Abstract
본 발명은 알로페론-피브로인 복합체에 관한 것으로, 피브로인과 복합체를 이루어 알로페론의 혈청을 포함한 수용상 내 안정성이 증가하여 낮은 함량으로 체내 투여되어도 우수하고 장기적인 면역 증진 효과를 갖는다.
Description
본 발명은 알로페론과 피브로인의 복합체에 관한 것으로서, 알로페론이 피브로인과 복합체를 이루어 알로페론의 혈청을 포함한 수용상 내 안정성이 개선되고 면역증진 활성이 보다 우수한 특징을 갖는다.
파리 유출 단계를 거치는 곤충은 항세균 또는 항진균 펩티드를 포함하는 여러 종류의 면역 유발 분자의 생산을 통하여 미생물 감염에 대응하는 것으로 알려져 있다. 알로페론(alloferon)은 이러한 감염된 곤충으로부터 분리된 펩티드를 연구하는 과정에서 발견된 면역조절 단백질이다. 알로페론은 항바이러스 및 항암효과를 나타내는 것으로 알려져 있는데, 구체적으로 2B4와 같은 NK 세포 활성 수용체의 발현을 증가시켜 NK 세포의 세포 살해 활성을 증가시키고, IFN-γ와 TNF-α의 생산을 증가시키고, NK 세포로부터의 과립 엑소시토시스(exocytosis)를 증가시켜 암세포 살해 활성을 증가시킨다. (Bae et al., Immunobiology, Vol. 218, Issue 8, August 2013, 1026-1033). 알로페론은 몇 가지의 아미노산 서열을 갖고, 각 아미노산 마다 체내 활성 효과 및 정도는 다르지만 공통적으로 면역 조절 기능을 갖는 것으로 알려져 있다.
그러나, 알로페론은 펩티드이므로 혈청을 포함한 수용상 내 안정성이 낮아 투여 후 빠른 시간 내 비활성화 되어 실제 기대할 수 있는 효과에 비해 과량으로 사용하여야 하는 문제점을 갖는다. 물, 생리식염수 및 혈청을 포함하는 수용상은 펩티드의 구조를 변형시켜 활성을 저해할 수 있으며, 특히 혈청의 경우 포함된 단백질, 효소, 지질 및 화학성분이 다른 수용상에 비해 월등히 많아 특히 펩티드의 활성을 저해하는 효과가 크다. 따라서 혈청에서 안정한 경우 물과 생리식염수를 포함한 수용상에서 펩티드의 안정성을 높이는 방법일 수 있다. 혈청 내에서 불안정한 알로페론의 단점은 알로페론의 우수한 효능에도 불구하고 의약이나 미용 또는 건강식품 산업에 적용하는데 장벽이 된다.
한국등록특허공보 10-0394864는 알로페론의 면역조절 기능을 개시한다. 그러나 알로페론을 포함한 펩티드의 혈청을 포함한 수용상 내 안정성을 증진시킬 경우 이러한 면역 조절기능을 더욱 증대시킬 수 있을 것이나 알로페론의 혈청을 포함한 수용상 내 안정성을 증가시킬 수 있는 방법은 개시하지 않는다.
본 발명의 일 목적은, 혈청을 포함한 수용상 내 안정성이 증진된 알로페론-피브로인 복합체를 제공하는 것이다.
본 발명의 다른 목적은, 알로페론-피브로인 복합체를 유효성분으로 포함하는 면역증진용 약학적 조성물과 건강기능식품을 제공하는 것이다.
본 발명의 또 다른 목적은, 알로페론-피브로인 복합체를 유효성분으로 포함하는 항바이러스 약학적 조성물과 건강기능식품을 제공하는 것이다.
본 발명의 또 다른 목적은, 상기 알로페론-피브로인 복합체를 유효 성분으로 포함하는 면역증진용 화장료 조성물을 제공하는 것이다.
본 발명의 또 다른 목적은, 알로페론-피브로인 복합체를 제조하는 방법을 제공하는 것이다.
상기 목적을 해결하기 위해, 일 양상은 알로페론 및 피브로인을 포함하는 복합체를 제공한다.
