Description
본 발명은 단풍나무 추출물을 유효성분으로 포함하는 항바이러스용 약학 조성물; 단풍나무 추출물을 유효성분으로 포함하는 인플루엔자 바이러스 감염질환의 예방 또는 치료용 약학 조성물; 상기 약학 조성물을 인플루엔자 바이러스 감염질환 증상이 나타나거나 또는 나타날 우려가 있는 개체에 투여하는 단계를 포함하는 인플루엔자 바이러스 감염질환의 예방 또는 치료 방법; 단풍나무 추출물을 유효성분으로 포함하는 조성물의 항바이러스 용도 에 관한 것이다.
바이러스(virus)는 라틴어로 독성물질을 의미하며, 세균여과지(0.22㎛)를 통과하는 일군의 감염형 병원성 입자이다. 바이러스는 숙주세포의 종류에 따라 박테리오파지, 식물 바이러스, 동물 바이러스로 분류하기도 하며, 핵산의 종류에 따라 DNA 바이러스, RNA 바이러스로 분류할 수 있다. 최근 신종플루, AI 및 구제역 등 다양한 바이러스 질병이 사회적으로 큰 문제를 일으켰으며, 이에 따라 바이러스 질병의 효과적인 대책에 대한 고민이 사회적으로 큰 관심을 불러일으키고 있다.
현재 바이러스성 질병을 예방하기 위한 가장 좋은 방법은 백신접종이지만, 바이러스에 의한 질병의 경우, 대체로 많은 바이러스 혈청형(아형) 생성 등에 따른 백신의 효율성 문제가 중요하게 제기되고 있다. 이러한 백신의 문제점을 보완해 줄 수 있는 바이러스 예방용 억제제의 개발 및 보급은 중요한 사항이며, 이를 위해 특히 바이러스에 대한 초기 방어시스템인 생체 내 선천적 면역시스템을 자극하여 개체 동물의 면역력을 높여주는 예방제제의 발굴 및 개발은 중요한 제제 개발 방법이 될 수 있다.
한편, 교통수단의 발달과 도시화에 따른 인구의 집중으로 인하여, 공기 중으로 전파되는 인플루엔자, 폐렴균 등에 의한 호흡기 감염성 질병이 공공에 심각한 위협이 되고 있다.
인플루엔자 바이러스(Influenza virus)는 오르소믹소 계통(Family Orthomyxoviridae)에 속하는 RNA 바이러스로서 혈청형은 A형, B형, C형 등 3가지로 구분된다. 인플루엔자바이러스의 증식을 억제하는 대표적인 항바이러스 제제로는 아만타딘(amantadine)과 리만타딘(rimantadine)이 있으나, 이들 두 가지 항바이러스 제제들은 혈청형 A형 인플루엔자바이러스에만 효과적이며, M2 단백질이 없는 혈청형 B형 인플루엔자바이러스에는 효과가 없는 것으로 확인되었다. 또한, 아만타딘과 리만타딘은 사용 시 인플루엔자바이러스 M2 단백질의 이온채널기능에 영향을 미치지 못하는 변이 바이러스의 출현이 매우 쉽게 일어나는 단점이 있는 것으로 확인되고 있다. 이러한 단점을 보완하기 위하여 16종의 모든 혈청형 A형 인플루엔자바이러스와 혈청형 B형 인플루엔자바이러스에 효과적인 항바이러스 제제로 자나미비르(zanamivir)와 오셀타미비르(oseltamivir)가 개발되었다. 그러나 자나미비르는 흡입 및 정맥 투여해야 하는 단점이 있으며, 오셀타미비르는 경구투여가 가능하나, 최근 내성 바이러스의 출현 보고와 경구투여 시 구토와 현기증 등의 부작용이 있어 단점으로 지적되고 있다.
