Abstract
본 발명은 코로나바이러스 유래 항원 단백질 또는 상기 단백질을 암호화하는 유전자를 포함하는 세포외소포체 및 그의 용도에 관한 것이다. 본 발명에서는 항원 단백질 또는 mRNA가 로딩된 활성화된 면역세포를 이용하여 이로부터 유래한 세포외소포체를 마우스에 투여해 면역화시킨 결과, 우수한 항원 특이적 중화항체 생성 및 T 세포 반응 유도 효과를 확인하였고, 동결건조 후 상온 및 냉장 조건에서 저장할 경우 우수한 안정성 및 항체 생성 효과를 실험적으로 확인하였다. 이로부터, 본 발명에 따른 세포외소포체 또는 이를 포함하는 백신 조성물은 우수한 항원 특이적 면역반응 유도 효과 및 안정성을 갖춘 다양한 질환에 적용 가능한 플랫폼으로써 감염성 질환, 특히 코로나바이러스 감염증을 포함한 다양한 질환의 예방 또는 치료용 백신 개발분야에서 유용하게 활용될 수 있을 것으로 기대된다.
Description
본 발명은 코로나바이러스 유래 항원 단백질 또는 상기 단백질을 암호화하는 유전자를 포함하는 세포외소포체 및 그의 용도에 관한 것이다.
백신이란 인간을 비롯한 동물에 특정 질병 혹은 병원체에 대한 후천성 면역을 부여하는 의약품으로, 주로 질병을 일으키는 미생물 병원체의 항원인식부위와 유사한 구조를 가지지만, 병원체와는 달리 병원성이 없다는 차이가 있다. 현재 다양한 병원체, 다양한 질병에 대한 여러 종류의 백신이 개발되어 있으나, 아직도 보다 효과적이면서 안전한 백신 개발이 필요한 질병들이 다수 존재한다.
세포외소포체는 이중지질막으로 이루어진 수십 nm에서 수백 nm 크기를 가진 나노소포체로 단백질, 지질 및 유전자 등 생물학적 활성을 가진 물질들로 구성되어 있다. 이러한 세포외소포체에는 대표적으로 엑소좀(exosome) 외에 엑토좀(ectosome), 미세소포(microvesicle) 또는 세포자멸체(apoptotic body) 등이 있다. 종래에는 이러한 세포외소포체가 세포에서 분비되는 찌꺼기로 여겨졌으나, 최근에는 세포외소포체들의 임상학적 의미가 대두되면서 다양한 연구가 진행되고 있다. 특히, 세포에 의해 배출되는 구형 소낭인 엑소좀은 모세포의 단백질, DNA 등과 같은 다양한 정보를 지니고 있어, 이를 바이오마커로 활용한 암진단 마커 및 센서 개발이 활발히 진행되고 있다.
엑소좀은 다양한 세포들로부터 분비되는 막 구조의 작은 소낭(대략30-100 nm의 직경)으로서, 전자현미경을 통한 연구에서 원형질막으로부터 직접 떨어져 나가는 것이 아니라 다낭체(multivesicular bodies, MVBs)라고 불리는 세포 내 특정 구획에서 기원하며 세포 밖으로 방출, 분비되는 것이 관찰되었다. 즉, 다낭체와 원형질막의 융합이 일어나면 소낭들은 세포 밖 환경으로 방출되는데, 이것을 엑소좀이라고 한다. 이러한 엑소좀은 적혈구 세포뿐만 아니라, B-림프구, T-림프구, 수지상 세포, 혈소판, 대식세포 등을 포함한 다양한 종류의 면역세포들과 종양세포, 줄기세포 등도 살아있는 상태에서 엑소좀을 생산하여 분비한다고 알려져 있다.
포유류 세포 유래 엑소좀은 지혈, 조직 재생, 줄기세포 유지, 염증/면역 반응 조절, 배아 발달 등의 다양한 생리적 기능에 관여한다고 알려져 있으며, 특히 면역세포에서 유래한 엑소좀은 염증성 사이토카인을 전달하거나, 직간접적으로 항원을 제시하여 면역 반응을 촉진할 수도 있음이 보고되어 있다(Nat Rev Immunol. 2009 Aug;9(8):581-93). 이러한 엑소좀의 특성 및 생체 내 기능에 기반하여, 엑소좀은 종양학, 면역요법 및 재생의학 분야 등에서 나노크기의 물질 전달 수단, 매개체 및 바이오마커 등의 용도로 연구개발되고 있다.
이와 같이 엑소좀을 백신 용도로 이용하기 위한 연구들이 이루어지고 있으나, 아직까지는 일부 질환에 국한되어 있으며, 감염성 질환을 비롯한 다양한 질환에 적용 가능한 백신 플랫폼 기술에 대한 연구개발은 미진한 실정이다.
본 발명자들은 다양한 질환에 범용적으로 적용 가능한 세포외소포체 기반 백신 플랫폼을 개발하기 위해 연구노력한 결과, 세포외소포체가 강력한 면역 자극제 및 항원 운반체로 작용하여 항원 특이적 중화항체 생성 및 항원 특이적 T 세포 반응을 유도하는 것을 확인하였는바, 이로써 본 발명을 완성하였다.
이에, 본 발명은 코로나바이러스(coronavirus) 유래 항원 단백질 또는 상기 단백질을 암호화하는 유전자를 포함하는 세포외소포체를 제공하는 것을 목적으로 한다.
또한, 본 발명은 상기 세포외소포체를 포함하는 코로나바이러스 감염증 예방 또는 치료용 백신 조성물을 제공하는 것을 다른 목적으로 한다.
또한, 본 발명은 상기 세포외소포체를 포함하는 코로나바이러스 감염증 예방 또는 개선용 건강기능식품을 제공하는 것을 또 다른 목적으로 한다.
또한, 본 발명은 상기 세포외소포체를 이를 필요로 하는 개체에 투여하는 단계를 포함하는 코로나바이러스 감염증 예방 또는 치료 방법을 제공하는 것을 또 다른 목적으로 한다.
또한, 본 발명은 코로나바이러스 감염증 예방 또는 치료용 약제의 제조를 위한 상기 세포외소포체의 용도를 제공하는 것을 또 다른 목적으로 한다.
그러나 본 발명이 이루고자 하는 기술적 과제는 이상에서 언급한 과제에 제한되지 않으며, 언급되지 않은 또 다른 과제들은 아래의 기재로부터 당업자에게 명확하게 이해될 수 있을 것이다.
상기와 같은 본 발명의 목적을 달성하기 위하여, 본 발명은 코로나바이러스(coronavirus) 유래 항원 단백질 또는 상기 단백질을 암호화하는 유전자를 포함하는 세포외소포체를 제공한다.
본 발명의 일 구현예로, 상기 코로나바이러스는 SARS-CoV-2(Severe Acute Respiratory Syndrome-Coronavirus-2)일 수 있다.
본 발명의 다른 구현예로, 상기 항원 단백질은 코로나바이러스 유래 스파이크 단백질(spike protein)의 수용체 결합 도메인(Receptor Binding Domain; RBD) 단백질일 수 있다.