상기 알로페론은 통상적으로 알려진 아미노산 서열을 갖는 폴리펩티드일 수 있다. 알로페론의 아미노산 서열은 예를 들어, 서열번호 5를 포함하는 것으로서, 서열번호 5의 아미노산 서열에서 N-말단 및 C 말단에 각각 0 내지 5 개의 아미노산을 더 포함하는 아미노산 서열로 이루어질 수 있다. 보다 구체적으로, 상기 알로페론은 서열번호 1, 서열번호 2, 서열번호 3, 서열번호 4, 및 서열번호 5로 이루어진 군으로부터 선택된 아미노산 서열을 갖는 폴리펩티드 또는 이들의 혼합물일 수 있다.
상기 피브로인은 천연 유래 생체물질로서, 누에(Bombyx mori)로 대표되는 견사충의 유충 견사샘에서 합성하는 섬유단백질로 분자 구조는 6개의 소수성 아미노산이 반복적으로 배열된 소수성 부위와 비교적 짧은 친수성 부위의 반복으로 이루어져 있으며, 소수성 부위는 자발적으로 안정화되기 위해 물리적 가교를 통한 베타-시트 구조를 기반으로 하는 단백질 결정구조를 형성할 수 있는 성질을 가지고 있다. 이러한 소수성 부위는 알라닌, 글라이신-알라닌, 글라이신-알라닌-세린과 같은 소수성 아미노산 또는 이러한 아미노산의 짧은 반복구조로 이루어져 있으며, 피브로인 단백질의 pI(isoelectirc point) 값은 약 4이고, 친수성을 띠는 하전된 아미노산은 대부분 N-과 C-말단 쪽에 위치하고 있다.
상기 알로페론은 알로페론 1 몰당 피브로인 2몰과 복합체를 이룰 수 있다. 알로페론-피브로인 복합체는 정전기적 결합으로 결합된 것일 수 있다.
상기 알로페론-피브로인 복합체는 300 내지 450 kDa일 수 있고, 바람직하게는 350 내지 400 kDa의 분자량을 갖는 것일 수 있다.
상기 알로페론-피브로인 복합체의 표면전하는 음전하로서, 예를 들어 -30 mV 내지 -10 mV, -25 mV 내지 -10 mV, 또는 -20 mV 내지 -15 mV 일 수 있다.
일 구체예에 따른 알로페론-피브로인 복합체는 면역활성을 증진시킬 수 있다. 상기 면역활성의 증진이란, T세포 또는 NK 세포의 세포독성활성을 증진시키는 것을 의미하고, 면역활성 증진에 따라 항바이러스 활성, 항박테리아 활성, 또는 암세포 살상능을 가질 수 있다.
다른 양상은 알로페론-피브로인 복합체를 유효성분으로 함유하는 면역증진용 약학적 조성물을 제공한다.
일 구체예에서, 상기 면역증진용 약학적 조성물은 암을 예방 또는 치료하기 위한 것일 수 있다.
상기 알로페론-피브로인 복합체는 복합체를 이루어도 T 세포 및 NK 세포의 세포살상능을 증진시키는 정도가 알로페론을 단독으로 이용한 경우에 비해 현저하게 우수하고, 그에 따라 우수한 암세포 살상능을 가짐을 확인하였다. 상기 알로페론-피브로인 복합체는 알로페론의 단독 대비 1.2 배 이상, 1.5 배 이상, 2 배 이상 또는 2.5 배 이상의 면역세포의 세포사멸능을 증진시키는 것일 수 있고, 예를 들어, T림프구의 세포사멸 효과를 500 ng/ml 농도를 기준으로 약 3 배 증진시킬 수 있는 것일 수 있다.
또 다른 양상은 상기 알로페론-피브로인 복합체를 유효성분으로 함유하는 면역증진용 건강기능식품을 제공한다.
일 구체예에 따른 알로페론-피브로인 복합체는 혈청을 포함한 수용상 내 안정성이 우수하므로 적은 양으로도 우수한 면역증진 효과를 나타내고, 부작용이 적어 약학적 조성물이나 건강기능식품의 유효성분으로서 적합하다.