이러한 상황에서, 최근 해외 및 국내에서 기존의 항바이러스 제제의 단점을 극복하고자 하는 많은 노력들이 이루어지고 있으며, 그 중의 하나로 국내에서는 생약 추출물 및 식물 추출물을 대상으로 항바이러스 효능에 대한 연구가 진행중이기는 하지만, 아직까지는 미비한 실정이므로, 기존의 항바이러스 제제의 단점을 극복하여 독성 및 부작용이 거의 없이 우수한 선천면역 증진 효과 및 항바이러스 활성을 발휘할 수 있는 조성물의 개발이 필요한 실정이다.
한편, 단풍나무(Acer palmatum Thunb.)는 낙엽 활목 교목으로, 한방에서는 단풍나무의 뿌리껍질과 가지를 해독 효과의 약재로 사용하고, 무릎관절염으로 통증이 심할 때 물에 넣고 달여서 복용하며, 골절상을 입었을 때 오가피를 배합해서 사용한다. 단풍나무의 치주질환 (KR 10-2021-0098887 A) 또는 전립선암 (KR 10-2021-0151509 A)에 대하여는 알려져 있으나, 항바이러스 활성에 관해 보고된 바 없다.
본 발명자들은 신규한 인플루엔자 바이러스 치료제를 개발하기 위한 노력을 계속한 결과, 안정성이 보장된 단풍나무 추출물이 인플루엔자 바이러스 증식을 억제하는 우수한 효과가 있음을 확인함으로써, 본 발명을 완성하였다.
본 발명의 목적은 단풍나무 추출물을 유효성분으로 포함하는 항바이러스용 약학 조성물을 제공하는 것이다.
본 발명의 다른 목적은 단풍나무 추출물을 유효성분으로 포함하는 인플루엔자 바이러스 감염질환의 예방 또는 치료용 약학 조성물을 제공하는 것이다.
본 발명의 또 다른 목적은 상기 조성물을 인플루엔자 바이러스 감염질환 증상이 나타나거나 또는 나타날 우려가 있는 개체에 투여하는 단계를 포함하는 인플루엔자 바이러스 감염질환의 예방 또는 치료 방법을 제공하는 것이다.
본 발명의 또 다른 목적은 단풍나무 추출물을 유효성분으로 포함하는 항바이러스용 건강기능식품 조성물을 제공하는 것이다.

본 발명의 용어, "항바이러스"는 바이러스 감역을 억제하는 성질을 말하며, 상기 "바이러스"란 동물, 식물, 세균 등 살아 있는 세포에 기생하고, 세포 안에서만 증식이 가능한 미생물을 의미한다. 본 발명에서 바이러스는 인플루엔자 바이러스일 수 있으나, 이에 제한되지 않는다.
본 발명에서, "인플루엔자 바이러스"는 오르소믹소 계통(Family Orthomyxoviridae)에 속하는 RNA 바이러스로서 혈청형은 A형, B형, C형 등 3가지로 구분된다. A형 인플루엔자 바이러스의 혈청형은 바이러스 표면의 두 가지 단백질인 해마글루티닌(Hemagglutinin: HA), 뉴라미니다제(Neuraminidase: NA)의 종류에 따라 구분되며, 혈청형에 따라 144종류(HA형 16종과 NA형 9종)로 분류할 수 있다. B, C형 바이러스의 경우 A형만큼 변이가 빠르게 일어나지 않기 때문에 이러한 구분수단을 따로 사용하지 않는다. 인플루엔자 바이러스는 독감, 감기, 인후염, 기관지염, 폐렴을 일으킬 수 있으며, 특히 조류독감, 돼지독감 또는 염소독감 등을 야기시킬 수 있다.
본 발명에서 인플루엔자 바이러스는 인플루엔자 A타입 또는 인플루엔자 B 타입일 수 있으며, 구체적으로, H1N1, H2N2, H3N2, H3N8, H5N1, H5N6, H7N2, H7N3, H7N7, H7N9 또는 인플루엔자 바이러스 B일 수 있으나, 이에 제한되지 않는다.