본 발명에서는 항원 단백질 또는 mRNA가 로딩된 활성화된 면역세포를 이용하여 이로부터 유래한 세포외소포체를 마우스에 투여해 면역화시킨 결과, 우수한 항원 특이적 중화항체 생성 및 T 세포 반응 유도 효과를 확인하였고, 동결건조 후 상온 및 냉장 조건에서 저장할 경우 우수한 안정성 및 항체 생성 효과를 실험적으로 확인하였다. 이로부터, 본 발명에 따른 세포외소포체 또는 이를 포함하는 백신 조성물은 우수한 항원 특이적 면역반응 유도 효과 및 안정성을 갖춘 다양한 질환에 적용 가능한 플랫폼으로써 감염성 질환, 특히 코로나바이러스 감염증을 포함한 다양한 질환의 예방 또는 치료용 백신 개발분야에서 유용하게 활용될 수 있을 것으로 기대된다.
도 1은 LPS를 처리하여 활성화시킨 THP-1 세포가 M1 대식세포의 표현형을 나타냄을 확인한 결과로서, 도 1a는 활성화되지 않은 THP-1 세포(Resting THP-1) 및 활성화된 THP-1 세포(Activated THP-1)의 형태를 보여주는 현미경 이미지이고, 도 1b는 상기 각 THP-1 세포에서 전염증성(pro-inflammatory cytokines) 마커인 IL-1b, IL-6, IL-8, TNF-α 및 iNOS의 mRNA 수준을 측정한 결과이다.
도 2는 활성화된 THP-1 세포로부터 분리된 엑소좀의 특성을 분석한 결과로서, 도 2a는 NTA(Nanosight Tracking Analyis)를 통해 상기 분리된 엑소좀의 직경을 측정한 결과이고, 도 2b는 상기 엑소좀을 보여주는 투과전자현미경(TEM) 이미지이며, 도 2c는 세포 용해물(cell lysate) 및 분리된 엑소좀(exosome)을 이용해 웨스턴 블롯을 실시하여 엑소좀 마커인 CD63, CD81 및 Flotillin 1 단백질의 발현수준을 측정한 결과이다.
도 3은 본 발명에 따른 SARS-CoV-2 유래 스파이크 단백질의 RBD가 탑재된 활성화된 THP-1 세포 유래 엑소좀(COVID 19-ImmunExo), 또는 보조제(adjuvant)와 RBD 단백질을 함께 투여하여 면역화시킨 마우스에서 항체의 생성을 확인한 결과로서, 도 3a는 상기 엑소좀을 이용한 마우스의 면역화 실험 방법 및 분석 일정을 그림으로 간략히 도시한 것이고, 도 3b는 마우스에 상기 엑소좀(ImmunExo)을 상이한 농도로 투여하거나 또는 보조제(adjuvant)와 RBD 단백질을 함께 투여(Adj+RBD 5ug)하고 7, 21, 30 및 50일 후에 혈청을 50배 희석한 샘플을 이용해 SARS-CoV-2 sVNT 테스트를 실시하여 항체 생성 효과를 분석한 결과이고, 도 3c는 도 3b와 동일한 실험군에서 50일 후에 분리한 혈청을 각각 10배 또는 50배 희석한 후 SARS-CoV-2 sVNT 테스트를 실시하여 항체 생성 효과를 분석한 결과이다.
도 4는 RBD 단백질 또는 mRNA가 탑재된 면역 세포(THP-1 세포 또는 KG-1 세포) 유래 엑소좀의 SARS-CoV-2의 RBD에 특이적인 혈청 내 항체의 결합 정도(antibody binding)를 나타낸 도로서, 구체적으로 (1) 도 4a는 도 3c와 동일하게 각 실험군 마우스에서 상기 상이한 농도의 THP-1 세포 유래 엑소좀, 또는 보조제(adjuvant)와 RBD 단백질을 함께 투여하고 50일 후에 혈청을 분리한 후 SARS-CoV-2의 RBD에 특이적인 혈청 내 항체의 결합 정도(antibody binding)를 흡광도를 측정하여 분석한 결과이고, 2) 도 4b는 KG-1 세포 유래 엑소좀, 또는 보조제(adjuvant)와 RBD 단백질을 함께 투여하고 7일 후에 혈청을 분리한 후 SARS-CoV-2의 RBD에 특이적인 혈청 내 항체의 결합 정도(antibody binding)를 흡광도를 측정하여 분석한 결과이며, 3) 도 4c는 THP-1 세포 유래 엑소좀, 또는 보조제(adjuvant)와 RBD mRNA를 함께 투여하고 7일 및 14일 후에 혈청을 분리한 후 SARS-CoV-2의 RBD에 특이적인 혈청 내 항체의 결합 정도(antibody binding)를 흡광도를 측정하여 분석한 결과이며, 4) 도 4d는 KG-1 세포 유래 엑소좀, 또는 보조제(adjuvant)와 RBD mRNA를 함께 투여하고 7일 및 14일 후에 혈청을 분리한 후 SARS-CoV-2의 RBD에 특이적인 혈청 내 항체의 결합 정도(antibody binding)를 흡광도를 측정하여 분석한 결과이다.
도 5는 SARS-CoV-2 유래 스파이크 단백질의 RBD가 로딩된 활성화된 THP-1 세포 유래 엑소좀(ImmunExo), 또는 보조제(adjuvant)와 RBD 단백질을 함께 투여하여 면역화시킨 마우스로부터 비장세포(splenocyte)를 분리하고 이에 대하여 IFN-γ ELISPOT(Enzyme-linked immunospot) 어세이를 실시한 결과로서, 도 5a는 IFN-γ ELISPOT 어세이를 실시한 후 반점의 생성여부를 보여주는 결과 및 반점 수의 정량화 그래프 결과이고, 도 5b는 비장세포 배양액을 이용해 ELISA를 실시하여 IFN-γ 단백질 수준을 측정한 결과이다.
도 6은 본 발명에 따른 SARS-CoV-2 유래 스파이크 단백질의 RBD가 로딩된 활성화된 THP-1 세포 유래 엑소좀의 저장에 따른 안정성을 확인한 결과로서, 도 6a는 엑소좀의 효과를 분석하기 위한 실험 일정을 그림으로 간략히 도시한 것이고, 도 6b는 동결건조 후 4℃에서 7일 동안 저장한 엑소좀을 마우스에 각각 0.1, 0.5, 또는 1 ug 투여하여 면역화시키고 투여 후 7일 및 21일에 각각 혈청을 얻어 SARS-CoV-2 sVNT 테스트를 실시해 동결건조하여 저장하지 않은 원래 상태의 엑소좀(fresh)과 항체 생성 효과를 비교하여 나타낸 결과이고, 도 6c는 동결건조 후 상온에서 7일 동안 저장한 엑소좀을 마우스에 1 ug 투여하여 면역화시키고 투여 후 7일 및 21일에 각각 혈청을 얻어 SARS-CoV-2 sVNT 테스트를 실시해 동결건조하여 저장하지 않은 원래 상태의 엑소좀(fresh)과 항체 생성 효과를 비교하여 나타낸 결과이다.
본 발명은 강력한 면역 자극제 및 항원 운반체로 기능하는 세포외소포체를 이용하여 다양한 감염성 질환에 대한 예방 또는 치료용 백신 개발분야에서 유용하게 활용될 수 있는 백신 플랫폼 기술에 관한 것이다.
이하, 본 발명을 상세히 설명한다.
본 발명은 코로나바이러스(coronavirus) 유래 항원 단백질 또는 상기 단백질을 암호화하는 유전자를 포함하는 세포외소포체를 제공한다.
본 발명에 있어서, 상기 코로나바이러스는 SARS-CoV-2(Severe Acute Respiratory Syndrome-Coronavirus-2)일 수 있다.