또 다른 양상은 상기 알로페론-피브로인 복합체를 유효성분으로 함유하는 항바이러스 약학적 조성물을 제공한다.
또 다른 양상은 상기 알로페론-피브로인 복합체를 유효성분으로 함유하는 항바이러스 건강기능식품을 제공한다.
일 구체예에서, 상기 항바이러스 약학적 조성물 및 건강기능식품을 이용하여, 바이러스 감염으로 인한 질환을 예방하거나 치료할 수 있다.

상기 알로페론-피브로인 복합체를 포함하는 약학적 조성물, 건강기능식품, 또는 화장료 조성물에서, 유효 성분은 알로페론이고, 피브로인은 담체로서 작용하는 것일 수 있다. 피브로인이 담체로서 작용하는 경우, 알로페론을 유리한 후 체내에서 효소 작용에 의해 쉽게 분해되어 배설될 수 있다.
또 다른 양상은, 상기 알로페론-피브로인 복합체를 유효 성분으로 포함하는 면역증진용 화장료 조성물을 제공한다. 상기 화장료 조성물은 진피균이나 바이러스에 의한 피부 감염증을 예방 또는 치료할 수 있다.
상기 조성물을 약학적 조성물로 이용하는 경우, 상기 조성물은 약학적으로 허용되는 담체를 더 포함할 수 있다. 상기 약학적으로 허용되는 담체는 예를 들어, 락토스, 덱스트로스, 수크로스, 솔비톨, 만니톨, 전분, 아카시아 고무, 인산 칼슘, 알기네이트, 젤라틴, 규산 칼슘, 미세결정성 셀룰로스, 폴리비닐피롤리돈, 셀룰로스, 물, 시럽, 메틸 셀룰로스, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 활석, 스테아르산 마그네슘 및 미네랄 오일과 같은 통상적으로 이용되는 것일 수 있으나, 이에 제한되지 않는다.
또한, 상기 약학적 조성물은 윤활제, 습윤제, 감미제, 향미제, 유화제, 현탁제, 보존제 등을 추가로 포함할 수 있다.
상기 약학적 조성물은 경구, 비경구 투여 또는 피부패치로 사용할 수 있으며, 비경구 투여인 경우에는 정맥내 주입, 피하 주입, 근육 주입, 복강 주입, 경피 투여, 점막 투여 및 점안 투여 등으로 투여할 수 있고, 적합한 투여량은 제형, 투여 방식, 환자의 연령, 체중, 성, 병적 상태, 음식, 투여 시간, 투여 경로, 배설 속도 및 반응 감응성과 같은 요인들을 고려하여 전문가의 지도하여 적절히 조절될 수 있다.
상기 약학적 조성물은 투여 목적에 따라 다양한 제형으로 제공될 수 있고, 예를 들어, 스프레이, 에어로졸, 오일 또는 수성 매질중의 용액, 현탁액, 시럽제 또는 유화액 형태이거나, 엑스제, 산제, 분말제, 과립제, 정제 또는 캅셀제가 있으나, 이에 제한되지 않는다. 상기 조성물이 스프레이 또는 에어로졸인 경우에는 추가적으로 클로로플루오로히드로카본, 질소, 프로판/부탄 또는 디메틸 에테르와 같은 추진체를 포함할 수 있으나, 이에 제한되지 않는다.
상기 건강기능식품의 예로는, 영양 보조제, 영양제, 파머푸드(pharmafood), 건강식품, 뉴트라슈티칼(nutraceutical), 디자이너 푸드, 식품 첨가제 등이 있으나 이에 제한되지 않는다. 상기 복합체가 식품 첨가제로 사용되는 경우, 식품으로는, 육류, 소세지, 빵, 쵸코렛, 캔디류, 스넥류, 과자류, 피자, 라면, 기타 면류, 껌류, 아이스크림류를 포함한 낙농제품, 각종 스프, 음료수, 차, 드링크제, 알콜 음료 및 비타민 복합제 등이 있으나 이에 제한되지 않는다.