본 발명의 일 구현예에서, 단풍나무 잎 추출물과 인플루엔자 바이러스를 동시 감염시킨 후 관찰한 결과, 단풍나무 잎 추출물을 처리한 경우 바이러스 증식에 의한 형광 발현 정도가 대조군 대비 감소하였고 (도 4), 인플루엔자 바이러스 단백질 발현을 억제시키는 것을 확인하였다 (도 5). 뿐만 아니라, 단풍나무 잎 추출물은 세포사멸 효과를 감소시켜 Raw 264.7 세포생존률이 현저히 증가함을 확인하였다. 이를 통해, 단풍나무 잎 추출물이 인플루엔자 바이러스 감염 억제 효과가 우수함을 확인하였다.
본 발명의 용어, "추출물"은 상기 단풍나무를 추출 처리하여 얻어지는 추출액, 상기 추출액의 희석액이나 농축액, 상기 추출액을 건조하여 얻어지는 건조물, 상기 추출액의 조정제물이나 정제물, 또는 이들의 혼합물 등, 추출액 자체 및 추출액을 이용하여 형성 가능한 모든 제형의 추출물을 포함한다.
상기 단풍나무를 추출하는 방법은 특별히 제한되지 않으며, 당해 기술 분야에서 통상적으로 사용하는 방법에 따라 추출할 수 있다. 상기 추출 방법의 비제한적인 예로는, 열수 추출법, 초음파 추출법, 상온 추출법, 냉침 추출법, 여과법, 환류 추출법, 증기 추출법 등을 들 수 있으며, 이들은 단독으로 수행되거나 2종 이상의 방법을 병용하여 수행될 수 있다.
본 발명에서 상기 단풍나무를 추출하는데 사용되는 추출 용매의 종류는 특별히 제한되지 않으며, 당해 기술 분야에서 공지된 임의의 용매를 사용할 수 있다. 상기 추출 용매의 비제한적인 예로는 물, 탄소수 1 내지 4의 알코올 또는 이들의 혼합 용매 등을 들 수 있으며, 이들은 단독으로 사용되거나 1종 이상 혼합하여 사용될 수 있다. 구체적으로 물 또는 에탄올이 사용될 수 있지만, 이에 제한되는 것은 아니며, 상기 에탄올은 10 내지 99%, 더욱 구체적으로 20 내지 50% 에탄올일 수 있지만, 이에 제한되는 것은 아니다.
본 발명에서, 상기 단풍나무 추출물은 단풍나무 알코올 또는 열수 추출물인 것일 수 있고, 더욱 구체적으로 에탄올 추출물일 수 있으나, 항바이러스 효과를 나타내는 한, 이에 제한되지 않는다.
본 발명의 일 구현예에서는, 단풍나무 추출물을 제조하기 위하여, 단풍나무 잎 분쇄물을 에탄올에 넣고 실온 침적하여 추출한 뒤, 상기 추출물을 감압농축하고 동결건조하여 단풍나무 잎 에탄올 추출물을 제조하였으며, 마우스 대식 세포주 RAW 264.7 세포에 대하여 세포 독성이 없음을 확인하였다 (도 1).
본 발명의 용어, "약학 조성물"은 총 조성물의 중량 대비 상기 단풍나무 추출물을 0.000001 내지 80 중량%로 포함할 수 있으며, 구체적으로 0.00001 중량% 내지 40 중량%로 포함할 수 있으나, 이에 제한되는 것은 아니다.
상기 약학 조성물은 약학 조성물의 제조에 통상적으로 사용하는 약학적으로 허용가능한 담체, 부형제 또는 희석제를 추가로 포함할 수 있고, 상기 담체는 비자연적 담체(non-naturally occuring carrier)를 포함할 수 있다.

이하 본 발명을 실시예 및 실험예를 통하여 보다 상세하게 설명한다. 그러나 이들 실시예 및 실험예는 본 발명을 예시적으로 설명하기 위한 것으로 본 발명의 범위가 이들 실시예 및 실험예에 한정되는 것은 아니다.