또한, 본 발명에 있어서, 상기 항원 단백질은 코로나바이러스 유래 스파이크 단백질(spike protein)의 수용체 결합 도메인(Receptor Binding Domain; RBD) 단백질일 수 있고, 상기 항원 단백질을 암호화하는 유전자는 코로나바이러스 유래 스파이크 단백질(spike protein)의 수용체 결합 도메인(Receptor Binding Domain; RBD) 단백질을 암호화하는 뉴클레오티드일 수 있다. 또한, 상기 뉴클레오티드는 DNA, RNA 또는 이들의 조합일 수 있다.
또한, 본 발명에 있어서, 상기 항원 단백질 또는 상기 단백질을 암호화하는 유전자는 상기 세포외소포체 내 탑재된 것일 수 있다.
상기 항원 단백질 또는 상기 단백질을 암호화하는 유전자를 세포외소포체로 탑재하는 방법은 구체적으로 제한되지 않으며 해당 기술분야에서 공지된 방법을 당업자가 적절히 선택하여 이용할 수 있다.
본 발명에서 사용되는 용어, "항원(antigen)"이란 숙주 개체 내에서 항체를 생성하기 위한 면역 반응을 유도할 수 있는 분자를 의미하며, 항원은 항체의 타겟으로 각 항체는 면역계의 세포가 항원과 접촉한 후 이에 대응하기 위해 항원 특이적으로 생산된다. 본 발명에 있어서, 상기 항원은 전술한 바와 같이 면역 반응을 유도할 수 있는 형태라면 단백질, 상기 단백질을 암호화하는 유전자, 상기 유전자의 mRNA 형태 모두를 포함하고, 항원의 기원은 구체적으로 한정되지 않으며, 비제한적인 예로 바이러스, 박테리아 또는 진균을 포함하는 미생물에서 유래된 것, 암세포에서 유래된 것을 모두 포함할 수 있다.
본 발명에서 사용되는 용어, "세포외소포체"란 세포에서 유래되는 막으로 둘러싸인 작은 구체를 의미하며, 이러한 구체는 그 명칭이 만들어지는 세포의 기원이나 만들어지는 방법에 따라 매우 다양하다. 본 발명에서는 세포에서 만들어지는 방법에 따라 명칭된 엑소좀(exosome), 엑토솜(ectosome), 미세소포(microvesicle) 및 세포자멸체(apoptotic body)로 이루어진 군으로부터 선택되는 하나 이상을 포함할 수 있고, 구체적으로는 엑소좀일 수 있으나, 이에 제한되지 않는다.
상기 세포외소포체는 자연계, 예컨대 동물, 식물 및 미생물로 이루어진 군에서 선택되는 하나 이상으로부터 유래된 세포외소포체 또는 인공적으로 제조된 세포외소포체일 수 있다. 또한, 상기 세포는 자연계 생물 개체로부터 분리된 세포일 수 있다. 또한, 상기 세포는 인간 및 비인간 포유류를 포함하는 임의 유형의 동물 또는 식물 유래일 수 있다. 상기 동물세포는 체세포, 생식세포, 면역세포, 신경세포 및 종양세포로 이루어진 군에서 선택되는 하나 이상의 세포일 수 있다.
또한, 상기 세포외소포체는 활성화된 면역세포 유래일 수 있고, 구체적으로는 활성화된 THP-1 세포 또는 KG-1 세포 유래 일 수 있다. 이 때, 상기 활성화는 지질다당류(lipopolysaccharide; LPS)에 의해 이루어지는 것일 수 있고, 상기 THP-1 세포가 활성화되는 경우, THP-1 세포는 대식세포와 유사하게 세포 형태가 변화함과 동시에 IL-1b, IL-6, IL-8, TNF-α 및 iNOS으로 이루어진 군에서 선택되는 하나 이상의 단백질을 발현할 수 있다.
본 발명에서는, 구체적인 실시예에서 일례로 항원 자극을 통해 활성화시킨 면역세포의 표현형 변화를 분석하고, 이로부터 분리된 엑소좀의 특성을 관찰하였다.
구체적으로, 본 발명의 일실시예에서는 인간 단핵구 세포주인 THP-1 세포에 지질다당류(LPS)를 처리한 결과, LPS를 처리하지 않은 세포와 비교할 때 세포의 형태가 변화하고 M1 대식세포의 마커들을 발현하는 것을 확인하였다(실시예 1 참조).
상기 체세포는 인간을 포함한 포유류를 포함하는 임의 유형의 동물의 신체를 구성하는 세포로서 본 발명에서는 생식세포, 면역세포, 신경세포 및 종양세포를 제외한 나머지 모든 종류의 세포를 포함할 수 있다.
상기 면역세포(immune cells)는 체내에서 면역 반응을 결정짓는 세포로서, 외부에서 침입한 병원균이나 이물질, 바이러스 등에 저항하여 이겨낼 수 있도록 방어하고 면역력을 조절하며, 체내에서 끊임없이 생성되는 암세포를 제거하는 역할을 하는 세포들을 의미한다. 구체적으로, NK세포(natural killer cell), 수지상세포(dendritic cell), T 세포, B 세포, 대식세포(macrophage) 및 림프구(lymphocyte)를 포함할 수 있으나, 이에 한정되지 않는다.
상기 신경세포(neuronal cells)는 신경계를 구성하는 세포로서, 나트륨 통로, 칼륨 통로 등의 이온 통로를 발현하여 다른 세포와는 달리 전기적인 방법으로 신호를 전달할 수 있으며 인접한 다른 신경세포와는 시냅스라는 구조를 통해 화학적 신호를 주고받음으로써 다양한 정보를 교환하는 세포를 말한다. 본 발명에 있어서, 상기 신경세포는 구체적인 종류가 제한되지 않으며 세포외소포체를 분비하는 세포라면 모두 포함될 수 있다.
상기 종양세포(tumor cells)는 암세포(cancer cells)와 동일한 의미로 사용되며, 정상세포와는 달리 세포주기가 조절되지 않아 빠르고 불규칙하게 성장하는 세포를 의미한다. 본 발명에 있어서 상기 종양세포는 기원 조직은 구체적으로 한정되지 않으며 세포외소포체를 분비하는 종양세포라면 모두 포함될 수 있다.
또한, 본 발명의 일 구현예에 있어서, 상기 세포외소포체는 CD63, CD81 및 Flotillin 1으로 이루어진 군에서 선택되는 하나 이상을 발현하는 것일 수 있고, 구체적으로는 CD63, CD81 및 Flotillin 1을 모두 발현하는 것일 수 있다.
본 발명의 다른 실시예에서는 상기 활성화된 면역세포 유래 엑소좀, 바람직하게 활성화된 TH-1 세포 유래 엑소좀이 평균 154.0nm의 직경을 가지며, CD63, CD81 및 Flotillin 1의 엑소좀 마커 단백질들을 발현하는 것을 확인하였다(실시예 2 참조).
또한, 본 발명은 상기 세포외소포체를 포함하는 코로나바이러스 감염증 예방 또는 치료용 백신 조성물을 제공한다.
상기 "세포외소포체", "코로나바이러스", "항원" 등은 전술한 범위 내일 수 있다.
본 발명에서 사용되는 용어, "예방"이란 본 발명에 따른 백신 조성물의 투여에 의해 특정 질병 또는 질환을 억제시키거나 발병을 지연시키는 모든 행위를 의미한다.