상기 화장료 조성물은 스킨, 세럼, 로션, 에멀전, 크림, 에센스, 화장수, 파운데이션, 팩, 비누, 계면활성제-함유 클린싱, 스프레이, 또는 파우더의 제형일 수 있으나, 이에 제한되지 않는다.
상기 화장료 조성물은 피부 흡수 속도를 증진시키고 사용감을 개선하기 위해 적절한 담체를 더 포함할 수 있다. 예를 들어, 상기 화장료 조성물이 페이스트, 크림 또는 겔인 경우에는 담체 성분으로서 동물성유, 식물성유, 왁스, 파라핀, 전분, 트라칸트, 셀룰로오스 유도체, 폴리에틸렌 글리콜, 실리콘, 벤토나이트, 실리카, 탈크 또는 산화아연 등을 더 포함할 수 있으나 이에 제한되지 않는다.
상기 화장료 조성물이 파우더 또는 스프레이인 경우에는 담체 성분으로서 락토스, 탈크, 실리카, 알루미늄 히드록시드, 칼슘 실리케이트 또는 폴리아미드 파우더가 이용될 수 있고, 특히 스프레이인 경우에는 추가적으로 클로로플루오로히드로카본, 프로판/부탄 또는 디메틸 에테르와 같은 추진체를 포함할 수 있으나, 이에 제한되지 않는다.
상기 화장료 조성물이 용액 또는 유탁액인 경우에는 담체 성분으로서 용매, 용해화제 또는 유탁화제가 이용될 수 있고, 예컨대 물, 에탄올, 이소프로판올, 에틸 카보네이트, 에틸 아세테이트, 벤질 알코올, 벤질 벤조에이트, 프로필렌글리콜, 1,3-부틸글리콜 오일, 글리세롤 지방족 에스테르, 폴리에틸렌 글리콜 또는 소르비탄의 지방산 에스테르가 있으나 이에 제한되지 않는다.
상기 화장료 조성물이 현탁액인 경우에는 담체 성분으로서 물, 에탄올 또는 프로필렌 글리콜과 같은 액상의 희석제, 에톡실화 이소스테아릴 알코올, 폴리옥시에틸렌 소르비톨 에스테르 및 폴리옥시에틸렌 소르비탄 에스테르와 같은 현탁제, 미소결정성 셀룰로오스, 알루미늄 메타히드록시드, 벤토나이트, 아가 또는 트라칸트 등이 이용될 수 있으나 이에 제한되지 않는다.
상기 화장료 조성물이 클렌징용인 경우에는 담체 성분으로서 지방족 알코올 설페이트, 지방족 알코올 에테르 설페이트, 설포숙신산 모노에스테르, 이세티오네이트, 이미다졸리늄 유도체, 메틸타우레이트, 사르코시네이트, 지방산 아미드 에테르 설페이트, 알킬아미도베타인, 지방족 알코올, 지방산 글리세리드, 지방산 디에탄올아미드, 식물성 유, 라놀린 유도체 또는 에톡실화 글리세롤 지방산 에스테르 등이 이용될 수 있으나 이에 제한되지 않는다.
또 다른 양상은 피브로인을 수득하는 단계; 상기 피브로인과 알로페론을 혼합하여 복합체를 형성하는 단계; 상기 형성된 복합체를 유기용매로 세척하는 단계를 포함하는 알로페론-피브로인 복합체의 제조 방법을 제공한다.
상기 피브로인과 알로페론은 각각 수성용액으로 제조한 후 혼합할 수 있고, 수성용액이란 물, 식염수, 완충액 등의 용액에 피브로인 또는 알로페론을 용해시킨 것을 의미할 수 있다. 복합체를 형성하는 단계에서는 제조시간이 길어져 펩티드가 변성되거나 분해되지 않도록 하기 위해, 알로페론과 피브로인을 진공 상태에서 혼합할 수 있고, 예를 들어, 1 내지 4 bar의 진공상태에서 약 5 - 7 시간 동안 혼합하고 반응할 수 있도록 10 내지 20 ℃에서 예치하는 것일 수 있다.