실험예 1. 단풍나무 잎 추출물 제조
단풍나무 (아처 팔마툼, Acer palmatum Thunb.) 잎 분쇄물 1700g을 17L의 25% 에탄올에 넣고 24시간 실온 침적하여 추출하였다. 그 후, 상기 추출물을 감압농축하고 동결건조하여 단풍나무 잎 25% 에탄올 추출물(이하, 'KIOM-2015E'로 명명함) 78.57g을 수득하였다.
실시예 1: RAW 264.7 세포 배양
본 발명의 단풍나무 잎 추출물의 항 바이러스 효능 실험을 위해, 마우스 대식 세포주 RAW 264.7 세포를 준비하여 10% 소 태아 혈청 (Fetal bovine serum, FBS), 100 U/ml 페니실린 (penicillin) 및 100 μg/ml 스트렙토마이신 (streptomycin)이 포함된 RPMI-1640 배지 (Hyclone)를 사용하여 배양하였다. 상기 세포는 37℃, 95% 습도, 5% CO2 조건의 배양기에서 배양하였으며 2 내지 3일마다 새로운 배양액으로 계대 배양하였다.
실시예 2: 단풍나무 잎 추출물의 세포 독성 분석
단풍나무 잎 추출물의 세포 독성이 없는 농도를 확인하기 위하여, RAW 264.7 세포를 96 well 세포 플레이트 seeding하였고 (1 x 105/ well), 단풍나무 잎 추출물을 1 μg/ml, 10 μg/ml, 50 μg/ml, 100 μg/ml, 200 μg/ml, 500 μg/ml, 1000 μg/ml 농도로 24시간 처리하였다. 24시간 배양 후, CCK-8 시약을 10 μl 넣고 2시간 반응시킨 후 450nm 파장에서 흡광도를 측정하였다 (도 1).
그 결과, 1 μg/ml 내지 500 μg/ml 농도에서 세포독성이 없음을 확인하였다. 본 발명의 단풍나무 추출물은 인플루엔자 바이러스 억제함에 있어 독성 없이 안전하게 사용될 수 있음을 알 수 있었다.
실시예 3: 단풍나무 잎 추출물의 항바이러스 효능 확인
단풍나무 잎 추출물의 인플루엔자 바이러스 감염에 억제 효능을 확인하기 위해 형광 현미경을 이용하여 확인하였으며, 구체적인 실험 방법은 하기와 같다.
12-well 플레이트에 Raw 264.7 세포(5X105 cell/well)를 배양한 후, 상기 실험예 1에서 제조한 단풍나무 잎 추출물 10 μg/ml, 50 μg/ml, 또는 100 μg/ml와 형광 단백질 (green fluorescence protein; gfp)를 부착한 인플루엔자 바이러스 (PR8-GFP)를 동시 감염시키고, 24시간 후 바이러스의 복제(replication)를 gfp 형광 발현을 통해 관찰하였다 (도 2).
이어서, 상기 세포를 원심분리하여 모아서 4% 파라포름알데하이드 용액으로 고정한 후, PBS (Phosphate buffered saline) 용액에 재현탁하여 유세포분석기 (FACS, Fluorescence activated cell sorter)를 활용하여 형광의 발현 정도를 분석하였으며 (도 3), 상기 유세포분석기로 얻은 형광값을 수치화하여 PR8-GFP 바이러스에 의한 형광값을 1로 기준으로 하여 상대적으로 비교 분석하였다 (도 4).
Mock은 음성 대조군, PR8-GFP는 인플루엔자 바이러스 감염군, PR8-GFP/단풍나무 잎 10 μg/ml은 바이러스와 단풍나무 잎 10 μg/ml 동시처리군, PR8-GFP/단풍나무 잎 50 μg/ml은 바이러스와 단풍나무 잎 50 μg/ml 동시처리군, PR8-GFP/단풍나무 잎 100 μg/ml은 바이러스와 단풍나무 잎 100 μg/ml 동시처리군을 나타낸다.
