- APC Goat anti-mouse IgG antibody Biolegend, Cat: 405308 (antibody concentration: 2 ⁇ g/ ml) and stained with antigen (4°C, 30 minutes). After the staining reaction, 300 ⁇ l of buffer was added per sample, and the antigen expression level was measured with a flow cytometer (BD bioscience, LSFRORESSA).
- E4re#1, 13, 14, 20, and 29 vectors were intramuscularly injected into 6-7 week-old BALB/c mice in each group. After administration, a blood sample of about 300 ⁇ l was collected by orbital blood sampling from the mice at 2-3 weeks. Thereafter, plasma was separated by centrifugation (8000 rpm, 10 minutes, 20° C.), and the amount of neutralizing antibody present in the blood was measured by enzyme-linked immunosorbent assay (ELISA).

- E1, E3 및 E4 유전자가 결실된 아데노바이러스에서 E4 유전자가 E1 유전자의 결실 위치에 삽입된 것을 특징으로 하는, 재조합 아데노바이러스 벡터.
- 제1항에 있어서, 상기 E4 유전자는 E4orf6(E4 open reading frame 6) 유전자인 것을 특징으로 하는, 재조합 아데노바이러스 벡터.
- 제1항에 있어서, 상기 결실된 E4 유전자는 E4orf6 유전자 영역의 10 내지 1,000bp 길이의 염기 서열인 것을 특징으로 하는, 재조합 아데노바이러스 벡터.
- 제1항에 있어서, 상기 아데노바이러스는 Ad2, Ad4, Ad5, Ad11, Ad26, Ad35, Ad5/35, ChAd68, FAd9 또는 PAd3인 것을 특징으로 하는, 재조합 아데노바이러스 벡터.
- 제1항에 있어서, 상기 결실된 E4 유전자는 E4orf6, E4orf6/7 또는 E4orf4 영역인 것을 특징으로 하는, 재조합 아데노바이러스 벡터.
- 제5항에 있어서, 상기 결실된 E4 유전자는 10 내지 1,500bp 길이의 염기 서열인 것을 특징으로 하는, 재조합 아데노바이러스 벡터.
- 제1항에 있어서, 상기 E1 유전자의 결실 위치에 삽입된 E4 유전자는 E4orf6 유전자인 것을 특징으로 하는, 재조합 아데노바이러스 벡터.
- 제7항에 있어서, 상기 E4orf6 유전자는 코작 서열(kozak sequence)이 제거된 것을 특징으로 하는, 재조합 아데노바이러스 벡터.
- 제1항에 있어서, 상기 E1 유전자의 결실 위치에 항원 단백질을 코딩하는 서열이 삽입된 것을 특징으로 하는, 재조합 아데노바이러스 벡터.
- 제9항에 있어서, 상기 항원 단백질을 코딩하는 서열은 E4orf6 유전자의 5' 위치에 삽입된 것을 특징으로 하는, 재조합 아데노바이러스 벡터.
- 제10항에 있어서, 상기 E4orf6 유전자는 정방향으로 발현되는 것을 특징으로 하는, 재조합 아데노바이러스 벡터.
- 제1항에 있어서, 상기 E1 유전자의 결실 위치에 삽입된 E4 유전자는 EF-1a, mPGK, RSV, CMV, E4orf6의 -0.25kb, E4orf6의 -0.5kb 및 E4orf6의 -1.0kb 로 이루어진 군으로부터 선택되는 어느 하나의 프로모터에 의해 발현되는 것을 특징으로 하는, 재조합 아데노바이러스 벡터.
- 제9항에 있어서, 상기 항원 단백질을 코딩하는 서열은 CMV(cytomegalovirus) 프로모터에 의해 발현되는 것을 특징으로 하는, 재조합 아데노바이러스 벡터.
- 제1항의 벡터에 코로나바이러스 스파이크 단백질을 코딩하는 서열이 삽입된, 코로나바이러스감염증 예방용 백신.
- 제14항에 있어서, 상기 코로나바이러스는 SARS-CoV, MERS-CoV 또는 SARS-CoV-2인 것을 특징으로 하는, 코로나바이러스감염증 예방용 백신.
- 제14항에 있어서, 상기 스파이크 단백질(Spike protein)은 스파이크 단백질의 S1 및 S2 유전자 사이의 절단 인지 서열이 제거되고 링커 서열을 도입한 재조합 스파이크 단백질인 것을 특징으로 하는, 코로나바이러스감염증 예방용 백신.
- 제16항에 있어서, 상기 링커는 (GGGGS)n으로 구성되는 것을 특징으로 하는, 코로나바이러스감염증 예방용 백신.
- 제17항에 있어서, 상기 n은 1 내지 5의 정수인 것을 특징으로 하는, 코로나바이러스감염증 예방용 백신.