상기 복합체를 형성한 후에는 복합체를 형성하지 않은 알로페론 및 피브로인을 제거하기 위해 유기용매로 세척할 수 있다. 상기 유기용매는 에탄올, 프로판올, 이소프로판올, 아세톤, 아세토니트릴, 클로로포름 또는 이들의 조합일 수 있으나 이에 제한되지 않는다.
형성된 복합체를 세척한 후에는, 순도를 높이기 위해 유기용매를 제거하는 건조 단계를 포함할 수 있다. 상기 건조 단계는 승화법을 이용할 수 있고, 선택적으로 동결건조제를 혼합하고 동결건조하여 유기용매를 제거하면서 보관안정성을 더욱 높일 수 있다.
일 양상에 따른 알로페론-피브로인 복합체는 혈청을 포함한 수용상 내 안정성이 높아서 체내에 투여 가능하고, 투여량에 비해 높은 효과를 얻을 수 있다. 따라서 알로페론을 의약, 건강기능식품, 또는 화장료 조성물로 사용 가능하도록 한다.

이하, 본 발명의 이해를 돕기 위하여 실시예 등을 들어 상세하게 설명하기로 한다. 그러나, 본 발명에 따른 실시예들은 여러 가지 다른 형태로 변형될 수 있으며, 본 발명의 범위가 하기 실시예들에 한정되는 것으로 해석되어서는 안 된다. 본 발명의 실시예들은 당업계에서 평균적인 지식을 가진 자에게 본 발명을 보다 완전하게 설명하기 위해 제공되는 것이다.

누에고치를 잘게 조각낸 후, 0.02 M 탄산나트륨 수용액으로 90℃에서 20 분간 정련하고, 표면에 부유하는 것을 제거하고, 20 분간 세척하는 단계를 2회 수행한 후 세레신과 탄산나트륨을 제거하여 실크 피브로인을 얻었다. 상기 실크 피브로인 견사를 12 시간 동안 후드에서 건조시키고, 9.3M LiBr 용액에서 60℃에서 4 시간 동안 용해시킨 후, Slide-a-Lyzer dialysis cassette (MWCO-3500)를 이용하여 상온에서 3일 동안 증류수를 6 번 교체하며 투석하였다. 투석 과정을 통해 얻은 실크 피브로인 용액을 5 ℃에서 원심분리하여 용해되지 않은 잔여물을 제거함으로써 높은 순도의 실크 피브로인 단백질 용액을 수득하였다. 수득한 실크 피브로인은 순도가 약 99%이고, 수득율은 약 93% 정도였다. 수득한 실크 피브로인 용액은 2mM 농도의 수용액으로 준비하였다. 알로페론은 de novo peptide synthesis 방법으로 하기의 표 1과 같은 서열로 합성하고, 각각 0.2 내지 20 mM 농도의 수용액으로 준비하였다.

각 준비된 피브로인과 알로페론을 혼합하고 상온에서 4 bar 이하의 진공 상태에서 6 시간 동안 복합체가 형성되도록 하였다. 복합체 형성 반응 후 100 % 에틸알코올로 2~3회 세척하고, 물에 녹여서 복합체의 구조를 확인하였다. 복합체 형성 반응에서 사용된 피브로인과 알로페론의 혼합비는 하기의 표 2와 같다. 표시된 비율은 피브로인 1 몰에 대한 알로페론의 몰비로서, 피브로인 농도를 2mM로 고정하고 알로페론의 농도를 조절하면서 몰비를 맞추었다. 에틸알코올로 세척 후 물에 녹인 샘플을 SDS-PAGE 겔 전기영동하여 분리하고, 피브로인 항체와 알로페론 항체를 처리하여 두 단백질이 모두 검출되는 경우를 구분함으로써 복합체가 형성되었는지 여부를 확인하여 하기의 표 2에 복합체의 형성 정도를 나타내었다.

[시험예 1] T림프구의 암세포에 대한 세포독성활성에 대한 효과 확인
상기 제조예 1 중 알로페론 1을 함유하고 피브로인 및 알로페론의 몰비가 2:1인 것을 실시예 1로하여 T림프구의 암세포에 대한 세포독성활성을 확인하였다.