본 발명에서 사용되는 용어, "치료"란 본 발명에 따른 백신 조성물의 투여에 의해 특정 질병 또는 질환에 대한 증세가 호전되거나 이롭게 변경되는 모든 행위를 의미한다.
본 발명에서 사용되는 용어, "백신"이란 인간을 포함하는 숙주에게서 해당 항원에 대한 면역 반응을 유도함으로써 해당 병원체 또는 항원에 의한 감염 또는 재감염의 예방, 증상의 중증도 감소 또는 증상의 제거, 또는 해당 병원체나 항원에 의한 질환의 실질적 또는 완전한 제거를 모두 포함하는 의미이다. 따라서 본 발명의 백신 조성물은 해당 병원체의 감염 전 또는 질환의 발병 전에 예방적으로 개체에 투여되는 것이 바람직하나, 해당 병원체의 감염 후 또는 질환 발병 후에 치료적으로 투여될 수 있다.
본 발명의 일실시예에서는, SARS-CoV-2 유래 스파이크 단백질(spike protein)의 수용체 결합 도메인(Receptor Binding Domain; RBD) 단백질 또는 mRNA가 로딩된 활성화된 THP-1 세포 또는 KG-1 세포 유래 엑소좀을 제조하여 마우스에 피하로 투여하여 면역화시킨 후, 중화항체의 생성 여부를 분석한 결과 보조제(Adjuvant)와 상기 RBD 단백질 또는 mRNA를 함께 투여한 경우와 비교해 훨씬 적은 양으로도 유사한 수준의 RBD 특이 IgG 및 중화항체 역가를 나타내는 것을 확인하였다(실시예 3 내지 5 참조).
또한, 본 발명에 있어서, 상기 백신 조성물은 항원 특이적 항체 생성 및 T 세포 매개 면역 반응을 동시에 유도하는 것일 수 있다.
본 발명의 다른 실시예에서는, 상기 엑소좀을 투여하여 면역화된 마우스에서 비장세포를 분리하여 RBD 항원 특이적 T 세포 반응을 분석한 결과 보조제(Adjuvant)와 상기 RBD를 함께 투여한 마우스에서는 상기 T 세포 반응이 유도되지 않은 반면 상기 엑소좀으로 면역화된 마우스에서는 강력한 T 세포 반응이 유도되는 것을 확인하였다(실시예 6 참조).
또한, 본 발명에 있어서, 상기 백신 조성물은 동결건조된 것일 수 있다.
본 발명자들은 실시예를 통해 상기 백신 조성물의 구체적인 면역반응 유도 및 저장 안정성을 확인하였다. 구체적으로, 본 발명에 따른 상기 엑소좀의 저장 안정성을 평가하기 위해 동결건조된 엑소좀(COVID19-ImmunExo)을 4℃ 냉장 조건에서 7일동안 보관하거나 또는 상온에서 보관한 후 마우스에 피하 면역화시켜 항체 생성 여부를 분석한 결과, 상기 조건에서 각각 저장한 후에도 우수한 안정성과 항체 생성 효과가 유지되는 것을 확인하였다(실시예 7 참조).
상기 본 발명의 실시예를 통해 활성화된 면역세포 유래 엑소좀은 피하 주사 후 림프절 방향성을 보였으며, 주로 국소 대식세포와 수지상 세포에 흡수되어 수지상 세포에 의해 엑소좀에서 MHC-I/펩티드 복합체가 나타나고, 이것은 SARS-CoV-2에 대한 중화 항체를 보다 효과적으로 생성하는 것과 더불어 면역화된 마우스의 비장세포에서 RBD 특이적 T 세포 반응을 유발할 수 있게 하는 것을 알 수 있었다.
이에, 본 발명은 실시예에서 확인한 결과에 근거하여, 세포에서 분비된 엑소좀을 포함하는 나노 크기의 세포외소포체가 강력한 면역 자극제 및 항원 운반체로 작용하여 표적 질환에 대한 효과적인 백신을 설계할 수 있는 플랫폼이 될 수 있을 것임을 강력히 시사한다.
본 발명의 백신 조성물은 임의의 적절한, 약학적으로 허용되는 제제로 제조될 수 있다. 예컨대 즉석 투여 용액 또는 현탁액 형태이거나, 투여 전 희석에 적절한 농축된 원액이거나, 또는 재구성 가능한 형태 예컨대 동결건조, 냉동-건조, 또는 냉동 제제 형태로 제조될 수 있다.
본 발명의 백신 조성물은 약학적으로 허용되는 담체를 추가로 포함하여 제제화될 수 있다. 구체적으로, 상기 담체는 예를 들어, 콜로이드 현탁액, 분말, 식염수, 지질, 리포좀, 미소구체(microspheres) 또는 나노 구형입자일 수 있다. 이들은 운반 수단과 복합체를 형성하거나 관련될 수 있고, 지질, 리포좀, 미세입자, 금, 나노입자, 폴리머, 축합 반응제, 다당류, 폴리아미노산, 덴드리머, 사포닌, 흡착 증진 물질 또는 지방산과 같은 당업계에 공지된 운반 시스템을 사용하여 생체 내 운반될 수 있다.

본 발명의 백신 조성물은 항원 보조제(adjuvant)를 추가로 포함할 수 있다. 항원 보조제는 항원에 대한 면역 반응을 증강시키는 하나 이상의 물질로 이루어진 것일 수 있다. 상기 항원 보조제는 예컨대 완전 프로인트(Freund), 불완전 프로인트, 사포닌, 겔 성상의 알루미늄 보조제, 표면 활성 물질(예: 리소레시틴, 플루론 글리콜, 다중 음이온, 펩티드, 오일 또는 탄화수소 에멀젼 등), 식물성 오일(면실유, 땅콩유, 옥수수유 등), 비타민 E 아세테이트 등일 수 있다.
본 발명의 백신 조성물은 본 발명이 속하는 기술 분야에서 통상적으로 이용되는 방법으로 제조할 수 있다. 상기 백신 조성물은 경구형 또는 비경구형 제제로 제조할 수 있고, 비경구형 제제는 바람직하게 경피, 근육내, 복막내, 정맥내, 피하내, 비강 또는 경막 외(eidural)로 이루어진 군으로부터 선택되는 어느 하나의 투여경로를 통해 투여될 수 있다.
용어 "투여"란 적절한 방법으로 개체에게 소정의 물질을 도입하는 것을 의미하며, "개체"란 코로나바이러스 감염증을 보유할 수 있는 인간을 포함한 쥐, 생쥐, 가축 등의 모든 생물을 의미한다. 구체적인 예로, 인간을 포함한 포유동물일 수 있다.
본 발명의 백신 조성물은 약제학적으로 유효한 양으로 투여한다. 본 발명의 용어 "약제학적으로 유효한 양"이란, 백신효과를 나타낼 수 있을 정도의 충분한 양으로, 부작용 또는 심각하거나 과도한 면역반응을 일으키지 않을 정도의 양을 의미하며, 유효 용량의 수준은 치료하려는 장애, 장애의 중증도, 백신 물질의 활성, 투여 경로, 단백질의 제거 속도, 치료 지속 기간, 백신 조성물과 조합되거나 동시에 사용되는 물질, 개체의 연령, 체중, 성별, 식습관, 일반적인 건강 상태 및 의학 분야에 공지된 인자를 비롯한 다양한 인자들에 따라 달라질 수 있다.
또한, 본 발명은 상기 세포외소포체를 포함하는 코로나바이러스 감염증 예방 또는 개선용 건강기능식품을 제공한다.

