に応じて適宜設定される。なお、本発明にかかる美容又は非治療的方法は、いわゆる医療行為を含まない。
さらに、本発明の一つの態様は、スペルミジンの産生を促進するためのマグネシウム化合物の使用に関する。本明細書に開示のデータは、マグネシウム化合物が優れたスペルミジン産生能を有することを実証している。よって、マグネシウム化合物を用いて、スペルミジン産生促進剤、スペルミジンの産生を促進する皮膚外用剤、スペルミジン産生能の低下による疾病を治療する治療剤を調製することができる。すなわち、本発明の一つの態様は、これら皮膚外用剤および治療剤の製造におけるマグネシウム化合物の使用にも関する。
さらに、本発明の一つの態様は、紫外線または活性酸素種発生による細胞障害から細胞を保護する皮膚外用剤の製造におけるマグネシウム化合物の使用に関する。本明細書に開示のデータは、紫外線または活性酸素種発生による細胞障害から細胞を保護することを実証している。よって、マグネシウム化合物を用いて、紫外線または活性酸素種発生による細胞障害から細胞を保護する皮膚外用剤を調製することができる。
さらに、本発明の一つの態様は、スペルミジンの産生を促進する化合物のスクリーニング方法であって、
細胞と該化合物を接触させる工程、
細胞内マグネシウム濃度を測定する工程、
該化合物の有無による細胞内マグネシウム濃度の増加の有無を決定する工程
を含む方法に関する。
スペルミジンの産生能は、細胞内マグネシウム化合物濃度を測定することにより測定することができる。具体的には、マグネシウムイオン濃度が増加すると、スペルミジン濃度が増大することから、スペルミジンの産生を促進する化合物のスクリーニングにおいて、候補化合物の有無による細胞内マグネシウム濃度の増加の有無を決定することにより、候補化合物の選択を行うことができる。
マグネシウム濃度は、細胞を候補化合物を含む培養液で培養した後の細胞内のマグネシウム濃度を測定することによって評価する。

を含む方法に関する。
スペルミジンの産生能は、細胞内マグネシウム化合物濃度を測定することにより測定することができる。具体的には、マグネシウムイオン濃度が増加すると、スペルミジン濃度が増大することから、スペルミジンの産生を促進する化合物のスクリーニングにおいて、候補化合物の有無による細胞内マグネシウム濃度の増加の有無を決定することにより、候補化合物の選択を行うことができる。
マグネシウム濃度は、細胞を候補化合物を含む培養液で培養した後の細胞内のマグネシウム濃度を測定することによって評価する。
スクリーニングの対象となる化合物は、例えば、化合物ライブラリーや、微生物・動植物等の抽出物のライブラリーに含まれるものが使用されうる。細胞内マグネシウム濃度の測定は、例えば、マグネシウム蛍光プローブなどを用いた蛍光測定法により行うことができる。マグネシウム蛍光プローブとしては、例えば、富士フイルム和光純薬(株)から購入可能なKMG-20-AMや、Thermo Fisher Scientificから購入可能なMag-Fura-2、Magnesium Green(商標)などが用いられうるが、これらに限定はされない。
さらに、本発明の一つの態様は、スペルミジンの産生量又はスペルミジンシンターゼ若しくはアデノシルメチオニンデカルボキシラーゼ1の発現量の変化を測定する工程をさらに含む、上記スクリーニング方法に関する。上記スクリーニング方法において、スペルミジンの産生量又はスペルミジンシンターゼ若しくはアデノシルメチオニンデカルボキシラーゼ1の発現量の変化を測定することにより、候補化合物の選択を行うことができる。
スペルミジンの産生量又はスペルミジンシンターゼ若しくはアデノシルメチオニンデカルボキシラーゼ1の発現量の変化は、例えば、細胞外のスペルミジン又はスペルミジンシンターゼ若しくはアデノシルメチオニンデカルボキシラーゼ1の発現量をELISAなどの公知の手法を用いて測定することで決定することができる。
本明細書において言及される全ての文献はその全体が引用により本明細書に取り込まれる。
以下に説明する本発明の実施例は例示のみを目的とし、本発明の技術的範囲を限定するものではない。本発明の技術的範囲は特許請求の範囲の記載によってのみ限定される。本発明の趣旨を逸脱しないことを条件として、本発明の変更、例えば、本発明の構成要件の追加、削除及び置換を行うことができる。
以下、本発明を実施例に基づきさらに詳細に説明するが、本発明は以下の実施例に限定されるものではない。
実施例1:マグネシウム化合物によるSRMとAMD1の発現量の同定
HaCaT細胞を35mmdishに播種後、4日間培養した。0.8mMのMgSO4を含有するDMEMに、0、2.5、5、10mMのMgCl2を添加後、24時間培養し、細胞を回収した。結果を図3に示す。MgCl2添加によりSRMとAMD1のタンパク質量の濃度依存的な増加が見られた。
HaCaT細胞を35mmdishに播種後、4日間培養した。0.8mMのMgSO4を含有するDMEMに、0、2.5、5、10mMのMgCl2を添加後、24時間培養し、細胞を回収した。結果を図3に示す。MgCl2添加によりSRMとAMD1のタンパク質量の濃度依存的な増加が見られた。
実施例2:SRM、AMD1mRNA量に対するマグネシウム化合物の影響