구체적으로, 면역화된 BALB/c 마우스로부터 비장을 분리하고 페트리 디쉬에 시린지를 이용하여 균일화하고, 정제수에 현탁한 후 70㎛ 나일론 필터를 이용해 여과하고 원심분리하여 비장 세포를 얻었다. 수득된 비장 세포에 알로페론과 상동성이 없는 랜덤서열의 펩티드(음성 대조군), 알로페론 1 펩티드, 및 실시예 1을 각각 0.5 ng/ml 내지 500 ng/ml로 처리하고 1 시간 인큐베이션 한 후, 암세포인 K562-s 세포에 처리하고 암세포의 세포 사멸율(%)을 측정하여 하기의 표 3 및 도 2에 나타내었다.

[시험예 2] 말초혈액림프구의 NK세포 활성 유도능 확인
상기 실시예 1을 이용하여 NK 세포 활성에 의한 암세포 사멸능을 평가하였다.
구체적으로, 사람의 혈액을 채취하여 원심분리하고 말초 혈액 단핵세포(peripheral blood mononuclear cell, PBMC)를 분리하였다. 그런 다음 유세포 분석기를 이용하여 NK 세포만을 분리한 다음, 알로페론과 상동성이 없는 랜덤서열의 펩티드(음성 대조군), 알로페론 1 펩티드, 및 실시예 1을 각각 0.5 ng/ml 내지 500 ng/ml로 처리하고 1 시간 인큐베이션한 후, 암세포인 K562-s 세포에 상기 NK 세포를 처리하여 암세포의 세포 사멸율(%)을 측정하여 하기의 표 4 및 도 3에 나타내었다. 양성대조군으로는 INF-a2b를 처리하였다.

[시험예 3] 항바이러스 활성 확인
상기 실시예 1을 이용하여 인간 인플루엔자 바이러스 A에 대한 항바이러스 활성을 평가하였다.
구체적으로, BALB/c 마우스를 준비하고 마취한 다음 인간 항바이러스 A를 비점막을 통해 감염시키고, 각 처리 군 및 농도에 따라 20마리씩 준비하고, 알로페론과 상동성이 없는 랜덤서열의 펩티드(음성 대조군), 알로페론 1 펩티드, 및 실시예 1을 각각 125㎍/kg 및 1,250㎍/kg 으로 복강 투여하였다. 그런 다음, 10일 간 지켜보면서 사망한 개체 수를 세어 하기의 표 5 및 도 4에 나타내었다. 양성대조군에는 독감 치료제인 리만타딘 1mg을 투여하였다.

[시험예 4] 바이러스 초기 면역 효과 확인
상기 실시예 1을 이용하여 인간 인플루엔자 바이러스 A에 대한 초기 면역 효과를 확인하였다.
구체적으로, BALB/c 마우스를 준비하고 마취한 다음 인간 항바이러스 A를 비점막을 통해 감염시키고, 각 처리 군 및 농도에 따라 8마리씩 준비하고, 알로페론과 상동성이 없는 랜덤서열의 펩티드(음성 대조군), 알로페론 1 펩티드, 및 실시예 1을 각각 125㎍/kg 으로 복강 투여하였다. 그런 다음, 24시간 후 혈액을 채취하여 인터페론(IFN) 존재량 (unit)을 측정하여 하기의 표 6 및 도 5에 나타내었다. 양성대조군에는 사이클로페론 1mg을 투여하였다.

알로페론 구조에 따른 면역 유도능의 차이를 확인하기 위해, 표 1과 같은 알로페론을 이용하되 실시예 1과 같이 복합체를 제조하고, 알로페론 1 내지 5를 포함하는 복합체를 각각 실시예 1 내지 5로 명명하였다. 그런 다음 시험예 2와 같이 NK세포의 암세포에 대한 세포독성활성을 평가하여 하기의 표 7에 나타내었다.

[시험예 6] 알로페론-피브로인 복합체의 독성 평가
(1) 급성독성 평가
본 발명의 복합체가 체내에서 급성독성을 갖는지 여부를 마우스를 이용해 평가하였다.