그 결과, 단풍나무 잎 추출물과 인플루엔자 바이러스를 동시에 감염시킨 경우, 농도의존적으로 바이러스 증식에 의한 형광이 감소되는 것을 확인하였으며, 특히 단풍나무 잎 추출물 50 μg/ml 농도 이상에서 인플루엔자 바이러스에 의한 GFP 형광값이 현저히 감소함을 확인하였다. 따라서 본 발명의 단풍나무 잎 추출물이 효과적으로 인플루엔자 바이러스의 감염률을 억제하는 효능이 있음을 확인하였다.
실시예 4: 형광면역분석법 (Immuno fluorescence technique)을 통한 인플루엔자 바이러스 단백질 발현 억제 효능 확인
단풍나무 잎 추출물의 인플루엔자 바이러스에 대한 항바이러스의 효능이 확인됨에 따라 단풍나무 잎 추출물이 바이러스 단백질 발현에도 영향을 미치는지 여부를 형광면역분석법을 이용하여 확인하였으며, 구체적인 실험 방법은 하기와 같다.
12-well 플레이트에 Raw 264.7 세포(5X105 cell/well)를 배양한 후, 상기 실험예 1에서 제조한 단풍나무 잎 추출물 100 μg/ml와 인플루엔자 바이러스 (PR8-GFP)를 Raw 264.7 세포에 동시에 처리하여 24시간 감염시킨 후 세포를 PBS로 세척하였다. 그 후, 100% 차가운 메탄올 용액에 5분, 3.7% 포르말린 용액에 10분간 고정하였다. 1% BSA와 0.05% tween-20이 함유된 PBS 용액에서 1시간 blocking, 바이러스 단백질 M2 (Matrix protein 2), NP (Nucleoprotein), HA (Hemagglutinin), NA (Neuraminidase))에 대한 1차 항체에 실온에서 1시간 반응시킨 후 PBS로 3번 세척한 후 Alexa594가 결합된 2차 항체와 1시간 반응하였다 (Red 형광). PBS로 3번 세척한 후 hoechst 33342 용액을 첨가하여 핵을 염색한 후 (blue 형광), 형광현미경으로 merge하여 두 가지 색의 형광을 겹쳐서 확인하였다 (도 5).
Hoechst 33342는 세포의 핵을 염색하며, Merge는 핵과 바이러스 단백질의 위치를 동시에 표시한 것이고, Brightfield는 일반현미경으로 세포를 확인한 결과이다. Medium은 음성 대조군, PR8-GFP은 인플루엔자 바이러스 감염군, PR8-GFP/단풍나무 잎은 바이러스와 단풍나무 잎 200 μg/ml을 동시에 처리한 군을 나타낸다.
그 결과, 단풍나무 잎 추출물과 인플루엔자 바이러스를 동시에 감염시킨 경우, 바이러스 단백질 M2, NP, HA, NA 모두 현저히 감소되었음을 알 수 있는바, 단풍나무 잎 추출물에 의해 바이러스 단백질의 발현이 억제됨을 확인하였다. 본 발명의 단풍나무 잎 추출물이 효과적으로 바이러스 단백질의 발현을 억제함으로 항바이러스 효능이 있음을 확인하였다.
실시예 5: 인플루엔자 A 타입의 H1N1, H3N2와 인플루엔자 B에 의한 세포살상효과 (CPE, cytopathic effect) 억제 효능 확인
바이러스에 감염된 세포가 죽으면서 나타나는 형태적 변화를 세포살상 현상 (CPE)이라 하고, 단풍나무 잎 추출물이 인플루엔자 바이러스에 의한 CPE가 억제되어 세포 생존률이 증가하는지 확인하기 위하여 CCK-8 어세이를 진행하였는바, 구체적인 실험 방법은 하기와 같다.
12-well 플레이트에 Raw 264.7 세포(5X105cell/well)를 배양한 후, 단풍나무 잎 추출물 0, 10 μg/ml, 50 μg/ml, 100 μg/ml, 또는 200 μg/ml와 인플루엔자 바이러스A의 H1N1 야생형 바이러스, H3N2 타입, 또는 인플루엔자 바이러스 B (IBV)와 동시 감염시켰다. 그 후, 24시간 배양한 세포에 CCK-8 용액을 넣어서 2시간 반응시킨 후, 450nm 파장에서 흡광도를 측정하였으며, 세포의 생존률을 배지에 대한 백분율로 나타내었다 (도 6).