용어 "개선"이란 치료되는 상태와 관련된 파라미터, 예를 들어, 증상의 정도를 적어도 감소시키는 모든 행위를 의미할 수 있다. 이때, 상기 건강기능식품은 코로나바이러스 감염증의 예방 또는 개선을 위하여 해당 질병의 발병 단계 이전 또는 발병 후, 치료를 위한 약제와 동시에 또는 별개로서 사용될 수 있다.
상기 건강기능식품에서, 유효성분은 식품에 그대로 첨가하거나 다른 식품 또는 식품 성분과 함께 사용될 수 있고, 통상적인 방법에 따라 적절하게 사용될 수 있다. 유효 성분의 혼합량은 그의 사용 목적(예방 또는 개선용)에 따라 적합하게 결정될 수 있다. 일반적으로, 식품 또는 음료의 제조 시에 상기 건강기능식품은 원료에 대하여 구체적으로 약 15 중량% 이하, 보다 구체적으로 약 10 중량% 이하의 양으로 첨가될 수 있다. 그러나, 건강 및 위생을 목적으로 하거나 또는 건강 조절을 목적으로 하는 장기간의 섭취의 경우에는 상기 양은 상기 범위 이하일 수 있다.
상기 건강기능식품은 담체, 희석제, 부형제 및 첨가제 중 하나 이상을 더 포함하여 정제, 환제, 산제, 과립제, 분말제, 캡슐제 및 액제 제형으로 이루어진 군에서 선택된 하나로 제형될 수 있다. 일 구현예에 따른 화합물을 첨가할 수 있는 식품으로는, 각종 식품류, 분말, 과립, 정제, 캡슐, 시럽제, 음료, 껌, 차, 비타민 복합제, 건강기능성 식품류 등이 있다.

상기 건강기능식품은 상기 유효성분을 함유하는 것 외에 특별한 제한없이 다른 성분들을 필수 성분으로서 함유할 수 있다. 예를 들어, 통상의 음료와 같이 여러 가지 향미제 또는 천연 탄수화물 등을 추가 성분으로서 함유할 수 있다. 상술한 천연 탄수화물의 예는 모노사카라이드, 예를 들어, 포도당, 과당 등; 디사카라이드, 예를 들어 말토스, 슈크로스 등; 및 폴리사카라이드, 예를 들어 덱스트린, 시클로덱스트린 등과 같은 통상적인 당, 및 자일리톨, 소르비톨, 에리트리톨 등의 당 알코올일 수 있다. 상술한 것 이외의 향미제로서 천연 향미제 (타우마틴, 스테비아 추출물 (예를 들어, 레바우디오시드 A, 글리시르히진 등)) 및 합성 향미제 (사카린, 아스파르탐 등)를 유리하게 사용할 수 있다. 상기 천연 탄수화물의 비율은 당업자의 선택에 의해 적절하게 결정될 수 있다.

이하, 본 발명의 이해를 돕기 위하여 바람직한 실시예를 제시한다. 그러나 하기의 실시예는 본 발명을 보다 쉽게 이해하기 위하여 제공되는 것일 뿐, 하기 실시예에 의해 본 발명의 내용이 한정되는 것은 아니다.
[실시예]
실시예 1. 활성화된 면역세포의 표현형 관찰
본 발명자들은 세포외소포체를 이용한 백신을 개발하기 위하여, 먼저 면역세포를 선정하고 면역세포를 항원으로 자극해 활성화된 면역세포로부터 분리된 엑소좀을 이용하여 백신을 제조하고자 하였다. 이를 위해, 상기 면역세포의 일례로 인간 단핵구 세포주인 THP-1 세포를 활성화시키고 활성화된 세포의 표현형을 관찰하였다.
구체적으로, THP-1 세포(8x105 cells/mL)에 지질다당류(lipopolysaccharide; LPS)를 100ng/mL로 처리하여 상기 세포를 활성화시킨 후 LPS를 처리하지 않아 활성화되지 않은 THP-1 세포와 현미경으로 세포 형태를 관찰하였다. 그 결과, 도 1a에 나타낸 바와 같이 활성화되지 않은 THP-1 세포(Resting THP-1)와 비교할 때 활성화된 THP-1 세포(Activated THP-1)의 경우 대식세포와 유사하게 세포 형태가 변화된 것을 확인하였다.
나아가 상기 결과에 근거하여, 해당 기술분야에 보고된 바와 같이 LPS 처리에 의해 활성화된 THP-1 세포가 M1 대식세포로 분화되었는지 여부를 확인하기 위해, M1 대식세포가 분비하는 전염증성(pro-inflammatory cytokines) 마커인 IL-1b, IL-6, IL-8, TNF-α 및 iNOS의 mRNA 수준을 측정하였다. 그 결과, 도 1b에 나타낸 바와 같이 활성화되지 않은 THP-1 세포의 경우 상기 마커들이 거의 발현되지 않은 것으로 나타난 반면에 활성화된 THP-1 세포의 경우 상기 마커들의 발현이 증가하였으며, 특히 IL-1b, IL-6 및 IL-8의 mRNA 수준이 현저히 증가한 것을 확인하였다.
상기 결과를 통해 LPS 처리를 통해 활성화된 THP-1 세포가 M1 대식세포의 표현형을 나타내는 것을 알 수 있었다.
실시예 2. 활성화된 면역세포 유래 엑소좀의 특성 확인
본 발명자들은 상기 활성화된 THP-1 세포 유래 엑소좀의 특성을 확인하기 위하여, 상기 세포로부터 하기 과정을 통해 엑소좀을 분리하였다. 구체적으로, 세포와 세포 파편을 제거하기 위해 세포배양액을 1200 rpm에서 5분 동안 원심분리하여 상층액을 수집한 후, 엑소좀보다 크기가 큰 미립자(microparticle)를 제거하기 위해 상층액을 0.2um 필터에 통과시킨 다음, Tangential flow filtration(TFF) system을 이용하여 엑소좀의 농축 및 세척을 진행하였다.
다음으로, NTA(Nanosight Tracking Analyis) 및 투과전자현미경 관찰을 수행하여 분리된 엑소좀의 특성을 분석하였다. 그 결과, 도 2a 및 도 2b에 나타낸 바와 같이 분리된 엑소좀의 평균 직경은 154.0nm이며, 원형의 형태를 갖는 것을 확인하였다.
또한, 분리된 엑소좀의 농도는 2.6 x 1011 particles/ml이었고, BCA 어세이를 통해 엑소좀 단백질 농도를 정량분석한 결과 3.5 mg/ml임을 확인하였다. 나아가 상기 분리된 엑소좀이 엑소좀 마커를 발현하는지 여부를 조사하기 위해 세포 용해물(cell lysate) 및 분리된 엑소좀(exosome)을 이용해 웨스턴 블롯을 실시하여 엑소좀 마커인 CD63, CD81 및 Flotillin 1의 발현수준을 측정하였다. 그 결과, 도 2c에서 볼 수 있는 바와 같이 세포 용해물에서는 CD63만이 약한 수준으로 발현된 반면에 분리된 엑소좀에서는 상기 마커 단백질들이 모두 높은 수준으로 발현되는 것을 확인하였다.
상기 결과들로부터, 상기 활성화된 THP-1 세포로부터 분리된 엑소좀은 상기와 같은 특성을 갖는 엑소좀인 것을 명확히 알 수 있었다.