구체적으로, BALB/c 마우스를 각 농도 별로 다섯 마리 준비하고 실시예 1을 500~6,000mg/kg 농도로 복강 단 회 투여하고, 10일간 지켜보았다. 그 결과, 가장 높은 투여량을 포함한 어떠한 투여량에서도 폐사하지 않았고, 성장 장애, 식이 장애, 뇌의 구조 변화, 주입 부위의 괴사, 내분비기관의 변화 등이 관찰되지 않았다. 동물 시험 결과, 약리독성 용량비는 1>1,200,000 (50㎍)으로 평가되었다.
(2) 아급성 및 만성독성 평가
본 발명의 복합체가 체내에서 아급성 또는 만성독성을 갖는지 여부를 마우스를 이용해 평가하였다.
구체적으로, BALB/c 마우스를 각 농도 별로 다섯 마리 준비하고 실시예 1을 각각 0.5, 5, 및 50 mg/kg로 90일 동안 이틀에 한 번씩 투여하여 아나필락시스 쇼크, 면역복합반응, 지방세포탈과립화의 간접적 반응, 결막탐색자(conjunctive probe), 및 지연성 과민증 반응을 관찰하였다.
그 결과, 90일 동안 폐사한 시험 동물은 없었고, 성장 장애, 식이 장애, 직장의 온도, 검안(점막표면상태 및 눈의 형태), 신경정신(흥분한계치, 자발적 운동활성), 심혈관(심장수축기 동맥압, 심장박동율), 간, 신장(오줌량), 혈액학적 지수(hemogram, leukocyte 조성,coagulogram), 말초혈액의 생화학적지수(단백질, 요소, 크레아틴, 포도당, 지질, 콜레스테롤, 빌리루빈, Alp, Alk, 전해질), 혈액형성지수, 병리학 및 조직학(심장, 폐, 기관지, 위, 간, 비장, 신장, 뇌, 정소, 난소)적으로 개체기능의 음성적 변화가 관찰되지 않았다. 또한 치료 용량의 10 배 및 100 배에 해당하는 용량에서도 어떠한 징후가 관찰되지 않았다.
(3) 배아 독성
본 발명의 복합체가 배아독성을 갖는지 여부를 마우스를 이용해 평가하였다.
구체적으로, 임신한 BALB/c 마우스를 각 농도 별로 세 마리 준비하고 실시예 1을 0.5, 5, 및 50 mg/kg의 농도로 피하 주사하고, 출산한 새끼를 관찰하였다.
그 결과, 모든 투여 군에서 출산한 새끼가 정상인 것을 확인하였다.
(4) 돌연변이 유발성
본 발명의 복합체가 돌연변이를 유발할 위험이 있는지 박테리아를 이용하여 AMES 시험(Bacterial reverse mutation test, 복귀돌연변이시험)에 따른 DNA SOS반응을 평가하였다.
구체적으로, 살모넬라 티피무리움(S. typhimurium)과 대장균(PQ37) 균주를 준비하고, 0.5~500 ㎍/dish의 농도로 처리한 후 소량의 히스티딘을 포함하는 배지에서 48 시간 동안 배양하고, 콜로니 수를 카운팅하였다.
그 결과 각 농도별로 3 회 테스트하였으나 콜로니가 발생한 배지가 없는 것을 확인함으로써, 돌연변이가 발생한 시험군은 없음을 확인하였다.
[시험예 7] 구조 안정성 평가
알로페론은 혈청 내 안정성이 낮다. 이에, 단독의 알로페론과 비교하여 본 발명의 복합체가 장기적으로 구조 안정성을 갖는지 평가하였다.
구체적으로, 인간혈청 10ml에 알로페론과 상동성이 없는 랜덤서열의 펩티드(음성 대조군), 알로페론 1 펩티드, 및 실시예 1을 각각 50 ng/ml로 녹이고, 동적 광산란법을 이용하여 입자의 크기를 측정하였다(0시간). 그런 다음 상기 각 샘플을 37℃ 5%, CO2 환경에서 1, 3, 5 및 7일 동안 방치 후 해당 시간마다 동적 광산란법을 이용하여 입자 크기 변화를 측정하였다. 각 샘플은 6개씩 평가하였고, 그 평균을 내어 혈청 첨가 직후의 입자 크기를 100%로 하고 이를 기준으로 입자의 크기를 %로 하기의 표 8 및 도 6에 나타내었다.