HaCaT細胞を4日間培養後、5mM塩化マグネシウム(MgCl2)、乳酸マグネシウム(Mglac)硫酸マグネシウム(MgSO4)を添加した培地で6時間培養した。RNAを抽出後、リアルタイムPCR法でSRMとAMD1のmRNA量を測定した。その結果を図4に示す。塩化マグネシウム(MgCl2)、乳酸マグネシウム(Mg-lac)、硫酸マグネシウム(MgSO4)のいずれでもSRM、AMD1の発現が促進された。
HaCaT細胞を4日間培養後、5mM塩化マグネシウム(MgCl2)、乳酸マグネシウム(Mglac)硫酸マグネシウム(MgSO4)を添加した培地で6時間培養した。RNAを抽出後、リアルタイムPCR法でSRMとAMD1のmRNA量を測定した。その結果を図4に示す。塩化マグネシウム(MgCl2)、乳酸マグネシウム(Mg-lac)、硫酸マグネシウム(MgSO4)のいずれでもSRM、AMD1の発現が促進された。
実施例3:UV照射による細胞障害に対するマグネシウム化合物の細胞保護作用
HaCaT細胞を4日間培養後、5、10mM塩化マグネシウムを添加した培地で24時間培養した。100、150mJ/cm2UVBを照射後、24時間培養した。WST-1試薬を用いて、細胞生存率を算出した。結果を図5に示す。UV照射で細胞の生存率は減少するが、MgCl2添加して培養すると生存率低下を有意に抑えることができた。
HaCaT細胞を4日間培養後、5、10mM塩化マグネシウムを添加した培地で24時間培養した。100、150mJ/cm2UVBを照射後、24時間培養した。WST-1試薬を用いて、細胞生存率を算出した。結果を図5に示す。UV照射で細胞の生存率は減少するが、MgCl2添加して培養すると生存率低下を有意に抑えることができた。
実施例4:UV照射による活性酸素種(ROS)産生量に対するマグネシウム化合物の細胞保護作用
HaCaT細胞を96well plateに播種後、4日間培養した。10mMのMgCl2(10Mg)、10mMのMgCl2+10μMシステアミン(10Mg+Cys)で24時間処理後、100mJUVBを照射した。100μMまたは200μMスペルミジン(SPR)、2mM N-アセチル-システイン(NAC)を、UVB照射の30分前に培地に添加した。H2DCFDAを負荷後、ROS産生量を蛍光強度で算出した。結果を図6に示す。UVB照射によりROS産生量が増加し、この効果はMgCl2、SPR、ROS産生阻害剤であるN-アセチル-システイン(NAC)の添加により抑制された。MgCl2による抑制効果は、スペルミジン合成阻害剤である、システアミンの共処理によって阻害された。
HaCaT細胞を96well plateに播種後、4日間培養した。10mMのMgCl2(10Mg)、10mMのMgCl2+10μMシステアミン(10Mg+Cys)で24時間処理後、100mJUVBを照射した。100μMまたは200μMスペルミジン(SPR)、2mM N-アセチル-システイン(NAC)を、UVB照射の30分前に培地に添加した。H2DCFDAを負荷後、ROS産生量を蛍光強度で算出した。結果を図6に示す。UVB照射によりROS産生量が増加し、この効果はMgCl2、SPR、ROS産生阻害剤であるN-アセチル-システイン(NAC)の添加により抑制された。MgCl2による抑制効果は、スペルミジン合成阻害剤である、システアミンの共処理によって阻害された。
実施例5:過酸化水素による細胞障害に対するマグネシウム化合物の細胞保護作用
HaCaT細胞を96well plateに播種後、4日間培養した。5mMのMgCl2、10mMのMgCl2、10μMシステアミンを添加後、24時間培養した。H2O2を添加してさらに24時間培養後、WST-1試薬を用いて細胞生存率を算出した。結果を図7に示す。H2O2による細胞生存率の低下が、MgCl2の前処理により改善した。
HaCaT細胞を96well plateに播種後、4日間培養した。5mMのMgCl2、10mMのMgCl2、10μMシステアミンを添加後、24時間培養した。H2O2を添加してさらに24時間培養後、WST-1試薬を用いて細胞生存率を算出した。結果を図7に示す。H2O2による細胞生存率の低下が、MgCl2の前処理により改善した。
図3~7の結果からも明らかなとおり、マグネシウム化合物を用いることで、アデノシルメチオニンデカルボキシラーゼ1(AMD1)またはスペルミジンシンターゼ(SRM)の発現量を促進することが確認された。さらに、マグネシウム化合物を用いることで、紫外線照射および活性酸素種による細胞障害から細胞を保護する効果が確認された。かかる作用には、マグネシウム化合物のスペルミジン産生促進作用が大きく関与していることが示唆された。

- マグネシウム化合物を有効成分として含有することを特徴とする、スペルミジン産生促進剤。
- マグネシウム化合物が、塩化マグネシウム、乳酸マグネシウムまたは硫酸マグネシウムである、請求項1に記載のスペルミジン産生促進剤。
- 表皮細胞および/またはケラチノサイトにおけるスペルミジンの産生を促進するために用いられる、請求項1または2に記載のスペルミジン産生促進剤。
- マグネシウム化合物を有効成分として含有するスペルミジン産生促進剤を適用することを含む、美容方法または非治療的方法。
- スペルミジンの産生を促進するためのマグネシウム化合物の使用。
- マグネシウム化合物を有効成分として含有することを特徴とする、スペルミジンの産生を促進する皮膚外用剤。