실시예 3. 재조합 SARS-CoV-2 유래 단백질이 로딩된 엑소좀 제조
본 발명자들은 상기 실시예 2에서 분리된 활성화된 THP-1 세포 또는 KG-1 세포 유래 엑소좀에 SARS-CoV-2 유래 스파이크 단백질(spike protein)의 수용체 결합 도메인(Receptor Binding Domain; RBD) 단백질을 로딩하여 SARS-CoV-2 감염증에 대한 백신으로써 예방 효과를 검증하고자 하였다.
이를 위해, 먼저 SARS-CoV-2 유래 스파이크 단백질의 RBD(YP_009724390.1)(Ser325-Lys529)를 암호화하는 DNA 서열이 C-말단에서 마우스 IgG2a의 Fc 영역과 융합된 단백질의 컨스트럭트(Cat. No. Ab-P0018, AbClon)를 준비하였다. 이후 상기 컨스트럭트로부터 발현시킨 RBD 단백질을 상기 활성화된 THP-1 세포 또는 KG-1 세포 유래 엑소좀에 로딩하기 위해, 3 x 1011 (9 mg)의 엑소좀과 상기 RBD 단백질 300 ug을 준비하고 체외충격파(Extracorporeal shockwave; ESW)를 처리한 후, 초원심분리기(32000 rpm, 2시간)를 이용하여 RBD 단백질이 로딩된 엑소좀의 세척 및 농축을 진행하였다.
이후 ELISA를 이용해 상기 THP-1 세포 유래 엑소좀에 로딩된 RBD 단백질의 양을 정량적으로 분석한 결과, 총 40.754ug의 RBD 단백질이 엑소좀 내로 로딩된 것을 확인하였다. 본 발명자들은 상기에서 제조된, SARS-CoV-2 유래 스파이크 단백질의 RBD가 로딩된 활성화된 THP-1 세포 또는 KG-1 세포 유래 엑소좀(ImmunExo RBD 또는 COVID 19-ImmunExo)에 대하여, 하기 실험들을 통해 코로나바이러스(SARS-CoV-2) 감염증-19(COVID-19)에 대한 효과를 평가하고자 하였다.
실시예 4. 재조합 SARS-CoV-2 유래 mRNA가 로딩된 엑소좀 제조
본 발명자들은 상기 실시예 2에서 분리된 활성화된 THP-1 세포 또는 KG-1 유래 엑소좀에 SARS-CoV-2 유래 스파이크 단백질(spike protein)의 수용체 결합 도메인(Receptor Binding Domain; RBD) mRNA를 로딩하여 SARS-CoV-2 감염증에 대한 백신으로써 예방 효과를 검증하고자 하였다.
구체적으로, RBD mRNA는 DNA template로부터 In vitro transcription (IVT) 과정을 통해 제작하였다. 제작된 RBD mRNA는 THP-1 세포 또는 KG-1 세포 유래 엑소좀(9 mg)과 RBD mRNA 150 ug을 준비하고 체외충격파(Extracorporeal shockwave; ESW)를 처리한 후, 초원심분리기(32000 rpm, 2시간)를 이용하여 RBD mRNA가 로딩된 엑소좀의 세척 및 농축을 진행하였다.
이 후 qRT-PCR를 이용해 상기 THP-1 세포 또는 KG-1 세포 유래 엑소좀에 로딩된 RBD mRNA의 양을 정량적으로 분석한 결과, THP-1 세포 유래 엑소좀 내에는 총 81 ug, KG-1 세포 유래 엑소좀 내에는 64.5 ug이 로딩된 것을 확인하였다. 본 발명자들은 상기에서 제조된, SARS-CoV-2 유래 스파이크 단백질의 RBD mRNA가 로딩된 활성화된 THP-1 세포 또는 KG-1 세포 유래 엑소좀(Exo+RBD mRNA)에 대하여, 하기 실험들을 통해 코로나바이러스(SARS-CoV-2) 감염증-19(COVID-19)에 대한 효과를 평가하고자 하였다.
실시예 5. COVID-19에 대한 활성화된 면역세포 유래 엑소좀의 백신 효능 확인
본 발명자들은 상기 실시예 3에서 제조한 SARS-CoV-2 유래 스파이크 단백질의 RBD 단백질 또는 mRNA가 로딩된 활성화된 THP-1 세포 또는 KG-1 세포 유래 엑소좀에 대하여 COVID-19에 대한 백신으로써의 효능을 평가하고자 하였다.
이를 위해, 마우스를 이용해 도 3a에 도시된 일정에 따라 in vivo 면역화 실험을 진행하였다. 구체적으로, 해당 마우스에 스파이크 단백질의 RBD 단백질이 로딩된 활성화된 THP-1 세포 유래 엑소좀(COVID 19-ImmunExo, 또는 ImmunExo)은 다양한 농도(0.005, 0.01, 0.05, 0.1, 0.2, 0.5 또는 1ug)로 피하로 주입하였고, 이 때, 엑소좀 입자 수는 동일하게 주입하였다. 이에 대한 대조군으로 마우스에 보조제(adjuvant)와 RBD 단백질을 함께 투여(Adj+RBD) 하였는데, 보조제로는 Al(OH)3를 마우스당 500ug 투여하고 RBD 단백질은 5ug 투여하였다. 이후 하기와 같은 실험들을 통해 다각적인 측면에서 상기 엑소좀의 효과를 평가하였다.
또한, 스파이크 단백질의 RBD 단백질이 로딩된 활성화된 KG-1 세포 유래 엑소좀(COVID 19-ImmunExo, 또는 ImmunExo)은 1ug의 농도로 피하로 주입하였으며, 스파이크 단백질의 RBD mRNA가 로딩된 활성화된 THP-1 세포 또는 KG-1 세포 유래 엑소좀(Exo+RBD mRNA)은 10ug의 농도로 피하로 주입하였다. 이 때, 엑소좀 입자 수는 모두 동일하게 주입하였고, 이에 대한 대조군으로 마우스에 보조제(adjuvant)와 RBD 단백질 또는 mRNA를 함께 투여(Adj+RBD 또는 Adj+RBD mRNA) 하였는데, 보조제로는 Al(OH)3를 마우스당 500ug 투여하고 RBD 단백질은 5ug, RBD mRNA는 10ug 투여하였다. 이후 하기와 같은 실험들을 통해 다각적인 측면에서 상기 엑소좀의 효과를 평가하였다.
5-1. SARS-CoV-2 surrogate virus neutralization test(sVNT)
SARS-CoV-2 surrogate virus neutralization 테스트는 바이러스 중화 과정을 모방한 차단 ELISA 검출 수단으로서, 중화항체에 의한 SARS-CoV-2의 RBD와 인간 ACE2(hACE2) 수용체 단백질의 상호작용 억제 효과에 근거한 테스트이다. 상기 테스트의 검출 민감도는 93.80%이며, 상기 상호작용에 대한 억제 효율(inhibition %)은 하기 수학식 1을 통해 도출하였다. 상기 결과 값의 컷오프(Cutoff)는 20%를 기준으로 하여, 20% 이상인 경우에는 양성(Positive)로써 SARS-CoV-2에 대한 중화항체가 검출된 것으로 해석하였고, 20% 미만인 경우에는 중화항체가 검출되지 않은 것으로 해석하였다.