그 결과, 실시예 1의 경우 알로페론 단독 형태 보다 현저하게 우수한 혈청 안정성을 가짐을 확인하였다.[시험예 8] 다양한 바이러스에 대한 항바이러스 활성 평가
인플루엔자바이러스 외에 다양한 바이러스에 대해 면역 활성 유도에 의한 항바이러스 효과를 가질 수 있는지 평가하였다.
항바이러스 활성 분석에 사용된 엔테로바이러스로서는 코사키 A16 바이러스, 코사키 B3 바이러스, 코사키 B4 바이러스 및 엔테로바이러스 71(EV71), 헤르페스 바이러스로서 헤르페스 심플렉스 타입 1 및 2 (HSV 1/2, VR-734, 1383), 헤르페스조스터(HZV) (VR-1367DQ)는 ATCC(American Type Culture Collection)에서 구입하였다. 엔테로바이러스로서 코사키 A16 바이러스(A16), 코사키 B3 바이러스(B3), 코사키 B4 바이러스(B4), 엔테로바이러스 71(EV71)은 Vero 세포, 헤르페스 바이러스는 NIH3T3-E1 세포에서 증식 배양하고 각각의 바이러스 역가를 측정하여 TCID50에 해당하는 바이러스 용액을 산출하였다.
바이러스 증식 억제능을 측정하기 위해서, 각 배양 세포 2 × 104개를 96 웰 플레이트의 각 웰에 넣고 24 시간 배양하였다. 24 시간 후에 각 웰의 배양 상등액을 제거하고, TCID 50이 되도록 희석한 각 바이러스 용액 90 ㎕을 각 웰에 넣은 다음, 상기 실시예1, 알로페론1 펩티드 및 대조군을 각각 50 ng/ml의 농도로 배지 혼합액 10 ㎕에 희석하여 각 웰에 투여하고 3일 이후 바이러스 증식 억제능을 측정하였다.
바이러스 증식 억제능 측정은 권두한 등이 발명한 등록 특허 제682069호에 제시된 방법에 따라 실시하였다. 바이러스 감염시험이 종료된 후에 각 웰에 10% TCA(trichloroacetic acid)를 100㎕씩 첨가한 후 1 시간 동안 4℃에 방치하고 증류수로 수회 세척하였다. 실온에서 건조시킨 후 1%(v/v) 아세트산에 녹인 0.4%(w/v) SRB(sulforhodamine B) 용액 100 ㎕를 첨가해 30분 동안 염색시켰다. 세포와 결합하지 않은 SRB 염색액은 1%(v/v) 아세트산으로 수회 세척한 다음 다시 건조시켰다. 각 웰에 10 mM 트리스 용액(pH 10.5) 100 ㎕를 각 웰에 가하여 세포와 결합되어 있는 염색제를 충분히 녹인 후 560 nm에서 흡광도를 측정하여 그 값을 바이러스만을 처리한 대조군과 바이러스와 약물을 병행 처리한 군의 비율로서 바이러스 증식억제능을 평가하였다. 결과에 대한 통계는 동일한 조건에서 수행한 3번의 실험 결과 수치에 대한 평균값과 표준편차로 계산한 값을 제시하였다.

상기 표 9에 나타낸 바와 같이, 실시예 1은 코사키바이러스 A16 형에서는 88%, B3형에서는 82%, B5에서는 79%, 엔테로바이러스 EV71에서는 84%, 헤르페스심플렉스 1형에서는 90%, 헤르페스심플렉스 2형에서는 94% 그리고 헤르페스조스터에서는 98%의 감염억제능을 보여 음성대조군에 비해서는 5 ~ 60배, 알로페론1 처리보다 2~4배 이상 항바이러스능이 뛰어났다.
본 발명은 면역활성을 유도하는 장기 안정성을 가지는 알로페론 복합체에 관한 것으로서, 건강기능식품 관련 산업에서 이용 가능성이 있다.