Mock는 음성 대조군, H1N1, H3N2, IBV는 인플루엔자 바이러스 감염군, H1N1, H3N2, IBV/단풍나무 잎 10 μg/ml는 단풍나무 잎 10 μg/ml 처리군, H1N1, H3N2, IBV/단풍나무 잎 50 μg/ml는 단풍나무 잎 50 μg/ml 처리군, H1N1, H3N2, IBV/단풍나무 잎 100 μg/ml는 단풍나무 잎 100 μg/ml 처리군, H1N1, H3N2, IBV /단풍나무 잎 200 μg/ml는 단풍나무 잎 200 μg/ml 처리군을 나타낸다.
그 결과, 단풍나무 잎 추출물과 H1N1, H3N2, IBV 인플루엔자 바이러스를 동시에 감염시킨 경우, CPE를 농도의존적으로 억제하여 세포 생존률이 증가하는 것을 확인하였다.
본 발명의 단풍나무 잎 추출물을 처리한 후 H1N1, H3N2 및 인플루엔자 B 바이러스에 의한 CPE를 농도의존적으로 억제하여 세포 생존률을 증가시키는 것을 확인하였으며, 특히 단풍나무 잎 추출물 50 μg/ml 농도 이상에서 세포 생존률이 현저히 증가함을 확인하였다. 본 발명의 단풍나무 잎 추출물이 바이러스의 복제에 의한 CPE를 억제함으로 인플루엔자 바이러스 A 뿐만 아니라, 인플루엔자 바이러스 B에서도 항바이러스 효능이 있음을 확인하였다.
이상의 설명으로부터, 본 발명이 속하는 기술분야의 당업자는 본 발명이 그 기술적 사상이나 필수적 특징을 변경하지 않고서 다른 구체적인 형태로 실시될 수 있다는 것을 이해할 수 있을 것이다. 이와 관련하여, 이상에서 기술한 실시예들은 모든 면에서 예시적인 것이며 한정적인 것이 아닌 것으로 이해해야만 한다. 본 발명의 범위는 상기 상세한 설명보다는 후술하는 특허 청구범위의 의미 및 범위 그리고 그 등가 개념으로부터 도출되는 모든 변경 또는 변형된 형태가 본 발명의 범위에 포함되는 것으로 해석되어야 한다.
Claims (10)
- 단풍나무 추출물을 유효성분으로 포함하는 항바이러스용 약학 조성물.
- 제1항에 있어서, 상기 단풍나무 추출물은 단풍나무 잎 추출물인 것인, 약학 조성물.
- 제1항에 있어서, 상기 바이러스는 인플루엔자 바이러스인 것인, 약학 조성물.
- 제1항에 있어서, 상기 단풍나무 추출물은 물, 탄소수 1 내지 4의 알코올, 및 이들의 혼합용매로 구성되는 군으로부터 선택되는 용매로 추출하여 제조되는 것인, 약학 조성물.
- 제2항에 있어서, 상기 추출은 열수 추출, 초음파 추출, 상온추출, 냉침추출, 환류 냉각 추출 또는 증기 추출인 것인, 약학 조성물.
- 단풍나무 추출물을 유효성분으로 포함하는 인플루엔자 바이러스 감염질환의 예방 또는 치료용 약학 조성물.
- 단풍나무 추출물을 유효성분으로 포함하는 조성물을 인플루엔자 바이러스 감염질환 증상이 나타나거나 또는 나타날 우려가 있는 개체에 투여하는 단계를 포함하는 인플루엔자 바이러스 감염질환의 예방 또는 치료 방법.
- 단풍나무 추출물을 유효성분으로 포함하는 항바이러스용 건강기능식품 조성물.
- 단풍나무 추출물을 유효성분으로 포함하는 항바이러스용 의약외품 조성물.