이에, 마우스에 SARS-CoV-2 유래 스파이크 단백질의 RBD 단백질이 로딩된 활성화된 THP-1 세포 유래 엑소좀(COVID 19-ImmunExo)을 전술한 바와 같이 상이한 농도로 투여하거나 보조제(adjuvant)와 RBD 단백질을 함께 투여(Adj+RBD 5ug)하고 7, 21, 30 및 50일 후에 채혈한 후 혈청을 50배 희석(X50 dilution)하고 상기 수학식을 통해 억제 효율을 계산하였다.
그 결과, 도 3b에 나타낸 바와 같이 상기 엑소좀(ImmunExo)을 투여한 경우 투여 농도에 비례하게 억제 효율이 증가하는 것으로 나타났으며, 엑소좀을 1ug 투여한 경우 보조제(adjuvant) 및 RBD 단백질을 함께 투여한 경우와 매우 유사한 수준의 억제 효율을 확인하였다.
상기 결과에 근거하여, 50일 후에 채취한 혈액에서 혈청을 분리하고 각각 10배 및 50배 희석한 샘플에 대하여 억제 효율을 각각 분석한 결과, 도 3c에서 볼 수 있는 바와 같이 역시 상기 엑소좀(ImmunExo)을 1ug 투여한 경우 보조제(adjuvant) 및 RBD 단백질을 5 ug을 함께 투여한 경우와 매우 유사한 수준의 억제 효율이 나타난 것을 확인하였다.
5-2. SARS-CoV-2에 대한 혈청 항체 반응 분석
(1) SARS-CoV-2 유래 스파이크 단백질의 RBD 단백질이 로딩된 THP-1 세포 유래 엑소좀의 경우
본 발명자들은 상기 실시예 5-1에서 마우스에 SARS-CoV-2 유래 스파이크 단백질의 RBD 단백질이 로딩된 활성화된 THP-1 세포 유래 엑소좀(ImmunExo)을 각각 상이한 농도로 투여하거나 보조제(adjuvant) 및 RBD 단백질을 함께 투여한 각 마우스로부터 50일 후에 혈청 샘플을 얻은 후 1% BSA가 첨가된 PBS에 1:100~1:1000의 비율로 희석하여 SARS-CoV-2의 RBD에 특이적인 혈청 내 항체의 결합 정도(antibody binding)를 흡광도를 측정하여 분석하였다.
그 결과, 도 4a에 나타낸 바와 같이 모든 군에서 마우스유래 혈청을 희석할수록 항체의 결합 정도가 동일한 양상으로 감소하였고, 상기 엑소좀의 투여 농도에 대체적으로 비례하게 나타났다. 또한, 전반적인 항체의 결합은 상기 엑소좀을 1ug 투여한 경우 가장 높게 나타났고, 0.5ug 투여한 경우 보조제(adjuvant)와 RBD 단백질을 함께 투여한 군(Adj+RBD 5ug)과 유사하게 나타난 것을 확인하였다.
(2) SARS-CoV-2 유래 스파이크 단백질의 RBD 단백질이 로딩된 KG-1 세포 유래 엑소좀의 경우
본 발명자들은 상기 실시예 3에서 제조한 마우스에 SARS-CoV-2 유래 스파이크 단백질의 RBD 단백질이 로딩된 활성화된 KG-1 세포 유래 엑소좀(ImmunExo)을 1 ug 농도로 투여하거나 보조제(adjuvant) 및 RBD 단백질 1 ug을 함께 투여한 각 마우스로부터 7일 후에 혈청 샘플을 얻은 후 1% BSA가 첨가된 PBS에 1:100~1:1000의 비율로 희석하여 SARS-CoV-2의 RBD에 특이적인 혈청 내 항체의 결합 정도(antibody binding)를 흡광도를 측정하여 분석하였다.
그 결과, 도 4b에 나타낸 바와 같이 대조군 대비 SARS-CoV-2의 RBD에 특이적인 혈청 내 항체의 결합(antibody binding) 정도가 나타남을 확인할 수 있었다.
(3) SARS-CoV-2 유래 스파이크 단백질의 RBD mRNA가 로딩된 THP-1 세포 유래 엑소좀의 경우
본 발명자들은 상기 실시예 4에서 제조한 마우스에 SARS-CoV-2 유래 스파이크 단백질의 RBD mRNA가 로딩된 활성화된 THP-1 세포 유래 엑소좀(Exo+RBD mRNA)을 10 ug 농도로 투여하거나 보조제(adjuvant) 및 RBD mRNA 10ug을 함께 투여한 각 마우스로부터 7일 및 14일 후에 혈청 샘플을 얻은 후 1% BSA가 첨가된 PBS에 1:100~1:1000의 비율로 희석하여 SARS-CoV-2의 RBD에 특이적인 혈청 내 항체의 결합 정도(antibody binding)를 흡광도를 측정하여 분석하였다.
그 결과, 도 4c에 나타낸 바와 같이 7일 후에는 모두 SARS-CoV-2의 RBD에 특이적인 혈청 내 항체의 결합(antibody binding) 정도를 측정할 수 없었으나, 14일 후에는 보조제(adjuvant)와 RBD mRNA를 함께 투여한 군 또는 대조군 대비 현저하게 높은 SARS-CoV-2의 RBD에 특이적인 혈청 내 항체의 결합(antibody binding) 정도가 나타남을 확인할 수 있었다.
(4) SARS-CoV-2 유래 스파이크 단백질의 RBD mRNA가 로딩된 KG-1 세포 유래 엑소좀의 경우
본 발명자들은 상기 실시예 4에서 제조한 마우스에 SARS-CoV-2 유래 스파이크 단백질의 RBD mRNA가 로딩된 활성화된 KG-1 세포 유래 엑소좀(Exo+RBD mRNA)을 10 ug 농도로 투여하거나 보조제(adjuvant) 및 RBD mRNA 10ug을 함께 투여한 각 마우스로부터 7일 및 14일 후에 혈청 샘플을 얻은 후 1% BSA가 첨가된 PBS에 1:100~1:1000의 비율로 희석하여 SARS-CoV-2의 RBD에 특이적인 혈청 내 항체의 결합 정도(antibody binding)를 흡광도를 측정하여 분석하였다.
그 결과, 도 4d에 나타낸 바와 같이 7일 후에는 다른 군과 달리, SARS-CoV-2 유래 스파이크 단백질의 RBD mRNA가 로딩된 활성화된 KG-1 세포 유래 엑소좀(Exo+RBD mRNA) 군만이 SARS-CoV-2의 RBD에 특이적인 혈청 내 항체의 결합(antibody binding) 정도를 측정할 수 있었고, 14일 후에는 보조제(adjuvant)와 RBD mRNA를 함께 투여한 군 또는 대조군 대비 현저하게 높은 SARS-CoV-2의 RBD에 특이적인 혈청 내 항체의 결합(antibody binding) 정도가 나타남을 확인할 수 있었다.
5-3. SARS-CoV-2 RBD 특이적 T 세포 반응 분석
본 발명자들은 상기 실시예 5-1에서 SARS-CoV-2 유래 스파이크 단백질의 RBD 단백질이 로딩된 활성화된 THP-1 세포 유래 엑소좀(ImmunExo)을 투여하여 면역화시킨 마우스로부터 비장세포(splenocyte)를 분리하고 이에 대하여 IFN-γ ELISPOT(Enzyme-linked immunospot) 어세이를 실시해 SARS-CoV-2 RBD 특이적 T 세포 반응을 분석하고자 하였다. 상기 IFN-γ ELISPOT 어세이는 단일세포 수준에서 사이토카인을 분비하는 항원 특이적 T 세포를 검출하는 민감한 기술로서, Th1 세포 매개 면역반응을 직접적으로 측정할 수 있어 세포 매개 면역을 유도하는 백신의 효과를 모니터링하는데 유용하며, 비색반응을 통해 생성되는 반점의 수를 확인함으로써 사이토카인 생성 T 세포의 빈도를 직접 측정할 수 있다.
상기 어세이를 실시하기 위해, 상기 실시예 5-1의 각 마우스 유래 비장세포를 5 x 105/well로 플레이트의 각 웰에 분주하고 SARS-CoV-2 유래 스파이크 단백질의 RBD 단백질을 2ug/mL 농도로 각각 처리한 후 24시간 동안 배양하였다.
그 결과, 도 5a에 나타낸 바와 같이 보조제(adjuvant)와 RBD 단백질을 함께 투여한 군(Adj+RBD 5ug)에서는 반점이 전혀 나타나지 않은 반면, 본 발명에 따른 엑소좀(COVID19-ImmunExo)을 투여한 군에서는 투여 농도에 따라 반점 수의 차이는 있으나 모두 반점이 생성된 것으로 나타났으며, 특히 상기 엑소좀을 0.5ug 투여한 군에서 반점의 수가 가장 많은 것을 확인하였다.
나아가 상기와 동일한 방법으로 본 발명에 따른 엑소좀 또는 보조제와 RBD 단백질을 함께 투여한 마우스 유래 비장세포(1 x 107/well)에 SARS-CoV-2 유래 스파이크 단백질의 RBD 단백질을 2ug/mL 48시간 동안 처리한 후 상기 비장세포의 배양액을 이용해 ELISA를 실시하여 Th1 세포로부터 분비된 IFN-γ 단백질 수준을 측정하였다.
그 결과, 도 5b에서 볼 수 있는 바와 같이 상기 도 5a의 결과와 부합하게 보조제와 RBD 단백질을 함께 투여한 마우스 유래 비장세포의 배양액에서는 IFN-γ가 전혀 존재하지 않는 것으로 나타난 반면, 본 발명에 따른 엑소좀(ImmunExo)을 투여한 마우스 유래 비장세포의 배양액에는 IFN-γ가 존재하는 것을 확인하였다.
상기와 같은 결과는 보조제와 RBD 단백질을 함께 투여한 경우에는 Th1 세포 매개 면역반응이 유도되지 않은 반면, 본 발명에 따른 엑소좀 투여에 의해 강한 Th1 세포 매개 면역반응이 유도되었음을 시사하는 것이다.
실시예 6. 동결건조된 COVID19-ImmunExo의 저장 안정성 및 효과 검증
본 발명자들은 본 발명에 따른 SARS-CoV-2 유래 스파이크 단백질의 RBD 단백질이 로딩된 활성화된 THP-1 세포 유래 엑소좀(COVID19-ImmunExo)의 임상에서의 이용 가능성을 확인하기 위해, 상기 엑소좀의 저장 안정성을 검증하였다.
보다 구체적으로, 도 6a에 도시된 일정에 따라 동결건조시킨 본 발명의 엑소좀을 농도별로 1회 투여할 분량으로 제조하여 튜브에 나누어 담아 -70℃에서 하루 동안 냉동한 후에 동결건조를 수행하였다. 동결건조된 엑소좀을 4℃(도 6b) 또는 상온(도 6c)에서 7일 동안 보관한 후 다시 투여 제형으로 제조한 엑소좀을 각 마우스에 제시된 농도로 각각 투여하였다. 이후 7일 및 21일째에 상기 실시예 5-1에서와 동일한 방법으로 SARS-CoV-2 sVNT 테스트를 실시하였다.
그 결과, 도 6b에 나타낸 바와 같이 상기 4℃에서 7일간 보관한 동결건조된 엑소좀을 0.1ug, 0.5ug, 1ug 농도로 투여한 경우, 7일째부터 엑소좀을 1ug 투여한 것에서 억제효과를 보였으며, 21일째에는 엑소좀을 0.5ug 이상의 농도로 투여한 경우 21일째에 약 50%에 근접하는 억제 효율을 확인하였다. 또한 도 6c에 나타낸 바와 같이 상온에서 7일간 보관한 동결건조된 엑소좀을 1ug 농도로 투여한 경우, 7일째부터 억제 효과를 보였으며 21일째에는 약 40%에 근접하는 억제 효율을 확인할 수 있었다. 이러한 결과는 동결건조시키고 7일간 보관하지 않은 엑소좀(fresh)과 비교할 때 동일한 조건에서 억제 효율이 유사한 것을 통해 항체 생성 효과가 유사한 것을 알 수 있었으며, 따라서 본 발명에 따른 상기 엑소좀을 상기 조건으로 저장한 경우에도 안정적으로 그 효과가 유지되는 것을 알 수 있었다.
전술한 본 발명의 설명은 예시를 위한 것이며, 본 발명이 속하는 기술분야의 통상의 지식을 가진 자는 본 발명의 기술적 사상이나 필수적인 특징을 변경하지 않고서 다른 구체적인 형태로 쉽게 변형이 가능하다는 것을 이해할 수 있을 것이다. 그러므로 이상에서 기술한 실시예들은 모든 면에서 예시적인 것이며 한정적이 아닌 것으로 이해해야만 한다.
Claims (14)
- 코로나바이러스(coronavirus) 유래 항원 단백질 또는 상기 단백질을 암호화하는 유전자를 포함하는 세포외소포체.
- 청구항 1에 있어서, 상기 코로나바이러스는 SARS-CoV-2(Severe Acute Respiratory Syndrome-Coronavirus-2)인 것인, 세포외소포체.
- 청구항 1에 있어서, 상기 항원 단백질은 코로나바이러스 유래 스파이크 단백질(spike protein)의 수용체 결합 도메인(Receptor Binding Domain; RBD) 단백질인, 세포외소포체.
- 청구항 1에 있어서, 상기 항원 단백질 또는 상기 단백질을 암호화하는 유전자는 상기 세포외소포체 내 탑재된 것인, 세포외소포체.
- 청구항 1에 있어서, 상기 세포외소포체는 동물세포, 식물세포 또는 미생물로 이루어진 군에서 선택되는 하나 이상으로부터 유래된 것인, 세포외소포체.
- 청구항 5에 있어서, 상기 동물세포는 체세포, 생식세포, 면역세포, 신경세포 및 종양세포로 이루어진 군에서 선택되는 하나 이상의 세포인 것인, 세포외소포체.
- 청구항 1에 있어서, 상기 세포외소포체는 활성화된 면역세포 유래인 것인, 세포외소포체.
- 청구항 1에 있어서, 상기 세포외소포체는 CD63, CD81 및 Flotillin 1으로 이루어진 군에서 선택되는 하나 이상을 발현하는 것인, 세포외소포체.
- 청구항 1의 세포외소포체를 포함하는 코로나바이러스 감염증 예방 또는 치료용 백신 조성물.
- 청구항 9에 있어서, 상기 백신 조성물은 항원 특이적 항체 생성 및 T 세포 매개 면역 반응을 동시에 유도하는 것인, 백신 조성물.
- 청구항 9에 있어서, 상기 백신 조성물은 동결건조된 것인, 백신 조성물.
- 청구항 1의 세포외소포체를 포함하는 코로나바이러스 감염증 예방 또는 개선용 건강기능식품.
